Abstract:It is an interesting research topic to study the interfacial interactions between hemicellulose and cellulose, specifically how hemicellulose's structure affects its binding to cellulose nanofibers. Our research proposes that dispersion interaction play an important role in this interfacial interaction, more so than electrostatic forces when considering the adherence of cellulose to xylan. To quantify these interactions, the Atomic Force Microscope (AFM) colloidal probe technique is applied to measure the intermolecular forces between cellulose nanofibers, which are attached to the probe and xylan. These measured forces are then analyzed in relation to the length, diameter and functional groups of the nanocellulose, as well as the molecular weight and side chains of the xylan. Moreover, the predominance of dispersion forces by contrasting the adhesive forces before and after the grafting of a large nonpolar group onto xylan. This modification significantly reduces contact between the cellulose and xylan backbone, thereby markedly diminishing the dispersion interactions. Parallel to the AFM experiments, molecular dynamics (MD) simulations corroborate the experimental results and support our hypotheses. Collectively, these findings contribute to a deeper understanding of polysaccharide interactions within lignocellulose.
Progress and Opportunities in the Characterization of Cellulose - An Important Regulator of Cell Wall Growth and Mechanics

Abstract:The plant cell wall is a dynamic network of several biopolymers and structural proteins including cellulose, pectin, hemicellulose and lignin. Cellulose is one of the main load bearing components of this complex, heterogeneous structure, and in this way, is an important regulator of cell wall growth and mechanics. Glucan chains of cellulose aggregate via hydrogen bonds and van der Waals forces to form long thread-like crystalline structures called cellulose microfibrils. The shape, size, and crystallinity of these microfibrils are important structural parameters that influence mechanical properties of the cell wall and these parameters are likely important determinants of cell wall digestibility for biofuel conversion. Cellulose-cellulose and cellulose-matrix interactions also contribute to the regulation of the mechanics and growth of the cell wall. As a consequence, much emphasis has been placed on extracting valuable structural details about cell wall components from several techniques, either individually or in combination, including diffraction/scattering, microscopy, and spectroscopy. In this review, we describe efforts to characterize the organization of cellulose in plant cell walls. X-ray scattering reveals the size and orientation of microfibrils; diffraction reveals unit lattice parameters and crystallinity. The presence of different cell wall components, their physical and chemical states, and their alignment and orientation have been identified by Infrared, Raman, Nuclear Magnetic Resonance, and Sum Frequency Generation spectroscopy. Direct visualization of cell wall components, their network-like structure, and interactions between different components has also been made possible through a host of microscopic imaging techniques including scanning electron microscopy, transmission electron microscopy, and atomic force microscopy. This review highlights advantages and limitations of different analytical techniques for characterizing cellulose structure and its interaction with other wall polymers. We also delineate emerging opportunities for future developments of structural characterization tools and multi-modal analyses of cellulose and plant cell walls. Ultimately, elucidation of the structure of plant cell walls across multiple length scales will be imperative for establishing structure-property relationships to link cell wall structure to control of growth and mechanics.
Molecular and thermodynamic insights into interfacial interactions between collagen and cellulose investigated by molecular dynamics simulation and umbrella sampling

Abstract:Cellulose/collagen composites have been widely used in biomedicine and tissue engineering. Interfacial interactions are crucial in determining the final properties of cellulose/collagen composite. Molecular dynamics simulations were carried out to gain insights into the interactions between cellulose and collagen. It has been found that the structure of collagen remained intact during adsorption. The results derived from umbrella sampling showed that (110) and ( ) faces exhibited the strongest affinity with collagen (100) face came the second and (010) the last, which could be attributed to the surface roughness and hydrogen-bonding linkers involved water molecules. Cellulose planes with flat surfaces and the capability to form hydrogen-bonding linkers produce stronger affinity with collagen. The occupancy of hydrogen bonds formed between cellulose and collagen was low and not significantly contributive to the binding affinity. These findings provided insights into the interactions between cellulose and collagen at the molecular level, which may guide the design and fabrication of cellulose/collagen composites.

Tuning Secondary Bonding Forces for Accelerated Dispersion of Cellulose Nanofibers

Abstract:Cellulose nanofibers are elementary building blocks of a wide variety of advanced functional materials. Nevertheless, the strong interfacial secondary bonds between cellulose molecular chains are against harvesting cellulose nanofibers from cellulose bundles that are built by nature, such as in plants. To address the above challenge, this work combines first-principles calculations, molecular dynamics simulations, and experimental implementations to show that an external electric field can facilitate and accelerate the dispersion of cellulose nanofibers by reducing the interfacial secondary bonding forces among cellulose molecular chains. An electro-dispersion method is implemented to apply computational findings. Experimental results show that the electric field can not only accelerate the dispersion process of cellulose nanofibers but also tune the mechanical properties of cellulose nanopapers assembled from the obtained cellulose nanofibers. Our discovery suggests new possibilities for developing building blocks of advanced functional materials from sustainable biological polymers where secondary bonds are essential.
Xylan inhibition of cellulase binding and processivity observed at single-molecule resolution

Abstract:Efficient cellulose degradation by cellulase enzymes is crucial for using lignocellulosic biomass in bioenergy production. In the cell wall of plants, cellulose is bound by lignin and hemicellulose, which are key factors contributing to the recalcitrance of plant biomass. These non-cellulosic cell wall components are known to interfere with the function of cellulolytic enzymes. While the effects of lignin have been studied extensively, the contribution of xylan, the major hemicellulose in the secondary cell walls of plants, is often overlooked. To study those effects, we generated model cell wall composites by growing bacterial cellulose supplemented with varying concentrations of purified xylan. We used single-molecule microscopy to image and track fluorescently labeled Cel7A, a commonly used model cellulase, as it binds and hydrolyses cellulose in these synthetic composites. We found that minute amounts of xylan are sufficient to significantly inhibit the binding of Cel7A to cellulose. The inclusion of xylan also reduced considerably the proportion of moving enzyme molecules, without affecting their velocity and run length. We suggest that, when available at low concentrations, xylan thinly coats cellulose fibrils, and incorporates as continuous patches when available at higher concentrations. Non-productive binding of Cel7A to xylan was not found to be a major inhibition mechanism. Our results highlight the importance of targeting xylan removal during biomass processing and demonstrate the potential of using single-molecule imagining to study the activity and limitations of cellulolytic enzymes.

Abstract:The molecular architecture of plant secondary cell walls is still not resolved. There are several proposed structures for cellulose fibrils, the main component of plant cell walls and the conformation of other molecules is even less well known. Glucuronic acid (GlcA) substitution of xylan (GUX) enzymes, in CAZy family glycosyl transferase (GT)8, decorate the xylan backbone with various specific patterns of GlcA. It was recently discovered that dicot xylan has a domain with the side chain decorations distributed on every second unit of the backbone (xylose). If the xylan backbone folds in a similar way to glucan chains in cellulose (2-fold helix), this kind of arrangement may allow the undecorated side of the xylan chain to hydrogen bond with the hydrophilic surface of cellulose microfibrils. MD simulations suggest that such interactions are energetically stable. We discuss the possible role of this xylan decoration pattern in building of the plant cell wall.

Abstract:The plant cell wall (PCW) inspires the preparation of fiber-based biomaterials, particularly emphasizing exploiting the intrinsic interactions within the load-bearing cellulose and hemicellulose network. Due to experimental difficulties in studying and interpreting the interaction between these polysaccharides, this research presents a numerical model based on coarse-grained molecular dynamics that evaluates the mechanical properties of fiber composites. To validate the model and explain the structural and mechanical role of hemicelluloses, bacterial cellulose (BC) was synthesized in the presence of different concentrations of xylan, arabinoxylan, xyloglucan, or glucomannan and subjected to nano- and macroscale structural and mechanical characterization. The data obtained were used to interpret the effects of each hemicellulose on the mechanics of the BC-hemicellulose composite based on the sensitivity of the model. The mechanical properties of the resulting simulated networks agreed well with the experimental observations of the BC-hemicellulose composites. Increased xylan and arabinoxylan contents increased the macroscale mechanical properties, fiber modulus (xylan), and fiber width (arabinoxylan). The addition of xyloglucan increased the mechanical properties of the composites in the elastic deformation phase, associated with an increase in the fiber modulus. Adding glucomannan to the culture medium decreased all the mechanical properties studied while the fiber width increased.
Molecular modeling of the structural and dynamical properties of secondary plant cell walls: influence of lignin chemistry

Abstract:A modeling of lignified secondary plant cell walls adapted to grass has been achieved, using molecular dynamics for time up to 180 ns, applied to systems composed of cellulose, xylan, water, and lignin. The overall model, which was 70 nm thick for a volume of 74.4 nm(3), consisted of two crystalline cellulose layers, each being two molecules deep, separated by an interlayer space where the three other components were located. Whereas the cellulose and xylan chemistry was fixed, 18 lignin systems were considered that varied not only in guaiacyl, syringyl, and p-hydroxyphenyl composition, but also in chain length, linkage types, and the presence or absence of coumaryl units. The stabilized models showed a well-defined interface between xylan and cellulose, but some interpenetration of xylan into the lignin part of the models. A survey of the 18 models showed that their lignin component was amorphous and that their density profile was very variable and essentially model dependent. This variability was also reflected in the co-orientation of the lignin phenyl rings with respect to the cellulose surfaces, some systems showing some orientation whereas others did not. The pattern of void distribution accessible to water varied from one system to the next, but the overall void volume was systematically established at around 3%, accepting around 200 water molecules. The estimated mobility of the water molecules interacting with lignin was 1.5 times greater than that interacting with carbohydrates.

Abstract:Lignocellulosic biomass, encompassing cellulose, lignin and hemicellulose in plant secondary cell walls (SCWs), is the most abundant source of renewable materials on earth. Currently, fast-growing woody dicots such as Eucalyptus and Populus trees are major lignocellulosic (wood fiber) feedstocks for bioproducts such as pulp, paper, cellulose, textiles, bioplastics and other biomaterials. Processing wood for these products entails separating the biomass into its three main components as efficiently as possible without compromising yield. Glucuronoxylan (xylan), the main hemicellulose present in the SCWs of hardwood trees carries chemical modifications that are associated with SCW composition and ultrastructure, and affect the recalcitrance of woody biomass to industrial processing. In this review we highlight the importance of xylan properties for industrial wood fiber processing and how gaining a greater understanding of xylan biosynthesis, specifically xylan modification, could yield novel biotechnology approaches to reduce recalcitrance or introduce novel processing traits. Altering xylan modification patterns has recently become a focus of plant SCW studies due to early findings that altered modification patterns can yield beneficial biomass processing traits. Additionally, it has been noted that plants with altered xylan composition display metabolic differences linked to changes in precursor usage. We explore the possibility of using systems biology and systems genetics approaches to gain insight into the coordination of SCW formation with other interdependent biological processes. Acetyl-CoA, s-adenosylmethionine and nucleotide sugars are precursors needed for xylan modification, however, the pathways which produce metabolic pools during different stages of fiber cell wall formation still have to be identified and their co-regulation during SCW formation elucidated. The crucial dependence on precursor metabolism provides an opportunity to alter xylan modification patterns through metabolic engineering of one or more of these interdependent pathways. The complexity of xylan biosynthesis and modification is currently a stumbling point, but it may provide new avenues for woody biomass engineering that are not possible for other biopolymers.

Abstract:Abstract Structural organization of the plant cell wall is a key parameter for understanding anisotropic plant growth and mechanical behavior. Four imaging platforms were used to investigate the cell wall architecture of Miscanthus sinensis cv. internode tissue. Using transmission electron microscopy with potassium permanganate, we found a great degree of inhomogeneity in the layering structure (4–9 layers) of the sclerenchymatic fiber (Sf). However, the xylem vessel showed a single layer. Atomic force microscopy images revealed that the cellulose microfibrils (Mfs) deposited in the primary wall of the protoxylem vessel (Pxv) were disordered, while the secondary wall was composed of Mfs oriented in parallel in the cross and longitudinal section. Furthermore, Raman spectroscopy images indicated no variation in the Mf orientation of Pxv and the Mfs in Pxv were oriented more perpendicular to the cell axis than that of Sfs. Based on the integrated results, we have proposed an architectural model of Pxv composed of two layers: an outermost primary wall composed of a meshwork of Mfs and inner secondary wall containing parallel Mfs. This proposed model will support future ultrastructural analysis of plant cell walls in heterogeneous tissues, an area of increasing scientific interest particularly for liquid biofuel processing.

Abstract:Lignin and carbohydrate rich secondary plant cell walls are abundantly available in the biosphere, and is a notable renewable feedstock for biofuels, biomaterials with wide applicability. Particularly for construction applications, long-lived wood that is resistant to fungal degradation is highly desirable. Chemical modifications, such as acetylation, have been successfully demonstrated inhibit wood breakdown. X-ray fluorescence microscopy experiments have determined that acetylation hinders fungal attack by reducing moisture content while other experiments examining moisture-induced wood damage mechanisms observed that chemical transport through cell walls is reduced after acetylation. We investigate these hypotheses directly through molecular simulation, acetylating existing models for secondary plant cell wall structure to 5-18% weight-percent gain for additional acetylations on exposed hemicellulose and lignin hydroxyl groups. By comparing diffusive behavior for cell wall polymers, water, and select ions (Na+ and Fe3+), we can track the dynamics within the cell wall and identify the causal mechanisms for reduced transport and uptake by acetylated cell walls. We find that cell wall acetylation alone does not account for reduced transport. The most substantial changes in diffusion occur in a constant volume system where the additional acetylation displaces water, reducing the moisture content for the cell wall. Utilizing these simulations, we further analyze the interactions between ions and cell wall polymers and the evolution of water pockets within the structure. Ions interact more frequently with the acetyl group than the hydroxyl groups they replace, yielding to increased ion interactions on aggregate upon acetylation. These findings elucidate the molecular mechanism through which acetylation affects secondary plant cell wall at atomic resolution.

Abstract:Solvent-controlled extraction and precipitation are the most fundamental methods for obtaining hemicellulose from lignocellulosic biomass and purification processes. However, the dissolution and precipitation mechanisms involved have scarcely been mentioned. In this study, the molecular scale behavior of xylan-type hemicellulose during solvent-controlled extraction and precipitation is investigated using molecular dynamics (MD) simulations and density functional theory (DFT) calculations. To bring the model closer to the real extracted xylan, a high degree of polymerization (DP100) of xylan is established, and hemicelluloses with low DP (DP15 and DP50) are also investigated. Four phenomena are explained at the molecular level, including the influence of the polymerization degree and side chain on the solubility of xylan in water, the improvement of the xylan's solubility in NaOH, the precipitation of xylan in ethanol, and the acetyl group preservation of xylan in DMSO. This study contributes to an increased understanding of the dissolution and precipitation mechanisms of hemicellulose and provides a resource for the simulation of high DP hemicellulose, which gives a theoretical basis for the efficient extraction of high-purity hemicellulose as well as economic biorefining.

Direct Visualization of Straw Cell Walls by Afm

Abstract:The structural relationship of cellulose, hemicellulose, and lignin in plant cell walls is still a mystery needing to be explored. By using atomic force microscopy (AFM) the surface of straw at different layers was directly observed, and the structural characteristics were analyzed by topographic analysis and FT-IR spectra. It was found that a compact layer of wax covered the outside of the straw, which protects the straw from insects and microorganisms. At the boundary of the primary and second wall there appears a network structure of cellulose and hemicellulose, with some lignin localised on the surface of the network. It is consistent with the model of a cell wall suggested by Vincent. Inside the second cell wall, there is a layer mainly composed of a cellulose crystalline region. High-resolution AFM observation reveals that the crystalline structure consists of both triclinic and monoclinic unit cells. Abstract:The interaction mechanism of two plant cell wall polysaccharides, arabinoxylan and xyloglucan, with cellulose has been investigated by means of bacterial cellulose fermentation to mimic the cell wall biosynthesis process. The combination of small angle scattering techniques with XRD and SEM has enabled the identification of different structural features comprising hierarchically-assembled bacterial cellulose, i.e. cellulose microfibrils and ribbons. The SANS results have been described by a core–shell formalism, which accounts for the presence of regions with different solvent accessibility and supports the existence of microfibril sub-structure within the ribbons. Additionally, SAXS and XRD results suggest that the microfibril packing and crystalline structure are not affected by arabinoxylan, while xyloglucan interferes with the crystallization and assembly processes, resulting in less crystalline Iβ-rich microfibrils. This specific interaction mechanism is therefore crucial for the cellulose microfibril cross-linking effect of xyloglucan in plant cell walls. It is proposed that the distinct interaction mechanisms identified have their origin in the differential structural role of arabinoxylan and xyloglucan in plant cell walls.

Abstract:Polysaccharides play a crucial role in virtually all living systems. They also represent the biocompatible and fully sustainable component of a variety of nanoparticles, which are of increasing interest in biomedicine, food processing, cosmetics, and structural reinforcement of polymeric materials. The computational modeling of complex polysaccharide phases will assist in understanding the properties and behavior of all these systems. In this paper, structural, bonding, and mechanical properties of 10 wt % cellulose-callose hydrogels (β-glucans coexisting in plant cell walls) were investigated by atomistic simulations. Systems of this kind have recently been introduced in experiments revealing unexpected interactions between the polysaccharides. Starting from initial configurations inspired by X-ray diffraction data, atomistic models made of ∼1.6 × 10<sup>6</sup> atoms provide a qualitatively consistent view of these hydrogels, displaying stability, homogeneity, connectivity, and elastic properties beyond those of a liquid suspension. The simulation shows that the relatively homogeneous distribution of saccharide nanofibers and chains in water is not due to the solubility of cellulose and callose, but to the formation of a number of cross-links among the various sample components. The broad distribution of strength and elasticity among the links implies a degree of anharmonicity and irreversible deformation already evident at low external load. Besides the qualitative agreement with experimental observations, the simulation results display also quantitative disagreements in the estimation of elastic coefficients, such as the Young's modulus, that require further investigation. Complementary simulations of dense cellulose-callose mixtures (no hydrogels) highlight the role of callose in smoothing the contact surface of different nanofibers forming larger bundles. Cellulose-callose structures in these systems displayed an enhanced water uptake and delayed dye release when compared to cellulose alone, highlighting potential new applications as drug delivery scaffolds. The simulation trajectories provide a tuning and testing ground for the development of coarse-grained models that are required for the large scale investigation of mechanical properties of cellulose and callose mixtures in a watery environment.

Abstract:Interactions of water with cellulose are of both fundamental and technological importance. Here, we characterize the properties of water associated with cellulose using deuterium labeling, neutron scattering and molecular dynamics simulation. Quasi-elastic neutron scattering provided quantitative details about the dynamical relaxation processes that occur and was supported by structural characterization using small-angle neutron scattering and X-ray diffraction. We can unambiguously detect two populations of water associated with cellulose. The first is “non-freezing bound” water that gradually becomes mobile with increasing temperature and can be related to surface water. The second population is consistent with confined water that abruptly becomes mobile at ~260 K, and can be attributed to water that accumulates in the narrow spaces between the microfibrils. Quantitative analysis of the QENS data showed that, at 250 K, the water diffusion coefficient was 0.85 ± 0.04 × 10 −10 m 2 sec −1 and increased to 1.77 ± 0.09 × 10 −10 m 2 sec −1 at 265 K. MD simulations are in excellent agreement with the experiments and support the interpretation that water associated with cellulose exists in two dynamical populations. Our results provide clarity to previous work investigating the states of bound water and provide a new approach for probing water interactions with lignocellulose materials.

Abstract:Abstract We have performed molecular dynamics calculations using a revised version of the Gromos56Acarbo force field to understand the consequences of the different potential hydrogen bonding patterns on the structural stability and thermal behavior of the Iα and Iβ forms of native cellulose. For each allomorph, we considered three patterns of hydrogen bonds: two patterns obtained from neutron diffraction data refinement and a regular mixture of the two. Upon annealing, the hydrogen bonding schemes of cellulose Iβ, irrespective of the starting structure, re-arranged into the main hydrogen bond pattern experimentally observed (pattern A). On the other hand, the Iα structures, irrespective of the starting hydrogen bonding pattern, converged to a non-experimental structure where the adjacent chains are shifted along the chain direction by 0.12 nm in the hydrogen-bonded plane, and the hydroxymethyl group conformation alternates between gt and tg along the chain. The exotic structure in Iα might be a consequence of a deficiency in force field parameters and/or potential molecular arrangement in less crystalline cellulose.
