Abstract:1,2-Azaborines represent a unique class of benzene isosteres that have attracted interest for developing pharmaceuticals with better potency and bioavailability. However, it remains a long-standing challenge to prepare monocyclic 1,2-azaborines, particularly multi-substituted ones, in an efficient and modular manner. Here we report a straightforward method to directly access diverse multi-substituted 1,2-azaborines from readily available cyclopropyl imines/ketones and dibromoboranes under relatively mild conditions. The reaction is scalable, shows a broad substrate scope, and tolerates a range of functional groups. The utility of this method is demonstrated in the concise syntheses of BN isosteres of a PD-1/PD-L1 inhibitor and pyrethroid insecticide, bifenthrin. Combined experimental and computational mechanistic studies suggest that the reaction pathway involves boron-mediated cyclopropane ring-opening and base-mediated elimination, followed by an unusual low-barrier 6 π -electrocyclization accelerated by the BN/CC isomerism. This method is anticipated to find applications for the synthesis of BN-isostere analogues in medicinal chemistry, and the mechanistic insights gained here may guide developing other boron-mediated electrocyclizations.

Abstract:5-Azaindoles either with three different substituents at their 2-, 4-, and 6-positions or with two identical substituents at their 2- and 6-positions and a different one at the 4-position, were obtained in good to excellent isolated yields by a zirconocene-mediated multicomponent process. Each reaction involved four organic partners, comprising a Si-tethered diyne, one tBuCN component, and two (either different or identical) nitriles. All these four components were combined through the action of a Cp2ZrII species into a three-ring fused Zr/Si-containing organometallic complex in a perfectly chemo-and regioselective manner. This multicomponent reaction process consisted of three reaction steps, all of which were made clear through the isolation and characterization of their cor-responding organometallic intermediates: the zirconacyclopropene-azasila-cyclopentadienes 2, the allenyl-aza-zirconacycles 3, and the three-ring fused complexes 6. X-ray single-crystal structural analyses of two three-ring fused Zr/Si-containing intermediates and two 5-azaindoles unambiguously showed the positions of the different substituents and the regioselectivity. Iminopyrrole derivatives could be also highly selectively prepared from a Si-tethered diyne and two different nitriles.

Abstract:An efficient sequence based on the Ugi-azide reaction and rhodium(III)-catalyzed intermolecular annulation has been established for the preparation of tetrazole-isoquinolone/pyridone hybrids. Several N -acylaminomethyltetrazoles were reacted with arylacetylenes to form the hybrid products in moderate to very good yields. The method relies on the capacity of the rhodium catalyst to promote C(sp 2 )–H activation in the presence of a suitable directing group. The Ugi-azide reaction provides broad molecular diversity and enables the introduction of the tetrazole moiety, which may further assist the catalytic reaction by coordinating the metal center. The scope of the isoquinolones is very wide and may be extended to the preparation of complex compounds having heterocyclic moieties such as pyridone, furan, thiophene and pyrrole, as well as the corresponding benzo-fused derivatives. The developed procedure is simple, reproducible and does not require inert conditions.

Abstract:The (Fmes)BH2·SMe2 reagent (7) reacts sequentially with an acetylene and, e.g., xylylisonitrile in a convenient three-component reaction to give a series of unprecedented dihydro-1,3-azaborole derivatives 16. The tolane-derived example 16a was deprotonated and used as a ligand in organometallic chemistry. Compounds 16 served as the starting materials for the straightforward synthesis of various dihydro-1,3-azaborinine derivatives by treatment with an isonitrile. Abstract:A number of enamine-functionalized 1,2,3-triazole derivatives have been prepared via the Cu-catalyzed three-component reaction of terminal alkyne, azide, and 2H-azirine. The reaction proceeds through insertion of vinyl nitrene into the C-Cu bond of the triazolyl-Cu species, providing an efficient and step- and atom-economic approach to the enamine-bearing polysubstituted 1,2,3-triazoles. The resulting triazoles were easily transformed to trisubstituted pyrazoles in the presence of a Rh catalyst.

Abstract:The reaction of boroles with organic azides is known to straightforwardly yield 1,2‐azaborinines, which have generated significant interest as benzene bioisosteres. In this work, this powerful synthetic pattern is extended to a variety of structurally and chemically diverse boron azides. It provides a general, powerful borylation method to expand the chemistry of 1,2‐azaborinines, yielding boryl‐substituted 1,2‐azaborinines and 1,2‐azaborinine‐functionalized BN heteroarenes. The reaction of boroles with organic azides is known to straightforwardly yield 1,2‐azaborinines, which have generated significant interest as benzene bioisosteres. We show that this powerful synthetic pattern extends to a variety of structurally and chemically diverse boron azides, including recently reported diborane(4) azides. Abstract:A simple and efficient strategy for the chemoselective synthesis of (2 Z ,5 Z )-3-benzyl/alkyl-5-(2-oxo-5-arylfuran-3(2 H )-ylidene)-2-(phenylimino)thiazolidin-4-one derivatives is developed via a sequential three-component reaction of readily available phenacyl bromides, pyridine and methyl 2-(3-benzyl/alkyl-4-oxo-2-(phenylimino)thiazolidin-5-ylidene)acetates in acetonitrile at room temperature. The advantages of this one-pot sequential Michael addition/1,2-H shift/elimination of pyridine/intramolecular cyclization are highlighted by its low energy requirements (short reaction times at room temperature), excellent yields, the absence of a metal catalyst, and mild conditions. The product structures are characterized by spectroscopic techniques and single-crystal X-ray analysis. Abstract:A stereoselective N -alkenylation of azoles with alkynes and iodine(III) electrophile is reported. The reaction between various azoles and internal alkynes is mediated by benziodoxole triflate as the electrophile in a trans -fashion, affording azole-bearing vinylbenziodoxoles in moderate to good yields. The tolerable azole nuclei include pyrazole, indazole, 1,2,3-triazole, benzotriazole, and tetrazole. The iodanyl group in the product can be leveraged as a versatile synthetic handle, allowing for the preparation of hitherto inaccessible types of densely functionalized N -vinylazoles.

Abstract:A Pd-catalyzed highly selective tandem reaction of allenynes, aryl or alkenyl iodides, and sodium azide for the synthesis of bicyclic 1,2,3-triazole derivatives with a very broad substrate scope including monosubstituted, 1,3-disubstituted, and trisubstituted allenynes as well as different types of organic iodides has been reported. Control experiments showed that the reaction went through a cascade allene difunctionalization/Winstein allylic azide rearrangement/intramolecular azide-alkyne cycloaddition process.

Abstract:A pyrrolidine‐catalyzed three‐component reaction of azlactone, N,O‐acetal and alcohol for the synthesis of α,β‐diamino ester was reported. The N,O‐acetal severed as the imine equivalent to occur Mannich reaction with azlactone, and the in situ generated α‐functionalized azlactone subsequently underwent a ring‐opening process in the presence of alcohol. A series of α,β‐diamino esters were obtained in 36‐99% yields under mild reaction conditions. The product could be synthesized on gram‐scale and transformed to the α,β‐diamino acid.

Abstract:The first sequential acid-catalyzed propargylation/base-mediated aza-cycloisomerization between indolyl-benzimidazoles and propargylic alcohols is described. This protocol enables the one-pot construction of N-fused benzimidazo-β-carbolines in good yields. The synthetic utility of this approach is demonstrated by the assembly of an aza-helicene and also by a gram-scale reaction.
