Abstract:Müller cells are principal glial cells in rat retina and have attracted much attention in glaucoma studies. However, it is not clear whether adenosine and adenosine receptor (AR) antagonists play any roles in the regulation of potassium channels in Müller cells and subsequently in the promotion of glutamine synthetase (GS) and L-Glutamate/L-Aspartate Transporter (GLAST) functions. We found that chronic ocular hypertension (COH) in rat down-regulated Müller cells Kir2.1, Kir4.1, TASK-1, GS and GLAST expressions and attenuated the peak of inward potassium current. Retinal ganglion cells (RGC) count was lower in the COH rats than that in the sham operation animals. Intravitreal injection of selective A2A AR antagonist SCH442416 up-regulated Müller cell Kir4.1, TASK-1, GS and GLAST expressions and enhanced inward potassium currents compared with those in the COH rats with vehicle control. Meanwhile, the RGC count was higher following intravitreal injection of SCH442416 in the COH rats than that after vehicle injection. The fact that PKA inhibitor H-89 blocked these SCH442416 effects suggested that the PKA signaling pathway was involved in the observed ocular responses following the intravitreal SCH442416 injection.
Glycine Modulates the Center Response of ON Type Rod-Dominant Bipolar Cells in Carp Retina

Abstract:Effects of glycine on ON type rod-dominant bipolar cells (RBCs) were studied in isolated, superfused carp retina by intracellular recording technique and in carp retinal slice preparation by whole cell recording. Glycine of 4mM hyperpolarized RBCs and potentiated their light responses to large light spots, which was reversed by co-application of 10μM strychnine. It was further found that illumination of the receptive field surround did not affect the depolarizing center response of RBCs. The above result therefore suggests that glycine modulates the center response of RBCs. Focal application of glycine to either dendrites or axon terminals of RBCs failed to induce any currents in both isolated cell and retinal slice preparations. On the other hand, glycine of 4mM increased the amplitude of the scotopic electroretinographic PIII component, which reflects the activity of rod photoreceptors. It seems likely that modulation by glycine of the RBC center response may be in part ascribed to a consequence of the potentiation of rod responses by glycine.

Each subject was administered the Sixteen Personality Factor Questionnaire as a pre- and post-test measurement. Results reveal that some aspects of personality are affected by Rational Therapy group programs while others are not.
Effects of Convergent Strabismus on the Development of Physiologically Identified Retinogeniculate Axons in Cats.

Abstract:We have studied the effects of surgically induced convergent strabismus (esotropia) on the morphological development of retinogeniculate X and Y axon arbors in cats. Single axons were recorded in the lateral geniculate nucleus or in the optic tract adjacent to the nucleus, classified physiologically, and injected intracellularly with horseradish peroxidase. The arbors of recovered axons were compared with X and Y axon arbors from normally reared adult cats. Our data demonstrate that while X axon arbors are relatively normal, the arbors of Y axons are profoundly affected by rearing with strabismus. Y axons, whether originating from the deviated or the nondeviated eye, have substantially smaller arbors and fewer boutons in the A-laminae of the lateral geniculate nucleus compared to Y axons in normal cats. The C-lamina terminations of contralaterally projecting Y axons in the strabismic cats are unaffected. These results suggest that the postnatal development of retinogeniculate Y axon arbors in the A-laminae is strongly influenced by abnormalities in postnatal visual experience. Furthermore, the present data suggest that, in addition to intraocular competitive interactions between X and Y axons previously proposed to account for the effects of other rearing conditions, interactions between afferents from the two eyes must also be involved in the development of at least Y axons.
GLYCINE MODULATES THE ERG Bwaves AND ONTYPE BIPOLAR CELLS IN CARP RETINA

Abstract:In the present work we investigated the effects of glycine on the electroretinograms (ERG) and light responses of ON-type bipolar cells under dark- and light-adapted conditions in superfused, isolated crucian carp retinas. It was revealed that application of 4 mmol/L glycine significantly suppressed the b-waves and the responses of ON-type bipolar cells and the effects were blocked by co-application of 20 mumol/L strychnine. In addition, glycine had no apparent action on the PIII component (photoreceptor potential) isolated by application of 3 mmol/L glutamate. These results suggest that glycine may act on bipolar cells directly and thus modulate the activity of the ON pathway in the retina.
Responses to sinusoidal gratings of two types of very nonlinear retinal ganglion cells of cat

Abstract:Perhaps 35% of all of the ganglion cells of the cat do not have classical center-surround organized receptive fields. This paper describes, quantitatively, the responses of two such cell types to stimulation with sinusoidal luminance gratings, whose spatial frequency, mean luminance, contrast, and temporal frequency were varied independently. The patterns were well-focused on the retina of the anesthetized and paralyzed cat. In one type of cell, the maintained discharge was depressed or completely suppressed when a contrast pattern was imaged onto the receptive field (suppressed-by-contrast cell). In the other type of cell, the introduction of a pattern elicited a burst of spikes (impressed-by-contrast cell). When stimulated with drifting gratings, the cell's mean rate of discharge was reduced (suppressed-by-contrast cell) or elevated (impressed-by-contrast cell) over a limited band of spatial frequencies. There was no significant modulated component of response. The reduction in mean rate of suppressed-by-contrast cells caused by drifting gratings had a monotonic dependence on contrast, a relatively low-pass temporal-frequency characteristic and was greater under photopic than mesopic illuminance. If grating of spatial frequency, that when drifted evoked a response from these cells, were instead held stationary and contrast-reversed, the mean rate of a suppressed-by-contrast cell was also reduced and that of an impressed-by-contrast cell increased. But, for contrast-reversed gratings, the discharge contained substantial modulation at even harmonic frequencies, the largest being the second harmonic. The amplitude of this second harmonic did not depend on the spatial phase of the grating, and its dependence on spatial frequency, at least for suppressed-by-contrast cells, was similar to that of the reduction in mean rate of discharge. Our results suggest that the receptive fields of suppressed-by-contrast and impressed-by-contrast cells can be modeled with the general form of the nonlinear subunit components of Hochstein and Shapley's (1976) Y cell model.
Structure and Function of Retinal Ganglion Cells Innervating the Cat's Geniculate Wing: an in Vitro Study

Abstract:We have examined in vitro the morphology and visual response properties of retinal ganglion cells innervating a component of the cat's lateral geniculate nucleus known as the geniculate wing (or retinorecipient zone of the pulvinar). Ganglion cells were first labeled in situ by retrograde transport of fluorescent microspheres from the geniculate wing. Labeled cells were injected intracellular with Lucifer yellow and biocytin in the isolated retina and visualized immunohistochemically. With one exception, stained cells appeared to belong to a single morphological class that corresponded closely to the epsilon cell of earlier descriptions (Leventhal et al., 1980; Rodieck and Watanabe, 1986). They had somas comparable in size to those of beta cells and large, sparse dendritic trees that ramified in the inner (ON) sublayer of the inner plexiform layer. Dendritic fields increased in size with eccentricity, but only within the central retina, and were among the largest so far reported for cat ganglion cells, exceeding those of alpha cells at most eccentricities. Dendritic profiles were typically elliptical with long axes pointing toward the area centralis. Axons were about as thick as those of beta cells and thicker than those of other varieties of non-alpha, non-beta ganglion cells. We recorded extracellularly from microsphere-labeled wing-projecting ganglion cells in a superfused, flattened eyecup preparation. All such cells exhibited sustained responses to standing contrast and had very large, concentric receptive fields with ON-centers and OFF-surrounds. Their response to gratings showed that they have relatively poor spatial resolution and a moderate amount of nonlinearity of spatial summation. These cells thus have many physiological response properties in common with ganglion cells previously termed “on-center tonic W-cells,” “on-center sluggish sustained cells,” and “Q-cells.” These findings indicate that ganglion cells innervating the cat's geniculate wing form a structurally and functionally homogeneous class. Their large dendritic and receptive fields and low-pass spatial frequency tuning suggest that fine spatial resolution is not required for the execution of their functional role(s).
D1 and D2 receptor-mediated dopaminergic modulation of visual responses in cat dorsal lateral geniculate nucleus

Abstract:The modulatory effects of dopamine (DA) on the visual responses of relay cells of the dorsal aspect of cat lateral geniculate nucleus (dLGN) were tested using local micro-iontophoretic application of DA and application of the receptor-specific agonists SKF38393 (SKF, D1/D5) and quinpirole (QUIN, D2/D3/D4) in the anaesthetized alcuronium-treated cat. The effects of DA and QUIN were clearly dose-dependent: small amounts caused a weak and transient facilitation of visual activity (10-30% increase) preferentially in Y-type relay cells, which changed to a moderate reduction of visual responses when the dose was increased (50%, maximal 70%). The effect of SKF was mainly suppressive and increased with the amount of drug applied (up to 90% reduction). The selective antagonists SCH23390 (SCH, D1) and sulpiride (SULP, D2) reduced the effects of co-applied DA agonists. We found little evidence for a specific dopaminergic modulation of the surround inhibition (stimulus-driven lateral inhibition) although DA slightly facilitated the transmission of weak signals (small stimuli). Nevertheless, some dopaminergic effects seem to be mediated via inhibitory interneurons regulating the strength of sustained or recurrent inhibition. Application of DA agonists during blockade of GABA(A) receptors indicates a direct suppression of relay cells via D1 receptors, an excitation of relay cells via D2 receptors and--with increasing amounts of D2 agonist--probably also an excitation of inhibitory interneurons, which results in an indirect inhibition of dLGN relay cells (predominantly of the X-type). The results are discussed in relation to the impairment of visual functions in Parkinson's disease.

Abstract:Rheumatoid arthritis (RA), the most common autoimmune disease, is characterized by persistent synovitis and systemic inflammation. Genetic predisposition as well as autoantibodies and environmental factors, such as smoking, are associated with an increased risk of RA. Traditionally RA has been treated with disease modifying anti-rheumatic drugs (DMARDs) but in the last 15 years or so the introduction of biological response modifiers has revolutionized the treatment of RA. Among these anti-tumor necrosis factor (TNF) agents were the first to be successfully used in treating RA. The goal in treating RA is to induce remission or very low disease activity; remission is now accepted as the ultimate therapeutic goal by adoption of a "treat to target" strategy to achieve tight disease control. Therefore early diagnosis, as well as immediate intervention, are of the utmost importance. This review of the role of TNF in RA pathogenesis describes the mechanisms of action of currently used anti-TNF agents and the adverse events and safety of these drugs. Guidance on the use of anti-TNFs during pregnancy and prior to surgical procedures is also discussed. The intense efforts currently being made to identify biomarkers of response to anti-TNF therapy and recent progress in defining genetic predictors of response using genome- wide association studies (GWAS) are covered. However, so far, none of these studies have been translated into clinical application. The development of biosimilars or follow-on biologicals is also discussed and the first reported study of a biosimilar, involving a multicenter study of an etanercept biosimilar, Etanar, is described.
Evidence for glycine modulation of excitatory synaptic inputs to retinal ganglion cells

Abstract:The actions of glycine on the NMDA receptor-mediated synaptic responses of ganglion cells were studied in the tiger salamander retinal slice. Ganglion cell excitatory postsynaptic currents (EPSCs) were elicited either by exciting bipolar cells with potassium puffs or by light stimulation, and were measured using whole-cell patch-clamp techniques. Increasing bath glycine concentrations to 10 microM had little effect on the amplitude of the puff-evoked EPSCs, indicating either that synaptic glycine concentrations were saturating or that the added glycine was buffered by uptake mechanisms. However, 5,7-dichlorokynurenic acid (5,7-DCK), an antagonist for the glycine site on the NMDA receptor, reduced the ganglion cell responses to NMDA puffs, and reduced the potassium puff- and light-elicited EPSCs. The IC50 values for 5,7-DCK became larger with increasing glycine concentrations, but not with increasing NMDA concentrations, indicating that 5,7-DCK acted at the glycine site. The IC50 values for 5,7-DCK were increased with stronger potassium puffs or light stimuli, suggesting that synaptic glycine levels increased with the strength of the stimuli. EPSCs measured in ON-OFF ganglion cells at light ON and OFF were reduced by 5,7-DCK. For dim light stimuli, the IC50 values were lower for the OFF EPSCs compared to the ON EPSCs, indicating that glycine concentrations were different at the times of OFF and ON activity. Estimates of synaptic glycine concentrations suggest that for dim light stimuli, concentrations at the OFF synapses were not saturated, but concentrations at the ON synapses were saturated.
[The Pharmacology of the GABA-induced Biphasic Response in Guinea-Pig Celiac Ganglion Neurons in Vitro].

Abstract:In guinea-pig celiac ganglion neurons, GABA induced a biphasic response, i.e. a depolarizing response associated with a resistance decrease followed by a hyperpolarizing response associated with a resistance increase. Such response persisted in low Ca2+ high Mg2+ medium. The GABA-induced biphasic response was mimicked by muscimol and inhibited by bicuculline methiodide (100 mumol/L) and picrotoxin (100-300 mumol/L). At doses producing almost equal depolarizing responses, muscimol was weaker in producing hyperpolarizing responses than GABA. Baclofen had no effect on membrane potentials of the ganglion cells. These results suggest that the depolarizing phase of the GABA-induced biphasic response is mediated by GABAA/Cl- ionophore receptor complex and the hyperpolarizing phase may be mediated by a novel type of GABA receptor possessing similar pharmacological properties as the classical GABAA receptor.
The Effect of A2AReceptor Antagonist on Microglial Activation in Experimental Glaucoma

Abstract:PURPOSE:We investigated the effect of A2A receptor (A2AR) antagonist on microglial activation and retinal ganglion cell (RGC) survival under chronic ocular hypertension (COH), and explored the relationship between microglial activation and RGC survival by means of in vitro and in vivo experiments.METHODS:An animal model of COH was induced in one eye of male Sprague-Dawley (SD) rats by ligation of three episcleral veins. The survival of RGCs and the activation of microglia under COH without or with intravitreous injection of A2AR antagonist ZM241385 were assessed by fluorescent labeling, real time PCR and Western blot. ELISA was used to measure the secretion of inflammatory mediators by microglia when glutamate and/or ZM241385 was added into the culture system.RESULTS:Compared to the baseline, RGC density 2 weeks after COH induction decreased at the central (2436 ± 143 cells/mm2 pre- and 2130 ± 148 cells/mm2 post-COH induction) and peripheral (2219 ± 140 cells/mm2 pre- and 1953 ± 142 cells/mm2 post-COH induction) retina. The microglia changed their ramified morphology to an amoeboid form with increase in TNF-α and IL-1β expression after COH. These changes, however, were ameliorated with intravitreous ZM241385 (RGC density only dropped to 2287 ± 135 cells/mm2). The upregulation of those proinflammatory cytokines secreted by microglia in vitro under high concentration of glutamate was downregulated when ZM241385 was added into the culture system.CONCLUSIONS:A2AR antagonist ZM241385 could reduce the activation of microglia and downregulate the proinflammatory cytokines expression under the conditions of COH and high concentration of glutamate, which may be one of the mechanisms that protected RGCs in experimental glaucoma.
Effects of Cortical Stimulation on Auditory‐responsive Thalamic Neurones in Anaesthetized Guinea Pigs

Abstract:In the present study, we investigated neuronal responses to acoustic stimuli and cortical stimulation in the medial geniculate body (MGB) through in vivo intracellular recordings in anaesthetized guinea pigs. Of the 54 neurones examined with acoustic stimuli, 36 showed excitatory postsynaptic potential (EPSP) responses and 19 showed inhibitory postsynaptic potential (IPSP) responses to acoustic stimuli. Of the 36 EPSP neurones examined with corticofugal modulation, 29 received corticofugal depolarization, 3 corticofugal inhibition, and 4 showed no effect. Of the 19 IPSP neurones, 17 received corticofugal inhibition and 2 were not affected. The mean amplitude of the EPSPs evoked by acoustic stimuli was similar to that evoked by the electrical cortical stimulation (9.19 ± 5.55 mV versus 9.22 ± 5.16 mV). There was a significant correlation between the parameters of the EPSPs evoked by an acoustic stimulus and those evoked by cortical stimulation. The mean amplitude of the IPSP evoked by electrical cortical stimulation was significantly greater than that evoked by acoustic stimuli (11.6 ± 3.8 mV versus 9.1 ± 3.7 ms, P < 0.05). Seven auditory EPSP and 7 IPSP neurones were examined with corticofugal modulation and labelled with Neurobiotin. Of the 7 EPSP neurones, 5 showed excitatory responses to cortical stimulation and 2 demonstrated no effects. Four of the 5 neurones that received corticofugal depolarization were located in the lemniscal MGB and 1 in the non‐lemniscal MGB; of the remaining 2, 1 was located in the lemniscal and the other in the non‐lemniscal MGB. Of the 7 IPSP neurones, 1 received an excitatory corticofugal input followed by an inhibitory input and 4 received only an inhibitory corticofugal input, while the remainder demonstrated no corticofugal effects. All 7 neurones were located in the non‐lemniscal MGB. The result that both ascending and descending inputs caused similarly shaped EPSPs reflects a neuronal endogenous characteristic irrespective of the physical locations of the synapses. The IPSP responses to both acoustic stimuli and electrical cortical stimulation are likely to be caused by feedback from the thalamic reticular nucleus.
Dramatically Reducing Training Data Size Through Vocabulary Saturation

Abstract:Our field has seen significant improvements in the quality of machine translation systems over the past several years. The single biggest factor in this improvement has been the accumulation of ever larger stores of data. However, we now find ourselves the victims of our own success, in that it has become increasingly difficult to train on such large sets of data, due to limitations in memory, processing power, and ultimately, speed (i.e., data to models takes an inordinate amount of time). Some teams have dealt with this by focusing on data cleaning to arrive at smaller data sets (Denkowski et al., 2012a; Rarrick et al., 2011), “domain adaptation” to arrive at data more suited to the task at hand (Moore and Lewis, 2010; Axelrod et al., 2011), or by specifically focusing on data reduction by keeping only as much data as is needed for building models e.g., (Eck et al., 2005). This paper focuses on techniques related to the latter efforts. We have developed a very simple n-gram counting method that reduces the size of data sets dramatically, as much as 90%, and is applicable independent of specific dev and test data. At the same time it reduces model sizes, improves training times, and, because it attempts to preserve contexts for all n-grams in a corpus, the cost in quality is minimal (as measured by BLEU ). Further, unlike other methods created specifically for data reduction that have similar effects on the data, our method scales to very large data, up to tens to hundreds of millions of parallel sentences.
Visual Response Properties of Y Cells in the Detached Feline Retina

Abstract:PURPOSE:To evaluate early changes in the visual response properties of Y cells in the detached feline retina.METHODS:The retinas of young adult cats were detached by injection, with a glass micropipette, of a solution of 0.004% sodium hyaluronate in a balanced salt solution between the neural retina and the retinal pigment epithelium. At 1, 3, and 7 days after detachment, the eyes were removed. The eyecup was prepared as a flat mount in a recording chamber and superfused with medium. Extracellular single-unit responses from Y cells in the retinas were recorded.RESULTS:One, 3, and 7 days after retinal detachment surgery, Y cells showed clear signs of functional deterioration. At each time point, more ON center cells than OFF cells were encountered. Y cells in the detached retinas showed a statistically significant elevation in the average threshold irradiance after 1-, 3-, and 7-day detachment, respectively. The average contrast threshold recorded from cells in the normal retina was 3.6%, but it increased to 14.5%, 21.8%, and 47.5% after 1-, 3-, and 7-day detachment, respectively. Furthermore, at each time point, the capability of Y cells to process contrast information decreased significantly more because of detachment than because of luminance task performance.CONCLUSIONS:Retinal detachment induced rapid functional remodeling that resulted in degenerated Y-cell function, including an elevated luminance threshold and a deteriorated contrast threshold. Detachment had a greater impact on the latter. These physiological changes after retinal detachment could be used as objective indicators of early deterioration of visual function in future studies of retinal remodeling.

Abstract:Current responses of carp retinal ganglion cells (RGCs) retrogradely labeled and freshly dissociated to rapid application of glycine were recorded by whole-cell patch clamp techniques and effects of glycine antagonists on these responses were analyzed. The current response to maintained application of glycine at a concentration higher than 30 μM exhibited desensitization, which was well fitted to a monoexponential function. Strychnine (1 μM), a glycine receptor antagonist, completely blocked the response to 100 μM glycine. Strychnine at a concentration range between 10 and 200 nM suppressed the response to 100 μM glycine in a dose-dependent manner, and only a slow-activated and sustained current eventually remained in the presence of 200 nM strychnine. Power spectral density (PSD) analysis revealed no changes in the density–frequency dependence caused by strychnine. It was further shown that dissociation of strychnine from glycine receptors was rather slow. Moreover, Zn2+ exerted similar dual action on this sustained response and the response in Ringer's: potentiating and reducing them at low and high concentrations of Zn2+, respectively. 5,7-Dichlorokynurenic acid (DCKA, 500 μM), a selective blocker of the glycine recognition site at the NMDA receptor, partially reduced the glycine response, but without changing its kinetics. These results suggest that glycinergic input to carp ganglion cells may be mediated by strychnine-sensitive glycine receptors with homologous kinetics, and slow dissociation of strychnine from glycine receptors may partially account for the changes in glycine response kinetics occurring in the presence of strychnine.

Abstract:We studied the responses of isolated and intact luminosity-type horizontal cells (L-HC) in the Xenopus retina to L-glutamate (L-glu) and its analogs. Isolated L-HCs studied with whole-cell patch clamp responded to L-glu, kainate (KA), AMPA, or quisqualate (quis) with inward currents from a holding potential of -60 mV, associated with a conductance increase. The current elicited by KA was relatively large and sustained, whereas AMPA or quis evoked a desensitizing current. Coapplication of quis and KA resulted in a smaller current and conductance change than that evoked by a pulse of either alone at the same concentration. This finding suggests that the L-HC has a single subtype of glutamate receptor that responds to both quis and KA. Prior exposure to dopamine enhanced the KA-evoked current about twofold. In the superfused eyecup we found that L-HC responses to quinoxalinediones (CNQX or DNQX) and to L-glu, KA, AMPA, and quis varied as a function of adaptational state. When driven exclusively by either cones or by rods, CNQX/DNQX hyperpolarized the L-HC and reduced its light response, without altering response kinetics, indicating that both rods and cones communicate with L-HCs at ionotropic glutamatergic synapses. Under mesopic conditions, however, as CNQX or DNQX reduced cone input, the rod input to the L-HC increased up to fivefold in magnitude and had slowed kinetics. The depolarizing response of the L-HC to L-glu, AMPA, or quis was relatively small and transient under photopic conditions, but was much larger and sustained when the eyecup was dark adapted. The D1 dopamine antagonist SCH 23390 potentiated the response to quis. In contrast, responses to KA were largest in light-adapted eyecups, were potentiated by a D1 dopamine agonist, SKF 38393, and were reduced by SCH 23390. We hypothesize that the segregated populations of glutamate receptors in the L-HC opposite cone and rod synaptic endings can be separately modulated to respond differentially to the native transmitter, glutamate. In photopic and mesopic states the dominant cone input tonically inhibits rod to L-HC communication. This inhibition appears to occur at the postsynaptic membrane and may be mediated by second messengers.
GABAergic Inhibition Controls Receptive Field Size, Sensitivity, and Contrast Preference of Direction Selective Retinal Ganglion Cells Near the Threshold of Vision

Abstract:Information about motion is encoded by direction-selective retinal ganglion cells (DSGCs). These cells reliably transmit this information across a broad range of light levels, spanning moonlight to sunlight. Previous work indicates that adaptation to low light levels causes heterogeneous changes to the direction tuning of ON–OFF (oo)DSGCs and suggests that superior-preferring ON–OFF DSGCs (s-DSGCs) are biased toward detecting stimuli rather than precisely signaling direction. Using a large-scale multielectrode array, we measured the absolute sensitivity of ooDSGCs and found that s-DSGCs are 10-fold more sensitive to dim flashes of light than other ooDSGCs. We measured their receptive field (RF) sizes and found that s-DSGCs also have larger receptive fields than other ooDSGCs; however, the size difference does not fully explain the sensitivity difference. Using a conditional knock-out of gap junctions and pharmacological manipulations, we demonstrate that GABA-mediated inhibition contributes to the difference in absolute sensitivity and receptive field size at low light levels, while the connexin36-mediated gap junction coupling plays a minor role. We further show that under scotopic conditions, ooDSGCs exhibit only an ON response, but pharmacologically removing GABA-mediated inhibition unmasks an OFF response. These results reveal that GABAergic inhibition controls and differentially modulates the responses of ooDSGCs under scotopic conditions.

Excitatory amino acid receptors on sustained retinal ganglion cells in the kitten during the critical period of development

Abstract:Effects of iontophoretically applied excitatory amino acid analogues, kainate, quisqualate and N-methyl-D-aspartate (NMDA) and their receptor antagonists on the sustained class of retinal ganglion cells were studied in the optically intact eye of pentobarbitone-anaesthetized kittens (7-9 weeks of age). These results were compared with the effects obtained in adult cats. All 3 excitatory amino acid agonists had excitatory actions on the majority of On- and Off-sustained ganglion cells in the kitten but at higher current levels than those required for adult cells, suggesting all 3 types of receptors of weaker sensitivity are present on the kitten cells. Whilst the relative potency of kainate, quisqualate and NMDA was 15:3:1 in the adult cells, it was 5:2:1 in the kitten cells. As for other neurones in the CNS, an increase in the potency of kainate receptors and a decrease in that of NMDA receptors appear, therefore, to characterize the postnatal development of the excitatory amino acid receptors on the retinal ganglion cells. In accordance with the agonist results, a broadband receptor antagonist, kynurenate, powerfully antagonised responses of kitten cells as well as those of adult cells. The pure NMDA receptor antagonist, 3((+-)-2-carboxypiperazin-4-yl)propyl-1-phosphonate (CPP), however, only suppressed spontaneous firing of kitten cells. Furthermore, in kitten cells, the visually-driven firing was depressed while the level of firing was raised by these excitatory amino acid analogous, and a long period of inhibition of firing followed the agonist-induced excitation.(ABSTRACT TRUNCATED AT 250 WORDS)
