Abstract:Regulatory factor X 6 (RFX6) is crucial for pancreatic endocrine development and differentiation. The RFX6 variant p.His293LeufsTer7 is significantly enriched in the Finnish population, with almost 1:250 individuals as a carrier. Importantly, the FinnGen study indicates a high predisposition for heterozygous carriers to develop type 2 and gestational diabetes. However, the precise mechanism of this predisposition remains unknown.

What problem does this paper attempt to address?
The problem this paper attempts to address is: **How does RFX6 gene haploinsufficiency lead to diabetes**. Specifically, the researchers focus on a particular variant of the RFX6 gene (p.His293LeufsTer7), which is relatively common in the Finnish population and is associated with a high risk of type 2 diabetes and gestational diabetes. However, the specific mechanism by which this variant leads to diabetes is not yet clear. Therefore, the researchers generated pluripotent stem cell models with different allele series using CRISPR technology to explore the impact of this variant on the development and function of pancreatic β-cells. ### Main Questions: 1. **How does the RFX6 gene variant affect the development and function of pancreatic β-cells?** 2. **Does RFX6 gene haploinsufficiency lead to a reduction in the number of pancreatic β-cells or impaired insulin secretion?** 3. **How does the RFX6 gene variant increase the risk of type 2 diabetes and gestational diabetes?** Through these studies, the authors hope to reveal the specific role of the RFX6 gene in the development and function of pancreatic endocrine cells, thereby providing new insights into the pathogenesis of diabetes.
