Abstract:An efficient enzymatic method for the synthesis of (S)-3-substituted glutaric acid monoesters which was aided by molecular docking has been described. The reaction was proceeded under mild conditions, and the desired products were afforded with up to 98% ee in the yield of 93%. The results demonstrate that molecular docking is efficient to facilitate selection of substrates in enzymatic reaction.

Abstract:A convenient approach for synthesis of raffinose, stachyose, and verbascose using sucrose as the starting material is presented. The key step is the regioselective de-O-benzylation with Co2(CO)8/Et3SiH/CO system, followed by a high α-selective glycosylation. The newly developed de-O-benzylation system is efficient in removing the primary benzyl groups of sucrose and raffinose under mild condition and with high selectivity. Using thioglycoside as donor, NIS/AgOTf as promoter and DTBMP as additive, glycosylation of acid labile sucrose substrate is achieved in high yield.

Abstract:Sucrose-6-acetate, a short-chain monoester, is an important intermediate for the preparation of sucralose. Regioselective transformations of mono- and oligosaccharides by enzymes have been performed in organic solvents. The present study was carried out with Candida rugosa lipase to synthesize sucrose-6-acetate by transesterification of sucrose and vinyl acetate in a two-phase system consisting of a buffer solution and an organic solvent. The screening results revealed that 2-butanol/Tris-HCl was the optimal two-phase system for biocatalysis, and other factors influencing the synthesis of sucrose-6-acetate were also studied. Three major factors (temperature, initial buffer pH, and molar ratio of substrates) were identified through Plackett-Burman design, and the response surface methodology was employed to optimize the reaction process. Under optimal conditions, the yield of sucrose-6-acetate reached up to 57.42%. The aqueous two-phase system was successfully applied to the synthesis of sucrose-6-acetate with high yield, low toxicity, good resolution, low cost, and simple scale-up.

Abstract:The stannylene based approach to regioselective sugar derivatization has been widely employed and reflects the satisfactory regioselectivity.The structure of dimer and polymer complex of tributyhin ether and dibutylstannylene acetal with sugar is well elucidated:the type of oxygen di-coordinated in an apical position of the dipyramid become more negative and hence more nucleophilic as the result of the electron-enriching effect of the tin atom redistribution,which account for the origin of regioselectivity. It is also discussed in detail with their regioselctive derivatization of tin-mediated complex systems of pyran-carbohydrate polyols of different types.

Abstract:Sucrose-6-acetate is an important intermediate in the preparation of sucralose (a finest sweetener). In our study, Candida rugosa lipase coated with surfactant was firstly immobilized on sol–gel supports. Then, the immobilized enzyme was used in the regioselective synthesis of sucrose-6-acetate by transesterification of sucrose and vinyl acetate. The screening results revealed that Tween 80 was an ideal surfactant to coat lipase immobilized in sol–gel and exhibited the highest yield of sucrose-6-acetate. Other factors that influenced the yield during the preparation process were also studied. Under optimal conditions, the yield of sucrose-6-acetate could reach up to 78.68 %, while free lipase was easily inactivated in polar solvent. Thermal and operational stabilities were also improved significantly. Surfactant-coated lipase immobilized in sol–gel remained stable when the temperature was higher than 60 °C. Moreover, they could maintain high catalytic activity after six recycles. This strategy is economical, convenient and promising for the food industry.

Abstract:Sucrose-6-acetate (S-6-A) is an important intermediate in the industrial production process of sucralose. In this work, the synthesis and reaction kinetics for both the high-temperature and low-temperature processes of sucrose acylation to produce S-6-A via the dibutyltin oxide method were investigated. The influences of the molar ratios of both dibutyltin oxide (DBTO)/sucrose and acetic anhydride/sucrose, reaction time, and temperature on the yield of S-6-A were evaluated. Based on the reaction mechanism of the high-temperature process and the solubility characteristic of DBTO, the first-order pseudohomogeneous kinetic model was proposed, which fitted experimental data well in the temperature range of 360.15 to 370.65 K. In addition, the second-order pseudohomogeneous kinetic model was established for the low-temperature process, and the model fitting was in good agreement with experimental data in the temperature range of 274.15 to 296.15 K. The results indicate that the early stage cooling and reduction of reaction time can significantly enhance the yield of S-6-A in the low-temperature process. Hopefully, the kinetic models developed in this work can provide valuable insights into the process optimization of the acylation reaction, the design of the reactors, and the continuous process.

Abstract:A highly efficient and mild method for the de-O-benzylation of protected saccharides was developed by transforming terminal benzyl ethers into silyl ethers using Co-2(CO)(8)-Et3SiH under 1 atm of CO. The method was successfully used for the de-O-benzylation of perbenzylated monosaccharides with various anomeric protecting groups, as well as natural disaccharides and trisaccharides such as sucrose, raffinose, and melezitose in good yields (>80%).

Abstract:Dibutyltin oxide reacts selectively with hydroxyl groups of carbohydrates to form dibutylstannylene acetal,and polymerization of the acetal in solution enlarges the electronegative and nucleophilic difference of hydroxyl groups situated on the different locations of polymer,which accounts for selective synthesis of sugar derivatives.Hence the chemical characteristic of dibutyltin oxide is well elucidated via its application on sides of protection of adjacent hydroxyl groups of sugars containing active functional groups,regioselective bridging of carbohydrates and glycosylation,which lays foundation for this reagent to extend its application into selective synthesis of sugar derivatives.

Abstract:This paper reviews the advances and up-to-date methods of synthesis and procedure of sucrose esters in addition to introduction to its reaction mechanism.Divided by its reaction ways,here are four ways to synthesize sucrose ester,that is,chloride esterification method,direct esterification method,transesterification method and enzyme catalyst method.Chloride esterification method:in the presence of a catalyst,sucrose ester is synthesized by the reaction of su- crose to fatty acid chloride.The need to use toxic solvents,as is known in this area,makes it difficult for the synthesis to come to the national standards for food products,cosmetics and pharmaceutical goods made in chloride esterification method.The direct esterification method as is mentioned above,in actual application,calls for an acid catalyst (e.g.tosylate).When sucrose ester is synthesized with fatty acid ester,it is necessary to use polar aprotic organic solvents (e.g. DMF) in the process,which seems simple and easy to operate for its synthesis but DMF proves toxic,for it is not easy to purify sucrose esters. The reason is that ester is decomposed easily in the acidic conditions, which result in low yield.Transesterification includes solvent method and solvent-free method.The former uses DMF,DMSO, propylene glycol or water as solvent,but both DMF and DMSO are somewhat toxic,which restricts the applications of sucrose esters in food industry.If propylene glycol or water is used as solvent,the emulsifying agent to be added to the process also makes it difficult to purify the sucrose esters.If non-solvent method is used,emulsifying agents or surfactants to be added to the reaction system tends to make the melting reactants homogenous.If the temperature of the reaction is used,the yield would also be made low,besides,the quality of the product can not be guaranteed.Seeing the shortcomings of all the above methods,a new biological synthesis method has been brought up forward,which is known as enzyme catalyst method.The enzyme catalyst,which is used for synthesis instead of the traditional catalysis, enjoys high selectivity and easy separation in the production process under moderated conditions.In spite of this,the paper has also made brief introductions to other auxiliary methods.

The results show the O-2 position of glucoside has advantage of being benzylated over the O-6 position. The major isomers of benzyl ether were separated by preparative HPLC method and characterized.

Abstract:This work studied the synthesis of sucroesters via transesterification of sucrose and methyl palmitate in a solvent-free process. The kinetic experiments were carried out under slight vacuum (80 kPa) to ensure an irreversible reaction, with K2CO3 as catalyst, and using a variety of surfactants (potassium palmitate, sucrose palmitate and glyceryl monostearate) with different Hydrophilic-Lipophilic Balance (HLB) as contacting agents. Reaction samples were analyzed with a novel isocratic HPLC method using a refraction index detector. The kinetic effects of temperature (369–409 K) and surfactant type and concentration (5–15%wt) were assessed. Results revealed that the reaction exhibited a lag period, consistent with a limiting sucrose activation step, followed by faster transesterification to monoester. Also, the different substitution steps on the sucrose backbone showed strong temperature dependence, and the corresponding large energies of activation. According to the product distribution, the reaction proceeded mostly to mono, di and tri substituted sucroesters. Particularly, potassium palmitate exhibited synergic contact-enhancing and catalytic effects, delivering a sucrose ester productivity of 55 g SE/h kg and a selectivity to monoesters in the range of 52–67%. It was observed that high HLB surfactants promoted a higher selectivity for monosubstituted sucroesters. The reaction kinetics was modeled as a set of steps involving sucrose activation, monoester formation, and lumped oligoester synthesis. The kinetic parameters were regressed for each surfactant, and the model showed good agreement with experiments. Obtained models can be used for further process analysis and design.

Abstract:The regio- and stereoselective, Lewis acid catalyzed Strecker reaction between Me3SiCN and different aldimines incorporating a 2,3,4,6-tetrakis-O-pivaloyl-D-glucopyranosyl (Piv(4)Glc) chiral auxiliary has been worked out. Depending on the conditions used, high yields (up to 95%) and good diastereoselectivities (de > 86%) were achieved under mild conditions (Table 1), especially with CuBr (.) Me2S as catalyst. Our protocol allows the ready preparation of asymmetric beta, gamma-unsaturated alpha-amino acids such as (R)-2-amino-4-phenylbut-3-enoic acid (13; Scheme 2) and congeners thereof.

Abstract:The transesterification of α-d-glucose with divinylsuccinate, divinyladipate and divinylsebacate in pyridine at 55 °C for 3 days was catalyzed by an alkaline protease from Bacillus subtilis to give corresponding 6- O -vinyl glucose esters at 30%, 53% and 35% yield, respectively. The stereo-selectivity of the alkaline protease toward the α-anomer was affected by the acyl donor chain length. 6- O -Vinylsuccinyl-d-glucose was mixture of α- and β-anomers (α/β=44/56), the other two products were the pure α-d-glucose derivatives.

Abstract:Phenylpropanoid sucrose esters are a large and important group of natural substances with significant therapeutic potential. This work describes a pilot study of the enzymatic hydroxycinnamoylation of sucrose and its derivatives which was carried out with the aim of obtaining precursors of natural phenylpropanoid sucrose esters, e.g., vanicoside B. In addition to sucrose, some chemically prepared sucrose acetonides and substituted 3′-O-cinnamates were subjected to enzymatic transesterification with vinyl esters of coumaric, ferulic and 3,4,5-trimethoxycinnamic acid. Commercial enzyme preparations of Lipozyme TL IM lipase and Pentopan 500 BG exhibiting feruloyl esterase activity were tested as biocatalysts in these reactions. The substrate specificity of the used biocatalysts for the donor and acceptor as well as the regioselectivity of the reactions were evaluated and discussed. Surprisingly, Lipozyme TL IM catalyzed the cinnamoylation of sucrose derivatives more to the 1′-OH and 4′-OH positions than to the 6′-OH when the 3′-OH was free and the 6-OH was blocked by isopropylidene. In this case, Pentopan reacted comparably to 1′-OH and 6′-OH positions. If sucrose 3′-O-coumarate was used as an acceptor, in the case of feruloylation with Lipozyme in CH3CN, 6-O-ferulate was the main product (63%). Pentopan feruloylated sucrose 3′-O-coumarate comparably well at the 6-OH and 6′-OH positions (77%). When a proton-donor solvent was used, migration of the 3′-O-cinnamoyl group from fructose to the 2-OH position of glucose was observed. The enzyme hydroxycinnamoylations studied can shorten the targeted syntheses of various phenylpropanoid sucrose esters.

Abstract:Sugar esters have recently received increased attention from the viewpoint of renewable resources. Specifically vinyl esters from carbohydrates by enzyme-catalyzed regioselective transformations offer a new family of functional water-soluble monomers for preparation of sugar-based polymers. Here we reported enzyme-catalyzed regloselective synthesis of novel polymerizable raffinose vinyl ester. Transesterification of raffinose with divinyl succinate, divinyl adipate and divinyl sebacate in anhydrous pyridine at 50 degreesC for 6 d was catalyzed by an alkaline protease from Bacillus subtilis. Three raffinose vinyl esters with different acyl donor chain lengths were regioselectively synthesized at 55%, 48% and 30% yields, respectively. Characterization of the products by H-1 NMR, C-13 NMR, and H-1-H-1 correlation spectroscopy and H-1-C-13 heteronuclear multiple quantum coherence NMR revealed that the ester was substituted at C-1 position of beta-fructofuranose moiety in raffinose. Reaction conditions for the esterification were investigated. Activities of. seven commercial enzymes from different sources catalyzing synthesis of raffinose ester Were compared. Lipase from porcine pancreas(PPL) and alkaline protease from Bacillus subtilis had high activities. We also studied the effect of temperature and incubation time on raffinose esters synthesis catalyzed by alkaline protease from Bacillus subtilis and found the optimal temperature and reaction time were 50 degreesC and 6 d respectively.

Abstract:The transesterification reaction of sucrose and fatty acid methyl esters to produce sucroesters was experimentally evaluated using commercial emulsifiers as compatibility agents. Reactions were carried out at temperatures between 100 and 140°C, using emulsifier concentrations in the range of 5 to 15 %wt, and potassium carbonate as catalyst. Fatty acid methyl esters consumption and sucroesters production was monitored by HPLC analysis of samples. Methyl esters conversions around 40 % were obtained with 68 %wt monoester content in sucroesters mixture. Despite the reaction times were reduced by operating at high temperatures and high emulsifier’s concentration, multiple substitution and color degradation were observed. Higher productivities of sucroester and higher selectivity to monoesters were obtained when potassium palmitate was used as contacting agent. The lower monoester content in the final product was obtained when using a commercial sucroester emulsifier. Results of this study can be used for preliminary process design in a solvent-free production of biobased sucroesters.
