Abstract:Maize mutants of the centromeric histone H3 ( CENP-A / CENH3 ) gene can form haploids that inherit only chromosomes of the pollinating parent but the cytoplasm from the female parent. We developed CENH3 haploid inducers carrying a dominant anthocyanin colour marker for efficient haploid identification and harbouring cytoplasmic male sterile cytoplasm, a type of cytoplasm that results in male sterility useful for efficient hybrid seed production. The resulting cytoplasmic male sterility cyto-swapping method provides a faster and cheaper way to convert commercial lines to cytoplasmic male sterile compared to conventional trait introgression.

Abstract:Rapeseed is a globally significant oilseed crop cultivated to meet the increasing demand for vegetable oil. In order to enhance yield and sustainability, breeders have adopted the development of rapeseed hybrids as a common strategy. However, current hybrid production systems in rapeseed have various limitations, necessitating the development of a simpler and more efficient approach. In this study, we propose a novel method involving the targeted disruption of Defective in Anther Dehiscence1 of Brassica napus (BnDAD1), an essential gene in the jasmonic acid biosynthesis pathway, using CRISPR/Cas9 technology, to create male-sterile lines. BnDAD1 was found to be dominantly expressed in the stamen of rapeseed flower buds. Disrupting BnDAD1 led to decreased levels of α-linolenic acid and jasmonate in the double mutants, resulting in defects in anther dehiscence and pollen maturation. By crossing the double mutant male-sterile lines with male-fertile lines, a two-line system was demonstrated, enabling the production of F 1 seeds. The male-sterile trait of the bndad1 double mutant lines was maintainable by applying exogenous methyl jasmonate and subsequently self-pollinating the flowers. This breakthrough holds promising potential for harnessing heterosis in rapeseed and offers a simpler and more efficient method for producing hybrid seeds.

Abstract:Capturing heterosis (hybrid vigor) is a promising way to increase productivity in many crops; hybrid crops often have superior yields, disease resistance, and stress tolerance compared with their parental inbred lines. The full utilization of heterosis faces a number of technical problems related to the specifics of crop reproductive biology, such as difficulties with generating and maintaining male-sterile lines and the low efficiency of natural cross-pollination for some genetic combinations. Innovative technologies, such as development of artificial in vitro systems for hybrid production and apomixis-based systems for maintenance of the resulting heterotic progeny, may substantially facilitate the production of hybrids. Genome editing using specifically targeted nucleases, such as clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated nuclease 9 (CRISPR/Cas9) systems, which recognize targets by RNA:DNA complementarity, has recently become an integral part of research and development in life science. In this review, we summarize the progress of genome editing technologies for facilitating the generation of mutant male sterile lines, applications of haploids for hybrid production, and the use of apomixis for the clonal propagation of elite hybrid lines.

Abstract:Heterosis boosts crop yield; however, harnessing additional progressive heterosis in polyploids is challenging for breeders. We bioengineered a 'mitosis instead of meiosis' ( MiMe ) system that generates unreduced, clonal gametes in three hybrid tomato genotypes and used it to establish polyploid genome design. Through the hybridization of MiMe hybrids, we generated '4-haplotype' plants that encompassed the complete genetics of their four inbred grandparents, providing a blueprint for exploiting polyploidy in crops.

Abstract:In plants, in vivo haploid induction has gained increasing attention for its significant potential applications in crop breeding and genetic research. This strategy reduces the chromosome number in progeny after fertilization, enabling the rapid production of homozygous plants through double haploidization, contrasting with traditional inbreeding over successive generations. Haploidy typically initiates at the onset of seed development, with several key genes identified as paternal or maternal factors that play critical roles during meiosis, fertilization, gamete communication, and chromosome integrity maintenance. The insights gained have led to the development of efficient haploid inducer lines. However, the molecular and genetic mechanisms underlying these factors vary considerably, making it challenging to create broadly applicable haploidy induction systems for plants. In this minireview, we summarize recent discoveries and advances in paternal and maternal haploid induction factors, examining their current understanding and functionalities to further develop efficient haploid inducer systems through the application of parental factor manipulation.

Abstract:When homozygously fertile plants were induced using doubled haploid (DH) induction lines Y3380 and Y3560, the morphology of the induced F 1 generation was basically consistent with the female parent, but the fertility was separated, showing characteristics similar to cytoplasmic male sterile (CMS) and maintainer lines. In this study, the morphology, fertility, ploidy, and cytoplasm genotype of the induced progeny were identified, and the results showed that the sterile progeny was polima cytoplasm sterile ( pol CMS) and the fertile progeny was nap cytoplasm. The molecular marker and test-cross experimental results showed that the fertile progeny did not carry the restorer gene of pol CMS and the genetic distance between the female parent and the offspring was 0.002. This suggested that those inductions which produced sterile and fertile progeny were coordinated to CMS and maintainer lines. Through the co-linearity analysis of the mitochondrial DNA (mtDNA), it was found that the rearrangement of mtDNA by DH induction was the key factor that caused the transformation of fertility ( nap ) into sterility ( pol ). Also, when heterozygous females were induced with DH induction lines, the induction F 2 generation also showed the segregation of fertile and sterile lines, and the genetic distance between sterile and fertile lines was approximately 0.075. Therefore, the induction line can induce different types of female parents, and the breeding of the sterile line and the maintainer line can be achieved through the rapid synchronization of sister crosses and self-crosses. The induction of DH inducer in B. napus can provide a new model for the innovation of germplasm resources and open up a new way for its application.

Abstract:The doubled haploid technique aims to generate pure inbred lines for basic research and as commercial cultivars. The doubled haploid technique first generates haploid plants and is followed by chromosome doubling, which can be separated in time or overlapped, depending the procedure for each species. For a long time, much effort has been focused on haploid production via androgenesis, gynogenesis, or parthenogenesis. The obtention of haploid plants has frequently required more optimization and has lagged behind research and improvements in chromosome doubling methods. Nevertheless, chromosome doubling has recently been of renewed interest to increase the rates and efficiency of doubled haploid plant production through trialing and optimizing of different procedures. New antimitotic compounds and application methods are being studied to ensure the success of chromosome doubling once haploid material has been regenerated. Moreover, a haploid inducer-mediated CRISPR/Cas9 genome-editing system is a breakthrough method in the production of haploid plant material and could be of great importance for species where traditional haploid regeneration methods have not been successful, or for recalcitrant species. In all cases, the new deployment of this system will demand a suitable chromosome doubling protocol. In this review, we explore the existing doubled haploid and chromosome doubling methods to identify opportunities to enhance the breeding process in major crops.

Abstract:Heterosis utilization in a large proportion of crops depends on the use of cytoplasmic male sterility (CMS) tools, requiring the development of homozygous fertile lines and CMS lines 1 . Although doubled haploid (DH) technology has been developed for several crops to rapidly generate fertile lines 2,3 , CMS lines are generally created by multiple rounds of backcrossing, which is time consuming and expensive 4 . Here we describe a method for generating both homozygous fertile and CMS lines through in vivo paternal haploid induction (HI). We generated in-frame deletion and restored frameshift mutants of BoCENH3 in Brassica oleracea using the CRISPR/Cas9 system. The mutants induced paternal haploids by outcrossing. We subsequently generated HI lines with CMS cytoplasm, which enabled the generation of homozygous CMS lines in one step. The BoCENH3 -based HI system provides a new DH technology to accelerate breeding in Brassica and other crops.

Abstract:Maize (Zea mays L.) is a plant that is grown for human and animal consumption, as well as biofuel and a range of industrial applications throughout the world. By 2020, it is predicted to outnumber wheat and rice in terms of demand. Farmers pick among two types of maize varieties in modern agriculture. Hybrids and open-pollinated varieties are mostly influenced by the current environmental and economic conditions and availability of seed for the preferred variety type. As a result, minimizing the time required for inbred development is critical in supplying varieties that fulfill the needs of modern farmers. The use of doubled haploid technology is the most effective way to accomplish this. The in vivo technique is widely regarded as a valuable tool for improving breeding efficiency. In maize, in vivo haploid induction has been quite successful and several commercial breeding programs currently use it extensively. Many factors such as anther stage, donor plant, and anther preparation were discovered to influence in vitro haploid induction (Androgenesis) in maize. Color indicators are widely used to distinguish typical fertilization kernels from those produced via haploid induction. Because gamete production is hindered in haploid plants, they are sterile. As a result, haploid chromosome duplication is required to permit self-pollination for seed production and genotype preservation. In the production of maize DH lines and colchicine is the most often used chemical as a chromosomal doubling agent. DHs are the ideal material for genetic improvement and genetic studies of maize because of their utility in homozygous line creation, recurrent selection, and induction of mutation, genetic enrichment, and compatibility with other technologies.

Abstract:Doubled haploid (DH) line production through in vivo maternal haploid induction is widely adopted in maize breeding programs. The established protocol for DH production includes four steps namely in vivo maternal haploid induction, haploid identification, genome doubling of haploid, and self-fertilization of doubled haploids. Since modern haploid inducers still produce relatively small portion of haploids among undesirable hybrid kernels, haploid identification is typically laborious, costly, and time-consuming, making this step the second foremost in the DH technique. This manuscript reviews numerous methods for haploid identification from different approaches including the innate differences in haploids and diploids, biomarkers integrated in haploid inducers, and automated seed sorting. The phenotypic differentiation, genetic basis, advantages, and limitations of each biomarker system are highlighted. Several approaches of automated seed sorting from different research groups are also discussed regarding the platform or instrument used, sorting time, accuracy, advantages, limitations, and challenges before they go through commercialization. The past haploid selection was focusing on finding the distinguishable marker systems with the key to effectiveness. The current haploid selection is adopting multiple reliable biomarker systems with the key to efficiency while seeking the possibility for automation. Fully automated high-throughput haploid sorting would be promising in near future with the key to robustness with retaining the feasible level of accuracy. The system that can meet between three major constraints (time, workforce, and budget) and the sorting scale would be the best option.

Abstract:Owing to their superior agronomic performance, the hybrids of vegetable crops are currently applied extensively. However, effective hybrid production requires a laborious manual emasculation to ensure the purity of hybrid seeds in tomato because of the lack of an effective male sterility system. Here, we created two types of tomato nuclear male-sterile lines with different screening markers in a clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) system. Co-knockouts of male sterile 1035 (Ms1035) and glutathione S-transferase (GSTAA) created a male-sterile line marked by a green hypocotyl. The Ms1035 biallelic mutation was introduced into the woolly tomato background, resulting in the linkage of male sterility and a non-woolly phenotype. Two types of male-sterile lines were easily selected at the seedling stage by hypocotyl color or trichome density and further showed high seed purity during hybrid seed production. Our work established the procedure for a rapid transfer of the male-sterile phenotype to the parents of hybrids without extra-modification by the CRISPR/Cas9 system that can be practically applied to hybrid seed production in tomato. This method will be the basis and example for sterile parent creation of multiple crops for hybrid production with the CRISPR/Cas9 system.

Abstract:The Brassica oleracea L. species embrace important horticultural crops, such as broccoli, cauliflower, and cabbage, which are highly valued for their beneficial nutritional effects. However, the complexity of flower emasculation in these species has forced breeders to adopt biotechnological approaches such as somatic hybridization to ease hybrid seed production. Protoplasts entail a versatile tool in plant biotechnology, supporting breeding strategies that involve genome editing and hybridization. This review discusses the use of somatic hybridization in B. oleracea L. as a biotechnological method for developing fusion products with desirable agronomic traits, particularly cytoplasmic male sterile (CMS) condition. These CMS lines are critical for implementing a cost-effective, efficient, and reliable system for producing F1 hybrids. We present recent studies on CMS systems in B. oleracea L. crops, providing an overview of established models that explain the mechanisms of CMS and fertility restoration. Additionally, we emphasize key insights gained from protoplast fusion applied to B. oleracea L. breeding. Key steps including pre-treatments of donor plants, the main tissues used as sources of parental protoplasts, methods for obtaining somatic hybrids and cybrids, and the importance of establishing a reliable plant regeneration method are discussed. Finally, the review explores the incorporation of genome editing technologies, such as CRISPR-Cas9, to introduce multiple agronomic traits in Brassica species. This combination of advanced biotechnological tools holds significant promise for enhancing B. oleracea breeding programs in the actual climate change context.

Abstract:The aim of any breeding process is to fully express the targeted, superior/desirable parent characteristic in the progeny. Hybrids are often used in this dynamic, and complex process for which homozygous parents-which may require up to eight generations of back crossing and selection-are required. Doubled haploid (DH) technologies can facilitate the production of true breeding lines faster and in a more efficient manner than the traditional back crossing and selection strategies. Sunflower is the third most important oilseed crop in the world and has no available double haploid induction procedure/technique that can be efficiently used in breeding programs. A reproducible and efficient doubled haploid induction method would be a valuable tool in accelerating the breeding of new elite sunflower varieties. Although several attempts have been made, the establishment of a sunflower doubled haploid induction protocol has remained a challenge owing recalcitrance to in vitro culture regeneration. Approaches for haploid development in other crops are often cultivar specific, difficult to reproduce, and rely on available tissue culture protocols-which on their own are also cultivar and/or species specific. As an out-crossing crop, the lack of a double haploid system limits sunflower breeding and associated improvement processes, thereby delaying new hybrid and trait developments. Significant molecular advances targeting genes, such as the centromeric histone 3 (CenH3) and Matrilineal (MTL) gene with CRISPR/Cas9, and the successful use of viral vectors for the delivery of CRISPR/Cas9 components into plant cells eliminating the in vitro culture bottleneck, have the potential to improve double haploid technology in sunflower. In this review, the different strategies, their challenges, and opportunities for achieving doubled haploids in sunflower are explored.
A biotechnology-based male-sterility system for hybrid seed production in tomato

Abstract:Incorporating male sterility into hybrid seed production reduces its cost and ensures high varietal purity. Despite these advantages, male-sterile lines have not been widely used to produce tomato (Solanum lycopersicum) hybrid seeds. We describe the development of a biotechnology-based breeding platform that utilized genic male sterility to produce hybrid seeds. In this platform, we generated a novel male-sterile tomato line by clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 (Cas9)-mediated mutagenesis of a stamen-specific gene SlSTR1 and devised a transgenic maintainer by transforming male-sterile plants with a fertility-restoration gene linked to a seedling-colour gene. Offspring of crosses between a hemizygous maintainer and the homozygous male-sterile plant segregated into 50% non-transgenic male-sterile plants and 50% male-fertile maintainer plants, which could be easily distinguished by seedling colour. This system has great practical potential for hybrid seed breeding and production as it overcomes the problems intrinsic to other male-sterility systems and can be easily adapted for a range of tomato cultivars and diverse vegetable crops.

Abstract:The practice of hybridization has greatly contributed to the increase in crop productivity. A major component that exploits heterosis in crops is the cytoplasmic male sterility (CMS)/nucleus-controlled fertility restoration (Rf) system. Through positional cloning, it is shown that heterozygous alleles (RsRf3-1/RsRf3-2) encoding pentatricopeptide repeat (PPR) proteins are responsible for restoring fertility to cytoplasmic male-sterile radish (Raphanus sativus L.). Furthermore, it was found that heterozygous alleles (RsRf3-1/RsRf3-2) show higher expression and RNA polymerase II occupancy in the CMS cytoplasmic background compared with their homozygous alleles (RsRf3-1/RsRf3-1 or RsRf3-2/RsRf3-2). These data provide new insights into the molecular mechanism of fertility restoration to cytoplasmic male-sterile plants and illustrate a case of overdominance.

Abstract:Wide or distant hybridization has been widely used as an important tool of chromosome manipulation for crop improvement. The chromosome behaviors in F1 hybrids provide us with the essential genetic basis for chromosome manipulation. The induction of homoeologous pairing in F1 hybrid plants followed by the incorporation of a single-chromosome fragment from an alien or a wild species into an existing crop species by translocating chromosomes has been used in the production of translocation lines. Most efforts to transfer a beneficial trait from wild plants into crops so far have bridged the species gap via alien chromosome translocation lines. Chromosome doubling in somatic cells or gametes of F1 hybrids followed by the incorporation of all alien chromosomes has been used in the production of amphidiploids. Amphidiploidy can be used for a bridge to move a single chromosome from one species to another or for the development of new crops. Chromosome elimination of a uniparental genome during the development of F1 hybrid embryos has been used in the production of haploids. Haploids are very useful in double-haploid breeding of a true-breeding crop such as wheat and rice since this method can quickly replace genetic recombination while enhancing breeding efficiency or facilitating genetic analysis.

However, the commercial application of male sterility in hybrid seed production has not been widely used in tomatoes. CRISPR/Cas9-mediated gene editing can facilitate acceleration for the practical application of male sterility in hybrid seed production. Here, by using the CRISPR-Cas9 system, two genes DYSFUNCTIONAL TAPETUM1(SlDYT1) and Glutathione S-transferase (SlGSTAA), which underly the two closely linked loci Male sterile 10 (Ms10) and Anthocyanin absent (AA), were knocked out simultaneously in two tomato parental lines. The generated dyt1gstaa double mutants developed green hypocotyl owing to anthocyanin deficiency and exhibited stable male sterility. Up to 92% effectiveness in selecting male sterility was achieved using green hypocotyl as a morphological marker, and thereafter an efficient and stable propagation strategy of male sterility with the aid of the morphological marker selection was developed. Furthermore, dyt1gstaa-derived hybrid seeds were produced and found to have comparable yield, weight, and germination rate with the corresponding WT-derived F1 seeds. The dyt1gstaa system not only increased hybrid seed purity to 100% but also facilitated its rapid and cost-effective determination. Moreover, this system was discovered to have no evident side effects on important agronomic traits. This study suggested that our CRISPR/Cas9-created dyt1gstaa system can be deployed in tomato hybrid seed production.

Abstract:Key messageA comprehensive understanding of CMS/Rf system enabled by modern omics tools and technologies considerably improves our ability to harness hybrid technology for enhancing the productivity of field crops.AbstractHarnessing hybrid vigor or heterosis is a promising approach to tackle the current challenge of sustaining enhanced yield gains of field crops. In the context, cytoplasmic male sterility (CMS) owing to its heritable nature to manifest non-functional male gametophyte remains a cost-effective system to promote efficient hybrid seed production. The phenomenon of CMS stems from a complex interplay between maternally-inherited (mitochondrion) and bi-parental (nucleus) genomic elements. In recent years, attempts aimed to comprehend the sterility-inducing factors (orfs) and corresponding fertility determinants (Rf) in plants have greatly increased our access to candidate genomic segments and the cloned genes. To this end, novel insights obtained by applying state-of-the-art omics platforms have substantially enriched our understanding of cytoplasmic-nuclear communication. Concomitantly, molecular tools including DNA markers have been implicated in crop hybrid breeding in order to greatly expedite the progress. Here, we review the status of diverse sterility-inducing cytoplasms and associated Rf factors reported across different field crops along with exploring opportunities for integrating modern omics tools with CMS-based hybrid breeding.
