Abstract:For the past 4 decades, intensive molecular studies of mostly leaf mesophyll cell-infecting pathogens and chewing insects have led to compelling models of plant-pathogen and plant-insect interactions. Yet, some of the most devastating pathogens and insect pests live in or feed on the phloem, a systemic tissue belonging to the plant vascular system. Phloem tissues are difficult to study, and phloem-inhabiting pathogens are often impossible to culture, thus limiting our understanding of phloem-insect/pathogen interactions at a molecular level. In this Perspective, we highlight recent literature that reports significant advances in the understanding of phloem interactions with insects and prokaryotic pathogens and attempt to identify critical questions that need attention for future research. It is clear that study of phloem-insect/pathogen interactions represents an exciting frontier of plant science, and influx of new scientific expertise and funding is crucial to achieve faster progress in this important area of research that is integral to global food security.

Abstract:Plant viruses transmitted by arthropods,as an important biotic factor,may not only directly affect the yield and quality of host plants,and development,physiological characteristics and ecological performances of their vector arthropods,but also directly or indirectly affect the non-vector herbivorous arthropods and their natural enemies in the same ecosystem,thereby causing influences to the whole agro-ecosystem. This paper reviewed the progress on the effects of plant viruses on herbivorous arthropods,including vector and non-vector,and their natural enemies,and on their ecological mechanisms to provide a reference for optimizing the management of vector and non-vector arthropod populations and sustainable control of plant viruses in agro-ecosystem.

Abstract:Attacks on plants by both viruses and their vectors is common in nature. Yet the dynamics of the plant-virus-vector tripartite system, in particular the effects of viral infection on plant-insect interactions, have only begun to emerge in the last decade. Viruses can modulate the interactions between insect vectors and plants via the jasmonate, salicylic acid and ethylene phytohormone pathways, resulting in changes in fitness and viral transmission capacity of their insect vectors. Virus infection of plants may also modulate other phytohormones, such as auxin, gibberellins, cytokinins, brassinosteroids and abscisic acid, with yet undefined consequences on plant-insect interactions. Moreover, virus infection in plants may incur changes to other plant traits, such as nutrition and secondary metabolites, that potentially contribute to virus-associated, phytohormone-mediated manipulation of plant-insect interactions. In this article, we review the research progress, discuss issues related to the complexity and variability of the viral modulation of plant interactions with insect vectors, and suggest future directions of research in this field.

Abstract:Like humans, animals use plants and other materials as medication against parasites. Recent decades have shown that the study of insects can greatly advance our understanding of medication behaviors. The ease of rearing insects under laboratory conditions has enabled controlled experiments to test critical hypotheses, while their spectrum of reproductive strategies and living arrangements – ranging from solitary to eusocial communities – has revealed that medication behaviors can evolve to maximize inclusive fitness through both direct and indirect fitness benefits. Studying insects has also demonstrated in some cases that medication can act through modulation of the host's innate immune system and microbiome. We highlight outstanding questions, focusing on costs and benefits in the context of inclusive host fitness.

Abstract:Insects are the host or vector of diverse viruses including those that infect vertebrates, plants, and fungi. Insect viruses reside inside their insect hosts and are vertically transmitted from parent to offspring. The insect virus-host relationship is intricate, as these viruses can impact various aspects of insect biology, such as development, reproduction, sex ratios, and immunity. Arthropod-borne viruses (arboviruses) that cause substantial global health or agricultural problems can also be vertically transmitted to insect vector progeny. Multiple infections with insect viruses and arboviruses are common in nature. Such coinfections involve complex interactions, including synergism, dependence, and antagonism. Recent studies have shed light on the influence of insect viruses on the competence of insect vectors for arboviruses. In this review, we focus on the biological effects of insect viruses on the transmission of arboviruses by insects. We also discuss the potential mechanisms by which insect viruses affect the ability of hosts to transmit arboviruses, as well as potential strategies for disease control through manipulation of insect viruses. Analyses of the interactions among insect vectors, insect viruses and arboviruses will provide new opportunities for development of innovative strategies to control arbovirus transmission.

Abstract:Insects are abundant members of all terrestrial and freshwater ecosystems and are also of immense economic importance, largely as pests and disease vectors, but occasionally having a more positive influence on human well-being as in the case of silk moths and honey bees. That they are subject to a variety of diseases has been understood for several centuries, but it has only been with the development of modern microbiology in the last sixty or so years that the widespread occurrence and biological diversity of insect diseases has been appreciated. Much of the impetus for the study of insect diseases has come from the possibility that they might be used as biological control agents to combat noxious pests, or from the need to preserve the health of economically valuable species. Thus the potential for insect diseases to control or regulate their host populations has been understood for many years. However, the ecological study of the population dynamics of insect-pathogen interactions is a relatively new field of biology and most work in this area has been carried out in the last fifteen years.The aim of this chapter is twofold: first, briefly to review the theory of insect-pathogen population dynamics and to try and identify areas that require further study; and second, to examine the most important case studies of insect-pathogen population dynamics and to discuss whether the predictions from theory are concordant with field experiments and observations. Abstract:Most plant viruses are transmitted by vectors,among which insect is the most important category. The tripartite interaction of plant-virus-vector is complex and specific. So far,virus has become a powerful booster for co-evolution of plant-virus-vector due to its rapid reproduction and fast variation,which then is conducive for its effective transmission. The effects of virus on vector insects can be direct and indirect. The indirect effect refers to such process,i.e.,virus infection to host plants results in the changes on the photosynthesis,secondary metabolism,nutrition components,and signal pathways of plants,which thus affects insects'biological properties,such as their behavior of host selection,growth and development,fertility and so on,and finally influences virus'transmission efficiency. The biological impacts of virus-infected host plants on piercing-sucking insects are reviewed here,aiming at providing evidences for the prevention and control of plant virus diseases.

Abstract:Bacteria associated with insects potentially provide many beneficial services and have been well documented. Mutualism that relates to insects is widespread in ecosystems. However, the interrelation between “symbiotic bacteria” and “mutualism” has rarely been studied. We introduce three systems of mutualism that relate to insects (ants and honeydew-producing Hemiptera, fungus-growing insects and fungi, and plant persistent viruses and vector insects) and review the species of symbiotic bacteria in host insects, as well as their functions in host insects and the mechanisms underlying mutualism regulation. A deeper understanding of the molecular mechanisms and role of symbiotic bacteria, based on metagenomics, transcriptomics, proteomics, metabolomics, and microbiology, will be required for describing the entire interaction network.

Molecular Plant-Microbe Interactions

Abstract:A successful viral infection requires complex, compatible molecular interactions between the invading virus and the host. A better understanding of such interactions may assist in the development of novel approaches to control viral diseases for sustainable crop production. In the past decade, the cell biology of virus-host and virus-vector interactions has been one of the most exciting areas of research in the molecular plant-microbe field. This is partially attributed to the availability of powerful cell biology techniques, including imaging tools like confocal microscopy and electron microscopy and tomography. As a result, there has been an unprecedented increase in knowledge in the areas of the bi- and tripartite interactions of virus, host, and vector. We now have a much clearer picture of viral virulence mechanisms, virus-induced host defenses, viral counteracting strategies, and viral circulations in the insect vectors. This Focus Issue highlights molecular virus-plant and virus-vector interactions in the areas of cell biology and closely related disciplines and explores biotechnology-based antiviral strategies using knowledge generated from these research areas.

Abstract:Of the approximately 1,200 plant virus species that have been described to date, nearly one-third are single-stranded DNA (ssDNA) viruses, and all are transmitted by insect vectors. However, most studies of vector transmission of plant viruses have focused on RNA viruses. All known plant ssDNA viruses belong to two economically important families, Geminiviridae and Nanoviridae, and in recent years, there have been increased efforts to understand whether they have evolved similar relationships with their respective insect vectors. This review describes the current understanding of ssDNA virus-vector interactions, including how these viruses cross insect vector cellular barriers, the responses of vectors to virus circulation, the possible existence of viral replication within insect vectors, and the three-way virus-vector-plant interactions. Despite recent breakthroughs in our understanding of these viruses, many aspects of plant ssDNA virus transmission remain elusive. More effort is needed to identify insect proteins that mediate the transmission of plant ssDNA viruses and to understand the complex virus-insect-plant three-way interactions in the field during natural infection.

Abstract:In the interaction between the viruses and their hosts,the viruses are more than just pathogens.They are also critical symbiotic partners in maintaining the health of their hosts.Mutualistic interactions are found between virus and microbes,plants,insects,and mammalians,and some interactions involve multiple players of the holobiont.Along with the discovery of new viruses,more mutualistic interactions are being described and more will undoubtedly be discovered.This review highlighted some of the recent and dramatic examples of beneficial viruses,demonstrating why viruses need to be taken seriously not just as pathogens but as integrated members of the holobiont.

Abstract:In nature, insects face a constant threat of infection by numerous exogeneous viruses, and their intestinal tracts are the predominant ports of entry. Insects can acquire these viruses orally during either blood feeding by hematophagous insects or sap sucking and foliage feeding by insect herbivores. However, the insect intestinal tract forms several physical and immunological barriers to defend against viral invasion, including cell intrinsic antiviral immunity, the peritrophic matrix and the mucin layer, and local symbiotic microorganisms. Whether an infection can be successfully established in the intestinal tract depends on the complex interactions between viruses and those barriers. In this review, we summarize recent progress on virus-intestinal tract interplay in insects, in which various underlying mechanisms derived from nutritional status, dynamics of symbiotic microorganisms, and virus-encoded components play intricate roles in the regulation of virus invasion in the intestinal tract, either directly or indirectly.

Abstract:There is growing evidence of the importance of microbial mutualistic symbioses in insect–plant interactions. Mutualists may affect host plant range and enable insects to manipulate plant physiology for their own benefit. The plant can also be a route for the horizontal transfer of mutualistic microorganisms among their host insects. Where this occurs, selection for improved transmission might cause the insect mutualist to damage the plant and become a plant pathogen. Insect microbial associates can influence ecological communities by changing the way the plant interacts with their hosts' competitors and natural enemies. We review recent research in this field and suggest that insect mutualists may be more important ‘hidden players' in insect–plant interactions than is currently realised.

Abstract:Insects are a group of arthropods and the largest group of animals on Earth, with over one million species described to date. Like other life forms, insects suffer from viruses that cause disease and death. Viruses that are pathogenic to beneficial insects cause dramatic economic losses on agriculture. In contrast, viruses that are pathogenic to insect pests can be exploited as attractive biological control agents. All of these factors have led to an explosion in the amount of research into insect viruses in recent years, generating impressive quantities of information on the molecular and cellular biology of these viruses. Due to the wide variety of insect viruses, a better understanding of these viruses will expand our overall knowledge of their virology. Here, we review studies of several newly discovered RNA insect viruses in China.

Abstract:The majority of plant viruses are transmitted by hemipteran insects. Bacterial symbionts in hemipteran hosts have a significant impact on the host life, physiology and ecology. Recently, the involvement of bacterial symbionts in hemipteran vector-virus and vector-plant interactions has been documented. Thus, the exploitation and manipulation of bacterial symbionts have great potential for plant viral disease control. Herein, we review the studies performed on the impact of symbiotic bacteria on plant virus transmission, including insect-bacterial symbiont associations, the role of these bacterial symbionts in viral acquisition, stability and release during viral circulation in insect bodies, and in viral vertical transmission. Besides, we prospect further studies aimed to understand tripartite interactions of the virus-symbiotic microorganisms-insect vector.

Abstract:Interactions between vector insects,plant viruses and host plants are complex and diverse.Although much work has been done to study the tripartite relationships,their roles in biological invasions have been rarely explored.The limited case studies available indicate that the interactions may be mediated by the host plant susceptibility to viruses,the suitability of host plants to vector insects,and the insects' capacity to utilize host plants.When a host plant is highly susceptible to the virus but shows a low level of suitability to the insect,and the insect has a strong capacity to use different host plants,an indirect mutualistic relationship is likely to occur between vector insect and plant virus via their shared host plants.This kind of mutualism can contribute to the widespread invasion of vector insects as well as the epidemics of plant viruses.In view of the significance of the tripartite interactions in biological invasions,future effort should be made to investigate comparatively many more combinations of different species,and various technologies can be used to reveal the physiological and molecular mechanisms of the interactions.
