Abstract:This study was aimed at investigating the hypoglycemic and hypolipidemic effects of a polysaccharide (RTFP) isolated from Rosa roxburghii Tratt fruit on type-2 diabetic db/db mice. The results indicated that the oral administration of RTFP could significantly decrease the body weight, fat, and liver hypertrophy and the levels of fasting blood glucose, serum insulin, and serum lipids of the db/db mice. Histopathological observation showed that RTFP could effectively protect the pancreas, liver, and epididymal fat against damage and dysfunction. Real-time quantitative polymerase chain reaction analysis confirmed that the gene expression levels of peroxisome proliferator-activated receptors-γ (PPAR-γ), sterol regulatory element-binding protein-1 (SREBP-1c), acetyl-CoA carboxylase-1 (ACC-1), fatty acid synthase (FAS), and glucose-6-phosphatase (G6 Pase) were significantly down-regulated in the liver of db/db mice after treatment with RTFP. Moreover, RTFP treatment reversed gut dysbiosis by lowering the Firmicutes-to-Bacteroidetes ratio and enhancing the relative abundances of beneficial bacteria including Bacteroidaceae, Bacteroidaceae S24-7 group, and Lactobacillaceae. These findings suggest that RTFP can be used as a promising functional supplement for the prevention and treatment of type-2 diabetes mellitus.

Abstract:Polysaccharide from Rosa roxburghii Tratt fruit (RTFP) ameliorates high-fat diet (HFD)-induced colitis in mice. However, it is still unknown whether the gut microbiota can mediate the anti-colitis effects of RTFP in mice. This research aims to investigate the role of gut microbes in modulating RTFP in colitis mice through fecal microbiota transplantation (FMT). The findings demonstrated that RTFP exhibited prebiotic effects on HFD-induced colitis mice. After FMT treatment (transplatation of the microbiota from the fecal sample to each recipient daily), the fecal microbiota of RTFP-treated donor mice remarkably alleviated colitis-related symptoms (e.g., colonic inflammation, loss of body weight, gut microbiota dysbiosis, and loss of barrier integrity) and upregulated the expression of tight junction proteins compared to the HFD-treated donor mice. Overall, RTFP can reduce the severity of HFD-induced colitis by regulating gut microbiota.

Abstract:In this study, the mitigative effect of Rhodomyrtus tomentosa (Ait.) Hassk fruit extract rich in phenolic compounds (RTE) on high fat diet (HFD)-induced intestinal barrier dysfunction of mice and the underlying mechanism were explored. The results revealed that RTE supplementation obviously improved gut microbiota dysbiosis induced by HFD, which was evidenced by elevated alpha diversity, suppressed Firmicutes/Bacteroidetes ratio, enriched short-chain fatty acid-producing bacteria (Odoribacter, Parabacteroides, Blautia and Akkermansia), and depleted harmful bacteria (Helicobacter, norank_f_ Desulfovibrionaceae and Mucispirillum). RTE intervention mitigated intestinal barrier dysfunction and inflammation by elevating tight junction proteins expression levels and decreasing proinflammatory cytokines levels. Furthermore, RTE administration inhibited the HFD-induced trigger of the lipopolysaccharide-toll-like receptor 4-nuclear factor kappa-B (LPS-TLR4-NF-κB) pathway in colonic tissue. Therefore, RTE supplementation may be an effective way to protect the intestinal tract in HFD-induced obese individuals.

Abstract:In the present study, the digestion properties of a polysaccharide from Rosa roxburghii fruit (RTFP-3) under saliva, simulated gastric and small intestinal conditions were studied for the first time. The results indicated that human saliva had no effect on RTFP-3. RTFP-3 was slightly degraded in simulated gastric and small intestinal juices. The molecular weight (Mw ) of RTFP-3 was decreased from 67.59 to 44.22 kDa. Meanwhile, the reducing sugars were increased from 0.503 to 1.744 mM. Then, the prebiotic effects of RTFP-3 were examined by in vitro fermentation model. RTFP-3 could significantly increase the production of total short-chain fatty acids from 23.49 to 44.29 mM, and 60.28% of total carbohydrate was consumed after 48 h fermentation. Furthermore, RTFP-3 could significantly modulate the microbial structure by lowering the ratio of Firmicutes to Bacteroidetes from 14.89 to 4.68 after 48 h fermentation, and improving the relative abundances of some beneficial gut microbiota.

Abstract:Ulcerative colitis (UC) is a non-specific inflammatory bowel illness characterized by intestinal mucosal barrier degradation, inflammation, oxidative damage, and gut microbiota imbalances. Rosa roxburghii Tratt Fruit extract (RRTE) was extracted from Rosa roxburghii Tratt fruit, exhibiting an excellent prevention effect against UC; RRTE could prevent the damage of DSS-induced human normal colonic epithelial (NCM 460) cells, especially in cell viability and morphology, and oxidative damage. Additionally, in UC mice, RRTE could limit the intestinal mucosal barrier by increasing the expression of intestinal tight junction proteins and mucin, reducing inflammation and oxidative damage in colon tissue. More importantly, RRTE can increase the abundance of beneficial bacteria to regulate gut microbiota such as Ruminococcus, Turicibacter, and Parabacteroides, and reduce the abundance of harmful bacteria such as Staphylococcus and Shigella. Furthermore, transcriptomics of colonic mucosal findings point out that the beneficial effect of RRTE on UC could be attributed to the modulation of inflammatory responses such as the IL-17 and TNF signaling pathways. The qPCR results confirm that RRTE did involve the regulation of several genes in the IL-17 signaling pathway. In conclusion, RRTE could prevent DSS-induced damage both in vitro and in vivo.

Abstract:Various plant polyphenols have been recognized by researchers as functional foods to improve the immune system. Our study showd that Rosa roxburghii Tratt fruit polyphenols (RRTP) could regulate the cellular and innate immune functions of mice. In addition, Compared with the normal control group, RRTP could affecting the balance of lymphocyte subsets. Metabolism studies showed that RRTP altered 46 different metabolites, mainly involving amino acid metabolism, carbohydrate metabolism, and lipid metabolism. Moreover, RRTP has a positive regulatory effect on the gut microbiota, the short-chain fatty acids(SCFAs) producing bacterias: Blautia, Bacteroides, Lachnospiraceae_NK4A136_group, Roseburia increased notably. Measurements of SCFAs in the cecum showed significant increases, suggesting that the immunomodulatory effects of RRTP are closely related to SCFAs. In Conclusion, RRTP could regulate the immune system of mice by regulating the balance of metabolites and intestinal microorganisms, RRTP has the potential to be developed as functional ingredients or foods to promote health.

A polysaccharide from salviae miltiorrhizae radix inhibits weight gain of mice with high-fat diet via modulating intestinal bacteria

Abstract:Background: Obesity, a global chronic disease, has been recognized as a severe risk to health. In our study, a novel polysaccharide named ARS was isolated and purified from aerial part of salviae miltiorrhizae radix. Our aim is to investigate the weight-reducing effect of a polysaccharide from salviae miltiorrhizae radix on mice fed a high-fat diet. Results: The novel polysaccharide ARS mainly consisted of glucose and galactose with a molar ratio of 0.59:1.00. We found that treatment with ARS could inhibit weight gain of mice fed a high-fat diet via modulating the intestinal bacteria. Moreover, we surveyed its mechanism in mice, and the gut microbiota sequencing results demonstrated that ARS can reverse or resist high-fat-diet-induced significant weight gain or obesity by increasing the diversity of gut microbiota and optimizing the ratio of Firmicutes to Bacteroidetes. Phylum and species analysis of gut microbiota demonstrated that obesity caused by a high-fat diet was accompanied by significant changes in the microbial communities, but ARS could reverse the disturbance of gut microbiota induced by the high-fat diet to maintain homeostasis. Conclusion: Overall, our findings suggested a new function of ARS in regulating gut microbiota, which provides a theoretical basis for the development of high-quality ARS functional foods and the application of dietary supplements. © 2023 Society of Chemical Industry.
Protective effect of soluble dietary fiber from Rosa roxburghii Tratt residue on dextran sulfate sodium-induced ulcerative colitis by regulating serum metabolism and NF-κB pathway in mice

Abstract:Background: Ulcerative colitis (UC) refers to an idiopathic chronic inflammatory bowel disease that starts with inflammation of the intestinal mucosa. Dietary fiber plays a crucial role in maintaining the normal architecture of the intestinal mucosa. In this study, the protective effect and potential mechanism of soluble dietary fiber from Rosa roxburghii Tratt residue (SDFR) on dextran sulfate sodium (DSS)-induced UC mice were explored. Results: The results revealed that SDFR could ameliorate body weight loss and pathological injury, improve the structure and crypt destruction in colon in DSS-induced mice. Moreover, the levels of NO, IL-1β, TNF-α, MPO and protein expression of iNOS and COX-2 were decreased after administration of SDFR. Notably, nontargeted metabolomics analysis indicated that there were significant differences in 51 potential metabolites in serum between the DSS and control groups. SDFR intervention could regulate aberrant alterations of these metabolites and mitigate UC via regulating metabolic pathways, including arachidonic acid and glycerophospholipid metabolism. Conclusion: This study provides novel evidence that SDFR could be used as a potential modulator to relieve UC. Also, the results provide a theoretical basis for the utilization of byproducts in Rosa roxburghii Tratt fruit processing. © 2024 Society of Chemical Industry.

Abstract:This study mainly investigated the regulatory effect of Rosa roxburghii Tratt fruit juice fermented by Lacticaseibacillus paracasei SR10-1 (LAB-RRTJ) on modulating gut microbiota in dextran sulfate sodium (DSS)-induced ulcerative colitis in mice. Compared to control group, DSS induction decreased body weight of mice, indexes of Shannon, Simpson, Chao1 and Faith_pd, and increased disease activity index (DAI) and levels of interleukin 1 beta (IL-1 beta), IL -6, tumor necrosis factor alpha (TNF-alpha) and interferon-gamma (IFN-gamma); And this induction also led to an increase in Proteobacteria, Verrucomicrobia and Actinobacteria at phylum level, harmful bacterial species richness at genus level, and relative richness of S. sciuri , Desulfovibrio C21_c20, R. gnavus and Akkermansia muciniphila at species level, and a decrease in Firmicutes at phylum level and relative richness of B. acidifaciens in mice. LAB-RRTJ increased body weight of mice with DSS induced ulcerative colitis (UC) and indexes of Shannon, Simpson, Chao1 and Faith_pd, reduced DAI and the content of four inflammatory factors and improved gut microbiota imbalance in DSS induced UC mice. Besides, the number of operational taxonomic units (OTUs) increased, alpha -diversity and fl -diversity were restored and similar to those in mice in the control group after LAB-RRTJ treatment. Compared with the positive drug treatment group, LAB-RRTJ has a better effect on regulating gut microbiota diversity in colitis mice. Correlation analysis showed that inflammatory factors were positively correlated with harmful bacteria and negatively correlated with beneficial bacteria which commonly found in some colitis mice. Taken together, our study demonstrated that LAB-RRTJ could alleviate DSS-induced colitis in mice through the modulation of inflammatory cytokines and gut microbiota composition. Abstract:Obesity and associated liver diseases are becoming global public health challenges. Raspberry (Rubus chingii Hu.), as a medicine food homology plant, possesses a series of health-promoting properties, but its protective effect on obesity-related liver injury and the potential mechanisms remain obscure. Herein high-fat diet (HFD)-fed mice were orally treated with raspberry polysaccharides (RCP) for 14 weeks. Treatment with RCP alleviated obesity and associated symptoms including hyperglycemia, hyperlipemia, endotoxemia, as well as hepatic inflammation and oxidant stress in HFD-induced obese mice. RCP restructured the gut microbiota and host metabolism especially by increasing the levels of Dubosiella and its metabolite butyrate. Besides, exogenous butyrate supplementation protected against intestinal barrier disruption, and thereby reduced inflow of lipopolysaccharide and mitigated inflammation and oxidative injury in the liver of obese mice. Therefore, we suggest that RCP can be utilized as a novel prebiotics to improve obesity-induced hepatic oxidative injury by enhancing butyrate-mediated intestinal barrier function.
Phytochemical profile, bioactivity and prebiotic potential of bound polyphenols released from Rosa Roxburghii fruit pomace dietary fiber during in vitro digestion and fermentation

Abstract:The aim of this study was to elucidate liberation and phytochemical profile of bound polyphenols existed in dietary fiber (RPDF) isolated from Rosa roxburghii fruit pomace during in vitro simulated digestion and colonic fermentation. The variation of biological activity and prebiotic potential of released polyphenols from RPDF were investigated. The results showed that bound polyphenols were released from RPDF at a higher ratio during colonic fermentation than gastrointestinal digestion. The released polyphenols showed antioxidant activity and α-glucosidase inhibitory activity. In vitro colonic fermentation assay showed that compared with dephenolized RPDF (DP-RPDF), RPDF exhibited a significantly stronger prebiotic effect through lowering Firmicutes to Bacteroidetes (F/B) ratio at phylum level, increasing the relative abundances of beneficial bacteria at genus level, and improving production of short chain fatty acids (SCFAs). These findings indicate that the bound polyphenols may act as important functional food ingredients that could contribute to the biological properties of RPDF. In addition, this work provides new insights into high-value utilization of Rosa roxburghii fruit pomace, which will reduce resource waste and make a great contribution to environmental protection.

Abstract:Rosa roxburghii fruit is an underutilized functional food abundant in polyphenols. Polyphenols have been proved to have antidiabetic effects. This study investigates the effects of Rosa roxburghii fruit polyphenols extract (RPE) on plasma metabolites and gut microbiota composition in streptozotocin (STZ)- and high-fat diet- induced type 2 diabetes using metabolomics and 16S rRNA gene sequencing. The induced diabetic mice were fed with 400 mg/kg body weight RPE for 8 weeks. RPE demonstrated hypoglycemic, hypolipidemic, and anti-inflammatory effects. Colonic oxidative stress biomarkers were also lowered by RPE. Besides, RPE decreased plasma ceramides and tyrosine levels and increased carnitine and phosphatidylinositols levels, indicating improved insulin resistance, lipid metabolism, and immune response. Furthermore, RPE decreased abundances of Lachnospiraceae and Rikenellaceae and increased abundances of Erysipelotrichaceae and Faecalibaculum. Metabolic function prediction of the gut microbiota by PICRUSt demonstrated that RPE downregulated the phosphotransferase system. Taken together, these findings demonstrated that RPE has the potential to prevent type 2 diabetes by regulating the plasma metabolites and gut microbes.

Abstract:Rosa roxburghii Tratt fruit is a highly valued fruit that contains abundant functional and nutritional constituents. In this study, a novel polysaccharide, named RTFP-1, was isolated and purified from R. roxburghii Tratt fruit. Structural characterization indicated that RTFP-1 was a homogeneous heteropolysaccharide with the molecular weight (Mw) of 128.7 kDa and consisted of arabinose, galactose, glucose, mannose, xylose, and fucose with molar ratio percentages of 34.84, 40.59, 12.11, 5.06, 3.39, and 4.01%, respectively. A CCK-8 assay indicated that RTFP-1 inhibited the cell growth of HepG2 cells in a dose-dependent manner. Morphological analysis and flow cytometry experiment showed that RTFP-1 promoted the apoptosis of HepG2 cells and increased reactive oxygen species (ROS) level. The underlying molecular mechanisms indicated that RTFP-1 activated the apoptosis of HepG2 cells through ROS-mediated MAPK, STAT, and p53 apoptotic pathways. These results suggest that RTFP-1 might be a potential chemopreventive and antitumor agent.

Abstract:A new Rosa roxburghii Tratt fruit polysaccharide (RTFP)-iron (III) complex was synthesized and characterized by various physicochemical, thermal, and structural analyses. The results showed that RTFP-iron complex was formed by the Fe-O bonding between hydroxyl and carboxyl groups in RTFP and iron. The Fe content of RTFP-iron (III) complex was 16.17%, and its surface morphology displayed relatively scattered and lumpy. In addition, the scavenging activities of RTFP and RTFP-iron (III) on DPPH, hydroxyl radicals and ABTS were examined. The results demonstrated that the antioxidant activity of RTFP-iron complex was weaker than that of RTFP. The release of iron from RTFP-iron (III) complex was observed during simulated gastrointestinal digestion. Furthermore, polyphenol and/or trypsin could enhance the stability and solubility of RTFP-iron (III) complex compared to FeCl3. Altogether, our findings revealed RTFP-iron (III) could serve as a new promising iron supplement. Practical applications As a new type of iron supplement, the polysaccharide-iron complex is an efficient water-soluble iron supplement with less adverse reaction and higher bioavailability. In our work, polysaccharides from Rosa roxburghii Tratt fruit and polysaccharide-iron (III) complex were prepared. The results demonstrated the iron in RTFP-iron (III) complex could have better stability and solubility. Apart from this, the bioavailability of RTFP-iron (III) complex was higher than that of FeCl3 and the complex had good antioxidant activity. The RTFP-iron (III) complex might be used as iron supplementary to relieve the side effects and oxidative stress. These results might support the future applications of RTFP-iron(III) in the pharmaceutical industry.
A Polysaccharide PRCP from Rosa Cymosa Tratt Fruit: Structural Characteristics and Immunomodulatory Effects Via MAPK Pathway Modulation in Vitro

Abstract:The Rosa cymosa Tratt, an herbal plant from the Rosaceae family, has historically been valued in China for its medicinal and edible properties. In this study, a novel polysaccharide from R. cymosa fruit, termed PRCP (purified R. cymosa polysaccharide), was isolated using water extraction, decolorization, deproteinization, and ion- exchange chromatography. The structural characteristics of PRCP were investigated using monosaccharide composition analysis, methylation, GPC, FTIR, CD, and NMR spectroscopy. The immunomodulatory effect and potential mechanism of PRCP were evaluated in vitro using a macrophage cell model. Results indicated that PRCP (37.28 kDa) is a highly branched polysaccharide (72.61 %) primarily composed of arabinogalactan, rhamnogalacturonan, and galactoglucan domains with 13 types of glycosidic linkage fragments. Furthermore, PRCP appears to modulate immunomodulatory effects by influencing the phosphorylation of P38 and JNK proteins in the MAPK pathway. Collectively, these findings highlight the potential of PRCP as a promising natural functional food ingredient for immunostimulation.
Flavonoids Extract from Rosa Roxburghii Fruit Ameliorates Carrageenan-Induced Thrombosis in Mice

Abstract:The fruits of Rosa roxburghii contain a high amount of flavonoids. We conducted a study using a carrageenan-induced thrombus model to investigate how Rosa roxburghii fruit flavonoids (RF) inhibit thrombosis using methods such as ELISA and 16S. The results showed that RF intervention reduced the length of the black tail in mice and controlled four hemagglutination parameters. Furthermore, RF treatment effectively reduced oxidative stress and inflammatory cytokines in the serum and tissues. RF also significantly inhibited platelet aggregation and promoted the synthesis of anticoagulant proteins. Pathological observations revealed that RF effectively alleviated tissue lesions and thrombosis. Through Western blot and qPCR analysis, we found that RF reduced inflammation by suppressing the NF-κB pathway in mice with thrombosis. The analysis of intestinal flora in mice, further revealed that RF intervention reduced the abundance of Bacteroides and Clostridium. Overall, these findings indicate that RF has biological activity in preventing carrageenan-induced thrombosis.
