Abstract:Good sense: A highly sensitive and selective ratiometric fluorescent probe for Fe2+ was developed by linking an Fe2+-insensitive fluorophore to an Fe2+-sensitive reporter–fluorophore fragment. The fluorescence intensity ratio increased linearly with the concentration of Fe2+ (see scheme). The probe was applied to the ratiometric imaging of intracellular and adscititious fluctuations of Fe2+ in live HL-7702 and HepG2 cells.

Abstract:A water-soluble rhodamine-based fluorescent probe (RD2) was designed and synthesized. Compared with the precursor RD1, RD2 shows excellent selectivity for Fe3+ over Cr3+ during the detection. The enhanced selectivity was achieved by incorporating another coordination moiety to the chelating moiety of the precursor RD1. Furthermore, bioimaging studies indicated that RD2 was cell permeable and suitable for the real-time imaging of Fe3+ in the living cells by confocal microscopy. (C) 2014 Elsevier B.V. Abstract:A novel rhodamine-based fluorescent probe 1 was synthesized, which was designed as a fluoroionophore and chromophore for the optical detection of Fe3+ ions in water with a broad pH span (4–8). As expected, it exhibited high selectivity and sensitivity for Fe3+ over other commonly coexistent metal ions in water. The detection limit was measured to be 1.5μM. The significant changes in the fluorescence color could be used for naked-eye detection. Furthermore, fluorescence imaging experiments of Fe3+ ions in living MGC803 cells demonstrated its value of practical applications in biological systems.

Abstract:Four novel rhodamine-based fluorescent probes (RE1-RE4) were designed and synthesized for sensitive detection of Fe3+ in water. RE1-RE4 exhibited low detection limits in the range of 18.6 nM to 29.4 nM. In addition, the binding modes of the probes (RE1-RE4) with Fe3+ were proved to be 1:1 stoichiometry of the complexes by Job's plot and also their complexing ability was determined using the binding constants (RE2 > RE4 > RE3 > RE1). RE2 was selected to study the proposed binding mechanism with Fe3+ by ESI-MS, H-1 NMR and DFT calculations. Furthermore, RE2 was further utilized to detect Fe3+ in living cells and real water samples, indicating its promising prospect in biological and environmental fields.

Abstract:Iron is the most abundant transition metal ion in the human body. It has unique redox activity and plays an important role in maintaining the normal function of cells. However, excessive iron produces highly reactive oxygen, which is harmful to organisms and can even cause cell damage or ferroptosis. The intracellular iron ion, especially Fe2+ which is not bound or weakly bound to the protein, has greater influence on the cellular function, and its metabolic abnormality is implicated in occurrence of a series of critical diseases. Detection the Fe2+ is of great demand for the investigation of aberrant iron related diseases. To this end, researchers have designed various fluorescent probes that can monitor Fe2+ in vitro and in vivo which substantially the accelerates the investigation and understanding of Fe2+ related diseases. In this review we summarize and classify Fe2+ fluorescent probes based on their different interaction mechanism with Fe2+, and introduce their applications in bioimaging, so as to provide clues for developing more reliable and applicable Fe2+ probes.

Abstract:A rhodamine derivative bearing a thiazole receptor (probe 1) was developed and characterized. Probe 1 rendered a prominently chromogenic and turn-on fluorescent signal toward Fe3+ ion in aqueous media based on coordination reaction. Probe 1 also showed high sensitivity toward Fe3+ ion with the detection limit as low as 92nM. By means of the Job's plot, 2:1 binding mode of 1-Fe3+ complex was determined and a possible sensing mechanism was further proposed based on the changes in the Fourier Transform Infrared (FT-IR) and mass spectra. The turn-on fluorescence and sensing mechanism for 1-Fe3+ complex were well demonstrated by theoretical calculations. Importantly, probe 1 has been tested highly suitable for mapping Fe3+ ion in human colon cancer cells SW480, providing a wonderful candidate for tracking Fe3+ in biological processes.

Abstract:We herein present an effective strategy to create water-soluble fluorescent bioimaging dyes by introducing the imidazolium-based ionic liquid (IL) pendants into a fluorescent skeleton. A new type of water-soluble imidazolium-anchored squaraine dye was synthesized accordingly. The relationship between the aggregate of squaraines and their fluorescent cell imaging application was elucidated in detail. Firstly, the aggregation behavior of squaraines in water solutions could be suppressed by varying the alkyl chain attached to the imidazolium unit. Secondly, the capability of cellular uptake and staining of dyes was also dramatically enhanced upon increasing the length of the paraffinic chain. These squaraine dyes displayed an excellent photostability that could permit real-time fluorescence bioimaging experiments to be monitored over a long time period with constant sample irradiation. Additionally, we designed for the first time an Fe(II)-ion probe on the basis of an attack of the hydroxyl radical to the four-membered ring of squaraine. The results demonstrated that the imidazolium-anchored squaraines could perform "naked-eye" detection of the Fe(2+) ion over a wide range of other interfering metals in aqueous media. More surprisingly, this process showed a fluorescence "turn-off" and "-on" response through the regeneration of squaraines in cells.

Abstract:A s the most abundant transition metal element in mammals, iron(Fe) plays a vital role in life activities. It is of great significance to study the variation of Fe 3+ level in living organisms. In virtue of the advantages of high sensitivity, good selectivity and low damage to living systems, the fluorescence detection of Fe 3+ has attracted much attention. Compared with the intensity-based fluorescent probe, the ratiometric fluorescent probe has less interference of environmental and can realize quantitative detection. In this study, four ratiometric Fe 3+ fluorescent probes, R 1 , R 2 , R 3 and R 4 , were designed and synthesized using fluorescence resonance energy transfer(FRET) mechanism to achieve quantitative detection of Fe 3+ . In the FRET systems, 1,8-naphthalimide fluorophore derivatives were adopted as donors while rhodamine B derivatives were selected as receptors. The connection sites of the donor and acceptor in R 3 and R 4 are different from those in R 1 and R 2. All the four probes showed good response and selectivity to Fe 3+ . The energy transfer efficiencies of R 3 and R 4 were obviously higher than those of R 1 and R 2. This work provided a promising strategy for the development of fluorescent ratiometic Fe 3+ sensors.

Abstract:Fe2+ plays a crucial role in maintaining physiological activities in the human body. However, abnormal Fe2+ levels can induce a variety of diseases. Therefore, the detection of Fe2+ in living organisms has attracted much attention in recent years. In this paper, we constructed two high-performance fluorescent probes based on 2-Hydroxy-3-pinanone for different channels detection of exogenous and endogenous Fe2+. These two probes could achieve the specific recognition of Fe2+ through introducing the nitrogen oxide group in the probes, which can serve as a highly reactive site for Fe2+. Both probes possessed very fast response times (<35 s), good sensitivity and selectivity, and low cytotoxicity for the detection of Fe2+. The exploration of detection conditions suggested that both probes can be well applied to the fluorescence imaging of Fe2+ in living cells and zebrafish. In addition, both probes successfully monitored the abnormal levels of Fe2+ in HeLa cells in a model of iron ferroptosis disease. These excellent properties enable these two probes to has highly promising for studying related diseases induced by Fe2+ abnormalities.

Abstract:A multifunctional fluorescent probe BHN-Fe 3 O 4 @SiO 2 nanostructure for Fe 3+ was designed and developed. It has a good selective response to Fe 3+ with fluorescence quenching and can be recycled using an external magnetic field. With adding EDTA (2.5 × 10 −5 M) to the consequent product Fe 3+ -BHN-Fe 3 O 4 @SiO 2 , Fe 3+ can be removed from the complex, and its fluorescence probing ability recovers, which means that this constituted on-off type fluorescence probe could be reversed and reused. At the same time, the probe has been successfully applied for quantitatively detecting Fe 3+ in a linear mode with a low limit of detection 1.25 × 10 −8 M. Furthermore, the BHN-Fe 3 O 4 @SiO 2 nanostructure probe is successfully used to detect Fe 3+ in living HeLa cells, which shows its great potential in bioimaging detection.

Abstract:A water-soluble "turn-on" fluorescent probe (RD1) for Fe(3+) based on rhodamine B was designed and synthesized. The fluorescent probe showed "turn-on" fluorescent and colorimetric responses to Fe(3+) with a high selectivity in water containing less than 1% organic cosolvent. Furthermore, bioimaging investigations indicated that the new probe was cell permeable and suitable for monitoring intracellular Fe(3+) in living cells by confocal microscopy with low cytotoxicity.

Abstract:A new fluorescent probe based on rhodamine was designed and synthesized. Upon addition of Fe3+,the probe exhibited selective fluorescence enhancement behavior,which indicated that the probe effectively avoided the fluorescence quenching for the paramagnetic nature of Fe3+ via its strong binding capability toward Fe3+. Further,the competing experiment showed that the probe will not be interfered by other common metal ions. Abstract:Fe3+ ions play an important role in both biological and environmental field. In this work, two novel rhodamine-based colorimetric and fluorescent probes (RBA2 and RBA3) were designed and synthesized for the efficient detection of Fe3+. Upon the addition of Fe3+, the fluorescence intensity of RBA2 and RBA3 enhanced 108-fold and 222-fold, respectively. RBA2 and RBA3 exhibited a low detection limit which could achieve 12.8 nM and 11.0 nM. In addition, the binding modes of RBA2 and RBA3 with Fe3+ were proved to be 1:1 stoichiometry in the complexes by Job's plot, ESI-MS and 1H NMR results. The complexing ability of RBA3 with Fe3+ excessed to that of RBA2 that was determined by the binding association constants, and highly consistent with DFT calculations results. Furthermore, RBA2 and RBA3 were further utilized to detect Fe3+ in living cells and real water samples, indicating their promising prospects in biological and environmental field.

Abstract:Two new rhodamine-based fluorescent probes were synthesized and characterized by NMR, high resolution mass spectrometer (HR-MS) and IR. The probes displayed a high selectivity for Fe3+ among environmentally and biologically relevant metal ions in aqueous solution (CH3OH-H2O = 3 : 2, v/v). The significant changes in the fluorescence color could be used for naked-eye detection. Job's plot, IR and H-1 NMR indicated the formation of 1: 1 complexes between sensor 1 and Fe3+. The reversibility establishes the potential of both probes as chemosensors for Fe3+ detection. The probe showed highly selectivity in aqueous solution and could be used over the pH range between 5 and 9. Abstract:A novel fluorescent composite () was synthesized for the selective detection of Fe 3+ based on the encapsulation of Carbon dots (CDs) into Zeolitic imidazolate frameworks (ZIF) with a two-dimensional leaf-like structure. The physicochemical structure, surface morphology, and composition of the adsorbent were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, and thermogravimetric analysis. The results showed that unlike the conventional three-dimensional rhombic structure (ZIF-8), the synthesized two-dimensional leaf-like nanosheets (ZIF-L) not only maintained the structural integrity but also had a permeable pore structure. Importantly, at an excitation wavelength of 350 nm, the composite's had significant fluorescence emission at 445 nm and was effectively quenched in the presence of Fe 3+ , which can be used as an effective fluorescent scaling probe. Under the optimal conditions, was used as a fluorescent probe to detect Fe 3+ with good linearity in the range of 0.02–2 mM (R 2 = 0.9948) and a low detection limit of 0.891 μM. The fluorescent probe can also be used for the determination of Fe 3+ in fruit samples with good recovery and accuracy, showing great potential for applications in environmental monitoring and agricultural pollutant control. Abstract:Three Rhodamine B derivatives were synthesized and characterized by ESI-MS, NMR, HR-MS and IR. The probes exhibit high selectivity and sensitivity towards Fe3+ over other metal ions in CH3CN-water. Upon the addition of Fe3+, the spirocyclic ring of the probe was opened and a significant enhancement of visible color and fluorescence within the range of 540-700 nm was observed. The colorimetric and fluorescent response to Fe3+ can be conveniently detected even by the naked eye, which provides a facile method for the visual detection of Fe3+. Job's plot, fluorescence titration and MS indicated the formation of 1:2 complexes between the probes and Fe3+. Abstract:A simple and versatile ratiometric fluorescent Fe(3+) detecting system, probe 1, was rationally developed based on the Fe(3+)-mediated deprotection of acetal reaction. Notably, this reaction was firstly employed to design fluorescent Fe(3+) probe. Upon treatment with Fe(3+), probe 1 showed ratiometric response, with the fluorescence spectra displaying significant red shift (up to 132 nm) and the emission ratio value (I522/I390) exhibiting approximately 2362-fold enhancement. In addition, the probe is highly sensitive (with the detection limit of 0.12 μM) and highly selective to Fe(3+) over other biologically relevant metal ions. The sensing reaction product of the probe with Fe(3+) was confirmed by NMR spectra and mass spectrometry. TD-DFT calculation has demonstrated that the ratiometric response of probe 1 to Fe(3+) is due to the regulation of intramolecular charge transfer (ICT) efficiency. Moreover, the practical utility in fluorescence detection of Fe(3+) in human blood serum was also conducted, and probe 1 represents the first ratiometric fluorescent probe that can be used to monitor Fe(3+) level in human blood serum. Finally, probe 1 was further employed in living cell imaging with pancreatic cancer cells, in which it displayed low cytotoxicity, satisfactory cell permeability, and selective ratiometric response to Fe(3+).

Abstract:Abnormal sulfur dioxide (SO 2 ) and microenvironmental viscosity are detrimental to human health, so the effective monitoring of these indicators in relevant foods and organisms is of great significance. The dual-functional fluorescent probes that are sensitive to active sulfur and microenvironment at the subcellular organelle level can link the spatiotemporal changes of different species, which are crucial for understanding complex biological processes. In this work, a high-performance bifunctional red fluorescent probe named Probe 2 was rationally designed and constructed by screening different electron-donating groups. The probe exhibits good chromogenic and turn-on fluorescence responses to SO 2 and viscosity, and can target mitochondria well. The probe has been successfully applied for accurate and rapid determination of sulfite content in water and food samples with good recovery (93.2–98.1 % and 94.2–100.4 %). Furthermore, Probe 2 enables real-time tracking and dual-color fluorescence imaging of SO 2 and viscosity in cells and zebrafish, especially its high sensitivity to changes in these two parameters during mitochondrial dysfunction and apoptosis.
