Abstract:In recent years, photothermal therapy (PTT) particularly nanomaterial-based PTT is a promising therapeutic modality and technique for cancer tumor ablation. In addition to killing tumor cells directly through heat, PTT also can induce immunogenic cell death (ICD) to activate the whole-body anti-tumor immune response, including the redistribution and activation of immune effector cells, the expression and secretion of cytokines and the transformation of memory T lymphocytes. When used in combination with immunotherapy, the efficacy of nanomaterial-based PTT can be improved. This article summarized the mechanism of nanomaterial-based PTT against cancer and how nanomaterial-based PTT impacts the tumor microenvironment and induces an immune response. Moreover, we reviewed recent advances of nanomaterial-based photothermal immunotherapy and discussed challenges and future outlook.

Recent strategies for nano-based PTT combined with immunotherapy: from a biomaterial point of view

Abstract:Cancer has been a great threat to humans for decades. Due to the limitations of monotherapy, combinational therapies such as photothermal therapy (PTT) and immunotherapy have gained increasing attention with expectation to overcome the shortfalls of each other and obtain satisfactory therapeutic outcomes. PTT can inhibit primary tumors by thermal ablation but usually fails to achieve complete eradication and cannot prevent metastasis and recurrence. Meanwhile, the efficacy of immunotherapy is usually attenuated by the weak immunogenicity of tumor and the immunosuppressive tumor microenvironment (ITM). Therefore, many recent studies have attempted to synergize PTT with immunotherapy in order to enhance the therapeutic efficacy. In this review, we aim to summarize the cutting-edge strategies in combining nano-based PTT with immunotherapy for cancer treatment. Herein, the combination strategies were mainly classified into four categories, including 1) nano-based PTT combined with antigens to induce host immune responses; 2) nano-based PTT in combination with immune adjuvants acting as in situ vaccines; 3) nano-based PTT synergized with immune checkpoint blockade or other regulators to relieve the ITM; 4) nano-based PTT combined with CAR-T therapy or cytokine therapy for tumor treatment. The characteristics of various photothermal agents and nanoplatforms as well as the immunological mechanisms for the synergism were also introduced in detail. Finally, we discussed the existing challenges and future prospects in combined PTT and immunotherapy.

Nanomedicine potentiates mild photothermal therapy for tumor ablation

Abstract:The booming photothermal therapy (PTT) has achieved great progress in non-invasive oncotherapy, and paves a novel way for clinical oncotherapy. Of note, mild temperature PTT (mPTT) of 42-45 °C could avoid treatment bottleneck of the traditional PTT, including nonspecific injury to normal tissues, vasculature and host antitumor immunity. However, cancer cells can resist mPTT via heat shock response and autophagy, thus leading to insufficient mPTT monotherapy to ablate tumor. To overcome the deficient antitumor efficacy caused by thermo-resistance of cancer cells and mono mPTT, synergistic therapies towards cancer cells have been conducted with mPTT. This review summarizes the recent advances in nanomedicine-potentiated mPTT for cancer treatment, including strategies for enhanced single-mode mPTT and mPTT plus synergistic therapies. Moreover, challenges and prospects for clinical translation of nanomedicine-potentiated mPTT are discussed.

Abstract:Nanoparticle-mediated photothermal therapy (PTT) has shown promising capability for tumor therapy through the high local temperature at the tumor site generated by a photothermal agent (PTA) under visible or near-infrared (NIR) irradiation. Improving the accumulation of PTA at the tumor site is crucial to achieving effective photothermal treatment. Here, we developed temperature-activatable engineered neutrophils (Ne) by combining indocyanine green (ICG)-loaded magnetic silica NIR-sensitive nanoparticles (NSNP), which provide the potential for dual-targeted photothermal therapy. The combined effect of neutrophil targeting and magnetic targeting increased the accumulation of PTA at the tumor site. According to magnetic resonance imaging (MRI), the retention of intravenous injected NSNP-incorporated neutrophils within the tumor site was markedly augmented as compared to free NSNP. Furthermore, when irradiated by NIR, NSNP could cause a high local temperature at the tumor site and the thermal stimulation of neutrophils. The heat can kill tumor cells directly, and also lead to the death of neutrophils, upon which active substances with tumor-killing efficacy will be released to kill residual tumor cells and thus reduce tumor recurrence. Thereby, our therapy achieved the elimination of malignancy in the mouse model of the pancreatic tumor without recurrence. Given that all materials used in this system have been approved for use in humans, the transition of this treatment method to clinical application is plausible.

Abstract:Although photothermal therapy (PTT) can act on tumors alone, resulting in tumor shrinkage or even eradication, this effect is short‐lived, and the tumor may regrow. Thus, the most helpful contribution of PTT to cancer therapy is to employ it in conjunction with other tumor therapies, such as chemotherapy, immunotherapy, photodynamic therapy, gene therapy, radiotherapy therapy, and multiple combination therapy to boost treatment efficacy. This paper covers the benefits and drawbacks of combining photothermal therapy with various therapeutic approaches by the nanodrug delivery system and the possibilities and obstacles of clinical combination therapy. Abstract:Nanomaterial-synergized photodynamic therapy (PDT) and photothermal therapy (PTT), as efficient and non-invasive treatment modalities, have shown significant advantages in fighting different types of cancer. However, neither PTT nor PDT can completely eradicate tumors due to distant metastasis and recurrence of tumors. Recently, photo-immunotherapy have attracted great attention as phototherapy has been reported to participate in immunotherapy by triggering immunogenic cell death (ICD), resulting in the secretion of tumor specific antigen (TSAs) and damage-associated molecular patterns (DAMPs). In particular, emerging interests are biased towards manipulating nanomaterials to form unique drug delivery systems, which are necessary for the combination of phototherapy and immunotherapy to eliminate metastatic tumor cells by promoting the maturation of dendritic cells (DCs) and the infiltration of cytotoxic T lymphocytes (CTLs). This review elaborates on the latest strategies on engineering nanomaterials to enhance the anti-cancer efficiency of synergistic photo-immunotherapy, with emphasis on the activation of anti-tumor immune response, the reversal of tumor immunosuppressive microenvironment (TIME), the regulation of the interaction between immunosuppressive cells and tumor cells, the infiltration of immune cells and improved efficiency of photo-immunotherapy-induced ICD. Current challenges and future opportunities in engineering nanomaterials to modulate synergistic photo-immunotherapy are also discussed.
Research Progress of Nanomedicine-Based Mild Photothermal Therapy in Tumor

Abstract:With the booming development of nanomedicine, mild photothermal therapy (mPTT, 42-45°C) has exhibited promising potential in tumor therapy. Compared with traditional PTT (>50°C), mPTT has less side effects and better biological effects conducive to tumor treatment, such as loosening the dense structure in tumor tissues, enhancing blood perfusion, and improving the immunosuppressive microenvironment. However, such a relatively low temperature cannot allow mPTT to completely eradicate tumors, and therefore, substantial efforts have been conducted to optimize the application of mPTT in tumor therapy. This review extensively summarizes the latest advances of mPTT, including two sections: (1) taking mPTT as a leading role to maximize its effect by blocking the cell defense mechanisms, and (2) regarding mPTT as a supporting role to assist other therapies to achieve synergistic antitumor curative effect. Meanwhile, the special characteristics and imaging capabilities of nanoplatforms applied in various therapies are discussed. At last, this paper puts forward the bottlenecks and challenges in the current research path of mPTT, and possible solutions and research directions in future are proposed correspondingly.

Abstract:Recently, photothermal therapy (PTT) has demonstrated excellent results in the treatment of preclinical cancer models and thus attracted intensive interests from the research fields of material science, biochemistry, oncology, and pharmacy. Also, its combination with other cancer therapies shows great promise for clinical application: PTT is able to make up for the drawbacks of others and improve the prognosis through various synergistic effects. In these combination therapies, design of nanoscale multifunctional therapeutics is key to achieve enhanced or even synergistic therapeutic effect. Herein, the recent progress is summarized in the development of strategies involving combination of PTT with other cancer therapies, conventional or emerging, with increased efficacy. The highlight lies in integrating various functional units to form multifunctional nanomaterials that play the role of photothermal agents as well as other therapeutics in a combination therapy and the design of an effective targeted delivery system for them. Smart combination of therapies will undoubtedly improve the therapeutic efficacy and make a contribution to the advancement in fighting cancer.

Abstract:Immuno‐oncotherapy has shown great promise for the cure of late‐stage and metastatic cancer. Great efforts have tried to improve the overall response rate (ORR) and to reduce the immune‐related adverse events (irAEs). Antigen presentation, T cell activation and killing are interlocking and distinct steps to initiate effective anti‐tumor immune responses. Aiming to overcome the tumor immune evasion whose mechanisms include limited release of neoantigen, suppressed infiltration of antigen‐presenting cells (APCs) and T cells, and the expression of immune checkpoints (ICPs), combinational therapeutic strategies have shown great potential by activating the anti‐tumor immune responses together with deactivating immunosuppressive conditions simultaneously. In this direction, photothermal therapy (PTT) has attracted attention due to the efficient ablation of tumor cells, of which the released immunogenic tumor debris can activate host immune responses. The combination of immunoadjuvants and/or ICP inhibitors can boost the anti‐tumor immune responses, realizing PTT‐synergized immuno‐oncotherapy. In this regard, numerous multifunctional nanomaterials have been designed with integration of photothermal and immuno‐oncotherapeutic agents into one package via well‐designed surface modification and functionalization. Abstract:Cancer nanomedicines have provided promising treatment strategies for the regression of several types of cancer, although there still exists a need for significant advances to improve their therapeutic outcomes and reduce side effects. In this context, photothermal therapy (PTT) that uses near-infrared (NIR) photoirradiation of photothermal agents to generate heat for localized thermal damages has been established as a safe therapeutic modality. In addition to direct ablation of tumors, the heat generated during PTT process can achieve on-demand release of other therapeutic compounds, regulation of gene transcription and enzyme activity and enhancement of chemical reactions in tumor tissues, thereby resulting in photothermal synergistic cancer therapy with obviously improved benefits. In this review, we summarize the recent developments in NIR photoactivated nanomedicines for photothermal synergistic cancer therapy. We introduce the designing principles and the working mechanisms of nanoparticles upon NIR photoirradiation and their applications in photothermal synergistic chemotherapy, enzyme therapy, gene therapy, photodynamic therapy, chemodynamic therapy, thermodynamic therapy, immunotherapy and their multimodal therapies for cancer. Moreover, we discuss the existing challenges and further perspectives in this field.

Abstract:Immunotherapy has revolutionized oncology remarkably and gained great improvements in cancer therapy. However, tumor immunotherapy still encounters serious challenges, especially certain tumors barely respond to immunotherapy. The lack of immunogenicity and subsequent insufficient antitumor immune activation is a pivotal reason. Here, a general introduction and the strengthening strategies of immunogenicity of a tumor for enhanced immunotherapy are reviewed. Specifically, nanotechnology nowadays is playing important roles in increasing the antitumor efficacy of various treatments, including immunotherapy. This review highlights how nanomedicines integrating one or more anticancer therapeutic methods (e.g., cancer vaccines, chemotherapy, phototherapy, and radiotherapy) to increase the tumor immunogenicity for rousing T cell related immune responses and achieving inspiring antitumor efficacy. Given the sophisticated immune evasion mechanisms, rational designed nanodrugs with combinational formulations are summarized to improve therapeutic efficacy in synergistic ways. Nanoplatforms taking advantage of the distinct features of tumor tissue or tumor cell with stimuli-responsiveness and targeting functions are introduced to accelerate tumor accumulation of drugs successfully and greatly promote therapeutic efficacy with low-dose administration and programmed drug release. Finally, the related challenges and personal perspectives of nanomedicines for tumor immunotherapy are concluded.

Abstract:Benefiting from the rapid development of nanotechnology, photodynamic therapy (PDT) is arising as a novel non-invasive clinical treatment for specific cancers, which exerts direct efficacy in destroying primary tumors by generating excessive cytotoxic reactive oxygen species (ROS). Notably, PDT-induced cell death is related to T cell-mediated antitumor immune responses through induction of immunogenic cell death (ICD). However, ICD elicited via PDT is not strong enough and is limited by immunosuppressive tumor micro environment (ITM). Therefore, it is necessary to improve PDT efficacy through enhancing ICD with the combination of synergistic tumor therapies. Herein, the recent progress of nanomaterials-based PDT combined with chemotherapy, photothermal therapy, radiotherapy, and immunotherapy, employing ICD-boosted treatments is reviewed. An outlook about the future application in clinics of nanomaterials-based PDT strategies is also mentioned.

Abstract:Cancer immunotherapy has made tremendous clinical progress in advanced-stage malignancies. However, patients with various tumors exhibit a low response rate to immunotherapy because of a powerful immunosuppressive tumor microenvironment (TME) and insufficient immunogenicity of tumors. Photodynamic therapy (PDT) can not only directly kill tumor cells, but also elicit immunogenic cell death (ICD), providing antitumor immunity. Unfortunately, limitations from the inherent nature and complex TME significantly reduce the efficiency of PDT. Recently, smart nanomedicine-based strategies could subtly modulate the pharmacokinetics of therapeutic compounds and the TME to optimize both PDT and immunotherapy, resulting in an improved antitumor effect. Here, the emerging nanomedicines for PDT-driven cancer immunotherapy are reviewed, including hypoxia-reversed nanomedicines, nanosized metal-organic frameworks, and subcellular targeted nanoparticles (NPs). Moreover, we highlight the synergistic nanotherapeutics used to amplify immune responses combined with immunotherapy against tumors. Lastly, the challenges and future expectations in the field of PDT-driven cancer immunotherapy are discussed.

Abstract:Phototherapy, referring to photodynamic/photothermal therapy, has been extensively validated to promote enhanced immunotherapeutic effects by stimulating tumor cell immunogenic death. Photoimmunotherapy has been persistently investigated to establish potent antitumor effects against primary and distant tumors, synchronously eliciting powerful immunological memory effects, thus ultimately preventing and eradicating rechallenged tumors. Phototherapeutic nanoagents play essential roles in ensuring the sufficient efficacy of photoimmunotherapy, which provides a flexible platform to integrate multifunctional types of phototherapy into a single platform. In particular, tailored nanoparticles are available to amplify tumor immunogenicity and to modulate the immunosuppressive tumor microenvironment simultaneously and spatiotemporally for the treatment of cancers. In this review, we summarized commonly adopted strategies to achieve enhanced cancer immunotherapies induced by conventionally designed phototherapeutic nanoagents. We also analyzed the immunotherapeutic performance and characteristics of phototherapy in detail. The manuscript implies our thoughts on the following aspects: directional design of photosensitizing agents, functional construction of nanomedicines, rational modulation of immunotherapy, and augmented phototherapeutic effects.
New Nano Photothermal Agent Integrates Immunotherapy Against Tumor by Modulating Immunosuppressive Microenvironment

Abstract:Photothermal therapy (PTT) was reported to induce synergistic immunogenic cell death (ICD) which may transfer tumor cells into therapeutic vaccines. However, PTT often fails to induce sufficient immune response to prevent tumor recurrence, partially due to the immunosuppressive microenvironment in tumors. Therefore, remodeling tumor microenvironment is of great importance to enhance the therapy efficiency of PTT. Herein, we fabricated a photosensitizer by assembling quercetin and Ferrum ion (QFN). The released quercetin from QFN inhibits the phosphorylation of JAK2 and STAT3 to reduce programmed death ligand 1 (PD-L1) in tumor cells, and reshape extracellular matrix (ECM) by down-regulating Î±-SMA + fibroblast in tumors. Moreover, QFN could capture tumor antigen and transfer it to the tumor-draining lymph nodes after PTT, further enhancing the antigen-presenting cell activation. As a result, the PTT-induced immunogenic cell death of tumor cells and its consequent immune responses is significantly enhanced, and QFN-based PTT eliminated melanoma and induced long-term immune memory to prevent tumor metastasis and recurrence. Recent advances in nanoparticles-based photothermal therapy synergizing with immune checkpoint blockade therapy

Abstract:Several breakthroughs have been achieved in anti-tumor immune checkpoint blockade (ICB) therapy in the past decade. Despite the considerable therapeutic effects, ICB is still limited by the low benefit rates and severe systemic toxicity. Pre-clinical studies have shown that photothermal therapy (PTT) mediated by nanoparticles (NPs) can counterbalance the drawbacks of ICB therapy and produce synergistic anti-tumor effects. NPs offer several advantages such as increased tumor-targeting ability, high drug-loading capacity and satisfying biocompatibility. Therefore, combining NPs-based PTT and ICB therapy can achieve a better outcome against tumors, and even prevent metastatic and recurrence by inducing immune memory. Here, we have discussed the current progress in ICB therapy and NPs-based PTT, along with the pre-clinical trials on PTT/ICB combinational therapy. The synthesis and mechanisms of different photothermal NPs have also been summarized. Moreover, the current challenges, deficiencies and future improvement of this novel treatment modality have been analyzed.

Abstract:Abstract Treatment of malignant tumors has always been a worldwide challenge. The complexity, diversity, and heterogeneity of tumors have prompted the treatment strategies to gradually shift from monotherapy to combination therapy that can synergize multiple treatment modalities. With unique physical and chemical properties, nanomaterials have been widely used in different cancer therapies, including chemotherapy, immunotherapy, and phototherapy. Especially, the variability and integrity of nanomaterials make them ideal media for synergistic collaboration strategies. Here, we provide our perspective on the synergistic strategy of nanoplatform-mediated phototherapies and related combination therapies, specifically photochemotherapy, photoradiotherapy, and photoimmunotherapy. Besides, we provide an analysis and outlook on the current challenges faced by synergistic nanophototherapy.

Abstract:Conventional chemotherapy for cancer treatment is usually compromised by shortcomings such as insufficient therapeutic outcome and undesired side effects. The past decade has witnessed the rapid development of combination therapy by integrating chemotherapy with hyperthermia for enhanced therapeutic efficacy. Near-infrared (NIR) light-mediated photothermal therapy, which has advantages such as great capacity of heat ablation and minimally invasive manner, has emerged as a powerful approach for cancer treatment. A variety of nanomaterials absorbing NIR light to generate heat have been developed to simultaneously act as carriers for chemotherapeutic drugs, contributing as heat trigger for drug release and/or inducing hyperthermia for synergistic effects. This review aims to summarize the recent development of advanced nanomaterials in chemo-photothermal combination therapy, including metal-, carbon-based nanomaterials and particularly organic nanomaterials. The potential challenges and perspectives for the future development of nanomaterials-based chemo-photothermal therapy were also discussed.

Abstract:In recent decades, cancer has posed a challenging obstacle that humans strive to overcome. While phototherapy and immunotherapy are two emerging therapies compared to traditional methods, they each have their advantages and limitations. These limitations include easy metastasis and recurrence, low response rates, and strong side effects. To address these issues, researchers have increasingly focused on combining these two therapies by utilizing a nano-drug delivery system due to its superior targeting effect and high drug loading rate, yielding remarkable results. The combination therapy demonstrates enhanced response efficiency and effectiveness, leading to a preparation that is highly targeted, responsive, and with low recurrence rates. This paper reviews several main mechanisms of anti-tumor effects observed in combination therapy based on the nano-drug delivery system over the last five years. Furthermore, the challenges and future prospects of this combination therapy are also discussed.
