Abstract:The purpose of this study was to design novel drug nanocrystals (NCs) stabilized by glycyrrhizic acid (GL) for achieving liver targeted drug delivery due to the presence of GL receptor in the hepatocytes. Quercetin (QT) exhibits good pharmacological activities for the treatment of liver diseases, including liver steatosis, fatty hepatitis, liver fibrosis, and liver cancer. It was selected as a model drug owing to its poor water solubility. QT NCs stabilized by GL (QT-NCs/GL) were fabricated by wet media milling technique and systemically evaluated. QT-NCs stabilized by poloxamer 188 (QT-NCs/P188) were prepared as a reference for comparison of in vitro and in vivo performance with QT-NCs/GL. QT-NCs/GL and QT-NCs/P188 with similar particle size around 130 nm were successfully fabricated by wet media milling technique. Both of QT-NCs/GL and QT-NCs/P188 showed irregular particles and short rods under SEM. XRPD revealed that QT-NCs/GL and QT-NCs/P188 remained in crystalline state with reduced crystallinity. QT-NCs/GL and QT-NCs/P188 exhibited significant solubility increase and drug release improvement of QT as compared to raw QT. No significant difference for the plasma concentration-time curves and pharmacokinetic parameters of QT were found following intravenous administration of QT-NCs/GL and QT-NCs/P188. However, a significantly higher liver distribution of QT following intravenous administration of QT-NCs/GL was observed in comparison to QT-NCs/P188, indicating QT-NCs stabilized by GL could achieve liver targeted delivery of QT. It could be concluded that GL used as stabilizer of QT NCs have a great potential for liver targeted drug delivery.

Abstract:The aim of this study was to develop a new drug nanocrystals self-stabilized Pickering emulsion (NSSPE) for improving oral bioavailability of quercetin (QT). Quercetin nanocrystal (QT–NC) was fabricated by high pressure homogenization method, and QT–NSSPE was then prepared by ultrasound method with QT–NC as solid particle stabilizer and optimized by Box-Behnken design. The optimized QT–NSSPE was characterized by fluorescence microscope (FM), scanning electron micrograph (SEM), X-ray diffraction (XRD), and differential scanning calorimetry (DSC). The stability, in vitro release, and in vivo oral bioavailability of QT–NSSPE were also investigated. Results showed that the droplets of QT–NSSPE with the size of 10.29 ± 0.44 μm exhibited a core-shell structure consisting of a core of oil and a shell of QT–NC. QT–NSSPE has shown a great stability in droplets shape, size, creaming index, zeta potential, and QT content during 30 days storage at 4, 25, and 40 °C. In vitro release studies showed that QT–NSSPE performed a better dissolution behavior (65.88% within 24 h) as compared to QT–NC (50.71%) and QT coarse powder (20.15%). After oral administration, the AUC0–t of QT–NSSPE was increased by 2.76-times and 1.38 times compared with QT coarse powder and QT–NC. It could be concluded that NSSPE is a promising oral delivery system for improving the oral bioavailability of QT.

Abstract:Quercetin (QCT) has antioxidant, anti-inflammatory, anti-tumor and other important pharmacological activities, but the poor water solubility limits its application. In this work, the amphiphilic dextran (ADEX) was prepared by grafting L-cysteine and octadecylamine onto carboxymethyl dextran with the grafting rate of 21.29 % and 19.35 %. Then, the QCT-loaded nanomicelles (QNMs) were prepared by using ADEX as wall material and the QCT as core material via ultrasonic self-assembly method. The particle size and zeta potential of QNMs were 372 nm and 31.4 mV. Under simulated gastric and simulated intestinal fluids, the cumulative release QNMs were 37.54 % and 52.13 % within 180 min, and the QNMs showed better stability in simulated gastric fluid. The QNMs showed significantly better PTIO, OH and O<sub>2</sub><sup>-</sup> scavenging activities than QCT. In addition, QNMs could effectively down-regulate the expression of pro-inflammatory cytokines and promoted the expression of anti-inflammatory cytokine. The cellular uptake results proved that the QNMs were more easily absorbed by cells than free QCT, indicating that the nano-encapsulation procedure effectively improved the uptake efficiency of QCT by cells.

Abstract:Quercetin, an essential flavonoid compound, exhibits diverse biological activities including anti-inflammatory and antioxidant effects. Nevertheless, due to its inadequate solubility in water and vulnerability to degradation, pure quercetin is constrainedly utilized in pharmaceutical formulations and functional foods. Considering the existing scarcity of nanoparticles consisted of zein and hydrophobic biopolymers, this study developed a quercetin-loaded nanoencapsulation based on zein, shellac, and chitosan (QZSC). When the mass ratio of zein to chitosan was 4:1, the encapsulation efficiency of QZSC reached 74.95%. The ability of QZSC for scavenging DPPH radicals and ABTS radicals increased from 59.2% to 75.4% and from 47.0% to 70.2%, respectively, compared to Quercetin. For QZSC, the maximum release amount of quercetin reached 59.62% in simulated gastric fluid and 81.64% in simulated intestinal fluid, achieving controlled and regulated release in vitro. In summary, this study offers a highly promising encapsulation strategy for hydrophobic bioactive substances that are prone to instability.
Quercetin nanocrystals for bioavailability enhancement: impact of different functional stabilizers on in vitro/in vivo drug performances

Abstract:The purpose of this study was to investigate the impact of different functional stabilizers on in vitro/in vivo drug performances after oral administration of drug nanocrystals. Quercetin nanocrystals (QT-NCs) respectively stabilized by five types of functional stabilizers, including hydroxypropyl methyl cellulose E15 (HPMC E15), poloxamer 407 (P407), poloxamer 188 (P188), D-α-tocopherol polyethylene glycol succinate (TPGS), and glycyrrhizin acid (GL), were fabricated by wet media milling technique. The particle size, morphology, physical state, drug solubility, drug dissolution in vitro , and orally pharmacokinetic behaviors of all QT-NCs were investigated. All QT-NCs with similar particle size about 200 nm were obtained by controlling milling speed and milling time. No significant differences in particles shape and crystalline nature were found for QT-NCs stabilized by different functional stabilizers. But the solubility and dissolution of QT-NCs were significantly influenced by the different functional stabilizers. The AUC 0∼ t of all QT-NCs after oral administration was in the following order: QT-NCs/P188 ≈ QT-NCs/HPMC E15 > QT-NCs/GL > QT-NCs/P407 ≈ QT-NCs/TPGS, and the C max showed an order of QT-NCs/P407 > QT-NCs/P188 ≈ QT-NCs/GL > QT-NCs/HPMC E15 > QT-NCs/TPGS. Both of QT-NCs/P407 and QT-NCs/TPGS exhibited faster oral absorption with T max at 0.5 h and 0.83 h, respectively, while the other three QT-NCs (QT-NCs/P188, QT-NCs/GL and QT-NCs/HPMC E15) showed a relatively slow absorption with same T max at 5.33 h. The longest MRT 0∼ t (11.72 h) and t 1/2z (32.22 h) were observed for QT-NCs/HPMC E15. These results suggested that the different functional stabilizers could significantly influence on drug solubility, drug dissolution in vitro and orally pharmacokinetic behavior of QT-NCs, and it is possible to alter the drug dissolution in vitro , oral absorption and drug retention in vivo by changing the type of functional stabilizers in NCs preparation.

Controlled preparation of curcumin nanocrystals by detachable stainless steel microfluidic chip

Abstract:Microfluidic technology has not been extensively utilized in nanocrystals manufacture, although it has been used in the production of liposomes and LNPs. This is mainly due to concerns including blockage of narrow pipes and corrosion of organic solvents on chips. In this study, a detachable stainless steel microfluidic chip with split-and-recombine (SAR) structure was engraved and used to prepare curcumin nanocrystal suspensions by a microfluidic-antisolvent precipitation method. A simulation study of the mixing activities of three chip structures was conducted by COMSOL Multiphysics software. Then the curcumin nanocrystals preparation was optimized by Box-Behnken design to screen different stabilizers and solvents. Two curcumin nanocrystals formulations with an average particle size of 59.29 nm and 168.40 nm were obtained with PDIs of 0.131 and 0.058, respectively. Compared to curcumin powder, the formulation showed an increase in dissolution rate in 0.1 M HCL while pharmacokinetic study indicated that Cmax was increased by 4.47 and 3.14 times and AUC0-∞ were 4.26 and 3.14 times greater. No clogging or deformation of the chip was observed after long usage. The results demonstrate that the stainless steel microfluidic chips with SAR structure have excellent robustness and controllability. It has the potential to be applied in GMP manufacturing of nanocrystals.
Co-crystallization of quercetin and isonicotinamide using solvent evaporation method

Abstract:Purpose: To obtain quercetin-isonicotinamide co-crystal (CQINA) with improved physicochemical and in-vitro dissolution characteristics. Methods: Co-crystallization of quercetin (Q) and isonicotinamide (INA) in molar ratio of 1:1 was performed using solvent evaporation method with the addition of 50 mL of ethanol (99.9%, v/v). The resultant solution was thoroughly mixed and stirred at room temperature for 48 h to slowly evaporate the solvent until CQINA was obtained. The co-crystal phase was characterized using differential scanning calorimetry (DSC), powder x-ray diffractometry (PXRD), scanning electron microscopy (SEM), and fourier transform infrared (FTIR) spectroscopy. In-vitro dissolution was performed by USP method II in 900 mL citrate buffer (pH 5.0 ± 0.05), with stirring at 100 rpm and at 37 ± 0.5 °C. Results: Computational approach predicted the formation of hydrogen bonds between Q and coformers used, and the interaction involved minimum energy. In CQINA thermogram, a new endothermic peak was formed with a melting point of 255.26 °C, while Q (314.85 °C) and INA (156.62 °C). Images from DSC, PXRD, SEM and FTIR showed that the crystal habits and morphologies of the CQINA differed from those of the original components. There was an improvement in the dissolution profile of CQINA, when compared with those of the original components. Conclusion: Q and INA subjected to solvent evaporation result in the formation of a CQINA with different crystal habit, which possess physicochemical characteristics different from those of its constituents. Modification of Q crystals in CQINA increases its in vitro dissolution, making it a potential pharmaceutical agent.

Abstract:The in vivo fate of nanocrystals is a controversial topic, i.e. dissolving versus integral absorption through the intestinal membrane. This is due to the lack of functional strategies to identify integral nanocrystals. In this study, the in vivo fate of quercetin hybrid nanocrystals (QT-HNCs) via the oral route is explored by physically embedding an environment-responsive probe in the crystal lattices of quercetin. The specific property of the probe is the water-initiated aggregation-caused quenching (ACQ) ability, by which integral QT-HNCs can be self-discriminated. Instead of dissolving instantly, QT-HNCs can be retained in the gastrointestinal tract for 12-16 h, and can then be absorbed and distributed into various organs with the liver as the primary terminal. The ileum provides better absorption than the jejunum. Cellular studies prove that both trans-epithelial and M cell-mediated routes are involved in the absorption of integral QT-HNCs, which may be impeded by the mucous layer. Moreover, the particle size affects the in vivo behavior and the ex vivo cellular interaction of QT-HNCs, with moderate size, such as 550 nm, being preferred. The results not only validate the idea of using ACQ fluorophores for bioimaging of integral nanocrystals but also support the intestinal absorption of nanocrystals.
Simple preparation and greatly improved oral bioavailability: The supersaturated drug delivery system of quercetin based on PVP K30

Abstract:Quercetin, as a representative flavonoid, is widely present in daily diet and has been developed as a dietary supplement due to its beneficial physiological activities. However, the application of quercetin is limited due to its poor water solubility and extensive metabolism. So far, the nano-drug delivery systems designed to improve its bioavailability generally have the shortcomings of low drug loading content and difficulty in industrial production. In order to tackle these problems, quercetin supersaturated drug delivery system (QSDDS) was successfully prepared using solvent method, for which PVP K30 was employed as a crystallization and precipitation inhibitor to maintain the supersaturated state of quercetin in aqueous system. The obtained QSDDS, with a relative high drug loading content of 13%, could quickly disperse in water and form colloidal system with the mean particle size of about 200 nm, meanwhile induce the generation of supersaturated quercetin solution more than 12 h. In vivo pharmacokinetic study proved that QSDDS achieved a high absolute bioavailability of 36.05%, 10 times as that of physical quercetin suspension, which was dose-dependent with higher bioavailability at higher dose. Considering the simple preparation method, QSDDS provided a feasible strategy and a simple way to improve oral absorption of insoluble flavonoids.

Abstract:Objective: Quercetin, a wonder flavanoid despite numerous pharmacological actions, has limited clinical applications due to solubility and permeability issues and additionally having shorter biological half-life. The goal of the current work was to design Quercetin lipid-based spherical crystals, to improve its oral bioavailability and sustain its in vivo plasma levels. Methods: An anti-solvent precipitation method was employed to prepare quercetin spherical agglomerates using ethanol and distilled water as good and bad solvents, respectively. As bridging liquid chloroform, dichloromethane, hexane and gelucire 43/01, compritol 888 as lipid carrier were screened. The drug-to-lipid polymer proportion and stirring speed effect were optimized by 3-level, 2-factor, experimental design. Numerical optimization function was employed to identify the optimum level of independent variables. Spectroscopic, micromeritic, surface morphology, size distribution, saturated solubility, in vitro dissolution, in vivo pharmackokinetic and stability studies were performed. Results: Surface morphology studies indicated the agglomeration of quercetin needle-like fragments into a spherical shape, which further showed smooth surfaces due to entrapment of QC in lipid carrier. The spherical agglomerates of quercetin showed a four-fold improvement in aqueous solubility compared to pure drug and showed 92.13% release in 8 h. The optimised formulation showed a 3.69-fold enhancement in relative bioavailability in contrast to the marketed preparation in an in vivo pharmacokinetic analysis in male Wistar rats. Conclusion: The obtained lipid-based spherical crystals of quercetin with enhanced bioavailability could be effectively used for its various potential pharmacological applications. The designed system can also be utilized to deliver other phytochemicals with poor bioavailability due to limited solubility and permeability.
Combination of co-crystal and nanocrystal techniques to improve the solubility and dissolution rate of poorly soluble drugs

Abstract:Purpose Solubility and dissolution rate are essential for the oral absorption and bioavailability of poorly soluble drugs. The aim of this study was to prepare nano-co-crystals by combination of nanocrystal and co-crystal technologies, and investigate its effect, in situ , on increased kinetic solubility and dissolution rate. Methods Co-crystals of itraconazole-fumaric acid, itraconazole-succinic acid, indomethacin-saccharin and indomethacin-nicotinamide were prepared and nano-sized by wet milling. The particle size and solid state of the co-crystals were characterized by optical microscope, LD, PCS, DSC and XRPD before and after milling. Results 300-450 nm sized nano-co-crystals with a stable physical solid state were successfully prepared. Nano-co-crystals exhibited a lower crystallinity reduction than nanocrystals after wet milling. The particle size effect on the kinetic solubility of co-crystals was analysed for macro-, micro- and nano-co-crystals with in situ kinetic solubility studies. The maximum kinetic solubility of nano-co-crystals increased with excess conditions until a plateau. The highest increase was obtained with itraconazole-succinic acid nano-co-crystals with a kinetic solubility of 263.5 ± 3.9 μg/mL which was 51.5 and 6.6 times higher than the solubility of raw itraconazole and itraconazole-succinic acid co-crystal. Conclusions The synergistic effect of nanocrystals and co-crystals with regard to increased kinetic solubility and dissolution rate was proven. The combination of the advantages of nanocrystals and co-crystals is a promising formulation strategy to increase both the solubility and dissolution rate of poorly soluble drugs.

Antioxidant Effects of Quercetin Nanocrystals in Nanosuspension against Hydrogen Peroxide-Induced Oxidative Stress in a Zebrafish Model

Abstract:Quercetin, a flavonoid compound rich in hydroxyl groups, possesses antioxidant properties, whereas its poor water solubility limits its bioavailability. In pursuit of addressing the water solubility of quercetin and comprehending the impact of nanocrystal particle size on antioxidant efficacy, we prepared three different-sized quercetin nanocrystals, namely small (50 nm), medium (140 nm), and large (360 nm), using a nanosuspension method in this study. Within the in vitro setting, assessments employing solubility and radical scavenging assays revealed that quercetin nanocrystals displayed superior solubility (26, 21, and 13 fold corresponding to small, medium, and large particle sizes) and antioxidant performance compared to the coarse quercetin. Furthermore, quercetin nanocrystals of three particle sizes all demonstrated significant protection effects on the survival rate of H2O2-treated zebrafish at 72 h (77.78%, 73.33%, and 66.67% for small, medium, and large particle sizes, respectively), while the coarse quercetin group exhibited a low survival rate (53.3%) similar to the H2O2-treated group (47.8%). Moreover, all quercetin nanocrystals exhibited potent antioxidant capacity on both the antioxidants and enzymatic antioxidant system in H2O2-treated zebrafish to restore zebrafish to a normal state under oxidative stress. For instance, the levels of reactive oxygen species were reduced to 101.10%, 108.83%, and 109.77% of the normal levels for small, medium, and large particle-sized quercetin nanocrystals, respectively. In conclusion, quercetin nanocrystals demonstrated enhanced solubility, robust antioxidant capacity, and protective effects in zebrafish compared to coarse quercetin.

Abstract:Resveratrol (Res) has a variety of pharmacological activities, but it has not been widely used in clinical practice due to its poor water solubility and low bioavailability. Therefore, improving the solubility and utilization of Res is a key factor in increasing its clinical application. Microneedles (MNs) are a new type of transdermal drug delivery system, which has been widely noticed for its advantages of avoiding hepatic first-pass effect, reducing gastrointestinal side effects, and improving patient compliance. In recent years, the integration of nanomedicine with MNs has also become a novel therapy for drug delivery. In this study, a novel transdermal drug delivery system (Res NC MNs) was obtained by altering the crystalline form of resveratrol by media milling method, preparing resveratrol nanocrystals (Res NC), and loading Res NC into soluble microneedles prepared from sodium hyaluronate. Methods Res NC was prepared by media milling method, and the experimental conditions, such as the type of stabilizer, the ratio of stabilizer to the drug, the number of milling beads, and the milling time, were screened to determine the optimal preparation process, using the particle size and dispersion of nanocrystals as indicators sodium hyaluronate. Dissolvable microneedles were prepared by vacuum casting method and loaded with Res NC, and then the morphology, skin puncture performance, and skin dissolution performance of Res NC MNs were examined. Results From the in vitro release experiments, it can be seen that the resveratrol nanocrystals prepared using the optimal process had a small particle size of about 110 nm, good stability, and good release characteristics. The Res NC MNs had a good morphology, a full and uniform tip, and good skin-piercing properties and dermal solubilization properties. The Res NC MNs prepared in this study improved the drawbacks of poor water solubility and low bioavailability of resveratrol APIs, and combined with MNs, overcame the inherent defects of the traditional drug delivery methods such as oral and injection, which provided a novel idea for the integration of nanomedicine and transdermal drug delivery system.

Tea saponin-Zein binary complex as a quercetin delivery vehicle: preparation, characterization, and functional evaluation

Abstract:In this study, in order to solve the application problems of poor water solubility and low bioavailability of quercetin, we prepared a nano-delivery system with core-shell structure by anti-solvent method, including a hydrophilic shell composed of tea saponin and a hydrophobic core composed of Zein, which was used to improve the delivery efficiency and biological activity of quercetin. Through the optimal experiments, the loading rate and encapsulation rate of nanoparticles reached 89.41 % and 7.94 % respectively. And the water solubility of quercetin is improved by 30.16 times. At the same time, the quercetin acted with Zein through non-covalent interaction and destroyed its spatial network through structural characterization, while tea saponin covered the surface of Zein through electrostatic interaction, making it change into amorphous state. In addition, the addition of tea saponin makes the nanoparticles remain stable under the changes of external environment. During simulating gastrointestinal digestion procedure, ZQTNPs has higher release rate and bioavailability than free quercetin. Importantly, ZQTNPs can overcome the limitations of a single substance through synergy. These results will promote the innovative development of quercetin precision nutrition delivery system.
Polysaccharide Based Self-Driven Tubular Micro/Nanomotors as a Comprehensive Platform for Quercetin Loading and Anti-inflammatory Function

Abstract:Quercetin (QR) is a natural flavonoid with strong anti-inflammatory properties, but it suffers from poor water solubility and bioavailability. Micro/nanomotors (NMs) are tiny devices that convert external energy or chemical fuels into an autonomous motion. They are characterized by their small size, rapid movement, and self-assembly capabilities, which can enhance the delivery of bioactive ingredients. The study synthesized natural polysaccharide-based nanotubes (NTs) using a layer-by-layer self-assembly method and combined with urease (Ure), glucose oxidase (GOx), and Fe3O4 to create three types of NMs. These NMs were well-dispersed and biocompatible. In vitro experiments showed that NMs-Fe3O4 has excellent photothermal conversion properties and potential for use in photothermal therapy. Cellular inflammation model results demonstrated that QR-loaded NMs were not only structurally stable but also improved bioavailability and effectively inhibited the release of inflammatory mediators such as IL-1β and IL-6, providing a safe and advanced carrier system for the effective use of bioactive components in food.
Pharmaceutical and pharmacokinetic characteristics of different types of fenofibrate nanocrystals prepared by different bottom-up approaches.

Abstract:Low dissolution rate of a poorly water soluble drug often leads to low and variable oral bioavailability. Formulating drugs as nanocrystals can help to overcome these problems by increasing the solubility and dissolution velocity. But different preparation approaches may result in different nanocrystals with different characteristics. In this study, three types of fenofibrate nanocrystals (FNT-NCs) were prepared by bottom-up methods, antisolvent and thermal precipitation under different conditions. These FNT-NCs were characterized by scanning electron micrography, dissolution testing, differential scanning calorimetry and powder X-ray diffractometry. A significant increase of dissolution rate was observed in the drug nanocrystals compared to the crude FNT powder (from 20% to 80% in 5 min). The crystallinity of the FNT-NCs prepared by antisolvent precipitation increased slightly, while that by thermal precipitation decreased. The oral bioavailability of two types of FNT-NCs prepared by antisolvent precipitation in rats increased notably compared to that of the crude powder (5.5-folds and 5.0-folds, respectively). However, the oral absorption of FNT-NCs prepared by thermal precipitation did not increase, although its dissolution rate was higher than that of the crude powder. In conclusion, different bottom-up methods produce different FNT-NCs with different crystallinity, which results in different oral bioavailability. Namely, a careful study and rational choice on preparation approaches are significant for the nanocrystal techniques.
Preparation and characterization of quercetin nanosuspensions using gypenosides as novel stabilizers

Abstract:To study the potential of gypenosides as natural stabilizers for the production of nanosuspensions, quercetin (QUE) was chosen as a model drug to prepare nanosuspensions stabilized by gypenosides. Here, high-speed shearing and high-pressure homogenization were used to obtain QUE nanosuspensions (QUE-NS). Different QUE-NS were prepared by changing the concentration and type of stabilizer to evaluate the stabilization efficiency of gypenosides. The results showed that the ability of gypenosides to stabilize nanosuspensions was superior to that of the conventional stabilizers at low concentrations. Using transmission electron microscopy, QUE-NS were observed to have the nanosized particles (less than 500 nm) and present a lumpy morphology. Due to the reduction of particle size, the in vitro dissolution rate and antioxidant activity of QUE-NS were significantly higher than that of QUE coarse drug. Powder X-ray diffraction and fourier-transform infrared spectroscopy showed that there was no significant difference in the chemical structure and crystal state of QUE during the production process. According to the results of stability studies, QUE-NS stabilized with gypenosides exhibited good storage stability. In conclusion, preparation of the nanosuspensions was feasible using gypenosides as novel stabilizers. Nanosuspensions stabilized by gypenosides could be used as a potential strategy for improving the solubility of poorly water-soluble drugs.

In vitro-in vivo study of CoQ10-loaded lipid nanoparticles in comparison with nanocrystals

Abstract:The present work described the effect of CoQ10 dissolution characteristics in nanocrystals and lipid nanoparticles (LNs) on its oral absorption in rats. Nanocrystals and LNs were prepared by melt-high pressure homogenization and sucrose monolaurate was used as a stabilizer in all formulations. Witepsol(®)W35 and medium-chain triglycerides (MCT) were selected as lipid additives to form LN(CoQ10+W35) and LN(CoQ10+MCT), respectively. From the results obtained, the particle size of CoQ10 nanocrystals was 285 nm, while it was reduced to 150 nm by mixture with an equal amount of lipid additives due to their lower melting points. In vitro dissolution results indicated that the drug release from two LNs was delayed compared with that from nanocrystals, and LN(CoQ10+W35) exhibited the highest drug release over 4h. Finally, in vivo evaluation demonstrated that the oral absorption of CoQ10 was markedly increased by using nanocrystals and LNs compared with a coarse suspension. A good relationship was found between the in vitro dissolution and in vivo evaluation. The enhanced oral absorption of CoQ10 by nanocrystals and LNs was due to improved dissolution. In conclusion, Witepsol(®)W35 was shown to be a better lipid additive for the preparation of LNs to increase the oral absorption of CoQ10.
Comparison of two nanocarriers for quercetin in morphology, loading behavior, release kinetics and cell inhibitory activity

Abstract:Recently, nanocarrier loading drugs have caused widespread concern. However, there are few reports about comparison of drug loading behavior of different nanoparticles. In this study, we prepared two nanocarriers (starch nanoparticles and F127 nanoparticles) for oral and intravenous injections, respectively. We used dynamic light scattering and transmission electron microscopy to study the morphology of the nanocarriers. Moreover, the loading behavior of nanoparticles on quercetin (QC) and the release behavior of nanoparticles at different pH levels were studied. In artificial gastric (pH 2.0) and intestinal (pH 6.8) juices, QC showed a sustained release in two nanoparticles. Meanwhile starch nanoparticle-QC and F127-QC nanosystems both showed relatively higher inhibitory activities than free QC on five kinds of cells, especially for cell A549, the cell inhibition ratio achieved to 55.16% and 64.06%, respectively. This study provided a reference for oral and injection of drug-loaded nanoparticles.
