Abstract:Bovine Th2 cells have usually been characterized by IL4 mRNA expression, but it is unclear whether their IL4 protein expression corresponds to transcription. We found that grass-fed healthy beef cattle, which had been regularly exposed to parasites on the grass, had a low frequency of IL4+ Th2 cells during flow cytometry, similar to animals grown in feedlots. To assess the distribution of IL4+ CD4+ T cells across tissues, samples from the blood, spleen, abomasal (draining), and inguinal lymph nodes were examined, which revealed limited IL4 protein detection in the CD4+ T cells across the examined tissues. To determine if bovine CD4+ T cells may develop into Th2 cells, naïve cells were stimulated with anti-bovine CD3 under a Th2 differentiation kit in vitro. The cells produced primarily IFNγ proteins, with only a small fraction (<10%) co-expressing IL4 proteins. Quantitative PCR confirmed elevated IFNγ transcription but no significant change in IL4 transcription. Surprisingly, GATA3, the master regulator of IL4, was highest in naïve CD4+ T cells but was considerably reduced following differentiation. To determine if the differentiated cells were true Th2 cells, an unbiased proteomic assay was carried out. The assay identified 4212 proteins, 422 of which were differently expressed compared to those in naïve cells. Based on these differential proteins, Th2-related upstream components were predicted, including CD3, CD28, IL4, and IL33, demonstrating typical Th2 differentiation. To boost IL4 expression, T cell receptor (TCR) stimulation strength was reduced by lowering anti-CD3 concentrations. Consequently, weak TCR stimulation essentially abolished Th2 expansion and survival. In addition, extra recombinant bovine IL4 (rbIL4) was added during Th2 differentiation, but, despite enhanced expansion, the IL4 level remained unaltered. These findings suggest that, while bovine CD4+ T cells can respond to Th2 differentiation stimuli, the bovine IL4 pathway is not regulated in the same way as in mice and humans. Furthermore, Ostertagia ostertagi (OO) extract, a gastrointestinal nematode in cattle, inhibited signaling via CD3, CD28, IL4, and TLRs/MYD88, indicating that external pathogens can influence bovine Th2 differentiation. In conclusion, though bovine CD4+ T cells can respond to IL4-driven differentiation, IL4 expression is not a defining feature of differentiated bovine Th2 cells.

What problem does this paper attempt to address?
### Problems the paper attempts to solve This paper mainly explores the in - vitro differentiation and regulatory mechanisms of bovine Th2 cells. Specifically, the researchers focus on the following aspects: 1. **Characteristics of bovine Th2 cells**: - Bovine Th2 cells are usually characterized by IL4 mRNA expression, but it is not clear whether their IL4 protein expression is consistent with transcription. The study found that healthy grass - fed beef cattle frequently exposed to grassland parasites showed a lower frequency of IL4+ Th2 cells in flow cytometry detection, which was similar to that of feed - fed cattle. - The researchers further examined the distribution of IL4+ CD4+ T cells in the blood, spleen, pyloric lymph nodes (draining) and inguinal lymph nodes, and found that the detection of IL4 protein in CD4+ T cells in these tissues was very limited. 2. **Th2 - differentiating ability of bovine CD4+ T cells**: - To determine whether bovine CD4+ T cells can differentiate into Th2 cells, the researchers stimulated naïve cells with anti - bovine CD3 antibody under Th2 differentiation kit conditions. The results showed that these cells mainly produced IFNγ protein, and only a few (< 10%) co - expressed IL4 protein. - Quantitative PCR confirmed an increase in IFNγ transcription level, but no significant change in IL4 transcription. Notably, GATA3 (the main regulator of IL4) was highest in naïve CD4+ T cells but decreased significantly after differentiation. 3. **Proteomic analysis of Th2 cells**: - To verify whether the differentiated cells were true Th2 cells, the researchers conducted an unbiased proteomic analysis. The analysis identified 4,212 proteins, of which 422 were differentially expressed compared with naïve cells. Based on these differential proteins, upstream components related to Th2 differentiation were predicted, including CD3, CD28, IL4 and IL33, indicating typical Th2 differentiation. 4. **Influence of Th2 - differentiation conditions**: - By reducing the anti - CD3 concentration to decrease the T - cell receptor (TCR) stimulation intensity, it was found that weak TCR stimulation almost completely inhibited the expansion and survival of Th2 cells. - Adding recombinant bovine IL4 (rbIL4) enhanced cell expansion, but the IL4 level did not change. These results indicate that although bovine CD4+ T cells can respond to Th2 - differentiation stimuli, the regulatory mode of the bovine IL4 pathway is different from that of mice and humans. 5. **Influence of external pathogens**: - Ostertagia ostertagi (OO) extract (a bovine gastrointestinal nematode) inhibited CD3, CD28, IL4 and TLRs/MYD88 signal transduction, indicating that external pathogens can affect the differentiation of bovine Th2 cells. ### Conclusion Although bovine CD4+ T cells can respond to IL4 - driven differentiation, IL4 expression is not a defining feature of differentiated bovine Th2 cells. Therefore, IL4 may not be the only signature cytokine of bovine Th2 cells. ### Keywords Bovine; Th2 differentiation; IL4; IFNγ; Proteomics ### Method overview 1. **Animal sources**: - The Wye Angus cattle herd was used and divided into two groups: grass - fed and feed - fed. 2. **CD4+ T - cell isolation**: - Blood was collected from the bovine jugular vein, PBMCs were extracted, and surface and intracellular staining was performed with fluorescent - labeled antibodies, and CD4+ T cells were sorted by flow cytometry. 3. **CD4+ T - cell differentiation**: - CD4+ T cells were cultured with anti - bovine CD3 antibody and Th1 or Th2 - differentiating media, and cells were collected on days 5 and 13 for analysis respectively. 4. **Flow cytometry analysis**: - Th1 and Th2 cells were subjected to surface and intracellular staining and analyzed using a FACSCalibur™ flow cytometer. 5. **Proteomic analysis**: - Proteomic analysis was performed using TMT tags and high - pH fractionation methods, proteins were identified by LC - MS/MS, and pathway analysis was performed using Ingenuity Pathway Analysis software. 6. **Quantitative PCR**: - Total RNA was extracted, cDNA was synthesized, and SYBR Green Superm was used.
