Abstract:Dissolved organic matter (DOM) represents one of the largest active organic carbon pools in the global carbon cycle. Although extensively studied, only <10% of DOM has been chemically characterized into individual dissolved compounds due to its molecular complexity. This study introduced a more comprehensive DOM characterization method by coupling both ion chromatography (IC) and liquid chromatography (LC) with high mass accuracy and resolution mass spectrometry. We presented a new on-the-fly...

What problem does this paper attempt to address?
This paper aims to address several key issues in the chemical characterization of dissolved organic matter (DOM): 1. **Enhancing the comprehensiveness of DOM characterization methods**: By combining ion chromatography (IC) with liquid chromatography (LC) and using high-precision, high-resolution Orbitrap mass spectrometry (OT-FTMS), a more comprehensive characterization of DOM compounds can be achieved. 2. **Improving mass accuracy**: A new "in-flight calibration" technique is introduced, utilizing the "lock mass" feature to ensure high-precision mass measurements during each scan. 3. **Enhancing reproducibility and reducing systematic errors**: The Random Forest algorithm is used to process quality control samples, reducing intra-day and inter-day variations of the instrument. Through these improvements, the study aims to better identify and quantify DOM compounds with different physicochemical properties in environmental samples, covering both positive and negative ionization modes. Additionally, this method is not limited to the chemical characterization of DOM but can also be used to analyze other complex mixtures, such as metabolites and environmental pollutants.
