Abstract:We experimentally investigate the influence of O(2) interferences on laser-induced fluorescence measurements of NO in lean methane-fueled flames at a range of pressures for both narrow-band and broadband fluorescence detection. We identify NO excitation schemes that minimize O(2) interferences. From detection scans we obtain interference spectra for the different NO excitation schemes. We then identify optimum excitation-detection schemes for narrow-band detection measurements of NO. To simulate broadband detection experiments, we numerically apply five different filter combinations to the experimentally obtained detection scans. We develop filter-assessment parameters to judge the effectiveness of the different filtering schemes, and we establish a methodology for evaluating broadband excitation-detection strategies. From this research we identify optimum excitation-detection schemes for broadband detection measurements of NO.
Effectiveness of a Worksite Wellness Program on Health Behaviors and Personal Health

Abstract:Objective: To evaluate the effectiveness of a worksite wellness program at improving health behavior and personal health. Methods: Analyses are based on 472 (71% men and 29% women) workers employed in 2009 through 2010. Results: Participants showed significant improvement in frequency of exercise, consumption of whole grains, vegetables and fruits, restful sleep, and seat belt use. Life satisfaction and perceived health also significantly increased, but job satisfaction significantly decreased and there was no change in smoking or body mass index. In addition, the percentage with borderline/high blood pressure significantly decreased. Conclusions: Participation in well-structured worksite wellness programs, such as the one evaluated in this study, may increase health and life satisfaction for employees. This type of wellness program appears to help employees develop and maintain healthy behaviors.

Abstract:The objective of this study was to investigate the factors affecting ball velocity at the final instant of the impact phase (t1) in full instep soccer kicking. Five experienced male university soccer players performed maximal full instep kicks for various foot impact points using a one-step approach. The kicking motions were captured two dimensionally by a high-speed camera at 2,500 fps. The theoretical equation of the ball velocity at t1 given in the article was derived based on the impact dynamics theory. The validity of the theoretical equation was verified by comparing the theoretical relationship between the impact point and the ball velocity with the experimental one. Using this theoretical equation, the relationship between the impact point and the ball velocity was simulated. The simulation results indicated that the ball velocity is more strongly affected by the foot velocity at the initial instant of the impact phase than by other factors. The simulation results also indicated that decreasing the ankle joint reaction force during ball impact shifts the impact point that produces the greatest ball velocity to the toe side and decreasing the ankle joint torque during ball impact shifts the impact point that produces the greatest ball velocity to the ankle side.
Interaction of fibrinogen with n-alkylagaroses and its purification by critical hydrophobicity hydrophobic interaction chromatograpy

Abstract:A rational application of hydrophobic interaction chromatography (HIC) to the purification of proteins has remained an enigma in spite of over 30 years of research. The critical hydrophobicity parameter, which can be determined from a concentration series of n-alkyl Sepharose 4B (Seph-Cn) offers the possibility of adapting the HIC gel to the needs of purification. To this end a library of HIC gels (Seph-C4 to Seph-C6) of different immobilized alkyl residue concentrations was synthesized and tested with purified bovine fibrinogen. Binding of fibrinogen to such a concentration series resulted in sigmoidal binding curves. Analysis of the Seph-C5 data according to the lattices-site binding model yielded adsorption coefficients (nS) between 5 and 10 indicating that 5-10 lattice-sites (alkyl residues) interact multivalently with a fibrinogen molecule for adsorption at low ionic strength. The apparent lattice-site half-saturation constant of dissociation lies between 21 and 25 micromol/ml packed gel. For each alkyl chain length a critical hydrophobicity could be determined. For fibrinogen purification the critical hydrohobicity gel, Seph-C5 (13 micromol/ml packed gel), was selected. With the help of the cosolvents NaCl or glycine a fully reversible adsorption of fibrinogen could be facilitated on the critical hydrophobicity gel. Application of the method to human and bovine blood plasma resulted in a single step purification of fibrinogen in high yields. A comparison of the classical purification of fibrinogen with the critical hydrophobicity HIC (CHIC) method demonstrates a reduction in preparation time from several days to ca. 1 h. The subunit structure of HIC-purified human fibrinogen is identical to the classically purified protein. In the case of bovine fibrinogen however HIC-purified fibrinogen displayed a different subunit structure in that the Aalpha chain of fibrinogen had a ca. 5 kDa higher molecular mass. This may be due to the rapidity of the new one-step method and an avoidance of proteolysis.

Abstract:Hirudin is the most potent natural inhibitor of thrombin and a powerful anticoagulant. Large-scale production of recombinant hirudin is desirable for therapy. In this study, the gene encoding hirudin variant III was redesigned and synthesized by usingBacillus subtilispreferred codons, and the recombinant hirudin variant III (rHV3) was overexpressed inB. subtilisDB403 with strong anticoagulation activity for the first time. The hirudin activity from the supernatant of culture with optimized expression conditions could reach 210 ATU/ml. The protein in culture supernatant was precipitated by trichloroacetic acid, then desalted by ultrafiltration and purified by anion exchange chromatography. Strong anion Q F.F. performed better than weak anion DEAE F.F. The proper pH and conductivity was determined at pH 8 and 6 ms/cm, respectively. The maximum applied sample was 240 ATU/ml to medium of strong anion Q F.F. This optimized procedure was employed in strong anion exchange HiPrep 16/10Q with the 90% recovery rate and 70.2% purity. After gel filtration, the purity of rHV3 checked by HPLC could reach 95.1%, and the recovery rate was 93% for this step. The purified recombinant rHV3 showed a single band in SDS-PAGE. The rHV3 was stable at 100 °C and acidity condition, but was unstable under the condition of both heating and alkalinity. In conclusion, theses studies suggests thatB.subtilismight be useful for the production of biologically active medicine peptides in secretion facilitating purification procedures, and that this isolation method was suitable for scale-up purification process at a low cost.

Abstract:The conformation of a new recombinant RGD-hirudin, which has the activities of anti-thrombin and anti-platelet aggregation, was investigated by multi-dimensional NMR spectroscopy. The H-1 NMR spectra of this protein are assigned in a sequential manner by using a combination of 2D NMR techniques to demonstrate through-bond and through-space (<5 angstrom) connectivities. The secondary structure of recombinant RGD-hirudin was deduced from chemical shift indices, sequential NOEs and (3)J(HN alpha) coupling constants. The results show that the recombinant RGD-hirudin has two anti-parallel beta-sheets and no alpha-helix, and also that the Arg-Gly-Asp (RGD) binding motif of this protein is located at the end of a long arm, which consists of two anti-parallel beta-strands (residues 26-31 and 36-41). As the strands are connected by a beta-turn, the recombinant RGD-hirudin acquires high flexibility and inhibits platelet aggregation more effectively. Copyright (c) 2005 John Wiley & Sons, Ltd.

Abstract:Recombinant RGD-Hirudin ( r-RGD-Hirudin ) has double functions: anti-thrombin activity and anti-platelet aggregation activity. To identify these functions, the expression plasmid, RGD-Hirudin-pPIC9K, was constructed by inserting cDNA of RGD-hirudin in yeast expression vector pPIC9K. The high expression clone was gained after screening. This clone was fermented for 3 days. The r-RGD-hirudin was secreted into the culture. It was ultra-filtrated from culture supernatant, then after gel filtration chromatography and anion exchange chromatography, the purified r-RGD-hirudin was gained. Its purity was larger than 97% and its specific activity was 12 000 ATU/mg. The yield per liter culture of purified r-RGD-hirudin was 1 g and overall recovery yield was more than 75% . The purified r-RGD-hirudin was identified by reductive SDS-PAGE, anti-thrombin activity assay, anti-platelet aggregation assay, LC/MS and isoelectrofocusing assay. It is proved that r-RGD-Hirudin is ramification of wt-Hirudin and it has anti-thrombin activity and anti-platelet aggregation activity.

Abstract:The solution structure of a new recombinant RGD-hirudin, which has the activities of anti-thrombin and anti-platelet aggregation, was determined by (1)H nuclear magnetic resonance spectroscopy and compared with the conformations of recombinant wild-type hirudin and hirudin (variant 2, Lys47) of the hirudin thrombin complex. On the basis of total 1284 distance and dihedral angle constraints derived from a series of NMR spectra, 20 conformers were computed with ARIA/CNS programs. The structure of residues 3-30 and 37-48 form a molecular core with two antiparallel beta-sheets as the other two hirudins. However, significant differences were found in the surface electrostatic charge distributions among the three hirudins, especially in the RGD segment of recombinant RGD-hirudin. This difference may be greatly beneficial to its additional function of anti-platelet aggregation. The difference in extended C-terminal makes its both ionic and hydrophobic interactions with the fibrinogen recognition exosite of thrombin more effective.
Studies on the Separation and Purification of Recombinant Hirudin Variant 3 from the Bacillus Subtilis

Abstract:[Objective] The research aimed to get the optimized separation and purification conditions of the hirudin produced from Bacillus subtilis DB403(pUBH5).[Method]Through the systemic pretreatment,preliminary chromatography and fine chromatography.[Result]The optimized separation and purification conditions were that:Supernatant was treated by trichloroacetic acid,then by ultrafiltration desalt and anion exchange chromatography.Strong anion Q F.F.was better than weak anion DEAE F.F.The proper balanced solution was Tris-HCl(pH 8.0).The proper conductivity was 6.0 mS/cm.The maximum applied sample was 240.0 ATU/ml to matrix of strong anion Q F.F.This optimized procedure was magnified in strong anion exchange HiPrep 16/10Q with the 90.0% recovery and 70.2% purity.The purification of gel filtration of Sephacryl S-100 to hirudin was not relative to flow rate within certain scope.The application size of sample was 10ml.The purity checked by HPLC was 95.1%,and the recovery was 93.0%,and the band of SDS-PAGE was single.[Conclusion] The research provided the reference of the further industrialization separation and purification of hiruin.

Abstract:Background Hirudin is an anti-coagulation protein produced by the salivary glands of the medicinal leech Hirudomedicinalis. It is a powerful and specific thrombin inhibitor. The novel recombinant hirudin, RGD-hirudin, which contains an RGD motif, competitively inhibits the binding of fibrinogen to GPIIb/IIIa on platelets, thus inhibiting platelet aggregation while maintaining its anticoagulant activity. Results Recombinant RGD-hirudin and six mutant variants (Y3A, S50A, Q53A, D55A, E57A and I59A), designed based on molecular simulations, were expressed in Pichia pastoris. The proteins were refolded and purified to homogeneity as monomers by gel filtration and anion exchange chromatography. The anti-thrombin activity of the six mutants and RGD-hirudin was tested. Further, we evaluated the binding of the mutant variants and RGD-hirudin to thrombin using BIAcore surface plasmon resonance analysis (SPR). Kinetics and affinity constants showed that the K D values of all six mutant proteins were higher than that of RGD-hirudin. Conclusions These findings contribute to a novel understanding of the interaction between RGD-hirudin and thrombin.

Abstract:A novel recombinant hirudin, RGD-hirudin, inhibits the activity of thrombin and the aggregation of platelets. Here, we successfully expressed (15)N, (13)C-labeled RGD-hirudin in Pichia pastoris in a fermenter. The protein was subsequently purified to yield sufficient quantities for structural and functional studies. The purified protein was characterized by HPLC and MALDI-TOF mass spectroscopy. Analysis revealed that the protein was pure and uniformly labeled with (15)N and (13)C. A bioassay showed that the anti-thrombin activity and the anti-platelet aggregation ability of the labeled protein were the same as those of unlabeled RGD-hirudin. Multidimensional heteronuclear NMR spectroscopy has been used to determine almost complete backbone (15)N, (13)C and (1)H resonance assignments of the r-RGD-Hirudin. The (15)N-(1)H HSQC spectrum of uniformly (15)N, (13)C-labeled RGD-hirudin allowed successful assignment of the signals. Examples of the quality of the data are provided for the (15)N-(l)H correlation spectrum, and by selected planes of the CBCA(CO)NH, CBCANH, and HNCO experiments. These results provide a basis for further studies on the structure-function relationship of RGD-hirudin with thrombin and platelets.
Surface plasmon resonance analysis to evaluate the importance of heparin sulfate groups' binding with human aFGF and bFGF

Abstract:Human acidic and basic fibroblast growth factors (aFGF and bFGF) are classic and well characterized members of the heparin-binding growth factor family. Heparin is generally thought to play an extremely important role in regulating aFGF and bFGF bioactivities through its strong binding with them. In order to unravel the mechanism of the interactions between heparin and FGFs, and evaluate the importance of heparin sulfate groups' binding with FGFs, surface plasmon resonance analyses were performed using IAsys Cuvettes System. Heparin and its regioselectively desulfated derivatives were immobilized on the cuvettes. aFGF and bFGF solutions with different concentrations were pipetted into the cuvettes and the progress of the interaction was monitored in real-time by Windows-based software, yielding kinetic and equilibrium constants for these interactions. In addition, in order to reduce the delicate difference among the cuvettes, inhibition analyses of mixtures of FGFs and immobilized native heparin by modified heparins were also done. The data from these two methods were similar, indicating that all sulfate groups at 2-O, 6-O and N- in heparin were required for the binding to aFGF; and that their contribution to the binding was in the order 2-O, N- and 6-O-sulfate group. In contrast, definite contribution of the 6-O-sulfate group to the binding with bFGF was most apparent, while the other two sulfate groups appeared to be necessary in the order 2-O and N-sulfate group. These methods established here can be used for analysing the effect of sulfate groups in heparin on the binding with other human FGF members or other heparin-binding proteins.
[Purification of Fusion Protein HSA/IL1ra and Its Bioactivity].

Abstract:OBJECTIVE:To purify the recombinant human serum albumin fusion protein with interleukin 1 (HAS/IL1ra) and to detect the bioactivity of the fusion protein.METHODS:The recombinant HAS/IL1ra protein was purified by affinity chromatography and ion exchange chromatography, the bioactivity of the fusion protein was detected by IL1-induced A375 S2 cell killing.RESULT:The purity of the fusion protein was at least 98 % as assessed by HPLC and the protective effect from the IL1-induced A375 S2 cell killing was similar to natural IL1ra.CONCLUSION:The purified recombinant HAS/IL1ra protein in this study has a satisfactory bioactivity.
A Method of Purifying Intact Complement Factor H from Human Plasma

Abstract:The aim of this study was to establish a method of purifying intact complement factor H (CFH) from human plasma. CFH was isolated from human plasma by polyethylene glycol (PEG) precipitation, following three sequential chromatographic columns, which consisted of l-lysine Sepharose column, Resource Q column and Sephacryl S-300 High Resolution HiPrep 16/60 column. All the above steps were performed at 4°C by Fast Protein Liquid Chromatography (FPLC) AKTA Purifier 10 with Frac-900. Identification of the purified CFH was confirmed by SDS-PAGE and Western blot. The following functions of the purified CFH were further analyzed compared with the commercial CFH in vitro: (1) binding ability with C3b; (2) binding ability with mCRP; (3) the protecting function of the hemolysis of sheep red blood cells; (4) the cofactor role for complement factor I-mediated proteolytic inactivation of C3b. Homogeneous CFH was purified from the plasma fraction through the above four steps. The purity and the functions of the purified CFH were comparable to the commercial CFH. The yield of CFH was 26±3% in our study. Compared with previous methods, our method was high yield with high purity. We established a stable and feasible system for purifying intact CFH, which could be used in the lab and clinical investigations.
Separation, Purification and Identification of Human Apolipoprotein H

Abstract:Objective To explore a method to separate and purify conveniently and efficiently apolipoprotein H from human plasma. Methods We added perchloric acid into human plasma.Apolipoprotein H was separated and purified by ion exchange chromatography and affinity chromatography.The purified apolipoprotein H was analyzed by SDS-polyacrylamide gel electrophoresis,Western blot and BCA kit. Results We repeated the experiment three times.The purity of the apolipoprotein H was all over 98%.Western blot demonstrated that anti β2-Glycoprotein Ⅰ monoclonal antibody reacted with the purified apolipoprotein H.We obtained a total of 12.56 mg purified apolipoprotein H,average 21 mg/L. Conclusions This method has good repeatability,reliability and more convenient and economical than the traditional method.
