Abstract:The field of molecular cages has attracted increasing interest in relation to the development of biological applications, as evidenced by the remarkable examples published in recent years. Two key factors have contributed to this achievement: First, the remarkable and adjustable host–guest chemical properties of molecular cages make them highly suitable for biological applications. This allows encapsulating therapeutic molecules to improve their properties. Second, significant advances have been made in synthetic methods to create water-soluble molecular cages. Achieving the necessary water solubility is a significant challenge, which in most cases requires specific chemical groups to overcome the inherent hydrophobic nature of the molecular cages which feature the organic components of the cage. This can be achieved by either incorporating water-solubilizing groups with negative/positive charges, polyethylene glycol chains, etc.; or by introducing charges directly into the cage structure itself. These synthetic strategies allow preparing water-soluble molecular cages for diverse biological applications, including cages' anticancer activity, anticancer drug delivery, photodynamic therapy, and molecular recognition of biological molecules. In the review we describe selected examples that show the main concepts to achieve water solubility in molecular cages and some selected recent biological applications.

Abstract:Supramolecular architectures have flowered over the past few decades as an inventive class of functional materials for various applications. Discrete porous molecular cages are anticipated to be potential supramolecular catalysts that mimic enzyme-like catalytic activities. In this context, dynamic covalent chemistry (DCC)-driven organic cages received an enormous research interest due to the easy engineering of these porous cages with desired shapes, sizes, and functionalities. Herein, we have highlighted the role of DCC-driven stable porous cages as catalysts where catalysis occurs via the activation of the encapsulated substrates. As a result, in many cases these hosts result in better yields and stereoselectivity compared to conventional reactions without cages. This perspective presents the use of dynamic covalent bond-driven organic cages as efficient supramolecular catalysts under both homogeneous and heterogeneous reaction conditions. Specifically, the implementation of imine/amine organic cages as solid supports for ultrafine metal nanocatalysts is briefly focused on. Furthermore, various aspects of postsynthetically modified stable organic cages as efficient supramolecular hosts in different organic transformations are emphasized. Overall, we have critically analyzed the potential of porous organic cages as suitable platforms for organic transformations and for generating catalytically active metal nanoparticles.

Abstract:Photocatalytic organic conversion is considered an efficient, environmentally friendly, and energy-saving strategy for organic synthesis. In recent decades, the molecular cage has emerged as a creative functional material with broad applications in host-guest recognition, drug delivery, catalysis, intelligent materials and other fields. Based on the unique properties of porous molecular cage materials, they provide an ideal platform for leveraging pre-structuring in the catalytic reactions and show great potential in various photocatalytic organic reactions. As a result, they have emerged promising alternatives to conventional molecules or inorganic photocatalysts in redox processes. In this Review, the synthesis strategies based on the coordination cage and organic cage, as well as their recent progress in photocatalytic organic conversion are comprehensively summarized. Finally, we deliver the persistent challenges associated with porous molecular cage compounds that need to be overcome for further development in this field.

Abstract:Supramolecular metal–organic cages, a class of molecular containers formed via coordination-driven self-assembly, have attracted sustained attention for their applications in catalysis, due to their structural aesthetics and unique properties. Their inherent confined cavity is considered to be analogous to the binding pocket of enzymes, and the facile tunability of building blocks offers a diverse platform for enzyme mimics to promote organic reactions. This minireview covers the recent progress of supramolecular metal–organic coordination cages for boosting organic reactions as reaction vessels or catalysts. The developments in the utilizations of the metal–organic cages for accelerating the organic reactions, improving the selectivity of the reactions are summarized. In addition, recent developments and successes in tandem or cascade reactions promoted by supramolecular metal–organic cages are discussed.

Abstract:Supramolecular cavitands and organic cages having a well-defined cavity and excellent host–guest complexing ability have been explored for a myriad of applications ranging from catalysis to molecular separation to drug delivery. On the other hand, porous organic polymers (POPs) having tunable porosity and a robust network structure have emerged as advanced materials for molecular storage, heterogeneous catalysis, water purification, light harvesting, and energy storage. A fruitful marriage between guest-responsive discrete porous supramolecular hosts and highly porous organic polymers has created a new interface in supramolecular chemistry and materials science, confronting the challenges related to energy and the environment. In this mini-review, we have addressed the recent advances (from 2015 to the middle of 2020) of cavitand and organic cage-based porous organic polymers for sustainable development, including applications in heterogeneous catalysis, CO2 conversion, micropollutant separation, and heavy metal sequestration from water. We have highlighted the "cavitand/cage-to-framework" design strategy and delineated the future scope of the emerging new class of porous organic networks from "preporous" building blocks.This article has not yet been cited by other publications.

Abstract:The interest in shape-persistent organic cages is nearly as old as the interest in supramolecular chemistry. In the beginning, organic cages have often been synthesized in a stepwise manner, which is not only laborious but very often also accompanied by low overall yields. In 1988, MacDowell published the one pot high-yielding synthesis of [2 + 3] imine cages based on TREN and aromatic dialdehydes, exploiting the reversible condensation of amines and aldehydes to imines, which was later used by others to make even larger cages on the basis of resorcinarenes. In 2008, the synthesis and characterization of an adamantoid [4 + 6] imine cage by condensation of a C3 v-symmetric triaminotriptycene and commercially available 4- tert-butyl salicyldialdehyde was introduced by the author, which was the ignition of our group activities in this research area. In 2011, we published the first gas-sorption data for this [4 + 6] imine cage: with a measured specific surface area of SABET = 1377 m2/g according to the model of Brunauer-Emmett-Teller (BET) this was twice as high as for the reported smaller cages of Cooper. For a second desolvated polymorph of the same cage, an even higher SABET = 2071 m2/g was determined; still one of the highest surface areas until date for porous organic molecular materials. Subsequently, the influence of the substituent in 4-position of the salicyldialdehyde for the reaction to [4 + 6] imine cages was investigated as well as the role of the phenolic hydroxyl group. It turned out that the phenolic hydroxyl group is crucial as directing group to increase the formation of the cage as well as stabilize the structure by cyclic six-membered intramolecular hydrogen bonds. The concept was extended to other imine-based cages of different geometry and size. For instance, a [4 + 4] cubic structure from triptycene trissalicylaldehyde and triptycene triamine was accessible as an amorphous insoluble solid, able to adsorb 18.2 wt % CO2 at ambient conditions. To gain further insight into the structural needs of the molecular precursors, rigidity and preorientation of reacting sites were investigated on prismatic [2 + 3] and truncated tetrahedral [4 + 4] imine cages, showing that rigidity and preorientation is beneficial or even crucial for cage formation. Furthermore, chiral self-sorting was studied on the basic of racemic triamines. Besides imine condensation, we explored the reversible formation of boronic esters from boronic acids and diols. Triptycene tetraol with its 120° angle between the aromatic units has been used in the condensation with benzene triboronic acid to achieve a large cuboctahedral [12 + 8] cage with pore dimensions of 2 nm, which are by IUPAC definition mesoporous. After activation the measured specific surface area was SABET = 3758 m2/g, a number rarely achieved even for other porous compounds such as threedimensional framework materials. Smaller tetrahedral [4 + 6] boronic ester cages were synthesized too. These cages show a selective gas sorption with preference of saturated hydrocarbon ethane over ethylene and acetylene. What distinguishes porous materials derived from molecular cages from three-dimensional frameworks or networks the most is their solubility; thus, the cages are soluble porous units (SPUs) in a broader sense. Taking advantage of this, [4 + 6] imine cages were postfunctionalized in solution to change the gas sorption properties in the crystalline state. Furthermore, cage solutions were spray-coated onto quartz crystal microbalances to enhance affinity and selectivity for sensing of airborne analytes. Abstract:Porous organic cages (POCs) are constructed from purely organic synthons by covalent linkages with intrinsic cavities and have shown potential applications in many areas. However, the majority of POC synthesis methods reported thus far have relied on dynamically reversible imine linkages, which can be metastable and unstable under humid or harsh chemical conditions. This instability significantly hampers their research prospects and practical applications. Consequently, strategies to enhance the chemical stability of POCs by modifying imine bonds and developing robust covalent linkages are imperative for realizing the full potential of these materials. In this review, we aim to highlight recent advancements in synthesizing chemical-stable POCs through these approaches and their associated applications. Additionally, we propose further strategies for creating stable POCs and discuss future opportunities for practical applications.

Abstract:This account deals with our work on cage systems that was reported during the past two decades. Here, we summarized various new synthetic routes to diverse cage polycycles by employing Claisen rearrangement, Diels–Alder (DA) reaction, [2+2] photocycloaddition, Grignard addition, Fischer indolization, ring‐closing metathesis (RCM), Van Leusen oxazole synthesis, and rearrangement reaction as key steps. Several heterocyclic cage frameworks (oxacycles and oxazoles), polycyclic bisindoles, cage spirocycles, cage energetic systems, D3‐trishomocubanes, cage propellanes and unusual polycycles were designed starting with inexpensive and easily accessible starting materials. In some occasions, we also prepared biologically important triquinane frameworks starting with readily available cage diones via photo‐thermal olefin metathesis strategy under microwave irradiation conditions. This new protocol avoids specialized equipment such as flash vacuum pyrolysis. Additionally, we have also shown interesting rearrangements in cage frameworks to generate unusual new and cage systems with the aid of the Lewis acids such as BF3.OEt2. Moreover, these functionalized cage systems can be considered as a useful candidates in material science, medicinal chemistry, and also in drug in design.

Abstract:Green solvent is a topic of increasing attention in modern chemistry because of the consciousness of the effect of solvents on air quality, energy usage, pollution, and continuous climate change. Over the past few years, remarkable progress has been made to accomplish these goals by giving interest in sustainable synthesis. For sustainable development, continuous efforts have been put forward for addressing these issues and the chemical approaches are now analysed for the usage of solvents. Realizing solvent property is a crucial area of green chemistry, and therefore the solvents were categorized by their handling techniques, ecological, and healthiness (ESH) features. The focus of this ranking is to help the researchers in choosing their reaction solvent efficiently, keeping in mind the ESH values of the solvents and discouraging them from using environmentally harmful and toxic solvents which are commonly used in laboratories. In this review article, we are trying to consider various synthetic methodologies of the most protuberant sustainable solvents such as polyethylene glycol, supercritical CO2, and water. We herein examine not only the chemical reactions but also represent the mechanism and synthetic utility of each class of solvent inside the background of the reactions for which it is used, but also documented the widespread substrate scope of the reaction. A broad range of economic and environmental factors are also demonstrated, giving a wider display of the current status of sustainable solvent research and development.

Abstract:N-Heterocyclic carbene (NHC)-based cages have emerged as a prominent and dynamic research area within the research field of chemistry. Leveraging the distinctive electronic and steric properties of NHC ligands, the design, synthesis, and application of these corresponding cages have garnered substantial scholarly interest. In recent years, we have witnessed the successful fabrication of diverse NHC-based cages through a range of synthetic methodologies, which hold significant potential for applications in molecular recognition and catalysis. In this review, we delve into the foundational synthetic strategies that underlie the creation of NHC-based cages, employing approaches encompassing metal–NHC chelation, coordination assembly, and covalent bonding. Additionally, we compile the diverse applications of these cages within catalytic processes and molecular recognition. Lastly, we shed light on the current limitations of synthesis and outline future trends in the development of NHC-based cages.

Abstract:Shape‐persistent organic cages are an intriguing class of molecular porous materials. Through hierarchical molecular design, size and shape of the intrinsic molecular voids are controlled by dynamic covalent chemistry, while pore structure and topology are governed by noncovalent alignment in the solid state. However, the predictable and reliable crystallization of organic cages is still challenging since long‐range superstructures are solely based on weak and rather unidirectional supramolecular interactions. In this tutorial review, we provide a general classification of porous solid‐state materials and discuss specific design principles regarding the dynamic covalent reactions, the small‐molecule building blocks and solid‐state engineering. Furthermore, we introduce the most important analytical techniques for porous materials with a special focus on organic cages.

Abstract:The emergence of supramolecular chemistry has led to the discovery of a rising number of macrocycles and cages with a range of functionalities. Most of these supramolecular aggregates are metal coordination networks, whereas pure organic assemblies are less developed. Organic macrocycles and cages have the advantages of chemical robustness, processability in organic solvents, and suitability for pilot-scale applications. They are constructed primarily from covalent bonds, with irreversible and reversible bond types. We herein highlight the use of different versatile bonding approaches in engineering these soft materials, as well as their emerging applications, such as gas storage, thin films, liquid crystals, and catalysis.

Abstract:The recent advancements in utilizing organocatalysts for the synthesis of organic compounds have been described in this review by focusing on their simplicity, effectiveness, reproducibility, and high selectivity which lead to excellent product yields. The organocatalytic methods for various derivatives, such as indoles, pyrazolones, anthrone-functionalized benzylic amines, maleimide, polyester, phthalimides, dihydropyrimidin, heteroaryls, N -aryl benzimidazoles, stilbenoids, quinazolines, quinolines, and oxazolidinones have been specifically focused. The review provides more understanding by delving into potential reaction mechanisms. We anticipate that this collection of data and findings on successful synthesis of diverse compound derivatives will serve as valuable resources and stimulating current and future research efforts in organocatalysis and industrial chemistry.

Rapid and scalable synthesis of porous organic cages with solvent recyclability

Abstract:Herein, we report the feasible synthesis of a series of porous organic cages (POCs, e.g., CC1, CC2, CC4, and CC3–OH) through a straightforward and effective dynamic solvent-mediated strategy. Notably, the preparation of POC nanocrystals can be significantly shortened from days/weeks (by traditional methods) to just 4 h, even at a large scale (around 2 g), due to the accelerated imine-bond formation. The easy-purification of POCs involving a filtration procedure also enables good solvent recyclability, which has been seldomly discussed in previous reports, but may be promising in potential industrial production. Moreover, the solvent media induce increased disorder of cage molecules within the nanocrystals, imparting expanded porosity with enhanced gas sorption property. Our finding may provide a universal tool for the synthesis and utilization of certain imine-based POCs, potentially advancing this field.

Abstract:Porous materials with well-defined pore structures have received considerable attention in the past decades due to their unique structures and wide applications. Most porous materials such as zeolites, metal-organic frameworks, covalent organic frameworks and porous organic polymers are extended to infinite frameworks or networks by robust covalent or coordination bonds. Porous molecular cages compose of discrete molecules with permanent cavities are an emerging class of porous material and the discrete molecules assemble into solids by weak intermolecular interaction. In comparison to porous extended solids such as metal-organic frameworks and covalent organic frameworks, porous molecular cage solids are generally soluble in organic solvents thus allowing solution processing, making them more convenient to apply in many fields. This review mainly focuses on the recent advance of application of porous molecular cages (porous organic cages and metal-organic cages) for enantioselective recognition and separation from 2010 to present, including gas chromatography, capillary electrochromatography, chiral fluorescent recognition, chiral potentiometric sensing and enantioselective adsorption. Abstract:Selective anion recognition remains a key challenge in supramolecular chemistry: only a very small number of systems that can function in water are known, and these nearly always preferentially bind hydrophobic anions. In this work, we report three robust hexa-cationic cages that can be prepared on scales up to 14 g in two simple and high-yielding steps from commercially-available materials. One of these cages displays unusually strong sulfate binding in water (Ka = 12,000 M–1), and demonstrates high selectivity for this anion over H2PO4–/HPO42– in DMSO/buffer mixtures. These results demonstrate that relatively large, three-dimensional supramolecular hosts can be prepared in high yields and on large scales, and can be highly potent receptors.

Abstract:Porous organic cages have shape persistent cavities which provide a suitable platform for encapsulation of guest molecules with size suitably fitting to the cavity. The interactions of the guest molecule with the porous organic cage significantly alter the properties of the guest molecule. Herein, we report the effect of encapsulation on the kinetics of various organic transformations including 2 + 4 cycloaddition, 1,5-sigmatropic, 6π-electrocyclization, ring expansion, cheletropic, dyotropic, trimerization and tautomerization reactions. Non-bonding interactions are generated between the CC2 cage and encapsulated species. However, the number and nature/strength of interactions are different for reactant and TS with the CC2 cage and this difference detects the reaction to be accelerated or slowed down. A significant drop in the barrier of reactions is observed for reactions involving strong interactions of the transition state within the cage. However, for some reactions such as the Claisen rearrangement, reactants are stabilized more than the transition state and therefore an increase in activation barrier is observed. Furthermore, non-covalent analyses of all transition states (inside the cage) confirm the interaction between the CC2 cage and substrate. The current study will promote further exploration of the potential of other porous structures for similar applications.
