Alkali-metal hexamethyldisilazide initiated polymerization on alpha-amino acid N-substituted N-carboxyanhydrides for facile polypeptoid synthesis

Abstract:<p>Polypeptoids have been explored as mimics of polypeptides, owing to polypeptoids' superior stability upon proteolysis. Polypeptoids can be synthesized from one-pot ring-opening polymerization of amino acid <em>N</em>-substituted <em>N</em>-carboxyanhydrides (NNCAs). However, the speed of polymerization of NNCAs can be very slow, especially for NNCAs bearing a bulky <em>N</em>-substitution group. This hindered the exploration on polypeptoids with more diverse structures and functions. Therefore, it is in great need to develop advanced strategies that can accelerate the polymerization on inactive NNCAs. Hereby, we report that lithium/sodium/potassium hexamethyldisilazide (Li/Na/KHMDS) initiates a substantially faster polymerization on NNCAs than do commonly used amine initiators, especially for NNCAs with bulky <em>N</em>-substitution group. This fast NNCA polymerization will increase the structure diversity and application of polypeptoids as synthetic mimics of polypeptides.</p>

What problem does this paper attempt to address?
This paper aims to solve the problem of slow polymerization of **N - substituted N - carboxylic anhydrides (NNCAs)**, especially for those NNCAs with bulky N - substituents. Since traditional amine - based initiators are less efficient in polymerizing these inactive NNCAs, the process of synthesizing polypeptoids is time - consuming and the structural diversity is limited. Therefore, researchers are in urgent need of developing new strategies that can accelerate the polymerization of such NNCAs to improve the structural diversity and application range of polypeptoids. The paper proposes that using lithium, sodium or potassium hexamethyldisilazane (Li/Na/KHMDS) as an initiator can significantly accelerate the polymerization rate of NNCAs, especially for those NNCAs with bulky N - substituents. This finding can not only shorten the reaction time, but also improve the uniformity of the molecular weight distribution of the product, thereby providing a broader material basis for the research and application of polypeptoids.
