Understanding the microscopic thermal effects of the hydrogel is important for its application in diverse fields, including thermal‐related studies in tissue engineering and thermal management for flexible electronic devices. In recent decades, localized thermal properties, such as thermal conductivity, have often been overlooked due to technical limitations. To tackle this, the study proposes a new hybrid diamond microsensor that is capable of simultaneous temperature control and readout in a decoupled manner. Specifically, the sensor consists of a silicon pillar (heater) at ≈10 microns in length, topped by a micron‐sized diamond particle that contains silicon‐vacancy (SiV) centers (thermometer) with 1.29 KHz−12 temperature measurement sensitivity. Combining this innovative, scalable sensor with a newly established simulation model that can transform heating‐laser‐induced temperature change into thermal conductivity, an all‐optical decoupled method is introduced with ≈0.05 W m−1 K−1 precision, which can reduce laser crosstalk. For the first time, the thermal conductivity change of hydrogels during the gelation process is tracked and the existence of variation is demonstrated. The study introduces a rapid, undisturbed technique for measuring microscale thermal conductivity, potentially serving as a valuable tool for cellular thermometry, and highlights the idea that decoupling can reduce crosstalk from different lasers, which is helpful for quantum sensing.

Abstract:Intracellular thermometry provides important information about the physiological activity of single cells and has been implemented using diverse temperature-sensitive materials as nanoprobes. However, measuring the temperature of specific organelles or subcellular structures is challenging because it requires precise positioning of the nanoprobes. Here, it is shown that dispersed fluorescent nanodiamonds (FNDs) endocytosed in living cells can be aggregated into microspheres using optical forces and used as intracellular temperature probes. The aggregation of the FNDs and electromagnetic resonance between individual nanodiamonds in the microspheres lead to a sevenfold intensity enhancement of 546-nm laser excitation. With the assistance of a scanning optical tweezing system, the FND microspheres can be precisely patterned and positioned within the cells. By measuring the fluorescence spectra of the microspheres, the temperatures at different locations within the cells are detected. The method provides an approach to the constructing and positioning of nanoprobes in an intracellular manner, which has potential applications in high-precision and flexible single-cell analysis.

Abstract:A suspended system for measuring the thermal properties of membranes is presented. The sensitive thermal measurement is based on the 3$\omega$ dynamic method coupled to a V$\ddot{o}$lklein geometry. The device obtained using micro-machining processes allows the measurement of the in-plane thermal conductivity of a membrane with a sensitivity of less than 10nW/K (+/-$5x10^{-3}$Wm$^{-1}K^{-1}$ at room temperature) and a very high resolution ($\Delta K/K =10^{-3}$). A transducer (heater/thermometer) centered on the membrane is used to create an oscillation of the heat flux and to measure the temperature oscillation at the third harmonic using a Wheatstone bridge set-up. Power as low as 0.1nanoWatt has been measured at room temperature. The method has been applied to measure thermal properties of low stress silicon nitride and polycrystalline diamond membranes with thickness ranging from 100 nm to 400 nm. The thermal conductivity measured on the polycrystalline diamond membrane support a significant grain size effect on the thermal transport.

Abstract:We report a new approach to controllable thermal stimulation of a single living cell and its compartments. The technique is based on the use of a single polycrystalline diamond particle containing silicon-vacancy (SiV) color centers. Due to the presence of amorphous carbon at its intercrystalline boundaries, such a particle is an efficient light absorber and becomes a local heat source when illuminated by a laser. Furthermore, the temperature of such a local heater is tracked by the spectral shift of the zero-phonon line of SiV centers [2]. Thus, the diamond particle acts simultaneously as a heater and a thermometer. In the current work, we demonstrate the ability of such a Diamond Heater-Thermometer (DHT) to locally alter the temperature, one of the numerous parameters that play a decisive role for the living organisms at the nanoscale. In particular, we show that the local heating of 11-12 °C relative to the ambient temperature (22 °C) next to individual HeLa cells and neurons, isolated from the mouse hippocampus, leads to a change in the intracellular distribution of the concentration of free calcium ions. For individual HeLa cells, a long-term (about 30 s) increase in the integral intensity of Fluo-4 NW fluorescence by about three times is observed, which characterizes an increase in the [Ca2+]cyt concentration of free calcium in the cytoplasm. Heating near mouse hippocampal neurons also caused a calcium surge - an increase in the intensity of Fluo-4 NW fluorescence by 30% and a duration of ~0.4 ms.

Abstract:Real-time temperature monitoring inside living organisms provides a direct measure of their biological activities, such as homeostatic thermoregulation and energy metabolism. However, it is challenging to reduce the size of bio-compatible thermometers down to submicrometers despite their potential applications for the thermal imaging of subtissue structures with single-cell resolution. Light-emitting nanothermometers that remotely sense temperature via optical signals exhibit considerable potential in such \textit{in-vivo} high-spatial-resolution thermometry. Here, using quantum nanothermometers based on optically accessible electron spins in nanodiamonds (NDs), we demonstrate \textit{in-vivo} real-time temperature monitoring inside \textit{Caenorhabditis elegans} (\textit{C. elegans}) worms. We developed a thermometry system that can measure the temperatures of movable NDs inside live adult worms with a precision of $\pm 0.22^{\circ}{\rm C}$. Using this system, we determined the increase in temperature based on the thermogenic responses of the worms during the chemical stimuli of mitochondrial uncouplers. Our technique demonstrates sub-micrometer localization of real-time temperature information in living animals and direct identification of their pharmacological thermogenesis. The results obtained facilitate the development of a method to probe subcellular temperature variation inside living organisms and may allow for quantification of their biological activities based on their energy expenditures.

Abstract:It has been known that cells have mechanisms to sense and respond to environmental noxiousness and mild temperature changes, such as heat shock response and thermosensitive TRP channels. Meanwhile, new methods of measuring temperature at the cellular level has recently been developed using fluorescent nanothermometers. Among these thermometers, fluorescent polymeric thermometers and fluorescent nanodiamonds excel in the properties required for intracellular thermometry. By using these novel methods to measure the temperature of single cells in cultures and tissues, it was revealed that spontaneous spatiotemporal temperature fluctuations occur within cells. Furthermore, the temperature fluctuations were related to organelles such as mitochondria and cellular and physiological functions, revealing a close relationship between intracellular temperature and cellular functions.

Abstract:Luminescence thermometry usually suffer from cellular complexity of the biochemical environment (such as pH and ionic strength), and thus the accuracy and reliability of the determined intracellular temperature are directly affected. Herein, a photoluminescent nanothermometer composed of polymer encapsulated quantum dots (P-QD) has been developed. And the prepared nanothermometer exhibits some advantages: such as non-sensitivity to pH and ionic strength, as well as high detection sensitivity and ultrahigh reversibility. The intracellular temperature was accurately determined under physiological conditions with different pH and ionic strength, and direct measurement of thermogenesis in individual cells has been achieved.

Abstract:Viscoelasticity of the cytoplasm plays a critical role in cell morphology and division. In parallel, local temperature is coupled to viscoelasticity and influences cellular bioenergetics. Probing the interdependence of intracellular temperature and viscoelasticity provides an exciting opportunity for the study of metabolism and disease progression. Here, we present a dual-mode quantum sensor, capable of performing simultaneous nanoscale thermometry and rheometry in a dynamic cellular environment. Our technique uses nitrogen-vacancy centres in nanodiamond, combining sub-diffraction resolution single-particle tracking in a fluidic environment with optically detected magnetic resonance spectroscopy. We demonstrate nanoscale sensing of temperature-dependent viscoelasticity in complex media. We then use our sensor to investigate the interplay between intracellular forces and cytoplasmic rheology in live cells, revealing details of active trafficking and nanoscale viscoelasticity.

Abstract:In this paper, a new method for in situ thermal conductivity measurements were established in diamond anvil cells (DAC) and achieve measurements at high pressure and high temperature under steady heat flow conditions. In this method, the traditional heat flux calibration was replaced by temperature measurement of anvils and finite element analysis. Furthermore, a DAC set-up with two heating furnaces was developed to better serve the proposed method. The thermal conductivity of four diamond anvils with different nitrogen content was measured and compared to reference to verify the method’s feasibility and serve as calibration for future measurements.

Abstract:Energy transformation, which can be described as temperature variations, is present in all activities of cellular metabolism. Detecting the temperature of numerous cells over the course of their lives is useful for studying the cellular physiological function and investigating the pathophysiology of illnesses. Current cell thermometric technologies, such as luminous temperature sensors, infrared thermometers, and thermocouple thermometers, have their own limitations, such as complicated operation, low repeatability, and inability to measure cell temperature in normal situations. Here, we have developed a high-precision platinum temperature sensor with small heat capacity, good heat conduction, excellent stability and biological compatibility, which made it possible to sense cells heat signal. The platinum resistance sensor's temperature coefficient of resistance was around 2800 ppm degrees C-1, with a good linear correlation (R (2) > 0.999) between resistance and temperature from 35 degrees C to 43 degrees C. The noise of the sensor system was below 0.02 degrees C, giving a small and stability noise in detection. The detecting cells were CHO-K1 cells. They caused a temperature increase of about 0.075 degrees C, which is higher than the sensor without cells. This platinum sensor has been found to be useful for in situ temperature measurement of cells, which is important for investigating cell informatics and pathology.

Abstract:Medical applications of nanotechnology typically focus on biosensors and drug delivery. Here, we combine nanotechnology and chemical synthesis to create a novel type of thermo-sensitive nanohybrid (CD-MSN@UP38) particle for intracellular temperature sensing and temperature-controlled drug release, which could lead to new cancer diagnosis techniques and therapies. The nanohybrid particles were first developed as a sensitive thermometer by loading them with a polarity-sensitive dye (BBD). The presented thermometer possessed high temporal resolution and temperature resolution down to 0.2 °C; combined with fluorescence lifetime imaging microscopy, the particles are well suited for mapping and sensing temperature distributions at the single-cell level. More interestingly, the nanohybrid particles can be further applied as a smart temperature-controlled drug delivery system for cancer therapy, through simply being triggered by the thermal effects of the cancer cells themselves. Our drug delivery and therapy system demonstrates highly efficient therapeutics with minimal side effects.
Single Silicon Nanowires As Inherent Heaters and Thermometers for Thermal Conductivity Measurements

Abstract:It is traditionally challenging to measure the thermal conductivity of nanoscale devices. In this Letter, we demonstrate a simple method for the thermal conductivity measurements of silicon nanowires by using the silicon nanowire under test as the thermometer and heater. The silicon nanowire (SiNW) arrays are patterned out of a silicon-on-insulator (SOI) wafer by standard microfabrication processes. The thermal conductivity of SiNWs with a width from 150nm to 400nm (while the thickness is fixed at 220nm) are measured in the temperature range of 20-200K. At low temperature range, the nanowire thermal conductivity exhibits a strong size dependency since the effective phonon mean-free path is dominated by the nanowire radial size. At high temperature range, the phonon mean-free path is significantly smaller than the nanowire radial size. The nanowire thermal conductivity is strongly temperature correlated and nearly independent of the nanowire size. Density function theory calculations are also performed on the SiNWs and the calculated thermal conductivity of SiNWs are largely consistent with the experimental data, showing that our method is valid for the thermal conductivity measurements of nanoscale devices.

Abstract:In this article, we present a kind of dual-emission fluorescent nanothermometer, which is made of europium (Eu3+)-doped silicon nanoparticles (Eu@SiNPs), allowing the detection of intracellular temperature in living cells with high accuracy. In particular, the presented SiNP-based thermometer features dual-emission fluorescence (blue (455 nm) and red (620 nm) emission), negligible toxicity (cell viability of treated cells remains above 90% during 24 h of treatment) and robust photostability in living cells (i.e., preserving >90% of fluorescence intensity after 45 min of continuous UV irradiation). More significantly, the fluorescence intensity of the Eu@SiNPs exhibits a linear ratiometric temperature response in a broad range from 25 to 70 °C. Taking advantage of these attractive merits, the Eu@SiNP-based nanothermometer is able to accurately (∼4.5% change per °C) determine dynamic changes in intracellular temperature in a quantitative and long-term (i.e., 30 min) manner.

Abstract:Sensitive probing of temperature variations on nanometer scales represents an outstanding challenge in many areas of modern science and technology. In particular, a thermometer capable of sub-degree temperature resolution as well as integration within a living system could provide a powerful new tool for many areas of biological research, including temperature-induced control of gene expression and cell-selective treatment of disease. Here, we demonstrate a new approach to nanoscale thermometry that utilizes coherent manipulation of the electronic spin associated with nitrogen-vacancy (NV) color centers in diamond. We show the ability to detect temperature variations down to 1.8 mK (sensitivity of 9 mK/sqrt(Hz)) in an ultra-pure bulk diamond sample. Using NV centers in diamond nanocrystals (nanodiamonds), we directly measure the local thermal environment at length scales down to 200 nm. Finally, by introducing both nanodiamonds and gold nanoparticles into a single human embryonic fibroblast, we demonstrate temperature-gradient control and mapping at the sub-cellular level, enabling unique potential applications in life sciences.

Abstract:Based on the transient hot-wire method and well developed microfabrication techniques, a new thermal sensor, in which the microfabricated gold wire serves as both a heater and a thermometer, has been developed for measuring the thermal conductivities of semi-rigid materials. The intention that only one calibration constant is needed for an entire batch of sensors has been validated. Thermal conductivities of distilled water, ethylene glycol, dimethyl sulfoxide (DMSO), and apple at different temperatures have been obtained. Measurements are in agreement with suggested values. Complete results will be presented at the conference. The operating temperature of 0 degrees C to 27 degrees C has been tested and is anticipated to be -20 degrees C to 60 degrees C. In spite of conventional advantages, the presented sensor offers new exciting superior performance characteristics, such as reduced thermal resistivity and higher sensitivity, easier manufacturing for mass production, flexibility to reconfigure the sensor for samples with various sizes and shapes, and much reduced workload for system calibration.

Abstract:The measurements of temperature directly influence the reasonability of experiments at high pressure and high temperature. In this article, we proposed a new integration design, the built-in thermocouple, for in situ temperature measurements in high-pressure-high-temperature experiments by fusing the characteristics of thermocouples and diamond anvil cells together. By integrating an S-type thermocouple inside the gasket of a diamond anvil cell, we successfully measured the temperature of the sample straight inside the pressure chamber at high pressure and high temperature. The setup underwent multiple experimental tests using internal and external heating techniques, the results of which revealed its capability to directly characterize the temperature of the sample with comparable accuracy and reliability to that of the typical external thermocouple setup. The proposed setup has also resolved the issue of the discrepancy of temperatures inside and outside the sample chamber and enormously expedited the temperature measurements by significantly reducing the response time of the thermocouple. In conclusion, the built-in thermocouple is a promising approach toward high-efficiency, in situ temperature measurements under extreme conditions.

Abstract:Temperature is an essential parameter in all biological systems, but information about the actual temperature in living cells is limited. Especially, in photothermal therapy, local intracellular temperature changes induce cell death but the local temperature gradients are not known. Highly sensitive nanothermometers would be required to measure and report local temperature changes independent of the intracellular environment, including pH or ions. Fluorescent nanodiamonds (ND) enable temperature sensing at the nanoscale independent of external conditions. Herein, we prepare ND nanothermometers coated with a nanogel shell and the photothermal agent indocyanine green serves as a heat generator and sensor. Upon irradiation, programmed cell death was induced in cancer cells with high spatial control. In parallel, the increase in local temperature was recorded by the ND nanothermometers. This approach represents a great step forward to record local temperature changes in different cellular environments inside cells and correlate these with thermal biology.
