Abstract:The health of the Dayang River ecosystem, critical freshwater resource for Dandong City, is intertwined with local water management and coastal development. In June 2023, we conducted a biodiversity survey in the Dayang River basin using a combined morphological and environmental DNA (eDNA) approach. We assessed both species and functional diversity of summer macroinvertebrates. A total of 81 macroinvertebrates species were identified, with some overlap in species detection between the methods. The trunk stream exhibited lower species diversity with uneven distribution across sites, whereas the tributaries displayed a more stable species composition. Functional diversity analysis mirrored this pattern, revealing lower functional diversity in the trunk stream, indicative of intense competition and niche occupation. Conversely, the tributaries displayed more uniform and complete functional diversity. This study highlights the differential influence of hydrological environments on macroinvertebrate communities during the summer season, with the trunk stream favoring certain species with specific habitat preferences and adaptations. Furthermore, it underscores the value of combining traditional morphological surveys and eDNA techniques for comprehensive biodiversity assessments.

What problem does this paper attempt to address?
The paper aims to study the biodiversity of macrobiotic communities in the summer of the Dayang River Basin, combining morphological surveys and environmental DNA (eDNA) technology for a comprehensive assessment. Specifically, the paper attempts to address the following issues: 1. **Assessment of Biodiversity**: Identify the macrobiotic species and their distribution in the main river and tributaries of the Dayang River Basin through morphological surveys and eDNA technology. 2. **Species Richness and Functional Diversity**: Compare the species richness, evenness, and functional diversity of macrobiotic communities in the main river and tributaries, exploring the impact of different hydrological environments on the structure of macrobiotic communities. 3. **Impact of Environmental Factors**: Analyze the influence of water quality parameters (such as pH, dissolved oxygen, total dissolved solids, etc.) on the composition of macrobiotic communities, especially the regulatory role of temperature on the distribution of representative species. 4. **Complementarity of Morphological and eDNA Techniques**: Validate the complementary advantages of morphological methods and eDNA technology in monitoring macrobiotic diversity, and evaluate the effectiveness of combining the two methods. In summary, through a comprehensive survey of the summer macrobiotic diversity in the Dayang River Basin, the paper aims to provide scientific evidence for the formulation of conservation strategies for freshwater ecosystems in the region, and emphasizes the importance of integrating traditional morphological and modern molecular biological techniques.
