Abstract:Cells inhabit a mechanical microenvironment that they continuously sense and adapt to. The plasma membrane (PM), serving as the boundary of the cell, plays a pivotal role in this process of adaptation. In this Review, we begin by examining well-studied processes where mechanoregulation proves significant. Specifically, we highlight examples from the immune system and stem cells, besides discussing processes involving fibroblasts and other cell types. Subsequently, we discuss the common molecular...

What problem does this paper attempt to address?
The paper primarily explores how cells perceive and adapt to mechanical signals in their microenvironment through the plasma membrane (PM). Specifically, the paper focuses on the following aspects: 1. **Role of the Plasma Membrane in Mechanical Regulation**: - The paper first introduces the critical role of the plasma membrane as the boundary between the cell and the external environment in adapting to the mechanical microenvironment. 2. **Mechanisms of Mechanical Regulation in Different Cell Types**: - The study highlights examples of mechanical regulation in the immune system, stem cells, and other cell types such as fibroblasts. 3. **Molecular Mechanisms**: - It discusses in detail how cells sense mechanical signals through molecules like integrins, YAP/TAZ, and Piezo, and convert these signals into chemical responses. 4. **Applications in Drug Discovery and Tissue Engineering**: - The paper explores how understanding these molecular components can drive advancements in drug discovery and tissue engineering, and discusses methods for measuring cellular mechanical properties. 5. **Role of Endocytosis in Mechanical Regulation**: - It analyzes how cells respond to changes in membrane mechanical properties through endocytosis and exocytosis, and how these processes affect membrane mechanics. 6. **Mechanical Integrity of Myocyte Fusion and Myofibers**: - Finally, the study examines the importance of mechanical regulation during myocyte fusion and the roles of various molecular components and processes. In summary, the paper aims to provide a comprehensive overview of how cells perceive and adapt to mechanical signals through the plasma membrane, and to explore the specific mechanisms of this process in different cell types and its applications in biology and medicine.
