Abstract:Enhancing angiogenesis may be an effective strategy to promote functional recovery after ischemic stroke. Inflammation regulates angiogenesis. Microglia are crucial cells that initiate inflammatory responses after various brain injuries. Long noncoding RNA nuclear paraspeckle assembly transcript 1 (NEAT1) plays a role in regulating brain injury. This study aimed to explore the effects of NEAT1-regulated microglial polarization on the neovascularization capacity of cerebrovascular endothelial cells and the underlying molecular regulatory mechanisms.

What problem does this paper attempt to address?
The problem this paper attempts to address is to explore how the long non-coding RNA NEAT1 affects the angiogenic capacity of cerebral arterial endothelial cells by regulating the polarization state of microglia, and the molecular mechanisms behind it. Specifically, the study aims to investigate the impact of NEAT1-regulated microglial M1 polarization on the angiogenic capacity of cerebral vascular endothelial cells and to reveal whether this effect is mediated through the AMPK signaling pathway. The background of the paper mentions that ischemic stroke is the second leading cause of disability and death worldwide, especially in low- and middle-income countries. Enhancing angiogenesis may be an effective strategy to promote functional recovery after ischemic stroke. Inflammation plays a critical role in angiogenesis, and microglia are key cells that initiate the inflammatory response after various brain injuries. NEAT1 plays a role in regulating brain injury, but its specific mechanisms are not yet fully understood. Therefore, this study not only helps to understand the role of NEAT1 in ischemic stroke but also may provide new targets for the prevention and treatment of brain injury caused by ischemic stroke.
