Abstract:Aims: The effects of exogenously applied salicylic acid (SA) on gas exchange characteristics, photosynthetic pigments and chloroplast ultrastructure were investigated in gerbera at their reproductive stage under salt-stressed conditions. Methodology: A pot experiment was conducted under glasshouse conditions at the Zhejiang University, Hangzhou, China, (30° N/120° E) between February 2008 and March 2009.Plants, pretreated with foliar applications of 0, 0.5, and 1.0 mmoldm-3 SA at the onset of flower initiation were irrigated with 100 mmoldm-3NaCl(aq) for two weeks, starting after three days from the SA pretreatment. Control did not receive either NaCl or SA.Photosynthetic rate, gas exchange, photosynthetic pigments content and chloroplast ultrastructure were investigated against treatments. All data were subjected to analysis of variance (ANOVA) and Generalized Linear Model (GLM) using SAS statistical software. Pearson’s correlation test was carried out to study the relationships among the parameters. The means were compared using Duncan’s multiple range test (DMRT). For all the tests, P< .05 was considered statistically significant. Results: Salt stress adversely affected the gas exchange characteristics, photosynthetic pigment contents and chloroplast ultrastructure. SA application significantly increased the net photosynthesis, stomatal conductivity, intra-cellular CO2 content and transpiration rate but decreased the stomatal limitation, compared to those of untreated salt-stressed plants. Further, the enhanced photosynthetic pigment contents and notably undamaged chloroplast ultrastructure were evident of the ameliorative effects of SA on photosynthetic system under salt stress. Of the two concentrations tested, 0.5 mmoldm-3 SA concentration seemed to have greater effect throughout the experiment showing no significant variation from control in some attributes (chlorophyll contents and chloroplast ultrastructure). Conclusion: Responses of plants pretreated with SA spraying and significant correlation among them plausibly suggest SA-induced enhancement of photosynthetic system as another target for conferring salt tolerance in crop plants.

Abstract:Small molecules in plants - such as metabolites, phytohormones, reactive oxygen species (ROS), and inorganic ions - participate in the processes of plant growth and development, physiological metabolism, and stress response. Wearable electrochemical sensors, known for their fast response, high sensitivity, and minimal plant damage, serve as ideal tools for dynamically tracking these small molecules. Such sensors provide producers or agricultural researchers with noninvasive or minimally invasive means of obtaining plant signals. In this review we explore the applications of wearable electrochemical sensors in detecting plant small molecules, enabling scientific assessment of plant conditions, quantification of environmental stresses, and facilitation of plant health monitoring and disease prediction.

Abstract:Using PEDOT as the conductive polymer, an innovative small-scale sensor for directly measuring salicylate ions in plants was developed, which avoided the complicated sample pretreatment of traditional analytical methods and realized the rapid detection of salicylic acid. The results demonstrate that this all-solid-state potentiometric salicylic acid sensor is easy to miniaturize, has a longer lifetime (≥1 month), is more robust, and can be directly used for the detection of salicylate ions in real samples without any additional pretreatment. The developed sensor has a good Nernst slope (63.6 ± 0.7 mV/decade), the linear range is 10 −2 ~ 10 −6 M, and the detection limit can reach (2.8 × 10 −7 M). The selectivity, reproducibility, and stability of the sensor were evaluated. The sensor can perform stable, sensitive, and accurate in situ measurement of salicylic acid in plants, and it is an excellent tool for determining salicylic acid ions in plants in vivo.

Abstract:The detection of phytohormones in real time has attracted increasing attention because of their critical roles in regulating the development and signaling of plants, especially in defense against biotic stresses. Herein, stainless steel sheet electrodes modified with carbon cement were coupled with paper-based analysis devices for direct and simultaneous detection of salicylic acid (SA) and indole-3-acetic acid (IAA) in plants. We demonstrated that the excellent conductivity of stainless steel sheet electrodes enabled us to simultaneously differentiate IAA and SA at a level of 10 nM. With our approach, the content of IAA and SA in Arabidopsis thaliana leaves infected or not infected with Pst DC3000 could be rapidly quantified at the same time. Our experimental results on differentiation of IAA and SA at different time points showed that there were antagonistic interactions between the IAA and SA after infection of Arabidopsis leaves with Pst DC3000. By offering a cost-effective approach for rapid and sensitive detection of IAA and SA, this study suggests that electrochemical detection can be used in the study and development of precision agriculture technology.

Abstract:Salicylic acid (SA) is a biological substance that acts as a phytohormone and plays an important role in signal transduction in plants. It is important to accurately and sensitively detect SA levels. A gold electrode modified with copper nanoparticles was used to assay the electrocatalytic oxidation of salicylic acid. It was found that the electrochemical behavior of salicylic acid was greatly improved at copper nanoparticles, indicating that anodic oxidation could be catalyzed at copper nanoparticles. And the pH had remarkable effect on the electrochemical process, a very well-defined oxidation peak appeared at pH 13.3 (0.2M NaOH). The kinetics parameters of this process were calculated and the heterogeneous electron transfer rate constant (k) was determined to be 1.34×10−3cms−1, and (1−α)nα was 1.22. The gold electrode modified with copper nanoparticles could detect SA at a higher sensitivity than common electrodes. The electrode was used to detect the SA levels in oilseed rape infected with the fungal pathogen Sclerotinia sclerotiorum. The results showed that the SA concentration reached a maximum during the 10th–25th hours after infection. This result was very similar to that determined by HPLC, indicating that the gold electrodes modified with copper nanoparticles could be used as salicylic acid sensors.
Disposable Stainless Steel-Based Electrochemical Microsensor for in Vivo Determination of Indole-3-acetic Acid in Soybean Seedlings.

Abstract:In vivo detecting of plants signal molecules is of great importance for the precision farming, crop management and plant phenotyping. In this work, for in vivo detecting indole-3-acetic acid (IAA), one of phytohormones, fine stainless steel (SS) wire was used as electrode material. Highly ordered nanopores, popcorn-like Au nanostructures, Pt nanoparticles and reduced graphene oxide (ERGO) nanocomposite films, and polymerized ST film (PST) were fabricated on the SS microelectrode in turn for improving the detection effect. Using the as-prepared SS microelectrode as working electrode, two untreated SS wires as reference electrode and counter electrode respectively, a disposable electrochemical microsensor for IAA were developed. The microsensor exhibited excellent selectivity and high sensitivity with low detection limit (LOD) of 43 pg mL-1. The limit of quantity (LOQ) is 143 pg mL-1. The RSD was 7% for 12 different PST/Pt-ERGO/Au/a-SS microsensors in presence of 100 µg mL-1 IAA. Using this microsensor, IAA of the stem of soybean seedlings was detected in vivo under salt stress. Our result was also confirmed by ultra-performance liquid chromatography-mass spectrum (UPLC-MS). This is the first report for the in vivo detection of IAA in plants using SS-based electrochemical microsensor. Our sensor provides an excellent sensing platform for detecting IAA in plants in vivo.
An Activatable NIR‐II Fluorescent Probe for Tracking Heavy‐metal Ion and High‐level Salt‐induced Oxidative Stress in Plant Sprouts

Abstract:Humans and plants have become enfolded and inseparable. Abiotic stresses in particular oxidative stress caused by heavy-metal ions or high-level salt contamination deleteriously impact plants' growth process and have become a major threat to sustaining food security. Sprouting is the first step in plants' growth process. When plant sprouts endure oxidative stress induced by toxic heavy-metal ions or high-level salt, accelerated generation of reactive oxygen species (e.g., H2O2) occurs inside plant sprouts; hence in-situ H2O2 in plant sprouts could serve as the in-vivo biomarker for tracking the oxidative stress in plant sprouts. Herein, we design an activatable probe CT-XA-H2O2 to track the oxidative stress in plant sprouts via in vivo NIR-II fluorescent imaging. In CT-XA-H2O2, cyano-thiazole acts as the electron-accepting moiety and xanthane-aminodiphenyl as the electron-donating moiety, and dioxaborolane as the biomarker-responsive unit and fluorescence quencher. The probe CT-XA-H2O2 shows weak fluorescent emission. When H2O2 is present, the dioxaborolane in the probe is cleaved, consequently, the dye CT-XA-OH is generated and brings about significant fluorescent signals for detecting and imaging the in-situ biomarker. Moreover, the aminodiphenyl group endues the chromophore (the activated probe) with aggregation-induced emission characteristics, which ensures stronger fluorescence in the aggregated state in the aqueous milieu. The probe CT-XA-H2O2 has been employed in the Cd2+-ion or high-level salt (NaCl) induced oxidative stress models of soybean sprouts and peanut sprouts, and the experimental results evidently reveal the probe's ability for in-situ biomarker-activatable in-vivo detection and imaging in the plants' sprouts.

Abstract:Indole-3-acetic acid (IAA) and salicylic acid (SA), as critical plant hormones, are involved in multiple physiological regulatory processes of plants. Simultaneous and continuous in vivo detection of IAA and SA will help clarify the mechanisms of their regulation and crosstalk. First, this study reports the development and application of an electrochemical microsensor for simultaneous and continuous in vivo detection of IAA and SA. This electrochemical microsensor system consisted of a tip (length, 2 mm) of platinum wire (diameter, 0.1 mm) modified with carbon cement and multi-walled carbon nanotubes, an untreated tip (length, 2 mm) of platinum wire (diameter, 0.1 mm), as well as a tip (length, 2 mm) of Ag/AgCl wire (diameter, 0.1 mm). It was capable of detecting IAA in the level ranging from 0.1 to 30 mu M and SA ranging from 0.1 to 50 mu M based on the differential pulse voltammetry or amperometric i-t., respectively. The dynamics of IAA and SA levels in tomato leaf veins under high salinity stress were continuously detected in vivo, and very little damage occurred. Compared to conventional detection methods, the constructed microsensor is not only suitable for continuously detecting IAA and SA in microscopic plant tissue in vivo, it also reduces the damage done to plants during the detection. More importantly, the continuous and dynamic changes in IAA and SA data obtained in stiu through this system not only can help clarify the interaction mechanisms of IAA and SA in plants, it also helps to evaluate the health status of plants, which will promote the development of basic research in botany and precision agriculture.
In vivo surface-enhanced Raman scattering nanosensor for the real-time monitoring of multiple stress signalling molecules in plants

Abstract:When under stress, plants release molecules to activate their defense system. Detecting these stress-related molecules offers the possibility to address stress conditions and prevent the development of diseases. However, detecting endogenous signalling molecules in living plants remains challenging due to low concentrations of these analytes and interference with other compounds; additionally, many methods currently used are invasive and labour-intensive. Here we show a non-destructive surface-enhanced Raman scattering (SERS)-based nanoprobe for the real-time detection of multiple stress-related endogenous molecules in living plants. The nanoprobe, which is placed in the intercellular space, is optically active in the near-infrared region (785 nm) to avoid interferences from plant autofluorescence. It consists of a Si nanosphere surrounded by a corrugated Ag shell modified by a water-soluble cationic polymer poly(diallyldimethylammonium chloride), which can interact with multiple plant signalling molecules. We measure a SERS enhancement factor of 2.9 × 107 and a signal-to-noise ratio of up to 64 with an acquisition time of ~100 ms. To show quantitative multiplex detection, we adopted a binding model to interpret the SERS intensities of two different analytes bound to the SERS hot spot of the nanoprobe. Under either abiotic or biotic stress, our optical nanosensors can successfully monitor salicylic acid, extracellular adenosine triphosphate, cruciferous phytoalexin and glutathione in Nasturtium officinale, Triticum aestivum L. and Hordeum vulgare L.-all stress-related molecules indicating the possible onset of a plant disease. We believe that plasmonic nanosensor platforms can enable the early diagnosis of stress, contributing to a timely disease management of plants.

Abstract:The determination of hormone-binding sites in plants is essential in understanding the mechanisms behind hormone function. Salicylic acid (SA) is an important plant hormone that regulates responses to biotic and abiotic stresses. In order to label SA-binding sites in plant tissues, a quantum dots (QDs) probe functionalized with a SA moiety was successfully synthesized by coupling CdSe QDs capped with 3-mercaptopropionic acid (MPA) to 4-amino-2-hydroxybenzoic acid (PAS), using 1-ethyl-3-(3-dimethyllaminopropyl) carbodiimide (EDC) as the coupling agent. The probe was then characterized by dynamic light scattering and transmission electron microscopy, as well as UV/vis and fluorescence spectrophotometry. The results confirmed the successful conjugation of PAS to CdSe QDs and revealed that the conjugates maintained the properties of the original QDs, with small core diameters and adequate dispersal in solution. The PAS-CdSe QDs were used to detect SA-binding sites in mung bean and Arabidopsis thaliana seedlings in vitro and in vivo. The PAS-CdSe QDs were effectively transported into plant tissues and specifically bound to SA receptors in vivo. In addition, the effects of the PAS-CdSe QDs on cytosolic Ca(2+) levels in the tips of A. thaliana seedlings were investigated. Both SA and PAS-CdSe QDs had similar effects on the trend in cytosolic-free Ca(2+) concentrations, suggesting that the PAS-CdSe QDs maintained the bioactivity of SA. To summarize, PAS-CdSe QDs have high potential as a fluorescent probe for the in vitro/in vivo labeling and imaging of SA receptors in plants.

Abstract:Hydrogen sulfide (H2S) is one of the typical reactive sulfur species, which exhibits an important role in regulating both physiological and pathological processes. Recent studies indicate that H2S also serves as a key signaling molecule in a broad range of regulatory processes in plants. However, in situ imaging and detection of the levels of H2S in plant tissues remains a challenge. In this work, a NIR fluorescent probe (HBTP-H2S) was synthesized to achieve H2S imaging in living plant tissues. HBTP-H2S exhibited high sensitivity toward H2S with a large Stokes shift (250 nm). HBTP-H2S could be applied to HeLa cells to detect the fluctuation of endogenous H2S levels in response to physiological stimulations. Importantly, HBTP-H2S was utilized for direct H2S imaging of rice roots and revealed the upregulation of H2S signaling in response to aluminum ions and flooding stresses. Our work thus provides a new tool to investigate H2S-involved signal interaction and protective resistance of crops under environmental stresses.
Exogenous Salicylic Acid Alleviates Salt Stress by Improving Leaf Photosynthesis and Root System Architecture in Cucumber Seedlings

Abstract:Exogenous salicylic acid (SA) can alleviate salt stress in plants. Previous researches have been focused on SA's role in leaf morphological and physiological characteristics, however, little information is available on its role in both leaf photosynthesis and root system architecture (RSA). A hydroponic study was conducted to analyze biomass, leaf photosynthesis, RSA parameters and expression of RSA genes in cucumber seedlings grown under four treatments, including control (CK), 50 mM NaCl (NaCl), 0.3 mM SA (SA) and 50 mM NaCl plus 0.3 mM SA (NaCl + SA). The results showed that compared with CK, both NaCl and SA treatments severely reduced relative growth rate (RGR) and leaf photosynthetic parameters (F-v/F-m, Phi(II) and Pn). However, compared with NaCl treatment, NaCl + SA treatment significantly increased RGR, leaf photosynthetic (F-v/F-m, Phi(II) and Pn) and RSA (total root length, root surface area, number and total length of primary and secondary LR) parameters. In addition, compared with CK, although both NaCl and SA treatments down-regulated expression of GL2 gene and up-regulated expression of RHD2 gene, NaCl + SA treatment notably up-regulated expression of NAC1, NAC2, GL2, EXP and down-regulated expression of RHD2. It is concluded that exogenous SA alleviated salt stress via effectively promoting leaf photosynthesis and RSA in cucumber seedlings. SA-mediated increment in expression of RSA genes, total length and number of primary and secondary LR could contribute to the improved RSA.

Abstract:In recent years, great concerns have been raised about salicylic acid (SA) and its derivatives as plant regulators. Therefore, precise determination of the distribution of SAs in the living plants is necessary for not only fundamental researches but also the regulating mechanisms. In this study, a custom-made solid phase micro extraction (SPME) fiber based on diallyl dimethyl ammonium chloride-assembled graphene oxide-coated C18 composite (C18@GO@PDDA) was proposed for in vivo detection of salicylic acid, acetylsalicylic acid (ASA), 4 methyl salicylic acid (4-SA) and 3-methyl salicylic acid (3-SA) in aloe plants. Under the optimized conditions, the analytical performance evaluated in homogenized aloe plant tissues exhibited low detection limits (1.8-2.8 mu g g(-1)), wide linear ranges (10-5000 mu g g(-1)), and satisfactory reproducibility (relative standard deviations less than 8.4% and 9.3% for inter-fiber and intra-fiber assays, respectively). Under cadmium stress, the developed method was applied for the in vivo tracing of four salicylic acids in aloe plants. A 48-h in vivo tracing revealed that salicylic acids were involved in the pathway of cadmium stress tolerance. To our best knowledge, it is the first effort to realize the in vivo analysis of SA and its derivatives in plants, and it has a made a great step forward in the area of plant hormone analysis.

Abstract:Background: Indole-3-acetic acid (IAA) and salicylic acid (SA), pivotal regulators in plant growth, are integral to a variety of plant physiological activities. The ongoing and simultaneous monitoring of these hormones in vivo enhances our comprehension of their interactive and regulatory roles. Traditional detection methods, such as liquid chromatography-mass spectrometry, cannot obtain precise and immediate information on IAA and SA due to the complexity of sample processing. In contrast, the electrochemical detection method offers high sensitivity, rapid response times, and compactness, making it well-suited for in vivo or real-time detection applications. Results: A microneedle electrochemical sensor system crafted from disposable stainless steel (SS) wire was specifically designed for the real-time assessment of IAA and SA in plant in situ. This sensor system included a SS wire (100 μm diameter) coated with carbon cement and multi-walled carbon nanotubes, a plain platinum wire (100 μm diameter), and an Ag/AgCl wire (100 μm diameter). Differential pulse voltammetry and amperometry were adopted for detecting SA and IAA within the range of 0.1-20 μM, respectively. This sensor was applied to track IAA and SA fluctuations in tomato leaves during PstDC3000 infection, offering continuous data. Observations indicated an uptick in SA levels following infection, while IAA production was suppressed. The newly developed disposable SS wire-based microneedle electrochemical sensor system is economical, suitable for mass production, and inflicts minimal damage during the monitoring of SA and IAA in plant tissues. Significance: This disposable microneedle electrochemical sensor facilitates in vivo detection of IAA and SA in smaller plant tissues and allows for long-time monitoring of their concentrations, which not only propels research into the regulatory and interaction mechanisms of IAA and SA but also furnishes essential tools for advancing precision agriculture.
Development of a structure-switching aptamer-based nanosensor for salicylic acid detection

Abstract:<p>Salicylic acid (SA) is a phytohormone regulating immune responses against pathogens. SA and its derivatives can be found in diverse food products, medicines, cosmetics and preservatives. While salicylates have potential disease-preventative activity, they can also cause health problems to people who are hypersensitive. The current SA detection methods are costly, labor-intensive and require bulky instruments. In this study, a structure-switching aptamer-based nanopore thin film sensor was developed for cost-effective, rapid, sensitive and simple detection of SA in both buffer and plant extracts. SA is a challenging target for aptamer selection using conventional systemic evolution of ligands by exponential enrichment (SELEX) due to its small size and scarcity of reactive groups for immobilization. By immobilizing the SELEX library instead of SA and screening the library using a structure-switching SELEX approach, a high affinity SA aptamer was identified. The nanopore thin film sensor platform can detect as low as 0.1 μM SA. This is much better than the sensitivity of antibody-based detection method. This nanosensor also exhibited good selectivity among SA and its common metabolites and can detect SA in <em>Arabidopsis</em> and rice using only about 1 μl plant extracts within less than 30 min. The integration of SA aptamer and nanopore thin film sensor provides a promising solution for low-cost, rapid, sensitive on-site detection of SA.</p>

Abstract:Monitoring plant physiological information for gaining a comprehensive understanding of plant growth and stress responses contributes to safeguarding plant health. Light-emitting probes - in terms of small-molecule, nanomaterials-based, and genetically protein-based probes - can be introduced into plants through foliar and root treatment or genetic transformation. These probes offer exciting opportunities for sensitive and in situ monitoring of dynamic plant chemical information - for example, reactive oxygen species (ROS), calcium ions, phytohormones - with spatiotemporal resolution. In this review we explore the sensing mechanisms of these light-emitting probes and their applications in monitoring various chemical information in plants in situ. These probes can be used as part of a sentinel plant approach to provide stress warning in the field or to explore plant signaling pathways.

Abstract:Salt stress is one of the most common factors limiting plant cultivation. In this study, metabolic responses to salt stress in Arabidopsis thaliana (A. thaliana) leaves were analyzed in situ by neutral desorption-extractive electrospray ionization mass spectrometry (ND-EESI-MS) without any sample pretreatment. Metabolic changes of A. thaliana leaves were observed in response to salt stress conditions, including the levels of serine, glutamic acid, arginine, cinnamic acid, ferulic acid, caffeic acid, protocatechuic acid, epicatechin, morin, myricetin, apigravin, and β-cotonefuran. The content of serine increased under 50, 100, and 200 mM NaCl salt stress, reaching the highest level at 200 mM NaCl, but decreased under the maximum concentration of 300 mM NaCl. A similar phenomenon was observed for arginine, glutamic acid, cinnamic acid, caffeic acid, ferulic acid, and epicatechin, respectively, involved in the metabolic pathway of shikimate-phenylpropanoid. Both principal component analysis (PCA) and partial least-squares discrimination analysis (PLS-DA) showed that the salt stress treatment groups of the higher concentrations (200 and 300 mM) could be well distinguished from those of the lower concentrations (50 and 100 mM) and the control. Marker metabolites, like m/z 261 (apigravin) and m/z 305 (β-cotonefuran), were assistantly selected from the fingerprints by variable importance for the projection (VIP). Our results indicated the potential of the ND-EESI-MS method for the rapid recognition of metabolic conditions in plant leaves under salt stress.
