Abstract:Rh(III)-catalyzed C(sp(3)) - H bond aminocarbonylation of 8-methylquinolines and isocyanates has been realized under mild conditions. This approach is applicable to different aryl and alkyl isocyanates, leading to the synthesis of various alpha-quinolinyl amide compounds in moderate to excellent yields. A plausible mechanism for this transformation is proposed according to the experimental results obtained.

Abstract:A cobalt(III)-catalyzed C-H/N-H bond functionalization for the synthesis of 1-aminoisoquinolines from aryl amidines and diazo compounds has been developed. The reaction proceeds under mild reaction conditions, obviates the need for oxidants, produces only N2 and H2O as the byproducts, and features a broad substrate scope.

Abstract:Catalytic asymmetric 1,3-dipolar cycloaddition reactions of activated isocyanides with various 2 pi dipolarophiles have been intensively studied, affording a wide range of enantioenriched five-membered N-heterocycles. In sharp contrast, the catalytic enantioselective higher-order cycloaddition of activated isocyanides has not been achieved yet. We present here our recent work on the development of an unprecedented silver-catalyzed highly diastereo- and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. This method features high efficiency, good to excellent stereocontrol, wide substrate scope, as well as operational simplicity. It is also noteworthy that the same catalytic system was proved to be suitable for not only the late-stage functionalization of complex bioactive molecules but also the kinetic resolution of racemic azomethine imines.

Abstract:In this study, Cp*Co(iii)-catalyzed site-selective amidation of pyridones and isoquinolones using oxazolones as the amidation reagent is reported. This approach features mild conditions, high efficiency and good functional tolerance. Furthermore, gram-scale preparation and preliminary mechanism experiments were carried out. It provides a straightforward approach for the direct modification of pyridone derivatives.

Abstract:An interesting approach to functionalized imidazolidines is described. These compounds are obtained in a copper(I)-catalyzed domino three-component coupling and cyclization reaction involving two formaldehyde-derived imine units and a terminal alkyne. Alternatively, imidazolidines can be obtained from propargylamines and formaldehyde-derived imines. This strategy provides a straightforward and atom-economic pathway to construct imidazolidines with high yields and benefits from readily available starting materials, convenient one-pot operations.

Abstract:An efficient and straightforward procedure toward the synthesis of amides-substituted 2H-benzopyrans H-benzopyrans has been explored through palladium-catalyzed one-pot aminocarbonylation reaction. By using nitro compounds as attractive nitrogen sources, with Mo(CO)6 6 as both CO source and reductant, a variety of amides-containing 2H- H- benzopyrans were obtained in moderate to high yields with good functional group tolerance. This protocol has advantages including atom-economy, no manipulation of toxic CO gas, and broad substrate scope.

Abstract:Transition metal catalyzed C-H bond activation have become one of the important tools for constructing new chemical bonds. Introducing directing group to the substrates are the key to successful reaction, these directing groups can also be further transformed in the reaction. As amidines with unique structure and reactivity, makes them ideal substrates for transition metal-catalyzed C-H transformations. This review describes the major progress and mechanistic investigations on C-H activation/annulation tandem reaction of amidines until early 2024, focusing on metal-catalyzed C-H activation of amidines with unsaturated compounds, such as alkynes, olefins vinylene carbonate, cyclopropanols and their derivatives. Meanwhile this manuscript also with a view to the reaction of amidines with diffierent carbene precursor, for example diazo compounds, azide, triazoles, pyriodotriazoles, sulfoxonium ylides. And its own C-H bond activation/cyclization reaction. A bright outlook was provided at the end of the manuscript.

Abstract:An iminopyridine cobalt dichloride complex was synthesized and demonstrated as an effective precatalyst for hydrosilylation/cyclization of 1,6-enynes with silanes. Various functional groups such as amine, free aniline, ester, ether, cyano, halide, trifluoromethyl, and heterocycle were tolerated to afford a variety of silicon-containing compounds. The reaction could be scaled up to afford products on the gram scale which could undergo further derivatizations. A primary mechanism was proposed based on analysis of side products and a deuterated experiment.

Abstract:Catalytic methods to couple alkene and amine feedstocks are valuable in synthetic chemistry. The direct carbonylative coupling of alkenes and amines holds promise as a perfectly atom-economical approach to amide synthesis, but general methods remain underdeveloped. Herein, we report an alkene hydroaminocarbonylation catalyzed by unmodified, inexpensive cobalt carbonyl under mild conditions and low pressure promoted by light. Silane addition after the reaction enables sequential cobalt-catalyzed amide reduction, constituting a formal alkene hydroaminomethylation. These methods exhibit exceptional scope across both alkene and amine components with high chemo- and regioselectivity and proceed efficiently even in the absence of solvent. The formation of a hydridocobalt through photodissociation of a carbonyl ligand is proposed to enable catalytic activity under mild conditions, which addresses a long-standing challenge in catalysis.

Abstract:C-H/N-O functionalizations by cobalt(III) catalysis allowed the expedient synthesis of a broad range of isoquinolines. Thus, internal and challenging terminal alkynes proved to be viable substrates for an isohypsic annulation, which was shown to proceed by a facile C-H cobaltation.

Abstract:Herein, we report a simple method for the synthesis of 3-benzazepine derivatives via a cobalt-catalyzed, picolinamide-directed α-amidoacrylate C(sp2)-H bond functionalization approach. The reactions utilize calcium carbide as an inexpensive, easy-to-handle, and solid acetylene source, and simple CoCl(2) as the reaction catalyst. Excellent functional group tolerance is observed, yielding diverse substituted 3-benzazepine derivatives in good to excellent yields.

Abstract:The electrochemical incorporation of carbon dioxide into amines catalyzed by an electrogenerated Ni complex afforded carbamates in moderate yields under very mild conditions (p(CO2) = 1 atm, room temperature) without any addition of probases. Mechanistic and electrochemical studies revealed the role of reduced nickel species in the activation of CO2 and electrogenerated CO2 center dot- as a base in the synthesis of carbamates. The influence of number of faradays per mole of amine supplied to the electrodes (Q), cathode materials, temperatures, supporting salts and amounts of catalyst was studied to optimize the electrolytic conditions. Abstract:A procedure for the [Cp*CoIII]-catalyzed, 1,2,3-triazole directed C-H amidation of arenes, using dioxazolone as an efficient amidating reagent, has been developed. This reaction provides a step-and atomeconomical protocol for C-N bond formation, releasing carbon dioxide as a single byproduct. Yet a variety of functional groups are well tolerated, obtaining the desired products in moderate to excellent yields.

Abstract:Highly chemoselective intramolecular amination of propargylic C(sp(3))-H bonds has been demonstrated for N-bishomopropargylic sulfamoyl azides through cobalt(II)-based metalloradical catalysis. Supported by D(2h)-symmetric amidoporphyrin ligand 3,5-Di(t)Bu-IbuPhyrin, the cobalt(II)-catalyzed C-H amination proceeds effectively under neutral and nonoxidative conditions without the need of any additives, and generates N2 as the only byproduct. The metalloradical amination is suitable for both secondary and tertiary propargylic C-H substrates with an unusually high degree of functional-group tolerance, thus providing a direct method for high-yielding synthesis of functionalized propargylamine derivatives.

Abstract:It is of great significance to the efficient and convenient synthesis of amides as their great application in medicinal chemistry. However, some problems still exist in the preparation of amides, such as atom -eco-nomic. Here, a new and efficient method to prepare amides by the carbonylation of amines was devel-oped without CO gas and palladium catalyst. Mo(CO)6 was employed as carbonyl source, different amines could tolerate the reaction conditions, and the corresponding amides were obtained with good to excellent yields. Abstract:α-Tertiary amides are of great importance for medicinal chemistry. However, they are often challenging to access through conventional methods due to reactivity and chemoselectivity issues. Here we report a single-step approach towards such amides via cobalt-catalyzed intermolecular oxidative hydroamidation of unactivated alkenes, using nitriles of either solvent- or reagent-quantities. This protocol is orthogonal to groups that rapidly react under known carbocation amidation conditions such as tertiary alcohols, electron-rich alkenes, ketals, weak C−H bonds, and carboxylic acids. Straightforward access to a diverse array of hindered amides is demonstrated, including a rapid synthesis of an aminoadamantane-derived pharmaceutical intermediate.

Abstract:A cationic cobalt(III)-catalyzed direct C-H amidation of unactivated (hetero)arenes and alkenes by using 1,4,2-dioxazol-5-ones as the amidating reagent has been developed. This transformation proceeds efficiently under external oxidant-free conditions with a broad substrate scope. Moreover, 6-arylpurine compounds, which often exhibit high potency in antimycobacterial, cytostatic, and anti-HCV activities, can be smoothly amidated, thus offering a mild protocol for their late stage functionalization.

Abstract:Molecules with an allylic amine motif provide access to important building blocks and versatile applications of biologically relevant chemical space. The need for diverse allylic amines requires the development of increasingly general and modular multicomponent reactions for allylic amine synthesis. Herein, we report an efficient catalytic multicomponent coupling reaction of simple alkenes, aldehydes, and amides by combining nickel catalysis and Lewis acid catalysis, thus providing a practical, environmentally friendly, and modular protocol to build architecturally complex and functionally diverse allylic amines in a single step. The method is remarkably simple, shows broad functional-group tolerance, and facilitates the synthesis of drug-like allylic amines that are not readily accessible by other methods. The utilization of accessible starting materials and inexpensive Ni(II) salt as the alternative precatalyst offers a significant practical advantage. In addition, the practicality of the process was also demonstrated in an efficient, gram-scale preparation of the prostaglandin agonist.
