Abstract:Biocatalytic reactions in living cells involve complex transformations in the spatially confined microenvironments. Inspired by biological transformation processes, we demonstrate effective biocatalytic cascade driven photodynamic therapy in tumor-bearing mice by the integration of an artificial enzyme (ultrasmall Au nanoparticles) with upconversion nanoparticles (NaYF4@NaYb0.92F4:Er0.08@NaYF4)zirconium/iron porphyrin metal-organic framework core-shell nanoparticles (UMOF NPs) which act as biocatalysts and nanoreactors. The construction of core-shell UMOF NPs are realized by using a unique "solvent-assisted self-assembly" method. The integration of ultrasmall AuNPs on the UMOFs matrix leads to glucose depletion, providing Au-mediated cancer therapy via glucose oxidase like catalytic activity. Meanwhile, the UMOF matrix acts as a near-infrared (NIR) light photon-activated singlet oxygen generator through a continuous supply of oxygen via hydrogen peroxide decomposition upon irradiation. Such kinds of biocatalysts offer exciting opportunities for biomedical, catalytical ,and energy applications.

Abstract:Porphyrins are a class of macromolecular heterocyclic compounds formed by the inter-carbon atoms of four pyrrole-like subunits through the submethyl bridge (〓CH▬). Porphyrin rings have 26 electrons in highly conjugated system and are easily modified peripheral structures, often serve as ideal building blocks to construct self-assembled nanostructures with excellent physical and chemical properties. Porphyrin nanostructures have excellent visible light absorption properties, which will significantly improve the efficiency of electron–hole separation, and are also commonly used in photocatalysis fields. Porphyrin photosensitizers have superior strong phototoxicity and little side effects, and are widely used in tumor photothermal/photodynamic treatment. This chapter summarizes the self-assembly methods of porphyrins, the applications progress of porphyrin self-assembled nanomaterials in photocatalysis and tumor therapy, and discusses the development trend in future of porphyrin nanomaterials.

Abstract:Porphyrins are cyclic molecules with large π-conjugated structure. They can form ordered nanostructures through controllable self-assembly via multiple non-covalent interactions, to regulate their photoelectric properties effectively. However, it is still a great challenge to accurately regulate the self-assembly process to obtain self-assembled nanostructures with controllable morphology, uniform size and ordered internal structure, and further to clarify the key factors controlling the performance. This paper reviews the development of solution phase self-assembly strategy of porphyrin self-assemblies in recent years and its applications in the fields of photocatalysis, chemical sensor and biological phototherapy, and preliminarily summarizes the structure-performance relationship between the morphology, size and internal structure of porphyrin self-assembled nanostructures and related applications.

Abstract:The development of intelligent nano-platforms with high tumor-targeting capability, enhanced solubility, and efficacious photochemotherapy is urgently demanded for clinical anticancer treatments. Herein, we report an amphiphilic porphyrin ( P ) to complex with camptothecin ( CPT ) to form supra-amphiphile-assembled nanoparticles (). The host–guest complexation between P and CPT promotes the singlet oxygen production of P and enhances the solubility of CPT , which should be beneficial to improve PDT efficiency and chemotherapy effect. Cancer cell membrane ( M ) is further used to camouflage , which endows the resulting "M with excellent stability, homologous targeting ability, and specific co-delivering of the chemotherapeutic drug and photosensitizer to tumor cells. The presented work takes advantage of the unique structure and intrinsic performance of P to construct an integrative co-delivery system with the assistance of membrane-coating technology, which not only extends the application scopes of porphyrin photosensitizers but also provides a promising nanoplatform for cancer therapy.

Abstract:Self-assembly in subcellular organs to regulate their metabolic processes for regulated cell death become one of increasingly significant topics but remains a formidable challenge in chemical and biological science. Herein, an in situ self-assembly system in mitochondria of cancer cell was successfully constructed by perylene diimide derivative (PDI-NH2), which could self-assemble into nanoparticles with multi-negativlely charged molecules in the mitochondria of cancer cells, resulting in metabolic dysfunction for cancer cell ferroptosis. Due to the directive interaction between mitochondria and in situ formed nanoparticles, the mitochondrial function would be damaged, impairing the cellular metabolism systems and iron homeostasis, which ultimately led to elevated ROS level, lipid peroxidation as well as the down regulation of GPX4 protein for cancer cell ferroptosis. Finally, this self-assembly process could also be realized in vivo to suppress the growth of tumors. This work reveals that in situ self-assembly in subcellular organelles could interfere the metabolic process for regulating the cellular behavior, providing strong support for supramolecular self-assembly in bio-application.
