Abstract:Immunobiosensors were developed for detection of Escherichia coli O157:H7 based on the surface immobilization of monoclone antibodies onto indium tin oxide (ITO) electrodes. The immobilization of antibodies onto ITO chips was carried out by silanization. The effects of epoxysilane monolayer, the antibody layer on the electrochemical properties of the electrode, and the combined target bacteria were analyzed through cyclic voltammetry and electrochemical impedance spectroscopy. By using Randles model as the equivalent circuit, the concentration of the target bacteria can be quantitatively analyzed in terms of the change of electron transfer resistance. The biosensor could detect the target bacteria with a detection limit of 4times103 CFU/mL. A linear response was found between 4times1034times106 CFU/mL. This biosensor was characterized with high sensitivity, excellent selectivity, short detection time and easy operation. It has a promising application in clinical laboratory diagnoses, environmental detection and food safety

Abstract:In recent years, quartz crystal microbalance (QCM) plays an important role in many biological detections, and the enhancement of a single strain DNA probe linked to the sensor surface is of crucial importance in DNA molecule recognition. By means of nanogold modification and mass amplification, a QCM biosensor for Escherichia coli O157:H7 DNA detection was explored and developed in this work. As a suitable surfactant, 1,6-Hexanedithiol was introduced onto the Au surface of QCM, then self-assembled to provide a thioled interface for the inner goldnano immobilization. Based on the Au-SH binding mechanism, thiolated single-stranded DNA (ssDNA) probes which was specific to E. coli O157:H7 eaeA gene would link to the nanoparticle-modified electrode surface. The hybridization was induced by exposing the ssDNA probe to the complementary target DNA, but the frequency change caused by the captured target DNA was negligible. In order to amplify the signal, outer nanogold as the mass enhancer were applied to combine with the target DNA by the biotin-avidin system. The electrochemical techniques, cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS), are adopted to manifest and character each step, which supplied some reasonable and powerful supports for the frication and detection of this QCM biosensor. The target DNA corresponding to 2.0 times 103 colony forming unit (CFU)/ml E. coli O157:H7 can be detected by this biosensor, so it is practical to develop a sensitive and effective QCM biosensor for pathogenic bacteria detection based on specific DNA analysis. In order to attract more attention, this DNA biosensor should be combined with some micro- and nano-fabrication techniques to realize more promising and pratical applications, and more attention should be focused on the further improvement of the sensitivity and the shorten of analysis time.

Abstract:Sensitive detection of endotoxin is especially critical in the fields of food security, therapeutics and medical science. In this work, a new sandwich-type electrochemical aptasensor was designed for ultrasensitive detection of endotoxin. For the first time, nitrogen-doped carbon nanotubes (N-CNTs) was functionalized with polydiallyldimethylammonium chloride (PDDA), forming P-N-CNTs nanocomposite. Then, Fe-based metal–organic framework (Fe-MOF) and silver nanoparticles (AgNPs) was decorated with P-N-CNTs to form the MOF/Ag-P-N-CNTs nanohybrid, which was employed as an enhanced signal indicator, not only exhibited outstanding electrochemical activity, but also could directly label with thiol-modified signal probes (TSPs) to form tracer label. Additionally, the avidin–biotin recognition system was introduced to increase the amount of biotin-modified capture probes (BCPs). Differential pulse voltammetry (DPV) was utilized to investigate the detection performances of the proposed aptasensor, which suggested a wide linear range from 1 fg/mL to 100 ng/mL with a low detection limit of 0.55 fg/mL toward endotoxin.

Abstract:Endotoxin in the blood can cause unexplained fever, and even death due to endotoxemia and bacteremia. Rapid and sensitive detection of endotoxin has become a priority event to intervene in the occurrence of dangerous diseases in time. In this context, a carbon nanotubes-based field-effect transistor (CNTs FET) nanosensor is developed to realize the rapid, label-free and sensitive detection of endotoxin. The CNTs FET was fabricated by assembling the polymer-sorted high-purity semiconductor CNT films onto the sensing channel. In order to improve the detection sensitivity, carboxylated graphene quantum dots (cGQDs) were coupled to the CNT surface via the poly- l -lysine (PLL). After that, polymyxin B (PMB), which is highly specific for endotoxin, was covalently combined with cGQDs, thus enabling the capture and detection of endotoxin. The method not only displayed an extremely low level of limitation of detection in PBS (4.6 fg/mL) and serum (30.3 fg/mL), respectively, with superior resistance to interference, but also enabled the analysis of Gram-negative bacterial infections in blood samples in a short duration of time. Meanwhile, the receiver operating characteristic curve (ROC) proved the superior diagnostic accuracy of this method (AUC = 0.990). Considering its excellent performance, the constructed CNTs FET biosensor is a promising tool to provide early warning of disease for clinical Gram-negative bacteremia and endotoxemia.

Abstract:Bacterial endotoxin (a type of lipopolysaccharide, LPS) that acts as the strongest immune stimulant exhibits high toxicity to human health. The golden standard detection methods rely heavily on the use of a large amount of tachypleus amebocyte lysate (TAL) reagents, extracted from the unique blue blood of legally protected horseshoe crabs. Herein, a cost-effective distance-based lateral flow (D-LAF) sensor is demonstrated for the first time based on the coagulation cascade process of TAL induced by endotoxin, which causes the generation of gel-state TAL. The gelation process can increase the amount of trapped water molecules and shorten the lateral flow distance of the remaining free water on the pH paper. The water flow distance is directly correlated to the concentration of endotoxin. Noteworthy, the D-LAF sensor allows the detection of endotoxin with the reduced dosage of TAL reagents than the golden standard detection methods. The detection limit of endotoxin is calculated to be 0.0742 EU/mL. This method can be applied to the detection of endotoxin in real samples such as household water and clinical injection solution with excellent performance comparable to the commercial ELISA kit.

Abstract:An impedimetric aptasensor for the detection of endotoxin in a microfluidic chip was proposed, in which the Apt/AuNPs/SPCE sensing surface was fabricated in a screen-printed electrode with good biological activity and stability. The quantitative detection of endotoxin was accomplished by electrochemical impedance spectroscopy (EIS) measurement before and after exposing to samples. The impedance biosensor offers an ultrasensitive and selective detection of endotoxin down to 500 pg mL-1 with a wide linear range from 500 pg mL-1 to 200 ng mL-1. According to the Langmuir isotherm model, the interactions between the target molecules and the sensing surface had been analyzed and strong binding was concluded. Compared to the traditional static incubation methods, the microfluidic biosensor realizes the enrichment of endotoxin owing to the confined space and continuous flow nature, so that the lowest detection concentration is reduced from 5 ng mL-1 to 500 pg mL-1, which is much lower than the existing technology, and the total assay time is shortened from 1.0 h to 0.5 h. The proposed microfluidic impedance biosensor provides a new strategy for the design of an aptasensor to realize the rapid detection of target biomolecules with high sensitivity and it can be integrated into wearable medical devices due to its flexible properties.

Abstract:Shiga toxin type II (Stx2), the major virulence component of enterohemorrhagic Escherichia coli , is strongly associated with the life-threatening hemolytic uremic syndrome thus posing a substantial risk to food safety and human health. In this work, a dual-mode aptasensor with colorimetric and surface-enhanced Raman scattering was developed for Stx2 specific detection based on noble metal nanoparticles and Raman reporter loaded metal-organic framework (Mn/Fe-MIL(53)@AuNSs-MBA). The Mn/Fe-MIL(53)@AuNSs could catalyze the H 2 O 2 -mediated oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB), thereby enabling visual detection. Meanwhile, the SERS signal from MBA can be enhanced by the decorated AuNSs. Under optimal conditions, a linear range of 0.05–500 ng/mL with limit of detection (LOD) of 26 pg/mL was achieved in colorimetric mode and a linear range of 5–1000 ng/mL with LOD of 0.82 ng/mL in SERS mode, in which the dual-mode results complement each other, widening the linear range, increasing the accuracy and reliability of the detection. The method was further applied to the detection of Stx2 in milk with average recovery of 101.1 %, demonstrating its superior potential for bacterial toxin monitoring.

Abstract:Endotoxin is a deadly pyrogen, rendering it crucial to monitor with high accuracy and efficiency. However, current endotoxin detection relies on multistep processes that are labor-intensive, time-consuming, and unsustainable. Here, we report an aptamer-based biosensor for the real-time optical detection of endotoxin. The endotoxin sensor exploits the distance-dependent scattering of gold nanoparticles (AuNPs) coupled to a gold nanofilm. This is enabled by the conformational changes of an endotoxin-specific aptamer upon target binding. The sensor can be used in an ensemble mode and single-particle mode under dark-field illumination. In the ensemble mode, the sensor is coupled with a microspectrometer and exhibits high specificity, reliability (i.e., linear concentration to signal profile in logarithmic scale), and reusability for repeated endotoxin measurements. Individual endotoxins can be detected by monitoring the color of single AuNPs via a color camera, achieving single-molecule resolution. This platform can potentially advance endotoxin detection to safeguard medical, food, and pharmaceutical products.

Abstract:Endotoxin can lead to irreversible shock and death, underlying the necessity for development of facile, rapid, sensitive and in situ methods to detect endotoxin. We used electrochemical impedance spectroscopy (EIS) to detect interaction of endotoxin with polymyxin B (PmB) immobilized on 4,4-dithiodibutyric acid coated gold electrodes. An equivalent circuit model with a constant phase element was used to interpret the obtained impedance spectra. The results indicated that the gold electrode after electrochemical polishing pretreatment in both 2-(N-Morpholino) ethanesulfonic acid (MES) and HClO4 solutions had a more effective surface state than that treated in H2SO4 solution. The changes in capacitance associated with a constant phase element on the PmB-modified electrodes were more sensitive, compared to those in the charge-transfer resistance, for detecting endotoxin over the 0.2–0.8ng/mL concentrations. The activated and modified gold electrodes might be used to EIS detect endotoxin in real-time.

Abstract:In both our folk psychology and mental health practice, we adhere to the modernist view that a crisis is something that an individual has. A crisis is either something that happens to people or which people bring on themselves because of character pathology. On the other hand, postmodern theorists now draw our attention to how processes of interaction provide opportunities for certain characterizations to emerge and disipate. Both the conceptualization of an event as a crisis and the scenarios of how it is best understood arise within a community and emerge out of the communal construction of meaning. In this process in Oklahoma City, bystanders, politicians, pilgrims and news media played a role of great significance. Whether local or from afar, they became a community of observers who interacted with one another and looked to one another for verification as to what counted as a crisis and what behavior was appropriate. Lindy has observed that traumatized individuals are often surrounded quickly by a small network of trusted people who serve to buffer and protect them and who define what is helpful and what constitutes further trauma. Here, in Oklahoma, however, a wider public defined the event as a crisis, elaborated on it, and have gradually transformed it into almost mythic proportions, a process now exemplified in the construction of the Oklahoma City National Monument. However, they have also endorsed diverse other scenarios, not all of which have facilitated appropriate grieving, repair of life-disruption, or moving on. All, however, seem to have given a sense of meaning to the event. In this rich matrix, some survivors and their families have felt helped. Others have felt exploited. Still others have wanted to forget the whole thing and to avoid anything that reminded them of it. Of the would-be helpers, some felt unjustly rejected and unappreciated. Others felt appreciated, useful, and even enriched. However, a true legacy of the bombing may well be the metalogue it can precipitate and advance about the relationship between individuals and society, and especially the relationship between our individual life-stories and larger public scenarios, and the effects these scenarios have on the questions we can ask, what we can perceive and what solutions we can create.
A dual-mode composite nanozyme-based cascade colorimetric-fluorescence aptasensor for Salmonella Typhimurium detection

Abstract:Background Salmonella Typhimurium poses a serious threat to human health worldwide, necessitating the development of a rapid, sensitive, and convenient method for S . Typhimurium detection. Nanozymes are considered ideal signal report elements, which are extensively used for developing colorimetric methods. However, single-component nanozymes display low catalytic activity, and colorimetric methods are susceptible to environmental interference, reducing the sensitivity and accuracy of detection results. To address these drawbacks, this study constructed a dual-mode composite nanozyme-based cascade colorimetric-fluorescence aptasensor for S . Typhimurium detection in food. Results In this study, the composite Fe 3 O 4 @MIL-100(Fe) nanozymes were successful synthesized and demonstrated substantial peroxide-like activity, with 4.76-fold higher specificity activity (SA) than that of Fe 3 O 4 nanozymes. Then, a glucose oxidase (GOx)-Fe 3 O 4 @MIL-100(Fe) cascade reaction was developed for colorimetric detection via an aptamer to facilitate the formation of Fe 3 O 4 @MIL-100(Fe)/ S . Typhimurium/carboxylated g-C 3 N 4 (CCN)-GOx sandwich complexes. Meanwhile, the fluorescence mode was achieved by measuring the fluorescence intensity of the sandwich complexes. In optimum conditions, the dual-mode detection limits (LOD) were 1.8 CFU/mL (colorimetric mode) and 1.2 CFU/mL (fluorescence mode), respectively, with the S . Typhimurium concentration ranging from 10 CFU/mL to 10 7 CFU/mL. Finally, the feasibility of the dual-mode colorimetric-fluorescence method was validated via three actual samples, yielding recovery rates of 77.32 % to91.17 % and 82.17 % to 103.7 %, respectively. Significance and novelty This study successfully develops a composite nanozyme-based cascade colorimetric and fluorescence dual-mode aptasensor for S . Typhimurium detection. It presents several distinct benefits, such as a broader linear range (10–10 7 CFU/mL), a lower LOD value (1.2 CFU/mL), and more accurate results. More importantly, the proposed dual-mode method displays a low LOD in colorimetric mode, demonstrating considerable potential for S . Typhimurium on-site detection in food.

Abstract:The release of lipopolysaccharide (LPS) during the proliferation and death of bacterial growth could cause acute versatile disorders like anaphylaxis and multiple organ failure. Analysis methods with high sensitivity and selectivity are expected to be developed to replace the limulus lysate test. Classical colorimetric method was equipped with portable visualization but limited with low sensitivity, while incredibly sensitive fluorescence sensing strategy suffered from negligible selectivity. Delicate design of the dual channels analysis would make the ends of such dilemma based on a fluorescent nanozyme of iron doped carbon quantum dots (Fe-CQDs) with both excellent fluorescence and peroxidase-mimic catalytic activity. Accordingly, a complementary dual channel analytical platform of LPS was prepared. The excellent peroxidase-like activity imparted the dominant selectivity for LPS (available under 6 interferences), while the powerful fluorescence property endowed the excellent sensitivity with the limit of detection (LOD) of an unprecedented level (0.0023 EU/mL). Our study has broadened a new path for design of efficient analytical methods via complement way.

Abstract:An electrochemical biosensor that used an aptamer as a biological element was constructed to detect endotoxin. Biolayer interferometry was used to obtain the affinity constant of an aptamer for lipopolysaccharide, which had an equilibrium dissociation constant of 22.9 nM. The amine-terminated aptamer was then assembled on a gold electrode surface using 3-mercaptopropionic acid as an intermediate linker. The modification of the gold electrode was confirmed by cyclic voltammetry and electrochemical impedance spectroscopy. In the range of 0.001-1 EU/mL, the increase in electron transfer resistance of the biosensor was linear with the logarithmic value of the endotoxin concentration. The constructed biosensor exhibits sensitivity and a low limit of detection.

Abstract:Bacterial contamination is a serious issue for public health and food safety. In this work, a simple and label-free fluorescence detection nanoplatform for Escherichia coli ( E. coli ) was established on the basis of the competitive relationship for the reduction of Cu 2+ in CuS-BSA between E. coli and O -phenylenediamine (OPD). OPD could be directly oxidized by Cu 2+ to produce 2,3-diaminophenazine (ox OPD) with fluorescence properties. When OPD was introduced into an aqueous solution containing CuS-BSA and E. coli , the oxidation of OPD was inhibited owing to the reduction of Cu 2+ to Cu + /Cu 0 by NADH-2 dehydrogenase in the bacterial copper homeostasis mechanism, thus decreasing the fluorescence response signal of the system. Meanwhile, our strategy exhibited a satisfactory performance with a broad linear response to E. coli ranging from 12 to 1.2 × 10 7 CFU mL −1 , and the limit of detection was 9 CFU mL −1 . The practicability of the developed fluorescence biosensing platform in real samples was evaluated by successful determination of E. coli in drinking water and orange juice. These findings provide a new sensing strategy for analyzing other foodborne bacteria and ensuring food safety assessment.

Abstract:The emergence of nanoenzymes has catalyzed the robust advancement of the lateral flow immunoassay (LFIA) in recent years. Among them, multifunctional nanocomposite enzymes with core-shell architectures are considered preferable for promoting the sensing ability due to their good biocompatibility, precise control over size, and surface properties etc. Herein, we developed a dual-channel ensured lateral flow immunoassay (DFLIA) platform utilizing a magnetic, colorimetric, and catalytic multifunctional nanocomposite enzyme (Fe3O4@TCPP@Pd) [TCPP, Tetrakis (4-carboxyphenyl) porphyrin] for the ultrasensitive and highly accurate rapid detection of Escherichia coli O157:H7 (E. coli O157:H7). Fe3O4@TCPP@Pd-mAb exhibits superior performance compared to traditional AuNPs, including enhanced sensitivity and an extended linear detection range, benefiting from its high brightness signal, strong magnetic separation ability, and high peroxidase activity (Vmax = 2.32 μM S1-). Moreover, the Fe3O4@TCPP@Pd-labeled mAb probe exhibited exceptional stability and high affinity toward E. coli O157:H7 (with an affinity constant of approximately 1.723 × 109 M-1), indicating its potential for the efficient capture of the pathogen. Impressively, the developed Fe3O4@TCPP@Pd-DFLIA achieved ultrasensitive detection for E. coli O157:H7 with pre- and postcatalytic naked-eye detection sensitivities of 255 cfu/mL and 77 cfu/mL, respectively, representing an approximately 41-fold improvement over the conventional AuNP-based LFIA and also possessed good specificity and reproducibility [relative standard deviation (RSD) < 10%]. Additionally, the established DFLIA exhibited satisfactory recoveries in detecting pork and milk samples, further validating the reliability of this platform for immunoassays and demonstrating its potential for utilization in bioassays and clinical diagnostics.

Abstract:Due to their superparamagnetism and enzyme-like activity, iron oxide (Fe3O4) nanozymes can be readily used for sample pretreatment and the generation of detection signals, and have, thus, attracted much attention in the field of bioanalysis and diagnosis. However, the low catalytic activity of Fe3O4 nanozymes does reduce the sensitivity of Fe3O4-based methods, limiting their application. In this study, Fe3O4@Cu@poly(pyrrole-2-carboxylic acid) yolk-shell nanozymes (Fe3O4@Cu@PCPy YSNs) were synthesized using a facile approach and selective chemical etching technology. Compared with Fe3O4 nanozymes, the Fe3O4@Cu@PCPy YSNs demonstrated a three-fold increase in the peroxidase-like activity, good dispersity and strong superparamagnetism. In addition, the flower-shaped structure of aptamer-complementary strand (Apt-CS) conjugates was designed on the surface of the Fe3O4@Cu@PCPy YSNs, which effectively inhibited their peroxidase-like activity by creating a physical barrier that hindered the access of substrates to the center of the Fe3O4@Cu@PCPy YSNs. Based on this principle, a robust and facile colorimetric aptasensor was developed for detecting Salmonella Typhimurium. The flower-shaped Apt-CS were dissociated in the presence of S. Typhimurium, promoting the recovery of Fe3O4@Cu@PCPy YSN catalytic activity. Under optimized conditions, this proposed aptasensor successfully detected S. Typhimurium in a linear range of 3 to 3 × 106 CFU/mL, achieving a detection limit of 1 CFU/mL. Finally, the feasibility of this novel aptasensor was further validated by three actual samples, with recoveries of between 84.3% and 102%, thereby demonstrating the huge potential of the proposed aptasensor for detecting S. Typhimurium in foods.

Abstract:A novel non-enzyme electrochemical biosensor for the rapid detection of Gram-positive bacteria has been constructed that relys on a stable and efficient combination between the peptidoglycan layer and platinum-nickel-copper nanocubes (Pt-Ni-Cu NCs). Briefly, bacteria were first captured by a specific antibody. Then, the electrochemical signal materials (Pt-Ni-Cu NCs) were bound to the bacteria peptidoglycan layer using specific structural and surface features. The rapid and sensitive bacterial detection was then achieved using intrinsic electrochemical characteristics and superoxidase-like activity of the Pt-Ni-Cu NCs. Moreover, the nature of peptidoglycan covering the whole bacteria provided the premise for signal amplification. Under optimal conditions, the electrochemical signal variation was proportional to the concentration of bacteria ranging from 1.5 × 102 to 1.5 × 108 CFU/mL with a detection limit of 42 CFU/mL using a working potential of − 0.4 V. This electrochemical biosensor has been successfully applied to detect bacteria concentrations in urine samples, and the recoveries range from 90.4 to 107%. The proposed biosensor could be applied for broad-spectrum detection of Gram-positive bacteria since most Gram-positive bacteria possess a thick peptidoglycan layer. The developed electrochemical biosensing strategy might be used as a potential tool for clinical pathogenic bacteria detection and point-of-care testing (POCT).

Abstract:Sensitive, accurate, simple and quick monitoring of Salmonella typhimurium ( S. typhimurium ) in food is significant for preventing food poisoning, but still remains a challenge. Herein, a colorimetry/fluorescence dual-mode sensing strategy was fabricated to detect S. typhimurium by integrating the self-assembly of magnetic covalent organic framework (MCOF) with gold nanoparticles (Au NPs) as the peroxidase-mimicking nanozyme and aggregation-induced emission luminogen (AIEgen). S. typhimurium could competitive bind to aptamer conjugated Au NPs (Au NPs@apt), inhibit the self-assembly of MCOF with Au NPs, and shield the catalytic activity of AuNPs. After adding H 2 O 2 and TPE-4A, the dark green solution changed to light with increasing S. typhimurium concentration, on the contrary, the fluorescent signals were generated. As a result, in colorimetry/fluorescence modes, S. typhimurium could be detected in the linear ranges of 10 3 –10 8 CFU mL −1 and 10 1 –10 7 CFU mL −1 , with LODs of 1000 and 10 CFU mL −1 , respectively. Importantly, different colors consistent with various S. typhimurium concentrations can be accurately classified by smartphone app and linear discriminant analysis (LDA). The smartphone-assisted data interpretation can generate complementary colorimetry and fluorescence signals without any sophisticated equipment and achieve on-site detection. Moreover, the proposed strategy could be explored for S. typhimurium monitoring in milk with satisfactory recoveries (97.6–100.4 %) in colorimetry and fluorescence mode and good classification and prediction performance in smartphone/LDA system, suggesting the feasibility and potential applications of the sensing platform.
