Abstract:The authors describe a rapid enzyme-free colorimetric assay for food- and water-borne pathogens by using the multi-functional MnO2-doped magnetic ferrite nanoparticles (MCDs-MnO2 NPs). These nanoparticles not only can recognize, absorb, and separate the analyte from the matrix efficiently but also can directly catalyze the oxidation of TMB into a blue colored product without H2O2. In the presence of target, the nanoparticles preferentially bind to the target surface and the interaction between nanoparticles and TMB can be blocked, providing turn-off sensing of bacteria. By combining the superior capture efficiency of magnetic beads with the high catalytic activity of the enzyme mimic, the turn-off colorimetric assay can detect the gram-positive bacterium and the gram-negative bacterium by the naked eye at a low detection limit (102 cfu mL−1) with a wide line arrange from 10 to 106 cfu mL−1. With the advantage of simplicity, sensitivity, and specificity, this proposed approach showed promise in rapid instrumental and on-site visual detection of bacteria for determination of water contamination.

Abstract:Food safety problems attributed to foodborne pathogenic bacteria seriously endanger human health and cause substantial economic losses. Novel assays for rapid and sensitive identification of foodborne pathogenic bacteria are highly desired. In this study, a multicolor sensing system has been established for simultaneous determination of four foodborne bacteria by exploiting oxidase mimicking activity of aptamer-functionalized manganese dioxide-coated ferriferrous oxide (apt-Fe3O4/MnO2) nanocomposites and oxTMB etching of gold nanorods (AuNRs). Apt-Fe3O4/MnO2 nanocomposites were used as capture probes to recognize and capture specific bacteria. The captured bacteria blocked the catalytic sites of the magnetic conjugate, which inhibited the catalyzation of oxTMB and further reduced the etching of AuNRs. Consequently, the longitudinal shift of AuNRs decreased linearly with the increase of the concentration of bacteria ranging from 10 to 106 CFU mL-1. Instrumental detection limits for S. aureus, L. monocytogenes, E. coli O157:H7 and V. parahaemolyticus reached down to 1.3 CFU mL-1, 1.2 CFU mL-1, 1.3 CFU mL-1 and 1.4 CFU mL-1, respectively. And their visual detection limit was as low as 10 CFU mL-1. The whole detection process only needs 40 min, suggesting that this method is promising in on-site detection of bacteria.

Abstract:Nitrite is a typical food additive and preservative used in the food industry, which has attracted considerable attention due to its severe adverse effects on human health. Here, a sensitive and highly selective ratiometric colorimetric sensing platform for the detection of nitrite was created based on a polymetallic oxide nanozyme CoMnO 3 nanofibers (CMO). The nanozyme has superior oxidase-like activity ( K m was 0.105 mM and V max was 63.7 × 10-8 M S -1 ) and could catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to oxidized TMB (oxTMB), which was because that CMO can achieve the conversion of oxygen in solution to superoxide anion (O 2 •- ). In addition, it is interesting to note that oxTMB can be diazotized in the presence of nitrite under acidic condition, causing a shift in the ratio of nitrite concentration to the absorbance peaks at 450 and 652 nm ( A 450 /A 652 ). The ratio of A 450 /A 652 exhibited a positive linear relationship with the concentration of nitrite within the concentration range of 0.2–200 μM, with a detection limit of 0.094 μM. Simultaneously, this method was also successful in quantifying the nitrite produced by brined and pickled foods, as well as dynamic tracking the nitrite levels in various types of dishes. The analysis method based on the polymetallic oxide nanozyme catalysis integrated with the particular diazotization reaction not only offers dual-signal ratio sensing with high sensitivity, but also holds the benefit of outstanding selectivity for the use of the particular reaction, which has a wide range of application prospects in food safety management.

Abstract:Infiltration of monocytes into the arterial wall is an early cellular event in atherogenesis. Recent evidence shows that C-reactive protein (CRP) is deposited in the arterial intima at sites of atherogenesis. In this study, we demonstrate that CRP deposition precedes the appearance of monocytes in early atherosclerotic lesions. CRP is chemotactic for freshly isolated human blood monocytes. A specific CRP receptor is demonstrated on monocytes in vitro as well as in vivo, and blockage of the receptor by use of a monoclonal anti-receptor antibody completely abolishes CRP-induced chemotaxis. CRP may play a major role in the recruitment of monocytes during atherogenesis.

Abstract:In recent years, nanozymes have been widely used in the field of biosensing and food safety testing due to their advantages of low cost, high stability, easy modification and adjustable catalytic activity. However, how to reduce the signal interference generated by reducing substances, macromolecules and colored substances in the food matrix in nanozymes-based colorimetric sensing is still a major challenge. In this paper, using Listeria monocytogenes as a model analyte, sodium sulfonyl methacrylate (SBMA) polymers were modified onto cotton swabs by photothermal polymerization and combined with Listeria monocytogenes-specific aptamer (Apt1) to prepare swabs that can specifically capture and isolate Listeria monocytogenes from complex matrices (SBMA/Apt1 cotton swab). In addition, in combination with the inhibitory effect of the aptamer (Apt2) on the oxidase activity of Mn3O4 NPs, a colorimetric biosensor based on nanozymes that can quantitatively, sensitively, and specifically identify Listeria monocytogenes in food products was constructed. The results showed that the colorimetric signal of the method was linear with the concentration of Listeria monocytogenes in the range of 2.83-2.83 × 105 CFU/mL, and the limit of detection was 2.64 CFU/mL, which can be used for the detection of Listeria monocytogenes in complex environments and food samples.

Abstract:Development of a simple and efficient dual-mode analytical technique with the built-in cross reference correction feature is benefit to achieve the highly accurate detection of the target pollutants and avoid the false-positive outputs in environmental media. Here, we synthesized a Fe-doped polydopamine (Fe@PDA)-based nanozyme with prominent peroxide-mimetic enzyme activity and high fluorescence emission ability. On this basis, we designed a dual-recognition strategy-driven fluorescence-colorimetric dual-mode detection platform, consisting of Listeria monocytogenes (L. monocytogenes) recognition aptamer-modified Fe@PDA (apt/Fe@PDA) and vancomycin-functionalized Fe3O4 (van/Fe3O4), for L. monocytogenes. Owing to van/Fe3O4-powered magnetic separation, there was a L. monocytogenes concentration-dependent fluorescence enhancement of apt/Fe@PDA for performing fluorescence assay in the precipitate. In this case, the prominent peroxide-mimetic enzyme activity of the residual apt/Fe@PDA in the precipitation could catalyze H2O2 to further oxidate colorless 3,3',5,5'-tetramethylbenzidine (TMB) into blue oxTMB, which displayed a L. monocytogenes concentration-dependent absorbance enhancement for carrying out colorimetric assay as well. As a result, a fluorescence-colorimetric dual-mode analytical platform was proposed to successfully detect the residual L. monocytogenes in real environmental media with acceptable results. This work showed the great prospects by integrating dual-recognition strategy into fluorescence nanozyme to develop efficient and reliable dual-mode analytical platforms for safeguarding environmental health.

Abstract:A multifunctional oxidase-mimicking Ag/Mn3O4 was prepared, catalyzing the 3, 3', 5, 5'-tetramethylbenzidine (TMB) chromogenic reaction. Six foodborne pathogenic bacteria species, including Escherichia coli, Staphylococcus aureus, Salmonella enterica, Listeria monocytogenes, Bacillus cereus, and Cronobacter sakazakii, were observed to differentially inhibit its oxidase-like activity, resulting in decelerating the TMB chromogenic reaction. Owing to these properties, the following achievements were achieved: colorimetric determination of these bacteria with high sensitivity can be achieved using Ag/Mn3O4 + TMB reaction system; precise identification of these bacteria at different concentrations, including individual bacterium, binary mixtures, and even multivariate mixtures, can be effectively realized by combining the Ag/Mn3O4-based colorimetric sensor array with principal component analysis (PCA); broad-spectrum inactivation of these bacteria can be remarkably realized through catalyzation of Ag/Mn3O4 to generate superoxide anion free radicals. Therefore, our proposed Ag/Mn3O4 holds significant application potential in the colorimetric determination, precise identification, and broad-spectrum inactivation of foodborne pathogenic bacteria.

Abstract:Listeria monocytogenes (L. monocytogenes) is one of the most common food-borne pathogens. The authors describe a sensitive and reliable multicolorimetric assay for L. monocytogenes using a sensing system based on TMB2+ etching of gold nanorods. Apt-MNP was used as the capture probe, and IgY-BSA-MnO2 NPs was chosen as an oxidase-like nano-artificial enzyme to oxidize TMB to generate TMB2+. Under the optimized conditions, the longitudinal shift of localized surface plasmon resonances had a linear correlation with the L. monocytogenes concentration in the range between 10 to 10(6) cfu mL(-1). Meanwhile, the sensing system can generate vivid color responses as colorful as a rainbow, and the limit of detection is as low as 10 cfu mL(-1) at a glance. Recoveries ranging from 97.4 to 101.3% are found when analyzing spiked food samples without pre-enrichment. In our perception, it shows promise in rapid instrumental and on-site visual detection of L. monocytogenes. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:Early detection of foodborne bacteria is urgently needed to ensure food quality and to avoid the outbreak of foodborne bacterial diseases. Here, a kind of metal-organic framework (Zr-MOF) modified with Pt nanoparticles (Pt-PCN-224) was designed as a peroxidase-like signal amplifier for microfluidic biosensing of foodborne bacteria. Taking Escherichia coli (E. coli) O157:H7 as a model, a linear range from 2.93 × 102 to 2.93 × 108 CFU/mL and a limit of detection of 2 CFU/mL were obtained. The whole detection procedure was integrated into a single microfluidic chip. Water, milk, and cabbage samples were successfully detected, showing consistency with the results of the standard culture method. Recoveries were in the range from 90 to 110% in spiked testing. The proposed microfluidic biosensor realized the specific and sensitive detection of E. coli O157:H7 within 1 h, implying broad prospects of MOF with biomimetic enzyme activities for biosensing.

Abstract:Listeria monocytogenes is a hazardous foodborne pathogen that is able to cause acute meningitis, encephalitis, and sepsis to humans. The efficient detection of 3-hydroxy-2-butanone, which has been verified as a biomarker for the exhalation of Listeria monocytogenes, can feasibly evaluate whether the bacteria are contained in food. Herein, we developed an outstanding 3-hydroxy-2-butanone gas sensor based on the microelectromechanical systems using Au/ZnO NS as a sensing material. In this work, ZnO nanosheets were synthesized by a hydrothermal reaction, and Au nanoparticles (~5.5 nm) were prepared via an oleylamine reduction method. Then, an ultrasonic treatment was carried out to modified Au nanoparticles onto ZnO nanosheets. The XRD, BET, TEM, and XPS were used to characterize their morphology, microstructure, catalytic structure, specific surface area, and chemical composition. The response of the 1.0% Au/ZnO NS sensors vs. 25 ppm 3-hydroxy-2-butanone was up to 174.04 at 230 °C. Moreover, these sensors presented fast response/recovery time (6 s/7 s), great selectivity, and an outstanding limit of detection (lower than 0.5 ppm). This work is full of promise for developing a nondestructive, rapid and practical sensor, which would improve Listeria monocytogenes evaluation in foods.

Abstract:The inability to integrate detection and disinfection hindered building a unified pathogen monitoring platform, risking secondary contamination. Herein, a novel "four - in - one" platform for monitoring foodborne Listeria monocytogenes (L. monocytogenes) was presented. The magnetic daptomycin - functionalized Fe3O4 (Dap/Fe3O4) could selectively bind to L. monocytogenes, enhancing detection accuracy. The separated bacteria were captured by aptamers - functionalized Fe - doped - silica nanoparticles (Apt/Fe@SiNPs) for tri - mode detection. Besides fluorescence, the Apt/Fe@SiNPs converted 3,3',5,5' - tetramethylbenzidine (TMB) to oxidized TMB (oxTMB) via peroxidase activity, allowing colorimetric and subsequent photothermal detection upon irradiation, as low as 2.06 CFU/mL. Magnetic - induced aggregation of Apt/Fe@SiNPs generated toxic hydroxyl radicals around L. monocytogenes, achieving ∼99.6% disinfection. Furthermore, the biofilm of L. monocytogenes was effectively inhibited by the action of hydroxyl radicals. The platform might offer a promising prospect to control L. monocytogenes in food industries.
Facile Synthesis of Iron Oxide Nanozymes for Synergistically Colorimetric and Magnetic Resonance Detection Strategy

Abstract:Iron oxide nanomaterials with mimic enzymes activity have been paid more attention in the clinical diagnosis field. The modified surface molecules would influence the catalytic activity of nanozyme, which is worth studying. Furthermore, the traditional detection strategy is based on colorimetric change of substrates, however, the optical signal is easy to be interfered in complex biological applications. In our research, an efficient and facile preparation strategy was developed to obtain functional artificial nanozymes. Herein, three kinds of surfactants, including citrate acid, poly(ethylene glycol) bis (carboxymethyl) ether and tannic acid have been applied to modify these nanomaterials that showed uniform size, high soluble dispersity and stability. Furthermore, these nanozymes exhibited different peroxidase-like activity to catalyze the hydrogen peroxide and 3,3',5,5'-tetramethylbenzidine. More importantly, magnetic relaxation effect of iron oxide nanozymes was found to be changed during the catalytic reaction. In addition, the relationship between the magnetic signal of nanozymes and the substrate concentration showed a good linear dependence. Combined with the natural enzymes, the magnetic detection of iron oxide nanozymes also exhibited excellent substrate specificity. On these bases, a dual-function specific assay was constructed and further used for glucose detection. In conclusion, this study demonstrated an efficient iron oxide nanozymes preparation method and constructed a new synergistically colorimetric-magnetic diagnosis strategy.
[Oesophagogastroduodenoscopy in swine--technique, methods, indications].

Abstract:The aim of this study was to investigate in 20 healthy pigs the practicability of the oesophagogastroduodenoscopic examination technique in regard to equipment, method of examination, indications and its suitability as a diagnostical tool for the assessment of the oesophagus, stomach and upper duodenum in one procedure. Preparation of the patient for endoscopy, the procedure of the endoscopic examination as well as the topographical findings of the upper intestinal tract including the duodenum until the flexura duodenojejunalis are described. Flexible oesophagogastroduodenoscopy is a suitable method for the observation and natural visualisation of mucosal surfaces and for the digital documentation of peristaltic movements. The procedure is easy to perform in anaesthetized animals, is in most cases completed within 15 min, and can be repeated in the same animal. Indications of this interesting diagnostic imaging technique are discussed.
Trifunctional Nanocomposites with Colorimetric Magnetic Catalytic Activities Labels in Sandwich Immunochromatographic Detection of Escherichia coli O157:H7

Abstract:The emergence of nanoenzymes has catalyzed the robust advancement of the lateral flow immunoassay (LFIA) in recent years. Among them, multifunctional nanocomposite enzymes with core-shell architectures are considered preferable for promoting the sensing ability due to their good biocompatibility, precise control over size, and surface properties etc. Herein, we developed a dual-channel ensured lateral flow immunoassay (DFLIA) platform utilizing a magnetic, colorimetric, and catalytic multifunctional nanocomposite enzyme (Fe3O4@TCPP@Pd) [TCPP, Tetrakis (4-carboxyphenyl) porphyrin] for the ultrasensitive and highly accurate rapid detection of Escherichia coli O157:H7 (E. coli O157:H7). Fe3O4@TCPP@Pd-mAb exhibits superior performance compared to traditional AuNPs, including enhanced sensitivity and an extended linear detection range, benefiting from its high brightness signal, strong magnetic separation ability, and high peroxidase activity (Vmax = 2.32 μM S1-). Moreover, the Fe3O4@TCPP@Pd-labeled mAb probe exhibited exceptional stability and high affinity toward E. coli O157:H7 (with an affinity constant of approximately 1.723 × 109 M-1), indicating its potential for the efficient capture of the pathogen. Impressively, the developed Fe3O4@TCPP@Pd-DFLIA achieved ultrasensitive detection for E. coli O157:H7 with pre- and postcatalytic naked-eye detection sensitivities of 255 cfu/mL and 77 cfu/mL, respectively, representing an approximately 41-fold improvement over the conventional AuNP-based LFIA and also possessed good specificity and reproducibility [relative standard deviation (RSD) < 10%]. Additionally, the established DFLIA exhibited satisfactory recoveries in detecting pork and milk samples, further validating the reliability of this platform for immunoassays and demonstrating its potential for utilization in bioassays and clinical diagnostics.

Abstract:Novel Fe3O4-Ag-MnO2 composites were successfully synthesized. It was noteworthy that the obtained Fe3O4-Ag-MnO2 composites were found to possess three types of enzyme-mimicking activities, including peroxidase-like, catalase-like and oxidase-like activities. Taking advantage of the oxidase properties of Fe3O4-Ag-MnO2, the direct oxidation of TMB could be catalyzed to generate blue oxidation products without H2O2. The oxidase-like activity of Fe3O4-Ag-MnO2 were carefully studied. Based on the Fe3O4-Ag-MnO2-TMB system, a fast, sensitive and intuitive multicolor colorimetric method for Staphylococcus aureus (S. aureus) detection was established under the optimized conditions. The proposed method allows the detection of S. aureus with a detection limit of 3.7 cfu mL-1 and a linear range of 10-106 cfu mL-1. This new colorimetric method has been successfully proved to be applicable to the detection S. aureus of food samples.

Abstract:Rapid, sensitive and point-of-care detection of foodborne pathogenic bacteria is essential for food safety. In this study, we found that hemin-concanavalin A hybrid nanoflowers (HCH nanoflowers), as solid mimic peroxidase, could catalyze oxidation of 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt (ABTS) in the presence of H2O2 to a green-colored product. HCH nanoflowers, integrating the essential functions of both biological recognition and signal amplification, meet the requirements of signal labels for colorimetric immunoassay of bacteria. In view of the excellent peroxidase mimetic catalytic activity of HCH nanoflowers, a colorimetric biosensing platform was newly constructed and applied for sensitive detection of foodborne Escherichia coli O157:H7 (E. coli O157:H7). The corresponding detection limits was as low as 4.1 CFU/mL with wide linear ranges (101-106 CFU/mL).

Abstract:A highly sensitive and selective foodborne pathogen detection system is essential to ensure food quality and public health. Therefore, in this study, a dual recognition and highly sensitive detection of Listeria monocytogenes by fluorescence enhancement strategy based on Fe3O4 @ZIF-8 have been developed. Aptamer absorbed on Fe3O4 @ZIF-8 were used for the specific capture of Listeria monocytogenes, and the conjugates of anti-Listeria monocytogenes antibody-biotin and streptavidin-FITC were used as signal amplification and fluorescence recognition probes binding with target bacteria. Simultaneously, the quantitative detection of Listeria monocytogenes was realized by monitoring the increase of FITC fluorescence signal due to hydrophobic environment. The linear range of the detection of pure culture was from 1.4 × 101 to 1.4 × 107 CFU/mL, with the detection limits as low as 0.88 CFU/mL and the coefficient of variation of each concentration no more than 2%. A good specificity of the detection system was also showed in the results, and the detection took about 70 min. Furthermore, the spiked samples of pork and milk were also tested by the detection system, and the recoveries ranged from 88.0% to 103.8%, with the coefficient of variation less than 12.8%. Taking advantages of sensitivity, rapidly and good selectivity, the proposed strategy in this study has great application potential in detection of foodborne pathogens in food.

Abstract:Considering the importance of rapid and effective detection of nitrite in food samples, herein, we report a multistep heterophase synthetic strategy for constructing crystalline-amorphous zinc/cobalt iron porous nanosheets (C-A Zn/Co-Fe PNSs) on carbon cloth. Notably, the C-A Zn/Co-Fe PNSs@CC electrochemical interface consists of interlaced carbon fibers with many uniformly distributed hybrid nanosheets containing crystalline-amorphous interfacial sites. This particular hybrid structure permits local enrichment of nitrite for enhanced nitrite capture efficiency, laying a solid foundation for the ultrasensitive detection of nitrite. In proof-of-concept experiments, we verified that C-A Zn/Co-Fe PNSs@CC could be used to analyze nitrite with good sensitivity and reproducibility. It is worth mentioning that an ultra-low LOD of 0.44 μM was obtained for the detection of nitrite. The designed electrochemical sensor worked well in complex samples such as sausages and tap water under optimal parameters.

Abstract:In this work, we constructed an enhanced electrochemical signal sensing platform using Au/ZnO nanoflake arrays coated on carbon cloth for the rapid detection of nitrite in food. Based on a stepwise synthesis strategy of electrodeposition and magnetron sputtering technique, AuNPs were sputtered onto ZnO nanoflower-like array sheets. Combining the high catalytic performance of AuNPs with the morphology of ZnO significantly increased the surface area and electrocatalytic activity of the electrodes. The prepared sensor showed a linear response range of 0.2-4986 μΜ, a limit of detection of 0.09 μM, and a high sensitivity of 5677 μA mM-1 cm-2. It is worth noting that the sensor can precisely detect nitrite in the presence of interfering substances and has excellent stability and reproducibility. In addition, the nitrite residues in several food samples were analyzed using this method and spectrophotometric method, and the results of the two methods were not significantly different.

Abstract:Foodborne microbial contaminants exist in foods variously and pose a major threat to human health. Therefore, the fast and touchy orientation of microbial contaminants in food is critical for avoiding the emergence and spreading of foodborne maladies. Nanozymes have the advantages of multifunction, which show broad application potential in the analysis of microbial contaminants and the prevention of infectious diseases. In this review, the various materials applied to fabricate nanozymes and diverse types of enzyme-like are briefly introduced. Further, the catalytic mechanism and regulatory behavior are summarized, while the methods in the combination with recognition elements are also been explored. Nanozymes build a multifunctional monitoring platform that effectively improves the detection level and shortens the analysis time and realizes on-site inspection to provide a feasible path for foodborne microbial contaminants detection. However, problems in the creation of superior and biosafe nanozymes, diversification of recognition-to-signal methods, and practical use in complicated food matrices must be overcome.
