Abstract:It is essential to develop ultrasensitive biosensors for cancer detection and treatment monitoring. In the development of sensing platforms, metal-organic frameworks (MOFs) have received considerable attention as potential porous crystalline nanostructures. Core-shell MOF nanoparticles (NPs) have shown different diversities, complexities, and biological functionalities, as well as significant electrochemical (EC) properties and potential bio-affinity to aptamers. As a result, the developed core-shell MOF-based aptasensors serve as highly sensitive platforms for sensing cancer biomarkers with an extremely low limit of detection (LOD). This paper aimed to provide an overview of different strategies for improving selectivity, sensitivity, and signal strength of MOF nanostructures. Then, aptamers and aptamers-modified core-shell MOFs were reviewed to address their functionalization and application in biosensing platforms. Additionally, the application of core-shell MOF-assisted EC aptasensors for detection of several tumor antigens such as prostate-specific antigen (PSA), carbohydrate antigen 15-3 (CA15-3), carcinoembryonic antigen (CEA), human epidermal growth factor receptor-2 (HER2), cancer antigen 125 (CA-125), cytokeratin 19 fragment (CYFRA21-1), and other tumor markers were discussed. In conclusion, the present article reviews the advancement of potential biosensing platforms toward the detection of specific cancer biomarkers through the development of core-shell MOFs-based EC aptasensors.

Abstract:Breast cancer detection at an early stage significantly increases the chances of successful treatment and survival. This study presents an electrochemical biosensor for detecting breast cancer cells, utilizing silver nanoclusters encapsulated by hemoglobin and Cu (II)-porphyrin-metal organic framework (BioMOF) in a graphene-incorporated nanohybrid probe. This Hb-AgNCs@MOF-G probe demonstrates high electrochemical activity, superior dispersity, porosity, and a large surface area for effective functionalization. Using a green ultrasonic-assisted stirring method, we fabricate ultra-small 5 nm particles that readily immobilize on a glassy carbon electrode, generating a detection signal when interacting with ferricyanide/ferrocyanide redox probes. The resulting immunosensor detects as few as 2 cells/mL using Electrochemical Impedance Spectroscopy (EIS) "signal on" and 16 cells/mL via Square Wave Voltammetry (SWV) "signal off", within a broad range of cell concentrations (10 2 –5 × 10 4 cells/mL). Our designed sensor shows improved selectivity (5- to 16-fold) and robust detection in human blood with a recovery efficiency between 94.8–106% (EIS method) and 95.4–111% (SWV method). This sensor could streamline early cancer diagnosis and monitor patient treatment without requiring labelling or signal amplification. As a pioneering endeavor, we've utilized integrated porous MOFs with Hb-encapsulated silver nanoclusters in cancer detection, where these components collectively enhance the overall functionality.

Abstract:A cobalt metal-organic framework (Co MOF)-ionic liquid (IL) nanocomposite comprising the base matrix of the electrode was developed with electropolymerized molecularly imprinted polymer consisting of o-phenylenediamine (oPD) to detect carcinoembryonic antigen (CEA), a biomarker of breast cancer. First, Co MOF was synthesized using hydrothermal synthesis, which was used as an electrically conductive skeleton and functionalized with an IL. Then, CEA was employed as a template in the electropolymerization of oPD-imprinted films on the surface of screen-printed carbon electrode. Finally, the template protein was removed to form a molecularly imprinted film capable of qualitatively and quantitatively signaling CEA. Under optimized conditions, the sensor for CEA exhibits a wide detection range of 1.0×10-4-10 ng·mL-1 (0.32 V vs. Ag/AgCl) with a limit of detection and limit of quantification of 0.024 pg·mL-1 and 0.082 pg·mL-1, respectively. Besides, other parameters including the selectivity, reproducibility (RSD 2.2%), and recovery rate (92.16-103.35%) are all satisfactory. The proposed strategy provides a new route for the development of rapid breast cancer diagnostic tools.

Abstract:Nanocomposite materials have provided a wide range of conductivity, sensitivity, selectivity and linear response for electrochemical biosensors. However, the detection of rare cells at single cell level requires a new class of nanocomposite-coated electrodes with exceptional sensitivity and specificity. We recently developed a construct of gold nanoparticle-grafted functionalized graphene and nanostructured polyaniline (PANI) for high-performance biosensing within a very wide linear response and selective performance. Further, replacing the expensive gold nanoparticles with low-cost silver nanoparticles as well as optimizing the nanocomposite synthesis and functionalization protocols on the electrode surface in this work enabled us to develop ultrasensitive nanocomposites for label-free detection of breast cancer cells. The sensor presented a fast response time of 30 min within a dynamic range of 10 - 5 × 106 cells mL-1 and with a detection limit of 2 cells mL-1 for the detection of SK-BR3 breast cancer cell. The nano-biosensor, for the first time, demonstrated a high efficiency of > 90% for the label-free detection of cancer cells in whole blood sample without any need for sample preparation and cell staining. The results demonstrated that the optimized nanocomposite developed in this work is a promising nanomaterial for electrochemical biosensing and with the potential applications in electro-catalysis and super-capacitances.

Abstract:Developing ultrasensitive and highly selective biosensors is significant for the early diagnosis of cancers and monitoring of the treatment process of cancer patients. Metal–organic frameworks (MOFs), as emerging porous crystalline materials, have received increased attention on account of their potential applications in many fields, such as energy, environment, storage, separation, and sensing. In light of their unique merits, such as large surface areas, tunable pore scales, and good adsorption capabilities, MOFs have been applied as sensitive platforms for anchoring diverse probes (e.g., antibodies, DNA, or aptamers) for the construction of electrochemical (EC) biosensors. Herein, we present a comprehensive overview of MOFs-based EC biosensors for detecting diverse targets (e.g., cancer markers, microRNA, and living cancer cells) that are considered as the indicators for early diagnosis of cancers. Different strategies for construction of EC biosensors with monometallic MOFs, bimetallic MOFs, luminophore-combined MOFs, and MOFs-based nanocomposites are reviewed. Further, the fabrication of MOFs-based EC biosensors toward specific analytes is described to promote and guide future studies on MOFs for biosensing applications.

A sensitive sandwich-type electrochemical immunosensor using nitrogen-doped graphene/metal-organic framework-derived CuMnCoOx and Au/MXene for the detection of breast cancer biomarker

Abstract:In terms of cancer-related deaths among women, breast cancer (BC) is the most common. Clinically, human epidermal growth receptor 2 (HER2) is one of the most commonly used diagnostic biomarkers for facilitating BC cell proliferation and malignant growth. In this study, a disposable gold electrode (DGE) modified with gold nanoparticle-decorated Ti3C2Tx (Au/MXene) was utilized as a sensing platform to immobilize the capturing antibody (Ab1/Au/MXene). Subsequently, nitrogen-doped graphene (NG) with a metal-organic framework (MOF)-derived copper-manganese-cobalt oxide, tagged as NG/CuMnCoOx, was used as a probe to label the detection antibody (Ab2). A sandwich-type immunosensor (NG/CuMnCoOx/Ab2/HER2-ECD /Ab1/Au/MXene/DGE) was developed to quantify HER2-ECD. NG/CuMnCoOx enhances the conductivity, electrocatalytic active sites, and surface area to immobilize Ab2. In addition, Au/MXene facilitates electron transport and captures more Ab1 on its surface. Under optimal conditions, the resultant immunosensor displayed an excellent linear range of 0.0001 to 50.0 ng. mL-1. The detection limit was 0.757 pg·mL-1 with excellent selectivity, appreciable reproducibility, and high stability. Moreover, the applicability for determining HER2-ECD in human serum samples indicates its ability to monitor tumor markers clinically.

Abstract:Timely identification of cancers is pivotal in optimizing treatment efficacy and reducing their widespread impact. This study introduces a novel biosensor for the sensitive electrochemical detection of cancer cells overexpressing mucin 1 (MUC1), a well-established model for breast cancer. The sensor substrate comprises gold columnar nanostructures obtained through glancing angle deposition (GLAD) of copper nanostructures, subsequently replaced by gold via a facile galvanic replacement process. Functionalizing these gold nanostructures with aptamers targeting the MUC1 glycoproteins, a prominent cancer biomarker, enables specific recognition of MCF-7 breast cancer cells. The proposed electrochemical sensing platform offers several advantages, including high selectivity, a wide linear range of detection, a low detection limit of 30 cells per mL, and long-term stability, rendering this sensor highly desirable for definitive breast cancer diagnosis.
Ketone body utilization drives tumor growth and metastasis

Abstract:We have previously proposed that catabolic fibroblasts generate mitochondrial fuels (such as ketone bodies) to promote the anabolic growth of human cancer cells and their metastasic dissemination. We have termed this new paradigm “two-compartment tumor metabolism.” Here, we further tested this hypothesis by using a genetic approach. For this purpose, we generated hTERT-immortalized fibroblasts overexpressing the rate-limiting enzymes that promote ketone body production, namely BDH1 and HMGCS2. Similarly, we generated MDA-MB-231 human breast cancer cells overexpressing the key enzyme(s) that allow ketone body re-utilization, OXCT1/2 and ACAT1/2. Interestingly, our results directly show that ketogenic fibroblasts are catabolic and undergo autophagy, with a loss of caveolin-1 (Cav-1) protein expression. Moreover, ketogenic fibroblasts increase the mitochondrial mass and growth of adjacent breast cancer cells. However, most importantly, ketogenic fibroblasts also effectively promote tumor growth, without a significant increase in tumor angiogenesis. Finally, MDA-MB-231 cells overexpressing the enzyme(s) required for ketone re-utilization show dramatic increases in tumor growth and metastatic capacity. Our data provide the necessary genetic evidence that ketone body production and re-utilization drive tumor progression and metastasis. As such, ketone inhibitors should be designed as novel therapeutics to effectively treat advanced cancer patients, with tumor recurrence and metastatic disease. In summary, ketone bodies behave as onco-metabolites, and we directly show that the enzymes HMGCS2, ACAT1/2 and OXCT1/2 are bona fide metabolic oncogenes.

Abstract:Ratiometric electrochemical biosensors have attracted the attention of researchers due to their ability to self-calibrate, which can improve accuracy of the detection. We constructed a radiometric electrochemical aptasensor based on functionalized bimetallic Zn–Co-MOF@ferrocene (Fc) and Fe–Co-MOF@methylene blue (MB) composites with high specific surface area, high electrical conductivity and strong adsorption properties to detect human epidermal growth factor receptor 2 (HER2). In this paper, we synthesized bimetallic Zn–Co-MOF@Fc by in situ electrochemical synthesis and bimetallic Fe–Co-MOF, and the MB signal was wrapped in Fe–Co-MOF, making it immune to the interference of the external environment. In addition, bimetallic MOFs can also immobilize aptamer on their surfaces through adsorption. Therefore, we used Zn–Co-MOF@Fc/aptamer as the capture probe for HER2 and Fe–Co-MOF@MB as the signal labeling probe to jointly construct the radiometric electrochemical aptasensor. By calculating the ratio of signal peaks, simple, fast and accurate detection of HER2 is achieved. The linear range of the sensor is 0.75–250 pg/mL, and the detection limit is 0.37 pg/mL. It provides a new idea for the detection of breast cancer tumor biomarkers.

Abstract:An ultra-sensitive ratiometric electrochemical aptasensor was constructed based on metal-organic frameworks (MOFs) and bimetallic oxides for the detection of the human epidermal growth factor receptor 2 (HER2), a breast cancer marker. The aluminum metal-organic framework (Al-MOF) and cerium-metal-organic framework (Ce-MOF) have higher specific surface area, which is conducive to load more aptamers or complementary DNA (cDNA), and realize the amplification of internal reference signal Fc. Furthermore, nanoflower-like bismuth copper oxide (Bi<sub>2</sub>CuO<sub>4</sub>) with abundant active sites was introduced to modify more aptamers on its surface, which were then fixed to the glassy carbon electrode (GCE) to amplify the detection signal. The quantitative detection of HER2 was achieved by differential pulse voltammetry (DPV), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The materials were characterized by scanning electron microscope, transmission electron microscope, Zeta potential analyzer, X-ray diffraction and X-ray photoelectron spectroscopy. The ratiometric electrochemical aptasensor based on nanomaterial and chain displacement signal amplification technology could discern HER2 in a very wide range (0.001-20.0 ng/mL) with an extremely low detection limit (0.049 pg/mL) and has demonstrated good performance in clinical serum analysis. This strategy also provides a feasible idea for sensitive analysis of other clinical tumor markers.

Abstract:Carbohydrate antigen 15-3 (CA15-3) is an important biomarker for early diagnosis of breast cancer. Herein, a label-free electrochemical immunosensor was built based on three-dimensional (3D) urchin-like core-shell Au@PdCu nanocrystals (labeled Au@PdCu NCs) for highly sensitive detection of CA15-3, where K 3 [Fe(CN) 6 ] behaved as an electroactive probe. The Au@PdCu NCs were synthesized by a simple one-pot wet-chemical approach and the morphology, structures, and electrocatalytic property were investigated by several techniques. The Au@PdCu NCs prepared worked as electrode material to anchor more antibodies and as signal magnification material by virtue of its exceptional catalytic property. The developed biosensor exhibited a wide linear detection range from 0.1 to 300 U mL −1 and a low limit of detection (0.011 U mL −1 , S / N = 3) for determination of CA15-3 under the optimal conditions. The established biosensing platform exhibits some insights for detecting other tumor biomarkers in clinical assays and early diagnosis.

Abstract:An electrochemiluminescence-electrochemistry (ECL-EC) dual-mode sensing platform based on a vertically-ordered mesoporous silica films (VMSF) modified electrode was designed here for the sensitive and selective determination of cancer antigen 15-3 (CA 15-3), a specific biomarker of breast cancer. VMSF was assembled through a rapid electrochemically assisted self-assembly (EASA) method and plays a crucial role in signal amplification via a strong electrostatic interaction with the positively charged bifunctional probe Ru(bpy)32+. To construct the biorecognition interface, epoxy functional silane was linked to the surface of VMSF for further covalent immobilization of the antibody. As a benefit from the specific combination of antigen and antibody, a non-conductive immunocomplex layer was formed in the presence of CA 15-3, leading to the hinderance of the mass and electron transfer of the probes. Based on this strategy, the dual-mode determination of CA 15-3 ranging from 0.1 mU/mL to 100 mU/mL with a LOD of 9 μU/mL for ECL mode, and 10 mU/mL to 200 U/mL with a LOD of 5.4 mU/mL for EC mode, was achieved. The proposed immunosensor was successfully employed for the detection of CA 15-3 in human serum without tedious pretreatment.

A multifunctional N-GO/PtCo nanocomposite bridged carbon fiber interface for the electrochemical aptasensing of CA15-3 oncomarker

Abstract:The development of integrated analytical devices is crucial for advancing next-generation point-of-care platforms. Herein, we describe a facile synthesis of a strongly catalytic and durable Nitrogen-doped graphene oxide decorated platinum cobalt (NGO-PtCo) nanocomposite that is conjugated with target-specific DNA aptamer (i-e. MUC1) and grown on carbon fiber. Benefitting from the combined features of the high electrochemical surface area of N-doped GO, high capacitance and stabilization by Co, and high kinetic performance by Pt, a robust, multifunctional, and flexible nanotransducer surface was created. The designed platform was applied for the specific detection of a blood-based oncomarker, CA15-3. The electrochemical characterization proved that nanosurface provides a highly conductive and proficient immobilization support with a strong bio-affinity towards MUC1 aptamer. The specific interaction between CA15-3 and the aptamer alters the surface properties of the aptasensor and the electroactive signal probe generated a remarkable increase in signal intensity. The sensor exhibited a wide dynamic range of 5.0 × 10-2 -200 U mL-1, a low limit of detection (LOD) of 4.1 × 10-2 U mL-1, and good reproducibility. The analysis of spiked serum samples revealed outstanding recoveries of up to 100.03 %, by the proposed aptasensor. The aptasensor design opens new revelations in the reliable detection of tumor biomarkers for timely cancer diagnosis.

Abstract:Electrochemical immunosensors, surpassing conventional diagnostics, exhibit significant potential for cancer biomarker detection. However, achieving a delicate balance between signal sensitivity and operational stability, especially at the heterostructure interface, is crucial for practical immunosensors. Herein, porous carbon (PC) integration with Ti3C2Tx-MXene (MX) and gold nanoparticles (Au NPs) constructs a versatile immunosensing platform for detecting extracellular matrix protein-1 (ECM1), a breast cancer-associated biomarker. The inclusion of PC provided robust structural support, enhancing electrolytic diffusion with an expansive surface area while synergistically facilitating charge transfer with Ti3C2Tx. The biosensor optimized with 1.0 mg PC demonstrates a robust electrochemical redox response to the surface-bound thionine (th) redox probe, utilizing an inhibition-based strategy for ECM1 detection. The robust antibody-antigen interactions across the PC-integrated Ti3C2Tx-Au NPs platform (MX-Au-C-1) enabled robust ECM1 detection within 0.1-7.5 nM, with a low limit of detection (LOD) of 0.012 nM. The constructed biosensor shows improved operational stability with a 98.6 % current retention over 1 h, surpassing MXene-integrated (MX-Au) and pristine Au NPs (63.2 % and 44.3 %, respectively) electrodes. Moreover, the successful adaptation of the artificial neural network (ANN) model for predictive analysis of the generated DPV data further validates the accuracy of the biosensor, promising its future application in AI-powered remote health monitoring.

Abstract:Breast cancer continues to be a significant contributor to global cancer deaths, particularly among women. This highlights the critical role of early detection and treatment in boosting survival rates. While conventional diagnostic methods like mammograms, biopsies, ultrasounds, and MRIs are valuable tools, limitations exist in terms of cost, invasiveness, and the requirement for specialized equipment and trained personnel. Recent shifts towards biosensor technologies offer a promising alternative for monitoring biological processes and providing accurate health diagnostics in a cost-effective, non-invasive manner. These biosensors are particularly advantageous for early detection of primary tumors, metastases, and recurrent diseases, contributing to more effective breast cancer management. The integration of biosensor technology into medical devices has led to the development of low-cost, adaptable, and efficient diagnostic tools. In this framework, electrochemical screening platforms have garnered significant attention due to their selectivity, affordability, and ease of result interpretation. The current review discusses various breast cancer biomarkers and the potential of electrochemical biosensors to revolutionize early cancer detection, making provision for new diagnostic platforms and personalized healthcare solutions.

Abstract:Cancer stands as one of the most impactful illnesses in the modern world, primarily owing to its lethal consequences. The fundamental concern in this context likely stems from delayed diagnoses in patients. Hence, detecting various forms of cancer is imperative. A formidable challenge in cancer research has been the diagnosis and treatment of this disease. Early cancer diagnosis is crucial, as it significantly influences subsequent therapeutic steps. Despite substantial scientific efforts, accurately and swiftly diagnosing cancer remains a formidable challenge. It is well known that the field of cancer diagnosis has effectively included electrochemical approaches. Combining the remarkable selectivity of biosensing components—such as aptamers, antibodies, or nucleic acids—with electrochemical sensor systems has shown positive outcomes. In this study, we adapt a novel electrochemical biosensor for cancer detection. This biosensor, based on a glassy carbon electrode, incorporates a nanocomposite of reduced graphene oxide/Fe 3 O 4 /Nafion/polyaniline. We elucidated the modification process using SEM, TEM, FTIR, RAMAN, VSM, and electrochemical methods. To optimize the experimental conditions and monitor the immobilization processes, electrochemical techniques such as CV, EIS, and SWV were employed. The calibration graph has a linear range of 10 2 –10 6 cells mL −1 , with a detection limit of 5 cells mL −1 .

Abstract:Breast cancer is a rapidly spreading disease and early detection of associated biomarkers in body fluids, particularly blood, is crucial. Electrochemical biosensors, known for their high sensitivity, selectivity, cost-effectiveness, and miniaturization potential, have emerged as a promising tool for breast cancer screening. This review article focuses on the integration of polymers and nanosized objects to enhance the performance of electrochemical biosensors in analyzing breast cancer biomarkers in serum samples, breast cancer cell lysates, and tumor tissues. Recent developments in this field have demonstrated the significant benefits of these sensors, including improved sensitivity and selectivity. By incorporating nanocomposites into the electrochemical sensing interface, signal amplification is achieved, resulting in enhanced detection limits and accuracy. The use of different dimensionalities of nanomaterials (0D, 1D, 2D, and 3D nanostructured interfaces) in polymer-based nanocomposites has further contributed to the development of sensors with unique properties for sensitive and rapid detection. The potential of electrochemical biosensors based on polymer nanocomposites to revolutionize disease management lies in their ability to enable user-friendly breast cancer screening and facilitate early detection. However, there are still limitations that need to be addressed, and further research is required to optimize the performance of these sensors. The article also discusses the future perspectives and real-world applications of electrochemical biosensors based on polymer nanocomposites while considering the challenges that lie ahead.

Abstract:The development of high-efficient technologies for cancer biomarkers detection has attracted tremendous research effort for its great clinic significance. In this work, we designed a new type of flexible and robust nanohybrid microelectrode by modifying carbon fiber with dual nanoenzyme, i.e., AuPd alloy nanoparticles (AuPd-ANPs) decorated graphene quantum dots (GQDs) assembly, and explored its practical application in electrochemical sensing system for sensitive detection of cancer biomarker hydrogen peroxide (H2O2) in human breast cancer cells and tissue. For the preparation of dual nanoenzyme modified microelectrode, ionic liquid was used as the electrolyte for the effective electrodeposition of GQDs on carbon fiber substrate to form a close-packed assembly under a very negative potential, then the highly dense AuPd-ANPs were uniformly decorated on GQDs assembly by electrodeposition. In virtue of the structural merits and synergistic contribution of dual nanoenzyme in enhancing the electrocatalytic activity to H2O2, the resultant nanohybrid microelectrode exhibited good sensing performances for electrochemical detection of H2O2, including a high sensitivity of 371 μA cm-2 mM-1, a wide linear range from 1.0 μM to 18.44 mM, a low detection limit of 500 nM (a signal-to-noise ratio of 3:1), as well as good selectivity and biocompatibility, which could be used for real-time tracking H2O2 released from different types of human breast cells and in situ sensitive detection of H2O2 in clinical breast cancer tissue.
