Abstract:Abstract Hydrogels are a three-dimensional and crosslinked network of hydrophilic polymers. They can absorb a large amount of water or biological fluids, which leads to their swelling while maintaining their 3D structure without dissolving (Zhu and Marchant, Expert Rev Med Devices 8:607–626, 2011). Among the numerous polymers which have been utilized for the preparation of the hydrogels, polysaccharides have gained more attention in the area of pharmaceutics; Sodium alginate is a non-toxic, biocompatible, and biodegradable polysaccharide with several unique physicochemical properties for which has used as delivery vehicles for drugs (Kumar Giri et al., Curr Drug Deliv 9:539–555, 2012). Owing to their high-water content and resembling the natural soft tissue, hydrogels were studied a lot as a scaffold. The formation of hydrogels can occur by interactions of the anionic alginates with multivalent inorganic cations through a typical ionotropic gelation method. However, those applications require the control of some properties such as mechanical stiffness, swelling, degradation, cell attachment, and binding or release of bioactive molecules by using the chemical or physical modifications of the alginate hydrogel. In the current review, an overview of alginate hydrogels and their properties will be presented as well as the methods of producing alginate hydrogels. In the next section of the present review paper, the application of the alginate hydrogels will be defined as drug delivery vehicles for chemotherapeutic agents. The recent advances in the application of the alginate-based hydrogels will be describe later as a wound dressing and bioink in 3D bioprinting.

Abstract:Gastrointestinal tumors are the main causes of death among the patients. These tumors are mainly diagnosed in the advanced stages and their response to therapy is unfavorable. In spite of the development of conventional therapeutics including surgery, chemotherapy, radiotherapy and immunotherapy, the treatment of these tumors is still challenging. As a result, the new therapeutics based on (nano)biotechnology have been introduced. Hydrogels are polymeric 3D networks capable of absorbing water to swell with favorable biocompatibility. In spite of application of hydrogels in the treatment of different human diseases, their wide application in cancer therapy has been improved because of their potential in drug and gene delivery, boosting chemotherapy and immunotherapy as well as development of vaccines. The current review focuses on the role of hydrogels in the treatment of gastrointestinal tumors. Hydrogels provide delivery of drugs (both natural or synthetic compounds and their co-delivery) along with gene delivery. Along with delivery, hydrogels stimulate phototherapy (photothermal and photodynamic therapy) in the suppression of these tumors. Besides, the ability of hydrogels for the induction of immune-related cells such as dendritic cells can boost cancer immunotherapy. For more specific cancer therapy, the stimuli-responsive types of hydrogels including thermo- and pH-sensitive hydrogels along with their self-healing ability have improved the site specific drug delivery. Moreover, hydrogels are promising for diagnosis, circulating tumor cell isolation and detection of biomarkers in the gastrointestinal tumors, highlighting their importance in clinic. Hence, hydrogels are diagnostic and therapeutic tools for the gastrointestimal tumors.

Abstract:Cancer is one of the most intractable diseases in the world because of its high recurrence rate, high metastasis rate and high lethality rate. Traditional chemotherapy, radiotherapy and surgery have unsatisfactory therapeutic effects and cause many severe side effects at the same time. Hydrogel is a new type of biomaterial with the advantages of good biocompatibility and easy degradation, which can be used as a carrier of functional nanomaterials for tumor therapy. Herein, we represent the progress of hydrogels with different skeletons and their application as carrier in tumor treatment. The hydrogels are listed as polyethylene glycol-based hydrogels, chitosan-based hydrogels, peptide-based hydrogels, hyaluronic acid-based hydrogels, steroid-based hydrogels and other hydrogels by skeletons, and their properties, modifications and toxicities were introduced. Some representative applications of combined hydrogels with nanomaterial for chemotherapy, photodynamic therapy, photothermal therapy, sonodynamic therapy, chemodynamic therapy and synergistic therapy are highlighted.

Abstract:Cancer, as leading cause of death, has a high rate of mortality worldwide. Although there is a wide variety of conventional approaches for the treatment of cancer (such as surgery and chemotherapy), they have considerable drawbacks in terms of practicality, treatment efficiency, and cost-effectiveness. Therefore, there is a fundamental requirement for the development of safe and efficient treatment modalities based on breakthrough technologies to suppress cancer. Chitosan (CS) and hyaluronic acid (HA) polysaccharides, as FDA-approved biomaterials for some biomedical applications, are potential biopolymers for the efficient treatment of cancer. CS and HA have high biocompatibility, bioavailability, biodegradability, and immunomodulatory function which guarantee their safety and non-toxicity. CS-/HA-based hydrogels (HGs)/composites stand out for their potential anticancer function, versatile preparation and modification, ease of administration, controlled/sustained drug release, and active and passive drug internalization into target cells which is crucial for efficient treatment of cancer compared with conventional treatment approaches. These HGs/composites can respond to external (magnetic, ultrasound, light, and thermal) and internal (pH, enzyme, redox, and ROS) stimuli as well which further paves the way to their manipulation, targeted drug delivery, practicality, and efficient treatment. The above-mentioned properties of CS-/HA-based HGs/composites are unique and practical in cancer treatment which can ignore the deficiencies of conventional approaches. The present manuscript comprehensively highlights the advances in the practical application of stimuli-responsive HGs/composites based on CS/HA polysaccharides.

Abstract:When hydrogel materials with excellent biocompatibility and biodegradability are used as excellent new drug carriers in the treatment of cancer, they confer the following three advantages. First, hydrogel materials can be used as a precise and controlled drug release systems, which can continuously and sequentially release chemotherapeutic drugs, radionuclides, immunosuppressants, hyperthermia agents, phototherapy agents and other substances and are widely used in the treatment of cancer through radiotherapy, chemotherapy, immunotherapy, hyperthermia, photodynamic therapy and photothermal therapy. Second, hydrogel materials have multiple sizes and multiple delivery routes, which can be targeted to different locations and types of cancer. This greatly improves the targeting of drugs, thereby reducing the dose of drugs and improving treatment effectiveness. Finally, hydrogel can intelligently respond to environmental changes according to internal and external environmental stimuli so that anti-cancer active substances can be remotely controlled and released on demand. Combining the abovementioned advantages, hydrogel materials have transformed into a hit in the field of cancer treatment, bringing hope to further increase the survival rate and quality of life of patients with cancer.

Abstract:With the development in the field of biomaterials, research on alternative biocompatible materials has been initiated, and alginate in polysaccharides has become one of the research hotspots due to its advantages of biocompatibility, biodegradability and low cost. In recent years, with the further understanding of microscopic molecular structure and properties of alginate, various physicochemical methods of cross-linking strategies, as well as organic and inorganic materials, have led to the development of different properties of alginate hydrogels for greatly expanded applications. In view of the potential application prospects of alginate-based hydrogels, this paper reviews the properties and preparation of alginate-based hydrogels and their major achievements in delivery carrier, dressings, tissue engineering and other applications are also summarized. In addition, the combination of alginate-based hydrogel and new technology such as 3D printing are also involved, which will contribute to further research and exploration.
Anti-melanoma Effect and Action Mechanism of a Novel Chitosan-Based Composite Hydrogel Containing Hydroxyapatite Nanoparticles

Abstract:Hydroxyapatite nanoparticles (HANPs) have been increasingly regarded and reported due to their potential anti-tumor ability. Previously, we found that the rod-like HANPs had good application potential for cutaneous melanoma (CMM). To satisfy the actual requirements in repairing post-operative skin defects and inhibiting CMM recurrence after tumorectomy, we constructed a novel chitosan/alginate (CS/Alg) hydrogel containing the aforementioned HANPs. The in vitro cell experiments confirmed that activated mitochondrial-dependent apoptosis was tightly related to the anti-tumor ability of HANPs. Specifically, we further discovered several target proteins might be involved in abnormal activating Wnt, proteoglycans in cancer, oxidative phosphorylation and p53 signaling pathways. The in vivo animal experiments demonstrated that the HANPs-loaded CS/Alg hydrogel (CS/Alg/HANPs) had a similar effect on inhibiting tumor growth as HANPs, and CS/Alg hydrogel as well as phosphate buffered saline (PBS) group (control) not showed any effect, proving the key role of HANPs. The immunohistochemical staining demonstrated a tumor inhibition via the mitochondria-mediated apoptosis pathway, consistent with the in vitro evaluation. Moreover, CS/Alg/HANPs exhibited no additional biosafety risk to the functions of major organs. Overall, this CS/Alg/HANPs hydrogel has substantial application potential for treating CMM.

Abstract:Traditional cancer treatment is confronted with the problem of limited therapeutic effect, tissue defects, and lack of drug screening. Hydrogel scaffolds from biological macromolecules based on microfluidic technology are a promising candidate, which can mimic tumor microenvironments to screen personalized drugs, promote the regeneration of healthy tissues, and deliver drugs for enhanced localized antitumor treatment. This review summarizes the latest research on the composition of biomacromolecular hydrogel scaffolds, the architecture of hydrogel scaffolds from microfluidic technology, and their application in cancer therapy, including anti-tumor drug screening, anti-tumor treatment, and anti-tumor treatment and tissue repair. In addition, the potential breakthroughs of this innovative platform in the clinical transformation of cancer therapy are further discussed. The insights revealed in this review are intended to guide the utilization of microfluidic technology-based biomacromolecular hydrogel scaffolds in cancer therapy.

Abstract:The need for tumor postoperative treatments aimed at recurrence prevention and tissue regeneration have raised wide considerations in the context of the design and functionalization of implants. Supramolecular hydrogel is an ideal choice due to its compatibility, injectability, and ease of functionalization. Herein, we developed supramolecular hydrogel-loaded Prussian blue nanoparticles (PB NPs) for postoperative treatments. Biocompatible gellan gum was modified with host (cyclodextrin) or guest (adamantane) molecules to form supramolecular hydrogels (GEL) for local therapy. Hollow mesoporous Prussian blue nanoparticles (HMPB NPs) were prepared for use as photothermal agents and reactive oxygen species (ROS) scavengers. Further supramolecular hydrogels containing HMPB NPs loading doxorubicin (HMPB-DOX@GEL) were designed as multifunctional postoperative implants to ensure effective residual tumor elimination and oxidative relief. In vitro and in vivo experiments were carried out to verify the elimination of cancer cells and tumor recurrence prevention through combinational chemotherapy/photothermal therapy with HMPB-DOX@GEL. The tissue regeneration acceleration and in vivo ROS scavenging of HMPB@GEL were verified using diabetic mouse models of wound healing. The findings thus show that, because of the effective tumor recurrence prevention and wound healing acceleration of the multifunctional hydrogels developed here, they are promising for application in tumor postoperative treatments.

Abstract:Ovarian cancer, the most deadly gynecologic malignancy, is often resistant to conventional antitumor therapy due to various factors such as severe side effects, unexpected recurrence, and significant tissue damage. The limitations of current treatments and the resistance of invasive tumor cells contribute to these challenges. Hydrogel therapy has recently emerged as a potential treatment option for ovarian cancer, offering advantages such as controllability, biocompatibility, high drug loading capacity, prolonged drug release, and responsiveness to specific stimuli. Hence, the utilization of biodegradable hydrogels as carriers for chemotherapeutic agents has emerged as a significant concern in the field. Injectable hydrogel-based drug delivery systems, in particular, have demonstrated superior efficacy compared to traditional systemic chemotherapy for cancer treatment. The pliability of hydrogel therapy allows for access to anatomical regions that may be challenging for surgical intervention. This review article examines recent advancements in the application of hydrogels for diagnosing and treating ovarian cancer, while also proposing a novel direction for the use of hydrogel technology in this context. The objective of this article is to offer a novel point of reference and serve as a source of inspiration for the advancement of more precise and individualized cancer therapies.

Abstract:Technology in Cancer Research &Treatment, Volume 22, Issue , January-December 2023. Gastrointestinal cancer is one of the most malignant tumors with high morbidity and mortality, especially colorectal cancer, which has become the second leading cause of cancer-related deaths worldwide. Targeted drug treatment and precise endoscopic resection can significantly improve the overall survival rate and greatly extend the life span. Promising biomedical applications of hydrogels would represent hopeful therapeutic alternatives for patients with different kinds of diseases, particularly providing precise therapy for cancer patients. Although the intersection field of material science and biomedical science has made tremendous advances, major challenges remain. In this review, the application of hydrogel-based technology in cancer precision medicine is the focus of attention, which is the development trend of multidisciplinary cooperation in the future. First, we provide the current clinical landscape of hydrogel applications, and then we highlight precision oncology, including personalized drug treatment and accurate endoscopic intervention. Finally, we discuss major challenges for their clinical translation that have not yet been overcome and future perspectives on cancer precision medicine.

Abstract:The application of sodium alginate (SA) in the field of hydrogels has attracted much attention. However, it remains challenging to fabricate sodium alginate‐based biocompatible hydrogels with improved strength, high elasticity, porosity, and extraordinary adhesiveness. Herein, a hydrogel is constructed by SA and a copolymer of acrylic acid (AA) and meth acrylic acid (MAA), was synthesized via a free‐radical polymerization (FRP) and reinforced by using dynamic cross‐linker (Fe2+/Fe3+) with their carboxylate groups (COO−) like a chelating complex. The XPS validates the presence of dynamic Fe2+ (711 eV)/Fe3+ (714 eV) ions in the hydrogel scaffold. Porous structure contributes to improving the swelling rate (400%) which assists in drug delivery (80%) applications. The hydrogel has a well‐interconnected network with a crossover point (G′ = G′′) at 120 Pa with 8.52% strain and various factors viz. frequency temperature and time sweep study affect the gelation. The hydrogel exhibits a substantial surface area (25m2/g), pore depth size up to 350 nm, and height distribution histogram average size of 394 nm. The poly(AA‐co‐MAA) copolymer found actively targeting breast cancer MDA‐MB‐231 cells and exhibited biocompatibility against HEK‐293 cells and useful in water soluble controlled drug delivery.

Abstract:Immunotherapy represents a revolutionary paradigm in cancer management, showcasing its potential to impede tumor metastasis and recurrence. Nonetheless, challenges including limited therapeutic efficacy and severe immune‐related side effects are frequently encountered, especially in solid tumors. Hydrogels, a class of versatile materials featuring well‐hydrated structures widely used in biomedicine, offer a promising platform for encapsulating and releasing small molecule drugs, biomacromolecules, and cells in a controlled manner. Immunomodulatory hydrogels present unique capability for augmenting immune activation and mitigating systemic toxicity through encapsulation of multiple components and localized administration. Notably, hydrogels based on biopolymers have gained significant interest owing to their biocompatibility, environmental friendliness, and ease of production. This review delves into the recent advances in bio‐based hydrogels in cancer immunotherapy and synergistic combinatorial approaches, highlighting their diverse applications. We anticipate that this review will guide the rational design of hydrogels in the field of cancer immunotherapy, fostering clinical translation and ultimately benefiting patients. Abstract:<p>Immunotherapy that boosts the body's immune system to treat local and distant metastatic tumors has offered a new treatment option for cancer. However, cancer immunotherapy <em>via</em> systemic administration of immunotherapeutic agents often has two major issues of limited immune responses and potential immune-related adverse events in the clinic. Hydrogels, a class of three-dimensional network biomaterials with unique porous structures can achieve local delivery of drugs into tumors to trigger the antitumor immunity, resulting in amplified immunotherapy at lower dosages. In this review, we summarize the recent development of polymer-based hydrogels as drug release systems for local delivery of various immunotherapeutic agents for cancer immunotherapy. The constructions of polymer-based hydrogels and their local delivery of various drugs in tumors to achieve sole immunotherapy, and chemotherapy-, and phototherapy-combinational immunotherapy are introduced. Furthermore, a brief conclusion is given and existing challenges and further perspectives of polymer-based hydrogels for cancer immunotherapy are discussed.</p>

Abstract:Most existing hydrogels, even recently developed injectable hydrogels that undergo a reversible sol‒gel phase transition in response to external stimuli, are designed to gel immediately before or after implantation/injection to prevent the free diffusion of materials and drugs; however, the property of immediate gelation leads to very weak tumour-targeting ability, limiting their application in anticancer therapy. Therefore, the development of tumour-specific responsive hydrogels for anticancer therapy is imperative because tumour-specific responses improve their tumour-targeting efficacy, increase therapeutic effects, and decrease toxicity and side effects. In this review, we introduce the following three types of tumour-responsive hydrogels: 1) hydrogels that gel specifically at the tumour site; 2) hydrogels that decompose specifically at the tumour site; and 3) hydrogels that react specifically with tumours. For each type, their compositions, the mechanisms of tumour-specific responsiveness and their applications in anticancer treatment are comprehensively discussed.

Abstract:Locally administered drug delivery systems are promising as they allow to circumvent the side effects associated with systematic administration. In this study, we constructed multifunctional hydrogels by simply mixing commercial alginate (ALG) sols with glucose oxidase (GOx)-conjugated polyacrylic acid-stabilized iron oxide nanoparticles (GPI NPs) and Toll-like receptor 7/8 agonist resiquimod (R848). The injectable sols were able to transform into hydrogels (GPI/R848@ALG) by the ionic cross-linking between ALG and physiological Ca2+ to trap the therapeutic components within the hydrogel framework. Upon intratumoral injection, the hydrogels were employed for starvation therapy, promoted chemodynamic therapy and tumor-associated macrophages (TAMs) repolarization. The energy supply was blocked by consuming the intratumoral glucose via the GOx-catalyzed conversion of glucose into gluconic acid and hydrogen peroxide (H2O2). In vitro results showed that the generated H2O2 could be further converted into highly cytotoxic hydroxyl radicals (·HO) by the Fenton reaction to induce enhanced chemodynamic therapy. The TAMs repolarization studies in vitro exhibited that the GPI/R848@ALG hydrogels up-regulated the expression of CD86 by 63% and down-regulated the proportion of CD206 by 14% with a synergistic effect of the presence of Fe3O4 and R848, suggesting that the multifunctional hydrogels exert functions to direct the remodeling of TAMs from the tumor supportive M2-like phenotype to the tumor destructive M1-like phenotype to further contribute to the antitumor effect. Moreover, both in vitro and in vivo experiments demonstrate that the multifunctional hydrogels exhibit admirable antitumor performance towards 4T1 tumors. This work thereby provides a promising multifunctional nanoplatform for synergistic cancer starvation therapy, chemodynamic therapy and TAMs repolarization.

Abstract:Supramolecular peptide hydrogel (SPH) is a class of biomaterials self‐assembled from peptide‐based gelators through non‐covalent interactions. Among many of its biomedical applications, the potential of SPH in cancer therapy has been vastly explored in the past decade, taking advantage of its good biocompatibility, multifunctionality, and injectability. SPHs can exert localized cancer therapy and induce systemic anticancer immunity to prevent tumor recurrence, depending on the design of SPH. This review first gives a brief introduction to SPH and then outlines the major types of peptide‐based gelators that have been developed so far. The methodologies to tune the physicochemical properties and biological activities are summarized. The recent advances of SPH in cancer therapy as carriers, prodrugs, or drugs are highlighted. Finally, the clinical translation potential and main challenges in this field are also discussed.

Abstract:The proportion of individuals with bladder cancer has drastically increased, thereby rendering bladder cancer a focus of social concern and great challenge for human health management. Hydrogels with three-dimensional network structures have attracted considerable attention in the field of drug delivery for tumor therapy. Therefore, exploring hydrogel drug-delivery systems with excellent biocompatibility and targeting ability is a current research hotspot. Herein, a chitosan/dialdehyde sodium alginate/magnetic dopamine hydrogel was investigated for this purpose. The hydrogel was prepared from chitosan using sodium dialdehyde alginate and dopamine via grafting, crosslinking, and compounding. Furthermore, characterization of the product structure, targeted adhesion, and evaluation of drug delivery properties were performed. The prepared wall-adherent hydrogel materials exhibited excellent targeted drug-delivery and controlled-release properties. In addition, the effect of dopamine dosage on the performance of drug-loaded hydrogels was examined. In conclusion, the drug-delivery materials exhibited strong organ-wall adhesion, hydrogel properties, targeting ability, antibacterial and antimicrobial abilities (98%), and biocompatibility (99%). Based on these excellent properties, the constructed targeted hydrogels demonstrate considerable potential for drug delivery and bladder cancer treatment.
