Abstract:As nanomedicine-based clinical strategies have continued to develop, the possibility of combining chemotherapy and singlet oxygen-dependent photodynamic therapy (PDT) to treat pancreatic cancer (PaC) has emerged as a viable therapeutic modality. The efficacy of such an approach, however, is likely to be constrained by the mechanisms of drug release and tumor oxygen levels. In the present study, we developed an Fe(III)-complexed porous coordination network (PCN) which we then used to encapsulate PTX (PCN-Fe(III)-PTX) nanoparticles (NPs) in order to treat PaC via a combination of chemotherapy and PDT. The resultant NPs were able to release drug in response to both laser irradiation and pH changes to promote drug accumulation within tumors. Furthermore, through a Fe(III)-based Fenton-like reaction these NPs were able to convert H2O2 in the tumor site to O2, thereby regulating local hypoxic conditions and enhancing the efficacy of PDT approaches. Also these NPs were suitable for use as a T1-MRI weighted contrast agent, making them viable for monitoring therapeutic efficacy upon treatment. Our results in both cell line and animal models of PaC suggest that these NPs represent an ideal agent for mediating effective MRI-guided chemotherapy-PDT, giving them great promise for the clinical treatment of PaC.

Abstract:A novel dual‐modal imagingprobe in the second near‐infrared (NIR‐II) window is developed with "double assurances" for the accurate imaging and monitoring of H2S in vivo. High hydrogen sulfide (H2S) expression has been shown to play a crucial role in tumor growth and proliferation in colon cancer. So far, most probes used for monitoring H2S offer single‐modal imaging, which makes the collection of accurate information for tumor diagnosis difficult. In the present study, a novel dual‐modal imaging probe in the second near‐infrared (NIR‐II, 950‐1700) window is proposed and constructed by coating a uniform shell of polydopamine (PDA) on the surface of cuprous oxide (Cu2O)‐conjugated downconversion nanoparticles (DCNPs). Of note, DCNP‐Cu2O@PDA is in the "on" state, with emission centered at 1550 nm (irradiation using a 980nm laser) in non‐neoplastic tissues. Colon tumors with endogenous H2S overexpression can induce the in situ transformation of Cu2O to copper sulfide (Cu2‐XS). Owing to the competitive absorption between Cu2‐XS and DCNPs at a wavelength of 980 nm, the generated DCNP‐Cu2‐XS@PDA exhibits a quenched NIR‐II fluorescence (FL) signal. Meanwhile, Cu2‐XS exhibits a strong absorbance at 1250 nm, enhancing the NIR‐II photoacoustic (PA) signal accompanied by NIR‐II FL attenuation. The switching in PA and FL signals at the tumor site offered "double assurances" for the accurate monitoring of H2S.

Abstract:Hydrogen sulfide (H2S) content has been shown to be significantly increased in colon cancer tissue; imaging H2S in vivo enables the accurate diagnosis of colon cancer disease. Photoacoustic (PA) imaging is a biological imaging mode combining photoexcitation and ultrasonic detection, and high-resolution imaging in deep biological tissues can be achieved using PA probes in the second near-infrared biowindow (NIR-II). Nevertheless, existing materials can only be used as contrast agents to achieve the PA imaging of biomolecules in NIR-II, which still remains a challenge. To this end, water-soluble MoO3 nanoparticles were prepared, and it was found that PA imaging and photothermal therapy (PTT) of colon cancer tissue can be specifically activated in NIR-II by endogenous H2S. Therefore, MoO3 nanoparticles were used as an NIR-II nanoprobe for PA imaging and PTT of colon cancer tissue. Furthermore, by comparing the efficiency for the PA imaging and PTT of colon cancer in NIR-I and NIR-II, it was proven that in the NIR-II window, the background interference of the PA imaging for biological tissue was low, the photothermal efficiency was high, and the PTT effect was good. Abstract:A study has been made of the levels of 6-methylaminopurine and 5-methyl-cytosine in the DNA of Bacillus subtilis during thymine deprivation. While DNA synthesis was inhibited by thymine deprivation, DNA methylation continued. Base analysis indicated that this aberrant methylation involved an increase solely in the amount of 5-methylcytosine. These aberrant 5-methylcytosine residues were removed from the DNA during continued growth of bacteria in medium lacking thymine. In contrast, 5-methylcytosine residues synthesized during normal growth were relatively unaffected by thymine deprivation. The results are interpreted to indicate that the extensive DNA damage which occurs during thymine deprivation is due in part to exonuclease digestion of regions of DNA containing aberrant 5-methylcytosine residues.
Hydrogen Sulfide (H2S) activatable Photodynamic Therapy Against Colon Cancer by tunable FRET Effect

Abstract:Developing activatable photodynamic agents is becoming a promising mode for realizing selective photodynamic therapy (PDT) in cancer treatment. However, till now only a few H2S‐activatable photodynamic agents have been involved in this field. Here, we fabricated H2S‐activatable nano‐assemblies (P@TPPCY) for the treatment of colon cancer containing high concentration H2S. The H2S‐activatable photosensitizer (TPPCY) was synthesized by covalent conjugation between tetraphenylporphyrin (TPP) and heptamethine cyanine (Cy7), and then TPPCY was encapsulated by an amphiphilic polymer DSPE‐mPEG to form P@TPPCY nano‐assemblies. We demonstrated that the photosensitizing effect of TPP was efficiently quenched by Cy7 with strong absorption in the NIR region via fluorescence resonance energy transfer (FRET) effect. Interestingly, the FRET effect between Cy7 and TPP was attenuated by the decrease of absorption of Cy7 in the NIR region after the Cy7 reacted with H2S, and then the photosensitizing effect of TPP in P@TPPCY was activated. Strikingly, the P@TPPCY showed efficient H2S‐activatable photodynamic therapy (PDT) in vitro against HCT116 cells (human colon carcinoma cell line), thus this work provides a new method for realizing accurate treatment of colon cancer in virtue of high H2S concentration microenvironment.

Abstract:INTRODUCTION: Photothermal therapy can be synergistically combined with chemotherapy to improve the therapeutic effect for colon cancer. However, conventional therapeutic agents have side effects in normal tissues, limiting their application.OBJECTIVES: To reduce these side effects, a smart agent (Cur@HKUST-1@PVP) whose functionality is triggered by the high content of endogenous hydrogen sulfide in colon tumors was engineered for photoacoustic imaging-guided combination of photothermal therapy and chemotherapy for colon tumors.METHODS: After reacting with hydrogen sulfide, Cur@HKUST-1@PVP simultaneously generates CuS and releases curcumin. The generated CuS serves as an imaging agent for both photothermal therapy and photoacoustic imaging, while the released curcumin is used for chemotherapy.RESULTS: In vivo photoacoustic imaging experiments demonstrated that Cur@HKUST-1@PVP can be used for selectively imaging colon cancer tumors. In vivo experiments in mice for treatment suggested that the endogenous hydrogen sulfide-activated combination of photothermal therapy and chemotherapy has a better treatment effect that photothermal therapy or chemotherapy treatment alone.CONCLUSION: The endogenous hydrogen sulfide-activated Cur@HKUST-1@PVP agent developed herein shows great potential for the accurate diagnosis and effective treatment of colon cancer.

Abstract:Nanoscale metal-organic frameworks (nano MOFs) have emerged as a promising biomedical nanoplatform because of their unique properties. However, the exploration of nano MOFs in photoacoustic (PA) imaging is still limited. Here, a novel hydrogen sulfide (H2S)-activated nano copper-based MOF (Cu-MOF) was developed as a near-infrared (NIR) ratiometric PA probe for in vivo monitoring of endogenous H2S level and orthotopic colorectal cancer imaging via in situ reaction of nano Cu-MOFs with endogenous H2S that is closely associated with tumor growth and proliferation in colon cancer. The synthesized nano Cu-MOFs displayed excellent PA responsiveness towards tumor H2S level with high selectivity and rapid kinetics. The result suggests the developed probe may provide a unique opportunity to investigate the malignant behaviors of H2S-associated events in vivo.

Abstract:Developing nanomedicines with high-performance is a promising therapeutic strategy for cancer treatment. Herein, a multifunctional nanoplatform (CuS@COFs-BSA-FA/DOX) based on copper sulfide nanoparticles engineered covalent organic frameworks (CuS@COFs) has been established for synergistic photothermal therapy (PTT), chemotherapy and chemodynamic therapy (CDT). The high photothermal effect and excellent Fenton-like catalytic activity enabled CuS@COFs to generate a new minimally invasive PTT/CDT synergistic therapy. Moreover, with the pH-dependent fluorescence behavior and the inherited mesoporous structure, the nanocomposites can also be utilized as fluorescent probes for cancer cell imaging and drug-delivery carriers to encapsulate the chemotherapeutic agent, DOX. The densely coated PEI and BSA-FA layers on the surface of CuS@COFs not only increased the cancer-targeting efficiency but also prevented the nonspecific release of DOX from CuS@COFs. The acid tumor microenvironment and near-infrared light triggered the release of DOX for chemotherapy, which simultaneously augmented the CDT efficiency of CuS@COFs. Notably, the localized hyperthermia induced by PTT can further improve the CDT efficiency of the nanoplatform, leading to a synergistic PTT/chemotherapy/CDT effect. The nanoplatform possessed the capability of cancer cell-targeted imaging and achieved better therapeutic efficacy with negligible systemic toxicity both in vitro and in vivo. Our work opens up a powerful avenue for developing multifunctional COFs-based theranostic platform and shows strong potential in practical applications.

Abstract:The clinical applications of the photothermal therapy (PTT) and photodynamic therapy (PDT) are restricted by the limited tissue penetration depth of excitation light, while the enzyme catalysis therapy is hindered on account of the low treatment efficiency. Herein, we modify artificial nanozyme Pt nanoparticles on the surface of CuCo 2 S 4 nanoparticles to prepare ultrasmall copper-based ternary bimetallic chalcogenides CuCo 2 S 4 -Pt-PEG nanocomposites for PTT/PDT/enzyme catalysis synergistic therapy. The as-fabricated CuCo 2 S 4 -Pt-PEG nanocomposites display peroxidase (POD)-like and catalase (CAT)-like activities, which can catalyze hydrogen peroxide to emerge highly toxic hydroxyl radicals and oxygen, respectively. The integration of Pt nanozyme significantly improves the POD-like and CAT-like activity of CuCo 2 S 4 −PEG that can produce more hydroxyl radicals and oxygen to improve the effects of PDT. Furthermore, the nanocomposites exhibit outstanding photothermal properties derived from the enhanced photothermal conversion efficiency ( η = 78.46 %) exposed to 1064 nm NIR-II light irradiation with a low power density (0.8 W cm −2 ). In particular, the photothermal performance of CuCo 2 S 4 -Pt-PEG significantly enhances the enzyme-like activity. Additionally, the T 1 -weighted magnetic resonance and photoacoustic imaging endow CuCo 2 S 4 -Pt-PEG with the advantages of guiding the treatment process. The synergistic treatment demonstrates significant anticancer treatment efficiency while also elicits immune response that suppressed metastasis. This study could further promote the application of ternary bimetallic chalcogenides in cancer diagnosis and treatment.

Abstract:Photodynamic therapy (PDT) is a promising alternative and palliative therapeutic strategy for colorectal cancer (CRC). A novel photosensitizer with higher selectivity for CRC and fewer side effects is vital for clinical application. Given that the overexpression of hydrogen sulfide (H 2 S) in CRC, it is expected to provide a selective stimulus for activatable photosensitizers that in respond to the specific microenvironment. Herein, we report a novel development of metal-organic frameworks (MOFs) composed of meso-Tetra (4-carboxyphenyl) porphine (TCPP) and ferric ion (Fe 3+ ) through a facile one-pot process. Experiments both in vitro and in vivo reveal that the MOF is capable of depredating in response to the high content of H 2 S in tumor microenvironment of CRC. Accompanying with the degradation and release of TCPP, the fluorescence and photosensitivity effect is switched from "off" to "on", enabling the MOF to serve as a H 2 S activatable nano-photosensitizer for real-time fluorescence imaging-guided and targeted PDT of CRC.

Abstract:Simultaneous photodynamic/photothermal therapy (PDT/PTT) is a combination cancer treatment that combines the principles of both PDT and PTT to achieve an effective and comprehensive attack on cancer cells. However, both approaches require an external light source to generate reactive oxygen species (ROS) and thermal heat, which could potentially hinder the therapeutic efficacy due to the low tissue penetration of light in vivo. Chemodynamic therapy (CDT) is a type of therapy that uses the ROS generated by the chemical reaction between a prodrug and a catalyst to kill cancer cells. We developed a combination therapy of three modalities (PTT/PDT/CDT) using the metal-organic framework (MOF). The Cu2+-doped MOF PCN-224 nanostructures modified with platinum (Pt) cluster and folic acid (FA) provided excellent tumor targeting, photothermal conversion ability, and ROS generated by the Pt cluster and Cu2+ under 650 nm light irradiation. Moreover, this multimodel nanocomposite had an extremely low dark toxicity but excellent phototoxicity under the combination laser irradiation at 650 nm, both in vitro and in vivo. Therefore, our prepared folic acid-conjugated PCN-224/Pt/Cu2+ (FA-PPC) nanosphere could be applied as a very promising multimodal phototherapeutic agent for enhanced cancer therapy in future clinical applications.

Abstract:Photodynamic therapy (PDT) and catalytic therapy were promising treatment modes, but tumor hypoxia and poor catalytic activity severely limited their efficacies. Herein, using a porphyrin metal–organic framework (PCN-224) as nanocarrier, a platinum/palladium (Pt/Pd) dual-modified PCN-224 nanoprobe (PCN-224-Pt@Pd) with strong peroxidase (POD)/catalase (CAT)-like activities was developed, achieving photothermal-promoted PDT/catalytic therapy. Compared with single ultrasmall Pt modifying, CAT-like activity of Pt/Pd dual-modifying increased oxygen concentration from 6.24 to 9.35 mg/L, which improved singlet oxygen ( 1 O 2 ) yield from 63.8 % to 82.9 %. Moreover, POD-like activity of Pt/Pd dual-modifying significantly accelerated hydroxyl radicals (·OH) generation. Importantly, PCN-224-Pt@Pd possessed near-infrared II (NIR-II) photothermal effect with a high efficiency (55.6 %), which further promoted ·OH production. Under combined therapy of PCN-224-Pt@Pd, the cell survival rate greatly reduced to 5.8 %, and the tumors were cured, suggesting NIR-II photothermal-enhanced PDT/catalytic therapy.

Abstract:Explorations on ferroptosis combined with other tumor therapies has attracted much attention in recent years. To enhance the cargo loaded capacity of the nanoparticles and integrate the ferroptosis as well as apoptosis, here, we reported the Pt-doped porous FeS 2 nanoparticles (Pt/pFeS 2 NPs) synthesized by solvothermal method to realize the photothermal enhanced photodynamic/ferroptosis antitumor combination therapy, with glucose oxidase (GOX) loaded and 4T1 cell membrane coated as the multifunctional Pt/pFeS 2 @ nanovesicles (NVs). The porous structure provided more sites for Fenton reaction as well as photodynamic effect, which dramatically improve the level of reactive oxygen species (ROS). Additionally, the loaded GOX enhanced the generation of endogenous hydrogen peroxide (H 2 O 2 ) and reduced the pH value of the tumor microenvironment (TME), further promoting the Fenton reaction for ferroptosis. Moreover, Pt catalyzed the degradation of H 2 O 2 to released oxygen, even facilitating the photodynamic therapy (PDT) efficacy in the hypoxic TME. The rate of Fenton reaction and photocatalytic reaction were both significantly improved, leading to the acceleration of ferroptosis and PDT with the increase of temperature under laser irradiation at 808 nm. Additionally, the in vivo antitumor therapy proved that Pt/pFeS 2 @ NVs efficiently accumulated at tumor sites for photoacoustic imaging. To sum up, the multifunctional Pt/pFeS 2 @ NVs could achieve photothermal enhanced photodynamic/ferroptosis combination antitumor therapy and significantly inhibit the growth of 4T1 tumor without side effects, shedding light on the promising application in photoacoustic image-guided tumor therapy system.

Abstract:Overproduced hydrogen sulfide (H2 S) is of vital importance for the progress of colon cancer and promotes cancer cellular proliferation. Devising pharmacological nanomaterials for tumor-specific H2 S activation will be significant for precise colon cancer treatment. Herein, a biocompatible fusiform iron oxide-hydroxide nanospindles (FeOOH NSs) nanosystem for magnetic resonance imaging (MRI), ferroptosis, and H2 S based cascade reaction-enhanced combinational colon cancer treatment is developed. The FeOOH NSs can effectively scavenge endogenous H2 S via the reduction reaction to prohibit the growth of CT26 colon cancer. The cascade produced FeS driven by overexpressed H2 S exhibits near-infrared-triggered photothermal therapy capability and Fe2+ -mediated ferroptosis functionality. Meanwhile, the as-prepared FeOOH NSs can light up tumor tissues as a potent MRI contrast agent. Additionally, FeOOH NSs present desirable biosafety in a murine model for up to three months and avoid any long-term toxicity. Furthermore, it is found that these H2 S-responsible nanotheranostics do not cause any cure effects on other cancer types, such as 4T1 breast cancer. Overall, the findings illustrate that the biocompatible FeOOH NSs can be successfully employed as a theranostic for specifically treating colon cancer, which may promote the clinical translation and development of H2 S-responsive nanoplatforms.
Photothermal and ferroptosis synergistic therapy for liver cancer using iron-doped polydopamine nanozymes

Abstract:An innovative nanozyme, iron-doped polydopamine (Fe-PDA), which integrates iron ions into a PDA matrix, conferred peroxidase-mimetic activity and achieved a substantial photothermal conversion efficiency of 43.5 %. Fe-PDA mediated the catalysis of H2O2 to produce toxic hydroxyl radicals (•OH), thereby facilitating lipid peroxidation in tumour cells and inducing ferroptosis. Downregulation of solute carrier family 7 no. 11 (SLC7A11) and solute carrier family 3 no. 2 (SLC3A2) in System Xc- resulted in decreased intracellular glutathione (GSH) production and inactivation of the nuclear factor erythroid 2-related factor 2 (NRF2)-glutathione peroxidase 4 (GPX4) pathway, contributing to ferroptosis. Moreover, the application of photothermal therapy (PTT) enhanced the effectiveness of chemodynamic therapy (CDT), accelerating the Fenton reaction for targeted tumour eradication while sparing adjacent non-cancerous tissues. In vivo experiments revealed that Fe-PDA significantly hampered tumour progression in mice, emphasizing the potential of the dual-modality treatment combining CDT and PTT for future clinical oncology applications.

Abstract:The specific in situ generation and activation of therapeutic agents with high spatiotemporal precision is expected to revolutionize cancer treatment. Here, we develop an intelligent nanoplatform (termed as NP-Cu), which is constructed by assembling photosensitizer chlorin e6 (Ce6), hypoxia-responsive prodrug banoxantrone (AQ4N) with clickable dibenzocyclooctyne (DIBO) functionalized lysine (D-K), and cyclen-Cu2+ complex, for improving combination anticancer therapy. Cyclen-Cu2+ complex-induced photodynamic therapy (PDT) quenching in NP-Cu can be effectively and selectively activated by tumor-overproduced hydrogen sulfide (H2S). More importantly, the reaction of endogenous H2S with Cu2+ can generate photothermal agent copper sulfide (CuS) for photothermal therapy (PTT). Furthermore, with the activation of PTT and PDT, intracellular hypoxic stress is amplified to trigger AQ4N-associated chemodynamic therapy (CDT), leading to light-enhanced cascade therapy of PDT, PTT and CDT. Therefore, we present a simple and practical strategy for developing pathological stimuli responsive combination therapy, which has the potential of advancing precision cancer medicine.

Abstract:Photoacoustic agents combing photodynamic therapy (PDT) and photothermal therapy (PTT) functions have emerged as potent theranostic agents for combating cancer. The molecular approaches for enhancing the near-infrared (NIR)-absorption and maximizing non-radiative energy transfer are essential for effective photoacoustic imaging (PAI) and therapy applications. In addition, such molecules with high specificity and affinity to cancer cells are urgently needed, which would further decrease the side effect during treatments. In this study, we applied a heavy-atom engineering strategy and introduced p-aminophenol, -thio, and -seleno moieties into NIR heptamethine cyanine (Cy7) skeleton (Cy7-X-NH2, X = O, S, Se) to significantly increase photothermal conversion efficiency for PTT and promote intersystem crossing for PDT. Additionally, we designed a series of nitroreductase (NTR)-activated photoacoustic probes (Cy7-X-NO2, X = O, S, Se), and target hypoxic tumors with NTR overexpression. Our prostate cancer targeting probe, Cy7-Se-NO2-KUE, exhibited specific tumor photoacoustic signals and effective tumor killing through outstanding synergistic PTT/PDT in vivo. These findings highlighted a versatile strategy for cancer photoacoustic diagnosis and enhanced phototherapy.

Abstract:During ground maintenance on an F-14 aircraft, a worker was removing the Mark 124 cartridge activated devices (CADS) from the aircraft when a second worker entered the cockpit of the aircraft and energized the electrical system, causing the four CADS to detonate. One of the four CADS became an airborne projectile. It struck the first worker in front of the right arm, passed through his chest, and became embedded in his thoracic spine. An immediate concern at autopsy was whether or not the device retained any explosive potential. Recommendations for autopsy procedures in cases involving military ordnance are discussed.
