Abstract:The genus Polygonatum, a well-known homology plant of medicine and food, has confirmed several biological activities and great market utilization value. A growing number of researches have shown that polysaccharides are one of the primary bioactive components of the Polygonatum spp. and present multiple biological activities both in vitro and in vivo methods, such as antioxidant activity, immunity enhancement effect, anti-inflammatory effect, anti-osteoporosis effect, and treatment of diabetes mellitus. This review summarizes the available information on the approaches used for GPPs extraction, purification, and structural characterization, and discusses the diverse biological activities of GPPs to support their potential application value in the functional foods, pharmaceuticals field, and cosmetics areas. New valuable insights for future research regarding GPPs were also proposed in the fields of the natural production of bio-based functional materials, functional foods, and therapeutic agents.

Abstract:Tea polysaccharide conjugates (TPS conjugates) are receiving worldwide attention in the recent decades, after they were recognized as critical components of coarse tea, and were responsible for their therapeutic effects in patients with diabetes. Existing evidences indicate that TPS conjugates not only benefit human health, but also can be used as carriers for drugs and as films for food packaging, and as emulsifiers. Despite their many practical applications, molecular structures of TPS conjugates are not well worked out yet, making their mechanisms of action difficult to understand. Their structural variations are attributed mainly to the differences in the processing of different tea. This comprehensive review summarises the compositions, structural characteristics, and bioactivities of TPS conjugates from different tea-leaf products, discusses the relationships between their compositions and structures, and the specific functions, and suggests the conjugation among polysaccharides, proteins and polyphenols occurring in tea leaves during processing which is mainly responsible for the alteration in composition and structure of TPS conjugates derived from different kinds of tea.

By conducting a comprehensive analysis of existing literature and research achievements, we strive to establish a comprehensive and systematic framework that can serve as a reference and guide for further investigations into pectic polysaccharides. Abstract:In recent decades, the polysaccharides from the medicinal plants have attracted a lot of attention due to their significant bioactivities, such as anti-tumor activity, antioxidant activity, anticoagulant activity, antidiabetic activity, radioprotection effect, anti-viral activity, hypolipidemic and immunomodulatory activities, which make them suitable for medicinal applications. Previous studies have also shown that medicinal plant polysaccharides are non-toxic and show no side effects. Based on these encouraging observations, most researches have been focusing on the isolation and identification of polysaccharides, as well as their bioactivities. A large number of bioactive polysaccharides with different structural features and biological effects from medicinal plants have been purified and characterized. This review provides a comprehensive summary of the most recent developments in physiochemical, structural features and biological activities of bioactive polysaccharides from a number of important medicinal plants, such as polysaccharides from Astragalus membranaceus, Dendrobium plants, Bupleurum, Cactus fruits, Acanthopanax senticosus, Angelica sinensis (Oliv.) Diels, Aloe barbadensis Miller, and Dimocarpus longan Lour. Moreover, the paper has also been focused on the applications of bioactive polysaccharides for medicinal applications. Recent studies have provided evidence that polysaccharides from medicinal plants can play a vital role in bioactivities. The contents and data will serve as a useful reference material for further investigation, production, and application of these polysaccharides in functional foods and therapeutic agents.

Characterization and macrophage immunomodulatory activity of two polysaccharides from the flowers of Paeonia suffruticosa Andr.

Abstract:Paeonia suffruticosa Andr. has a long history of cultivation in China and its flower possesses high nutritional and medicinal value. Characterization and macrophage immunomodulatory activity of the two polysaccharides from the flowers of Paeonia suffruticosa Andr., PSAP-1 and PSAP-2, purified by anion exchange chromatography followed by size exclusion chromatography, were investigated in the present study. Results showed that PSAP-1, with a molecular weight of 155 kDa, was mainly composed of glucuronic acid, glucose, arabinose and galactose with molar percentages of 0.83, 11.53, 18.98 and 68.96% respectively, and PSAP-2, with a molecular weight of 210 kDa, consisted of rhamnose, galacturonic acid, glucose, arabinose and galactose with molar percentages of 9.13, 8.35, 12.20, 14.75 and 55.57% respectively. Immunostimulatory activity evaluation in vitro revealed that the PSAP-1 and PSAP-2 could significantly stimulate the proliferation of Raw264.7 cells in dose-dependent manner and further activate Raw264.7 cells by releasing immunoactive molecules such as nitric oxide, tumor necrosis factor (TNF)-α and interleukin (IL)-6. In addition, PSAP-2 had higher immunomodulatory activity than PSAP-1. The above results suggested that both PSAP-1 and PSAP-2 had potent immunostimulatory activity and could be explored as a potential natural immunomodulatory agent in medicine or functional food fields.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Ethnopharmacological relevance</h3><p><em>Paeonia</em>, which comprises approximately 52 shrubs or herbaceous perennials around the world, is the only genus of the Paeoniaceae and is pervasively distributed in Asia, southern Europe, and North America. Many species of the genus <em>Paeoni</em>a have been used for centuries in ethnomedical medical systems.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Aim of the review</h3><p>The present study aims to summarize the traditional uses, clinical applications, and toxicology of the genus <em>Paeonia</em>, to critically evaluate the state-of-the-art phytochemical and pharmacological studies of this genus published between 2011 and 2020, and to suggest directions for further in-depth research on <em>Paeonia</em> medicinal resources.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Materials and methods</h3><p>Popular and widely used databases such as PubMed, Scopus, Science Direct, and Google Scholar were searched using the various search strings; from these searches, a number of citations related to the traditional uses, phytochemistry, biological activities, clinical application, and toxicology of the genus <em>Paeonia</em> were retrieved.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The use of 21 species, 2 subspecies, and 7 varieties of the genus <em>Paeonia</em> as traditional herbal remedies has been reported, and many ethnomedicinal uses, such as the treatment of hematemesis, blood stasis, dysmenorrhea, amenorrhea, epilepsy, spasms, and gastritis, have been recorded. The roots and root bark are the most frequently reported parts of the plants used in medicinal applications. In phytochemical investigations, 451 compounds have been isolated from <em>Paeonia</em> plants to date, which contains monoterpenoid glucosides, flavonoids, tannins, stilbenes, triterpenoids and steroids, and phenols. Studies of their pharmacological activities have revealed the antioxidant, anti-inflammatory, antitumour, antibacterial, antiviral, cardiovascular protective, and neuroprotective properties of the genus <em>Paeonia.</em> In particular, some bioactive extracts and compounds (total glucosides of peony (TGP), paeonol, and paeoniflorin) have been used as therapeutic drugs or tested in clinical trials. In addition to the "incompatibility" of the combined use of "shaoyao" and <em>Veratrum nigrum</em> L. roots in traditional Chinese medicine theory, <em>Paeonia</em> was considered to have no obvious toxicity based on the available toxicological tests.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>A large number of phytochemical and pharmacological reports have indicated that <em>Paeonia</em> is an important medicinal herb resource, and some of its traditional uses including the treatment of inflammation and cardiovascular diseases and its use as a neuroprotective agent, have been partially confirmed through modern pharmacological studies. Monoterpenoid glucosides are the main active constituents. Although many compounds have been isolated from <em>Paeonia</em> plants, the biological activities of only a few of these compounds (paeoniflorin, paeonol, and TGP) have been extensively investigated. Some paeoniflorin structural analogues and resveratrol oligomers have been preliminarily studied. With the exception of several species (<em>P. suffruticosa</em>, <em>P. ostii</em>, <em>P. lactiflora</em>, and <em>P. emodi)</em> that are commonly used in folk medicine, many medicinal species within the genus do not receive adequate attention. Conducting phytochemical and pharmacological experiments on these species can provide new clues that may lead to the discovery of medicinal resources. It is necessary to identify the effective phytoconstituents of crude extracts of <em>Paeonia</em> that displayed pharmacological activities by bioactivity-guided isolation. In addition, comprehensive plant quality control, and toxicology and pharmacokinetic studies are needed in the future studies.</p>

Abstract:The genus Polygonatum is gaining increasing attention from nutrition experts as well as health-conscious consumers because of its excellent performance in providing nutrients. Among these plants, Polygonatum sibiricum and Polygonatum odoratum have been selected for inclusion in China's Medicinal Food Directory due to their high safety profile. Polysaccharides are considered the main functional component and one of the main active ingredients of the plant. In addition, polysaccharides from genus Polygonatum have a variety of nutritional, biological and health-promoting properties, such as immunomodulatory, anti-inflammatory, cardiovascular protective, neuroprotective, antitumor, antidiabetic, antiosteoporosis, and hepatoprotective properties. This paper reviews the origin, extraction, purification, structural characteristics, biological activity, safety, toxicological evaluation, and structure-activity relationship of polysaccharides from the genus Polygonatum . Ultimately, we hope that this work can provide a more useful reference for understanding the polysaccharide structure and developing of new functional foods from polysaccharides of the genus Polygonatum .

Abstract:Two fractions of polysaccharides, named PTPS-1 and PTPS-2, was extracted and purified from puerh tea by Sephacryl S-300 column chromatography.The physicochemical properties of these two polysaccharides were investigated by high-performance liquid chromatography (HPLC), Fourier Transform IR spectra, scanning electron microscope (SEM) and atomic force microscope (AFM).Analysis of chemical compositions (protein, neutral sugars, uronic acid and monosaccharide composition) suggested that they were both kinds of acid heteropolysaccharides bound with protein, and contained seven monosaccharides with different molar ratio.Meanwhile, evaluation of antioxidant activities by in vitro assays of DPPH, ABTS and FRAP showed that PTPS-1 demonstrated stronger antioxidant ability than PTPS-2.Similarly, PTPS-1 exhibited remarkable inhibitory potential on α-glycosidase in vitro, significantly stronger than that of PTPS-2 and acarbose.Moreover, the results from animal test indicated PTPS-1 possessed significant inhibition on postprandial hyperglycemia in diabetic mice compared with the model group.

Abstract:Platycodon grandiflorum , a globally recognized medicinal and edible plant, possesses significant nutritional value and pharmacological value. In traditional Chinese medicine, it has the effects of tonifying the spleen and replenishing the Qi , moistening the lung and relieving the cough, clearing the heat and detoxifying, and relieving the pain. Accumulating evidence has revealed that the polysaccharides from P. grandiflorum (PGPs) are one of the major and representative biologically active macromolecules and have diverse biological activities, such as immunomodulatory activity, anti-inflammatory activity, anti-tumor activity, regulation of the gut microbiota, anti-oxidant activity, anti-apoptosis activity, anti-angiogenesis activity, hypoglycemic activity, anti-microbial activity, and so on. Although the polysaccharides extracted from P. grandiflorum have been extensively studied for the extraction and purification methods, structural characteristics, and pharmacological activities, the knowledge of their structures and bioactivity relationship, toxicologic effects, and pharmacokinetic profile is limited. The main purpose of the present review is to provide comprehensively and systematically reorganized information on extraction and purification, structure characterizations, and biological functions as well as toxicities of PGPs to support their therapeutic potentials and sanitarian functions. New valuable insights for future research regarding PGPs were also proposed in the fields of therapeutic agents and functional foods.

Abstract:Background Genus Paeonia, which is the main source of Traditional Chinese Medicine (TCM) Paeoniae Radix Rubra (Chishao in Chinese), Paeoniae Radix Alba (Baishao in Chinese) and Moutan Cortex (Mudanpi in Chinese), is also rich in active pharmaceutical ingredient such as monoterpenoid glycosides (MPGs). MPGs from Paeonia have extensive pharmacological effects, but the pharmacological effects and molecular mechanisms of MPGs has not been comprehensively reviewed. Purpose MPGs compounds are one of the main chemical components of the genus Paeonia , with a wide variety of compounds and strong pharmacological activities, and the structure of the mother nucleus-pinane skeleton is similar to that of a cage. The purpose of this review is to summarize the pharmacological activity and mechanism of action of MPGs from 2012 to 2023, providing reference direction for the development and utilization of Paeonia resources and preclinical research. Methods Keywords and phrases are widely used in database searches, such as PubMed, Web of Science, Google Scholar and X-Mol to search for citations related to the new compounds, extensive pharmacological research and molecular mechanisms of MPGs compounds of genus Paeonia. Results Modern research confirms that MPGs are the main compounds in Paeonia that exert pharmacological effects. MPGs with extensive pharmacological characteristics are mainly concentrated in two categories: paeoniflorin derivatives and albiflflorin derivatives among MPGs, which contains 32 compounds. Among them, 5 components including paeoniflorin, albiflorin, oxypaeoniflorin, 6'-O-galloylpaeoniflorin and paeoniflorigenone have been extensively studied, while the other 28 components have only been confirmed to have a certain degree of anti-inflammatory and anticomplementary effects. Studies of pharmacological effects are widely involved in nervous system, endocrine system, digestive system, immune system, etc., and some studies have identified clear mechanisms. MPGs exert pharmacological activity through multilateral mechanisms, including anti-inflammatory, antioxidant, inhibition of cell apoptosis, regulation of brain gut axis, regulation of gut microbiota and downregulation of mitochondrial apoptosis, etc. Conclusion This systematic review delved into the pharmacological effects and related molecular mechanisms of MPGs. However, there are still some compounds in MPGs whose pharmacological effects and pharmacological mechanisms have not been clarified. In addition, extensive clinical randomized trials are needed to verify the efficacy and dosage of MPGs.

Abstract:Two carboxymethylated polysaccharides (CPSP30, CPSP60) were obtained from the petals of Paeonia x suffruticosa Andrews (PSP30, PSP60, respectively) with different molecular weight. Chemical characterization, morphological properties and antioxidant activities of PSP and CPSP were determined. PSP were mainly composed of galactose, glucose, fructose and galacturonic with different molar percentages. CPSP were mainly composed of arabinose, galactose, glucose with different molar percentages. The signals of carboxymethyl groups were found in Fourier transform infrared spectroscopy (FT-IR) and 13 C nuclear magnetic resonance (NMR) spectra. Scanning electron microscopy (SEM) indicated that the original polysaccharides had a smooth surface, while the modified polysaccharides had an irregular lamellar structure. Thermal analysis indicated that the modified polysaccharides (CPSP30, CPSP60) presented higher thermal stability than the original polysaccharides. All polysaccharides presented typical structures of non-crystalline compounds. Carboxymethylation did not destroy the triple-helical structures of the polysaccharides. With its lower molecular weight, CPSP30 had higher scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH), OH, and Fe 2+ chelating ability than PSP30. The scavenging DPPH and OH abilities of both derivatives were higher than that of original polysaccharides. This study demonstrates that carboxymethylation improved the solubilities and antioxidant abilities of the original polysaccharides, thus enhancing their potential for application in food and pharmaceutical industry.

Abstract:Protein aggregation is a major bottleneck during the bacterial production of recombinant proteins. In general, the induction of gene expression at sub‐optimal growth temperatures improves the solubility of aggregation‐prone polypeptides and minimizes inclusion body (IB) formation. However, the effect of low temperatures on the quality of the recombinant protein, especially within the insoluble cell fraction, has been hardly ever explored. In this work, we have examined the conformational status of a recombinant GFP protein when produced in Escherichia coli below 37°C. As expected, the fraction of aggregated protein largely decreased at lower temperatures, while the conformational quality of both soluble and aggregated GFP, as reflected by its specific fluorescence emission, progressively improved. This observation indicates that physicochemical conditions governing protein folding affect concurrently the quality of the soluble and the aggregated forms of a misfolding‐prone protein, and that protein misfolding and aggregation are clearly not coincident events. Abstract:Plant polysaccharides (PPS) composed of more than 10 monosaccharides show high safety and various pharmacological activities, including immunoregulatory, antitumor, antioxidative, antiaging, and other effects. In recent years, emerging evidence has indicated that many PPS are beneficial for metabolic diseases, such as cardiovascular disease (CVD), diabetes, obesity, and neurological diseases, which are usually caused by the metabolic disorder of fat, sugar, and protein. In this review, we introduce the common characteristics and functional activity of many representative PPS, emphasize the common risks and molecular mechanism of metabolic diseases, and discuss the pharmacological activity and mechanism of action of representative PPS obtained from plants including Aloe vera, Angelica sinensis, pumpkin, Lycium barbarum, Ginseng, Schisandra chinensis, Dioscorea pposite, Poria cocos, and tea in metabolic diseases. Finally, this review will provide directions and a reference for future research and for the development of PPS into potential drugs for the treatment of metabolic diseases.

Abstract:Phellinus linteus (P. linteus) is a famous Chinese medicine and has a long history in China. In recent years, P. linteus polysaccharides (PLPs) have attracted extensive attention because of their biological activities such as anti-bacteria, anti-aging, anti-oxidation, anti-inflammation, anti-tumor, hepatoprotective effect and hypoglycemic effect. In this review, we systemically summarized the advances in extractions, purifications and structural characterizations of PLPs, and also analyzed their biological functions and molecular mechanisms. Meanwhile, the structure-activity relationships of PLPs are closely related to their anti-oxidation and anti-tumor activities. So far, the applications of PLPs are still very limited, further exploring structure-activity relationships, biological functions and their mechanisms of PLPs will promote to develop functional foods.

Abstract:Phellinus spp. is one of the largest genera of Hymenochaetaceae with approximately 220 species, such as P. vaninii, P. buamii, P. linteus, and P. igniarius, these species are considered as precious food supplements and medicinal ingredients in China, Korea, Japan, and other Asian countries for over 2000 years. Phellinus spp. contains abundant bioactive polysaccharides and other key components (e.g., phenolics, terpenes, steroids, etc.). Pharmacological investigations have confirmed that bioactive polysaccharides and other important secondary metabolites from Phellinus spp. possess multiple health-promoting benefits, including antitumor, immunomodulatory, anti-inflammatory, antidiabetic, antioxidant, and antimicrobial effects. However, comprehensive evaluations on the preparation and structural characteristics, bioactivities, and toxicology of these functional components (e.g., polysaccharides, phenolics, terpenes, steroids) from various Phellinus spp. species are very limited, which may restrict the practical application of Phellinus spp. This review summarizes the physicochemical characteristics, pharmacological activities, and possible mechanisms of bioactive components from Phellinus spp. according to published studies from 2017 to 2022. It also surveyed the toxicological assessment for safety and applications of different Phellinus spp. species. This review aims to provide useful references and promising directions for the comprehensive development and utilization of Phellinus spp. in functional foods, pharmaceuticals, and cosmetics.

Abstract:Pueraria lobata (Willd.) Ohwi is an important resource with dual functions in medicine and food since ancient times. Polysaccharides are the main bioactive component of P. lobata and have various bioactivities, such as antidiabetic, antioxidant, immunological activities, etc. Due to the distinctive bioactivity of P. lobata polysaccharides (PLPs), the research on PLPs is booming. Although a series of PLPs have been isolated and characterized, the chemical structure and mechanism are unclear and need further study. Here, we reviewed recent progress in isolation, identification, pharmacological properties, and possible therapeutic mechanisms of PLPs to update awareness of these value-added natural polysaccharides. Besides, the structure-activity relationships, application status, and toxic effects of PLPs are highlighted and discussed to afford a deeper understanding of PLPs. This article may provide theoretical insights and technical guidance for developing PLPs as novel functional foods.

Abstract:Abstract Penthorum chinense Pursh is a well-known traditional Chinese medicine, however, little attention has been paid to the polysaccharides of P. chinense (PCP). Therefore, it is great significance to investigate the characteristics and activities of PCP. In this study, response surface methodology was applied to optimize the extraction parameters of PCP. Then, two polysaccharides fractions (PCP-1 and PCP-2) were purified from PCP by using DEAE-52 chromatography, and there preliminary chemical properties and in vitro antioxidant activities were investigated. Results revealed that the optimum extraction conditions of PCP were identified as follow: ratio of water to raw material 20.6 mL/g; extraction time 3.5 h and extraction temperature 85°C. Based on these conditions, the maximum yield of PCP was 3.12% ± 0.19%. Purified fractions PCP-1 and PCP-2 were all acidic heteropolysaccharides, and PCP-1 mainly consisted of galactose and arabinose while PCP-2 mainly consisted of rhamnose, galacturonic acid, galactose and arabinose. Moreover, PCP-2 exhibited stronger scavenging activities against DPPH radical, hydroxyl radical and superoxide anion radical and chelating activity on Fe 2+ in vitro .
