Abstract:Largemouth bass iridovirus (LMBV) can cause high mortality and lead to heavy economic loss in the cultivation of largemouth bass, but there was no effective treatment. Here, the present study constructed a recombinant Pichia pastoris expressing LMBV major capsid protein ( MCPD ). The recombinant GS115-pW317- MCPD was then used to immunize largemouth bass via oral administration, and mucosal immune response mediated by immunoglobulins (Igs) was measured after oral immunization. Serum antibody levels were measured by ELISA, neutralizing antibody titers were determined by serum neutralization test (SNT), antigen presentation-related gene expressions were detected by RT-PCR, and the histopathological characteristics of immunized fish were assessed after challenging with 0.1 ml 10 7.19 TCID 50 /ml LMBV. The relative percentage survival (RPS) was also determined. Our results showed that the serum antibody titers of immunized fish were significantly higher than that of control groups (P < 0.05). IgT and IgM expressions in gut were increased significantly after vaccination with GS115-pW317- MCPD ; however, much stronger response in gut was observed as compared with gill. The expression levels of major histocompatibility complex ( MHC ) II, CD 8, and T-cell receptor ( TCR ) were significantly elevated in GS115-pW317- MCPD group (P < 0.05), while CD 4 and MHC I transcription levels remained unchanged after oral immunization ( P > 0.05). The RPS of fish orally immunized with 1.0 × 10 8 CFU/g GS115-pW317- MCPD was reached up to 41.6% after challenge with 0.1 ml 10 9.46 TCID 50 /ml LMBV. Moreover, orally immunizing with GS115-pW317- MCPD can relieve the pathological damage caused by LMBV. Therefore, GS115-pW317- MCPD showed a promising potential against LMBV.

Abstract:Largemouth bass ranavirus (LMBV) seriously affects the development of largemouth bass (Micropterus salmoides) industry and causes huge economic losses. Oral vaccine can be a promising method for viral disease precaution. In this study, MCP2α was identified as a valuable epitope region superior to MCP and MCP2 of LMBV by neutralizing antibody experiments. Then, recombinant Lactobacillus casei expressing the fusion protein MCP2αC (MCP2α as antigen, C represents flagellin C from Aeromonas hydrophila as adjuvant) on surface was constructed and verified. Further, PLA microsphere vaccine loading recombinant MCP2αC L. casei was prepared. The PLA microspheres vaccine were observed by scanning electron microscopy and showed a smooth, regular spherical surface with a particle size distribution between 100 and 200 μm. Furthermore, we evaluated the tolerance of PLA-MCP2αC vaccine in simulated gastric fluid and simulated intestinal fluid, and the results showed that PLA-MCP2αC can effectively resist the gastrointestinal environment. Moreover, the protective effect of PLA-MCP2αC against LMBV was evaluated after oral immunization and LMBV challenge. The results showed that PLA-MCP2αC effectively up-regulated the activity of serum biochemical enzymes (T-SOD, T-AOC, LZM, complement C3) and induced the mRNA expression of representative immune genes (IL-1β, TNF-α, IFN-γ, MHC-IIα, Mx, IgM) in spleen and head kidney tissues. The survival rate of largemouth bass vaccinated with PLA-MCP2αC increased from 24 % to 68 %. Meanwhile, PLA-MCP2αC inhibited the LMBV burden in spleen, head kidney and liver tissues and attenuated tissue damage in spleen. These results suggested that PLA-MCP2αC can be used as a candidate oral vaccine against LMBV infection in aquaculture.

Abstract:Largemouth bass virus (LMBV) poses a significant threat to largemouth bass farming, leading to substantial economic losses. In December 2022, massive largemouth bass juveniles died at a fish farm in the city of Xinxiang, China. Through a series of experiments, we conclusively identified LMBV as the causative pathogen. The affected fish displayed anorexia, lethargy, and hemorrhage at the pectoral and caudal fin base. No parasites or pathogenic bacteria were detected on the body surface or gills, or isolated from the diseased fish. Severe hemorrhage, lymphocyte infiltration, and extensive necrosis were observed in the liver, spleen, intestine, and stomach of the moribund fish. The tissue homogenate from the diseased fish induced epithelioma papulosum cyprini cells (EPC) cell death, while no such effects were observed in grouper spleen (GS) cells. Sequence similarity analysis of the major capsid protein (MCP) indicated the virus shared 100% similarity with the LMBV-FS2021 strain, placing it within the Ranavirus genus. Transmission electron microscopy (TEM) observations revealed plenty of hexagonal virions accumulated in the cytoplasm of infected EPC cells. Artificial infection demonstrated that LMBV-XX01 was highly fatal to Micropterus salmoides juveniles, with an LD 50 of 10 3.081 TCID 50 /fish. RT-qPCR detection confirmed that LMBV appeared in all sampled tissues of the challenged largemouth bass, with significantly higher viral loads detected in the liver and heart compared to other tissues. Additionally, we successfully obtained a highly purified recombinant MCP of LMBV and developed two strains of monoclonal antibodies targeting MCP of LMBV-XX01. Overall, our findings provide valuable materials and insights for the design of prevention strategies and the development of detection methods for LMBV.

Abstract:Frontotemporal dementia and Parkinsonism linked to chromosome 17 (FTDP‐17) is an autosomal dominant condition clinically characterized by behavioral, cognitive and motor disturbances. It was recently discovered that the majority of the FTDP‐17 families carry missense or 5′ splice mutations in the exons coding for the microtubule‐binding domains of the tau protein. However, in at least five FTDP‐17 families, no such mutations could be identified. In the present study, we aimed at further investigate abnormalities in the tau gene in a Swedish FTDP‐17 family, where no mutations in the tau gene previously have been identified. Initially, we searched for larger deletions by Southern blot hybridization. Furthermore, possible abnormal splicing events was investigated by RT‐PCR from brain tissue of affected individuals. In addition, we investigated the presence of mutations in other genes in the FTDP‐17 candidate region on chromosome 17q21; Gamma‐tubulin, Glial Fibrillary Acid Protein (GFAP), Human dual specificity phosphatase tyrosine/serine (VHR), Rap‐interacting protein 8 (RPIP8), P35, and the recently identified FTDCG1. In conclusion, no pathological changes in the tau gene were observed, neither was any mutations segregating with the disease detected in the investigated candidate genes. Further investigation of extended intron sequences or promoter regions of the tau gene and additional candidate genes on chromosome 17q21, therefore seems to be necessary in order to identify the additional causes of FTDP‐17. © 2003 Wiley‐Liss, Inc.

Abstract:Largemouth bass virus (LMBV) infections has resulted in high mortality and economic losses to the global largemouth bass industry and has seriously restricted the healthy development of the bass aquaculture industry. There are currently no antiviral therapies available for the control of this disease. In this study, we developed three types of vaccine against LMBV; whole virus inactivated vaccine (I), a subunit vaccine composed of the major viral capsid protein MCP (S) as well as an MCP DNA vaccine(D), These were employed using differing immunization and booster strategies spaced 2 weeks apart as follows: II, SS, DD and DS. We found that all vaccine groups induced humoral and cellular immune responses and protected largemouth bass from a lethal LMBV challenge to varying degrees and DD produced the best overall effect. Specifically, the levels of specific IgM in serum in all immunized groups were elevated and significantly higher than those in the control group. Moreover, the expression of humoral immunity (CD4 and IgM) and cellular immunity (MHC-1) as well as cytokines (IL-1β) was increased, and the activity of immunity-related enzymes ACP, AKP, LZM, and T-SOD in the serum was significantly enhanced. In addition, the relative percent survival of fish following an LMBV lethal challenge 4 weeks after the initial immunizations were high for each group: DD(89.5%),DS(63.2%),SS(50%) and II (44.7%). These results indicated that the MCP DNA vaccine is the most suitable and promising vaccine candidate for the effective control of LMBV disease.

Abstract:Diseases caused by pathogens severely hampered the development of aquaculture, especially largemouth bass virus (LMBV) has caused massive mortality and severe economic losses to the culture of largemouth bass (Micropterus salmoides). Considering the environmental hazards and human health, effective and environmentally friendly therapy strategy against LMBV is of vital importance and in pressing need. In the present study, a novel nanobody (NbE4) specific for LMBV was selected from a phage display nanobody library. Immunofluorescence and indirect ELISA showed that NbE4 could recognize LMBV virions and had strong binding capacity, but RT-qPCR evidenced that NBE4 did not render the virus uninfectious. Besides, antiviral drug ribavirin was used to construct a targeted drug system delivered by bacterial nanocellulose (BNC). RT-qPCR revealed that NbE4 could significantly enhance the antiviral activity of ribavirin in vitro and in vivo. The targeted drug delivery system (BNC-Ribavirin-NbE4, BRN) reduced the inflammatory response caused by LMBV infection and improved survival rate (BRN-L, 33.3 %; BRN-M, 46.7 %; BRN-H, 56.7 %)compared with control group (13.3 %), ribavirin group (RBV, 26.7 %) and BNC-ribavirin (BNC-R, 40.0 %), respectively. This research provided an effective antiviral strategy that improved the drug therapeutic effect and thus reduced the dosage.

Abstract:Largemouth bass ranavirus (LMBV) infection results in huge economic losses in largemouth bass (Micropterus salmoides) industry. Nanopeptide C-I20 and anthocyanins have a positive effect on promoting immune responses and antioxidant mechanisms in several aquatic organisms, and are therefore used to inhibit LMBV infection. In this study, we developed an LMBV immersion challenge model using three different viral concentrations (1 × 104 copies/mL, 1 × 105 copies/mL, and 1 × 106 copies/mL) to infect largemouth bass, and LMBV-MCP mRNA expression was detected in infected fish. Following infection, the fish exhibited severe external ulceration, redness swelling, and darkening of the skin. Histopathological examination revealed significant necrosis and inflammation in muscle tissue, epithelial cell shedding in renal tubules, macrophage aggregation centers and cellular vacuolization in spleen and head kidney, and cellular hypertrophy in liver. To mitigate LMBV infection, we explored the protective effects of a combined treatment strategy involving C-I20 and anthocyanin. Overall, the combination of anthocyanin and C-I20 demonstrated the highest protective efficacy, significantly reducing viral loads in muscle, liver, spleen, and head kidney. Moreover, this treatment regimen enhanced antioxidant enzyme activities (T-AOC, TSOD, GSH-Px, CAT) and modulated important immune genes (IL-1, IL-8, TNF-α, IL-10, Mx, and IgM) expression. In conclusion, the synergistic application of anthocyanin and C-I20 demonstrates significant efficacy in mitigating LMBV infection. This research introduces a novel and promising approach to managing infectious diseases in aquaculture settings.
Comparison and Evaluation of DNA Vaccines Against Nocardia Seriolae Infection in Largemouth Bass (micropterus Salmoides)

Abstract:Chronic granulomatous diseases caused by Nocardia seriolae have resulted in substantial economic losses for the aquaculture industry. DNA vaccines are considered a promising approach for controlling infectious diseases. To develop effective DNA vaccines against N. seriolae, we selected Ag85L, MmaA4, HspX, Rv3407, and HtpG as candidate antigens. Among these, pcDNA-Ag85L and pcDNA-MmaA4 were the most effective against N. seriolae infection in largemouth bass (Micropterus salmoides), achieving the highest relative percentage survival (RPS) rates of 78.6 % and 92.9 %, respectively. Tissue bacterial loads and histopathological tests revealed a significant reduction in bacterial abundance in the liver, spleen, head kidney, and trunk kidney of the immunised groups compared to that in the control group. Additionally, no distinct pathological changes were observed in the spleen or head kidney tissues. The expression levels of immune-related genes including IL-1 beta, IL-8, MHCI, MHCII, CD4, and CD8 alpha were significantly upregulated in the pcDNA-Ag85L and pcDNA-MmaA4 immunised groups, thus indicating that the two DNA vaccines mediated immune protection by promoting both humoral and cellular immune responses. Overall, the pcDNA-Ag85L and pcDNA-MmaA4 DNA vaccines demonstrated strong potential for protecting largemouth bass against N. seriolae infection, offering an innovative solution for controlling granulomatous diseases in fish.
Artificial Infection Pathway of Largemouth Bass LMBV and Identification of Resistant and Susceptible Individuals

Abstract:Identifying disease-resistant and disease-susceptible materials in largemouth bass can provide fundamental analytical information for disease-resistant breeding research on this species. To date, no stable and effective artificial infection pathway has been established against largemouth bass virus (LMBV) in largemouth bass. In this study, using the pathogen at a concentration of 1.60 x 105 TCID50/mL, largemouth bass juveniles were infected using three different methods: muscle injection, feeding, and immersion. A comprehensive comparative study was conducted on parameters such as disease progression, viral load, pathological changes, and electron microscopy. The results revealed that the injection group experienced rapid mortality, with all individuals dying within 6 days after the infection. The feeding group showed minimal mortality, with an overall mortality rate of 28.57%. The immersion group reached its peak mortality on the 9th day after infection, with an accumulated mortality rate of 63.24%. Viral gene quantification results showed that while in the injection group muscles had the highest viral load followed by the liver and spleen, the other infection methods led to the highest viral loads in the liver and spleen. Tissue pathology and electron microscopy observations revealed that the liver and spleen exhibited the most significant pathological changes and were the easiest organs to detect viral particles, indicating that the liver and spleen are the primary target organs of LMBV in largemouth bass. In conclusion, this study suggests that immersing largemouth bass in LMBV at a concentration of 1.99 x 103 c/mL (diluted 80 times) closely mimics the natural infection rate, allowing for a clear differentiation between causes of death and survival, making it an almost non-invasive artificial infection method that closely resembles natural infection conditions. These findings can provide crucial support for the accurate identification of disease-resistant and disease-susceptible materials in largemouth bass and the breeding of disease-resistant varieties.

Abstract:In order to understand the immune response of largemouth bass against Micropterus salmoides Rhabdovirus (MSRV), assisting disease resistance breeding, three largemouth bass breeding varieties Micropterus salmoides “Youlu No 3” (U3), “Youlu No 1” (U1) and “Zhelu No 1” (P1) were challenged intraperitoneally with MSRV. Serum and tissues were sampled to study the changes in non-specific immune parameters, viral loads, and transcript levels of immune-related genes, and the cumulative mortality rate was recorded daily for 14 days. The results showed that the cumulative mortality rates in the U1, P1, and U3 groups were 6.66% ± 2.89%, 3.33% ± 2.89%, and 0, respectively. The higher mortality may attribute to the increased viral loads after infection in the liver (2.79 × 105 and 2.38 × 105 vs. 1.3 × 104 copies/mg), spleen (2.14 × 105 and 9.40 × 104 vs. 4.21 × 103 copies/mg), and kidney (3.59 × 104 and 8.40 × 103 vs. 2.42 × 103 copies/mg) in the U1 and P1 groups compared to the U3 group. The serum non-specific immune parameters (lysozyme, catalase, and acid phosphatase) were found to be increased significantly in the U3 group. In addition, the transcripts of interferon-related genes (IFN-γ, IRF3, and IRF7) and pro-inflammatory-related genes (TNF-α and IL-1β) exhibited up-regulation and peaked at 6 h post infection in the U3 group, which also exhibited up-regulation but peaked at 12–24 h post infection in the U1 and P1 groups. In conclusion, these findings indicate that earlier activation of interferon and pro-inflammatory response is beneficial to largemouth bass against MSRV infection. This experiment may provide an insight into understanding the immune mechanism of largemouth bass against MSRV infection and contributes to molecular-assisted selection.

Abstract:Largemouth bass ( Micropterus salmoides ) has emerged as a significant economic fish species, with a rise in Aeromonas veronii infections in farming. However, research on adjuvants for vaccines against A. veronii in largemouth bass remains scarce. In present study, recombinant largemouth bass IL-1β (LbIL-1β) was expressed to explore its adjuvant effect on the A. veronii inactivated vaccine. Following vaccination with recombinant LbIL-1β (rLbIL-1β) and the inactivated A. veronii , higher serum SOD levels and lysozyme activities were observed in largemouth bass from inactivated A. veronii + rLbIL-1β vaccinated group. Furthermore, it was discovered that rLbIL-1β was able to boost the serum-specific antibody levels induced by the inactivated A. veronii . The qRT-PCR analysis revealed that rLbIL-1β also enhanced the expression of IgM, CD4, and MHC II in largemouth bass triggered by the inactivated A. veronii . After challenged with live A. veronii , the outcomes demonstrated that the relative percentage survival (RPS) for largemouth bass resulting from the inactivated A. veronii in combination with rLbIL-1β was 76.67 %, surpassing the RPS of 60 % in the inactivated A. veronii group. Collectively, these findings indicate that rLbIL-1β enhances the protective effect of the A. veronii inactivated vaccine on largemouth bass, showcasing potential as an adjuvant for further development.

Abstract:Largemouth bass is an important commercially farmed fish in China, but the rapid expansion of its breeding has resulted in increased incidence of diseases caused by bacteria, viruses and parasites. In this study, moribund largemouth bass containing ulcer foci on body surfaces indicated the most likely pathogens were iridovirus and rhabdovirus members and this was confirmed using a combination of immunohistochemistry, cell culture, electron microscopy and conserved gene sequence analysis. We identified that these fish had been co-infected with these viruses. We observed bullet-shaped virions (100–140 nm long and 50–100 nm in diameter) along with hexagonal virions with 140 nm diameters in cell culture inoculated with tissue homogenates. The viruses were plaque purified and a comparison of the highly conserved regions of the genome of these viruses indicated that they are most similar to largemouth bass virus (LMBV) and hybrid snakehead rhabdovirus (HSHRV), respectively. Regression infection experiments indicated fish mortalities for LMBV-FS2021 and HSHRV-MS2021 were 86.7 and 11.1%, respectively. While co-infection resulted in 93.3% mortality that was significantly (p 100-fold. Overall, we simultaneously isolated and identified LMBV and a HSHRV-like virus from diseased largemouth bass, and our results can provide novel ideas for the prevention and treatment of combined virus infection especially in largemouth bass.

Abstract:The largemouth bass virus (LMBV) isolate of Santee-Cooper ranavirus showed evidence of widespread infection in adult fish, but disease presentation caused by different viral strains exhibited considerable difference. In this study, a highly pathogenic LMBV-like resembling Santee-Cooper ranavirus was isolated and identified from juvenile largemouth bass. The pathogenicity and dynamic distribution of LMBV-like strain, histopathological analysis and host immune response of juvenile largemouth bass infected with LMBV-like were investigated. The results show that LMBV-like was highly pathogenic to juvenile fish, and the infected fish showed typical signs of acute haemorrhages and visceral enlargement. LMBV-like positive cells were found in the liver, spleen, kidney, gills, and intestinal tissue, and the virus content in spleen was the highest. Histopathological analysis showed different pathological changes in major tissues of diseased fish, mostly manifested as infiltration of inflammatory cell and histiocyte necrosis. In addition, humoral immune factors such as superoxide dismutase (SOD), catalase (CAT) and acid phosphatase (ACP) were used as serum indicators to evaluate the immune response of juvenile fish after infection. Quantitative real-time PCR (qRT-PCR) was used to evaluate the expression patterns of immune-related genes (CD40, IFN-γ, IgM, IL-1β, IL-8, IL-12a, Mxd3, TGF-β, and TNFα) in liver, spleen, and head kidney tissues. The results showed that immunological activity of the juvenile largemouth bass was significantly enhanced by LMBV-like infection. This research comprehensively systematically revealed the pathogenic characteristics of LMBV-like separated from juvenile largemouth bass and properties of the host's immune response caused by the virus infection, which providing a basis for further exploring the interaction between the virus and the host, and prevention and treatment of disease caused by Santee-Cooper ranavirus.
Antiviral innovations in largemouth bass aquaculture: Unveiling the potential of arctigenin derivatives against Micropterus Salmoides rhabdovirus

Abstract:Aquaculture confronts an escalating threat from viral infections, necessitating effective strategies to ensure fish health. This study investigated the comprehensive antiviral properties of compound A28 against Micropterus Salmoides Rhabdovirus (MSRV) in largemouth bass. Synthesized as a derivative of arctigenin, A28 was characterized for its inhibitory effects on MSRV at both cellular and organismal levels. To elucidate A28's impact on MSRV, cytotoxicity was gauged using the CCK-8 assay, revealing a safe concentration of 2.5 mg/L for A28 on GCO cell viability. At this concentration, A28 treatment resulted in remarkable inhibition rates, notably 99.14% for the MSRV nucleoprotein ( N ) gene and 99.46% for the glycoprotein ( G ) gene expression in GCO cells. Additionally, RT-qPCR analysis demonstrated a dose-dependent inhibition of MSRV genes ( N and G ) with increasing A28 concentrations. Morphological assessment revealed a reduction in MSRV-induced cytopathic effects in the presence of 2.5 mg/L A28 after 48 h. Intriguingly, preventive and therapeutic experiments underscored A28's dual role against MSRV infection. Water stability assessment indicated a sustained inhibitory effect of A28 on MSRV for approximately 2 days. Notably, A28 demonstrated the capacity to alleviate MSRV-induced nuclear and mitochondrial damage, underscoring its cytoprotective effects. In vivo experiments on largemouth bass revealed that A28 significantly enhanced fish survival rates upon MSRV challenge, with a notable 38% increase following A28 treatment. Concurrently, A28 upregulated fish immune-related genes, furnishing molecular evidence of its immunomodulatory effects. Furthermore, A28 exhibited efficacy in suppressing the horizontal transmission of MSRV in fish, suggesting its potential as a therapeutic agent in aquaculture settings. In conclusion, compound A28 emerges as a promising antiviral agent against MSRV in largemouth bass, offering insights into its potential application in managing aquaculture diseases. The findings provide a foundation for further research on A28's pharmacokinetics, environmental impact, and scalability for practical aquaculture applications.
