Abstract:Biological nitrogen fixation (BNF) refers to a microbial mediated process based upon an enzymatic “Nitrogenase” conversion of atmospheric nitrogen (N 2 ) into ammonium readily absorbable by roots. N 2 -fixing microorganisms collectively termed as “diazotrophs” are able to fix biologically N 2 in association with plant roots. Specifically, the symbiotic rhizobacteria induce structural and physiological modifications of bacterial cells and plant roots into specialized structures called nodules. Other N 2 -fixing bacteria are free-living fixers that are highly diverse and globally widespread in cropland. They represent key natural source of nitrogen (N) in natural and agricultural ecosystems lacking symbiotic N fixation (SNF). In this review, the importance of Azotobacter species was highlighted as both important free-living N 2 -fixing bacteria and potential bacterial biofertilizer with proven efficacy for plant nutrition and biological soil fertility. In addition, we described Azotobacter beneficial plant promoting traits (e.g., nutrient use efficiency, protection against phytopathogens, phytohormone biosynthesis, etc.). We shed light also on the agronomic features of Azotobacter that are likely an effective component of integrated plant nutrition strategy, which contributes positively to sustainable agricultural production. We pointed out Azotobacter based-biofertilizers, which possess unique characteristics such as cyst formation conferring resistance to environmental stresses. Such beneficial traits can be explored profoundly for the utmost aim to research and develop specific formulations based on inoculant Azotobacter cysts. Furthermore, Azotobacter species still need to be wisely exploited in order to address specific agricultural challenges (e.g., nutrient deficiencies, biotic and abiotic constraints) taking into consideration several variables including their biological functions, synergies and multi-trophic interactions, and biogeography and abundance distribution.

Azotobacter vinelandii N2 fixation increases in co-culture with the PGPR Bacillus subtilis in a nitrogen concentration-dependent manner

Abstract:ABSTRACT Biological nitrogen fixation (BNF) is an essential source of new nitrogen (N) for terrestrial ecosystems. The abiotic factors regulating BNF have been extensively studied in various ecosystems and laboratory settings. Despite this, our understanding of the impact of neighboring bacteria on N 2 fixer activity remains limited. Here, we explored this question using a co-culture of the two model species: the free-living diazotroph Azotobacter vinelandii and the non-fixing plant growth-promoting rhizobacteria Bacillus subtilis . We observed that the interaction between the two bacteria was modulated by N availability. Under N-replete conditions, B. subtilis outcompeted A. vinelandii in the co-culture. Under N-limiting conditions, BNF activity by A. vinelandii was enhanced in the presence of B. subtilis . Reciprocally, the presence of A. vinelandii repressed sporulation by B. subtilis and supported its growth likely through N transfer. N inputs by A. vinelandii were doubled in the presence of B. subtilis compared to the monoculture, primarily due to the retention of a robust N 2 fixation activity in the stationary phase. A proteomic analysis revealed that A. vinelandii N metabolism, particularly the molybdenum nitrogenase isoform protein levels (NifK and NifD), was upregulated during the stationary growth phase in the presence of B. subtilis . This study revealed that N stress drives bacterial interactions and activity in a two-species community, especially in the stationary phase. IMPORTANCE Reducing inputs of chemical N fertilizers is essential to develop a more sustainable agriculture. The stimulation of biological nitrogen fixation by N2 fixers in multispecies cultures, here the plant growth-promoting rhizobacteria Azotobacter vinelandii and Bacillus subtilis , opens opportunities for the formulation of biofertilizers consortia. While most research on N2 fixation historically focussed on the exponential growth phase of microorganisms, we observed that Bacillus subtilis stimulated Azotobacter vinelandii N2 fixation mostly during the stationary phase. This result highlights that more research on the factors controlling N2 fixation repression during the stationary growth phase, especially bacteria-bacteria interactions, is eagerly needed.

Improved Stability of Engineered Ammonia Production in the Plant-Symbiont Azospirillum brasilense

Abstract:Bioavailable nitrogen is the limiting nutrient for most agricultural food production. Associative diazotrophs can colonize crop roots and fix their own bioavailable nitrogen from the atmosphere. Wild-type (WT) associative diazotrophs, however, do not release fixed nitrogen in culture and are not known to directly transfer fixed nitrogen resources to plants. Efforts to engineer diazotrophs for plant nitrogen provision as an alternative to chemical fertilization have yielded several strains that transiently release ammonia. However, these strains suffer from selection pressure for nonproducers, which rapidly deplete ammonia accumulating in culture, likely limiting their potential for plant growth promotion (PGP). Here we report engineered Azospirillum brasilense strains with significantly extend ammonia production lifetimes of up to 32 days in culture. Our approach relies on multicopy genetic redundancy of a unidirectional adenylyltransferase (uAT) as a posttranslational mechanism to induce ammonia release via glutamine synthetase deactivation. Testing our multicopy stable strains with the model monocot Setaria viridis in hydroponic monoassociation reveals improvement in plant growth promotion compared to single copy strains. In contrast, inoculation of Zea mays in nitrogen-poor, nonsterile soil does not lead to increased PGP relative to WT, suggesting strain health, resource competition, or colonization capacity in soil may also be limiting factors. In this context, we show that while engineered strains fix more nitrogen per cell compared to WT strains, the expression strength of multiple uAT copies needs to be carefully balanced to maximize ammonia production rates and avoid excessive fitness defects caused by excessive glutamine synthetase shutdown.

Abstract:ABSTRACT A grand challenge for the next century is in facing a changing climate through bioengineering solutions. Biological nitrogen fixation, the globally consequential, nitrogenase-catalyzed reduction of atmospheric nitrogen to bioavailable ammonia, is a vital area of focus. Nitrogen fixation engineering relies upon extensive understanding of underlying genetics in microbial models, including the broadly utilized gammaproteobacterium, Azotobacter vinelandii ( A. vinelandii ). Here, we report the first CRISPR interference (CRISPRi) system for targeted gene silencing in A. vinelandii that integrates genomically via site-specific transposon insertion. We demonstrate that CRISPRi can repress transcription of an essential nitrogen fixation gene by ~60%. Further, we show that nitrogenase genes are suitably expressed from the transposon insertion site, indicating that CRISPRi and engineered nitrogen fixation genes can be co-integrated for combinatorial studies of gene expression and engineering. Our established CRISPRi system fills an important gap for engineering microbial nitrogen fixation for desired purposes. IMPORTANCE All life on Earth requires nitrogen to survive. About 78% of the atmosphere alone is nitrogen, yet humans cannot use it directly. Instead, we obtain the nitrogen we need for our survival through the food we eat. For more than 100 years, a substantial portion of agricultural productivity has relied on industrial methods for nitrogen fertilizer synthesis, which consumes significant amounts of nonrenewable energy resources and exacerbates environmental degradation and human-induced climate change. Promising alternatives to these industrial methods rely on engineering the only biological pathway for generating bioaccessible nitrogen: microbial nitrogen fixation. Bioengineering strategies require an extensive understanding of underlying genetics in nitrogen-fixing microbes, but genetic tools for this critical goal remain lacking. The CRISPRi gene silencing system that we report, developed in the broadly utilized nitrogen-fixing bacterial model, Azotobacter vinelandii , is an important step toward elucidating the complexity of nitrogen fixation genetics and enabling their manipulation.

Abstract:The nitrogen (N)-use efficiency of agricultural plants is notoriously poor. Globally, about 50% of the N fertilizer applied to cropping systems is not absorbed by plants, but lost to the environment as ammonia (NH3), nitrate (NO3−), and nitrous oxide (N2O, a greenhouse gas with 300 times the heat-trapping capacity of carbon dioxide), raising agricultural production costs and contributing to pollution and climate change. These losses are driven by volatilization of NH3 and by a matrix of nitrification and denitrification reactions catalysed by soil microorganisms (chiefly bacteria and archaea). Here, we discuss mitigation of the harmful and wasteful process of agricultural N loss via biological nitrification inhibitors (BNIs) exuded by plant roots. We examine key recent discoveries in the emerging field of BNI research, focusing on BNI compounds and their specificity and transport, and discuss prospects for their role in improving agriculture while reducing its environmental impact.

Abstract:Biological nitrogen fixation, the microbial reduction of atmospheric nitrogen to bioavailable ammonia, represents both a major limitation on biological productivity and a highly desirable engineering target for synthetic biology. However, engineering of nitrogen fixation requires an integrated understanding of how the gene regulatory dynamics of host diazotrophs restrict the available sequence-function space of its central catalytic metalloenzyme, nitrogenase. Here, we interrogate this relationship by analyzing the transcriptome of Azotobacter vinelandii engineered with a phylogenetically inferred, ancestral nitrogenase protein variant. The engineered strain exhibits reduced cellular nitrogenase activity but recovers wild-type growth rates following an extended lag period. We find that expression of genes within the immediate nitrogen fixation network is resilient to nitrogenase sequence-level perturbations. Rather, physiological compatibility with the ancestral nitrogenase variant is restored by reducing trace metal and electron resource allocation to nitrogenase. Our results spotlight cellular processes adjacent to nitrogen fixation as productive engineering targets to improve compatibility between remodeled nitrogenase proteins and engineered host diazotrophs.

Abstract:Strains of the nitrogen‐fixing microbe Azotobacter vinelandii were constructed with the ability to up‐regulate biological nitrogen fixation in a galactose‐dependent manner resulting in strains that release extracellular ammonium through an external trigger. These strains produced ammonium while maintaining high levels of cell biomass and polyhydroxybutyrate production. The capture and reduction of atmospheric dinitrogen gas to ammonium can be accomplished through the enzyme nitrogenase in a process known as biological nitrogen fixation (BNF), by a class of microbes known as diazotrophs. The diazotroph Azotobacter vinelandii is a model organism for the study of aerobic nitrogen fixation, and in recent years has been promoted as a potential producer of biofertilizers. Prior reports have demonstrated the potential to partially deregulate BNF in A. vinelandii, resulting in accumulation and extracellular release of ammonium. In many cases, deregulation requires the introduction of transgenic genes or elements to yield the desired phenotype, and the long‐term stability of these strains has been reported to be somewhat problematic. In this work, we constructed two strains of A. vinelandii where regulation can be precisely controlled without the addition of any foreign genes or genetic markers. Regulation is maintained through native promoters found in A. vinelandii that can be induced through the addition of extraneous galactose. These strains result in varied degrees of regulation of BNF, and as a result, the release of extracellular ammonium is controlled in a precise, and galactose concentration‐dependent manner. In addition, these strains yield high biomass levels, similar to the wild‐type A. vinelandii strain and are further able to produce high percentages of the bioplastic polyhydroxybutyrate.

Abstract:“AGA Thermovision” has been used to record the temperature of the face and scalp in normal subjects and patients susceptible to vascular headache. Even when free of headache, the latter group showed asymmetry of heat loss from the forehead more often than normal controls. During migraine headache, the skin temperature was lower on the affected side by about 1°C in 8 out of 12 of the patients with hemicrania. The reduction of skin temperature on the side of migraine headache, in spite of dilatation of the extracranial arteries, Indicates that blood Is shunted away from the skin during the attack. Asymmetry of heat loss persisted for some time after the relief of migraine headache by intravenous serotonin or oral ergotamine tartrate therapy, suggesting that these agents achieve their effect through constriction of the extracranial arteries rather than dilatation of skin capillaries.

Abstract:Nitrogen (N2) fixation stands as an indispensable natural process for the survival of all life forms on Earth. Within the grand tapestry of the nitrogen cycle, biological nitrogen fixation plays a vital role. But its optimal manifestation may be constrained by the metabolic generation of reducing equivalents from chemical substrates. Therefore, it becomes important to propose reasoned and feasible strategies to improve the production of reducing equivalents and thereby enhance nitrogen-fixation efficiency. In a recent publication within Angewandte Chemie International Edition, Bazan and coworkers reported a light-driven N2-fixing bacteria biohybrid system supported by a broad light-harvesting conjugated oligoelectrolyte (COE-IC) and non-photosynthetic bacteria of Azotobacter vinelandii (A. vinelandii). The introduction of the optically active COE-IC enhances the conversion of N2 to ammonia (NH3) and improves the photoinduced generation of biomass and L-amino acids of biological nitrogen-fixation systems.

The engineered strain studied here responds by adjusting the expression of proteins involved in cellular processes adjacent to nitrogen fixation, rather than that of nitrogenase proteins themselves. These insights can inform future strategies to transfer nitrogenase variants to non-native hosts.

Abstract:Cyanobacteria, renowned for their nitrogen‐fixing characteristics, are important for sustainable biomanufacturing and agricultural innovation. This review explores the synergy between cyanobacteria and nitrogen fixation, highlighting their potential to revolutionize biobased compound production and reduce the ecological impact of traditional nitrogen sources. It focuses on genetic enhancements and synthetic biology techniques, which transform these microorganisms into sustainable nitrogen providers. Current applications range from agricultural enhancement to cutting‐edge biotechnology, highlighting the important consequences of cyanobacterial nitrogen fixation. Challenges persist, however, requiring a meticulous analysis of ecological, regulatory, and scalability concerns. The untapped potential of cyanobacteria in nitrogen fixation promises a significant shift in biomanufacturing and environmental stewardship. The aim of this article is to inspire high‐impact research and transformative applications in biotechnology and sustainability.

Abstract:Eight inorganic-organic hybrid compounds with a formula of [R-Bz-1-APy][PbI(3)] (R-Bz-1-APy(+) = mono-substituted benzylidene-1-aminopyridinium Schiff base derivative; R = m-CN (1), m-CH(3) (2), H (3), p-F (4), p-Cl (5), p-Br (6), o-Cl (7), o-Br (8)) have been synthesized and characterized structurally. The common characteristic of the crystal structures of 1-8 is that the inorganic components form straight and face-sharing octahedral [PbI(3)](∞) chains and the Schiff base cations surround the [PbI(3)](∞) chains to form molecular stacks. The substituent (R) on the phenyl ring of the Schiff base cation clearly influences the packing structures of 1-8, and the hybrid compound crystallizes in the space group P6(3) when R = CN (1) in the meta-position of the phenyl ring, and in a central symmetric space group when R is in the ortho- or para-position of the phenyl ring. The conformation of the Schiff base cation is related to the R position, and the dihedral angle between the phenyl and pyridyl rings increases in the order of para- < meta- < ortho-position substitution of the phenyl ring. The long molecular axis of the Schiff base cation adopts a manner approximately parallel to the straight inorganic [PbI(3)](∞) chain in the para-substituted hybrid compounds, and perpendicular to the straight inorganic [PbI(3)](∞) chain in the ortho-substituted hybrid compounds. 1 is second harmonic generation (SHG) active with a comparable response as that of urea and also exhibits ferroelectricity with larger P(s) and P(r) values; 1-8 emit multi-band luminescence in the 300-650 nm regions under the excitation of ultraviolet light.

Abstract:Agricultural productivity relies on synthetic nitrogen fertilizers, yet half of that reactive nitrogen is lost to the environment. There is an urgent need for alternative nitrogen solutions to reduce the water pollution, ozone depletion, atmospheric particulate formation, and global greenhouse gas emissions associated with synthetic nitrogen fertilizer use. One such solution is biological nitrogen fixation (BNF), a component of the complex natural nitrogen cycle. BNF application to commercial agriculture is currently limited by fertilizer use and plant type. This paper describes the identification, development, and deployment of the first microbial product optimized using synthetic biology tools to enable BNF for corn (Zea mays) in fertilized fields, demonstrating the successful, safe commercialization of root-associated diazotrophs and realizing the potential of BNF to replace and reduce synthetic nitrogen fertilizer use in production agriculture. Derived from a wild nitrogen-fixing microbe isolated from agricultural soils, Klebsiella variicola 137-1036 ("Kv137-1036") retains the capacity of the parent strain to colonize corn roots while increasing nitrogen fixation activity 122-fold in nitrogen-rich environments. This technical milestone was then commercialized in less than half of the time of a traditional biological product, with robust biosafety evaluations and product formulations contributing to consumer confidence and ease of use. Tested in multi-year, multi-site field trial experiments throughout the U.S. Corn Belt, fields grown with Kv137-1036 exhibited both higher yields (0.35 ± 0.092 t/ha ± SE or 5.2 ± 1.4 bushels/acre ± SE) and reduced within-field yield variance by 25% in 2018 and 8% in 2019 compared to fields fertilized with synthetic nitrogen fertilizers alone. These results demonstrate the capacity of a broad-acre BNF product to fix nitrogen for corn in field conditions with reliable agronomic benefits.

Abstract:The energetic requirements for biological nitrogen fixation necessitate stringent regulation of this process in response to diverse environmental constraints. To ensure that the nitrogen fixation machinery is expressed only under appropriate physiological conditions, the dedicated NifL-NifA regulatory system, prevalent in Proteobacteria, plays a crucial role in integrating signals of the oxygen, carbon and nitrogen status to control transcription of nitrogen fixation ( nif ) genes. Greater understanding of the intricate molecular mechanisms driving transcriptional control of nif genes may provide a blueprint for engineering diazotrophs that associate with cereals. In this study, we investigated the properties of a single amino acid substitution in NifA, (NifA-E356K) which disrupts the hierarchy of nif regulation in response to carbon and nitrogen status in Azotobacter vinelandii . The NifA-E356K substitution enabled overexpression of nitrogenase in the presence of excess fixed nitrogen and release of ammonia outside the cell. However, both of these properties were conditional upon the nature of the carbon source. Our studies reveal that the uncoupling of nitrogen fixation from its assimilation is likely to result from feedback regulation of glutamine synthetase, allowing surplus fixed nitrogen to be excreted. Reciprocal substitutions in NifA from other Proteobacteria yielded similar properties to the A . vinelandii counterpart, suggesting that this variant protein may facilitate engineering of carbon source-dependent ammonia excretion amongst diverse members of this family.

Genetic modification of diazotrophic bacteria has successfully enhanced NH 4 + release. In this study, we compared the competitive fitness of A. vinelandii mutant strains, which allowed us to analyze the burden of NH 4 + release under a broad dynamic range. Long-term competition assays under regular culture conditions confirmed a large burden for NH 4 + release, exclusion by the wt strain, phenotypic instability, and loss of the ability to release NH 4 + . In contrast, co-inoculation in mild autoclaved soil showed a much longer co-existence with the wt strain and a stable NH 4 + release phenotype. All genetically modified strains increased the N content and changed its chemical speciation in the soil. This study contributes one step forward towards bridging a knowledge gap between molecular biology laboratory research and the incorporation of N from the air into the soil in a molecular species suitable for plant nutrition, a crucial requirement for developing improved bacterial inoculants for economic and environmentally sustainable agriculture.

Abstract:Abstract Nitrogen is an essential element of life, and nitrogen availability often limits crop yields. Since the Green Revolution, massive amounts of synthetic nitrogen fertilizers have been produced from atmospheric nitrogen and natural gas, threatening the sustainability of global food production and degrading the environment. There is a need for alternative means of bringing nitrogen to crops, and taking greater advantage of biological nitrogen fixation seems a logical option. Legumes are used in most cropping systems around the world because of the nitrogen-fixing symbiosis with rhizobia. However, the world's three major cereal crops—rice, wheat, and maize—do not associate with rhizobia. In this review, we will survey how genetic approaches in rhizobia and their legume hosts allowed tremendous progress in understanding the molecular mechanisms controlling root nodule symbioses, and how this knowledge paves the way for engineering such associations in non-legume crops. We will also discuss challenges in bringing these systems into the field and how they can be surmounted by interdisciplinary collaborations between synthetic biologists, microbiologists, plant biologists, breeders, agronomists, and policymakers.

[Correlation between cell cycle and cell morphology of RKN cells].

Abstract:UNLABELLED In order to study the changes in the morphology of each phase of the cell cycle, RKN cells, derived from a human ovarian myosarcoma, were synchronized by a mitotic selection method and studied under time-lapse cinematography, light microscopy, scanning and transmission electron microscopy. RESULTS M-phase: The cells with relatively a few cytoplasmic organelles were small and spherical, and chromosome were seen in them. The cell surface was covered with numerous mivilli. Late G1,S and early G2 phase: The large, thin cells were flat and polygonal with various organelles inside and had central nuclei. Most of the chromatin patterns were finely granular. No microvilli were found on the cell surface. Early G1-phase: The paired daughter cells were fanshaped and the nuclei were eccentric with finely granular chromatin patterns. At the edges of the cells, rufflers which were related to cell movements were often seen. Late G2-phase: The thick cells were of a spindle shape and the chromatin patterns were a coarse granular type. Again on the cell surface microvilli increased and the slender filopodia were often seen.

Abstract:Low nitrogen content in soil is usually overcome by chemical fertilization. After long application period, high-dose and intensive use of N fertilizers can cause ammonia volatilization and nitrates accumulation in soil. In sustainable agriculture, the use of bacterial inoculant integrated with nutrient management system has a role in soil health and productivity. Azotobacter-based biofertilizer is suggested as a chemical nitrogen fertilizer substitute or addition in crop production to improve available nutrients in the soil, provide some metabolites during plant growth, and minimize fertilizer doses. The objective of this literature reviewed paper is to discuss the role of Azotobacter in agriculture; and the prospective of Azotobacter to increase yield and substitute the chemical fertilizer in food crops production. The results revealed that mechanisms by Azotobacter in plant growth enhancement are as biofertilizer, biostimulant, and bioprotectant. Nitrogen fixation by Azotobacter is the mechanism to provide available nitrogen for uptake by roots. Azotobacter stimulates plant growth through phytohormones synthesis; indole acetic acid, cytokinins, and gibberellins are detected in the liquid culture of Azotobacter. An indirect effect of Azotobacter is exopolysaccharide production and plant protection. Inoculation of Azotobacter in the field integrated with organic matter and reduced chemical fertilizer are reported to improve plant growth and yield.

Abstract:The soil environment adjacent to plant roots, termed the rhizosphere, is home to a wide variety of microorganisms that can significantly affect the physiology of nearby plants. Microbes in the rhizosphere can provide nutrients, secrete signaling compounds, and inhibit pathogens. These processes could be manipulated with synthetic biology to enhance the agricultural performance of crops grown for food, energy, or environmental remediation, if methods can be implemented in these nonmodel microbes. A common first step for domesticating nonmodel organisms is the development of a set of genetic engineering tools, termed a synthetic biology toolbox. A toolbox comprises transformation protocols, replicating vectors, genome engineering (e.g., CRISPR/Cas9), constitutive and inducible promoter systems, and other gene expression control elements. This work validated synthetic biology toolboxes in three nitrogen-fixing soil bacteria: Azotobacter vinelandii, Stutzerimonas stutzeri (Pseudomonas stutzeri), and a new isolate of Klebsiella variicola. All three organisms were amenable to transformation and reporter protein expression, with several functional inducible systems available for each organism. S. stutzeri and K. variicola showed more reliable plasmid-based expression, resulting in successful Cas9 recombineering to create scarless deletions and insertions. Using these tools, we generated mutants with inducible nitrogenase activity and introduced heterologous genes to produce resorcinol products with relevant biological activity in the rhizosphere.
