Abstract:Alpine grasslands are important ecosystems providing a variety of ecosystem services. However, alpine grassland degradation poses significant threats impacting soil health, stabilization and soil ecosystem function. This study aims to investigate the soil ecological stoichiometric characteristics and influencing factors in alpine grasslands, to find the primary contributing factors in soil degradation process. Soil samples were collected from alpine grassland areas with varying degrees of degradation, including non-, lightly, moderately and severely degraded (ND, LD, MD, SD). Soil characteristics including soil organic carbon (SOC), total nitrogen (TN), total phosphorus (TP), available phosphorus (AP), available nitrogen (AN), microbial biomass carbon (MBC), and microbial biomass nitrogen (MBN) were measured. The results revealed that soil TN, N:P ratios and C:P ratios were below the average level of China’s soil, while C:N ratio were above the average. With increasing aggravation of grassland degradation, SOC, TP, AN, MBN and MBC significantly decreased. Correlation were observed between SOC and C:N ratios as well as C:P ratios in alpine grassland with different degradation degrees (P < 0.05). The C:N ratios in degraded grasslands were lower than those in ND grasslands but higher than the average level of China’s soil. Additionally, the N:P and C:P ratios of LD and MD were significantly higher than those of ND, but lower than the China’s soil average level. In conclusion, soil degradation in alpine grasslands is associated with lower TN contents, higher C:N ratios, and lower N:P ratios, which can impact soil organic matter decomposition and act as plant growth restriction factor. This study provides comprehensive insights into the ecological stoichiometry of alpine grasslands, understanding these factors is essential for the sustainable management of alpine grasslands and restoration strategies for the ecosystem services.

Abstract:Soil nutrient storage and sequestration effects are closely related to the stability of alpine grassland ecosystems, and the study of soil nutrient storage and sequestration effects in alpine grasslands with different degradation levels is of great significance to the recovery of alpine grassland vegetation. On this basis, four alpine grasslands with different degrees of degradation (non-degraded, lightly degraded, moderately degraded, and heavily degraded) were selected in Maqu County, Gannan, and soil samples were collected from 0–10, 10–20, 20–40, and 40–60 cm, respectively, and analyzed for nutrients in terms of storage and sequestration effects. The results showed that soil water content decreased with increasing degradation, while soil pH, bulk density, and conductivity tended to increase. The storage and sequestration effects of soil organic carbon decreased with increasing degradation. And the storage and sequestration effects of soil total nitrogen and total phosphorus were smallest for moderately degraded soils in the 10–60 cm soil layer. Soil water content and soil bulk density were co-influences on soil organic carbon, total nitrogen, and total phosphorus storage. For the soil organic carbon sequestration effect, soil total nitrogen sequestration effect and soil pH were the two most important influencing factors, and the soil total nitrogen and total phosphorus sequestration effect were greatly influenced by soil organic carbon sequestration effect. In addition, soil bulk density was also the main influencing factor of soil total nitrogen sequestration effect, and the impact of soil total nitrogen sequestration on soil total phosphorus sequestration was also greatly influenced.

Abstract:Nitrogen is the crucially sustainable element for most ecosystem including grassland. Fenced alpine meadow grassland (FG), lightly, moderately and heavily degraded (LD, MD and HD) alpine meadow in spring, summer and winter were studied to clear the effects of alpine meadow degradation on soil total nitrogen (TN), available nitrogen (AN), ammonium nitrogen (NH 4 + -N), nitrate nitrogen (NO 3 – -N), inorganic nitrogen (IN) content and the ratio of inorganic nitrogen to available nitrogen (IN/AN), and to find practical management strategies on the background of alpine meadow degradation and ecological environment restoration in the Eastern Qilian Mountains area, northwest China. The results showed that soil total nitrogen contents of the alpine meadow types were at a low level, ranged from 1.60 g·kg −1 to 5.15 g·kg −1 , but soil available nitrogen contents was at a high level, ranged from 373.2 mg·kg −1 to 650.8 mg·kg −1 in the study area. Alpine meadow degradation can seasonally change soil nitrogen status. Degraded alpine meadow had more inorganic nitrogen than fenced meadow in surface soil. Degradation only changed soil NO 3 − -N profile distribution in winter and had no significant effect on profile distribution of TN, AN, NH 4 + -N and IN. There were significant interactions of alpine meadow types, seasons and soil depth on soil available nitrogen, NH 4 + -N, NO 3 – -N and inorganic nitrogen content. Although soil available nitrogen increased under degradation, nitrogen input is needed in the restoration of heavily degraded alpine meadow during the summer growing season to improve vegetation coverage because of the low soil inorganic nitrogen and IN/AN ratio in summer growing season in 10–20 cm and 20–30 cm depth.

Abstract:Abstract Soil moisture, nutrients and environmental conditions have extremely vital effects on vegetation growth and microbial activities in terrestrial ecosystems. To study the changes of soil physical and chemical properties as affected by degradation along the eastern margin of the Tibetan Plateau, using the largest wetland in Ruoergai as the research sample for two consecutive years, we tested the soil bulk density, the soil moisture content, the pH value, the content of organic C, total N and total P of 0~20 cm and 20~40 cm in different degradation degree (light, moderate, serious and non-degraded alpine meadow). The results show that: (1) Degradation had profound effects on the soil physical properties and chemical properties. with the intensification of the alpine meadow degradation in the eastern margin of the Qinghai-Tibet Plateau, the soil organic carbon, total nitrogen, total phosphorus and soil water content decreased significantly but soil bulk density increased significantly. (2) the soil organic carbon, total nitrogen, total phosphorus contents and soil water content increased with the increase of soil depth, however, the soil bulk density show the opposite regular. (3) The correlation analysis indicated that there was an extremely significant positive correlation between soil organic carbon and total nitrogen, total phosphorus, soil water, and the correlation coefficients were 0.822, 0.907 and 0.885 respectively ( P < 0.01). There was an extremely significant positive relationship between the pH and the soil bulk density ( r = 0.488 , p < 0.01), while a significant negative relationship between pH and soil water ( r = –0.387 , p < 0.05). Overall, these results suggested that soil degradation not only changed the soil physical properties but also affected the soil chemical properties indirectly. Protecting surface vegetation and reducing wind erosion could be a good way to curb degradation.

Abstract:A large number of bare patches (BP) were formed in alpine meadows on the Qinghai-Tibet Plateau (QTP) due to serious degradation. The recovery succession of BP may lead to changes in nutrient cycling in grassland ecosystems. However, changes in the intrinsic correlations of soil-microbe-enzyme stoichiometry in response to successional processes of bare patches recovery are uncertain. We selected five types of patches: bare patches (BP), initial recovery patches (IRP), mid-term recovery patches (MRP), long-term recovery patches (LRP) and healthy alpine meadow (AM), to analyze the variation characteristics of soil-microbe-enzyme carbon (C), nitrogen (N) and phosphorus (P) stoichiometry, as well as the changes in soil moisture at different natural succession stages of patchily degraded alpine meadows in the Source Zone of the Yellow River. The results showed that the successional recovery of patchily degraded alpine meadows promoted soil moisture, soil organic carbon (SOC) and total nitrogen (STN) content, soil microbial biomass carbon (MBC), nitrogen (MBN) and phosphorus (MBP), soil extracellular enzyme (alkaline phosphatase (AP) and L-leucine aminopeptidase (LAP)) activities, soil C/P, soil N/P, soil microbial biomass C/N. And C/P and N/P showed an opposite trend, while soil extracellular enzyme (β-1,4-glucosidase (BG), β-1,4- N -acetylglucosaminidase (NAG)) activity and enzyme C/N, C/P and N/P showed an opposite trend. Soil total phosphorus (STP), SOC, STN, MBC, MBN and MBP in the topsoil are higher than in the subsoil. The correlation between soil-microbe-enzyme ecological stoichiometry indicated that soil moisture was the main limiting factor affecting soil-microbe-enzyme nutrient cycling during the natural succession of degraded alpine meadows.

Driving Factors That Reduce Soil Carbon, Sugar, and Microbial Biomass in Degraded Alpine Grasslands

Abstract:Soil carbon and sugars play key roles in carbon (C) cycling in grassland ecosystems. However, little is known about their changes in quantity and composition in degraded alpine meadows in the Tibetan plateau. We compared vegetation C density, soil organic carbon (SOC) density, and soil sugars in nondegraded (ND), degraded (DA; following artificial restoration), and extremely degraded (ED) grasslands and analyzed the relation among these parameters by redundancy analysis (RDA) and structural equation models (SEMs). Belowground biomass, soil microbial biomass C, soil microbial biomass nitrogen (N), belowground biomass C density, SOC density, and soil sugars were lower in DA and ED grasslands than in ND grasslands. In addition, the ratio of belowground biomass to aboveground biomass (BAR) decreased with an increase in degradation. The ratio of belowground biomass to aboveground biomass was identified as the main indirect driving force of ecosystem C density by affecting total vegetation C and SOC densities. Soil dissolved organic carbon (DOC), microbial biomass carbon (SMBC), neutral sugars (NS), and total nitrogen (TN) were identified as main direct driving forces. The ratio of belowground biomass to aboveground biomass altered DOC, SMBC, NS, and TN and, consequently, was the primary driving force for the alpine meadows’ ecosystem C density. It was concluded that land management in alpine meadows should include practices that maintain a relatively high BAR in order to curb degradation and increase ecosystem C density.

A Study on Correlations Between Vegetation Degradation and Soil Degradation in the 'alpine Meadow' of the Qinghai-Tibetan Plateau

Abstract:This paper discusses a study into the characteristics of the soil and the plant communities of the‘alpine meadow’, located on the Qinghai-Tibetan Plateau. Meadows across the region were categorized as having from low to extreme degradation levels. With increasing degradation of the alpine meadow, the grassland quality index, percent plant cover, biomass percent of palatable herbage and the similarity index were all correspondingly shown to decrease across meadows in the alpine meadow region. The species diversity and evenness indexes had the largest values at the meadows characterized as being moderately degraded. The above-ground biomass was the greatest at lightly degraded meadows and lowest at extremely degraded meadows. Increasing degradation levels lead to an increase in the overall biomass of forbs whilst the biomass of sedges and graminoids significantly decreased. The total below-ground biomass was shown to decrease with increased degradation levels. The below-ground biomass change in sedges and graminoids corresponded with the change in total below-ground biomass, however the below-ground biomass of forbs was increased at moderate degradation levels, then decreased at extreme degradation levels. The below-ground biomass at different soil layers decreased, with root-systems displaying increased shallowness with increasing degradation levels. The relationship between above and below-ground biomass was shown to have a strong positive correlation. With increasing soil degradation, soil organic matter, available phosphorus, calcium and manganese, soil hardness and soil water all were shown to decrease whilst soil bulk density was shown to increase. Where soil organic matter losses were severe, available soil nitrogen was shown to be affected substantially, often present at deficient levels. Available copper content was not adequate for herbage growth in severely degraded meadows, however available zinc and manganese still appeared to satisfy plant growth requirements. The degradation of alpine soil was shown to be linked to the degradation of alpine vegetation.
Soil C, N, P and K stoichiometry affected by vegetation restoration patterns in the alpine region of the Loess Plateau, Northwest China

Abstract:The Grain-for-Green project is an important ecological restoration measure to address the degradation of alpine ecosystems in China, which has an important impact on the ecological stoichiometry of soil carbon (C), nitrogen (N), phosphorus (P) and potassium (K). However, soil stoichiometry changes under different vegetation restoration patterns and at different soil depths remain poorly understood in the alpine region of the Loess Plateau. To clarify these soil stoichiometry changes, a 0–60 cm soil profile was sampled from two typical vegetation restoration patterns: grassland (GL) and forestland (FL), including Picea crassifolia (PC), Larix principis-rupprechtii (LR), Populus cathayana (PR) and Betula platyphylla (BP). The control was a wheat field (WF). In all soil layers, the soil organic carbon (SOC), total nitrogen (TN), soil available nitrogen and potassium (AN and AK, respectively) and C:P, C:K, N:P and N:K ratios of FL were higher than those of GL and WF. The TN content and N:P and N:K ratios of GL were higher than those of WF in each soil layer. Additionally, the soil nutrients (except TK) of all vegetation types and stoichiometry of PR and GL (except the N:P ratio of GL) were greater at 0–20 cm than at 20–60 cm. Moreover, the SOC and TN showed the strongest correlation with the soil stoichiometry (except P:K ratio); thus, C and N had the greatest effect on the soil stoichiometry. Furthermore, soil fertility was limited by N. Our results indicated that different vegetation restoration patterns and soil depths had significant effects on the soil nutrients and stoichiometry in the alpine region of the Loess Plateau. The recovery of farmland to forestland promoted better improvements of soil nutrients, and PR had the most significant positive effect on soil surface nutrients.

Nutrients available in the soil regulate the changes of soil microbial community alongside degradation of alpine meadows in the northeast of the Qinghai-Tibet Plateau

Abstract:The alpine meadow in the Qinghai-Tibet Plateau has been seriously degraded due to human activities and climate change in recent decades. Understanding the changes of the soil microbial community in response to the degradation process helps reveal the mechanism underlying the degradation process of alpine meadows. We surveyed and analyzed changes of the vegetation, soil physicochemical properties, and soil microbial community in three degradation levels, namely, non-degradation (ND), moderate degradation (MD), and severe degradation (SD), of the alpine meadows in the northeastern Qinghai-Tibet Plateau. We found that as the level of degradation increased, plant cover, plant density (PD), above-ground biomass (AGB), plant Shannon-Wiener index (PS), soil water content (SWC), soil organic carbon (SOC), total nitrogen (TN), total phosphorus (TP), total potassium (TK), available nitrogen (AN), available phosphorus (AP), and available potassium (AK) decreased significantly, while the soil pH increased from 7.20 to 8.57. Alpine meadow degradation significantly changed the composition of soil bacterial and fungal communities but had no significant impact on the diversity of the microbial communities. Functional predictions indicated that meadow degradation increased the relative abundances of aerobic_chemoheterotrophy, undefined_saprotroph, and plant_pathogen, likely increasing the risk of plant diseases. Redundancy analysis revealed that in ND, the soil microbial community was mainly regulated by PS, PH, PD, SWC, and soil pH. In MD, the soil microbial community was regulated by the soil's available nutrients and SOC. In SD, the soil microbial community was not only regulated by the soil's available nutrients but also influenced by plant characteristics. These results indicate that during alpine meadow degradation, while the changes in the plants and soil environmental factors both affect the composition of the soil microbial community, the influence of soil factors is greater. The soil's available nutrients are the main driving factors regulating the change in the soil microbial community's composition alongside degradation levels.
Density and Stoichiometric Characteristics of Carbon, Nitrogen, and Phosphorus in Surface Soil of Alpine Grassland in Sanjiangyuan

Abstract:The balance and stoichiometry of carbon (C), nitrogen (N), and phosphorus (P) can reflect the quality and content of organic matter and is very important for understanding ecological processes and ecosystem response to climate change and disturbance. To explore the spatial distribution characteristics of C, N, P, and their stoichiometry of topsoil in Sanjiangyuan (SJY), we sampled 0-20 cm soil of alpine grassland in SJY, measured soil C, N, and P, and analyzed their correlation with latitude, longitude, altitude, and vegetation characteristics (diversity, height, coverage, biomass, etc.). The results showed that the average densities of soil organic C (SOC), total N (TN), total P (TP), and soil inorganic C (SIC) in SJY were 7.56 kg·m -2 , 0.71 kg·m -2 , 151.57 g·m -2 , and 1.77 kg·m -2 , respectively. The spatial distribution of SOC, TN, and TP in SJY showed a pattern of high in the east and low in the west, while SIC showed the opposite pattern. The heterogeneity of soil TP in the SJY region was small, which indicated that there may be little difference in soil parent materials in the whole region. The soil nutrients were mainly affected by longitude, soil water content, pH, and vegetation height and coverage. The C: N: P ratio of the 0-20 cm soil in the study area was approximately 48:5:1, and the C: N, C: P, and N: P ratios were significantly affected by soil water content and vegetation height. Soil C: N ratio (10.45) and N:P ratio (4.51) were lower than the national level, which indicated that the soil N mineralization capacity of SJY was higher than that of other regions in China, and the degree of soil N limit was small.

Abstract:With the interaction between global climate change and unreasonable human utilization, the alpine meadows on the Qinghai-Tibet Plateau have suffered various weathering degrees. Uncovering the degradation mechanism and restoration strategies can be facilitated by gaining insights into the diversity of soil microflora during meadow degradation. Therefore, we used Illumina sequencing technology to investigate the patterns of soil microbial diversity, microbial community composition, and the driving factors of microbial change in all non-degraded (ND), lightly degraded (LD), moderately degraded (MD), and severely degraded (SD) alpine meadows in the southeastern Qinghai-Tibet Plateau. Our results pointed out that with the intensification of degradation, vegetation characteristics were significantly reduced, and soil parameters significantly varied among all degraded meadows. The contents of soil organic carbon (SOC), total nitrogen (TN), available phosphorus (AN), and total phosphorous (AK) in soils decreased with the increase of degradation. The dominant bacterial phyla were the same regardless of the meadow degradation level with Actinobacteria (37.67%) and Proteobacteria (20.62%) having the highest relative abundance. Meanwhile, the dominant fungi were Ascomycota (49.9%). Based on the linear discriminant analysis (LDA) and effect size (LEfSe) method, 38 bacterial and 49 fungal species were found to be affected in the degraded alpine meadow, most of which belonged to Actinobacteria and Ascomycota , respectively. Mantel test analysis illustrated that the bacterial community was mainly significantly dependent on below-ground biomass, pH, soil organic carbon, and total nitrogen, while the fungal community was significantly dependent on soil organic carbon, total nitrogen, available nitrogen, and available potassium. These results suggest that the degeneration of alpine meadows contributes to the variability of the diversity and composition of microflora on the Tibetan plateau. Yet this effect is mainly dependent on soil factors.

Long‐term Degradation from Marshes into Meadows Shifts Microbial Functional Diversity of Soil Phosphorus Cycling in an Alpine Wetland of the Tibetan Plateau

Abstract:Soil microbes greatly contribute to the regulating of phosphorus (P) cycling, which plays a significant role in maintaining wetland ecosystem processes and function. The microbial functional diversity of soil P cycling in response to wetland degradation, however, remains largely unknown. We used metagenomic sequencing to investigate the microbial community and genes related to soil P cycling in un‐degraded marshes and meadows derived from long‐term marsh degradation in the Lalu alpine wetland of the Tibetan Plateau. When the marsh degraded into meadow, organic P (OP) mineralization genes increased while genes related to P‐starvation response regulation decreased. Proteobacteria (20.5–74.3%) and Actinobacteria (5.6–59.7%) were the dominant phyla in soils and were also the main contributors (39.7–84.1% in total) to soil P‐cycling genes. Soil pH was the primary factor influencing the P‐cycling functional genes. Soil pH negatively affected the genes related to the P‐uptake and transport system and had negative effects on the genes related to P‐starvation response regulation, OP mineralization, and inorganic P solubilization. These findings may deepen our understanding of the biogeochemical process of soil OP and may be beneficial for wetland management.
[Interaction Effects of Vegetation and Soil Factors on Microbial Communities in Alpine Steppe Under Degradation]

Abstract:To clarify the regulating effect of vegetation and soil factors on microbial communities in the alpine steppe under degradation on the Qinghai-Xizang Plateau, the alpine steppe in the Sanjiangyuan area of the Qinghai-Tibet Plateau was chosen. We analyzed the differences in vegetation and soil factors in different stages of degradation （non-degradation, moderate degradation, and severe degradation） and detected the variations in microbial community characteristics in the alpine steppe under different degradation stages using high-throughput sequencing technology. Eventually, redundancy analysis （RDA） and multiple regression matrixes （MRM） based on the similarity or dissimilarity matrix were used to identify key environmental factors regulating microbial （bacterial and fungal） community changes under degradation. The results showed that the degradation of the alpine steppe significantly changed the community coverage, height, biomass, and important value of graminae； significantly reduced the contents of soil organic matter, total nitrogen, total phosphorus, and silt； and increased the soil bulk density and sand content. Degradation did not change the composition of bacteria and fungi, but their composition proportions changed and also resulted in the loss of microbial richness （Chao1 index and Richness index） but did not significantly change the microbial diversity （Shannon index）. With the occurrence of degradation, the vegetation characteristics, soil physicochemical properties, and microbial diversity showed a consistent change trend. Combined with the characteristics of the network topology changes （the number of nodes and clustering coefficient significantly decreased）, it was found that degradation of the alpine steppe led to the decline of interspecies interactions, decentralization of network, and homogenization of microorganisms, but the cooperation relations among the species were maintained （positive correlation connections accounted for more than 90% in all degradation stages）. Under the alpine steppe degradation, the vegetation-soil interaction had the greatest effect on soil bacterial community, whereas soil physicochemical properties had the greatest influence on soil fungal community. Specifically, vegetation community height, biomass, and soil bulk density were the mutual factors regulating soil microorganisms, whereas the vegetation Simpson index, important value of graminae, soil total phosphorus, total potassium, and silt content were the unique factors affecting the soil bacterial community, and soil pH and total nitrogen content were the particular factors affecting the soil fungal community.
Effects of Different Ecological Restoration Pattern on Soil Organic Nitrogen Components in Alpine Sandy Land

Abstract:Ecological restoration can improve soil fertility and have a significant impact on the soil nitrogen cycle. Nitrogen (N) is an essential nutrient element for plant growth and development, and also an important factor limiting soil productivity. As an important part of soil nitrogen, the composition and proportion of soil organic nitrogen components can directly or indirectly affect the difficulty of soil organic nitrogen mineralization and nitrogen availability, and then affect soil fertility. However, the current studies on soil nitrogen under ecological restoration mainly focus on nitrogen accumulation and nitrogen mineralization, while there are relatively few studies on changes in soil organic nitrogen components, especially in alpine regions. Therefore, in this study, three restoration pattern of mixed forage (MG), single shrub (SA) and shrub combination (SG) that have been restored continuously for 15 years in northwest Sichuan, China, were taken as the research object, and natural sandy land (CK) without manual intervention was taken as the control. Through field sampling and laboratory analysis, the characteristics of the soil nitrogen content and its proportion to soil total nitrogen (TN) under ecological restoration in alpine sandy land in northwest Sichuan, China, were investigated, and the correlation between the nitrogen content and soil physicochemical properties was analyzed. The results showed that the three ecological restoration patterns significantly increased the contents of acylated ammonium nitrogen (AMMN), acid-lyzed amino sugar nitrogen (ASN), acid-lyzed amino acid nitrogen (AAN), acid-lyzed unknown nitrogen (HUN), acid-lyzed total nitrogen (AHN) and non-acid-lyzed nitrogen (NHN) in soil, and the change trend was consistent with that of soil TN. Ecological restoration improved soil nitrogen mineralization and storage capacity by increasing the proportion of AAN, HUN and NHN to soil TN, and the effect was most obvious in the MG pattern 20–40 cm and SG pattern 40–60 cm soil layers. In general, except ASN, the soil nitrogen content was positively correlated with the soil TN, soil water content (SWC) and soil organic carbon (SOC), and negatively correlated with the soil bulk density (BD) and pH. The results of this study will help us to understand the supply capacity of soil nitrogen under ecological restoration and provide a scientific basis for the selection of an ecological restoration mode and the improvement of the restoration effect and efficiency in alpine sandy land.

Abstract:As important terrestrial ecosystems on the Qinghai‒Tibet Plateau with important ecological and economic value, alpine meadows in some regions are in a degraded state due to climate change and unsustainable grazing. Due to the fragile ecological environment, exploring sustainable restoration models of degraded alpine meadows using close-to-nature restoration measures is the direction of ongoing efforts. Thus, in this study, three close-to-nature restoration measures, namely, manure application (M), Poa annua, Elymus dahuricus , and Puccinellia distans mixed with non-tillage reseeding (R), and the combined treatments (MR), were used to restore degraded alpine meadows from 2019 to 2021. The results showed that the M and MR treatments significantly enhanced plant biomass and reduced forbs dominance, thus improving community structure. Moreover, the M and MR treatments significantly affected plant and bacterial alpha diversity. However, the three restoration measures did not alter the correlation between the plant and bacterial communities. Furthermore, these three measures resulted in a significant increase in the plant carbon:nitrogen ratio and a significant decrease in the nitrogen:phosphorus ratio, while neither the plant carbon:phosphorus ratio nor the soil stoichiometric ratio changed significantly, indicating that nitrogen limitation was an important factor that limited the recovery process of the alpine meadows. The structural equation model indicated that the plant nitrogen:phosphorus ratio may regulate the response of plant and bacterial diversity to different restoration measures in alpine meadows. Our findings emphasize that the management of degraded alpine meadows using close-to-nature restoration measures should be a long-term effort, with more attention given to changes in the nitrogen content of meadow ecosystems.

Abstract:Microorganisms drive many important soil processes and ecosystem functioning; however, the soil microbial community can respond to environmental change, which can alter decomposition and storage of soil organic carbon (SOC). We assessed soil microbial taxonomic and functional attributes linked to SOC decomposition and concentration based on a grassland experiment fertilized with nitrogen (N) and phosphorus (P). There were divergent responses at both the taxonomic and functional level to fertilization, with a greater quantitative response in terms of the taxonomic attributes of bacteria and fungi than the abundance of SOC degradation genes. There was a negative correlation between SOC concentration and the abundance of SOC degradation genes. This indicated that N and P fertilization had the potential to stimulate SOC decomposition through modification of the soil microbial community, particularly by increasing the abundance of SOC degradation genes. Redundancy and variation partitioning analyses, as well as structural equation models revealed that soil total P was a pivotal factor shaping the microbial community both for taxonomic and functional gene attributes in the alpine meadow. The abundance of SOC degrading genes had stronger effects on SOC concentrations than did taxonomic attributes. Quantification of the abundance of SOC degradation genes can provide more direct and effective understanding of SOC decomposition processes and prediction of shifts in soil function as organic C pool in grasslands than a focus on the taxonomy of microbial communities. Given the important role of microbial community in regulating SOC decomposition and lower SOC concentrations under N and P fertilization, we recommend that the alpine meadow should be protected from mineral fertilizers, particularly P alone fertilization, to sustain high grassland ecosystem function as SOC sink in this region.

Abstract:Alpine wetland plays an important role in the global carbon balance but are experiencing severe degradation under climate change and human activities. With the aim to clarify the effect of alpine wetland degradation on carbon fluxes (including net ecosystem CO 2 exchange, NEE; ecosystem respiration, ER; gross ecosystem productivity, GEP, and CH 4 flux), we investigated 12 sites and measured carbon fluxes using the static chamber method in the Zoige alpine wetland during August 2018, including undegraded wetland (UD), lightly degraded wetland (LD), moderately degraded wetland (MD), and severely degraded wetland (SD). The results showed that carbon sink strengths differ among the Zoige wetlands with different degradation stages during the growing season. From UD to LD, the rate of carbon sequestration (mean value of NEE) increased by 25.70%; however, from LD to SD, it decreased by 81.67%. Wetland degradation significantly reduced soil water content (SWC), soil organic carbon (SOC), microbial biomass carbon (MBC), and microbial biomass nitrogen (MBN). NEE was significantly correlated with MBC and MBN, while ER was positively correlated with ST but negatively correlated with SOC ( P < 0.01). Among all measured environmental factors, GEP was positively correlated with pH ( P < 0.01), while CH 4 flux was most closely correlated with SOC, SWC, MBC, MBN, and ST ( P < 0.001), and was also affected by pH and NO 3 – content ( P < 0.01). These results suggest that the capacity of carbon sequestration in the Zoige wetlands reduced with intensification of the degradation. This study provides a reference for sustainably managing and utilizing degraded wetlands under climate change.

Abstract:Abstract Wet meadows provide opportunities to decrease carbon dioxide (CO 2 ) and methane (CH 4 ) released into the atmosphere by increasing the soil organic carbon (SOC) stored in wetland systems. Although wet meadows serve as the most important and stable C sinks, there has been very few investigations on the seasonal distributions of SOC fractions in high-altitude wet meadows. Here, we studied the effects of four vegetation degradation levels, non-degraded (ND), lightly degraded (LD), moderately degraded (MD), and heavily degraded (HD), on the measured vertical and seasonal changes of SOC and its different fractions. Among these vegetation degradation levels, 0–10 and 10–20 cm soil depths in ND plots had significantly higher SOC contents than the other degradation levels had throughout the year. This is attributed to the relatively greater inputs of aboveground plant litter and richer fine-root biomass in ND plots. Particulate organic carbon (POC) and light fraction organic carbon (LFOC) showed similar vertical and seasonal variations in autumn, reaching a minimum. Moreover, microbial biomass (MBC) and easily oxidizable organic carbon (EOC) contents were highest in summer and the smallest in winter, while dissolved organic carbon (DOC) content was highest in spring and lowest in summer, and were mainly concentrated in the 0–20 cm layer. Pearson correlation analysis indicated that soil properties and aboveground biomass were significantly related to different SOC fractions. The results indicate that vegetation degradation reduces the accumulation of total SOC and its different fractions, which may reduce carbon sink capacity and soil quality of alpine wet meadows, and increase atmospheric environmental pressure. In addition, vegetation biomass and soil characteristics play a key role in the formation and transformation of soil carbon. These results strengthen our understanding of soil C dynamics, specifically related to the different C fractions as affected by vegetation degradation levels and soil depth, in wet meadow systems.

Abstract:PurposeThe main objectives of this research are to decipher the stoichiometric characteristics of carbon (C), nitrogen (N), and phosphorus (P) in soils from the alpine ecosystem and to obtain information about nutrient limitation on plants and microbes.Materials and methodsThe soils were sampled along an altitudinal gradient (2000 to 4300 m above sea level) from the eastern slope of Gongga Mountain in eastern Tibetan Plateau. In total of 102 soil samples in profiles and 27 soil microbial biomass (SMB) samples from five vegetation zones were collected to analyze the concentrations of C, N, and P as well as their ratios. The concentrations of C and N were measured using an automated C/N analyzer, total P was detected by inductively coupled plasma-atomic emission spectrometer, and the concentrations of microbial biomass C, N, and P were measured by the chloroform fumigation-extraction method. Soil P fractions were extracted by modified Hedley sequential extraction method.Results and discussionThe concentrations of C, N, and P in the soils and SMB varied spatially, whereas the variation of their ratios was constrained. The C:N:P ratios were 556:22:1 for the O horizon, 343:16:1 for the A horizon, 154:7:1 for the B horizon, and 63:3:1 for the C horizon, indicating a significant decrease with depth. The mean ratio in the SMB was 51:6.6:1. Microbial biomass C, N, and P were important components of soil nutrients, especially the microbial biomass P which accounted for 40.8 % of soil available P. The C:P and N:P were higher in the soils of broadleaf-coniferous and coniferous forests, whereas the ratios in the SMB were higher in the broadleaf forest. The ratios of C and N to available P in the soils decreased significantly with altitude.ConclusionsThe local climate, vegetation succession, and soil development in the high mountain resulted in the soil nutrient cycling different from that in other terrestrial ecosystems. Among the different vegetation zones, the P-limitation of plants and microbial communities might be possible in the soils of lower land forests in the long term.

Abstract:Abstract Vegetation degradation, due to climate change and human activities, changes the biomass, vegetation species composition, and soil nutrient input sources and thus affects soil nutrient cycling and enzyme activities. However, few studies have focused on the responses of soil nutrients and enzymes to vegetation degradation in high-altitude wet meadows. In this study, we examined the effects of vegetation degradation on soil nutrients (soil organic carbon, SOC; total nitrogen, TN; total phosphorus, TP) and enzyme activities (i.e., urease, catalase, amylase) in an alpine meadow in the eastern margin of the Qinghai-Tibet Plateau. Four different levels of degradation were defined in terms of vegetation density and composition: primary wet meadow (CK), lightly degraded (LD), moderately degraded (MD), and heavily degraded (HD). Soil samples were collected at depth intervals of 0–10, 10–20, 20–40, 40–60, 60–80, and 80–100 cm to determine soil nutrient levels and enzyme activities. The results showed that SOC, TN, catalase and amylase significantly decreased with degradation level, while TP and urease increased with degradation level ( P < 0.05). Soil nutrient and enzyme activity significantly decreased with soil depth ( P < 0.05), and the soil nutrient and enzyme activity exhibited obvious "surface aggregation". The activities of soil urease and catalase were strongest in spring and weakest in winter. The content of TN in spring, summer, and autumn was significantly higher than observed in winter ( P < 0.05). The soil TP content increased in winter. Soil amylase activity was significantly higher in summerm than in spring, autumn, and winter ( P < 0.05). TP was the main limiting factor for plant growth in the Gahai wet meadow. Values of SOC and TN were positively and significantly correlated with amylase and catalase ( P < 0.05), but negatively correlated with urease ( P < 0.05). These results suggest the significant role that vegetation degradation and seasonal freeze–thaw cycle play in regulating enzyme activities and nutrient availability in wet meadow soil.
