Abstract:Bacillus anthracis is the causative agent of anthrax and can be contracted by humans and herbivorous mammals by inhalation, ingestion, or cutaneous exposure to bacterial spores. Due to its stability and disease potential, B. anthracis is a recognized biothreat agent and robust detection and viability methods are needed to identify spores from unknown samples. Here we report the use of smartphone-based microscopy (SPM) in combination with a simple microfluidic incubation device (MID) to detect 50 to 5000 B. anthracis Sterne spores in 3 to 5 hours. This technique relies on optical monitoring of the conversion of the ∼1 μm spores to the filamentous vegetative cells that range from tens to hundreds of micrometers in length. This distinguishing filament formation is unique to B. anthracis as compared to other members of the Bacillus cereus group. A unique feature of this approach is that the sample integrity is maintained, and the vegetative biomass can be removed from the chip for secondary molecular analysis such as PCR. Compared with existing chip-based and rapid viability PCR methods, this new approach reduces assay time by almost half, and is highly sensitive, specific, and cost effective.

Abstract:Hydrocyclone separation technique has commonly been applied in engineering for solid-liquid separation. In aquaculture, since Hydrodynamic Vortex Separator (HDVS) is able to control flocs concentration effectively, the Recirculating Biofloc Technology (RBFT) has been gradually acknowledged (Zhu et al., 2016). In order to operate RBFT system at maximum performance, the removal efficiency of HDVS at different Hydraulic Retention Time (HRT) must be fully predictable. The aim of this work is to investigate the relationship between separation efficiency and HRT of HDVS applied in RBFT system, through Computational Fluid Dynamics (CFD) method. For this, a three-dimensional unsteady transient model was developed to simulate the hydrodynamics in HDVS based on the commercial software Workbenching 17.0. A Two Fluid Model (TFM) using kinetic theory of granular flow (KTGF) has been developed to describe the dense solid-liquid (flocs-water) flow field of HDVS in RBFT system. Additionally, the Reynolds averaged Navier-Stokes (RANS) equations with Reynolds stress turbulence model (RSM) are solved by use of the finite volume method based on the SIMPLE pressure correction algorithm in the computational domain. Finally, pilot-scale studies were conducted to evaluate the accuracy and precision of simulation models applied. The results indicate that the flocs removal efficiency of HDVS obtained from tests were less than that from the simulation results at three different HRTs. But it decreased with the decline of HRT both for experiment and simulation. Additionally, the CFD model proposed can enhance the fundamental understanding of the effects of HRT on separation efficiency and at the same time, probably coupled with physical experiment whenever necessary, offer a more reliable way for optimum control of HDVS under varying conditions. Based on the simulation results, the flocs management in RBFT system is briefly discussed.

Abstract:Recirculating biofloc technology (RBFT) has been gradually acknowledged for its positive effect on the control of biofloc concentration using a hydrodynamic vortex separator (HDVS). To operate an RBFT system at maximum performance, the removal efficiency of an HDVS at different hydraulic retention times (HRTs) must be fully predictable. Hence, a numerical study of the fluid flow and particle dynamics was performed to characterize the performance of an HDVS at varying HRTs. First, flow simulation was conducted to determine an economical mesh size at an HRT of 248 s. Then, with respect to the total suspended solids (TSS) in the RBFT system and the physical properties of the flocs, two-way coupling of the dense discrete phase model (DDPM) and discrete element model (DEM) methods was used to predict floc tracking in an HDVS. Additionally, the Reynolds averaged Navier-Stokes (RANS) equations with the Reynolds stress turbulence model (RSM) were solved using the finite volume method based on the semi-implicit method pressure-linked equations (SIMPLE) pressure correction algorithm in the computational domain. Finally, pilot-scale studies were conducted to verify the simulation models. Based on the simulation results, floc management in an RBFT system is briefly discussed. Due to limited research on the numerical simulation and operating conditions of an HDVS in an RBFT system, this article describes an original investigation of the modeling approach.
Optimization design of aerated struvite-crystallizer for recovering phosphorus from swine manure using computational fluid dynamics

Abstract:The aerated struvite-crystallizer (ASC) is widely used to recover phosphorus from swine manure. However, there is a lack of knowledge on the optimization of ASC design in view of hydrodynamics. In this study, computational fluid dynamics (CFD) is used to describe the behavior of water and air bubble in terms of velocity magnitude and stagnant volume in a column-shape ASC with cone bottom under different conditions i.e., ratio of height to diameter in the reaction zone (Hi:Di ratio) and aeration intensity. Results show that decreasing Hi:Di ratio does harm to the settlement in the bottom of ASC for the vortex fluid, whereas increasing Hi:Di ratio will shorten the retention time for air. Additionally, the aeration intensity has influence not only on the mixture effectiveness within the inner cylinder reaction zone of ASC, but also on the fluid field in the bottom of ASC. The optimized Hi:Di ratio is determined as 2:1 and the suitable aeration intensity is 14.6m3/h for the ASC system mentioned in this study.
Effect of extracellular polymeric substances on sludge reduction in the humic-activated sludge anoxic-oxic-settling-anaerobic process

Abstract:The wide utilization of the activated sludge process for treating domestic wastewater has resulted in the huge production of excess sludge, posing a serious disposal problem. In the present study, a humic-activated sludge-anoxic-oxic-settling-anaerobic (HS-A-OSA) process was used to study the function of humic activated sludge on the efficiency of sludge reduction. For this purpose, two-anoxic-oxic-settling-anaerobic (A-OSA) experimental devices were fabricated. Between the two devices, one device with the composite humic biological filler served as the HS-A-OSA process and the other device served as the comparison. The processes were operated with a mixed liquor suspended solids concentration of 3,000 mg/L for a period of 60 days. The sludge internal recycling was 100% from the sedimentation basin to the anaerobic sludge reaction basin, followed by the anoxic basin. The return sludge residence time was 5.5 h with both devices. During the operation period, 23.2% of excess sludge reduction was observed. The chemical oxygen demand and total nitrogen removal efficiencies improved in the HS-A-OSA process. Based on the variations in polysaccharide, protein, humic acid and extracellular polymeric substances concentrations in different basins, the main reason for improved sludge reduction was that the sludge disintegration velocity of the HS-A-OSA process was higher than the A-OSA process.

Abstract:In this study, a swirl flocculation-enhanced reactor (SFR) was designed to address the problems of high dosage and poor pollutant removal in the pretreatment process of coal chemical wastewater (CCW). The effects of different height-to-diameter (H/D) ratios, stirring speed, and impeller length on SFR flow field were analyzed by computational fluid dynamics (CFD) simulation. The optimal H/D and operating condition of SFR were determined by comparing and analyzing fluid velocity, eddy viscosity, and turbulent kinetic energy. Then the lab-scale SFR was manufactured to conduct agglomeration tests. The results showed that the flocculation effect was the best when the H/D ratio of SFR is 8:5 and the mixing blade length was at 3/10 of the reactor as well as 180 r/min of agitating rate. The removal rates of oil, suspended solids (SS), chemical oxygen demand (COD) and phenol in CCW could reach 73.43%, 87.88%, 78.63% and 65.20%, respectively. In addition, the plausibility of SFR design was further validated by the fact that the reagent dose was reduced by 50–60% and the removal rate was increased by 20–30% in comparison with the beaker flocculation experiment. The SFR flow field flocculation enhancement mechanism was analyzed. It is expected that the reactor designed in this work, has a wide range of industrial applications in the CCW treatment.

Abstract:Conditioning is essential for achieving effective sludge dewatering and easier disposal. In this study, a combined pretreatment of acidification and oxidation using potassium permanganate (KMnO4) as oxidant was conducted to improve the effect of a cationic starch-based flocculant (St-WH) on the dewaterability of sewage sludge. Synergetic dewatering mechanisms by acidification, oxidation, and flocculation are discussed in detail according to the analysis of the changes in bound water content, extracellular polymeric substances (EPS) fractions and components, zeta potentials, floc size, and surface microstructures of sludge cakes in the dewatering process. Acidification and oxidation could destroy the sludge flocs, thereby causing the degradation of EPS and formation of fine particles. Original loosely and tightly bound EPS partially converted to soluble EPS, resulting in release of trapped water, which can be reflected by the significant correlation between loosely bound EPS and filter cake moisture content (FCMC) (R-s = 0.83, P < 0.05). Those fine particles, simultaneously produced, were adverse to filtration efficiency. In addition to enhancing the oxidation effect of KMnO4 , acidification treatment could still compress the protein-like materials in soluble EPS due to protonation effect, which was positively related to specific resistance to filtration (SRF) (R-s = 0.74, P < 0.05). The following flocculation using St-WH efficiently aggregated those fine particles and restrained the released EPS to bind with free water through charge neutralization and bridging effects, thereby resulting in improved filtration performance and enhanced removal of bound water (R-s = 0.88, P < 0.01). Response surface methodology was also applied to achieve an optimal condition and evaluate the effects of various environmental factors.

Abstract:Abstract Two series of binary graft cationic starch-based flocculants (CS-DMCs and CS-DMLs) with different hydrophilicity and charge density (CD) were prepared by graft copolymerization of acrylamide with 2-(Methacryloyloxy)- N , N , N -trimethylethanaminium chloride and methacrylic acid 2-(benzyldimethylaminio) ethyl chloride, respectively, on the starch (St) backbone. The sludge dewatering performance of CS-DMCs and CS-DMLs were evaluated and compared based on the changes in filter cake moisture content (FCMC), specific resistance of filtration (SRF), fractions and components of extracellular polymeric substances, and various physiochemical characteristics of sludge flocs and cakes. Increase in CD of the St-based flocculants caused improved sludge dewaterability. Under the similar CD, CS-DML with relatively high hydrophobicity exhibited lower FCMC and SRF, larger and denser sludge flocs, and better permeability of sludge cakes than CS-DMCs due to the synergistic effects of charge neutralization, bridging flocculation and hydrophobic association. Furthermore, a second-order polynomial model on the basis of phenomenological theory was successfully applied to quantitatively evaluate the influences of the two important structural factors of these St-based flocculants, i.e., hydrophobicity and CD, on the sludge dewaterability. The structure–activity relationship of the St-based flocculants in sludge dewatering was obtained according to the theoretic simulation. The dewatering mechanisms was discussed in depth on the basis of the experimental and simulated results; besides, the FCMC and optimal dose can be predicted by the established structure–activity relationship. This current work offered a novel and valuable way to exploit and design of low-cost and high-performance graft natural polymeric flocculants applied in efficient conditioning of sludge.

Abstract:Rheology characteristics of raw sludge and cationic polyacrylamide (CPAM) conditioned sludge were both studied by steady test, thixotropy test, and dynamic test, respectively. Results of steady test showed Herschel–Bulkley, Bingham and Casson models could accurately describe the flow behavior of raw sludge and conditioned sludge, as the CPAM dosage was lower than 3 mg/g. An initial peak point for CPAM conditioned sludge at a low shear rate was observed, where the higher this peak was, the more energy was needed to break floc network. However, at a larger shear rate, the trend of rheogram was similar to that of raw sludge. Thixotropy was prominent for conditioned sludge, and the thixotropic loop was expanded with the increase of CPAM dosage. The strain sweep test demonstrated that the strength of floc network was intensified, leading to the larger elastic modulus. MGKV model which connected Maxwell model and series of Kelvin–Voigt modules was developed to characterize the creep recovery of sludge before and after conditioned. Furthermore, CPAM has more significant effect on delay time λ1, which reduced by about 90% as the dosage was up to 3.0 mg/g. A smaller value of λ1 explicated that conditioned sludge could restore to a new stable status in a shorter time. Compared to λ2 and λ3, λ1 has more significant dominance during the entire creep recovery test.

Abstract:The activated sludge process is widely used for wastewater treatment, and the settleability and dewaterability of activated sludge are crucial to the operation of wastewater treatment plants. In this study the surfactant-mediated settleability and dewaterability of activated sludge were investigated. It is found that dose of anionic surfactant sodium dodecyl sulfate (SDS) caused substantial release of extracellular polymer substances (EPS), especially proteins, from sludge. Laser light scattering (LLS) results reveal that the released EPS became stretched and more expanded when the SDS dosage was increased. The sludge dewaterability was significantly deteriorated at a dosage of 2.084g/L SDS, and meanwhile the capillary suction time (CST) and specific resistance of filtration (SRF) increased by 6.3 and 5.1 times, respectively, than the raw sludge. The correlations between the EPS contents and the sludge properties, i.e., CST, SRF, moisture content of filter-cake and sludge volume index, were evaluated and the significant roles of proteins in governing the EPS properties as well as the settleability and dewaterability of sludge were further confirmed. This study elucidates how the settleability and dewaterability of activated sludge were affected by surfactants via changing the properties and structure of the released EPS from sludge.
Effects of contact/stabilization time ratio on organic carbon-methane conversion in high-rate contact stabilization system

Abstract:The high-rate contact stabilization (HRCS) process is a promising technique for recovering carbon from wastewater. However, its practical application faces challenges due to its low chemical oxygen demand (COD) removal efficiency, which requires further optimization. The objective of this study was to investigate the optimal process conditions for enhancing COD removal efficiency and carbon recovery rate (CRR) in the HRCS process. To achieve this, a sequencing batch reactor (SBR) was utilized to maximize the feast-famine regime. Although the HRCS process exhibited a lower COD removal efficiency compared to the high-rate activated sludge (HRAS) process in SBR mode, it demonstrated a 16.3 % higher CRR. The influence of the contact time to stabilization time (t c /t s ratio) on both COD removal efficiency and CRR was investigated. The optimum condition was achieved at a t c /t s ratio of 0.18, which resulted in a COD removal efficiency of 60.2 ± 3.0 % and a CRR of 0.417 ± 0.041 g-COD CH4 /g-COD inf (COD CH4 : produced methane as COD, COD inf : influent COD). The microbial communities in HRAS and HRCS exhibited simplification compared to conventional activated sludge. Distinct shifts were observed in response to changes in solids retention time and t c /t s ratio. Under harsher operating conditions, Lactococcus spp. became the predominant species, achieving the greatest dominance at a t c /t s ratio of 0.18. This is because they produce more extracellular polymeric substances and polyhydroxyalkanoates, which improve organic capture. The operating conditions identified in this study are anticipated to make a significant contribution to practical applications aimed at enhancing carbon recovery.

Characteristics of free cells and aggregated flocs for the flocculation and sedimentation of activated sludge

Abstract:Laboratory experiments were conducted to investigate the correlation of the free cells/aggregated flocs characteristics and the sludge flocculation and separation behavior during the activated sludge process. Activated sludge was cultured in three laboratory-scale batch reactors with same carbon sources of glucose but different sludge retention times (SRTs) of 5, 10 and 20 days. The variation in the operation condition produced sludge with different flocculation and separation characteristics. The reactor performance in terms of bioflocculation as measured by the amount of suspended solids in the effluent, sludge sedimentation and compression as measured by the sludge value index improved considerably as the SRT lengthened. The higher SRT was related to less negatively charged surface of free cells and more hydrophobic of aggregated flocs. The negative zeta potential of aggregated flocs (11–13 mV) was smaller than that of free cells (15–23 mV), and free cells carrying lesser negative surface charges resulted in effluent clarification. Hydrophobicity of aggregated flocs (10–17 %) in each reactor was higher than that of free cells (5–8 %). The increase in hydrophobicity of aggregated flocs had positive effect on settleability. Floc strength measured by breakage/erosion index was closely related to hydrophobicity of aggregated flocs, but no correlation could be established with zeta potential of aggregated flocs. In addition, different bacterial species existed in free cells and aggregated flocs. Microbial similarity between free cells and aggregated flocs increased with shorter SRT and was closely related to effluent clarification.
Enhancement of flocculation and dewaterability of a highly stable activated sludge using a hybrid system of organic coagulants and polyelectrolytes

Abstract:This study investigates the influence of hybrid coagulation-flocculation system on the flocculation and dewaterability efficiency of a real highly stable activated sludge obtained from the membrane bioreactor tank of a petroleum industry. Two organic coagulants, polyDADMAC (FL 4440) and polyamine (FL 2949), coupled with differently structured polyacrylamides (PAMs) are used. Residual turbidity, Zeta potential (ζ), flocs size, and capillary suction time (CST) are used to assess the impact of the hybrid system on the degree of flocculation and dewaterability of MBR sludge. The study reveals that electrical double layer compression, which lowers the electrostatic repulsion between the particles, contributes to the improvement in flocculation behavior. Addition of FL 4440 and FL 2949 prior to PAM has greatly reduced the turbidity and ζ of the sludge suspension by decreasing the required doses of PAMs from 67 % to 80 % depending on the type of the PAMs used. Addition of FL 2949 and FL 4440 has strong influence on the linear structured PAMs with 71 and 80 % reduction in the required PAM's dose for the 40 % Charge density (CD) (FO 4490 SSH) and 60 % CD (FO 4690 SSH), respectively followed by slightly and highly branched structured PAMs. Similar observations have been noted for flocs size and dewaterability (CST) measurements of the flocculated MBR sludge. Overall, this study has successfully determined the effect of hybrid coagulation-flocculation on the flocculation performance of MBR sludge.

Abstract:Activated sludge processes with an ultra-short sludge retention time (ultra-short-SRT) are considered to have potential for energy and resource recovery from wastewater. The present study focused on the sludge characteristics, system performance and microbial kinetics in ultra-short-SRT activated sludge (USSAS) processes using typical domestic wastewater (SRT = 0.5, 1, 2, 3 and 4 d). The results showed that compared with the sludge in conventional activated sludge (CAS) processes, the sludge structure in USSAS system was looser (fractal dimension, D2P, 1.19–1.33), the boundary was rougher (pore boundary fractal dimension, DB, 1.44–1.59), the sludge concentration was lower, and the sludge volume index (SVI) was higher; bacteria such as Thiothrix and Trichococcus that cause sludge bulking, which poses an operation risk, were extensively detected, especially at SRTs of 0.5 d and 1.0 d. The performance in terms of total chemical oxygen demand (tCOD) and phosphorus removal increased with increasing SRT, and the highest removal rate (approximately 85% for tCOD and 90% for phosphorus) was observed when the SRT was 4 d. Both bioconversion and biosorption were responsible for the C/P separation, and their roles were different for different types of organic matter and phosphorus under different SRT conditions. The proportion of phosphate-accumulating organisms (PAO) reached 2.4% when the SRT was 3 d, resulting in highly effective biological phosphorus removal. The values of microbial kinetic parameters such as YH and KdH in USSAS systems were higher than those in CAS systems, indicating faster microbial community renewal. This study was helpful for understanding the characteristics of USSAS process.

Abstract:This study addresses the challenge of the reverse effect observed in the dewatering process of waste slurry as poly-aluminum chloride (PAC) dosages increase. It investigates the flocculation mechanism of PAC at varying dosages in a high-water-content slurry using molecular dynamics simulation methods. The results indicate that PAC molecules primarily adsorb montmorillonite particles while exerting negligible influence on quartz. Dewatering efficiency is influenced by the adsorption-repulsion interaction at the PAC-montmorillonite interface. Efficiency increases with PAC dosage up to an optimum level due to enhanced adsorption but decreases thereafter due to increasing repulsion. Within this optimal range, montmorillonite interfaces exhibit a higher negative charge, facilitating stable PAC adsorption and leading to increased aggregation of montmorillonite particles, thereby forming flocs. The adsorption capacity of PAC on montmorillonite particles escalates with dosage until achieving the optimal flocculation effect. However, surpassing this optimal dosage prompts repulsion among outer PAC molecules, gradually causing montmorillonite particles to separate due to repulsion, thereby weakening the flocculation effect. This research provides theoretical support for the flocculation and dewatering of PAC in high-water-content waste slurry, emphasizing the rational and cost-effective addition of PAC in practical applications.

Abstract:Flocculation is a significant process in treating shale gas hydraulic fracturing flowback fluids. To achieve a high removal rate of pollutant, a range of multi-stage enhanced flocculation reactors that incorporated sequential intense, moderate, and weak mixing sections along the flow direction were devised, in which the aspect ratios (ARs) in the intense mixing section were varied. The results of the computer simulation indicated that the reactor with AR of 3.5 exhibited a gradual decrease in velocity and energy along the flow direction, and the flow field distribution is highly uniform and symmetrical. The generated flow field promoted the formation of small flocs in the intense mixing section and facilitated the collision of small flocs in the moderate mixing section, while preventing the fragmentation of flocs in the weak mixing section, therefore resulting in the generation of large and compact flocs. Moreover, the generated flocs demonstrated a good resistance to shear and oscillation, facilitating their separation from the simulated flowback fluids. The obtained reactor achieved remarkable removal efficiencies of 80.15 % for COD and 89.06 % for turbidity. Furthermore, when treating bentonite suspension, the multi-stage flocculation reactor achieved a turbidity removal rate of 99.34 % with a processing time of only 2.38 min and a settling time of just 20 min. This demonstrated the broad applicability of this reactor to various types of wastewater. This work provides valuable insights into the design of efficient flocculation reactors by integrating multiple mixing intensities within a reactor.

Abstract:Vacuum preloading combined with flocculation has shown great potential for improving the dredged sludge's dewatering and consolidation. To identify a suitable flocculant combined with vacuum preloading method, this study investigated the effects of various common flocculants on the sludge's filtration properties and consolidation behaviors. The results indicated that dual-use of polymeric aluminium chloride (PAC) and polyacrylamide (PAM) had the most significant improvement on sludge's filtration performance, as reflected by capillary suction time. In addition, a single dose of PAM had the most substantial improvement on the sludge's compressibility. While, lime improved the sludge's permeability the most. To predict the consolidation settlement of flocculated sludge during vacuum preloading, a method was proposed based on a small homemade vacuum preloading apparatus and the Taylor method. The influence of different flocculants on sludge properties was also analyzed based on their flocculation mechanisms and microstructure. Finally, an appropriate flocculant for the sludge in vacuum preloading projects was suggested in accordance with different project goals.

Abstract:Laboratory experiments were carried out on activated sludge (AS) to investigate the correlations between the content of extracellular polymeric substances (EPS) and the performance of biosolids–water separation, including sludge flocculation, sedimentation, compression, and dewatering, under non-steady-state conditions. On three stabilized AS reactors changes were made in sludge retention time (SRT), substrate composition, and loading rate, respectively, to bring about unstable operation to the reactors. A two-step heating method was used to extract from the sludge the easily extractable EPS, or loosely bound EPS (LB-EPS), and tightly bound EPS (TB-EPS), respectively. The experimental results demonstrate dynamic changes in sludge characteristic and EPS production under the non-steady-state conditions. During the early phase of transition after a change was imposed, the sludge became generally worse in flocculation, compressibility, and dewaterability. With the acclimatization of the biomass to the new process conditions, biosolids–water separation showed a general trend of improvement. Changes in AS process condition also resulted in considerable variations in EPS production. The change of the LB-EPS content appeared to be more significant than that of the TB-EPS. Throughout the non-steady-state operation, the sludge flocculating behavior, settleability, compressibility, and dewaterability had a positive correlation with the LB-EPS content; however, no correlation could be found between these properties and the TB-EPS content. The results suggest that although EPS is essential to biofloc formation, excessive EPS in the form of LB-EPS would weaken cell attachment and deteriorate the AS floc structure, resulting in poor biosolids–water separation.

Abstract:In this research, the effects of peracetic acid (PAA), polymeric flocculants, and their combined conditioning on improving the dewatering performance were comprehensively evaluated. The results showed that sludge cake moisture content, capillary suction time (CST), and specific resistance to filtration (SRF) were 70.6%, 48.1 s, and 3.42 × 1012 m/kg after adding 0.10 g/gMLSS PAA for 50 min, representing reductions of 12.60%, 40.32%, and 33.98%, respectively. Additionally, conditioning of sludge with polyferric sulfate (PFS), polyaluminum chloride (PAC), and cationic polyacrylamide (CPAM) enhanced sludge properties in the following order: CPAM > PAC > PFS. After the PAA oxidation and re-flocculation process, the optimal dosages of PFS, PAC, and CPAM were reduced to 1.5 g/L, 0.9 g/L, and 0.04 g/L, respectively. The sludge dewatering performance significantly improved, with sludge cake moisture content measuring 65.8%, 66.3%, and 61.7%, respectively. Moreover, the spatial multi-porous skeleton structures were formed via re-flocculation to improve the sludge dewatering. Furthermore, economic evaluation validated that the pre-oxidation and re-flocculation process could be considered an economically viable option. These research findings could serve as a valuable reference for practical engineering applications.
