Abstract:Wound ulceration caused by diabetes is a typical chronic wound wherein healing the local tissue is difficult due to lack of blood vessels and tissue necrosis caused by the long-term accumulation of free radicals. Near-field electrospinning (NFES) is an innovative technology used to produce micro-nano-scaled, controllable sequencing fibers. In this study, we constructed a novel wound dressing based on the NFES polycaprolactone (PCL) fiber network and modified gelatin with methacrylic anhydride (GelMA) hydrogel to promote angiogenesis and the re-epithelialization of diabetic wounds. An angiogenic and antioxidant drug named deferoxamine (DFO) was encapsulated in a GelMA hydrogel to achieve a slow-release effect that is more suitable for chronic wounds. The cell adhesion experiment showed that the cells could attach to the fibers in the dressing group having a network of PCL fibers on the surface and grow along the direction of the fibers, which in turn, effectively regulates cell behavior from the physical structure. Additionally, the large pore size (∼ 500 μm) of the network allowed the cells to penetrate the pores and enter the surface of the hydrogel without being blocked out. Besides, the composite dressing had a notable effect on angiogenesis. Furthermore, antioxidation experiments confirmed that the DFO-loaded hydrogel exhibited antioxidant activity. Experimental animal models of diabetes showed that rats treated with the PCL-GelMA-DFO (PGD) hydrogel had faster ability of hemostasis, scab formation, and wound healing. In conclusion, the PGD hydrogel effectively promoted the repair of chronic wounds.

Antibacterial coaxial hydro-membranes accelerate diabetic wound healing by tuning surface immunomodulatory functions

Abstract:Diabetic foot ulcers, typical non-healing wounds, represent a severe clinical problem. Advanced glycation end-products (AGEs), which create a prolonged pro-inflammatory micro-environment in defective sites, can be responsible for refractoriness of these ulcers. Macrophages are polarized to the M2 phenotype to facilitate the transition from a pro-inflammatory microenvironment to an anti-inflammatory microenvironment, which has been demonstrated to be an effective way to accelerate diabetic wound closure. Herein, we developed coaxial hydro-membranes mimicking the extracellular matrix structure that are capable of anti-inflammatory and antibacterial functions for diabetic wound repair. These fibrous membranes maintain a moist microenvironment to support cell proliferation. Macrophages grow in an elongated shape on the surface of the fibrous membranes. The fibrous membranes effectively impaired macrophage AGE-induced M1 polarization and induced macrophage polarization towards the M2 phenotype. The effects of the fibrous membranes on the interactions between macrophages and repair cells under a diabetic condition were also investigated. Furthermore, in vivo results from a full-thickness diabetic wound model confirmed the potential of the coaxial hydro-membranes to accelerate wound healing. This study's results indicate that the developed bioactive anti-inflammatory and antibacterial wound dressing can affect AGE-induced macrophage activation and crosstalk between macrophages and fibroblasts for treating diabetic wounds.

Abstract:Diabetes is a chronic disease affects nearly 10% of the global population, the prolonged metabolic abnormalities and the complex microenvironment in diabetic wounds often leading to impaired healing. To address the typical characteristics of diabetic wounds, such as hyperglycemia, low pH, excessive reactive oxygen species (ROS) production, and chronic inflammation, this study designed a baicalin derivative dynamically cross-linked natural polysaccharide hydrogel (CFPB) and explored its potential to promote diabetic wound healing. The framework of CFPB utilized chitosan and modified with dextran to introduce aldehyde groups via mild grafting, and crosslinked with the modified active substance baicalin to effectively extend the drug delivery time. A series of characterizations demonstrated that CFPB hydrogel possesses excellent mechanical properties, self-healing ability, and responsive release capability. Subsequently, multi-level biological experiments validated the promoting effect of CFPB hydrogel on diabetic wound healing in vitro and in vivo. This study not only offers a potential candidate for the local therapy of diabetic wounds, but also provides new insights for the development of advanced wound dressings.

Abstract:In patients with diabetes, chronic hyperglycemia impairs immune function at wound sites, increasing susceptibility to infections, prolonging inflammation, and delaying healing. This study aimed to develop wound dressings that control bacterial infections and accelerate healing. Phloretin (PHL), which has antibacterial and anti-inflammatory properties, was encapsulated with γ-cyclodextrin (γ-CD) to form a PHL@CD complex with enhanced bioavailability. This complex was incorporated into nanofiber wound dressings composed of polycaprolactone and natural silk protein. The resulting dressings exhibited favorable physical and chemical properties, including nutrient transport and gas exchange, which are essential for wound healing. The nanofiber membranes exhibited antibacterial activity against Staphylococcus aureus (90.31 ± 4.41 % inhibition), with high antioxidant capacity (91.48 ± 0.33 % ABTS scavenging) and blood compatibility. The membranes also promoted cell viability. Importantly, the nanofiber dressings accelerated wound healing in a diabetic mouse model by reducing the duration of inflammation. The novel nanofiber wound dressing can significantly improve the treatment of diabetic wounds.
Asymmetric Wettable Composite Wound Dressing Prepared by Electrospinning with Bioinspired Micropatterning Enhances Diabetic Wound Healing

Abstract:Wound dressings with asymmetric wettability surfaces can effectively prevent bacterial colonization and tissue dehydration and have shown great potential for diabetic wound healing applications. However, the construction of a highly hydrophobic outer surface with high biocompatibility and permeability is still the major challenge in the preparation of asymmetric wettable dressings. Inspired by the superhydrophobic surface structures existing in nature, an asymmetric wettable composite wound dressing with a highly hydrophobic outer layer was successfully prepared for diabetic wound healing in this study. The hydrophobic outer layer was fabricated by the electrospinning of poly(ε-caprolactone) (PCL) on a micron-pore-size nylon mesh template, and the hydrophilic inner layer was obtained by the electrospinning of pioglitazone-incorporated gelatin (Gel-pio). The hydrophobic outer layer of the dressing with a hierarchical micro–nanostructure exhibits excellent ability to waterproof and prevent bacterial adhesion, whereas the hydrophilic inner layer can promote cell proliferation, migration, and angiogenesis by its nanofiber structure and biocompatible gelatin composition. The presented dressing has good mechanical properties, permeability, and high biocompatibility. More importantly, the results of full-thickness skin wound model evaluation on db/db mice (type 2 diabetes) and STZ rats (type 1 diabetes) indicate that the developed dressing can promote wound healing by stimulating cell proliferation, angiogenesis, collagen deposition, and re-epithelialization. Abstract:Owing to hyperglycemia-related metabolic abnormalities that cause oxidative stress, recurrent infections, and vascular lesions, the therapeutic management of diabetic wounds is still unsatisfactory. Herein, we present a hydrogel (GCM) wound dressing loaded with melanin nanoparticles in a polysaccharide matrix (biguanide chitosan and oxidized β-glucan) for efficient healing of bacterially infected diabetic wounds. By a simple local injection, a protective GCM hydrogel barrier is established over the wound to rapidly stop bleeding and neutralize inflammatory cytokines. Subsequently, the GCM hydrogel achieves an accelerated transfer from the inflammatory process to the healing phase of the wound because of its excellent photothermal antimicrobial and reactive oxygen species scavenging properties. Additionally, GCM hydrogel has a three-dimensional network structure that dramatically enhances cell proliferation, migration, and angiogenesis by creating favorable ecological conditions for cell survival. Notably, our GCM hydrogel can help treat a variety of bacteria-infected diabetic wounds like skin and oral ulcer wounds. Collectively, this work provides a new option for diabetic wound treatment.

Abstract:Highlights • Spinnable bioactive glass (BG) particles were prepared and blended along with PLGA/Gel to afford nanofibrous membranes. • The BG particles afforded the sustained release of therapeutic ions as well as improved the anti-oxidative and anti-bacterial properties of membranes. • The BG containing membranes effectively accelerated hemostasis in multiple injury models. • The study exhibited the promising skin regenerative capability established through hemostasis, antibacterial, anti-inflammatory, and antioxidative properties. • The BG also accelerated wound healing in vivo , improved neovascularization, and suppressed inflammation. Excessive scar tissue formation along with bacterial infection, hemorrhage, and oxidative wound microenvironment pose adverse physiological and psychological effects on patients, which necessitate the advent of innovative anti-inflammatory, anti-bacterial, and anti-oxidative multifunctional wound dressings. The overarching objective of this study was to exploit bioactive glass (BG) and a natural anti-bacterial component namely "oregano essential oil (OEO)" to impart multifunctionality to poly(L-lactide-co-glycolide)/Gelatin (PLGA/Gel)-based nanofibrous dressings for excisional wound management. We performed a series of structural, morphological, and release studies as well as delineated angiogenic, hemostatic, anti-bacterial, and anti-oxidative properties of these bioactive dressings in vitro , which altogether revealed the beneficial effects of BG and OEO in terms of rapid hemostasis, improved chemotactic response, diminished bacterial colonization, and anti-inflammatory response. Impressively, in multiple injury models, including a rat tail-amputation model, an ear artery injury model, and a liver trauma model in rabbit in vivo , we reported BG-mediated rapid hemostasis. Moreover, dressings containing BG showed improved hemocompatibility and suppressed coagulation as revealed by activated partial thromboplastin assay (APTT) in vitro . In addition, the transplantation of these nanofibrous dressings in a full-thickness excisional wound model in rats showed significant tissue regeneration as evidenced by the more number of blood vessels, glands, and hair follicles, re-epithelialization, diminished inflammatory response, and less fibrotic tissue formation. Taken together our approach of simultaneously harnessing economical BG and OEO to enable multifunctionality to nanofibrous dressings for tissue repair may hold great promise for wound healing as well as the other bio-related disciplines. Abstract:The proficient handling of diabetic wounds, a rising issue coinciding with the global escalation of diabetes cases, poses significant clinical difficulties. A range of biofunctional dressings have been engineered and produced to expedite the healing process of diabetic wounds. This study proposes a multifunctional hydrogel dressing for diabetic wound healing, which is composed of Polyvinyl Alcohol (PVA) and N1-(4-boronobenzyl)-N3-(4-boronophenyl)-N1, N1, N3, N3-teramethylpropane-1, 3-diaminium (TSPBA), and a dual-drug loaded Gelatin methacryloyl (GM) microgel. The GM microgel is loaded with sodium fusidate (SF) and nanoliposomes (LP) that contain metformin hydrochloride (MH). Notably, adhesive and self-healing properties the hydrogel enhance their therapeutic potential and ease of application. In vitro assessments indicate that SF-infused hydrogel can eliminate more than 98% of bacteria within 24 h and maintain a sustained release over 15 days. Additionally, MH incorporated within the hydrogel has demonstrated effective glucose level regulation for a duration exceeding 15 days. The hydrogel demonstrates a sustained ability to neutralize ROS throughout the entire healing process, predominantly by electron donation and sequestration. This multifunctional hydrogel dressing, which integrated biological functions of efficient bactericidal activity against both MSSA and MRSA strains, blood glucose modulation, and control of active oxygen levels, has successfully promoted the healing of diabetic wounds in rats in 14 days. The hydrogel dressing exhibited significant effectiveness in facilitating the healing process of diabetic wounds, highlighting its considerable promise for clinical translation.

The management of diabetic wounds remains a critical therapeutic challenge. Platelet-rich plasma (PRP) gel, PRP-derived exosomes (PRP-Exos), and mesenchymal stem cell-derived exosomes (MSC-Exos) have demonstrated therapeutic potential in wound treatment. Unfortunately, their poor mechanical properties, the short half-lives of growth factors (GFs), and the burst release of GFs and exosomes have limited their clinical applications. Furthermore, proteases in diabetic wounds degrade GFs, which hampers wound repair. Silk fibroin is an enzyme-immobilization biomaterial that could protect GFs from proteases. Herein, we developed novel dual-crosslinked hydrogels based on silk protein (SP) (sericin and fibroin), including , , and , to promote diabetic wound healing synergistically. was prepared from PRP and SP using calcium gluconate/thrombin as agonist, while and were derived from exosomes and SP with genipin as crosslinker. SP provided improved mechanical properties and enabled the sustained release of GFs and exosomes, thereby overcoming the limitations of PRP and exosomes in wound healing. The dual-crosslinked hydrogels displayed shear-induced thinning, self-healing, and eradication of microbial biofilms in a bone-mimicking environment. In vivo , the dual-crosslinked hydrogels contributed to faster diabetic wound healing than PRP and SP by upregulating GFs expression, down-regulating matrix metalloproteinase-9 expression, and by promoting an anti-NETotic effect, angiogenesis, and re-epithelialization. Hence, these dual-crosslinked hydrogels have the potential to be translated into a new generation of diabetic wound dressings. Abstract:An injectable and microporous microgel‐fiber granular hydrogel (MFgel) loaded with bioglass and siRNA can promote diabetic wound healing through improving ingrowth of cell and tissue and regulating MMP‐9 and inflammation. Injectable hydrogels find extensive application in the treatment of diabetic wound healing. However, traditional bulk hydrogels are significantly limited due to their nano‐porous structure, which obstructs cell migration and tissue infiltration. Moreover, regulating inflammation and matrix metalloproteinase ‐9 (MMP‐9) expression in diabetic wounds is crucial for enhancing wound healing. This study marks the first instance of introducing an efficient, scalable, and simple method for producing microfiber‐gel granules encapsulating bioceramics powders. Utilizing this method, an injectable microporous granular microgel‐fiber hydrogel (MFgel) is successfully developed by assembling microgel‐fibers made from hyaluronic acid (HA) and sodium alginate (SA) loaded with small interfering RNA (siRNA) and bioglass (BG) particles. Compared to traditional hydrogels (Tgel), MFgel possesses a highly interconnected network with micron‐sized pores, demonstrating favorable properties for cell adhesion and penetration in in vitro experiments. Additionally, MFgel exhibits a higher compressive modulus and superior mechanical stability. When implanted subcutaneously in mice, MFgel promotes cellular and tissue infiltration, facilitating cell proliferation. Furthermore, when applied to skin defects in diabetic rats, MFgel not only effectively regulates inflammation and suppresses MMP‐9 expression but also enhances angiogenesis and collagen deposition, thereby significantly accelerating diabetic wound healing. Taken together, this hydrogel possesses great potential in diabetic wound healing applications.

Abstract:Diabetes-related slow healing of wounds is primarily driven by bacterial infections and angiogenesis disorder and presents a substantial hurdle in clinical treatment. To solve the above problems, an advanced multifunctional hydrogel system based on natural polymer was created here to facilitate wound healing in patients with chronic diabetes. The prepared dressing was composed of an outer hydrogel containing polyvinyl alcohol and hydroxypropyl methyl cellulose in dimethyl sulfoxide and water as binary solvents, and an inner hydrogel containing chitosan quaternary ammonium salt, flaxseed gum, and polyvinyl alcohol. Thus, a polysaccharide based bilayer hydrogel (BH) with superior mechanical strength and biocompatibility was created. This bilayer hydrogel could easily bind to dynamic tissue surfaces, thereby generating a protective barrier. Meanwhile, L-arginine-modified polyoxometalate (POM@L-Arg) nanoclusters were loaded in the inner hydrogel. They released NO when stimulated by the peroxide microenvironment of diabetic wounds. NO as a signal molecule regulated vascular tension and promoted cell proliferation and migration. Additionally, because of the synergistic effect of NO and the chitosan quaternary ammonium salt, the hydrogel system exhibited excellent antibacterial performance. The NO released reduced the levels of proinflammatory factors IL-6 and TNF-α in the diabetic wounds, which thus accelerated wound healing. In short, BH + POM@L-Arg is expected to serve as an ideal wound dressing as it exerts a good promotion effect on diabetes-related wound healing.
Immunoregulatory Hydrogel Decorated with Tannic acid/Ferric Ion Accelerates Diabetic Wound Healing via Regulating Macrophage Polarization

Abstract:Diabetic wound management still faces great challenges mainly due to immune disorders at wound area. Especially, macrophage polarization toward M2 phenotype is severely inhibited at the inflammatory stage. Thus, targeted regulation of macrophage polarization could be a useful approach to normalize the immune environment of diabetic wound to accelerate wound healing. Herein, a composited hydrogel derived from natural polymers, as an immunoregulatory dressing, was fabricated to treat diabetic wound. The composited hydrogel (GBTF) was synthesized by one step photo-polymerization of gelatin methacrylate (GelMA), Bletilla Striata polysaccharide (BSP) and tannic acid/ferric iron complex (TA/Fe3+). Attributed to the chemical and physical cross-linking, the GBTF hydrogel exhibited adequate mechanical properties. Thanks to the photothermal transition of TA/Fe3+ in GBTF, good antibacterial capacity was achieved. Meanwhile, the polyphenol in TA could help to attenuate excessive ROS and induce macrophage polarization toward M2 phenotype. Furthermore, the potential immunoregulatory mechanism of the GBTF hydrogel was investigated through RNA sequencing in vitro. In addition, BSP showed positive contribution in vascularization. In vivo, results from diabetic wound model showed that the wound treated by GBTF hydrogel presented the fastest closure rate, superior granulation tissue regeneration, re-epithelialization and collagen deposition. These results indicated GBTF hydrogel rapidly promoted wound healing through anti-inflammation and pro-angiogenesis. The GBTF hydrogel showed great potential to serve as promising dressing for treatment of diabetic wounds in clinic.

Abstract:Patients with diabetes suffer from a variety of complications and easily develop diabetic chronic wounds. The microenvironment of diabetic wounds is characterized by an excessive amount of reactive oxygen species (ROS) and an imbalance of proinflammatory and anti-inflammatory cells/factors, which hinder the regeneration of chronic wounds. In the present study, a wound dressing with immunomodulation and electroconductivity properties was prepared and assayed in vitro and in vivo. [2-(acryloyloxy) ethyl] Trimethylammonium chloride (Bio-IL) and gelatin methacrylate (GelMA) were 3D printed onto a doxycycline hydrochloride (DOXH)-loaded and ROS-degradable polyurethane (PFKU) nanofibrous membrane, followed by UV irradiation to obtain conductive hydrogel strips. DOXH was released more rapidly under a high ROS environment. The dressing promoted migration of endothelial cells and polarization of macrophages to the anti-inflammatory phenotype (M2) in vitro. In a diabetic rat wound healing test, the combination of conductivity and DOXH was most effective in accelerating wound healing, collagen deposition, revascularization, and re-epithelialization by downregulating ROS and inflammatory factor levels as well as by upregulating the M2 macrophage ratio. STATEMENT OF SIGNIFICANCE: The microenvironment of diabetic wounds is characterized by an excessive amount of reactive oxygen species (ROS) and an imbalance of proinflammatory and anti-inflammatory cells/factors, which hinder the regeneration of chronic wounds. Herein, a wound dressing composed of a DOXH-loaded ROS-responsive polyurethane membrane and 3D-printed conductive hydrogel strips was prepared, which effectively accelerated skin regeneration in diabetic wounds in vivo with better epithelialization, angiogenesis, and collagen deposition. DOXH regulated the dysfunctional wound microenvironment by ROS scavenging and polarizing macrophages to M2 phenotype, thereby playing a dominant role in diabetic wound regeneration. This design may have great potential for preparing other similar materials for the therapy of other diseases with excessive inflammation or damage to electrophysiological organs, such as nerve defect and myocardial infarction.
Multifunctional Injectable Hydrogel Loaded with Cerium-Containing Bioactive Glass Nanoparticles for Diabetic Wound Healing

Abstract:Diabetic foot wound healing is a major clinical problem due to impaired angiogenesis and bacterial infection. Therefore, an effective regenerative dressing is desiderated with the function of promoting revascularization and anti-bacteria. Herein, a multifunctional injectable composite hydrogel was prepared by incorporation of the cerium-containing bioactive glass (Ce-BG) into Gelatin methacryloyl (GelMA) hydrogel. The Ce-BG was synthesized by combining sol-gel method with template method, which maintained spherical shape, chemical structure and phase constitution of bioactive glass (BG). The Ce-BG/GelMA hydrogels had good cytocompatibility, promoted endothelial cells migration and tube formation by releasing Si ion. In vitro antibacterial tests showed that 5 mol % CeO2-containing bioactive glass/GelMA (5/G) composite hydrogel exhibited excellent antibacterial properties. In vivo study demonstrated that the 5/G hydrogel could significantly improve wound healing in diabetic rats by accelerating the formation of granulation tissue, collagen deposition and angiogenesis. All in all, these results indicate that the 5/G hydrogel could enhance diabetic wound healing. Therefore, the development of multifunctional materials with antibacterial and angiogenic functions is of great significance to promote the repair of diabetic wound healing.

Astragaloside/PVP/PLA nanofiber functional dressing prepared by coaxial electrostatic spinning technology for promoting diabetic wound healing

Abstract:Diabetic wound is often a chronic wound with complex healing process and long healing time. It is evident that conventional dressings are inadequate in promoting the healing of diabetic wounds. In this study, a novel nanofiber dressing, produced through coaxial electrostatic spinning technology, was created to facilitate the healing of diabetic chronic wounds. This dressing was composed of Astragaloside (AS), polyvinylpyrrolidone (PVP) and polylactic acid (PLA). The morphology of the nanofiber dressing was characterized by scanning electron microscopy (SEM). Confirmation of the core–shell structure was done with the help of transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD). A pharmacodynamic study was conducted both in vivo and in vitro. It was shown that the nanofibers were approximately 604.83 ± 127.96 nm and with a core–shell structure. Additionally, in vitro drug release experiments revealed that the coaxial AS/PVP/PLA nanofiber functional dressings had a first rapid and then slow release property. Antioxidant activity experiment showed that the free radical scavenging rate of Astragaloside-loaded nanofibers reached 61 %. Mechanical tests also indicated that the nanofibers had an elastic modulus of 38.37 ± 1.81 MPa. Coaxial AS/PVP/PLA nanofiber functional dressings had good antimicrobial activity, and its diameter of the circle of inhibition against Staphylococcus aureus was 17.17 ± 2.04 mm, and its diameter of the circle of inhibition against Escherichia coli was 6.80 ± 0.78 mm. In addition, the findings of in vitro cellular tests showed that coaxial AS/PVP/PLA nanofiber functional dressings could promote the multiplication, adhesion and the migration ability of L929 fibroblasts with better biocompatibility. Finally, the wound healing rate of diabetic rats in the coaxial AS/PVP/PLA nanofiber functional dressing group was 96.54 ± 0.29 % on day 14. Consequently, the coaxial AS/PVP/PLA nanofiber functional dressings prepared by coaxial electrostatic spinning technique had satisfactory morphology, met the requirements of performance, and had the potential to be developed as a dressing for improving diabetic chronic wound healing.

Abstract:Diabetic chronic wounds are difficult to heal because of the presence of excessive inflammation and high overexpression of matrix metalloproteinase-9 (MMP-9), which greatly affects the quality of life of patients with diabetes and increases the risk of death. Thus, the regulation of excessive inflammation and inhibition of MMP-9 overexpression are effective strategies to improve diabetic wound healing. The present study is the first to demonstrate that ion products of 45S5 Bioglass® (BG) can work with small interfering RNA of MMP9 (MMP9-siRNA) to reduce MMP-9 expression in tissue-forming cells and enhance the synthesis of extracellular matrix proteins (ECMs). Specifically, the BG ionic products can stimulate macrophages to convert to M2 phenotype, thereby creating a proregenerative inflammation microenvironment to indirectly suppress the expression of MMP-9 in tissue-forming cells. Chitosan nanoparticles encapsulating MMP9-siRNA (MMP9-siNP) can directly lower MMP-9 expression in tissue-forming cells. In addition, BG ionic products can promote the vascularization of endothelial cells and ECM protein synthesis by fibroblasts. Thus, injectable BG/sodium alginate (BG/SA) hydrogels loaded with MMP9-siNP can significantly accelerate the healing process of full-thickness excision wounds of diabetic rats by decreasing MMP-9 expression, improving collagen synthesis, and enhancing angiogenesis in the wounds, thereby demonstrating their great application potential in treating diabetic chronic wounds.Statement of SignificanceExcessive inflammation and high overexpression of MMP-9 have been considered as factors that severely hinder the healing process of diabetic chronic wounds. Effective strategies are required for the regulation of excessive inflammation and inhibition of MMP-9 overexpression to enhance diabetic wound healing. In the present work, an injectable bioglass/sodium alginate (BG/SA) hydrogel loaded with MMP9-siNP was developed; this hydrogel significantly accelerated the healing process of full-thickness excision wounds of diabetic rats by decreasing MMP-9 expression, improving collagen accumulation, and enhancing angiogenesis in the wounds. Thus, the BG/SA hydrogel loaded with MMP9-siNP has great potential for use in healing of diabetic chronic wounds.

Abstract:Background: Diabetic foot ulcer (DFU) is a major debilitating complication of diabetes. The lack of effective diabetic wound dressings has been a significant problem in DFU management. In this study, we aim to establish a phlorotannin-incorporated nanofibre system and determine its potential in accelerating hyperglycaemic wound healing. Methods: The effective dose of Ecklonia cava phlorotannins (ECP) for hyperglycaemic wound healing was determined prior to phlorotannin nanofibre fabrication using polyvinyl-alcohol (PVA), polyvinylpyrrolidone (PVP), and ECP. Vapour glutaraldehyde was used for crosslinking of the PVA/PVP nanofibres. The phlorotannin nanofibres were characterised, and their safety and cytocompatibility were validated. Next, the wound healing effect of phlorotannin nanofibres was determined with 2D wound scratch assay, whereas immunofluorescence staining of Collagen-I (Col-I) and Cytokeratin-14 (CK-14) was performed in human dermal fibroblasts (HDF) and human epidermal keratinocytes (HEK), respectively. Results: Our results demonstrated that 0.01 μg/mL ECP significantly improved hyperglycaemic wound healing without compromising cell viability and proliferation. Among all nanofibres, PVA/PVP/0.01 wt% ECP nanofibres exhibited the best hyperglycaemic wound healing effect. They displayed a diameter of 334.7 ± 10.1 nm, a porosity of 40.7 ± 3.3%, and a WVTR of 1718.1 ± 32.3 g/m2/day. Besides, the FTIR spectra and phlorotannin release profile validated the successful vapour glutaraldehyde crosslinking and ECP incorporation. We also demonstrated the potential of phlorotannin nanofibres as a non-cytotoxic wound dressing as they support the viability and proliferation of both HDF and HEK. Furthermore, phlorotannin nanofibres significantly ameliorated the impaired hyperglycaemic wound healing and restored the hyperglycaemic-induced Col-I reduction in HDF. Conclusion: Taken together, our findings show that phlorotannin nanofibres have the potential to be used as a diabetic wound dressing.

Abstract:As the incidence of chronic wounds increases, the requirements for wound dressings are rising. The specific aim of this study is to propose a novel gallic acid (GA) functionalized silk fibroin (SF) and gelatin (Gel) composite wound dressing in which GA is used as an antibacterial and wound healing substance. Via electrospinning, SF, Gel, and GA mixed solutions could be conveniently fabricated into a composite nanofiber mat (SF-Gel-GA), consisting of uniform fibers with an average diameter around 134.57 ± 84 nm. The internal mesh structure of SF-Gel-GA provides sufficient drug loading capacity, proper moisture permeability, and proper degradation rate. SF-Gel-GA presents excellent biocompatibility. NIH-3T3 fibroblast cells could adhere and spread stably on the SF-Gel-GA surface with slightly promoted proliferation. In the presence of SF-Gel-GA, the growth of both Gram-positive and Gram-negative bacteria, includingStaphylococcus aureusandPseudomonas aeruginosa, is significantly inhibited in both plate and suspension cultures. A cutaneous excisional mouse wound model proves the efficient ability of SF-Gel-GA to promote wound healing. Compared with pure SF dressing and commercial Tegaderm Hydrocolloid3Mdressing, the wound closure rate with SF-Gel-GA treatment is significantly improved. The histological assessments further demonstrate SF-Gel-GA could facilitate collagen deposition, neovascularization, and epithelialization at wound sites to promote wound healing. In conclusion, a novel SF-Gel-GA composite wound dressing with efficient wound healing activities have been developed for chronic wound treatment with broad healing potential.

Abstract:Bacterial infection and excessive reactive oxygen species (ROS) in diabetic wounds lead to a prolonged inflammatory phase, and injuries are highly susceptible to developing into chronic wounds. Improving the poor microenvironment is vital to achieving effective diabetic wound healing. In this work, methacrylated silk fibroin (SFMA) was combined with ε-polylysine (EPL) and manganese dioxide nanoparticles (BMNPs) to form an (EPL-BM) hydrogel with in situ forming, antibacterial and antioxidant properties. EPL imparted high antibacterial activity (>96 %) to the hydrogel. BMNPs and EPL showed good scavenging activity against a variety of free radicals. (EPL-BM) hydrogel had low cytotoxicity and could alleviate H 2 O 2 -induced oxidative stress in L929 cells. In diabetic wounds infected with Staphylococcus aureus ( S. aureus ), the (EPL-BM) hydrogel exhibited better antibacterial properties and reduced wound ROS levels more significantly than that of the control in vivo. In this process, the pro-inflammatory factor TNF-α was down-regulated, and the vascularization marker CD31 was up-regulated. H&E and Masson staining showed a rapid transition from the inflammatory to the proliferative phase of the wounds, with significant new tissue and collagen deposition. These results confirm that this multifunctional hydrogel dressing holds well potential for chronic wound healing.
