Abstract:In this paper, a comprehensive theoretical framework for understanding surface-enhanced Raman scattering (SERS) measurements in both solution and thin-film setups, focusing on electromagnetic enhancement principles, was presented. Two prevalent types of SERS substrates found in the literature were investigated: plasmonic colloidal particles, including spherical and spheroid nanoparticles, nanoparticle diameters, and thin-film-based SERS substrates, like ultra-thin substrates, bundled nanorods, plasmonic thin films, and porous thin films. The investigation explored the impact of analyte adsorption, orientation, and the polarization of the excitation laser on effective SERS enhancement factors. Notably, it considered the impact of analyte size on the SERS spectrum by examining scenarios where the analyte was significantly smaller or larger than the hot spot dimensions. The analysis also incorporated optical attenuations arising from the optical properties of the analyte and the SERS substrates. The findings provide possible explanations for many observations made in SERS measurements, such as variations in relative peak intensities during SERS assessments, reductions in SERS intensity at high analyte concentrations, and the occurrence of significant baseline fluctuations. This study offers valuable guidance for optimizing SERS substrate design, enhancing SERS measurements, and improving the quantification of SERS detection.

Abstract:Surface-enhanced Raman spectroscopy (SERS) is a powerful optical sensing technique used in various applications, including medicine, microbiology, and environmental analysis. Planar SERS substrates are of particular interest due to their ease of integration in lab-on-chips and better reproducibility compared to colloidal SERS. The performance of these SERS substrates is quantified using metrics such as enhancement factor, sensitivity, and reproducibility. However, there is yet to be a consensus on how to practically compare and interpret such metrics in publications and experiments. These performance metrics are strongly influenced by the nanostructures' material, architecture, element sizes, as well as the circumstances surrounding the experiments. Understanding the effect of these characteristics on the SERS substrates' performance could not only enable a better performance but also direct their development for different applications. Thus, we developed a planar SERS-substrate characterization protocol to explore the correlation between the nanostructures' physical characteristics and the performance metrics through coordinate-transformed spectroscopic measurements over structure-characterized areas. Seven commercial SERS substrates, with various surface architectures fabricated using different fabrication technologies, were studied using this benchmarking protocol. The results demonstrated how this protocol can indicate a SERS substrate's suitability for a specific application, thus, guiding the substrate's further adaptations or development.
High-Performance SERS Substrate Based on Gold Nanoparticles-Decorated Micro/Nano-Hybrid Hierarchical Structure

Abstract:Owing to its excellent localized surface plasmon resonance (LSPR) effect, noble metal nanoparticles (NPs) find extensive application in the preparation of surface-enhanced Raman scattering (SERS) substrates. However, due to process limitations, the practicality and testing effectiveness of SERS substrates still leaves much to be desired. Here, a wafer-scale gold (Au) NPs silicon (Si) micro/nano-hybrid structured substrate is prepared. This is achieved through two-step etching, followed by decorating Au-NPs onto the structure via self-assembly process induced by de-wetting. Finite-difference time-domain (FDTD) simulations reveal that the significant enhancement of local electric field in the voids between Au-NPs and Si is crucial for enhancing SERS. Using Rhodamine 6G (R6G) as the probe molecule, performance of the fabricated SERS substrate is investigated. It demonstrates a minimum detection limit of 10 −11 M, with a calculated enhancement factor of 4.40 × 10 8 , indicating its high sensitivity. The minimum relative standard deviation for the substrate is 5.254 %. After 20 days of placement, the SERS performance show tiny variation. Even after four cycles of cleaning experiments, it still maintains outstanding SERS performance. This demonstrates its excellent stability, uniformity and reusability. Our research provides guidance for the efficient and low-cost fabrication of high-performance SERS substrates.

Abstract:As a powerful spectroscopy technique, surface-enhanced Raman scattering (SERS) can provide non-destructive and sensitive characterization down to a single molecular level. Aiming to the main challenges of high-performance SERS-active substrates for their real-world applications involving the ultra-sensitive and reproducible signals detection and signal uniformity with large-area, herein, a facile and reliable strategy based on combination of thermal imprinting polycarbonate (PC) film with porous anodic aluminum oxide (AAO) mold and E-beam evaporation of gold is provided to fabricate a high-quality SERS-active substrate consisting of ultra-dense hot-spots with large-area uniformity. Two kinds of sub-10 nm gaps were obtained, including the nanogaps between the neighboring gold coated PC-nanopillars and those between gold on the top of the nanopillars and that on the base, which actually build up a three-dimensional (3D) hot-spot network for high-performance SERS detection. The effect of structural parameters on SERS enhancement was investigated numerically and experimentally, and by optimizing the structural parameters, a remarkable average SERS enhancement factor up to of 1.4×108 is achieved and it shows an excellent reproducibility with a relative standard deviation of 18%, which allows for enhanced practicability in the application of quantitative biochemical detection.

Abstract:Wih a view to improve the surface-enhanced Raman scattering (SERS) properties, including the sensitivity and uniformity, a simple and practical pretreatment method via optimizing the adsorption process of probe molecules and the morphology of SERS-active nanostructures was studied. Excellent SERS performances were obtained using this method such as high sensitivity (5 x 10(-14) M rhodamine 6G, 1 x 10(-8) M melamine), remarkable uniformity (similar to 9% relative standard deviation (RSD)) and reproducibility (similar to 10% RSD). Uniform and repeatable silver nanoparticle arrays were fabricated by an optimized filter-based method, which guaranteed the reliability of SERS detection. The SERS intensity was greatly improved by adjusting the adsorption capacity of the noble metal nanostructures, which has a linear relationship with the pH value of silver solution below 10. An accurate and reliable enhancement factor (EF) of 3.28 x 10(8) was calculated through the quantification of the adsorbed probe molecules. It is worth noting that the lifetime of the substrates prepared by this method could be prolonged to two months, which was definitely longer than other ordinary silver SERS substrates. The pretreatment method not only provided perfect SERS properties, but also made SERS a practical analysis tool because of its good homogeneity and ultralow detection limit.

Abstract:Colloidal plasmonic metal nanoparticles are capable of surface-enhanced Raman scattering (SERS) for various analytical fields. Despite steady efforts to establish hot spots and obtain optimal SERS signals, it remains a challenge to ensure the accessibility and high density for enhancing Raman activity. Here, pyramid silicon was utilized as a template to fabricate the graphene oxide (GO)/silver nanoparticles (AgNPs)/pyramid polymethyl methacrylate (PMMA) three-dimension (3D) flexible structure. Micrographs displayed the hybrid structure inherited the periodic 3D nanostructure with the pyramid microstructure, and the results exhibited that the proposed substrate possesses superior SERS performance with high sensitivity, homogeneity and stability. The enhancement factor reached up to 8.1 × 109 using rhodamine 6G as the probe molecules. Besides, the in situ detection of the malachite green (MG) on the shrimp’s surface was carried out to achieve and detect surface analytes. This work can provide a novel assembled SERS substrate as SERS-based biological detection in practical applications.

Abstract:Experimental results obtained in different laboratories world‐wide by researchers using surface‐enhanced Raman scattering (SERS) can differ significantly. We, an international team of scientists with long‐standing expertise in SERS, address this issue from our perspective by presenting considerations on reliable and quantitative SERS. The central idea of this joint effort is to highlight key parameters and pitfalls that are often encountered in the literature. To that end, we provide here a series of recommendations on: a) the characterization of solid and colloidal SERS substrates by correlative electron and optical microscopy and spectroscopy, b) on the determination of the SERS enhancement factor (EF), including suitable Raman reporter/probe molecules, and finally on c) good analytical practice. The authors hope that both newcomers and specialists will benefit from these recommendations in order to increase the inter‐laboratory comparability of experimental SERS results and further establish SERS as an analytical tool.

Abstract:Robust quantitative analysis methods are very attractive but challenging with surface-enhanced Raman scattering (SERS) technique till now. Quantitative analysis methods using absolute Raman scattering intensities tend to desire very critical reproducibility of SERS substrates and consistency of testing conditions, as batch differences and inhomogeneity of SERS substrates as well as the fluctuation of measuring parameters placed challenging obstacles. Relative Raman scattering intensities, on the other hand, can release the adverse interferences mentioned above and provide effective and robust information as it is independent of the reproducibility of SERS substrates. By establishing external calibration working curves, we achieved accurate molecule composition prediction of molecules in multi-component systems. Further, by choosing or adding a label molecule with known concentration as Raman internal standards, the concentration of target molecules can be easily predicted. This approach proved the effectiveness and robustness of quantitative analysis with the relative Raman scattering intensities, even carried out with a flexible inhomogeneous SERS substrate.
Design of Novel 3D SERS Probes with Drastically Improved Detection Limit by Maximizing SPP - Based Multiple Coupling Effects

Abstract:Quantifying formidable multiple coupling effects involved in Surface-enhanced Raman scattering (SERS) is a prerequisite for accurate design of SERS probes with superior detection limit and uniformity which are the targets for trace substance detection. Here, combining theory and experiments on novel 3D periodic Au/SiO2 hybrid nanogrids, we successfully develop a generalized methodology of accurately designing high performance SERS probes. Structural parameters and symmetry, Au roughness, and polarization are quantitatively correlated to intrinsic electromagnetic field (EMF) enhancements from surface plasmon polariton (SPP), localized surface plasmon resonance (LSPR), optical standing wave and their couplings theoretically, which is experimentally verified. The hexagonal SERS probes optimized by the methodology successfully detect 5*10^-11 M Hg ions in water, and 2.5*10^-11 M R6G with 40 times improvement of detection limit, an enhancement factor of 3.4*10^8 and uniformity of 5.56%, which results from the extra Au roughness - independent 144% contribution of LSPR effects excited by SPP interference waves as secondary sources, beyond the conventional recognization. This study opens up a pioneering way not only for providing the generalized design principles of SERS probe structures with high performance but for accurately designing their structures with particular purposes such as greatly improved detection limit and uniformity which are very significant for trace substance detection.

Abstract:We introduce a simple and cost-effective approach for fabrication of effective surface-enhanced Raman spectroscopy (SERS) substrates. It is shown that the as-fabricated substrates show excellent SERS effects in various probe molecules with high sensitivity, that is, picomolar level detection, and also good reliability. With a SERS enhancement factor beyond 108 and excellent reproducibility (deviation less than 5%) of signal intensity, the fabrication of the SERS substrate is realized on a four-inch wafer and proven to be effective in pesticide residue detection. The SERS substrate is realized first through the fabrication of quasi-periodic nanostructured silicon with dimension features in tens of nanometers using superaligned carbon nanotubes networks as an etching mask, after which a large amount of hot spots with nanometer gaps are formed through deposition of a gold film. With rigorous nanostructure design, the enhanced performance of electromagnetic field distribution for nanostructures is optimized. With the advantage of cost-effective large-area preparation, it is believed that the as-fabricated SERS substrate could be used in a wide variety of actual applications where detection of trace amounts is necessary.

Abstract:usually much lower than other, synchronized readings from the same scalp (Fig. 1 ) . The ability to achieve a reliable, synchronized reading was inversely related to the degree of caput present; in many instances, placing the probe just next to caput allowed synchronization and the readings became accurate. We suggest that the findings reported by Johnson et al. demonstrate the need to exclude artefact. Pulse oximetry is meant to reflect arterial saturation lcvels, but over caput a regular plcthysmograph may also reflect pulsations that are venous or transmitted through oedematous tissue. Synchronizing the readings to the truc heart rate from the ECG may avoid this problem. The clinical relevance of oximetry over caput is limited, as the latter usually develops after there is sufficient cervical dilatation to be able to avoid placing the probe onto it. If caput develops later undcrncath the probe, our system will show a loss of signal synchronization and the oxygen saturation rcadings are rejected as unreliable, indicating a need to reposition the probe. Highly effective and uniform SERS substrates fabricated by etching multi-layered gold nanoparticle arrays

Abstract:Gold nanoparticle multilayers printed on silicon substrates layer by layer were etched by a gold etchant to form highly effective and uniform substrates for surface-enhanced Raman scattering (SERS). The performance of the SERS substrates was systematically studied by adjusting the number of nanoparticle layers and the etching time. The optimized enhancement factor (EF) and the detection limit of the substrates were determined to be 8.6 x 10(6) and 1 x 10(-12) M, respectively. The high EF and low detection limit were attributed to the high density of "hot-spots" and the facile accession of probe molecules to these spots. Moreover, the SERS substrates exhibited a nice uniformity with a small spot-to-spot variation and a good sample-to-sample reproducibility as well. The experimental results were supported by finite-difference time domain (FDTD) simulations. Our study suggests that low-cost, large-scale, and uniform SERS substrates with a high EF and low detection limit can be achieved by using bottom-up chemical methods.

Abstract:Surface-enhanced Raman spectroscopy (SERS) with enormous enhancements has shown great potential in ultrasensitive detection technologies, but the fabrication of large-scale, controllable and reproducible substrates with high SERS activity is a major challenge. Here, we report the preparation of Au nanoparticle arrays for SERS-active substrates with tunable particle sizes and interparticle gaps, and the enhancement factor of the SERS signal obtained from 4-mercaptopyridine probe molecules was as high as 10(7). The experimental data points show the increase of enhancement factor as a function of the ratio of diameter to interparticle gap, which can be explained by the averaged electromagnetic field enhancement model. Furthermore, we demonstrated that this type of substrate merits its high uniformity, high reproducibility and excellent long-term stability. As the fabrication protocol of such a SERS substrate is simple and inexpensive, this substrate may anticipate a wide range of applications in SERS-based sensors.
Ag/ZIF-8 Substrate with Enhanced SERS via the Plasmonic Nanogap and MOF-Enabled Molecular Preconcentration Effect

Abstract:Surface-enhanced Raman scattering (SERS) is an enthralling phenomenon that significantly boosts the Raman signals of molecules near “hot spots”. Its sensitivity primarily relies on the design of “hot spots” and concentrating the analytes into these regions. Herein, a high-performance Ag/ZIF-8 SERS substrate is fabricated with a facile low-cost two-step method. After randomly dispersing ZIF-8 nanocrystals on the substrate, a Ag layer is sprayed. Plasmonic nanogaps as small as 10 nm are created between the Ag nanocap on the top of the ZIF-8 and the Ag layer on the substrate around the ZIF-8 crystal by optimizing the thickness of the Ag layer. Because of plasmonic coupling effects, the near-field interactions at the plasmonic nanogaps generate a greatly enhanced electromagnetic (EM) field, which is also interpreted by numerical simulations. Furthermore, the porous ZIF-8 nanoparticles’ preconcentration effect can enrich analytes in the nanogap. The measurement results show that this SERS substrate has a SERS enhancement factor (EF) of up to 2.84 × 107, the lowest detectable concentration for 4-ATP of 10–9 M, and a relative standard deviation (RSD) of only about 6.99%. This work may provide a facile strategy for the construction of metal–organic framework (MOF)-based nanostructure substrates as a potential SERS platform for molecular detection.

Abstract:Surface-enhanced Raman scattering (SERS) enhancement and the reproducibility of the SERS signal strongly reflect the quality and nature of the SERS substrates because of diverse localized surface plasmon resonance (LSPR) excitations excited at interstitials or sharp edges. LSPR excitations are the most important ingredients for achieving huge enhancements in the SERS process. In this report, we introduce several gold and silver nanoparticle-based SERS-active substrates developed solely by us and use these substrates to investigate the influence of LSPR excitations on SERS. SERS-active gold substrates were fabricated by immobilizing colloidal gold nanoparticles on glass slides without using any surfactants or electrolytes, whereas most of the SERS-active substrates that use colloidal gold/silver nanoparticles are not free of surfactant. Isolated aggregates, chain-like elongated aggregates and two-dimensional (2D) nanostructures were found to consist mostly of monolayers rather than agglomerations. With reference to correlated LSPR and SERS, combined experiments were carried out on a single platform at the same spatial position. The isolated aggregates mostly show a broadened and shifted SPR peak, whereas a weak blue-shifted peak is observed near 430 nm in addition to broadened peaks centered at 635 and 720 nm in the red spectral region in the chain-like elongated aggregates. In the case of 2D nanostructures, several SPR peaks are observed in diverse frequency regions. The characteristics of LSPR and SERS for the same gold nanoaggregates lead to a good correlation between SPR and SERS images. The elongated gold nanostructures show a higher enhancement of the Raman signal than the the isolated and 2D samples. In the case of SERS-active silver substrates for protein detection, a new approach has been adopted, in contrast to the conventional fabrication method. Colloidal silver nanoparticles are immobilized on the protein functionalized glass slides, and further SERS measurements are carried out based on LSPR excitations. A new strategy for the detection of biomolecules, particularly glutathione, under aqueous conditions is proposed. Finally, supramolecular J-aggregates of ionic dyes incorporated with silver colloidal aggregates are characterized by SERS measurements and correlated to finite-difference time-domain analysis with reference to LSPR excitations.FigureSPR and SERS images for isolated, elongated and two-dimensional gold nanostructures

Abstract:A surface plasmon resonance (SPR)-surface enhanced Raman scattering (SERS) microspectrometer was designed to obtain the incident angle dependence of SERS signals excited by the evanescent field. By simultaneous measurement of the SERS and SPR spectra of analytes, the highest SERS signal intensities were found to appear at the vicinity of the resonance angle. The enhancement factor was about 2.0x10(6). The simulated angle of the maximal SERS intensity based on Fresnel equation was found to be in good agreement with the experimental results. The SERS and SPR spectra captured simultaneously not only directly confirm the correlation between the SERS and SPR but also present a potential technique for obtaining the structure information about the analytes in molecule level with recording their SPR curves.

Abstract:Surface-enhanced Raman scattering (SERS) provides significantly enhanced Raman scattering signals from molecules adsorbed on plasmonic nanostructures, as well as the molecules' vibrational fingerprints. Plasmonic nanoparticle systems are particularly powerful for SERS substrates as they provide a wide range of structural features and plasmonic couplings to boost the enhancement, often up to >108-1010. Nevertheless, nanoparticle-based SERS is not widely utilized as a means for reliable quantitative measurement of molecules largely due to limited controllability, uniformity, and scalability of plasmonic nanoparticles, poor molecular modification chemistry, and a lack of widely used analytical protocols for SERS. Furthermore, multiscale issues with plasmonic nanoparticle systems that range from atomic and molecular scales to assembled nanostructure scale are difficult to simultaneously control, analyze, and address. In this perspective, we introduce and discuss the design principles and key issues in preparing SERS nanoparticle substrates and the recent studies on the uniform and controllable synthesis and newly emerging machine learning-based analysis of plasmonic nanoparticle systems for quantitative SERS. Specifically, the multiscale point of view with plasmonic nanoparticle systems toward quantitative SERS is provided throughout this perspective. Furthermore, issues with correctly estimating and comparing SERS enhancement factors are discussed, and newly emerging statistical and artificial intelligence approaches for analyzing complex SERS systems are introduced and scrutinized to address challenges that cannot be fully resolved through synthetic improvements.

Abstract:Surface-enhanced Raman spectroscopy (SERS) and related spectroscopies are powered primarily by the concentration of the electromagnetic (EM) fields associated with light in or near appropriately nanostructured electrically-conducting materials, most prominently, but not exclusively high-conductivity metals such as silver and gold. This field concentration takes place on account of the excitation of surface-plasmon (SP) resonances in the nanostructured conductor. Optimizing nanostructures for SERS, therefore, implies optimizing the ability of plasmonic nanostructures to concentrate EM optical fields at locations where molecules of interest reside, and to enhance the radiation efficiency of the oscillating dipoles associated with these molecules and nanostructures. This review summarizes the development of theories over the past four decades pertinent to SERS, especially those contributing to our current understanding of SP-related SERS. Special emphasis is given to the salient strategies and theoretical approaches for optimizing nanostructures with hotspots as efficient EM near-field concentrating and far-field radiating substrates for SERS. A simple model is described in terms of which the upper limit of the SERS enhancement can be estimated. Several experimental strategies that may allow one to approach, or possibly exceed this limit, such as cascading the enhancement of the local and radiated EM field by the multiscale EM coupling of hierarchical structures, and generating hotspots by hybridizing an antenna mode with a plasmonic waveguide cavity mode, which would result in an increased local field enhancement, are discussed. Aiming to significantly broaden the application of SERS to other fields, and especially to material science, we consider hybrid structures of plasmonic nanostructures and other material phases and strategies for producing strong local EM fields at desired locations in such hybrid structures. In this vein, we consider some of the numerical strategies for simulating the optical properties and consequential SERS performance of particle-on-substrate systems that might guide the design of SERS-active systems. Finally, some current theoretical attempts are briefly discussed for unifying EM and non-EM contribution to SERS.

Abstract:Since its discovery, the phenomenon of Surface Enhanced Raman Scattering (SERS) has gradually become an important tool for analyzing the composition and structure of substances. As a trace technique that can efficiently and nondestructively detect single molecules, the application of SERS has expanded from environmental and materials science to biomedical fields. In the past decade or so, the explosive development of nanotechnology and nanomaterials has further boosted the research of SERS technology, as nanomaterial-based SERS substrates have shown good signal enhancement properties. So far, it is widely recognized that the morphology, size, composition, and stacking mode of nanomaterials have a very great influence on the strength of the substrate SERS effect. Herein, an overview of methods for the preparation of surface-enhanced Raman scattering (SERS) substrates is provided. Specifically, this review describes a variety of common SERS substrate preparation methods and explores the potential and promise of these methods for applications in chemical analysis and biomedical fields. By detailing the influence of different nanomaterials (e.g., metallic nanoparticles, nanowires, and nanostars) and their structural features on the SERS effect, this article aims to provide a comprehensive understanding of SERS substrate preparation techniques.
