Abstract:Oncogenic mutations of HER2 have been identified in HER2-low tumors. Such mutations in the extracellular region could serve as tumor-specific antigens that are accessible to antibody therapeutics. Although multiple antibody therapeutics have been successfully developed against cancers that overexpress wild-type HER2, none is specifically applicable to treating tumors driven by HER2 mutants expressed at a low level. Here, we developed antibodies exquisitely selective to HER2 S310F and S310Y, two most common oncogenic mutations in the HER2 extracellular region. Cryo-EM structures of the homodimer of HER2 S310F and an antibody bound to HER2 S310F revealed that these antibodies recognize the mutations in a manner that mimic the dimerization arm of HER and inhibit HER2 dimerization. These antibodies as T-cell engagers selectively killed a HER2 S310F-driven cancer cell line in vitro and in a mouse xenograft model. These results validate HER2 extracellular region mutations as actionable therapeutic targets and offer promising candidates toward clinical development. More generally, they suggest that point mutations in the extracellular region of cell-surface receptors as actionable targets for anti-cancer therapy. Abstract:The human epidermal growth factor receptor (HER)2 provides an excellent target for selective delivery of cytotoxic drugs to tumor cells by antibody-drug conjugates (ADC) as has been clinically validated by ado-trastuzumab emtansine (Kadcyla(TM)). While selecting a suitable antibody for an ADC approach often takes specificity and efficient antibody-target complex internalization into account, the characteristics of the optimal antibody candidate remain poorly understood. We studied a large panel of human HER2 antibodies to identify the characteristics that make them most suitable for an ADC approach. As a model toxin, amenable to in vitro high-throughput screening, we employed Pseudomonas exotoxin A (ETA') fused to an anti-kappa light chain domain antibody. Cytotoxicity induced by HER2 antibodies, which were thus non-covalently linked to ETA', was assessed for high and low HER2 expressing tumor cell lines and correlated with internalization and downmodulation of HER2 antibody-target complexes. Our results demonstrate that HER2 antibodies that do not inhibit heterodimerization of HER2 with related ErbB receptors internalize more efficiently and show greater ETA'-mediated cytotoxicity than antibodies that do inhibit such heterodimerization. Moreover, stimulation with ErbB ligand significantly enhanced ADC-mediated tumor kill by antibodies that do not inhibit HER2 heterodimerization. This suggests that the formation of HER2/ErbB-heterodimers enhances ADC internalization and subsequent killing of tumor cells. Our study indicates that selecting HER2 ADCs that allow piggybacking of HER2 onto other ErbB receptors provides an attractive strategy for increasing ADC delivery and tumor cell killing capacity to both high and low HER2 expressing tumor cells.
Computational Engineering of a Therapeutic Antibody to Inhibit Multiple Mutants of HER2 Without Compromising Inhibition of the Canonical HER2

Abstract:Genomic germline and somatic variations may impact drug binding and even lead to resistance. However, designing a different drug for each mutant may not be feasible. In this study, we identified the most common cancer somatic mutations from the Catalogue of Somatic Mutations in Cancer (COSMIC) that occur in structurally characterized binding sites of approved therapeutic antibodies. We found two HER2 mutations, S310Y and S310F, that substantially compromise binding of Pertuzumab, a widely used therapeutics, and lead to drug resistance. To address these mutations, we designed a multi-specific version of Pertuzumab, that retains original function while also bindings these HER2 variants. This new antibody is stable and inhibits HER3 phosphorylation in a cell-based assay for all three variants, suggesting it can inhibit HER2-HER3 dimerization in patients with any of the variants. This study demonstrates how a small number of carefully selected mutations can add new specificities to an existing antibody without compromising its original function, creating a single therapeutic antibody that targets multiple common variants, making a drug that is not personalized yet its activity may be.

Targeting HER2 in solid tumors: Unveiling the structure and novel epitopes

Abstract:Human epidermal growth factor receptor-2 (HER2) is overexpressed in various solid tumor types, acting as an established therapeutic target. Over the last three decades, the fast-paced development of diverse HER2-targeted agents, notably marked by the introduction of the antibody-drug conjugate (ADC), yielding substantial improvements in survival rates. However, resistance to anti-HER2 treatments continues to pose formidable challenges. The complex structure and dynamic dimerization properties of HER2 create significant hurdles in the development of novel targeted therapeutics. In this review, we synthesize the latest insights into the structural intricacies of HER2 and present an unprecedented overview of the epitope characteristics of HER2-targeted antibodies and their derivatives. Furthermore, we delve into the correlation between anti-HER2 antibody binding epitopes and their respective functions, with a particular focus on their efficacy against resistant tumors. In addition, we highlight the potential of emerging anti-HER2 agents that target specific sites or non-overlapping epitopes, poised to transform the therapeutic landscape for HER2-positive tumors in the foreseeable future.

Abstract:Abstract Human epidermal growth factor receptor 2 (HER2) is a membrane tyrosine kinase that was found to be amplified and overexpressed in various human cancers including breast cancer. Monoclonal antibodies targeting HER2 such as trastuzumab and pertuzumab provide good therapeutic benefits in patients with high HER2 expression tumors, however, not all patients respond to the treatment, and many suffer recurrence. There is still a large unmet need for targeted therapies to treat HER2 high expression tumors. T cell engaging antibodies that direct T cells to kill cancer cells by simultaneously binding to CD3 on T cells and tumor-associated antigens (TAA) on cancer cells have emerged as a promising new approach. Such therapeutics are potent, but safe use of them depends on selectivity of the TAA engaged. Though HER2 is highly overexpressed in ~ 20% of breast cancer, expression of HER2 is not restricted to cancer cells and present at low level in normal tissues. The capability of selectively targeting cancer cells over HER2 low expressing normal tissues is critical to the safety of CD3 x HER2 antibodies. It is noteworthy to mention that unlike antibody-drug conjugate (ADC) therapeutics, CD3 x HER2 antibodies might not be a good approach to target HER2 low/neg expression tumors due to the difference in mechanism of action. CMG6A19 is a 150 kD IgG-like tri-specific CD3 x HER2 x HER2 T cell-engaging antibody generated on Chimagen’s TRIAD platform. It is capable of simultaneously recognizing and binding to two different epitopes on HER2 proteins. Multiple human cell lines with different levels of HER2 expression were used as targets in the antibody-mediated hPBMC killing assays in vitro and in vivo, among which SK-BR-3, KPL-4 and BT474 have high HER2 expression as seen in HER2 positive cancers, HT55 and JIMT1 have medium level of HER2 expression, and PC3 and MCF7 have HER2 expression level that is close to in normal tissues. Conventional 1:1 IgG-like CD3 x HER2 bispecific antibodies were used as control. Results showed that CMG6A19 demonstrated higher tumor inhibition effect compared to control antibodies in SK-BR-3/KPL-4/BT474 models, meanwhile remarkably weaker effect in PC3/MCF7 models suggesting better safety in HER2 low expressing tissues. This selectivity of reactivity towards HER2 high expressing cells is likely due to the unique orientation of the antibody which allows better T cell activation when antigen abundance is high. Preliminary preclinical study in cynomolgus monkeys suggested that the antibody was well tolerated in monkeys at the dosage of 20 mpk and had a half-life that is comparable to conventional antibodies. CD3 x HER2 x HER2 tri-specific antibodies selectively showed potent anti-tumor activity on HER2 high expressing cells and good safety on HER2 low expressing cells. Abstract:Inhibition of human epidermal growth factor receptor 2 mediated cell signaling pathway is an important therapeutic strategy for HER2-positive cancers. Although monoclonal antibodies are currently used as marketed drugs, their large molecular weight, high cost of production and susceptibility to proteolysis could be a hurdle for long-term application. In this study, we reported a strategy for the development of artificial antibody based on γ-AApeptides to target HER2 extracellular domain (ECD). To achieve this, we synthesized a one-bead-two-compound (OBTC) library containing 320,000 cyclic γ-AApeptides, from which we identified a γ-AApeptide, M-3-6, that tightly binds to HER2 selectively. Subsequently, we designed an antibody-like dimer of M-3-6, named M-3-6-D, which showed excellent binding affinity toward HER2 comparable to monoclonal antibodies. Intriguingly, M-3-6-D was completely resistant toward enzymatic degradation. In addition, it could effectively inhibit the phosphorylation of HER2, as well as the downstream signaling pathways of AKT and ERK. Furthermore, M-3-6-D also efficiently inhibited cell proliferation in vitro, and suppressed tumor growth in SKBR3 xenograft model in vivo, implying its therapeutic potential for the treatment of cancers. Its small molecular weight, antibody-like property, resistance to proteolysis, may enable it a new generation of artificial antibody surrogate. Furthermore, our strategy of artificial antibody surrogate based on dimers of cyclic γ-AApeptides could be applied to a myriad of disease-related receptor targets in future.

We aimed to investigate the prevalence, clinicopathologic characteristics, and functional relevance of novel HER2 mutations in breast cancer.Experimental Design: We performed Sanger sequencing of all exons of the HER2 gene in 1,248 primary tumors and 18 paired metastatic samples. Novel HER2 mutations were functionally characterized.Results: The total HER2 somatic mutation rate was 2.24% (28/1,248). Of the seven novel HER2 mutations, L768S and V773L were only detected in HER2-negative tumors, whereas K753E was found in HER2-positive disease. L768S and V773L mutations exhibited a significant increase in tyrosine kinase-specific activity and strongly increased the phosphorylation of signaling proteins in various cell lines. Xenograft experiments showed that NIH3T3 cells bearing the L768S and V773L mutations displayed more rapid growth. MCF10A, BT474, and MDA-MB-231 cells bearing the K753E mutation were resistant to lapatinib, but could be inhibited by neratinib. Finally, comparison of HER2 mutations in 18 pairs of primary and metastatic lesions revealed that the drug-resistant HER2 mutations (K753E and L755S) were enriched in metastatic lesions.Conclusions: HER2-negative breast cancer with activating mutations can benefit from HER2-targeted therapies. Meanwhile, mutations in the HER2 kinase domain might be a key mechanism of resistance to HER2-targeted therapy, and irreversible tyrosine kinase inhibitors such as neratinib may offer alternative treatment options. Abstract:Recent development of monoclonal antibodies as mainstream anticancer agents demands further optimization of their safety for use in humans. Potent targeting and/or effector activities on normal tissues is an obvious toxicity concern. Optimization of specific tumor targeting could be achieved by taking advantage of the extracellular acidity of solid tumors relative to normal tissues. Here, we applied a structure-based computational approach to engineer anti-human epidermal growth factor receptor 2 (Her2) antibodies with selective binding in the acidic tumor microenvironment. We used an affinity maturation platform in which dual-pH histidine-scanning mutagenesis was implemented for pH selectivity optimization. Testing of a small set of designs for binding to the recombinant Her2 ectodomain led to the identification of antigen-binding fragment (Fab) variants with the desired pH-dependent binding behavior. Binding selectivity toward acidic pH was improved by as much as 25-fold relative to the parental bH1-Fab. In vitro experiments on cells expressing intact Her2 confirmed that designed variants formatted as IgG1/k full-size antibodies have high affinity and inhibit the growth of tumor spheroids at a level comparable to that of the benchmark anti-Her2 antibody trastuzumab (Herceptin®) at acidic pH, whereas these effects were significantly reduced at physiological pH. In contrast, both Herceptin and the parental bH1 antibody exhibited strong cell binding and growth inhibition irrespective of pH. This work demonstrates the feasibility of computational optimization of antibodies for selective targeting of the acidic environment such as that found in many solid tumors.

Abstract:Aptamers provide a potential source of alternative targeting molecules for existing antibody diagnostics and therapeutics. In this work, we selected novel DNA aptamers targeting the HER2 receptor by an adherent whole-cell SELEX approach. Individual aptamers were identified by next generation sequencing and bioinformatics analysis. Two aptamers, HeA2_1 and HeA2_3, were shown to bind the HER2 protein with affinities in the nanomolar range. In addition, both aptamers were able to bind with high specificity to HER2-overexpressing cells and HER2-positive tumor tissue samples. Furthermore, we demonstrated that aptamer HeA2_3 is being internalized into cancer cells and has an inhibitory effect on cancer cell growth and viability. In the end, we selected novel DNA aptamers with great potential for the diagnosis and possible treatment of HER2-positive cancer.

Abstract:Bispecific antibodies are considered attractive bio-therapeutic agents owing to their ability to target two distinct disease mediators. Cross-arm avidity targeting of antigen double-positive cancer cells over single-positive normal tissue is believed to enhance the therapeutic efficacy, restrict major escape mechanisms and increase tumor-targeting selectivity, leading to reduced systemic toxicity and improved therapeutic index. However, the interplay of factors regulating target selectivity is not well understood and often overlooked when developing clinically relevant bispecific therapeutics. We show in vivo that dual targeting alone is not sufficient to endow selective tumor-targeting, and report the pivotal roles played by the affinity of the individual arms, overall avidity and format valence. Specifically, a series of monovalent and bivalent bispecific IgGs composed of the anti-HER2 trastuzumab moiety paired with affinity-modulated VH and VL regions of the anti-EGFR GA201 mAb were tested for selective targeting and eradication of double-positive human NCI-H358 non-small cell lung cancer target tumors over single-positive, non-target NCI-H358-HER2 CRISPR knock out tumors in nude mice bearing dual-flank tumor xenografts. Affinity-reduced monovalent bispecific variants, but not their bivalent bispecific counterparts, mediated a greater degree of tumor targeting selectivity, while the overall efficacy against the targeted tumor was not substantially affected.

Abstract:Background: EGFR and HER2 are highly expressed in many pancreatic cancers which strongly correlate with poorer prognoses. We tested homodimeric (EGFRxEGFR or HER2xHER2) and heterodimeric (EGFRxHER2) T cell-engaging bispecific antibodies (BsAbs) to direct polyclonal T cells to these antigens on pancreatic tumors. Methods: We used the 2+2 IgG(L)-scFv format to create BsAbs, previously shown to have exceptional anti-tumor properties compared to other structural platforms. These BsAbs utilize two anti-CD3 scFvs attached to the light chains of an IgG to engage T cells while retaining bivalent binding to tumor antigens with both Fab arms. Anti-EGFR was based on the Cetuximab and anti-HER2 on the Trastuzumab amino acid sequences. Using a 'knob-into-hole' strategy, we generated an EGFRxHER2 heterodimeric BsAb bearing one Fab specific for EGFR and one for HER2. T cells were tagged with luciferase for tracking by bioluminescence for in vivo trafficking studies. Results: The EGFRxEGFR, HER2xHER2, and EGFRxHER2 BsAbs demonstrate high avidity (KD: 2pM to 300 pM) and T cell-mediated cytotoxicity against several human pancreatic ductal adenocarcinoma (PDAC) cell lines in vitro with EC50s in the picomolar range (0.17pM to 18pM). They mediate potent T cell-mediated antitumor effects against subcutaneous PDAC xenografts and were highly efficient in driving human polyclonal T cells into tumors as measured by bioluminescence. To examine the importance of anti-tumor valency in antitumor effects, EGFR and HER2 BsAbs were heterodimerized with a CD33 control BsAb to generate EGFRxCD33 and HER2xCD33 BsAbs. Both EGFRxCD33 and HER2xCD33 BsAbs showed modest reductions in target avidity by surface plasmon resonance (2.7-fold and 12.6-fold, respectively), cell binding strength by FACS (8.0-fold and 63.6-fold) and cytotoxicity (7.7-fold and 47.2-fold) compared to their homodimeric counterparts and, remarkably, showed no therapeutic efficacy against SW1990 cell line-derived and patient-derived PDAC xenografts, correlating with reduced tumor-infiltrating bioluminescent T cells, when compared to EGFRxHER2 BsAb. When tested against SW1990 with CRISPR-mediated target KOs, only HER2xHER2 BsAb treatment could eliminate EGFR-KO tumors, while EGFRxEGFR and EGFRxHER2 BsAbs were ineffective. Conversely, in HER2-KO SW1990 tumors, while EGFRxEGFR BsAb demonstrated strong tumor control, HER2xHER2 and EGFRxHER2 BsAbs demonstrated only weak activity. Conclusion: EGFR and HER2 were useful targets for driving T cell infiltration and tumor ablation. HER2 was overall more effective as a target when compared to EGFR for pancreatic cancer in vivo. Bivalent Fab binding to tumor was critical for robust anti-tumor effect in vivo even when in vitro differences of monovalent BsAb were less obvious. EGFRxHER2 heterodimeric BsAb exhibited potent antitumor efficacy against EGFR(+)HER2(+) double positive PDAC xenografts, but not single-positive tumors, and could offer a strategy to spare healthy single-positive tissues to reduce 'on target/off tumor' side effects. Abstract:Hormone receptor positive (HR+) metastatic breast cancer remains a difficult clinical problem. Endocrine Therapies (ET) have remained the mainstay of therapy for decades. However, a large percent of these patients become resistant to the therapies and succumb to the disease. One of the most common resistance mechanisms is the ER alpha type I (ESR1) point mutations with amino acid substitutions such that they become constitutively active. About 40-50% of metastatic HR+ patients develop these mutations. Hence, there is a need to develop strategies that target WT and mutant ESR1 in order to achieve robust anti-tumor immune responses. Type I polarized dendritic cell (DC1) vaccine developed by our group has shown to be a powerful immunotherapeutic tool to treat breast cancer by boosting the CD4 Th1 responses against oncodrivers like HER2 and HER3. This is facilitated by identification of immunogenic human peptides using a novel peptide screening method targeting these oncodrivers and breaching the tolerance. Having strong evidence of the effectiveness of DC1 vaccines, our study focusses on testing efficacy of DC1 pulsed WT ESR1 peptides (DC1-WT ESR1) and clinically prevalent ESR1 mutations/neoantigens (DC1-Mutant ESR1) in pre-clinical mouse model. In this study, we tested the homology between human and murine WT and mutated ESR1 peptides and also assessed the efficacy of DC1-WT ESR1 and DC1-Mutant ESR1 vaccines in mice to generate CD4 Th1 response (IFNγ production). To develop pre-clinical ESR1 mouse tumor models, two different cell lines were validated for nuclear ESR1 expression. We tested the efficacy of DC1-WT ESR1 vaccine in orthotopic ESR1+ tumor bearing mice by administrating six DC1 vaccines intratumorally. Our results show around 90% and above homology between human and murine WT and mutated ESR1 peptides generating immunogenic responses in mice. In vitro treatment of ESR1+ mouse cell lines with IFNγ resulted in reduced proliferation. This effect was in line with the mouse tumor model, wherein we observed reduced tumor burden post administration of six DC1-WT ESR1 vaccines intratumorally in ESR1+ pre-clinical mouse models. In addition, we observed about a 12% of complete regression in these mice. Upon using combinatorial approaches such as fulvestrant and DC1-WT ESR1, we observed about a 14% of complete regression, however, the DC1 alone mice showed 62% responders and did a lot better than the fulvestrant alone. Overall, these findings suggest that DC1 vaccines targeting ESR1 could be a promising tool to treat aggressive ESR1+ breast cancer. We now aim to use combinatorial approaches such as CDK4/6 inhibitors and DC1-WT ESR1 to boost the anti-tumor responses and further test the efficacy of DC1-Mutant ESR1 vaccines using pre-clinical ESR1+ mouse models. Design of antigens to present a tumor-specific cryptic epitope

Abstract:In many cancers, the epidermal growth factor receptor (EGFR) gene is amplified, mutated, or both. The monoclonal antibody mAb806 binds selectively to cancer cells that overexpress EGFR or express the truncated mutant EGFRvIII, but not to normal cells. This suggests that a promising avenue for developing cancer vaccines may be to design antigens that elicit mAb806-like antibodies. In this study, we designed antigens that present the mAb806-binding epitope in the same conformation as in overexpressed or truncated EGFR. We first used molecular dynamics simulations to identify conformations of EGFR in which the residues of the mAb806-binding epitope are accessible. We then designed antigens by substituting that epitope in place of a structurally similar loop in a different protein and generating mutants that could potentially stabilize the mAb806-binding conformation in this new context. Two mutants in which the epitope remained stable in subsequent simulations were chosen for evaluation in vitro. Binding kinetics experiments with these designed antigens provided strong evidence that the epitope was successfully stabilized in the mAb806-binding conformation, suggesting that they could potentially form the basis of vaccines that elicit cancer-selective antibodies.

Structure-based Design of Peptides with High Affinity and Specificity to HER2 Positive Tumors.

Abstract:To identify peptides with high affinity and specificity against human epidermal growth factor receptor 2 (HER2), a series of peptides were designed based on the structure of HER2 and its Z(HER2:342) affibody. By using a combination protocol of molecular dynamics modeling, MM/GBSA binding free energy calculations, and binding free energy decomposition analysis, two novel peptides with 27 residues, pep27 and pep27-24M, were successfully obtained. Immunocytochemistry and flow cytometry analysis verified that both peptides can specifically bind to the extracellular domain of HER2 protein at cellular level. The Surface Plasmon Resonance imaging (SPRi) analysis showed that dissociation constants (K D) of these two peptides were around 300 nmol/L. Furthermore, fluorescence imaging of peptides against nude mice xenografted with SKBR3 cells indicated that both peptides have strong affinity and high specificity to HER2 positive tumors.
In Silico Exploration and Biological Evaluation of Bispecific Peptides Derived from Anti-HER2 Antibodies and Peptide-Camptothecin Conjugates for HER2-Positive Breast Cancer

Abstract:To enhance the affinity of the human epidermal growth receptor 2 (HER2) targeted peptide developed previously, bispecific fusion peptides P1GCGT1 and P1GCGCGT1 were designed using an in silico approach. Molecular dynamic simulation showed that both peptides strongly interacted with HER2 domains II and IV. Compared with peptides targeting each single domain, P1GCGT1 and P1GCGCGT1 could bind to HER2 more significantly and targeted HER2-positive cells specifically. Additionally, both peptides were used to generate peptide-drug conjugates with camptothecin (CPT), among which I-1 and I-4 were screened for enhanced cellular activity and selectivity. Biological evaluation demonstrated that I-1 and I-4 induced cell apoptosis, promoted cell cycle arrestin S-phase, and inhibited Topo I activity. The binding affinity assay and confocal analysis revealed that I-1 and I-4 were effective at targeting HER2. Moreover, I-1 and I-4 showed better stability than single targeting peptide and presented enhanced antitumor activity and safety than CPT in tumor-bearing mice.
Targeted Immunotherapy for HER2-low Breast Cancer with 17p Loss

Abstract:The clinical challenge for treating HER2 (human epidermal growth factor receptor 2)-low breast cancer is the paucity of actionable drug targets. HER2-targeted therapy often has poor clinical efficacy for this disease due to the low level of HER2 protein on the cancer cell surface. We analyzed breast cancer genomics in the search for potential drug targets. Heterozygous loss of chromosome 17p is one of the most frequent genomic events in breast cancer, and 17p loss involves a massive deletion of genes including the tumor suppressor TP53. Our analyses revealed that 17p loss leads to global gene expression changes and reduced tumor infiltration and cytotoxicity of T cells, resulting in immune evasion during breast tumor progression. The 17p deletion region also includes POLR2A, a gene encoding the catalytic subunit of RNA polymerase II that is essential for cell survival. Therefore, breast cancer cells with heterozygous loss of 17p are extremely sensitive to the inhibition of POLR2A via a specific small-molecule inhibitor, alpha-amanitin. Here, we demonstrate that alpha-amanitin-conjugated trastuzumab (T-Ama) potentiated the HER2-targeted therapy and exhibited superior efficacy in treating HER2-low breast cancer with 17p loss. Moreover, treatment with T-Ama induced immunogenic cell death in breast cancer cells and, thereby, delivered greater efficacy in combination with immune checkpoint blockade therapy in preclinical HER2-low breast cancer models. Collectively, 17p loss not only drives breast tumorigenesis but also confers therapeutic vulnerabilities that may be used to develop targeted precision immunotherapy.
Single domain antibodies specific for HER2 dimerization domain effectively disrupts HER2 dimerization

Abstract:Dimer-dependent phosphorylation of HER2 receptor is a key event for the signal transduction of HER family of receptors which correlates with tumor invasion and metastasis. New generation of therapies based on dimerization domain inhibition using monoclonal or fragment antibodies was introduced. A potent method for manufacturing antibodies and antibody fragments is the phage display antibody library method. A recombinant phage was generated using the phage display method from synthetic dAb library. Subtractive biopanning was performed on sepharose 4b resin. Evaluation of success of subtractive biopanning was confirmed by the PCR fingerprinting after the fourth round of biopanning. The fourth round of biopanning results in the isolation of several dimerization domain reactive clones based on the polyclonal phage ELISA results. Monoclonal phage cell ELISA was used to select the positive clones with the highest affinity, and they were subsequently employed for functional tests. Cell-ELISA, MTT assay and dimerization inhibition test revealed that the reactivity and specificity of the selected monoclonal phage to dimerization domain of HER2. Further, Annexin V/PI staining and gene expression analysis showed that increased apoptosis rates. Also, in silico binding of the selected clones to conformational structure of HER2 was applied, using protein-protein docking tool of the ICM-Pro software, and showed sdAbs were specifically interacted with dimerization domain of the receptor. In conclusion, we have identified a single domain targeting HER2 dimerization, which represents a promising therapeutic and diagnostic candidate for HER2-positive cancers. Purified sdAb needs to more research to evaluate it both in vivo and in vitro via functional tests to determine if it can be applied for treatment and diagnostics.
