Abstract:The smuggling of illicit drugs urges the development of new tools for rapid on-site identification in cargos. Current methods rely on presumptive color tests and portable spectroscopic techniques. However, these methods sometimes exhibit inaccurate results due to commonly used cutting agents, the colorful nature of the sample or because the drugs are smuggled in common goods. Interestingly, electrochemical sensors can deal with these specific problems. Herein, an electrochemical device is presented that uses affordable screen-printed electrodes for the electrochemical profiling of several illicit drugs by square-wave voltammetry (SWV). The identification of the illicit compound is based on the oxidation potential of the analyte. Hence, a library of electrochemical profiles is built upon the analysis of illicit drugs and common cutting agents. This library allows the design of a tailor-made script that enables the identification of each drug through a user-friendly interface (laptop or mobile phone). Importantly, the electrochemical test is compared by analyzing 48 confiscated samples with other portable devices based on Raman and FTIR spectroscopy as well as a laboratory standard method (i.e., gas chromatography–mass spectrometry). Overall, the electrochemical results, obtained through the analysis of different samples from confiscated cargos at an end-user site, present a promising alternative to current methods, offering low-cost and rapid testing in the field.

Abstract:The increasing demand for rapid and sensitive roadside identification of trace illegal drugs drives the development of high-performance miniature mass spectrometric instrumentation and methodology. Here, we report a synchronized flash-thermal-desorption purging and ion injection (SFTDPI) method to increase the sensitive and rapid screening of volatile and nonvolatile illegal drugs for miniature ion trap mass spectrometry (ITMS). The flash-thermal desorption could reach 290 degrees C in 2.5 s, which could achieve efficient vaporization of nonvolatile noscapine with boiling point at 565 degrees C. ITMS using discontinuous atmospheric pressure interface (DAPI) has an ion utilization ratio of less than 1%, the synchronized purging for flash-thermal desorption and ion injection with DAPI could accurately control the time interval along with the desorption, gas purging, ionization and ion injection, and the sample utilization ratio increases more than five times. The miniature SFTDPI-ITMS presents good performance: (1) more than 60 times improvement in sensitivity was achieved compared to the previously reported thermal-desorption acetone-assisted photoionization ion trap mass spectrometer for nonvolatile drugs, and the minimum detectable quantity reaches 50 pg for fentanyl. (2) Ten kinds of mixing drugs with boiling point difference of 300 degrees C can be simultaneously identified within 3 s under a single analysis. SFTDPI-ITMS was deployed at the roadside checkpoint of Sino-Burmese border; fentanyl in the captured encapsulated powder and the suspected opiate had been successfully identified.

Abstract:Most women who are diagnosed with breast cancer have either no risk factors or only one. Recommendations for breast cancer screening for women between the ages of 40 and 49 are controversial, because the National Institutes of Health (NIH) Consensus Conference made no specific recommendation regarding the frequency of screening. In so doing, a possible reduction in mortality or morbidity is balanced against known false-positive or false-negative rates and the detection of ductal carcinoma in situ. Although genetic testing is available, many questions surround its use, including its likely emotional and financial impact on the patient and her family and the unresolved issue of how to interpret the results.
Real-time electrochemical screening of cocaine in lab and field settings with automatic result generation

Abstract:This work presents the results of a novel application for the fast on-site screening of cocaine and its main cutting agents in suspicious and confiscated samples. The methodology behind the novel application consists of portable electrochemical detection coupled with a peak recognition algorithm for automated result output generation, validated both in laboratory and field settings. Currently used field tests, predominantly colorimetric tests, are lacking accuracy, often giving false positive or negative results. This presses the need for alternative approaches to field testing. By combining portable electrochemical approaches with peak recognition algorithms, an accuracy of 98.4% concerning the detection of cocaine was achieved on a set of 374 powder samples. In addition, the approach was tested on multiple "smuggled," colored cocaine powders and cocaine mixtures in solid and liquid states, typically in matrices such as charcoal, syrup, and clothing. Despite these attempts to hide cocaine, our approach succeeded in detecting cocaine during on-site screening scenarios. This feature presents an advantage over colorimetric and optical detection techniques, which can fail with colored sample matrices. This enhanced accuracy on smuggled samples will lead to increased efficiency in confiscation procedures in the field, thus significantly reducing societal economic and safety concerns and highlighting the potential for electrochemical approaches in on-the-spot identification of drugs of abuse.

Abstract:Heavy metals released from industries causes severe environmental pollution in developing countries. This study has been conducted to evaluate the intensity of heavy metals pollution in soil at 0, 30 and 60 m distances from waste carrying canal of Bhaluka industrial area of Mymensingh, Bangladesh. Lead (Pb), chromium (Cr), copper (Cu), zinc (Zn), iron (Fe) and manganese (Mn) concentrations in soils decreased gradually with the increase of distance from waste canal. Maximum concentration was found at 60 to 0 m distance varied from 67.13–90.93, 52.23–76.73, 32.75–133.85, 61.18–422.10, 26900–36900 and 240–540 µg g-1 for Pb, Cr, Cu, Zn, Fe and Mn, respectively. Geoaccumulation index showing that the soil was moderately polluted for Pb and Zn. Pollution load index values>1.0, explaining pollution load was increased adjacent to industrial area and deteriorate the quality of surface soils day by day. Contamination factor for Pb, Cu and Zn were very high indicating these metals as the major soil pollutants came from anthropogenic sources which was supported by enrichment factor values (>5). The extent of pollution in adjacent to this industrial area implies the condition becoming worse and alarming for biota and inhabitants of that area.

Abstract:Methamphetamine (MA) is one of the most virulent illicit drugs that can be synthesized from household materials leading to its prevalent trafficking and local manufacturing in clandestine drug laboratories (clan labs). The significant problems of tracing MA in clan labs and monitoring drug abusers lie in the lag time between sample collection and analysis and the number of tests done. Capillary electrophoresis (CE) is a rapid separation technique amenable to miniaturization and field testing. Herein, we developed a simple transient isotachophoretic (tITP)-CE method to detect MA and its precursor pseudoephedrine (PSE) in clan labs and non-invasive biological fluids. The method was implemented on the ETD-100, a commercial fully automated portable CE instrument with an integrated swab-based extraction system. Within 2 min of insertion of the swab, MA and PSE were automatically extracted with a leading electrolyte (LE) and then separated on covalently modified capillaries. The ETD-100 showed a limit of detection (LOD) and quantification (LOQ) of MA 0.02 and 0.05 μg/swab and 0.02 and 0.06 μg/swab of PSE, with an enhancement factor of 118 and 328, respectively, when compared to a normal non-tITP injection. The intra and inter-day relative standard deviation in terms of migration time were in the range of 0.75-1.93 % for both MA and PSE and were 2.0-2.4 % for both MA and PSE peak height. The method was demonstrated with the detection of spiked MA and PSE on different household materials as well as in non-invasive biological fluids with a recovery above 60 %.

Abstract:Among the drugs of abuse, cocaine (COC) is the second most commonly used worldwide due to its stimulating and psychoactive effects. Currently, this drug is classified, according to the World Drug Report (WDR), as an illicit substance and is frequently seized by law enforcement agencies. Additionally, it can cause social problems due to its abusive use. This study describes the development of an electroanalytical methodology using square wave voltammetry (SWV) associated with a single electrode of carbon paste modified with 10 % MWCNTs (multi-walled carbon nanotubes) to quantify COC in seized samples containing up to 20% cocaine (low purity index) in the state of Espírito Santo (Brazil). SWV parameters, including E s = 10 mV, a = 30 mV, and f = 15 Hz, were optimized utilizing a Britton-Robinson buffer solution of 0.1 mol/L (pH 10) as a supporting electrolyte through the implementation of experimental design (CCD). COC showed an oxidation peak at approximately 1.0 V, with low limits of quantification and detection (1.90 × 10 −6 mol L −1 and 5.75 × 10 −7 mol L −1 , respectively), in the linear range from 2.36 × 10 −6 mol L −1 to 1.38 × 10 −5 mol L −1 , with the proposed methodology. Thus, this research develops a fast and low-cost technique in favor of the police, allowing the R 2 = 99%. The results obtained in the intraday calibration curves present a relatively low standard deviation (RSD < 8%), indicating a good repetition of the value production of an electrode for less than USD 1.42 per analysis.

Abstract:In this work, we investigated the ability of an electrochemical sensor to recognize Cannabis sativa L. samples with different total content of Δ9-tetrahydrocannabinol (Δ9-THC), determined by the levels of the psychoactive cannabinoid and of its biosynthetic precursor Δ9-tetrahydrocannabinolic acid (Δ9-THCA), using a multivariate approach. The voltammetric responses recorded with screen-printed electrodes modified with carbon black reflected the compositional differences from the different samples, in terms of cannabinoids of the vegetal material. PLS-DA models allowed for the correct classification of most C. sativa samples into the classes of legal and illegal samples according to total Δ9-THC content, based on threshold limits defined by the EU/US (0.3 % w/w) and Italian (0.6 % w/w) regulations. Satisfactory results were achieved in both cases, obtaining classification efficiency values in prediction of the external test set equal to 85 % and 100 % for the EU/US and Italian thresholds, respectively. The obtained results suggest the possibility to consider the proposed method as a starting point for the implementation of an automated device for rapid prescreening of total Δ9-THC content directly on site.
Integrated Sensing Platform Validated for the Efficient and On-Site Screening of Amine-Containing Illicit Drugs

Abstract:Efficient and reliable technologies for the on-site detection of illicit drugs are important in drug-facilitated crime investigations. However, the development of such technologies is challenging. Based on the synthetic optimization, introducing a boron ester functional group to the two furanic indicators endows the stimulus-responsive properties synergistically. The ring-opening reaction of the indicators in the presence of amine-containing illicit drugs generated well-known donor-acceptor Stenhouse adducts, accompanied by strong color changes. A small-size and lightweight laminated sensor was integrated based on the outstanding ratiometric variations of the two active furanic indicators. A prototype platform was fabricated equipped with a circuit control, a mini pump, and a signal processing system. A user-friendly detection and efficient screening of amine-containing illicit drugs, including phenethylamines, amphetamines, cathinones, and tryptamines in the liquid states were conducted. The ratiometric response of the sensor was linear in the concentration range of 2.1-10.6 μg·mL-1 for methamphetamine·HCl and methcathinone ·HCl. The detection limits for the two illicit drugs at the sublevel (ng·mL-1) were found to be 8.4 and 9.0 ng·mL-1, respectively. Double-blind field tests and different illicit drugs were evaluated with good screening capability. Successful trials showed the potential applications of the developed prototype platform for efficient and on-site analytical determination.

Abstract:Screening tests in forensic laboratories are a critical step in ensuring an efficient and effective analytical scheme for presumptive identification. Electrochemical surface-enhanced Raman spectroscopy (EC-SERS) represents a novel workflow that can be applied both in the laboratory and on-site as a fast, inexpensive, and selective approach to seized drug screening. Using cyclic voltammetry and a 785 nm Raman spectrometer, a nontargeted screen was developed using silver screen-printed electrodes and tested on a panel of common drugs of abuse and adulterants. Following characterization of the analyte panel, in-house binary and tertiary mixtures were assessed and the effectiveness of the developed EC-SERS method was tested using common score-based algorithms including correlation, hit-quality-index, spectral angle mapper, and correlation of the 1st derivative. For in-house blind samples, this approach allowed for positive identification of at least one compound in 100 % of samples. Identification of all compounds was lower at 52 %. Seized drug samples from adjudicated casework were tested on-site at the Maryland State Police laboratory as a fit-for-purpose study. EC-SERS provided an accurate screening result of 86 % using the 1st derivative correlation. Applying knowledge of both the local drug landscape and the prevalence of specific adulterants, this value improved to a positive screening of 93 % for the authentic samples. EC-SERS represents a novel approach to drug screening that could impact forensic laboratories, customs and border patrol, public health, and scene investigations. Future work should focus on improved data processing and chemometric tools for data generated in EC-SERS methods.

Abstract:A deep ultraviolet-visible (DUV-Vis) reflected optical fiber sensor was developed for use in a simple spectrophotometric detection system to detect the absorption of various illegal drugs at wavelengths between 180 and 800 nm. Quantitative analyses performed using the sensor revealed a high specificity and sensitivity for drug detection at a wavelength of approximately 200 nm. Using a double-absorption optical path length, extremely small sample volumes were used (32 to 160 nL), which allowed the use of minimal amounts of samples. A portable spectrophotometric system was established based on our optical fiber sensor for the on-site determination and quantitative analysis of common illegal drugs, such as 3,4-methylenedioxymethamphetamine (MDMA), ketamine hydrochloride, cocaine hydrochloride, diazepam, phenobarbital, and barbital. By analyzing the absorbance spectra, six different drugs were quantified at concentrations that ranged from 0.1 to 1000 μg mL(-1) (16 pg-0.16 μg). A novel Matching Algorithm of Spectra Space (MASS) was used to accurately distinguish between each drug in a mixture. As an important supplement to traditional methods, such as mass spectrometry or chromatography, our optical fiber sensor offers rapid and low-cost on-site detection using trace amounts of sample. This rapid and accurate analytical method has wide-ranging applications in forensic science, law enforcement, and medicine.

Abstract:In this manuscript, a 3D-printed analytical device has been successfully developed to classify illicit drugs using smartphone-based colorimetry. Representative compounds of different families, including cocaine, 3,4-methylenedioxy-methamphetamine (MDMA), amphetamine and cathinone derivatives, pyrrolidine cathinones, and 3,4-methylenedioxy cathinones, have been analyzed and classified after appropriate reaction with Marquis, gallic acid, sulfuric acid, Simon and Scott reagents. A picture of the colored products was acquired using a smartphone, and the corrected RGB values were used as input data in the chemometric treatment. ANN using two active layers of nodes (6 nodes in layer 1 and 2 nodes in layer 2) with a sigmoidal transfer function and a minimum strict threshold of 0.50 identified illicit drug samples with a sensitivity higher than 83.4 % and a specificity of 100 % with limits of detection in the microgram range. The 3D printed device can operate connected to a rechargeable lithium-ion cell portable battery, is inexpensive, and requires minimal training. The analytical device has been able to discriminate the analyzed psychoactive substances from cutting and mixing agents, being a useful tool for law enforcement agents to use as a screening method.

Non-contact identification and differentiation of illicit drugs using fluorescent films

Abstract:Sensitive and rapid identification of illicit drugs in a non-contact mode remains a challenge for years. Here we report three film-based fluorescent sensors showing unprecedented sensitivity, selectivity, and response speed to the existence of six widely abused illicit drugs, including methamphetamine (MAPA), ecstasy, magu, caffeine, phenobarbital (PB), and ketamine in vapor phase. Importantly, for these drugs, the sensing can be successfully performed after 5.0 × 105, 4.0 × 105, 2.0 × 105, 1.0 × 105, 4.0 × 104, and 2.0 × 102 times dilution of their saturated vapor with air at room temperature, respectively. Also, presence of odorous substances (toiletries, fruits, dirty clothes, etc.), water, and amido-bond-containing organic compounds (typical organic amines, legal drugs, and different amino acids) shows little effect upon the sensing. More importantly, discrimination and identification of them can be realized by using the sensors in an array way. Based upon the discoveries, a conceptual, two-sensor based detector is developed, and non-contact detection of the drugs is realized.

Abstract:Drug abuse is a social issue worldwide, and there is an increasing demand for on-site rapid detection of illicit drugs. In this study, a rapid and simple analytical method for the detection of 6-monoacetylmorphine (6-MAM), methamphetamine (MA), methylenedioxymethamphetamine (MDMA), ketamine (K), norketamine (NK), and cocaine (COC) in urine was developed. The developed method combines C18 pipette-tip based solid-phase extraction (C18 PT-SPE) with a miniaturized mass spectrometer (miniMS), exhibiting remarkable simplicity, high sensitivity, and strong reliability, compared with the conventional method. The optimal extraction and elution conditions for C18 PT-SPE were considered as 9 and 3 aspirating-dispensing cycles, respectively. The miniMS parameters including spray voltage, isolation potential, and collision-induced dissociation energy for the detection of these six illicit drugs were optimized using a nano-electrospray ionization method. The limit of detection (LOD), limit of quantification (LOQ), linear range, and linearity for the analysis of six illicit drugs in urine with the proposed C18 PT-SPE-miniMS method were determined. Except for the LOD of K and COC was determined as 0.5 and 0.25 ng mL-1 respectively, and the LOD of 6-MAM MA, MDMA, and NK was determined as 1 ng mL-1. This method enables rapid on-site detection, providing easier operation, lower cost, and better portability compared to conventional methods, making it a potential tool in drug crime investigation and forensic science.
Evaluation of spectroscopic techniques for on-site drug testing of festival seizures

Abstract:Despite the fact that drugs of abuse are illegal, a drug-free festival still remains an utopia in most settings. For law enforcement purposes, it is necessary to rapidly determine whether controlled substances are involved. On-site testing is a challenging task because drugs appear in different physical forms and concentrations. The aim of this study was to compare the performance of two spectroscopic techniques, Raman and Fourier transform-infrared (FT-IR), for the testing of drug seizures at a dance festival. First, samples were measured through packaging with Raman. Subsequently, homogenized samples were analysed with FT-IR. For MDMA tablets, a chemometric model was applied on the FT-IR spectra for the dose estimation. After the festival, results were confirmed in the forensic laboratory with gas chromatography coupled with mass spectrometry (GC-MS) and gas chromatography with flame ionization detection (GC-FID). In total, 166 samples of which 90 tablets, 53 powders, 16 crystals and 7 liquids were analysed. MDMA, cocaine and ketamine were the top three drugs seized. The Raman technique was suitable for powders and crystals (sensitivity of 100% and 81%, respectively). However, in comparison with FT-IR, Raman performance was lower for the analysis of liquids (sensitivity of 67%) and 'ecstasy'-like tablets (sensitivity of 41%). Overall, sensitivities above 95% were obtained with FT-IR. The MDMA doses of the tablets, determined on-site, ranged between 52 mg and 336 mg MDMA hydrochloride. For a quick identification of a variety of drugs on-site, the combination of Raman and FT-IR is recommended. It should be emphasized that optimized settings, in-house libraries and analysis by trained operators are essential to obtain correct results.

Abstract:In this work, a novel coupled approach MicroNIR/Chemometrics based on a miniaturized and portable spectrometer is proposed for the on site detection of amphetamines (AMP) in non pretreated oral fluids. In particular, the coupling of MicroNIR with chemometrics was investigated with the aim of developing a fast and accurate approach able to perform the on-site prediction of AMP abuse. A predictive model to be used in real cases was developed by collecting specimens from volunteers and spiked samples with increasing amounts of AMP were prepared to optimize calibration. Partial Least Square-Discriminant Analysis (PLS-DA) and Partial Least Square regression (PLS) were involved to detect and quantify AMP. Results demonstrated that MicroNIR/Chemometric platform is statistically able to identify AMP abuse in simulated oral fluid samples containing, with the accuracy and sensitivity of the actual proposed official reference methods. The method was checked against false positive and true positive response and results proved to be those required for confirmatory analyses. This method would permit to simplify AMP abuse monitoring for roadside drug testing or workplace surveillance and may be of help at first aid points.
Portable Digital Linear Ion Trap Mass Spectrometer Based on Separate-Region Corona Discharge Ionization Source for On-Site Rapid Detection of Illegal Drugs

Abstract:As narcotic control has become worse in the past decade and the death toll of drug abuse hits a record high, there is an increasing demand for on-site rapid detection of illegal drugs. This work developed a portable digital linear ion trap mass spectrometer based on separate-region corona discharge ionization source to meet this need. A separate design of discharge and reaction regions was adopted with filter air as both carrier gas for the analyte and protection of the corona discharge needle. The linear ion trap was driven by a digital waveform with a low voltage (±100 V) to cover a mass range of 50–500 Da with a unit resolution at a scan rate of 10,000 Da/s. Eighteen representative drugs were analyzed, demonstrating excellent qualitative analysis capability. Tandem mass spectrometry (MS/MS) was also performed by ion isolation and collision-induced dissociation (CID) with air as a buffer gas. With cocaine as an example, over two orders of magnitude dynamic range and 10 pg of detection limit were achieved. A single analysis time of less than 10 s was obtained by comparing the information of characteristic ions and product ions with the built-in database. Analysis of a real-world sample further validated the feasibility of the instrument, with the results benchmarked by GC-MS. The developed system has powerful analytical capability without using consumables including solvent and inert gas, meeting the requirements of on-site rapid detection applications.
