Abstract:This work is concerned with investigating the effect of substrate hydrophobicity and zeta potential on the dynamics and kinetics of the initial stages of bacterial adhesion. For this purpose, bacterial pathogens Staphylococcus aureus and Escherichia coli O157:H7 were inoculated on the substrates coated with thin thiol layers (i.e., 1-octanethiol, 1-decanethiol, 1-octadecanethiol, 16-mercaptohexadecanoic acid, and 2-aminoethanethiol hydrochloride) with varying hydrophobicity and surface potential. The time-resolved adhesion data revealed a transformation from an exponential dependence to a square root dependence on time upon changing the substrate from hydrophobic or hydrophilic with a negative zeta potential value to hydrophilic with a negative zeta potential for both pathogens. The dewetting of extracellular polymeric substances (EPS) produced by E. coli O157:H7 was more noticeable on hydrophobic substrates, compared to that of S. aureus, which is attributed to the more amphiphilic nature of staphylococcal EPS. The interplay between the timescale of EPS dewetting and the inverse of the adhesion rate constant modulated the distribution of E. coli O157:H7 within microcolonies and the resultant microcolonial morphology on hydrophobic substrates. Observed trends in the formation of bacterial monolayers rather than multilayers and microcolonies rather than isolated and evenly spaced bacterial cells could be explained by a colloidal model considering van der Waals and electrostatic double-layer interactions only after introducing the contribution of elastic energy due to adhesion-induced deformations at intercellular and substrate-cell interfaces. The gained knowledge is significant in the context of identifying surfaces with greater risk of bacterial contamination and guiding the development of novel surfaces and coatings with superior bacterial antifouling characteristics.

Surface adhesion of viruses and bacteria: Defend only and/or vibrationally extinguish also?! A perspective

Abstract:Coronaviruses COVID-19, SARS-CoV and NL63 use spikes in their corona to bind to angiotensin converting enzyme 2 (ACE2) sites on cytoskeletal membranes of host cells to deliver their viral payload. While groups such as disulfides in ACE2's zinc metallopeptidase, and also in COVID-19's spikes, facilitate such binding, it is worth exploring how similar complementary sites on materials such as polymers, metals, ceramics, fabrics, and biomaterials promote binding of viruses and bacteria and how they could be further engineered to prevent bioactivity, or to act as agents to collect viral payloads in filters or similar devices. In that vein, this article offers a perspective on novel tools and approaches for chemically and topologically modifying most utilitarian surfaces via defensive topological vibrational engineering to either prevent such adhesion or to enhance adhesion and elicit vibrational characteristics/'musical signatures' from the surfaces so that the structure of the binding sites of viruses and bacteria is permanently altered and/or their cellular machinery is permanently disabled by targeted chemical transformations. Abstract:A recent study published in Nature Communications showed that essential modulatory roles of interfacial adhesion and mechanical microenvironments such as geometric constraints and extracellular matrix stiffness, in microbe-host cell interactions. This study utilized single-cell force spectroscopy and RNA sequencing to gain insight into the intrinsic mechanisms by which the mechanical microenvironment regulates bacterial-host interactions and therefore reveal potential interventions against bacterial entry. Meanwhile, the adhesion forces involved in the bacteria-host interactions were recognized as a new indicator for assessing the extent of bacterial infection. Taken together, these findings demonstrate that interfacial adhesion forces and mechanical microenvironments play a dominant role in modulating functions and behaviors of microorganisms and host cells, which also provide a mechanobiology-inspired idea for the development of subsequent drug-resistant antimicrobials and broad-spectrum antiviral drugs.

Abstract:The ability to control cell behaviour, cell fate and simulate reliable tissue models in vitro remains a significant challenge yet is crucial for various applications of high throughput screening e.g. drug discovery. Mechanotransduction (the ability of cells to convert mechanical forces in their environment to biochemical signalling) represents an alternative mechanism to attain this control with such studies developing techniques to reproducibly control the mechanical environment in techniques which have potential to be scaled. In this review, the use of techniques such as finite element modelling and precision interferometric measurement are examined to provide context for a novel technique based on nanoscale vibration, also known as "nanokicking". Studies have shown this stimulus to alter cellular responses in both endothelial and mesenchymal stem cells (MSCs), particularly in increased proliferation rate and induced osteogenesis respectively. Endothelial cell lines were exposed to nanoscale vibration amplitudes across a frequency range of 1-100 Hz, and MSCs primarily at 1 kHz. This technique provides significant potential benefits over existing technologies, as cellular responses can be initiated without the use of expensive engineering techniques and/or chemical induction factors. Due to the reproducible and scalable nature of the apparatus it is conceivable that nanokicking could be used for controlling cell behaviour within a wide array of high throughput procedures in the research environment, within drug discovery, and for clinical/therapeutic applications.STATEMENT OF SIGNIFICANCE: The results discussed within this article summarise the potential benefits of using nanoscale vibration protocols for controlling cell behaviour. There is a significant need for reliable tissue models within the clinical and pharma industries, and the control of cell behaviour and stem cell differentiation would be highly beneficial. The full potential of this method of controlling cell behaviour has not yet been realised.

Abstract:For bacterial adhesion and biofilm formation, a thorough understanding of the mechanism and effective modulating is lacking due to the complex extracellular electron transfer (EET) at bacteria-surface interfaces. Here, we explore the adhesion behavior of a model electroactive bacteria under various metabolic conditions by an integrated electrochemical single-cell force microscopy system. A nonlinear model between bacterial adhesion force and electric field intensity is established, which provides a theoretical foundation for precise tuning of bacterial adhesion strength by the surface potential and the direction and flux of electron flow. In particular, based on quantitative analyses with equivalent charge distribution modeling and wormlike chain numerical simulations, it is demonstrated that the chain conformation and unfolding events of outer membrane appendages are dominantly impacted by the dynamic bacterial EET processes. Abstract:ABSTRACT Introduction: Guanfacine is a noradrenergic agonist that is believed to improve symptoms of attention-deficit/hyperactivity disorder (ADHD) through selective actions at α2A-adrenoceptors in the prefrontal cortex. A recent double-blind, multicenter trial supports the efficacy and safety of guanfacine extended release (GXR) for pediatric ADHD. This long-term, open-label extension was conducted to study the safety profile and effectiveness of GXR for up to 2 years. Methods: Subjects were 240 children 6–17 years of age with a diagnosis of ADHD who participated in the preceding randomized trial. GXR was initiated at 2 mg/day and titrated as needed in 1-mg increments to a maximum of 4 mg/day to achieve optimal clinical response. Results: The most common adverse events were somnolence (30.4%), headache (26.3%), fatigue (14.2%), and sedation (13.3%). Somnolence, sedation, and fatigue were usually transient. Cardiovascular-related adverse events were uncommon, although small reductions in mean blood pressure and pulse rate were evident at monthly visits. ADHD Rating Scale, Version IV, total and subscale scores improved significantly from baseline to endpoint for all dose groups (P<.001 for all comparisons, intent-to-treat population). Conclusion: Long-term treatment with GXR was generally safe for up to 24 months of treatment, and effectiveness was maintained over this treatment period.

Abstract:The first stage of biofilm-associated infections is commonly caused by initial adhesion of bacteria to intravascular tubes, catheters and other medical devices. The overuse of antibiotics to treat these infections has led to the spread of antibiotic resistance, which has made infections difficult to eradicate. It is crucial to develop advanced strategies to inhibit biofilm formation, avoiding the emergence of antibiotic resistance. Previously, it has been reported that substrate stiffness plays an important role in the initial attachment of bacteria. However, the mechanism of how the stiffness modulates the initial adhesion of bacteria remains unclear. Here, we developed magnetic nanoprobe-based force-induced remnant magnetization spectroscopy (FIRMS) as a new platform to measure the adhesion force of bacteria. Through examining the initial adhesion force and the adhesive protein, fibronectin-binding protein (FnBP), of Staphylococcus aureus (S. aureus), we found that the increase of the substrate stiffness promoted the expression of FnBP, thus enhancing the initial adhesion force of bacteria. Following the formation of initial adhesion, the substrates with soft stiffness delayed the biofilm formation, whereas those with moderate stiffness showed preferential promotion of the biofilm formation. We expect this versatile platform to be beneficial to the study of adhesion behaviors of bacteria that sheds light on the design of new medical materials to treat microbial infections.
Harnessing electrical energy for anti-biofilm therapies: effects of current on cell morphology and motility

Abstract:Preferentially altering bacterial migration could be a successful approach for augmenting the natural wound-healing process. Inducing electrotactic behaviours through the application of physiologically safe currents is one possible strategy for altering bacterial movement. By controlling bacterial movement at the site of infection, healing times and the severity/extent of bacterial infection could be reduced. Here, we deployed microfluidics and atomic force microscopy to determine the effect of an applied electrical current on bacterial motility and cell morphology in wound pathogens namely Pseudomonas aeruginosa, Escherichia coli, and methicillin-resistant Staphylococcus aureus (MRSA). Nanoscale imaging combined with microfluidic platforms allows for the study of single-cell swimming dynamics. Current values of 0, 0.07, and 0.125 mA were applied to bacteria in suspension. E. coli exhibited an increase in directionality and a drop in mean cellular velocity across all voltages. P. aeruginosa showed a significant decrease in mean cellular velocity at 0.07 mA, while all currents increased the directionality of movement. Electrical current had no statistically significant effect on the width or length of individual bacterial cells. A better understanding of how electrical stimulation affects pathogenic bacteria at wound sites may lead to advancements in electrotherapy and help to identify new alternatives to traditional antimicrobial approaches.

Abstract:We report an in-vivo electromechanical Atomic Force Microscopy (AFM) study of charge distribution on the cell wall of Gram plus Rhodococcus wratislaviensis bacteria, naturally adherent to a glass substrate, in physiological conditions. The method presented in this paper relies on a detailed study of AFM approach-retract curves giving the variation of the interaction force versus distance between tip and sample. In addition to classical height and mechanical (as stiffness) data, mapping of local electrical properties, as bacterial surface charge, was proved to be feasible at a spatial resolution better than few tens of nanometers. This innovative method relies on the measurement of the cantilever's surface stress through its deflection far from (higher than 10nm) the repulsive contact zone. The variations of surface stress come from modification of electrical surface charge of the cantilever (as in classical electrocapillary measurements) likely stemming from its charging during contact of both tip and sample electrical double layers. This method offers an important improvement in local electrical and electrochemical measurements at the solid-liquid interface particularly in high-molarity electrolytes when compared to technics focused on the direct use of electrostatic force. It thus opens a new way to directly investigate in-situ biological electrical surface processes involved in numerous practical and fundamental problems as bacterial adhesion, biofilm formation, microbial fuel cell, etc.

Abstract:Biofilm formation is most commonly combatted with antibiotics or biocides. However, proven toxicity and increasing resistance of bacteria increase the need for alternative strategies to prevent adhesion of bacteria to surfaces. Chemical modification of the surfaces by tethering of functional polymer brushes or films provides a route toward antifouling coatings. Furthermore, nanorough or superhydrophobic surfaces can delay biofilm formation. Here we show that submicrometer-sized roughness can outweigh surface chemistry by testing the adhesion of E. coli to surfaces of different topography and wettability over long exposure times (>7 days). Gram-negative and positive bacterial strains are tested for comparison. We show that an irregular three-dimensional layer of silicone nanofilaments suppresses bacterial adhesion, both in the presence and absence of an air cushion. We hypothesize that a 3D topography can delay biofilm formation (i) if bacteria do not fit into the pores of the coating or (ii) if bending of the bacteria is required to adhere. Thus, such a 3D topography offers an underestimated possibility to design antibacterial surfaces that do not require biocides or antibiotics.

Mechanical stimuli activate gene expression via a cell envelope stress sensing pathway

Abstract:Mechanosensitive mechanisms are often used to sense damage to tissue structure, stimulating matrix synthesis and repair. While this kind of mechanoregulatory process is well recognized in eukaryotic systems, it is not known whether such a process occurs in bacteria. In Vibrio cholerae, antibiotic-induced damage to the load-bearing cell wall promotes increased signaling by the two-component system VxrAB, which stimulates cell wall synthesis. Here we show that changes in mechanical stress within the cell envelope are sufficient to stimulate VxrAB signaling in the absence of antibiotics. We applied mechanical forces to individual bacteria using three distinct loading modalities: extrusion loading within a microfluidic device, direct compression and hydrostatic pressure. In all cases, VxrAB signaling, as indicated by a fluorescent protein reporter, was increased in cells submitted to greater magnitudes of mechanical loading, hence diverse forms of mechanical stimuli activate VxrAB signaling. Reduction in cell envelope stiffness following removal of the endopeptidase ShyA led to large increases in cell envelope deformation and substantially increased VxrAB response, further supporting the responsiveness of VxrAB. Our findings demonstrate a mechanosensitive gene regulatory system in bacteria and suggest that mechanical signals may contribute to the regulation of cell wall homeostasis.

Abstract:The environmental adaption, growth, motion, and other dynamic behaviors of bacteria are closely associated with the coupled mechanical–electrochemical properties of their subcellular structures, but the underlying regulatory mechanisms remain unclear. The mechanical responses of bacteria are difficult to elucidate by traditional models without considering the mechanical, chemical, and electric coupling effects. In this paper, a mechanical–electrochemical theory is constructed to investigate the deformation behavior of bacterial cells. A bacterium is treated as a bilayer structure consisting of a negatively charged polysaccharide capsule and an elastic envelope subjected to turgor pressure. This model is used to reveal the regulating roles of the electrostatic double-layer force and osmosis under different electrolyte conditions. Good agreement is found between the theoretical predictions and the experimentally observed three-stage nanoindentation responses of Klebsiella pneumoniae (K. pneumoniae) bacterial cells. Furthermore, we investigate the mechanical–electrochemical coupling mechanisms in the compression resistance of the bacterial capsule. The results reveal that the osmosis induced by ionic imbalance and polysaccharide–solvent aggregates plays a significant role in the compression resistance of the capsule. The present model not only deepens our understanding of the mechanical–electrochemical coupling mechanisms of bacterial cells at the subcellular scale, but also holds promise for applications in characterizing their mechanical properties.

Abstract:Electroactive bacteria play a crucial role in electrocatalytic oxidation and reduction reactions within bioelectrochemical systems (BES). However, the influence of BES startup modes on the adhesion of electroactive bacteria and the underlying mechanisms remains elusive. We hypothesize that startup mode-induced microniches trigger changes in bacterial motility, which mediate bacterial adhesion on anodic surface. To test this hypothesis, we employed Shewanella oneidensis MR-1 as a model strain and launched investigations in single-chambered BES under both external voltage-free (EVF) and external voltage-applied (EVA) modes. Our results showed that the EVA mode facilitated MR-1 adhesion up to fourfold compared to the EVF mode. Surface chemistry analysis revealed that EVA mode, with proper external voltages (400–600 mV), enhanced substrate adsorption onto anodic surfaces by up to 410 %, significantly promoting bacterial cell movement and chemotactic migration towards anodic surfaces, thereby facilitating bacterial adhesion. Principal component analysis and structural equation modeling indicated that lactate adsorption capacity was the determining factor for bacterial adhesion under EVF mode, whereas external voltage diminished the effect of lactate on triggering cell motility and subsequent bacterial adhesion in the EVA mode. Our findings provide valuable insights into the initiation and enhancement of electrochemically active biofilm on electrode surfaces in BES.

Abstract:Microorganisms are very sensitive to the effect of metal ions and a deficit or an excess of essential metal ions may induce some cell disturbances. The main objective of this research is to evaluate the effect of toxic metal ions. Doing that, the effects of metal ions on the electrophysical properties of the cell membrane, probed by electroorientational spectroscopic techniques and analysis of the electrophoretic mobility of cells have been chosen for investigation. The purpose of this work is to determine the optimal conditions of investigation of different kinds of microorganisms exposed to Cu. pH had a great influence on the electrophysical changes induced by Cu and this may be related to the speciation of the metal, according to its degree of hydroxylation. It will now be possible to compare the effects of toxic metal ions (Cd, Hg, Pb, etc..) or therapeutic metal ions (Pt, Ga, Ru, Ti, Sn, etc..) with those observed for Cu, in the conditions optimized by this study.

Escherichia coli adhesion is mediated by appendages, including type-1 fimbriae and curli amyloid fibers. Antifouling surfaces prevent the adhesion of bacteria to combat biofilm formation. Here, we used single-cell force-spectroscopy to study the interaction between E. coli and glass or two antifouling surfaces: the tripeptide DOPA-Phe(4F)-Phe(4F)-OMe and poly(ethylene glycol) polymer-brush. Our results indicate that both antifoulants significantly deter E. coli initial adhesion. By using two mutant strains expressing no type-1 fimbriae or curli amyloids, we studied the adhesion mechanism. Our results suggest that the bacteria adhere to different antifoulants via separate mechanisms. Finally, we show that some bacteria adhere much better than others, illustrating how the variability of bacterial cultures affects biofilm formation. Our results emphasize how additional study at the single-cell level can enhance our understanding of bacterial adhesion, thus leading to novel antifouling technologies.

Abstract:Cell-substrate interaction plays a critical role in determining the mechanical status of living cell membrane. Changes of substrate surface properties can significantly alter the cell mechanical microenvironment, leading to mechanical changes of cell membrane. However, it is still difficult to accurately quantify the influence of the substrate surface properties on the mechanical status of living cell membrane without damage. This study addresses the challenge by using an electrochemical sensor made from an ultrasmall quartz nanopipette. With the tip diameter less than 100 nm, the nanopipette-based sensor achieves highly sensitive, noninvasive and label-free monitoring of the mechanical status of single living cells by collecting stable cyclic membrane oscillatory signals from continuous current versus time traces. The electrochemical signals collected from PC12 cells cultured on three different substrates (bare ITO (indium tin oxides) glass, hydroxyl modified ITO glass, amino modified ITO glass) indicate that the microenvironment more favorable for cell adhesion can increase the membrane stiffness. This work provides a label-free electrochemical approach to accurately quantify the mechanical status of single living cells in real-time, which may help to better understand the relationship between the cell membrane and the extra cellular matrix.

Abstract:In addition to their visible motion such as swimming (e.g., with the help of flagella), bacteria can also exhibit nanomotion that is detectable only with highly sensitive instruments, and this study shows that it is possible to detect bacterial nanomotion using an AFM detection system. The results show that the nanomotion characteristics depend on the bacterial strain, and that nanomotion can be used to sense the metabolic activity of bacteria because the oscillations are sensitive to the food preferences of the bacteria and the type of surrounding medium.

Abstract:Understanding how bacteria respond to ventilated environments is a crucial concept, especially when considering accurate airflow modeling and detection limits. To properly design facilities for aseptic conditions, we must minimize the parameters for pathogenic bacteria to thrive. Identifying how pathogenic bacteria continue to survive, particularly due to their multi-drug resistance characteristics, is necessary for designing sterile environments and minimizing pathogen exposure. A conserved characteristic among bacterial organisms is their ability to maintain intracellular homeostasis for survival and growth in hostile environments. Mechanosensitive (MS) channels are one of the characteristics that guide this phenomenon. Interestingly, during extreme stress, bacteria will forgo favorable homeostasis to execute fast-acting survival strategies. Physiological sensors, such as MS channels, that trigger this survival mechanism are not clearly understood, leaving a gap in how bacteria translate physical stress to an intracellular response. In this paper, we study the role of mechanosensitive ion channels that are potentially triggered by aerosolization. We hypothesize that change in antimicrobial uptake is affected by aerosolization stress. Bacteria regulate their defense mechanisms against antimicrobials, which leads to varying susceptibility. Based on this information we hypothesize that aerosolization stress affects the antimicrobial resistance defense mechanisms of Escherichia coli (E. coli). We analyzed the culturability of knockout E. coli strains with different numbers of mechanosensitive channels and compared antibiotic susceptibility under stressed and unstressed airflow conditions. As a result of this study, we can identify how the defensive mechanisms of resistant bacteria are triggered for their survival in built environments. By changing ventilation airflow velocity and observing the change in antibiotic responses, we show how pathogenic bacteria respond to ventilated environments via mechanosensitive ion channels.
