Abstract:Three E. coli O157:H7 outbreaks have been attributed to contaminated pork in Alberta, Canada, recently. This study investigates the phylogenetic relatedness of E. coli O157:H7 from pigs, cattle, and pork-production environments for source attribution. Limited strain diversity was observed using five conventional subtyping methods, with most or all strains being in one subgroup. Whole-genome single nucleotide polymorphism analysis confirmed the recent ancestry of the isolates from all three sources. Most environmental isolates clustered closer with pig isolates than cattle isolates. Also, a direct link was observed between 2018-outbreak environmental isolates and isolates collected from a pig farm in 2018. The majority of pig isolates harbor only one Shiga toxin gene, stx2a, while 70% (35/50) of the cattle isolates have both stx1a and stx2a. The results show some E. coli O157:H7 strains could establish persistence on pig farms and as such, pigs can be a significant source of the organism.

Abstract:ABSTRACT Although heat-stable (ST) and heat-labile (LT) enterotoxins produced by enterotoxigenic Escherichia coli (ETEC) have been documented as important factors associated with diarrheal diseases, investigations assessing the contributions of individual enterotoxins to the pathogenesis of E. coli infection have been limited. To address the individual roles of enterotoxins in the diarrheal disease caused by K88-positive ETEC in young pigs, enterotoxin-positive and -negative isogenic E. coli strains were constructed by using pBR322 to clone and express LT and STb. Four strains, K88 + astA , K88 + astA /pBR322, K88 + astA STb + , and K88 + astA LT + , were constructed and subsequently included in gnotobiotic piglet challenge studies, and their pathogenesis was assessed. The results indicated that all K88 + isogenic strains were able to colonize the small intestines of piglets exhibiting the K88 receptor. However, only LT- and STb-positive strains caused appreciable diarrhea. Piglets inoculated with the K88 + astA LT + strain became dehydrated within 18 h, while those inoculated with the K88 + astA STb + strain did not, although diarrhea developed in several piglets. The changes in the blood packed-cell volume and plasma total protein of gnotobiotic piglets inoculated with the LT-positive strains were significantly greater than those of pigs inoculated with the K88 astA /pBR322 strain ( P = 0.012, P = 0.002). Immunochemistry image analysis also suggested that LT enhanced bacterial colonization in a gnotobiotic piglet model. This investigation suggested that LT is a major contributor to the virulence of K88 + ETEC and that isogenic constructs are a useful tool for studying the pathogenesis of ETEC infection.
Serological and virulence factors analysis of Escherichia coli isolates causing post-weaning diarrhea in intensive pig farms

Abstract:In order to determine the prevalent serotypes and virulence factors of pathogenic Escherichia coli isolates from piglets with post-weaning diarrhea(PWD) in intensive pig farms of Zhejiang Province,rectum swabs from piglets with PWD in intensive pig farms were collected for isolations of pathogenic Escherichia coli.The isolations were identified by gram stain,biochemical reactions and pathogenicity tests in mice.The O serotyping and pili typing were conducted by slide agglutination with antiserum against 11 commonly seen swine Escherichia coli O antigens and anti-K88 and anti-F18 monoclonal antibodies,respectively.The types of adhesins and enterotoxins were further verified by Polymerase chain reactions(PCR) with specific primer pairs.Among 56 Escherichia coli isolates,33 were typable and distributed in 11 serotypes.The prevalent serotypes included O_(149), O_(139) and O_8,amounted to 51.5%(17/33) of the total typable ones.Slide agglutination tests and PCR analysis showed that the enterotoxigenic Escherichia coli accounted for 67.92%(36/53),of which harbored the genes for enterotoxin STa,STb,STa+STb,LT and SLT-2e amounted to 30.19%(16/53),35.85%(19/53),18.9%(10/53),1.89%(1/53)and 9.43%(5/53),respectively.Isolations with F18 and K88 adhesins contributed 28.30%(15/53),24.53%(13/53),respectively.It is concluded that pathogenic Escherichia coli isolates collected from piglets with PWD of Zhejiang Province covered at least 11 O serotypes and the predominant serotypes are O_(149),O_(139),and O_8.Escherichia coli causing PWD are mostly enterotoxigenic,with the predominant enterotoxins STa,STb,SLT-2e.The toxins and adhesins F18 and K88 are the most important virulence factors involved.

Abstract:Enterotoxigenic Escherichia coli (ETEC)-associated post-weaning diarrhea (PWD) is economically one of the most important diseases for the swine industry. Porcine ETEC strains typically express K88 or F18 fimbria and heat-labile (LT) and/or heat-stable (STa, STb) enterotoxins. However, recent studies indicate that EAST1 toxin, adhesin involved in diffuse adherence (AIDA-I) and porcine attaching and effacing-associated factor (paa) may also be expressed by ETEC strains associated with diarrhea. To better understand the virulence factors of E. coli strains that cause PWD, we applied PCR to screen for K88, F18, F41, 987P and K99 fimbrial genes; LT, STa, STb, Stx2e and EAST1 toxic genes; and AIDA-I, paa and EAE adhesin genes in E. coli strains recently isolated from young pigs with PWD in the US. Of 304 E. coli isolates from diarrheic pigs submitted for testing, 175 (57.6%) strains possessed fimbrial genes: K88 (64.6%), F18 (34.3%), F41 (0.57%), K99 (0.57%), 987P (0); toxin genes: LT (57.7%), STb (72.6%), STa (27.4%), STx2e (17.4%), EAST1 (35%); and adhesin genes: AIDA-I (26.9%), paa (60%), EAE (1.1%). All toxin genes except the EAST1 toxin gene, were almost exclusively associated with K88+ or F18+ isolates, and most of these isolates carried multiple toxin genes. The non-fimbrial adhesin paa was found present in over half of the K88+ isolates. A total of 129 (42%) isolates carried no fimbrial genes, including 66 (21.7%) isolates that did not have any of the above virulence genes. These results suggest a broad array of virulence genes associated with PWD in pigs.

Abstract:Extraintestinal pathogenic Escherichia coli (ExPEC) is an important zoonotic pathogen. Recently, ExPEC has been reported to be an emerging problem in pig farming. However, the mechanism of pathogenicity of porcine ExPEC remains to be revealed. In this study, we constructed a transposon (Tn) mutagenesis library covering Tn insertion in over 72% of the chromosome-encoded genes of a virulent and multi-drug resistant porcine ExPEC strain PCN033. By using a mouse infection model, a transposon-directed insertion site sequencing (TraDIS) assay was performed to identify in vivo fitness factors. By comparing the Tn insertion frequencies between the input Tn library and the recovered library from different organs, 64 genes were identified to be involved in fitness during systemic infection. 15 genes were selected and individual gene deletion mutants were constructed. The in vivo fitness was evaluated by using a competitive infection assay. Among them, ΔfimG was significantly outcompeted by the WT strain in vivo and showed defective adhesion to host cells. rfa which was involved in lipopolysaccharide biosynthesis was shown to be critical for in vivo fitness which may have resulted from its role in the resistance to serum killing. In addition, several metabolic genes including fepB, sdhC, fepG, gltS, dcuA, ccmH, ddpD, narU, glpD, malM, and yabL and two regulatory genes metJ and baeS were shown as important determinants of in vivo fitness of porcine ExPEC. Collectively, this study performed a genome-wide screening for in vivo fitness factors which will be important for understanding the pathogenicity of porcine ExPEC.

Abstract:Enterotoxigenic Escherichia coli (ETEC)-associated diarrhea causes a substantial economic loss to swine producers worldwide. The majority of ETEC strains causing porcine diarrhea, especially postweaning diarrhea (PWD), produce heat-labile toxin (LT) and heat-stable toxin b (STb). LT is commonly used in vaccine development, but STb has not been included because of its poor immunogenicity. As a virulence factor in porcine diarrhea, STb needs to be included as an antigen for development of broad-spectrum vaccines. In this study, we used an LT toxoid (LTR192G [hereafter, LT192]) derived from porcine ETEC to carry a mature STb peptide for LT192-STb fusions to enhance STb immunogenicity for potential vaccine application. Anti-LT and anti-STb antibodies were detected in immunized rabbits and pigs. In addition, when challenged with an STb-positive ETEC strain, all 10 suckling piglets borne by immunized gilts remained healthy, whereas 7 out 9 piglets borne by unimmunized gilts developed moderate diarrhea. This study indicates that the LT192-STb fusion enhanced anti-STb immunogenicity and suggests the LT192-STb fusion antigen can be used in future vaccine development against porcine ETEC diarrhea.
Progress and Challenges in Vaccine Development Against Enterotoxigenic Escherichia Coli (Etec)-Associated Porcine Post- Weaning Diarrhoea (PWD)

Abstract:Porcine Post-Weaning Diarrhea (PWD) continues to be one of the most important swine diseases worldwide. Entero toxigenic Escherichia coli (ETEC) strains are the primary cause of PWD. The key virulence factors of ETEC in PWD are bacterial fimbrial adhesins and enterotoxins. Adhesions mediate ETEC bacteria initial attachment to pig epithelial cells and subsequent colonization at pig small intestines. Enterotoxins including heat-labile toxin (LT) and heat-stable toxin (ST) disrupt fluid homeostasis in small intestinal epithelial cells to cause electrolyte-rich fluid hyper-secretion and diarrhea. Currently there are no effective prevention measures to protect weaned pigs against PWD. Vaccination would be the most practical and effective prevention approach, and vaccines inducing anti-adhesion immunity to block ETEC attachment and colonization and also antitoxin immunity to neutralize enterotoxicity are considered optimal against ETEC-associated PWD. Although progress has been made in past decades in developing effective vaccines against PWD, challenges continue to exist due to the disease complexity and immunological heterogeneity among ETEC strains. Recent progress in using safe toxoid antigens, toxoid fusion antigens and MEFA (multiepitope fusion antigen) approach to develop multivalent vaccines for broad protection, however, shows promising in developing new vaccines for effective protection against ETEC-associated PWD.

Abstract:To examine whether the heat-labile enterotoxin gene in porcine enterotoxigenic Escherichia coli (ETEC) strains is as divergent as in human ETEC strains, we sequenced the heat-labile and heat-stable toxin genes from 52 and 33 porcine ETEC strains, respectively. We found that the STa gene is identical, that the LT gene has only two mutations in 4 (of 52) strains, and that both mutations cause a reduction in GM1 binding and toxicity.

Abstract:Virulence of enterotoxigenic Escherichia coli (ETEC) is associated with fimbrial adhesins and enterotoxins such as heat-labile (LT) and/or heat-stable (ST) enterotoxins. Previous studies using a cell culture model suggest that exclusion of ETEC from attachment to epithelial cells requires expression of both an adhesin such as K88 (F4) fimbriae, and LT. To test the ability of non-pathogenic E. coli constructs to exclude virulent ETEC sufficiently to prevent clinical disease, we utilized a piglet ETEC challenge model. Thirty-nine 5-day-old piglets were inoculated with a placebo (control), or with either of the three K88+ E. coli strains isogenic with regard to modified LT expression: 8017 (pBR322 plasmid vector control), non-toxigenic mutant 8221 (LTR192G) in pBR322, or 8488, with the LT gene fused to the STb gene in pBR322 (LTR192G-STb). Piglets were challenged with virulent ETEC Strain 3030-2 (K88+/LT/STb) 24h post-inoculation. K88ac receptor-positive piglets in the control group developed diarrhea and became dehydrated 12–24h post-challenge. Piglets inoculated with 8221 or 8488 did not exhibit clinical signs of ETEC disease; most piglets inoculated with 8017 showed diarrhea. Control pigs exhibited significant weight loss, increased blood total protein, and higher numbers of colony-forming units of 3030-2 E. coli in washed ileum and jejunum than treated pigs. This study shows for the first time that pre-inoculation with an avirulent strain expressing adhesive fimbriae and a non-toxic form of LT provides significant short term protection from challenge with a virulent ETEC strain that expresses the same fimbrial adhesion and enterotoxin.

Piglets developed identical diarrheal diseases when inoculated with constructs expressing human or porcine enterotoxins.

Abstract:Shiga toxin-producing Escherichia coli (STEC) causes diarrhea and hemorrhagic colitis with life-threatening complications, such as hemolytic uremic syndrome. The aim of this study was to assess the molecular epidemiologic features of non-O157 STEC strains from different resources in China and illustrate the role of animal reservoirs or animal-derived foodstuffs in human STEC infections. A collection of 301 non-O157 STEC isolates from domestic and wild animals (i.e., cattle, goat, pig, yak, pika, and antelope), raw meats (i.e., beef, pork, mutton, chicken, and duck), diarrheal patients, and healthy carriers in different regions of China were selected in this study. Of the 301 analyzed STEC isolates, 67 serogroups, and 118 serotypes were identified; this included some predominant serogroups associated with human disease, such as O26, O45, O103, O111, and O121. Eighteen different combinations of stx subtypes were found. Eleven isolates carried the intimin gene eae, 93 isolates contained ehxA, and 73 isolates carried astA. The prevalence of other putative adhesion genes saa, paa, efa1, and toxB was 28.90% (87), 6.98% (21), 2.31% (7), and 1% (3), respectively. The phylogenetic distribution of isolates was analyzed by multilocus sequence typing (MLST). Ninety-four sequence types were assigned across the 301 isolates. A subset of isolates recovered from yak and pika residing in the similar wild environments, Qinghai-Tibetan plateau, showed similar genetic profiles and more tendencies to cluster together. Isolates from goat and mutton exhibited close genetic relatedness with those from human-derived isolates, providing evidence that transmission may have occurred locally within intraspecies or interspecies, and importantly, from animal reservoirs, or raw meats to humans. Comparing isolates in this study with highly virulent strains by MLST, along with serotyping and virulence profiles, it is conceivable that some of isolates from goat, yak, or raw meats may have potential to cause human diseases.

Abstract:This study was aimed to examine the genetic characteristics of 44 Streptococcus suis type 2 (SS2) isolates and the virulence attributes of 23 representative isolates. Multilocus sequence typing revealed five sequence types (ST1, ST7, ST28, ST86, and ST162) with 19 isolates assigned to ST7 (43.2%), 14 to ST1 (31.8%), and 9 to ST28 (20.5%). PCR typing of the pilus gene clusters classified the isolates into three types: A (72.7%), B (22.7%), and N (4.5%). All isolates of pilus types A and N were assigned to the ST1 complex containing ST1, ST7, and ST86, while those of type B belonged to the ST27 complex comprising ST28 and ST162. Only two strains had the putative pathogenicity island 89-kb cluster (89K) and were of type N. The type B strains had a significantly lower adhesion, were more readily killed by macrophage, and had lower virulence to mice than those of types A and N. We conclude that SS2 strains of both ST1 and ST27 complexes, parallel to pilus types A and B, were prevalent in the pig populations in Zhejiang Province, and ST7 and ST1 strains were the predominant genotypes in the diseased pigs with pneumonia.

Abstract:F4 (K88) and F18 fimbriaed enterotoxigenic Escherichia coli (ETEC) are the predominant causes of porcine post-weaning diarrhea (PWD), and vaccines are considered the most effective preventive approach against PWD. Since heterologous DNA integrated into bacterial chromosome could be effectively expressed with stable inheritance, we chose probiotic EcNc(E. coli Nissle 1917 prototype cured of cryptic plasmids) as a delivery vector to express the heterologous F4 or both F4 and F18 fimbriae and sequentially assessed their immune efficacy of anti-F4 and F18 fimbriae in both murine and piglet models. Employing the CRISPR-cas9 technology, yjcS, pcadA, lacZ, yieN/trkD, maeB, and nth/tppB sites in the chromosome of an EcNc strain were targeted as integration sites to integrate F4 or F18 fimbriae cluster genes under Ptet promotor to construct two recombinant integration probiotic strains(RIPSs), i.e. nth integration strain (EcNcΔnth/tppB::PtetF4) and multiple integration strain (EcNc::PtetF18x4::PtetF4x2). Expression of F4, both F4 and F18 fimbriae on the surfaces of two RIPSs was verified with combined methods of agglutination assay, Western blot and immunofluorescence microscopy. The recombinant strains have improved adherence to porcine intestinal epithelial cell lines. Mice and piglets immunized with the nth integration strain and multiple integration strain through gavage developed anti-F4, and both anti-F4 and anti-F18 IgG immune response. Moreover, the serum antibodies from the immunized mice and piglets significantly inhibited the adherence of F4+ or both F4+ and F18+ ETEC wildtype strains to porcine intestinal cell lines in vitro, indicating the potential of RIPSs as promising probiotic strains plus vaccine candidates against F4+/F18+ ETEC infection.

Abstract:Enterotoxigenic Escherichia coli (ETEC) is an important type of pathogenic bacteria that causes diarrhea in humans and young livestock. The pathogen has a high morbidity and mortality rate, resulting in significant economic losses in the pig industry. To effectively prevent piglet diarrhea, we developed a new tetravalent genetically engineered vaccine that specifically targets ETEC. To eliminate the natural toxin activity of ST 1 enterotoxin and enhance the preventive effect of the vaccine, the mutated ST 1 , K88ac , K99 , and LT B genes were amplified by PCR and site-specific mutation techniques. The recombinant strain BL21(DE3)(pXKK3SL) was constructed and achieved high expression. Animal experiments showed that the inactivated vaccine had eliminated the natural toxin activity of ST 1 . The immune protection test demonstrated that the inclusion body and inactivated vaccine exhibited a positive immune effect. The protection rates of the inclusion body group and inactivated vaccine group were 96 and 98%, respectively, when challenged with 1 minimum lethal dose, indicating that the constructed K88ac-K99-3ST 1 -LT B vaccine achieved a strong immune effect. Additionally, the minimum immune doses for mice and pregnant sows were determined to be 0.2 and 2 mL, respectively. This study suggests that the novel K88ac-K99-3ST 1 -LT B vaccine has a wide immune spectrum and can prevent diarrhea caused by ETEC through enterotoxin and fimbrial pathways. The aforementioned research demonstrates that the K88ac-K99-3ST 1 -LT B vaccine offers a new genetically engineered vaccine that shows potential for preventing diarrhea in newborn piglets.

Abstract:Diarrhea in pre-weaning and post-weaning piglets is still a frequently occurring disease on scaled farms,and result in great economic losses.Enterotoxigenic Escherichia coli(ETEC) is a major cause,and the virulence factors are the key to its pathogenicity.The traditional and commonly detected pili of ETEC are F4(K88),F5(K99),F6(987P),F18 and F41,and the enterotoxins include heat-stable STa and STb,heat-labile enterotoxin LT and Stx2e.However,new reports have shown that ETEC strains lacking of recognized fimbriae are becoming increasingly common,which may be linked to the use of vaccines incorporating fimbriated strains,providing selection pressure for the acquisition and carriage of novel,unrecognized virulence genes.Recently,EAST1(enteroaggregative Escherichia coli heat-stable enterotoxin 1),PAA(porcine attaching and effacing-associated factor),AIDA-I(adhesin involved in diffuse adherence)and intimin encoded by eae(Escherichia coli attaching and effacing) have been reported.Nevertheless,few related data have been available in China;furthermore,the prevention strategies have been devised largely based on the virulence factors.In order to gain more insight into the pathogenesis of colibacillosis in piglets and control the disease,it is of significant importance to track the changes of E.coli virulence factors and antimicrobial resistance,and elucidate the possible relationship between them. Samples were collected by rectum swabs from diarrheal piglets on intensive pig farms of Zhejiang Province.A total of 124 pathogenic isolates were obtained after bacteria isolation,purification,and identification based on the combination of morphology and PCR techniques.The virulence factors were verified by PCR with specific primer pairs.Susceptibility testing was carried out by K-B method,and analyzed by Whonet software.The correction between antibacterial resistance and virulence factors was evaluated by Logistic regression analysis. The PCR results showed that fimbriae positive(F4+ and F41+) amounted to 7.26%(9/124).EAST1,PAA and AIDA-I were detected for the first time in the region.EAST1 turned out to be as high as 19.68%(24/124) of the isolates,whereas only 4.84%(6/124) and 2.42%(3/124) of the isolates harbored PAA and AIDA-I,respectively.Isolates were highly resistant to oxacillin(100%),co-trimoxazole(97.6%),rifampicin(97.6%),doxycycline(96%),carbenicillin(93.5%),ampicillin(93.5%) and amoxicillin(93.5%).In addition,streptomycin showed serious cross-resistance with ampicillin(75.45%),enrofloxacin(65.45%),gentamicin(78.18%),doxycycline(96%) and florfenicol(50.91%).Besides,all isolates were multi-drug resistant among them,the majority were resistant to 14-16 antibacterial agents(43.31%).Moreover,virulence factor eae was found to be corrected with amoxicillin/clavulanic acid(P=0.046) and doxycycline(P=0.020) resistance,respectively.Similarly,there was a significant correction between EAST1 and polymyxin B(P=0.004),EAST1 and chloramphenicol(P=0.013),respectively. This investigation demonstrated that prevalent virulence factors had largely changed compared with previous studies,and resistance was still on the increase,furthermore,there existed a correlation between them.Further research is required to better understand the complicated virulence factors,bacterial resistance and its subtle correction.
Bacillus halotolerans SW207 alleviates enterotoxigenic Escherichia coli-induced inflammatory responses in weaned piglets by modulating the intestinal epithelial barrier, the TLR4/MyD88/NF-κB pathway, and intestinal microbiota

Abstract:ABSTRACT Enterotoxigenic Escherichia coli (ETEC) is one of the major pathogens contributing to piglet diarrhea, with significant implications for both piglet health and the economic aspects of the livestock industry. SW207 is an isolate of Bacillus halotolerans isolated from the cold- and disease-resistant Leixiang pigs in Northeastern China. We have discovered that SW207 can survive in the pig’s gastrointestinal fluid and under conditions of high bile salt concentration, displaying potent antagonistic activity against ETEC. In this study, we established a weaned piglet diarrhea model infected with ETEC to investigate the role of SW207 in preventing diarrhea and improving intestinal health. Results indicate that SW207 upregulates the expression of tight junction proteins, including claudin-1, occludin, and zonula occludens-1, at both the transcriptional and translational levels. Furthermore, SW207 reduces serum endotoxin, D-lactic acid, and various oxidative stress markers while enhancing piglet mechanical barrier function. In terms of immune barrier, SW207 suppressed the activation of the TLR4/MyD88/NF-κB pathway, reducing the expression of various inflammatory factors and upregulating the expression of small intestine mucosal sIgA. Concerning the biological barrier, SW207 significantly reduces the content of E. coli in the intestines and promotes the abundance of beneficial bacteria, thereby mitigating the microbiota imbalance caused by ETEC. In summary, SW207 has the potential to prevent weaned piglet diarrhea caused by ETEC, alleviate intestinal inflammation and epithelial damage, and facilitate potential beneficial changes in the intestinal microbiota. This contributes to elucidating the potential mechanisms of host-microbe interactions in preventing pathogen infections. IMPORTANCE Enterotoxigenic Escherichia coli (ETEC) has consistently been one of the significant pathogens causing mortality in weaned piglets in pig farming. The industry has traditionally relied on antibiotic administration to control ETEC-induced diarrhea. However, the overuse of antibiotics has led to the emergence of drug-resistant zoonotic bacterial pathogens, posing a threat to public health. Therefore, there is an urgent need to identify alternatives to control pathogens and reduce antibiotic usage. In this study, we assessed the protective effect of a novel probiotic in a weaned piglet model infected with ETEC and analyzed its mechanisms both in vivo and in vitro . The study results provide theoretical support and reference for implementing interventions in the gut microbiota to alleviate early weaned piglet diarrhea and improve intestinal health.

Abstract:Enterococci have the dual characteristics of being opportunistic pathogens and promising probiotics. The isolation from patients of CDC PNS-E2, a newly described Enterococcus species Enterococcus sanguinicola , may pose potential hazards. Enterococcus thailandicus from fermented sausage is a senior subjective synonym of E . sanguinicola . In this study, Enterococcus thailandicus TC1 was first isolated in healthy pigs in Tongcheng, China and identified by phenotypic analysis and 16S rRNA-based techniques. To evaluate the strain safety, an approach including virulence factors, antibiotic resistance, and animal experiments was adopted. The results show that cylA , gelE , esp , agg , ace , efaAfm , efaAfs , ptsD genes were undetected, and that the strain was sensitive or poorly resistant to some clinically relevant antibiotics. However, the isolated strain demonstrated β-hemolytic activity in rabbit blood agar plates. Analysis of animal experiments revealed that the isolated strain had no adverse effect on translocation and the internal organ indices, though significant differences in histology (villi height, crypts height) of ileum were observed. The data acquired suggest that E . thailandicus TC1 may be associated with a potential health risk.

Abstract:Introduction: Enterotoxigenic Escherichia coli (ETEC) infections are the most common cause of secretory diarrhea in suckling and post-weaning piglets. For the latter, Shiga toxin-producing Escherichia coli (STEC) also cause edema disease. This pathogen leads to significant economic losses. ETEC/STEC strains can be distinguished from general E. coli by the presence of different host colonization factors (e.g., F4 and F18 fimbriae) and various toxins (e.g., LT, Stx2e, STa, STb, EAST-1). Increased resistance against a wide variety of antimicrobial drugs, such as paromomycin, trimethoprim, and tetracyclines, has been observed. Nowadays, diagnosing an ETEC/STEC infection requires culture-dependent antimicrobial susceptibility testing (AST) and multiplex PCRs, which are costly and time-consuming. Methods: Here, nanopore sequencing was used on 94 field isolates to assess the predictive power, using the meta R package to determine sensitivity and specificity and associated credibility intervals of genotypes associated with virulence and AMR. Results: Genetic markers associated with resistance for amoxicillin (plasmid-encoded TEM genes), cephalosporins (ampC promoter mutations), colistin (mcr genes), aminoglycosides (aac(3) and aph(3) genes), florfenicol (floR), tetracyclines (tet genes), and trimethoprim-sulfa (dfrA genes) could explain most acquired resistance phenotypes. Most of the genes were plasmid-encoded, of which some collocated on a multi-resistance plasmid (12 genes against 4 antimicrobial classes). For fluoroquinolones, AMR was addressed by point mutations within the ParC and GyrA proteins and the qnrS1 gene. In addition, long-read data allowed to study the genetic landscape of virulence- and AMR-carrying plasmids, highlighting a complex interplay of multi-replicon plasmids with varying host ranges. Conclusion: Our results showed promising sensitivity and specificity for the detection of all common virulence factors and most resistance genotypes. The use of the identified genetic hallmarks will contribute to the simultaneous identification, pathotyping, and genetic AST within a single diagnostic test. This will revolutionize future quicker and more cost-efficient (meta)genomics-driven diagnostics in veterinary medicine and contribute to epidemiological studies, monitoring, tailored vaccination, and management.

Abstract:45 patients underwent urinary diversion by caecocystoplasty following simple or radical cystectomy with or without prostatectomy. The immediate postoperative course of such a major operation was surprisingly simple both in terms of the gastrointestinal tract (due to the use of automatic sutures) and in terms of the urinary tract. Micturition and diurnal continence were perfect. The few cases of incontinence related to high caecal pressure were controlled by perineal reeducation and/or the prescription of oxybutynin. A Peniflow apparatus controls the inevitable enuresis of these neobladders when the trigone has not been preserved. The upper tract is perfectly protected. The authors consider that the caecal reservoir warrants a place of choice in bladder replacement after cystectomy.
