Abstract:Glypican-3 (GPC3), which is a surface heparan sulfate proteoglycan, has been widely known the ideal biomarker for diagnosis and treatment of hepatocellular carcinoma (HCC). Accurate and sensitive detection of serum GPC3 level is an important and challenging task. In this paper, a sandwich electrochemical aptasensor was designed to specifically detect serum GPC3 level through aptamer-target-aptamer recognition. A GPC3 aptamer (GPC3 Apt ) immobilized on the surface of gold nanoparticles@reduced graphene oxide modified screen-printed electrode (Au NPs@rGO/SPCE) was used as the capture probe. Hemin-reduced graphene oxide-carboxymethyl chitosan@platinum nanoparticles (H-rGO-CMC@Pt NPs), with good mimicking peroxidases activity, were labeled with amination GPC3 aptamer, which was employed as the signal probe. Once the target GPC3 was anchored on the surface of GPC3 Apt /Au NPs@rGO/SPCE, the H-rGO-CMC@Pt NPs-GPC3 Apt was further specifically bound to the GPC3, forming the aptamer-target-aptamer sandwich structure, which can catalyze the oxidation of hydroquinone (HQ) to benzoquinone (BQ) and lead the oxidation current of HQ recorded by DPV to changing. The linear relationship between the current signal of oxidation of HQ and GPC3 concentration range of 0.0001–3.0 μg/mL and 3.0–60.0 μg/mL, with a low detection limit (LOD) of 0.0685 ng/mL. Additionally, the electrochemical aptasensor was successfully applied for the detection of GPC3 in human serum samples with the recovery of 99.95–104.06 % and relative standard deviation (RSD) of 1.31–5.22 %. Thus, the sandwich aptasensor could provide a new strategy for detection of serum GPC3 level and improve the early diagnosis rate of HCC.

Abstract:In this study, a new electrochemical aptasensor with specific identification and quantitative analysis of glypican-3 (GPC3) was constructed using reduced graphene oxide-chitosan-ferrocene deposition of Pt–Pd bimetallic nanoparticles (RGO-CS-Fc/Pt–Pd BNPs) as biosensing platform and GPC3 aptamer (GPC3apt) as recognition molecule. The GPC3apt could react specifically with GPC3 present in the experimental samples, resulting in the formation of GPC3-aptamer complexes on the biosensing platform, which would increase the electron transfer impedance and reduce the redox current of ferrocene. The electrochemical aptasensor was characterized by scanning electron microscopy (SEM), cyclic voltammetry (CV), electrochemical impedance spectrometry (EIS), and Raman spectroscopy. Under the optimal experimental conditions, the response redox current was linearly related to the concentration of GPC3 in the range from 0.001 to 10 µg mL−1 with good linear correlation coefficient (R2 of 0.9929), and had a low detection limit of 3.67 ng mL−1 (S/N = 3), high sensitivity of 0.149 µA µM−1 cm−2. In addition, the aptasensor showed long stability, good specificity, acceptable reproducibility and satisfactory recoveries (101.1–105.6%) in the detection of clinical serum samples, which developed a new research method for early clinical diagnosis.

Abstract:Glypican-3 (GPC3) protein is considered as an effective and specific biomarker for the accurate diagnosis of hepatocellular carcinoma (HCC), and there is a critical demand to construct strategies with shorter time, lower cost noise and higher sensitivity. In this study, a biosensor for label-free detection of GPC3 was prepared by modifying GPC3 aptamer (GPC3Apt) on the surface of Si-based light addressable potentiometric sensor (LAPS) chip modified by reduced graphene oxide/polyetherimide/gold nanoparticles (rGO/PEI/AuNPs). Among them, GPC3Apt with a low equilibrium dissociation constant was used as a biorecognition molecule to recognize and capture GPC3. The rGO/PEI/AuNPs decorated on the LAPS surface could enlarge the surface area for GPC3Apt immobilization and provide more active sites for GPC3Apt to bind effectively with the target GPC3. When a bias voltage was provided to the LAPS system, the forming GPC3-GPC3Apt complexs on the sensing interface of LAPS would cause the potential of the sensitive membrane surface to change and generate a corresponding offset response in photocurrent-voltage (I-V) curves. The potential shift and GPC3 concentration showed a good linear range from 0.1 to 100.0 μg/mL with a detection limit of 40.0 ng/mL (S/N = 3) and a sensitivity of 1.33638 mV/μg/mL. In addition, the LAPS detection system also has good specificity and stability, which can be used for the detection of GPC3 in spiked serum samples.

Abstract:Glypican-3 (GPC3), as an emerging biomarker, has been shown to be beneficial for the early diagnosis and treatment of hepatocellular carcinoma (HCC). In this study, an ultrasensitive electrochemical biosensor for GPC3 detection has been constructed based on the hemin-reduced graphene oxide-palladium nanoparticles (H-rGO-Pd NPs) nanozyme-enhanced silver deposition signal amplification strategy. When GPC3 specifically interacted with GPC3 antibody (GPC3Ab) and GPC3 aptamer (GPC3Apt), an "H-rGO-Pd NPs-GPC3Apt/GPC3/GPC3Ab" sandwich complex was formed with peroxidase-like properties which enhanced H2O2 to reduce the silver (Ag) ions in solution to metallic Ag, resulting in the deposition of silver nanoparticles (Ag NPs) on the surface of the biosensor. The amount of deposited Ag, which was derived from the amount of GPC3, was quantified by the differential pulse voltammetry (DPV) method. Under ideal circumstances, the response value was linearly correlated with GPC3 concentration at 10.0-100.0 μg/mL with R2 of 0.9715. When the GPC3 concentration was in the range from 0.01 to 10.0 μg/mL, the response value was logarithmically linear with the GPC3 concentration with R2 of 0.9941. The limit of detection was 3.30 ng/mL at a signal-to-noise ratio of three and the sensitivity was 1.535 μAμM-1cm-2. Furthermore, the electrochemical biosensor detected the GPC3 level in actual serum samples with good recoveries (103.78-106.52%) and satisfactory relative standard deviations (RSDs) (1.89-8.81%), which confirmed the applicability of the sensor in practical applications. This study provides a new analytical method for measuring the level of GPC3 in the early diagnosis of HCC.

Abstract:Golgi protein-73(GP73) is one of the most common biomarkers with outstanding sensitivity and specificity in the diagnosis of hepatocellular carcinoma. Herein an ultrasensitive label-free electrochemical aptasensing platform was constructed for GP73 detection based on reduced graphene oxide-ferrocene formic acid-tripterythrine tetroxide nanocomposites (RGO-Fc-Mn 3 O 4 ) and GP73 aptamer (GP73 Apt ). () was electrodeposited on the surface of screen-printed electrode (SPE) as the conductive substrate. RGO-Fc-Mn 3 O 4 was then drop-coated on /SPE as a signal probe to generate measurable redox peaks due to the electroactivity of Fc in RGO-Fc-Mn 3 O 4 . GP73 Apt was anchored on the surface of RGO-Fc-Mn 3 O 4 NPs//SPE to constitute an electrochemical aptasensing biosensor. When GP73 was captured via the specific affinity of GP73 Apt and formed GP73-GP73 Apt complex onto the aptasensing platform, the formation GP73-GP73 Apt complex impeded the electron transfer on the electrode interface, resulting in a decrease in the peak current recorded by differential pulse voltammetry method. Thus, GP73 can be detected by measuring the change of Fc redox peak current in RGO-Fc-Mn 3 O 4 . Log-linear calibration curve for GP73 concentration (0.01–1000 ng/mL) was obtained with good correlation coefficients of 0.99485 and a lower limit of detection of 0.01 ng/mL. The human serum samples were tested by direct detection method, and the relative error and relative standard deviation are 4.21–9.87% and 3.28–9.40%, respectively. All these results demonstrate that the proposed aptasensor would be a promising tool for clinical analysis of GP73 in the diagnosis of hepatocellular carcinoma.

Abstract:An ultrasensitive electrochemical immunosensor based on signal amplification of the deposition of the electroactive ferrocene-tyramine (Fc-Tyr) molecule, catalyzed by horseradish peroxidase (HRP), was constructed for the detection of the liver cancer marker Glypican-3 (GPC3). Functional electroactive molecule Fc-Tyr is reported to exhibit both the enzymatic cascade catalytic activity of tyramine signal amplification (TSA) and the excellent redox properties of ferrocene. In terms of design, the low matrix effects inherent in using the magnetic bead platforms, a quasi-homogeneous system, allowed capturing the target protein GPC3 without sample pretreatment, and loading HRP to trigger the TSA, which induced a large amount of Fc-Tyr deposited on the electrode surface layer by layer as a signal probe for the detection of GPC3. The concept of Fc-Tyr as an electroactive label was validated, GPC3 biosensor exhibited high selectivity and sensitivity to GPC3 in the range of 0.1 ng mL −1 –1 μg mL −1 . Finally, the sensor was used simultaneously with ELISA to assess GPC3 levels in the serum of clinical liver cancer patients, and the results showed consistency, with a recovery of 98.33–105.35% and a relative standard deviation (RSD) of 4.38–8.18%, providing a theoretical basis for achieving portable, rapid and point of care testing (POCT) of tumor markers.

Abstract:Hepatocellular carcinoma (HCC) is the second leading cause of cancer-related deaths in China. Glypican-3 (GPC3) is a specific antigen related to HCC, which is widely used in clinical detection as a reliable marker of HCC. In this paper, a highly sensitive homogeneous apatasensor was designed for GPC3 detection based on fluorescence resonance energy transfer (FRET) where the GPC3 aptamer labelled gold carbon dots (AuCDs-GPC3 Apt ) are used as a donor and magnetic graphene oxide (Fe 3 O 4 /GO) nanosheets are used as an acceptor. A one-step hydrothermal method was used to synthesize AuCDs to provide sufficient fluorescence. The FRET phenomenon exists between AuCDs-GPC3 Apt and Fe 3 O 4 /GO, which weakens the fluorescence intensity of the whole system. When the target GPC3 is added to the FRET system, the fluorescent AuCDs-GPC3 Apt binds to the GPC3 and forms a folded structure, which leads to AuCDs-GPC3 Apt separation from Fe 3 O 4 /GO nanosheets. The Fe 3 O 4 /GO is then magnetically separated so that the fluorescence of free labelled AuCDs-GPC3 Apt is restored. Under the optimum conditions, the fluorescence recovery rate is linearly correlated with the concentration of GPC3 (5–100 ng·mL −1 ) and the detection limit is 3.01 ng·mL −1 (S/N = 3). This strategy shows recoveries from 98.76 to 101.29% in real human serum samples and provides an immediate and effective detection method for the quantification of GPC3 with great potential applications for early diagnosis of HCC.

High-performance Assaying Phosphatidylinositol Proteoglycan 3 Based on a Dual-Mode Biosensor Coupling Near-Infrared Photoelectrochemical with Ratiometric Electrochemical Sensing

Abstract:A dual-model biosensor was prepared for assaying phosphatidylinositol proteoglycan 3 (GPC3) by coupling near-infrared photoelectrochemical with ratiometric electrochemical sensing techniques. Nd-MOFs synthesized with ionic liquid and Nd3+ ions were integrated with gold nanoparticles (AuNPs) to form Nd-MOF@AuNPs nano-composites. Nd-MOF@AuNPs nanocomposites were used as a near-infrared photoactive element to construct a photoelectrochemical interface. Fc-Apt was fixed on the Nd-MOF@AuNPs based NIR photoelectrochemical interface by Au-S bond to provide electrochemical signal through the oxidation of Fc. The specific recognition of GPC3 can lead to the decrease of the photocurrent and the oxidation peak current of Fc (IFc). Subsequently, the methylene blue-labeled aptamer (MB-Apt@AuNRs) was introduced into the interface to form a sandwich immunocomplex, MB-Apt@AuNRs/GPC3/FC-Apt, resulting in the amplification of the photocurrent and the oxidation peak current of MB (IMB) and a decrease on the value of IFc. Under optimized experimental conditions, the linear response ranges for the ratiometric electrochemical (IMB/IFc) and photoelectrochemical sensing stra-tegies were 0.005-10 and 0.001-10 ng mL-1, respectively. The calculated detection limits were 0.66 and 1.9 pg/ mL. The dual-model biosensing system provides good performances in determining GPC3 in human serum samples, and can be considered as a potential tool for the diagnosis of tumor.
Socioeconomic gradients in health for white and Mexican-origin populations.

Abstract:OBJECTIVES We assessed whether the few findings to date suggesting weak relationships between education and health-related variables among Hispanics are indicative of a more widespread pattern. METHODS We used logistic regression models to examine education differentials (i.e., education gradients) in health behaviors and outcomes among White and Mexican-origin adults, adolescents, and infants. We gathered information from 3 data sets: the Los Angeles Family and Neighborhood Survey, the Fragile Families and Child Wellbeing Study, and the National Health Interview Survey. RESULTS In contrast with patterns for Whites, education was weakly associated or not associated with numerous health-related variables among the US Mexican-origin population. Among adults, Mexican immigrants were especially likely to have weaker education gradients than Whites. CONCLUSIONS The weak relationships between education and health observed among individuals of Mexican origin may have been the result of several complex mechanisms: social gradients in health in Mexico that differ from those in the United States, selective immigration according to health and socioeconomic status, and particular patterns of integration of Mexican immigrants into US society.

Abstract:A three-dimensional graphene-based ratiometric signal amplification aptasensor for highly sensitive and selective detection of mucin1 (MUC1) by electrochemical method has been developed. Au-reduced graphene oxide (Au-RGO) composite, which was synthesized through hydrothermal reaction and freeze-drying treatment, was used as substrate for fabricating the sensor interface on glassy carbon electrode (GCE). The application of Au-RGO is beneficial for improving electrochemical performance and immobilizing aptamer. Ferrocene labeled aptamer (Fc-Apt), grafting on Au-RGO composite, worked for specific recognition of MUC1 and as internal reference to improve the detection accuracy. Au nanoparticle modified with methyl blue labeled aptamer (MB-Apt@Au) was adopted to combine with MUC1 that captured on the electrode surface to amplify the electrical signal. Under optimal experimental conditions, this sensor was applied to detect a series concentration of MUC1 solution by alternating current voltammetry method. The aptasensor shows satisfactory detection results with a wide linear range of 1 pM to 1 µM, a low detection limit of 0.25 pM and good specificity. The good performance of the prepared electrochemical aptasensor implies that this method has potential for MUC1 detection, and it would be promising for early screening and diagnosis of cancer diseases. The proposed method also lays an important foundation for preparing ratiometric signal amplification aptasensor for other biomarkers detection.

Abstract:A sandwich-type electrochemical immunosensor was developed in this study for sensitive detection of carcinoembryonic antigen (CEA) based on rGO/CS/AuNPs as a sensing platform and PTh-Au composites as signal labels. Reduced graphene oxide (rGO), Chitosan (CS) and AuNPs were layer by layer modified on glass carbon electrode (GCE), which effectively enhanced electron transfer and provided excellent large electrochemical active surface area. Polythionine-Au composites prepared by chemical reduction method greatly amplified electrochemical signal with high conductivity and large surface area. The prepared immunosensor showed high sensitivity, good stability and selectivity for detection of CEA with a wide range of 0.3 ng/mL to 30 ng/mL ( R 2 = 0.9972) and a detection limit as low as 0.1471 pg/mL. Furthermore, this sandwich-type immunosensor might be as a versatile platform for reliable early cancer diagnostics clinical.
Sensitive Electrochemical Aptamer Biosensor for Dynamic Cell Surface N-Glycan Evaluation Featuring Multivalent Recognition and Signal Amplification on A Dendrimer-Graphene Electrode Interface

Abstract:We demonstrate a multivalent recognition and highly selective aptamer signal amplification strategy for electrochemical cytosensing and dynamic cell surface N-glycan expression evaluation by the combination of concanavalin A (Con A), a mannose binding protein, as a model, conjugated poly(amidoamine) dendrimer on a chemically reduced graphene oxide (rGO-DEN) interface, and aptamer- and horseradish peroxidase-modified gold nanoparticles (HRP-aptamer-AuNPs) as nanoprobes. In this strategy, the rGO-DEN can not only enhance the electron transfer ability but also provide a multivalent recognition interface for the conjugation of Con A that avoids the weak carbohydrate protein interaction and dramatically improves the cell capture efficiency and the sensitivity of the biosensor for cell surface glycan. The high-affinity aptamer- and HRP-modified gold nanoparticles provide an ultrasensitive electrochemical probe with excellent specificity. As proof-of-concept, the detection of CCRF-CEM cell (human acute lymphoblastic leukemia) and its surface N-glycan was developed. It has demonstrated that the as-designed biosensor can be used for highly sensitive and selective cell detection and dynamic evaluation of cell surface N-glycan expression. A detection limit as low as 10 cells mL(-1) was obtained with excellent selectivity. Moreover, this strategy was also successfully applied for N-glycan expression inhibitor screening. These results imply that this biosensor has potential in clinical diagnostic and drug screening applications and endows a feasibility tool for insight into the N-glycan function in biological processes and related diseases.

Abstract:Achieving a biosensing interface without baseline drift caused by variables in matrix samples is essential for real-time detection of analytes. In this study, we developed a molecular beacon based electrochemical aptasensor to realize the ratiometric signal quantification of VEGF in serum by surface modification of nanocomposites of graphene oxide/methylene blue (GO/MB) and AuNPs followed by the attachment of ferrocene-labeled aptamer (aptamer-Fc) against VEGF. The presence of VEGF can trigger the configuration change of aptamer-Fc, resulting in the redox probe Fc being far away from the electrode surface to attenuate the electrochemical communication between electrode and Fc. Meanwhile, signal of MB also decreased due to the impediment of aptamer-Fc to electron transfer passage. The achieved GC-rGO/MB-AuNPs-aptamer-Fc sensing interface was successfully used for the sensitive detection of VEGF in real-time with a linear detection range 2-500 pg mL-1 and detection limit of 0.1 pg mL-1 based on ratiometric dual signal (Fc and MB) read-out. It was observed loading MB and AuNPs to the GO based sensing interface was favorable to enhance the analytical performance in terms of sensitivity and capability to effectively eliminate background interference. This electrochemical aptasensor provides a universal and reliable biosensing platform which is potential for real-time and sensitive tracking of various cytokines in vivo.

Abstract:P-glycoprotein (P-gp), a transmembrane glycoprotein widely expressed on the surface of various cells, is highly associated with multidrug resistance (MDR) that heralds the malignant progress of disease after drug treatment. Notably, there have been reported that serum P-gp is a potential marker for assessing the progression of disease resistance. Currently, there are few methods for point-of-care serum P-gp detection. In this study, we proposed a gold nanoparticles/electrochemically reduced graphene nanotube (AuNPs/) modified immunosensor based on a one-step electrochemical co-reduction method. The limit of detection (LOD) of our constructed electrochemical immunosensor for P-gp detection reached 0.13 ng/mL, and the detection results in serum were consistent with ELISA. The developed immunosensor is expected to provide a scientific basis for the clinical application of serum P-gp monitoring and integrated medicine.

A Luminol Electrochemiluminescence Aptasensor Based on Glucose Oxidase Modified Gold Nanoparticles for Measurement of Platelet-Derived Growth Factor BB.

Abstract:A sandwich-type luminol electrochemiluminescence (ECL) aptasensor for highly sensitive and selective detection of platelet-derived growth factor BB (PDGF-BB) is fabricated. For this proposed ECL aptasensor, a multilayered AuNPs-electrochemically reduced graphene (AuNPs-EG) nanocomposite film was formed on the GCE surface as the base of the aptasensor through a co-electrodeposition method. The AuNPs-EG composites possess high conductivity to promote the electron transfer at the electrode interface and good biocompatibility and large surface area to capture large amounts of primary aptamer (Apt1), thus amplifying the detection response. Moreover, glucose oxidase (GOD) functionalized AuNPs labeled secondary aptamer (GOD-Apt2-AuNPs) was designed as the signal probe for the sandwiched aptasensor. Enhanced sensitivity was obtained by in situ generation of H2O2 from reaction between GOD and glucose and the excellent catalytic behavior of AuNPs to the ECL of the luminol-H2O2 system. Under the optimal conditions, the as-prepared ECL aptasensor exhibited excellent analytical property for the detection of PDGF-BB in the range from 1.0×10(-13) to 5.0×10(-10) mol L(-1) with a detection limit of 1.7×10(-14) mol L(-1) (S/N=3). The application of the present protocol was demonstrated by analyzing PDGF-BB in human serum and human urine samples with the recoveries from 85.0% to 110%.

Abstract:Glycoproteins play important roles in a wide variety of biological processes. The change in the concentration levels has been associated with many cancers, as well as other diseases. Thus, rapid, sensitive and selective determination of glycoproteins is much preferred. In this work, we reported a sandwich-type electrochemical biosensor based on dual-amplification of 4-mercaptophenylboronic acid (MBA)-capped gold nanoparticles (MBA-AuNPs) and dopamine (DA)-capped AuNPs (DA-AuNPs). Biological recognition elements such as synthetic receptor and aptamer immobilized onto gold electrodes were used to capture glycoproteins. The captured glycoproteins were then derivatized with MBA-AuNPs through the formation of tight covalent bonds between the boronic acids of MBA-AuNPs and diols of glycoproteins. Electroactive DA-AuNPs were attached by the anchored MBA-AuNPs via the interaction of boronic acids with DA tags, which facilities the amplified voltammetric detection of glycoproteins. With avidin and prostate specific antigen (PSA) as model analytes, we demonstrated the feasibility and sensitivity of the proposed method. The results indicated that sub-picomolar avidin/PSA can be readily measured. We believe that this strategy will be valuable for the electrochemical detection of other glycoproteins.

Abstract:Septicemia is a serious disease that requires early diagnosis, and procalcitonin (PCT) serves as a diagnostic biomarker for this disease. Traditional clinical detection (via immune-gold chips) remains difficult and expensive. An electrochemical immunosensor based on new nanomaterials may provide a solving approach. Herein, an ultrasensitive sandwich electrochemical strategy for PCT detection was developed. Firstly, reduced graphene oxide (rGO)–gold (Au) nano-composite film was used as the immunosensor platform to increase the amount of PCT antibody 1(Ab1) immobilized. Next, single-walled carbon nanohorns (SWCNHs)/hollow Pt chains (HPtCs) complex was firstly utilized to label PCT Ab2 as signal tags. For SWCNHs with few side effects, high surface area and HPtCs with higher specific surface, better catalytic activity, complex synthesized from both may provide more advantages. Moreover, to amplify signal, HPtC catalytic activity with H2O2 was enhanced by horseradish peroxidase (HRP) for dual synergy amplification. The whole results demonstrated that the proposed immunosensor exhibited fast operation, high sensitivity, good reproducibility, acceptable stability and ideal selectivity compared with traditional method. The linear calibration of the immunosensor ranged from 1.00pg/mL to 2.00×101ng/mL with a detection limit of 0.43pg/mL. Analytical application results revealed that the immunosensor matched with the real concentrations of serum samples. Overall this immunosensor may provide a new alternative strategy for PCT detection.

Abstract:The early detection of tumor markers has an effective role in the treatment of cancer. Here, a new sandwich-type electrochemical immunosensor for early label-free detection of the cancer biomarker carcinoembryonic antigen (CEA) was developed. Dendritic tri-fan blade-like PdAuCu nanoparticles (PdAuCu NPs)/amine functionalized graphene oxide (NH2-GO) were the label of secondary antibodies (Ab2), and Au nanoparticle-decorated polydopamines (Au/PDA) were immobilized on a screen-printed carbon electrode (SPCE) as the substrate materials. Dendritic tri-fan blade-like PdAuCu NPs/NH2-GO was synthesized according to a simple hydrothermal procedure and used to immobilize antibodies (Ab2) with large surfaces areas, increased catalytic properties and good adsorption to amplify the current signals. Subsequently, Ab2/PdAuCu NPs/NH2-GO catalyzed the reduction of H2O2 in the sandwich-type immunoreactions. Under optimal conditions, the immunosensor exhibited a satisfactory response to CEA with a limit detection of 0.07 pg mL−1 and a linear detection range from 0.1 pg mL−1 to 200 ng mL−1. The proposed immunosensor could be suitable enough for a real sample analysis of CEA, and has clinical value in the early diagnosis of cancer.
