Abstract:The fall armyworm, Spodoptera frugiperda, is a notorious polyphagous pest that causes serious economic losses in crucial crops and has invaded Africa and Asia. Lufenuron is widely used for controlling S. frugiperda in China, owing to its high toxicity against this key pest, and less pollution and little impact on natural enemies. In the present study, the sublethal and transgenerational effects of lufenuron on S. frugiperda were investigated to provide in-depth information for the rational use of lufenuron. Results showed that the development time and pupae weight were not significantly affected following exposure of females to LC10 and LC25 and male S. frugiperda to the LC10 of lufenuron. However, LC25 exposure significantly reduced pupal and total development time and pupae weight of male S. frugiperda. The longevity of S. frugiperda adults was prolonged by lufenuron and the fecundity of S. frugiperda treated with LC10 of lufenuron was significantly increased by 40% compared to the control. In addition, our study demonstrated that the LC25 of lufenuron had transgenerational effects on the progeny generation. The development time of female S. frugiperda whose parents were exposed to LC25 of lufenuron was significantly decreased compared to the control. And then, the expression profiles of Vg, VgR, JHEH, JHE, JHAMT, JHBP, CYP307A1, CYP306A1, CYP302A1 and CYP314A1 genes involved in insect reproduction and development were analyzed using Quantitative Real-Time PCR (RT-qPCR). Results showed that Vg, VgR, JHE, JHAMT, and CYP306A1 were significantly upregulated at the LC10 of lufenuron, which revealed that these upregulated genes might be linked with increased fecundity of S. frugiperda. Taken together, these findings highlighted the importance of sublethal and transgenerational effects under laboratory conditions and these effects may change the population dynamics in the field. Therefore, our study provided valuable information for promoting the rational use of lufenuron for controlling S. frugiperda.
Monitoring of Early Stage Resistance to Novel Classes of Insecticides in Spodoptera Exigua (Lepidoptera:Noctuidae)

Abstract:Several classes of new insecticides with novel mode of action,such as emamectin benzoate,abamectin,indoxacarb,chlorfenapy,methoxyfenozide,JS118 and tebufenozide were developed or introduced to control the beet armyworm,spodoptera exigua(Hubner)in China.Resistance of this pest to many conventional insecticides has been reported in different countries and areas,but there is few evidence of resistance to these new insecticides in beet armyworm as yet.In order to make good use of these new insecticides and implement effective resistance management strategy,susceptibility to several classes of new insecticides was determined using leaf-dipping method in field populations of Spodoptera exigua from six areas of Jiangsu and Henan Province,in China.The results showed that in all six field populations tested with abamectin,three populations from Fengxian and Fu'ning counties,Jiangsu province and Henan province were susceptible or had decreased susceptibility(2.5~4.4-fold),one population from Dafeng county,Jiangsu province developed low level resistance(5.2-fold),and two populations from Jiangning and Sheyang counties,Jiangsu province first developed moderate levels of resistance(10.4~14.2-fold).In all four populations tested,three populations from Henan province and Jiangning,Fu'ning and Fengxian of Jiangsu province were susceptible(2.3~2.9-fold)to indoxacarb except that one population from Fengxian of Jiangsu province had low level of resistance(6.8-fold)to indoxacarb.Four populations from Henan province and Jiangning,Fu'ning and Sheyang counties of Jiangsu province were susceptible to emamectin benzoate(RR0.6~2.0-fold).In case of chlorfenapy,all four populations tested were susceptible(1.5~2.7-fold).Similarly,all populations tested were susceptible to methoxyfenozide(0.4~1.7-fold)and JS118(0.3~1.3-fold).In all six populations tested with tebufenozide,four populations from Dafeng,Jiangning and Fengxian of Jiangsu province and Henan province were susceptible(1.6~2.5-fold),and two populations from Fu'ning and Sheyang,Jiangsu province had decreased susceptibility(3.2-fold).These results provide a guideline for developing a more effective resistance management program for beet armyworm.

Abstract:Aqueous trivalent aluminum (Al) ions and their oligomers play important roles in diverse areas, such as environmental sciences and medicine. The geometries of octahedral Al(H(2)O)(6)(3+) and tetrahedral Al(OH)(4)(-) species have been studied extensively. However, structures of intermediate hydrolysis products of the Al(III) ion, such as the penta-coordinated Al(OH)(2+) species, which exists at pH values ranging from 3.0 to 4.3, and their mode of formation have been poorly understood. Here, we present that a trigonal bipyramidal Al(OH)(H(2)O)(4)(2+) structure is formed in aqueous solution and how this monomeric species dimerizes to a dinuclear [(H(2)O)(4)Al(OH)(2)Al(H(2)O)(4)](4+) complex in aqueous solution. The Gibbs free energy change calculations indicate that the formation of the dinuclear complex is preferred over the existence of two single trigonal bipyramidal Al(OH)(H(2)O)(4)(2+) species in aqueous solution. This study captures the solution dynamics and proton transfer in the oligomerization reactions of penta-coordinated Al(OH)(2+) species in aqueous solution.
One stone for two birds: Endophytic fungi promote maize seedlings growth and negatively impact the life history parameters of the fall armyworm, Spodoptera frugiperda

Abstract:The fall armyworm (FAW) Spodoptera frugiperda , is a voracious pest of cereals native to the Americas and which invaded Africa in 2016. Chemical control is the main management option, which however remains ineffective and unsustainable. Fungal endophytes are increasingly used as alternative for the management of insect pests of economic importance. This study assessed the potential of eight endophytic fungal isolates to colonize maize plant and their ability to promote seedlings growth through seed and foliar inoculations, as well as their suppressive effects on FAW. Fungal colonization rates of different plant parts by the endophytes varied as per the inoculation methods. Beauveria bassiana ICIPE 279 colonized more than 60% of all the seedling parts while B . bassiana G1LU3 only colonized stem (25%) and leaf (5%) tissues through foliar inoculation. Trichoderma atroviride F2S21, T. asperellum M2RT4, T. harzianum F2R41, Trichoderma sp. F2L41, Hypocrea lixii F3ST1 and Fusarium proliferatum F2S51 successfully colonized all the plant parts and therefore were selected and further evaluated through seed inoculation for their endophytic persistence, effect on plant growth, and pathogenicity to Spodoptera frugiperda immature and adult stages. Weekly assessment showed varied effect of the endophytes on maize plant growth parameters compared to the control. During the first week, percentage colonization of the plant parts ranges between 90%–100%, 65%–100%, and 60%–100%, in the roots, stems, and leaves, respectively for all the five tested isolates. However, the colonization pattern/rates significantly decreased over time for H. lixii F3ST1 in the stems and leaves, and for T. harzianum F2R41 in the leaves and for T. asperellum M2RT4 in the roots. In addition, T. harzianum F2R41 outperformed all the other isolates in boosting the plant height, whereas H. lixii F3ST1 and T. asperellum M2RT4 outperformed all the other isolates in increasing the wet and dry shoots weight. Furthermore, the number of egg masses laid on endophytically-colonized maize plants varied among the treatments. Trichoderma asperellum M2RT4 and H. lixii F3ST1 endophytically-colonized maize plants significantly reduced the number of egg masses and the defoliation/feeding rates of the pest compared to the control. Additionally, T. harzianum F2R41 had the highest negative impact on the pupation and adult emergence of S. frugiperda with a female-biased sex ratio. Our findings indicate that T. asperellum M2RT4, T. harzianum F2R41, and H. lixii F3ST1 hold a potential to be developed as endophytic-fungal-based biopesticides for sustainable management of S. frugiperda and as plant growth promoters.

Abstract:After being discovered in Taiwan for the first time in June 2019, the polyphagous invasive fall armyworm (FAW), Spodoptera frugiperda (Lepidoptera: Noctuidae), has since spread throughout the entire nation. In Taiwan, this insect has a significant impact on the quality and output of wheat, corn, sorghum, and millet. It may further infest more crops in Taiwan due to its diverse range of hosts and alternate hosts. Maize and other staple crops have already been the subject of several study. The biology of FAW has not yet been studied in relation to the alternative hosts, particularly those commonly found in Taiwanese farmlands. Therefore, this study proposed to investigate the effects of napier grass (Pennisetum purpureum), natal grass (Melinis repens), and sunn hemp (Crotalaria juncea) on the development, reproduction, survivorship, and population growth of FAW under laboratory conditions. According to the results, the developmental duration was considerably the shortest when FAW was reared on sunn hemp while the longest on natal grass. Furthermore, female adults reared on napier grass had a longer adult pre-oviposition period, total pre-oviposition period, oviposition period, longevity, highest fecundity, and highest net reproductive rate (Ro: 465.12). Among the tested three alternative host plants evaluated, sunn hemp had the highest intrinsic rate of increase (r: 0.1993), finite rate of increase (λ: 1.2206), and shortest mean generation time (T: 29.98). Therefore, this study suggests that all hosts plants can contribute to the development and outbreak of this pest in the absence of its primary host; however, sunn hemp was a relatively more suitable host plant for this insect. The possibilities for the FAW's growth and development vary depending on the host plant. Thereby, all potential host plants in the area should be extensively examined while developing an IPM program against FAW.

Abstract:In mid-May, 2019, the fall armyworm (FAW) Spodoptera frugiperda invaded Jiangxi Province, China, and caused extensive damage to corn crops. However, little attention has been given to the life-history traits of the FAW. In the present study, we systematically investigated the life-history traits of the newly invasive FAW on corn leaves at 19, 22, 25, 28, and 31°C under a photoperiod of LD 15:9 hr. The FAW thrived on the corn leaves with short developmental periods, high survival rates of larvae and pupae, very high mating success rates, and high fecundity. The pupal developmental stage was significantly longer in males than females at all temperatures, thus resulting in a protogyny phenomenon. The pupal weight was heaviest after a relatively shorter larval development stage at a higher temperature (25°C); thus, the FAW did not follow the temperature–size rule. Females were smaller than males, indicating sexual size dimorphism. A small proportion of females delayed their pre-oviposition period and began to lay eggs on the 7th to 9th day after adult emergence. There were positive relationships between pupal weight and larval developmental time and between adult weight and fecundity. There was a negative relationship between fecundity and longevity. These findings can help us to predict the population dynamics of the FAW on corn and to develop a suitable and practical management strategy.

Abstract:How herbivorous insects adapt to host plants is a key question in ecological and evolutionary biology. The fall armyworm, (FAW) Spodoptera frugiperda (J.E. Smith), although polyphagous and a major pest on various crops, has been reported to have a rice and corn (maize) feeding strain in its native range in the Americas. The species is highly invasive and has recently established in China. We compared behavioral changes in larvae and adults of a corn population (Corn) when selected on rice (Rice) and the molecular basis of these adaptational changes in midgut and antennae based on a comparative transcriptome analysis. Larvae of S. frugiperda reared on rice plants continuously for 20 generations exhibited strong feeding preference for with higher larval performance and pupal weight on rice than on maize plants. Similarly, females from the rice selected population laid significantly more eggs on rice as compared to females from maize population. The most highly expressed DEGs were shown in the midgut of Rice vs. Corn. A total of 6430 DEGs were identified between the populations mostly in genes related to digestion and detoxification. These results suggest that potential adaptations for feeding on rice crops, may contribute to the current rapid spread of fall armyworm on rice crops in China and potentially elsewhere. Consistently, highly expressed DEGs were also shown in antennae; a total of 5125 differentially expressed genes (DEGs) s were identified related to the expansions of major chemosensory genes family in Rice compared to the Corn feeding population. These results not only provide valuable insight into the molecular mechanisms in host plants adaptation of S. frugiperda but may provide new gene targets for the management of this pest.

Abstract:Fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is a major polyphagous pest with the potential to seriously damage various crops. A better understanding of FAW's performance on different hosts may help to predict which plants will be attacked when the preferred host is absent, and facilitate the development of effective pest management practices. We compared the larval performance of FAW fed on maize with that of FAW fed on potato and tobacco, which are important crops in China, using an age‐stage two‐sex life table and adult female oviposition preference experiments. In cage experiments with potato, tobacco, or maize as the host, FAW reared on maize exhibited the strongest performance with shorter developmental time in the larval stage, longer longevity, and a higher reproductive rate in adults. Females oviposited on maize in preference to potato or tobacco. Compared with larvae fed on maize, those fed on potato and tobacco exhibited significantly lower survival, with only 31.61% and 8.13% developing to the adult stage, respectively. Several life table parameters, including the mean generation time (T ), net reproductive rate (R 0), finite rate of increase (λ ), and intrinsic rate of natural increase (r m) were negatively affected in FAW fed on potato and tobacco. Our results support the preference–performance hypothesis, i.e., that herbivore females maximize fitness by choosing host plants associated with strong larval performance. Although larvae and adults performed poorly on potato and tobacco, their offspring will be capable of establishing populations on them, posing a potential threat to these crops in China.This article is protected by copyright. Investigation of the fall armyworm (Spodoptera frugiperda) gut microbiome and entomopathogenic fungus-induced pathobiome

Abstract:The gut microflora plays an important role in insect development and physiology. The gut bacterial microbiome of the fall armyworm (FAW), Spodoptera frugiperda, in both cornfield and laboratory-reared populations was investigated using a 16S metagenomic approach. The alpha- and beta-diversity of the cornfield FAW populations varied among sampling sites and were higher than those of the laboratory-reared FAW population, indicating that different diets and environments influence the gut bacterial composition. To better understand the interaction between the microbiome and entomopathogenic fungi (EPF), FAWs from organic and conventionally managed corn fields and from the laboratory-reared colony were inoculated with Beauveria bassiana NCHU-153 (Bb-NCHU-153). A longer median lethal time (LT50) was observed in the Bb-NCHU-153-infected cornfield FAW population than in the laboratory-reared FAWs. In terms of the microbiome, three Bb-NCHU-153-infected FAW groups showed different gut bacterial compositions compared to noninfected FAW. Further investigation of the cooccurrence network and linear discriminant analysis (LDA) of effect size (LEfSe) revealed that the enriched bacterial genera, such as Enterococcus, Serratia, Achromobacter, and Tsukamurella, in the gut might play the role of opportunistic pathogens after fungal infection; in contrast, some gut bacteria of Methylobacterium, Marinomonas, Paenochrobactrum, Pseudomonas, Acinetobacter, Delftia, Dietzia, Gordonia, Leucobacter, Paracoccus, and Stenotrophomonas might be probiotics against EPF infection. These results indicated that EPF infection can change the gut bacterial composition and lead to a pathobiome in the FAW and that some bacterial species might protect the FAW from EPF infection. These findings could be applied to the design of pathobiome-inducing biocontrol strategies.

Abstract:Although the role of nucleotides in the catalytic cycle of the GroESL chaperonin system has been extensively studied, the molecular effects of nucleotides in modulating exposure of sites on GroEL has not been thoroughly investigated. We report here that nucleotides (ATP, ADP, or adenosine 5′-(β,-imino)triphosphate) in the presence of Mg make the oligomer selectively sensitive to trypsin proteolysis in a fashion suggesting conformational changes in the monomers of one heptameric ring. The site of proteolysis in the monomer that is exposed upon nucleotide binding by the oligomer is in the apical domain (Arg-268). Further, complexes of GroEL with GroES or rhodanese display the same sensitivity to proteolysis, unlike the GroEL-GroES-rhodanese complex, which is protected from proteolysis. The influence of various cations on trypsin proteolysis is investigated to elucidate the differential effects that monovalent and divalent cations have on the oligomeric structure of the chaperonin. These results are discussed in relation to the molecular basis for the chaperonin activity.
Negative effects on the development of Chrysodeixis includens and Spodoptera cosmioides fed by peanut plants inoculated with entomopathogenic fungi

Abstract:Recent studies have shown that entomopathogenic fungi, as endophytes, can have beneficial effects on plants, protecting them from defoliating insects. The potential of endophytic association by entomopathogenic fungi with the peanut crop has been little explored. In our study, we conducted experiments by inoculation of peanut seeds through a soil drench method with nine strains/species of entomopathogenic fungi of the genera Metarhizium, Beauveria and Cordyceps, subsequently these plants were consumed by two larval pests, Chrysodeixis includens and Spodoptera cosmioides. The parameters of larval growth rates, mortality, foliar consumption and larval period were observed during the development of larvae. In addition, the endophytic capacity of these fungi in peanut plants and their persistence in soil were investigated. In two replicate greenhouse trials for each larva, peanut plants were inoculated with fungi by the soil-drench method. We evaluated the performance of C. includens and S. cosmioides feeding on inoculated peanut plants starting at the 2nd larval instar. The larval and pupal weights of C. includens and S. cosmioides were significantly different among the fungal treatment groups, where insects feeding on control plants exhibited higher larval and pupal weights than insects feeding on treated plants. The differences in larval period showed that Control larvae pupated faster than the larvae fed on fungal-inoculated plants, fungal treatments had a larval period of 3 to 5 days more than the control. The mortality rates of C. includens and S. cosmioides were significantly different among the fungal treatment groups, insects fed on Control plants exhibited higher survival than insects fed on fungal-inoculated plants. The persistence of all Metarhizium fungi was higher in the soil compared to other fungi, and only Metarhizium and B. bassiana IBCB215 emerged from the phyllosphere of peanut plants. Although the fungus Cordyceps presented the worst performance among the fungal treatments. Overall, our results demonstrate the negative effects on the development of C. includens and S. cosmioides that were fed on fungal-inoculated peanut plants, the best results recorded were for Metarhizium strains and the fungus B. bassiana IBCB215.
Comparative toxicity and enzymatic detoxification responses in Spodoptera frugiperda (Lepidoptera: Noctuidae) to two insecticides

Abstract:The fall armyworm (FAW), Spodoptera frugiperda Smith (Lepidoptera: Noctuidae), poses a significant threat to food security, necessitating effective management strategies. While chemical control remains a primary approach, understanding the toxicity and detoxification mechanisms of different insecticides is crucial. In this study, we conducted leaf-dipping bioassays to assess the toxicity of quinalphos and beta-cypermethrin·emamectin benzoate (β-cyp·EMB) on S. frugiperda larvae. Additionally, we assessed the response of alterations in CarE, GST, MFO, and AChE activities to sublethal concentrations of these insecticides over various treatment durations. Results indicated that β-cyp·EMB exhibited higher toxicity than quinalphos in S. frugiperda. Interestingly, the highest activities of GST, CarE, MFO, and AChE were observed at 6 h exposure to LC10 and LC25 of β-cyp·EMB, surpassing equivalent sublethal concentrations of quinalphos. Subsequently, GST and CarE activities exposure to β-cyp·EMB steadily decreased, while MFO and AChE activities exposure to both insecticides was initially decreased then increased. Conversely, two sublethal concentrations of quinalphos notably enhanced GST activity across all exposure durations, with significantly higher than β-cyp·EMB at 12-48 h. Similarly, CarE activity was also increased at various durations. Our research has exhibited significant alterations in enzyme activities exposure to both concentration and duration. Furthermore, Pearson correlation analysis showed significant correlations among these enzyme activities at different treatment durations. These findings contribute to a better understanding of detoxification mechanisms across different insecticides, providing valuable insights for the rational management of S. frugiperda populations.

Abstract:Serial passaging of Semlike Forest virus in BHK cells at a constant input multiplicity of 50 p.f.u./cell resulted in a 4 log10 drop in yield of infectious virus by passage 9. An interference analysis showed that this drop was due to the presence of defective-interfering (DI) particles. Attempts were made to separate the DI particles from standard virus by equilibrium and velocity centrifugation. Only equilbrium centrifugation on CsCl resolved the DI particles (identified by interference analyses) from standard virus. The buoyant density of the DI particles (1.23 g/ml) was higher than that of standard virus (p = 1-20 g/ml). No difference was observed between the structural porteins of standard virus and DI particles. Analysis of the RNA of standard virus and DI particles showed that whereas standard virus contained only 42S RNA (mol. wt. approx. 4-2 X 10(6), DI particles contained two small pieces of RNA of mol. wt. 0.81 and 0-75 X 10(6) respectively. Infectivity assays showed that these low mol. wt. species were not only non-infectious but also interfered with the infectivity of 42S RNA from standard virus. Nucleocapsids derived from purified DI particles had a buoyant density 0-02 g/ml greater than the nucleocapsids from standard virus. Analysis of the RNA from DI nucleocapsids showed it to be entirely of the low mol. wt. class. Metagenomic profiling of gut microbiota in Fall Armyworm (Spodoptera frugiperda) larvae fed on different host plants

Abstract:Background: The fall armyworm (FAW, Spodoptera frugiperda) is a polyphagous pest known for causing significant crop damage. The gut microbiota plays a pivotal role in influencing the biology, physiology and adaptation of the host. However, understanding of the taxonomic composition and functional characteristics of the gut microbiota in FAW larvae fed on different host plants remains limited. Methods: This study utilized metagenomic sequencing to explore the structure, function and antibiotic resistance genes (ARGs) of the gut microbiota in FAW larvae transferred from an artificial diet to four distinct host plants: maize, sorghum, tomato and pepper. Results: The results demonstrated significant variations in gut microbiota structure among FAW larvae fed on different host plants. Firmicutes emerged as the dominant phylum, with Enterococcaceae as the dominant family and Enterococcus as the prominent genus. Notably, Enterococcus casseliflavus was frequently observed in the gut microbiota of FAW larvae across host plants. Metabolism pathways, particularly those related to carbohydrate and amino acid metabolism, played a crucial role in the adaptation of the FAW gut microbiota to different host plants. KEGG orthologs associated with the regulation of the peptide/nickel transport system permease protein in sorghum-fed larvae and the 6-phospho-β-glucosidase gene linked to glycolysis/gluconeogenesis as well as starch and sucrose metabolism in pepper-fed larvae were identified. Moreover, the study identified the top 20 ARGs in the gut microbiota of FAW larvae fed on different host plants, with the maize-fed group exhibiting the highest abundance of vanRC. Conclusions: Our metagenomic sequencing study reveals significant variations in the gut microbiota composition and function of FAW larvae across diverse host plants. These findings underscore the intricate co-evolutionary relationship between hosts and their gut microbiota, suggesting that host transfer profoundly influences the gut microbiota and, consequently, the adaptability and pest management strategies for FAW.
Feeding experience on hairy vetch induces a loss of host-specific adaptation to maize and changes in the gut microbiota communities of the fall armyworm

Abstract:Developing ecological control technologies against the fall armyworms (FAWs) is crucial. The ecological role of green manure crops has gained attention for their indirect contribution to enhancing crop resistance against specific pests and diseases in rotation systems. We hypothesize that green manure application directly affects FAWs host utilization. By using two-sex life tables, we measured the host adaptation of FAWs to hairy vetch and their offspring's adaptation to the original host (maize) as breeding generations increased during larval stages fed on hairy vetch. When FAWs' adaptability to maize was significantly affected by feeding on hairy vetch, we used 16s rDNA sequencing to detect the gut microbiota composition of current generation of FAW populations that fed continuously on either hairy vetch or maize. Our results suggest that continuous feeding of FAWs on hairy vetch does not enhance their adaptation to the host plant across generations, indicating no increased risk of damage to hairy vetch in maize-hairy vetch rotation. As successive generations of FAWs fed on hairy vetch, their host-specific adaptation to maize decreased or even lost, which was shown by a decrease in pupal weight and fecundity, as well as an inability of emergent adults in completing mating. FAWs' gut microbiota exhibited lower diversity when fed on hairy vetch compared to maize. Cyanobacteria were not detected in "Hairy_vetch" group, while Oxyphotobacteria_unclassified , Acinetobacter , Pseudomonas , Serratia, and Klebsiella showed significantly differences in abundance between the "Maize" and "Hairy_vetch" groups. Additionally, gut microbial taxa and FAWs performance were significantly correlated. Overall, the feeding experience of FAWs on hairy vetch resulted in a reduced or was lost maize-specific adaptation, as shown through changes in gut microbiota. Our study provides experimental evidence for the ecological role of maize-green manure rotation systems in pest management.

Abstract:The fall armyworm (FAW), Spodoptera frugiperda, has become one of the most important pests on corn in China since it invaded in 2019. Although FAW has not been reported to cause widespread damage to rice plants in China, it has been sporadically found feeding in the field. If FAW infests rice in China, the fitness of other insect pests on rice may be influenced. However, how FAW and other insect pests on rice interact remains unknown. In this study, we found that the infestation of FAW larvae on rice plants prolonged the developmental duration of the brown planthopper (BPH, Nilaparvata lugens (Stål)) eggs and plants damaged by gravid BPH females did not induce defenses that influenced the growth of FAW larvae. Moreover, co-infestation by FAW larvae on rice plants did not influence the attractiveness of volatiles emitted from BPH-infested plants to Anagrus nilaparvatae, an egg parasitoid of rice planthoppers. FAW larvae were able to prey on BPH eggs laid on rice plants and grew faster compared to those larvae that lacked available eggs. Studies revealed that the delay in the development of BPH eggs on FAW-infested plants was probably related to the increase in levels of jasmonoyl-isoleucine, abscisic acid and the defensive compounds in the rice leaf sheaths on which BPH eggs were laid. These findings indicate that, if FAW invades rice plants in China, the population density of BPH may be decreased by intraguild predation and induced plant defenses, whereas the population density of FAW may be increased.
Effectiveness of Entomopathogenic Fungi on Immature Stages and Feeding Performance of Fall Armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae) Larvae

Abstract:Maize is a major staple crop in China, and the sustainable productivity of this primary crop has been recently threatened by fall armyworm (FAW), Spodoptera frugiperda, invasion. The five fungal isolates, Aspergillus sp. BM-3 and SE-2-1, Cladosporium tenuissimum SE-10, Penicillium citrinum CTD-24, and Beauveria bassiana ZK-5 were assessed for their efficacy in causing mortality against first to sixth instar eggs and neonate larvae seven days post-treatment, and their effects on the feeding performance of sixth instar S. frugiperda larvae at 48 h post-treatment at three concentrations (1 × 106, 1 × 107, and 1 × 108 conidia mL−1) were also assessed. The six instar S. frugiperda larvae were not susceptible to the five tested fungal isolates. However, B. bassiana ZK-5 caused the highest egg mortality of 40, 70, and 85.6% at 1 × 106, 1 × 107, and 1 × 108 conidia mL−1, respectively, followed by P. citrinum CTD-24 (30.6, 50, and 75.6%) and C. tenuissimum SE-10 (25.6, 40, and 55.6%). In addition, B. bassiana ZK-5 caused the highest neonate mortality of 54.3% at 1 × 108 conidia mL−1. B. bassiana ZK-5 and P. citrinum CTD-24 caused cumulative mortality, including 93.3 and 83.3% mortality of eggs and neonates, respectively, at 1 × 108 conidia mL−1. Furthermore, B. bassiana ZK-5 reduced the feeding efficacy of first to third instar S. frugiperda larvae by 66.7 to 78.6%, while P. citrinum CTD-24 and C. tenuissimum SE-10 reduced larval feeding by 48.3 to 57.1% at 1 × 108 conidia mL−1. However, these fungal isolates were less potent in reducing the feeding activity of fourth to sixth instar S. frugiperda larvae (>46% with B. bassiana at 48 h post-treatment). The tested fungal isolates could play an essential role as microbial biopesticides in suppressing the S. frugiperda population in China after further investigations on their efficacy are obtained in the field.

Abstract:Introduction: Spodoptera frugiperda is an important nomadic agricultural pest with a diverse host range and resistance against several insecticides. The current study investigated the life history traits of two strains of the field-collected population against chlorantraniliprole using an age-stage two-sex life table. Method: For this, we established the chlorantraniliprole-susceptible (Crp-SUS G 12 ), and chlorantraniliprole-reduced susceptible (Crp-RES G 12 ) strains derived from the sixth generation of the QJ-20 population having a resistance ratio (RR) of 10.39-fold, compared with the reported susceptible population. Results: The results showed that the chlorantraniliprole-reduced susceptible strain attained a 4.0-fold RR, while the chlorantraniliprole-susceptible strain attained an RR of 0.85-fold, having overlapped fiducial limits (FLs) with the referred susceptible baseline. Meanwhile, the present study revealed that the development time of the susceptible strain was significantly longer than that of the reduced susceptible strain. Similarly, the mean longevity, adult pre-oviposition period (APOP), and total pre-oviposition period (TPOP) of the female chlorantraniliprole-susceptible strain were considerably longer than those of the female chlorantraniliprole-reduced susceptible strain. Contrarily, the population parameters, including the intrinsic rate of increase ( r ), finite rate of increase ( λ ), and net reproductive rate ( R ), of the chlorantraniliprole-susceptible strain were considerably lower than those of the chlorantraniliprole-reduced susceptible strain, while the mean generation time ( T ) of the chlorantraniliprole-susceptible strain was substantially longer than the chlorantraniliprole-reduced susceptible strain. The age-stage characteristic survival rate ( s xj ) and age-stage characteristic life expectancy ( e xj ) of the chlorantraniliprole-susceptible strain were longer than those of the chlorantraniliprole-reduced susceptible strain, but the age-stage-specific reproductive value ( v xj ) of the chlorantraniliprole-susceptible strain was shorter than that of the chlorantraniliprole-reduced susceptible strain. Moreover, the contents of vitellogenin (Vg) and VgR in the chlorantraniliprole-reduced susceptible strain were higher than those in the chlorantraniliprole-susceptible strain. Discussion: These findings showed that reducing susceptibility to chlorantraniliprole promoted population growth in S. frugiperda . Therefore, this study could provide conceptual support for the integrated pest management (IPM) approach to control S. frugiperda in the field.

Abstract:As an agricultural pest, the fall armyworm (FAW), Spodoptera frugiperda, poses a severe threat to agriculture in China. Chlorantraniliprole has been widely used to control this pest. In our previous studies, we discovered that LD10, LD20, and LD30 chlorantraniliprole promoted encapsulation in the 4th instar larvae of the FAW, with LD30 chlorantraniliprole having the most significant effect. To further investigate the molecular mechanism underlying the sublethal effects of chlorantraniliprole on encapsulation in the FAW, this study conducted the effects of encapsulation in 4th instar larvae of the FAW exposed to LD30 chlorantraniliprole. Then, we analyzed the transcriptome of the FAW hemolymph treated with LD30 chlorantraniliprole and identified genes related to encapsulation using RNAi. Our results showed that the encapsulation in the FAW was enhanced at 6, 12, 18, 24, and 48 h after exposure to LD30 chlorantraniliprole. Additionally, LD30 chlorantraniliprole significantly affected the expression of certain immune-related genes, with the heat shock protein 70 family gene SfHSP68.1 showing the most significant upregulation. Subsequent interference with SfHSP68.1 resulted in a significant inhibition of encapsulation in FAW. These findings suggested that LD30 chlorantraniliprole can promote encapsulation in the FAW by upregulating SfHSP68.1 expression. This study provides valuable insights into the sublethal effects of chlorantraniliprole on encapsulation in the FAW and the interaction between encapsulation and heat shock proteins (HSPs).
