Abstract:Summary The neural mechanisms underlying novelty detection are not well understood, especially in relation to behavior. Here, we present single-unit responses from the primary auditory cortex (A1) from two monkeys trained to detect deviant tones amid repetitive ones. Results show that monkeys can detect deviant sounds, and there is a strong correlation between late neuronal responses (250–350 ms after deviant onset) and the monkeys' perceptual decisions. The magnitude and timing of both neuronal and behavioral responses are increased by larger frequency differences between the deviant and standard tones and by increasing the number of standard tones preceding the deviant. This suggests that A1 neurons encode novelty detection in behaving monkeys, influenced by stimulus relevance and expectations. This study provides evidence supporting aspects of predictive coding in the sensory cortex.
Delay‐period Activities in Two Subdivisions of Monkey Inferotemporal Cortex During Pair Association Memory Task

Abstract:The macaque inferotemporal cortex, which is involved in encoding and retrieval of visual long-term memory, consists of two distinct but mutually interconnected areas: area TE (TE) and area 36 (A36). In the present study, we compared delay-period activities of the two subdivisions in terms of their signal contents. We recorded single-unit activities from TE and A36 during a delayed pair association task, in which monkeys were required to choose the paired associate of a cue stimulus after a delay period. The stimulus-selective delay-period activities of single neurons were characterized by using partial correlation coefficients of delay-period activities for each cue stimulus with the cue-period responses to that stimulus (cue-holding index, CHI) and with the cue-period responses to its paired associate (pair-recall index, PRI). The delay-period activities of TE neurons preferentially represented the paired associate (PRI, median = 0.54) rather than the cue stimulus itself (CHI, 0.23) (P < 0.001, n = 70), while the delay-period activities of A36 neurons retained both the cue stimulus and its paired associate equivalently (CHI, 0.44; PRI, 0.46) (P = 0.78, n = 38). These results indicate that the signal contents of delay-period activities differ between the two subdivisions: TE mostly represents a sought target that is retrieved from long-term memory, while A36 in addition retains cue-stimulus that is transmitted from earlier visual areas.
Forward processing of long-term associative memory in monkey inferotemporal cortex.

Abstract:The macaque inferotemporal (IT) cortex, which serves as the storehouse of visual long-term memory, consists of two distinct but mutually interconnected areas: area TE (TE) and area 36 (A36). In the present study, we tested whether memory encoding is put forward at this stage, i.e., whether association between the representations of different but semantically linked objects proceeds forward from TE to A36. To address this question, we trained monkeys in a pair-association (PA) memory task, after which single-unit activities were recorded from TE and A36 during PA trials. Neurons in both areas showed stimulus-selective cue responses (347 in TE, 76 in A36; "cue-selective neurons") that provided, at the population level, mnemonic linkage between the paired associates. The percentage of neurons in which responses to the paired associates were significantly (p < 0.01) correlated at the single-neuron level ("pair-coding neuron") dramatically increased from TE (4.9% of the cue-selective neurons) to A36 (33%). The pair-coding neurons in A36 were further separable into Type1 (68%) and Type2 (32%) on the basis of their initial transient responses after cue stimulus presentation. Type1 neurons, but not Type2 neurons, began to encode association between paired stimuli as soon as they exhibited stimulus selectivity. Thus, the representation of long-term memory encoded by Type1 neurons in A36 is likely substantiated without feedback input from other higher centers. Therefore, we conclude that association between the representations of the paired associates proceeds forward at this critical step within IT cortex, suggesting selective convergence onto a single A36 neuron from two TE neurons that encode separate visual objects.
The Effect of Learning on the Face Selective Responses of Neurons in the Cortex in the Superior Temporal Sulcus of the Monkey.

Abstract:Neurophysiological studies have shown that some neurons in the cortex in the superior temporal sulcus and the inferior temporal gyrus of macaque monkeys respond to faces. These neurons provided a consistently identifiable substrate with which studies of the storage of visual information were performed. To determine whether face responsive neurons change how much they respond to different novel faces as they become familiar, neurons were tested with two experimental designs. In the first experiment, 22 neurons were tested on their responsiveness to the different members of a large set of novel faces as the set was presented repeatedly until the faces became familiar. 6 neurons altered the relative degree to which they responded to the different members of the set between the first two presentations and subsequent presentations. In a control condition, only 1 out of 17 neurons showed a significant response difference between the first two presentations and subsequent presentations when the experiment started with faces which were already familiar to the monkey. In the second experiment, 26 neurons were tested on their responsiveness to the different members of a set of familiar faces before and after the addition of a novel face to the set. 5 neurons altered the relative degree in which they responded to the different members of the set of familiar faces after addition of a novel face. It is suggested that these changes in neuronal responsiveness to different stimuli reflect the setting up of an ensemble encoded representation of face stimuli. This alteration of neuronal responsiveness as novel faces become familiar suggests that face responsive neurons may store information useful in visual recognition.
Backward Signal from Medial Temporal Lobe in Neural Circuit Reorganization of Primate Inferotemporal Cortex

Abstract:Neuropsychological theories proposed a critical role of the interaction between the medial temporal lobe and neocortex in the formation of long-term memory for facts and events, which has often been tested by learning of a series of paired words or figures in humans. We identify neural mechanisms of this long-term memory formation process by single-unit recording and molecular biological methods in an animal model of visual pair-association task in monkeys. In our previous studies, we found a group of neurons that manifested selective responses to both of the paired associates (pair-coding neuron) in the anterior inferior temporal (IT) cortex. It provides strong evidence that single IT neurons acquire the response-selectivity through associative learning, and suggests that the reorganized neural circuits for the pair-coding neurons serve as the memory engram of the pair-association learning. In this article, we investigated further mechanisms of the neural circuit reorganization. First, we tested the role of the backward connections from the medial temporal lobe to IT cortex. lbotenic acid was injected unilaterally into the entorhinal and perirhinal cortex which provided massive backward projections ipsilaterally to IT cortex. We found that the limbic lesion disrupted the associative code of the IT neurons between the paired associates, without impairing the visual response to each stimulus. Second, we ask why the limbic-neocortical interactions are so important. We hypothesize that limbic neurons would undergo rapid modification of synaptic connectivity and provide backward signals that guide reorganization of neocortical neural circuits. We then investigated the molecular basis of such rapid synaptic modifiability by detecting the expression of immediate-early genes. We found strong expression of zif268 during the learning of a new set of paired associates, most intensively in area 36 of the perirhinal cortex. All these results with visual pair-association task support our hypothesis, and demonstrate that the ‘consolidation’ process, which was first proposed on the basis of clinico-psychological evidence, can now be examined in the primate with neurophysiolocical and molecular biological approaches.
The Spatial Frequency Representation Predicts Category Coding in the Inferior Temporal Cortex

Abstract:Understanding the neural representation of spatial frequency (SF) in the primate cortex is vital for unraveling visual processing mechanisms in object recognition. While numerous studies concentrate on the representation of SF in the primary visual cortex, the characteristics of SF representation and its interaction with category representation remain inadequately understood. To explore SF representation in the inferior temporal (IT) cortex of macaque monkeys, we conducted extracellular recordings with complex stimuli systematically filtered by SF. Our findings disclose an explicit SF coding at single-neuron and population levels in the IT cortex. Moreover, the coding of SF content exhibits a coarse-to-fine pattern, declining as the SF increases. Temporal dynamics analysis of SF representation reveals that low SF (LSF) is decoded faster than high SF (HSF), and the SF preference dynamically shifts from LSF to HSF over time. Additionally, the SF representation for each neuron forms a profile that predicts category selectivity at the population level. IT neurons can be clustered into four groups based on SF preference, each exhibiting different category coding behaviors. Particularly, HSF-preferred neurons demonstrate the highest category decoding performance for face stimuli. Despite the existing connection between SF and category coding, we have identified uncorrelated representations of SF and category. In contrast to the category coding, SF is more sparse and places greater reliance on the representations of individual neurons. Comparing SF representation in the IT cortex to deep neural networks, we observed no relationship between SF representation and category coding. However, SF coding, as a category-orthogonal property, is evident across various ventral stream models. These results dissociate the separate representations of SF and object category, underscoring the pivotal role of SF in object recognition.

Abstract:Functional magnetic resonance imaging (fMRI) has been used extensively to identify regions in the inferior temporal (IT) cortex that are selective for categories of visual stimuli. However, comparatively little is known about the neuronal responses relative to these fMRI-defined regions. Here, we compared in nonhuman primates the distribution and response properties of IT neurons recorded within versus outside fMRI regions selective for four different visual categories: faces, body parts, objects, and places. Although individual neurons that preferred each of the four categories were found throughout the sampled regions, they were most concentrated within the corresponding fMRI region, decreasing significantly within 1-4 mm from the edge of these regions. Furthermore, the correspondence between fMRI and neuronal distributions was specific to neurons that increased their firing rates in response to the visual stimuli but not to neurons suppressed by visual stimuli, suggesting that the processes associated with inhibiting neuronal activity did not contribute strongly to the fMRI signal in this experiment.

Abstract:There are populations of neurons in the temporal lobe of the macaque which appear to be specialised to respond to faces. These neurons are relatively rare in the inferior temporal visual cortex, area TE, but are more common in the cortex in the middle and anterior parts of the superior temporal sulcus. In some cases these neurons can respond to parts of faces such as the eyes, and in other cases a fuller complement of the features present in faces, with even the correct configuration of these features, appears to be necessary for these neurons to respond. In experiments to investigate whether the responses of these neurons would be useful for recognition of an individual, it has been shown that in many cases (77% of one sample), these neurons are sensitive to differences between faces. In experiments designed to investigate the information required for these neurons to respond, it has been shown that these neurons can respond to blurred images of faces, and to faces with only the high spatial frequencies included. A population of neurons with face-selective responses has also been found in the amygdala, which receives inputs from the temporal lobe visual cortex. In many cases these neurons respond differently to different faces. It is suggested that the importance of this type of neuron may be in the different emotional and social responses which occur to different faces, and which are disrupted by damage to the amygdala.

Abstract:Current studies investigating electroencephalogram correlates associated with categorization of sensory stimuli (P300 event-related potential, alpha event-related desynchronization, theta event-related synchronization) typically use an oddball paradigm with few, familiar, highly distinct stimuli providing limited insight about the aspects of categorization (e.g. difficulty, membership, uncertainty) that the correlates are linked to. Using a more complex task, we investigated whether such more specific links could be established between correlates and learning and how these links change during the emergence of new categories. In our study, participants learned to categorize novel stimuli varying continuously on multiple integral feature dimensions, while electroencephalogram was recorded from the beginning of the learning process. While there was no significant P300 event-related potential modulation, both alpha event-related desynchronization and theta event-related synchronization followed a characteristic trajectory in proportion with the gradual acquisition of the two categories. Moreover, the two correlates were modulated by different aspects of categorization, alpha event-related desynchronization by the difficulty of the task, whereas the magnitude of theta -related synchronization by the identity and possibly the strength of category membership. Thus, neural signals commonly related to categorization are appropriate for tracking both the dynamic emergence of internal representation of categories, and different meaningful aspects of the categorization process.

Midbrain activity supports high-level visual properties in primate temporal cortex

Abstract:Abstract The evolution of the primate brain is marked by a dramatic increase in the number of neocortical areas that process visual information 1 . This cortical expansion supports two hallmarks of high-level primate vision – the ability to selectively attend to particular visual features 2 and the ability to recognize a seemingly limitless number of complex visual objects 3 . Given their prominent roles in high-level vision for primates, it is commonly assumed that these cortical processes supersede the earlier versions of these functions accomplished by the evolutionarily older brain structures that lie beneath the cortex. Contrary to this view, here we show that the superior colliculus (SC), a midbrain structure conserved across all vertebrates 4 , is necessary for the normal expression of attention-related modulation and object selectivity in a newly identified region of macaque temporal cortex. Using a combination of psychophysics, causal perturbations and fMRI, we identified a localized region in the temporal cortex that is functionally dependent on the SC. Targeted electrophysiological recordings in this cortical region revealed neurons with strong attention-related modulation that was markedly reduced during attention deficits caused by SC inactivation. Many of these neurons also exhibited selectivity for particular visual objects, and this selectivity was also reduced during SC inactivation. Thus, the SC exerts a causal influence on high-level visual processing in cortex at a surprisingly late stage where attention and object selectivity converge, perhaps determined by the elemental forms of perceptual processing the SC has supported since before there was a neocortex.

Abstract:The activity of neurones in the inferotemporal cortex of the alert rhesus monkey was recorded while the monkey was shown visual stimuli, which included both food and non-food objects for comparison with the activity of neurones in the lateral hypothalamus and substantia innominata. In the anteroventral part of the inferotemporal cortex, neurones were found with visual responses which were sustained while the animal looked at the appropriate visual stimuli. The latency of the responses was 100 msec or more. The majority (96/142 or 68%) of these neurones responded more strongly to some stimuli than to others. These units usually had different responses when objects were shown from different views, and physical factors such as shape, size, orientation, colour and texture appeared to account for the responses of some of these units. Association of visual stimuli with a food reward (glucose solution) or an aversive taste (5% saline solution) did not affect the magnitude of the responses of the neurones to the stimuli either during the learning or after the period of learning. Nor did feeding the monkey to satiety affect the responses of the neurones to their effective stimuli.
Bottom-up saliency and top-down learning in the primary visual cortex of monkeys.

Abstract:Early sensory cortex is better known for representing sensory inputs but less for the effect of its responses on behavior. Here we explore the behavioral correlates of neuronal responses in primary visual cortex (V1) in a task to detect a uniquely oriented bar-the orientation singleton-in a background of uniformly oriented bars. This singleton is salient or inconspicuous when the orientation contrast between the singleton and background bars is sufficiently large or small, respectively. Using implanted microelectrodes, we measured V1 activities while monkeys were trained to quickly saccade to the singleton. A neuron's responses to the singleton within its receptive field had an early and a late component, both increased with the orientation contrast. The early component started from the outset of neuronal responses; it remained unchanged before and after training on the singleton detection. The late component started ∼40 ms after the early one; it emerged and evolved with practicing the detection task. Training increased the behavioral accuracy and speed of singleton detection and increased the amount of information in the late response component about a singleton's presence or absence. Furthermore, for a given singleton, faster detection performance was associated with higher V1 responses; training increased this behavioral-neural correlate in the early V1 responses but decreased it in the late V1 responses. Therefore, V1's early responses are directly linked with behavior and represent the bottom-up saliency signals. Learning strengthens this link, likely serving as the basis for making the detection task more reflexive and less top-down driven.

Abstract:The current understanding of sensory and motor cortical areas has been defined by the existence of topographical maps across the brain surface, however, higher cortical areas, such as the prefrontal cortex, seem to lack an equivalent organization, and only limited evidence of functional clustering of neurons with similar stimulus properties is evident in them. We thus sought to examine whether neurons that represent similar spatial and object information are clustered in the monkey prefrontal cortex and whether such an organization only emerges as a result of training. To this end, we analyzed neurophysiological recordings from male macaque monkeys before and after training in spatial and shape working memory tasks. Neurons with similar spatial or shape selectivity were more likely than chance to be encountered at short distances from each other. Some aspects of organization were present even in naïve animals, however other changes appeared after cognitive training. Our results reveal that prefrontal microstructure automatically supports orderly representations of spatial and object information.

Abstract:How is the representation of complex visual objects organized in inferotemporal (IT) cortex, the large brain region responsible for object recognition? Areas selective for a few categories such as faces, bodies, and scenes have been found, but the vast majority of IT is “wild,” lacking any known specialization, leading to uncertainty over whether any general principle governs IT organization. We recorded responses of IT cells in macaque monkeys to a set of objects, and built a low dimensional object space to describe these objects using a deep network Alexnet, by performing principal component analysis on the responses of fc6 layer. We found that responses of single IT cells could be well-modeled by projection onto specific axes of this object space. Remarkably, cells were spatially clustered into networks according to their preferred axes, and this clustering was determined by the topography of object space. Furthermore, this topography was replicated across at least three hierarchical stages of increasing view invariance. Finally, pooling cells across this newly identified object-topic map allowed reconstruction of general objects. Taken together, these results provide a unified picture of IT organization in which category-selective regions are part of a topographic continuum.
Category learning can alter perception and its neural correlates

Abstract:Learned Categorical Perception (CP) occurs when the members of different categories come to look more dissimilar (between-category separation) and/or members of the same category come to look more similar (within-category compression) after a new category has been learned. To measure learned CP and its physiological correlates we compared dissimilarity judgments and Event Related Potentials (ERPs) before and after learning to sort multi-featured visual textures into two categories by trial and error with corrective feedback. With the same number of trials and feedback, about half the participants succeeded in learning the categories (learners: criterion 80% accuracy) and the rest did not (non-learners). At both lower and higher levels of difficulty, successful learners showed significant between-category separation in pairwise dissimilarity judgments after learning compared to before; their late parietal ERP positivity (LPC, usually interpreted as decisional) also increased and their occipital negativity (N1) (usually interpreted as perceptual) decreased. LPC increased with response accuracy and N1 amplitude decreased with between-category separation for the Learners. Non-learners showed no significant changes in dissimilarity judgments, LPC or N1, within or between categories. This is behavioral and physiological evidence that category learning can alter perception. We sketch a neural net model for this effect.

Abstract:The superior colliculus is an evolutionarily conserved midbrain region that is thought to mediate spatial orienting, including saccadic eye movements and covert spatial attention. Here, we reveal a role for the superior colliculus in higher-order cognition, independent of its role in spatial orienting. We trained rhesus macaques to perform an abstract visual categorization task that involved neither instructed eye movements nor differences in covert attention. We compared neural activity in the superior colliculus and the posterior parietal cortex, a region previously shown to causally contribute to abstract category decisions. The superior colliculus exhibits robust encoding of learned visual categories, which is stronger than in the posterior parietal cortex and arises at a similar latency in the two areas. Moreover, inactivation of the superior colliculus markedly impaired animals' category decisions. These results demonstrate that the primate superior colliculus mediates abstract, higher-order cognitive processes that have traditionally been attributed to the neocortex.
Formation of brain-wide neural geometry during visual item recognition in monkeys

Abstract:Neural dynamics reflect canonical computations that relay and transform information in the brain. Previous studies have identified the neural population dynamics in many individual brain regions as a trajectory geometry in a low-dimensional neural space. However, whether these populations share particular geometric patterns across brain-wide neural populations remains unclear. Here, by mapping neural dynamics widely across temporal/frontal/limbic regions in the cortical and subcortical structures of monkeys, we show that 10 neural populations, including 2,500 neurons, propagate visual item information in a stochastic manner. We found that the visual inputs predominantly evoked rotational dynamics in the higher-order visual area, the TE and its downstream striatum tail, while curvy/straight dynamics appeared more frequently downstream in the orbitofrontal/hippocampal network. These geometric changes were not deterministic but rather stochastic according to their respective emergence rates. These results indicated that visual information propagates as a heterogeneous mixture of stochastic neural population signals in the brain.

Neuronal Correlates of the Set-Size Effect in Monkey Lateral Intraparietal Area

Abstract:It has long been known that the brain is limited in the amount of sensory information that it can process at any given time. A well-known form of capacity limitation in vision is the set-size effect, whereby the time needed to find a target increases in the presence of distractors. The set-size effect implies that inputs from multiple objects interfere with each other, but the loci and mechanisms of this interference are unknown. Here we show that the set-size effect has a neural correlate in competitive visuo-visual interactions in the lateral intraparietal area, an area related to spatial attention and eye movements. Monkeys performed a covert visual search task in which they discriminated the orientation of a visual target surrounded by distractors. Neurons encoded target location, but responses associated with both target and distractors declined as a function of distractor number (set size). Firing rates associated with the target in the receptive field correlated with reaction time both within and across set sizes. The findings suggest that competitive visuo-visual interactions in areas related to spatial attention contribute to capacity limitations in visual searches.
