Abstract:N,N'-Substituted p-phenylenediamines (PPDs) are widely used as antioxidants in the rubber industry and are released into the environment in large quantities during the production and use of rubber products. We quantified PPDs and PPD quinone derivatives (PPD-Qs) in rubber consumer products, including car tires, rubber belts, rubber gloves, rubber cables, and rubber hoses, to obtain information on the degree of weathering over time during their use. Additionally, we investigated the occurrences and sources of PPDs and PPD-Qs in dust samples collected from four typical urban environments (roads, parking lots, automotive repair shops, and residences). The detected compounds included the highly toxic N-(1,3-dimethylbutyl)-N'-phenyl-1,4-phenylenediamine quinone, which can cause acute mortality of coho salmon (Oncorhynchus kisutch). Concentrations of PPDs in the automotive repair shops reached 56.0 μg/g, and were much higher than in the other environments, while the residential samples had the lowest contaminant concentrations. In road and residential samples, N,N'-di-2-naphthyl-p-phenylenediamine accounted for 17 %-30 % of the PPDs, and may have originated from different sources. We preliminarily identified 32 transformation products, and 11 of these were N,N'-di-2-naphthyl-p-phenylenediamine transformation products. The average daily intakes of PPDs and PPD-Qs were calculated to assess the health risks of dust exposure in each environment. Workers had high total intakes of PPD [60.3 ng/(kg day)] and PPD-Qs [20.1 ng/(kg day)], and were at some risk of occupational exposure. These results improve our understanding of the environmental occurrences, sources, transformation, and health risks of PPDs and PPD-Qs.
Plastic additives and personal care products in south China house dust and exposure in child-mother pairs

Abstract:Indoor environment constitutes an important source of industrial additive chemicals to human exposure. We hypothesized that the influence of residential environment on human exposure varies among different types of additive chemicals and differs between children and mothers. This study determined a suite of additive chemicals in house dust from South China dwellings (n = 47) and urine from child-mother pairs. Concentrations of phthalates (PAEs; median 601 μg/g) were 2 - 3 orders of magnitude greater than those of parabens (0.82 μg/g), bisphenols (3.31 μg/g), and benzophenone-related chemicals (2.69 μg/g). Urinary concentrations differed between children and mothers, but the pattern of differences varied between chemical groups. Children exhibited greater urinary levels of mono-PAEs than mothers (510 versus 395 ng/mL, p = 0.152), while the latter population exhibited greater levels of parabens and benzophenones. Regression analyses indicate a lack of association between dust and urinary levels for most chemicals, suggesting that other exposure pathways can complicate human exposure scenarios. Indeed, we estimated that the daily intake via dust ingestion only constituted 0.002 - 0.81% of total daily intake estimated based on urine data for mothers and 0.04 – 5.61% for children. Future efforts are needed to better characterize source-specific exposure for different populations.

Abstract:The wide use and continuous abrasion of rubber-related products appears to be leading to an incredible release of p-phenylenediamine (PPD) antioxidants in the environment. However, no related research has been conducted on the pollution characteristics and potential health risks of PM2.5-bound PPDs. We report for the first time the ubiquitous distributions of six emerging PPDs and a quinone derivative, N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPDQ), in PM2.5 from urban areas of China. Atmospheric contamination levels of PM2.5-bound PPDs were found to be mostly in pg m-3 amounts between 2018 and 2019. Urban vehicle rubber tire abrasion was found to probably contribute to the PPDs in PM2.5 and accounted for their significant spatiotemporal-dependent concentration variations. Furthermore, 6PPDQ, an emerging oxidation product of 6PPD in the environment, was first quantified (pg m-3) with a total detection rate of 81% in the urban PM2.5, demonstrating its broad existence. On the basis of the determined ambient concentrations, the annual intakes of PPDs and 6PPDQ for adults were not low, indicating their possible human health risks induced by long-term exposure. This study confirms the widespread occurrence of PPDs and 6PPDQ in PM2.5, showing that the pollution of such compounds in urban air should not be underestimated.

Abstract:Tire wear particles (TWPs) enter road surface with the friction between tires and road surfaces. Under the volatilization, leaching, and transformation action on TWPs by sunlight and rain, tire additives are released into urban water systems, such as surface rainfall runoff, wastewater treatment plants (WWTPs), receiving surface waters, and drinking water treatment plant (DWTP). In this study, we investigated the occurrence of 23 tire additives and their transformation products in the urban water system of the Pearl River Delta region, South China. Nineteen target compounds were detected in the surface runoff, with 1,3-Diphenylguanidine (DPG) showing highest maximum concentration of 58780 ng/L. Benzothiazole and its transformation products are detected at the frequency of 100% with the total concentrations of 480–42160 ng/L. The antioxidant derivative N-(1,3-dimethylbutyl)-N′-phenyl-p-phenylenediamine-quinone (6PPD-Q) was also detected up to 1562 ng/L, which was considerably higher than that of the parent compound 6PPD (the maximum concentration of 7.52 ng/L). Eleven and 8 compounds were detected in WWTPs influents and effluents, respectively, with removal rates of −62-100%. Seventeen compounds were detected in the receiving Zhujiang and Dongjiang rivers, while 9 compounds were detected in drinking water sources and DWTP samples. Road runoff, with total concentrations of target compounds up to 79200 ng/L, is suggested as the main non-point source for receiving rivers, while WWTPs effluents are the point sources due to incomplete removal of target compounds after accepting the initial runoff. 6PPD-Q and other 10 compounds displayed median to high ecological risks in surface waters, and the human daily intake of tire additives was estimated to be 2.63×10 −8 -3.16×10 −5 mg/(kg d) via drinking water. This is the first report of the 6PPD-Q and 1,3-Diphenylurea levels in surface waters in China.

Abstract:Aniline antioxidants (ANs) are widely used as industrial chemicals in products composed of rubber. ANs originate mainly from vehicles, where tire wear particles end up in dust and soil after being deposited on roads. Nowadays, limited information is available on the fate and seasonal variation of ANs in the road environment. In this study, we investigated the occurrence of 32 ANs in dust and soil from different road environments, including road dust, garage dust, parking lot dust, and green-belt soil. The total concentrations of ANs were 369 ng g-1 in road dust, 712 ng g-1 in garage dust, and 687 ng g-1 in parking lot dust. These concentrations are several times higher than that in green-belt soil (128 ng g-1). The highest concentrations of N-(1,3-dimethylbutyl)-N'-phenyl-1,4-phenylenediamine (6PPD) were found in dust and soil. Furthermore, notable seasonal differences were observed, with significantly higher concentrations of ANs in autumn than those in spring. In the main urban area, roads with high traffic volume exhibited higher concentrations of ANs than those with low traffic volume, indicating that ANs were produced by vehicle-related sources.

Abstract:There is increasing research focused on rubber additives, predominantly originating from tire and road wear particles. Other consumer products including sports equipment also contain rubber additives and the overall human exposure to these compounds is of concern due to demonstrated toxicity to animal species. Rubber additives are intentionally incorporated into climbing shoes for specific performance. We found high concentrations of rubber additives in shoe sole samples, aerosol particulate matter, and settled dust in indoor climbing halls. The estimated daily intake via inhalation for climbers and employees of these facilities exceeds the intake of rubber additives from all other known sources. Abrasion powder resulting from friction of climbing shoes on the holds is responsible for the high concentrations of rubber additives observed in aerosol particulate matter and settled dust, while other emission sources could be excluded. We also show that atmospheric transformation of rubber additives occurs in indoor environments. These findings identify a previously unknown human exposure route to rubber additives and emphasize the global problem of the toxicity burden of plastic additives.

Abstract:N , N ′-substituted p -phenylenediamines (PPDs) are widely used antioxidants in rubber tires, which could be released and accumulated in road dusts with rubber tires wear. As ozonation product of N -(1,3-dimethylbutyl)- N′ -phenyl- p -phenylenediamine (6PPD), 6PPD-quinone (6PPD-Q) exhibited higher toxicity to coho salmon. However, studies on their environmental behaviors are still limited. Road dust is the major medium PPDs exist, which significantly affects the levels of PPDs in other mediums, especially surface water and particulate matter. In this study, road dust samples were collected in 55 major cities of China to explore the distribution characteristics of PPDs and 6PPD-Q. The concentrations of total PPDs (ΣPPDs) and 6PPD-Q in urban trunk road dust samples were in the ranges of 7.90–727 and 3.00–349 ng/g, with median concentrations of 68 and 49 ng/g, respectively. 6PPD and 6PPD-Q are the dominant components in most road dusts. The functional region-dependent pollution characteristics of PPDs and 6PPD-Q give the first finding that urban tunnel road was the highly polluted region, followed by urban trunk roads. Suburban road dusts had a lower pollution level. Moreover, the estimated daily intake (EDI) of PPDs and 6PPD-Q for children was much higher than adults.

Abstract:Indoor dust has been extensively used for assessment of indoor contamination, especially for semi-volatile organic compounds (SVOCs). In the present study, the occurrence of four groups of SVOCs, i.e. organophosphorus flame retardants (PFRs), emerging PFRs (ePFRs), legacy phthalates (LPs), and alternative plasticizers (APs), was investigated in the indoor dust and air collected from floors, table surfaces, windows, and air conditioner (A/C) filters in bedrooms and offices in Guangzhou, South China. In bedrooms, A/C filter dust showed the highest median concentrations of PFRs (4670 ng/g) and ePFRs (586 ng/g), whilst the highest median concentrations of LPs and APs were found in floor (240,880 ng/g) and window dust (157,160 ng/g), respectively. In offices, A/C filter dust showed the highest median concentrations for PFRs (6750 ng/g) and APs (504,520 ng/g), while the highest ePFR median level was found in PC table dust (5810 ng/g) and LPs in floor dust (296,270 ng/g). Median air concentrations of PFRs, ePFRs, LPs, and APs were measured at 4.6, 0.12, 399, and 25 ng/m3 in bedrooms, and at 8.0, 0.05, 332, and 43 ng/m3 in offices, respectively. Tris(1-chloro-iso-propyl) phosphate (TCIPP) was the predominant PFRs/ePFRs in both dust and air. Di(2-ethylhexyl) phthalate (DEHP), di-iso-decyl phthalate (DIDP) and di-iso-nonyl phthalate (DINP) were the main LP/AP compounds in dust, whilst di-iso-butyl phthalate (DIBP) and di-n-butyl phthalate (DNBP) were the most abundant LPs/APs in air. A significant correlation (p < 0.05) was found between dust and air levels for chemicals with log Koa < 14, indicating that equilibrium was achieved for these chemicals but not for those with log Koa > 14. Among the investigated human exposure pathways (i.e. dust ingestion, dermal absorption, and air inhalation), dust ingestion was the predominant one for all chemicals. Human exposures of this magnitude to these chemicals through the investigated pathways was unlikely to present a health risk in the present study.

Abstract:Wearing of vehicle parts could release many chemical additives into the environment, such as benzotriazoles (BTRs), benzothiazoles (BTHs), and p-phenylenediamines (PPDs), which are potentially toxic to wildlife and humans. This study investigated the occurrence, source, and risks of BTRs, BTHs, and PPDs in a source catchment providing water to Guangzhou, a megacity in South China, covering groundwater, surface water, and stormwater. The results showed that BTRs and BTHs were predominant in surface water and groundwater. Unexpectedly, the BTR and BTH concentrations were lower in surface water than groundwater in a third of the paired samples. For the first time, 6PPD-quinone, a toxic ozonation product of N-(1,3-dimethylbutyl)-N'-phenyl-1,4-phenylenediamine (6PPD), was extensively detected in source waters. Stormwater decreased the BTR concentrations but increased the 6PPD-quinone concentrations in surface water owing to their affiliation to suspended particles. From natural to urban segments of Liuxi river, a downstream increasing trend in BTR and BTH concentrations was observed, confirming that they are indicative of urban anthropogenic activities. Strong correlations between industrial activities and BTR or BTH concentrations in surface water indicated that industrial activities were their main sources. Six compounds were prioritized as potentially persistent, mobile, and toxic (PMT) chemicals, combing our monitoring results and REACH criterion. This study improves our understanding of the environmental fates and risks of water-soluble tire-wear chemicals, which provides important information for chemical management, and indicates attention should be paid to the risk posed by 6PPD-quinone in the source water.

Abstract:A comparative study of internal and external exposure is a good method to comprehensively understand human exposure to environmental contaminants that may trigger oxidative stress in human body. Information is limited regarding the influences of reactive oxygen species (ROS) on human health from the environment. In addition, data on the contribution of polycyclic aromatic hydrocarbons (PAHs) from indoor environments, especially air, to total human exposure are still insufficient. The present study measured PAHs in paired indoor dust (n = 101), gas (polyurethane foams, n = 100), and particle samples (quartz fiber filters, n = 100) and their hydroxy metabolites (OH-PAHs) in 205 urine samples from 101 families in Guangzhou, South China. The oxidative potential (OP) in dust samples was quantified with a dithiothreitol (DTT) assay to reflect the oxidizability of ROSs, and explore the relationship between environmental ROSs and oxidative stress in humans (using urinary 8-hydroxy-2′-deoxyguanosine (8-OHdG) as a biomarker). The estimated daily intakes (EDIs) of Σ16PAH via air inhalation were much higher than those from gas dermal contact, dust dermal contact, and dust ingestion (mean: 19.5 > 4.27 > 3.75 > 1.60 ng/kg_bw/day). Generally, approximately 16% of naphthalene, 28% of fluorene, 9% of phenanthrene, and 3% of pyrene were derived from indoor environments for all residents when compared with the total PAH exposure amount from all sources. Significantly positive relationships were found between OH-PAHs and 8-OHdG (coefficients β: 0.129–0.366, p < 0.05) checked by linear mixed effect models, and males seemed to be more susceptible than females to the DNA oxidative damage related to PAH exposure. The mean OP value in dust was 7.14 ± 6.68 pmol/(min·μg). Individual PAHs in dust gradually intensified the oxidizability of dust particles as their molecular weight increased. A potential but not significant dose-relationship was found between dusty OP and urinary 8-OHdG. Further work should determine the impact of chemical profiles on OP in different environmental media and continuously explore the potential to use OP as a useful indicator to reflect the total oxidizability of several groups of environmental pollutants.

New Evidence of Rubber-Derived Quinones in Water, Air, and Soil

Abstract:p-Phenylenediamines (PPDs) have been extensively used in the rubber industry and found to be pervasive in various environmental compartments for decades, while their transformation products and associated ecological and human health risks remain largely unknown. Herein, we developed and implemented a mass spectrometry-based platform combined with self-synthesized standards for the investigation of rubber-derived quinones formed from PPD antioxidants. Our results demonstrated that five quinones are ubiquitously present in urban runoff, roadside soils, and air particles. All of the identified sources are closely related to mankind's activities. Among the identified quinones, N-(1,3-dimethylbutyl)-N′-phenyl-p-phenylenediamine quinone has been recently found to be highly toxic, causing acute mortality of coho salmon in the Pacific Northwest. Ultrahigh-performance liquid chromatography coupled with triple quadrupole mass spectrometry was then applied for quantification of the five quinones and their corresponding PPD antioxidants. The results revealed interesting distinct distribution and concentration patterns of PPD-derived quinones in different environmental matrices. Daily intake rates of these quinones in a compact city of Hong Kong were estimated to be varied from 1.08 ng/(kg·day) for adults to 7.30 ng/(kg·day) for children, which were higher than the exposure levels of their parent compounds. Considering the prevalence of the use of rubber products, the outcome of this study strongly suggests for additional toxicological studies to investigate potential ecological and human health risks of the newly discovered quinones.Five p-phenylenediamine-derived quinones have been identified to be pervasive in roadside soil, runoff water, and air particles, while their health risks remain elusive. Research findings demonstrate the coexistence of five quinones from globally ubiquitous p-phenylenediamine antioxidants in urban runoff, roadside soils, and air particles.This article has not yet been cited by other publications.

Abstract:Background Polycyclic aromatic hydrocarbons (PAHs) are typical residential pollutants mainly from biofuel combustion that impose inevitable risk to children. The PAHs in residential dust is universal in most Chinese households with an obvious public health concern. Methods In this observational study, a total of 235 residential dust samples from 8 Chinese cities (Panjin, Shijiazhuang, Lanzhou, Luoyang, Xi'an, Wuxi, Mianyang, and Shenzhen) were collected from April 2018 to March 2019, which were extracted and analyzed for 16 priority PAHs by HPLC/FD-UV. Diagnostic ratios, hierarchical clustering analysis and principal component analysis were applied simultaneously for source apportionments. Incremental lifetime cancer risk was employed to estimate children's health risks based on the assumed exposure scenarios. Spearman correlation, Mann-Whitney U test, Kruskal-Wallis H test and Partial Least Squares were used to screen the factors affecting the concentration of PAHs in residential dust. Results The median concentration of ∑ 16 PAHs in residential dust from 8 cities was 44.11 μg/g (0.04–355.79 μg/g). ∑ 16 PAHs were found both higher in dust samples in heating season and from downwind households only in Mianyang ( p < 0.05). The leading two sources of PAHs were combustion processes and automobile exhaust emissions based on four principal components that accounted for 74.29 % of the total variance. Indoor air environmental factors, household characteristics, and residents' behavioral lifestyles may be the influencing factors of residential dust PAHs. The carcinogenic risk of children aged 0–5 years, under the moderate exposure level of PAHs in residential dust, exceeded the acceptable level (10 −5 - 10 −4 for dermal contact and 10 −6 - 10 −5 for ingestion). Conclusions There was serious PAHs pollution in residential dust under actual living conditions in eight cities across China. More evidence-based measures were needed to control PAHs pollution to safeguard children's health according to appointed sources and influencing factors in residential dust.

As an antioxidant and antiozonant widely used, 6PPD would be released during the production or use of rubber-related products. Because of the mass production and wide use of rubber-related products, 6PPD and 6PPD-quinone have been identified to be ubiquitous in the environment. In this study, we firstly reviewed the current available literature on the analytical procedures, concentrations and distribution of 6PPD and 6PPD-quinone, and then investigated the potential toxic effects of these two compounds on aquatic organisms. Current studies have been mainly focused on the occurrence of 6PPD and 6PPD-quinone in dust and water, while available information on atmosphere, soil, sediments and organisms is limited. The fate and distribution of 6PPD and 6PPD-quinone would be influenced by environmental factors such as temperature, illumination, and storm events, etc. Although 6PPD and 6PPD-quinone have potential adverse effects on aquatic organisms, and 6PPD-quinone has species-specific toxicity, toxicological mechanisms of these compounds are still unclear. Based on the review and analysis of current studies, some suggestions for future research of 6PPD and 6PPD-quinone are given.

Abstract:Phthalates are typical air pollutants in vehicular environment since numerous synthetic materials that might contain phthalates are widely used to fabricate vehicle interiors (e.g., seat cushions, floor mats and dashboards). Hitherto, the importance of phthalate pollution in vehicular environment is not well-recognized because people spend only a small portion (around 8%) of their time in vehicles. In this study, the mass fractions of six phthalates in nine materials commonly used in Chinese vehicles (floor mats and seat cushions) were measured. Two phthalates, di-n-butyl phthalate (DnBP) and di-2-ethylhexyl phthalate (DEHP), were identified in most materials (the other phthalates were not detected). The emission characteristics of DnBP and DEHP from these materials were further investigated. The measured emission parameters were used as input for a mass-transfer model to estimate DnBP and DEHP concentrations in cabin air. Finally, the ratios between human exposures (via inhalation and dermal absorption from the gas phase) in vehicular environment and the total exposures in typical indoor environments (e.g., residences and offices) were estimated to be up to 110% and 20% for DnBP and DEHP, respectively. Based on these results, the vehicular environment might be a considerable site for human exposure to airborne phthalates.

Abstract:Background: N,N'-disubstituted p-phenylenediamine-quinones (PPDQs) are oxidization derivatives of p-phenylenediamines (PPDs) and have raised extensive concerns recently, due to their toxicities and prevalence in the environment, particularly in water environment. PPDQs are derived from tire rubbers, in which other PPD oxidization products besides reported PPDQs may also exist, e.g., unknown PPDQs and PPD-phenols (PPDPs). Results: This study implemented nontarget analysis and profiling for PPDQ/Ps in aged tire rubbers using liquid chromatography-high-resolution mass spectrometry and a species-specific algorithm. The algorithm took into account the ionization behaviors of PPDQ/Ps in both positive and negative electrospray ionization, and their specific carbon isotopologue distributions. A total of 47 formulas of PPDQ/Ps were found and elucidated with tentative or accurate structures, including 25 PPDQs, 18 PPDPs and 4 PPD-hydroxy-quinones (PPDHQs). The semiquantified total concentrations of PPDQ/Ps were 14.08-30.62 μg/g, and the concentrations followed the order as: PPDPs (6.48-17.39) > PPDQs (5.86-12.14) > PPDHQs (0.16-1.35 μg/g). Significance: The high concentrations and potential toxicities indicate that these PPDQ/Ps could seriously threaten the eco-environment, as they may finally enter the environment, accordingly requiring further investigation. The analysis strategy and data-processing algorithm can be extended to nontarget analysis for other zwitterionic pollutants, and the analysis results provide new understandings on the environmental occurrence of PPDQ/Ps from source and overall perspectives.

Abstract:Phthalate acid esters (PAEs), as a common group of plasticizers, are widely present in indoor environments and pose a risk to human health. Indoor dust samples collected from dormitory, classroom, laboratory, and office in several universities in China, were analyzed for seven types of PAEs. The total concentrations of seven PAEs (Σ 7 PAEs) ranged from 4.87 to 360 μg/g, with a median concentration of 51 μg/g, which is lower than that reported by other studies. Using the median concentration of Σ 7 PAEs as a metric, we assessed the levels of contamination in different microenvironments, resulting in the following ranking: dormitory > classroom > laboratory > office. There are significant differences in the levels of individual PAEs in different microenvironments. Radiation from sunlight, ventilation rates, cleaning frequency, and sprays were influential factors for the concentrations of individual PAEs in indoor dust. The indoor environmental conditions and consumption patterns profoundly affect PAEs levels. The sources of PAEs in classroom and office were more complex than in dormitory and laboratory. Daily intakes of PAEs were used to calculate carcinogenic and non-carcinogenic human risk for males and females, indicating a low health risk to humans. This is the first study to assess the risk of PAEs in university microenvironments and provides a valuable reference for further research.
