Abstract:Rhubarb, a well-known Chinese herbal medicine, is also used in Europe and other places of the world. Anthraquinones derivatives are thought to be the major active components. A pressurized liquid extraction (PLE) and capillary zone electrophoresis (CZE) separation were developed for simultaneous determination of five anthraquinones including aloe-emodin, emodin, chrysophanol, physcion, and rhein in Rhubarb. The effects of the experimental variables on PLE and CZE have been optimized. The optimum conditions of PLE were: solvent, methanol; temperature, 140 degrees C; particle size, 0.13-0.2 mm; static extraction time, 5 min; pressure, 1500 psi; and one extraction. The best separation of the five anthraquinones could be obtained using 50 mm borate buffer (pH 8.2) containing 25% isopropyl alcohol and 25% acetontrile as modifier, while the separation voltage was 25 kV and the temperature was at 20 degrees C. The method developed is accurate, simple, and reproducible, and could be used for quality control of Rhubarb and its medical preparations.

Abstract:A validated liquid chromatography method was first developed to evaluate the quality of crude and processed Radix Scrophulariae extracts through establishing chromatographic fingerprint and simultaneous determination of five bioactive compounds, namely 5-hydroxymethylfurfural (5-HMF), acteoside, angroside C, harpagoside and cinnamic acid. The chromatographic were separated on an Agilent Zorbax Extend C(18) column (250 mm × 4.6 mm, 5 μm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.03%, v/v) and (B) acetonitrile using a gradient elution. Analytes were performed at 30 °C with a flow rate of 1.0 mL/min and UV detection at 280 nm. All calibration curves showed good linear regression (r(2) ≥0.9996) within the tested ranges, and the recovery of the method was in the range of 98.12-103.38%, with RSD values ranging from 0.6 to 2.8%. In addition, the contents of those five bioactive compounds in crude and processed Radix Scrophulariae prepared by different locations of China were determined to establish the effectiveness of the method. The results demonstrate that the developed method is accurate and reproducible and could be readily utilized as a suitable quality control method for the quantification of Radix Scrophulariae.
Identification and Determination of Active Anthraquinones in Chinese Teas by Micellar Electrokinetic Capillary Chromatography

Abstract:The simultaneous determination of emodin, aloe-emodin and rhein, in Semen Cassiae, Qinghai Wild Dahuang tea, Ningxia Juemingzi tea, Lanzhou Juemingzi tea and Surong Juerningzi tea, has been investigated by micellar electrokinetic capillary chromatography for the first time. With an electrolyte containing 15 mm borax, 30 mm SDS, 10% (v/v) ethanol, at pH 9.60 and 20 kV applied voltage, the three analytes were completely separated within 12 min. The effects of the concentration of borax, electrolyte pH, the concentrations of SDS and organic modifier and the applied voltage on electropheoretic behavior and separation were studied. The linear calibration range was 4-120 mug/mL (r = 0.9921) for emodin, 10-200 mug/mL (r = 0.9970) for aloe-emodin and 2-100 mug/mL (r = 0.9971) for rhein, respectively. Under the optimum conditions, the relative standard deviation (RSD) values (n = 6) of the migration time and the peak area of each peak was 0.59-0.80% and 1.30-3.22%, respectively. The contents of the analytes in Rheum, Qinghai Wild Dahuang tea, Semen Cassiae and three other kinds of teas were easily determined with recoveries ranging from 95.3 to 104.4%. Determination of anthraquinones in Rhamnus purshiana using high-performance liquid chromatography coupled to diode array detector and simple ultraviolet spectroscopic analysis

Abstract:A new method based on Ultraviolet spectrophotometry was developed and compared with that based on high-performance liquid chromatography for the determination and quantification of anthraquinones in the extracts of Rhamnus purshiana bark. A validated quantitative analysis of cascaroside A, cascaroside B, emodin, and aloe-emodin in these herbal products has been previously performed using high-performance liquid chromatography coupled with a diode array detector. In the high-performance liquid chromatography analysis, all the anthraquinones showed satisfactory regression (r2 > 0.98) within the test ranges, and the recovery was in the range of 94-117%. The limits of detection and quantification were 0.008-0.010 and 0.029-0.035 μg/mL, respectively. Hierarchical cluster analysis and principal component analysis showed differences in the anthraquinones determined from herbal samples. Subsequently, a simple and low-cost ultraviolet spectrophotometric methodology for the quantitative analysis of the same compounds in the extracts was applied, and all the contents were determined. A paired t-test confirmed that there were no significant differences between the two methods. Our results revealed that the developed method is simple and provides the ability to discriminate and control the quality of anthraquinones in herbal products.

Abstract:A method for quantitative determination of the main active ingredient in traditional Chinese medicine was developed and validated, using liquid-liquid chromatography combined with high performance liquid chromatography, which was successfully applied to synephrine in Fructus Aurantii Immaturus. A biphasic solvent system composed of ethyl acetate - n-butanol - water - trifluoroacetic acid (1:2:3:0.003, v/v) was used to purify the crude extract of the samples by liquid-liquid chromatography. Then, quantitative analysis of synephrine in Fructus Aurantii Immaturus could be well determined by high performance liquid chromatography. Moreover, the collection time and retention time of synephrine could be well predicted using a continuous-stirred tank reactors model. Compared with previous studies, the present method showed advantages of good recovery, good repeatability, high selectivity to target components and simple operation.
Novel Approaches for the Analysis and Isolation of Benzylisoquinoline Alkaloids in Chelidonium majus

Abstract:Benzylisoquinoline alkaloids are the major bioactive components in Chelidonium majus , a plant that has a long usage history for the treatment of gastrointestinal ailments in European and Asian phytomedicine. This study reports on the development and application of a supercritical fluid chromatography technique for the simultaneous qualitative and quantitative determination of seven benzylisoquinoline alkaloids in under six minutes using a Viridis BEH 2-EP column and a modifier comprising methanol with 30% acetonitrile and 20 mM ammonium formate. The method was fully validated according to ICH guidelines showing, e.g., excellent linearity (≥ 0.9997) and maximum deviations for intraday and inter-day precision of 2.99 and 2.76%, respectively. The new supercritical fluid chromatography assay was not only employed for the analysis of several C. majus samples but was also used for the subsequent development of a fast centrifugal partition chromatography technique, whereby five benzylisoquinoline alkaloids could be isolated within approximately 2.5 h, with only two of them, protopine and chelidonine, requiring an additional purification step. To achieve this, a solvent system composed of chloroform/methanol/0.3 M hydrochloric acid was used in descending mode. By injecting 500 mg of crude extract, stylopine (1.93 mg), sanguinarine (0.57 mg), chelidonine (1.29 mg), protopine (1.95 mg), and coptisine (7.13 mg) could be obtained. Abstract:High-performance liquid chromatography (HPLC) and capillary electrophoresis (CE) were compared to simultaneously determine and separate 11 anthraquinones from rhubarb, including emodin, chrysophanol, rhein and their glucosides, aloe-emodin, sennoside A, and sennoside B. A UV-diode array detector (DAD) at 254 nm with a gradient elution of acetonitrile/water (method A: 0 min 6:94, 12 min 12:88, 15 min 20:80, 40 min 25:75, 53 min 55:45, 55 min 100:0; method B: 0 min 5:95, 2 min 15:85, 5 min 20:80, 12 min 25:75, 15 min 50:50, 19 min 98:2) at 28(+/-1) degrees C (method A) and 30-60 degrees C (method B) in HPLC or with 0.03 M borate buffer (pH 10.0) containing 25% (v/v) acetonitrile with 0.002 M 2,6-di-O-methyl-beta-cyclodextrin (CD) and 0.005 M alpha-CD in CE effectively detected this separation in 25 min. The detection limits of anthraquinones from rhubarb were in the 0.02-0.2 microg/mL and 0.1-0.8 microg/mL ranges for HPLC and CE, respectively. The established HPLC and CE methods are suitable for quantitative determination of emodin, chrysophanol, aloe-emodin, emodin-1-beta-d-glucoside, emodin-8-beta-d-glucoside, chrysophanol-1-beta-d-glucoside, chrysophanol-8-beta-d-glucoside, and rhein-8-beta-d-glucoside.

Determination of Active Constitutents in Folium Eucommiae by RP-HPLC

Abstract:OBJECTIVE To establish a new reversed phase high-performance liquid chromatography(RP-HPLC) method for the determination of chlorogenic acid and genipoide,the active constitutents in folium eucommiae simultaneously.METHODS YMC-pack ODS-A column(250 mm×4.6 mm,5 μm) was used and the mobile phase was methanol-water-acetic acid(20:80:1.5) at the flow rate of 1.0 mL·min-1.The detection wavelength was 237 nm and the temperature of column was 30 ℃.RESULTS The linear range of chlorogenic acid and genipoide were 0.041 6-0.624 0 μg(r=0.999 2) and 0.081 6-1.224 0 μg(r=0.998 9).The average recoveries were 102.92% and 102.35%,respectively.The RSD were 1.49% and 1.20%.The content of chlorogenic acid and genipoide in folium eucommiae were 2.801 mg·g-1 and 0.273 mg·g-1.CONCLUSION The method is simple,accurate and reliability,with good repeatability,and can be used for the quality control of folium eucommiae.
Rapid Determination of Two Triterpenoid Acids in Chaenomelis Fructus Using Supercritical Fluid Extraction On-line Coupled with Supercritical Fluid Chromatography.

Abstract:In this study, an on-line supercritical fluid extraction (SFE) and supercritical fluid chromatography (SFC) method was developed for the rapid determination of oleanoic acid and ursolic acid in Chaenomelis Fructus. After optimization of the conditions, the two triterpenoid acids was obtained by SFE using 20% methanol as a modifier at 35°C in 8 min. They were resolved on a Shim-pack UC-X Diol column (4.6 × 150 mm, 3 μm) in 14 min (0 - 10 min, 5 - 10%; 10 - 14 min, 10% methanol in CO2) with a backpressure of 15 MPa at 40°C. The on-line SFE-SFC method could be completed within 40 min (10.79 mg/g dry plant, Rs = 2.36), while the ultrasound-assisted extraction and HPLC method required at least 90 min (3.55 mg/g dry plant, Rs = 1.92). This on-line SFE-SFC method is powerful to simplify the pre-processing and quantitative analysis of natural products.

Abstract:For the European Pharmacopoeia (Ph. Eur.) herbal monograph draft of Cassia angustifolia Vahl. and Cassia senna L. leaves and pods, a safety limitation of aloe-emodin and rhein was proposed, due to toxicological concerns. A quantitative, analytical method of the anthraquinone aglycones in all Ph. Eur. monographed herbal laxatives is of interest. A rational method development for the aglycones aloe-emodin, rhein, emodin, chrysophanol, and physcion in five herbal drugs was realized by using 3D chromatographic modelling (temperature, solvent, and gradient time) and design of experiment (DOE) software (DryLab® 4). A methodical approach suitable for the challenging peak tracking in the chromatograms of the herbal drugs in dependence on the changes in the chromatographic conditions is described by using a combination of mass spectroscopy (MS) data (UHPLC-QDa), UV/Vis-spectra, and peak areas. The model results indicate a low robust range and showed that with the selected chromatographic system, small interferences could not be averted. The separation achieved shows a pure UV/Vis spectrum for all aglycones except for chrysophanol in Aloe barbadensis and emodin in Cassia angustifolia fruit. A gradient with the best resolution of the aglycones in all five drugs is proposed, and its suitability demonstrated for the quantification of aglycones in these herbal drugs.
A Simple Liquid Chromatography Coupled with Tandem Mass Spectrometry Approach for the Simultaneous Quantification of Thirteen Compounds in Rats Following Oral Administration of Raw and Processed Fructus Xanthii: Application in a Comparative Pharmacokinetic Study

Abstract:A simple and sensitive analysis using ultra high performance liquid chromatography with a tandem mass spectrometric system operated in selected reaction monitoring mode was developed for the determination of 11 phenolic acids, atractyloside, and carboxyatractyloside in rat plasma. The two classes of analytes were then separated on a Waters ACQUITYTM UPLC HSS T3 column (50 mm x 2.1 mm, 1.8 mu m) using gradient elution with a mobile phase of 0.2% formic acid in water containing 10 mM ammonium acetate and methanol. Detection was accomplished by selected reaction monitoring scanning via an electrospray source operating in negative ionizationmode. The calibration curve was linear (R-2 = 0.990) over a concentration range of 1.20-3500 ng/mL, while the validated lower limit of quantification was 1.20 ng/mL. The precision varied from 0.84 to 4.62%, and the accuracy varied within +/- 5%. The method proved robust with sample freezing and thawing and with short- and long-term sample storage. The established method was used for simultaneous quantification and was successfully used for the first time for the pharmacokinetic evaluation of 13 compounds after the intragastric administration of raw and processed Fructus Xanthii in rats. The results indicated that processing affects the absorption and metabolism of Fructus Xanthii extract. Importantly, the results also indicated the importance of processing for the clinical application of traditional Chinese medicine.

Abstract:A new on-line method for simultaneous identification and monitoring of antioxidants in Fructus aurantii was established by coupling high performance liquid chromatography-diode array detector-electrospray ionisation-ion trap-time of flight-mass spectrometry with post-column derivatisation and luminol–potassium ferricyanide chemiluminescence (HPLC-DAD-ESI-IT-TOF-MS-PCD-LPFCL). While the HPLC fingerprint, structural identification and radical scavenging profile were rapidly obtained by an on-line assay using ultraviolet (UV) absorption, MS and LPFCL, details of the precise substitution patterns of various structures were achieved through UV absorption using PCD addition of shift reagents. Twenty-five flavonoids were identified by either their PCD and MS data or comparison with reference substances. Data collected both from chromatograms and activity profiles of 12 samples revealed significant differences among samples from different habitats. The results showed that this method was rapid and precise, and therefore would be an effective and sensitive method for biocompounds analysis and quality evaluation for complex food and medicinal samples.
Simultaneous Determination of Hydroxyanthraquinones in Rhubarb and Experimental Animal Bodies by High-Performance Liquid Chromatography

Abstract:A simple and reliable high-performance liquid chromatographic (HPLC) method was developed for the simultaneous determination of five hydroxyanthraquinones (aloe-emodin, rhein, emodin, chrysophanol, and physcion) in Rhubarb and experimental animal bodies. A Zorbax SB-C18 column (250 mm × 4.6 mm i.d., 5 µm) and a methanol–0.5% acetic acid (85:15, v/v) mobile phase were used for the separation. The detection wavelength of a diode array detector (DAD) was set at 254 nm. Regression equations revealed a linear relationship (R2 > 0.9996) between the mass of hydroxyanthraquinones injected and the peak areas detected by DAD. The detection limits (S/N = 3) ranged from 0.35 ng to 3.13 ng, and the recoveries ranged from 83% to 103% for different hydroxyanthraquinones. This method is simple, sensitive and suitable for the analysis of hydroxyanthraquinones in medicinal materials and pharmacological experiment samples.
Comparison of three different extraction methods and HPLC determination of the anthraquinones aloe-emodine, emodine, rheine, chrysophanol and physcione in the bark of Rhamnus alpinus L. (Rhamnaceae)

Abstract:Introduction: Rhamnus alpinus L. (Rhamnaceae), a traditional plants in the flora of the Abruzzo region, is known to contain active anthraquinone secondary metabolites. However, the content of anthraquinones varies among R. alpinus samples depending on collection season and site. Thus, using simple, reliable and accurate analytical methods for the determination of anthraquinones in R. alpinus extracts allows comparative study of different methods of extraction. Objective: After a partial validation of an HPLC method for the simultaneous determination of five anthraquinones, aloe-emodine, rheine, emodine, chrysophanol and physcione, in the bark of R. alpinus, we compared three different methods of extraction. Methodology: Anthraquinones were extracted from the bark of R. alpinus using different techniques (methanol maceration, ultrasonic and supercritical CO(2) extraction). Separation and quantification of anthraquinones were accomplished using a reversed-phase C(18) column with the mobile phase of H(2)O-methanol (40 : 60, v/v, 1% formic acid) at a wavelength of 254 nm. The qualitative analyses were also achieved at wavelength of 435 nm. Results: All calibration curves were linear over the concentration range tested (10-200 mM) with the determination coefficients >or=0.991. The detection limits (S/N = 3) were 5 mM for each analytes. All five anthraquinones were found in the samples tested at concentrations reported in experimental data. Conclusion: The described HPLC method and optimised extraction procedure are simple, accurate and selective for separation and quantification of anthraquinones in the bark of R. alpinus and allow evaluation of the best extraction procedure between the tested assays.
One Single Standard Substance for the Determination of Multiple Anthraquinone Derivatives in Rhubarb Using High-Performance Liquid Chromatography-Diode Array Detection.

Abstract:Due to the complexity of the chemical constitution of traditional Chinese medicines (TCMs), now there is a trend to establish methods, using multi-components analysis, for the effective quality control of TCMs. However, the limited availability of multiple reference substances hinders the wide popularization for routine quality control of TCMs. Using an easily available single component contained in the analyte as an internal standard to determine multiple analogues should be a practical option. In this study, we selected rhubarb rhizome as an example, and used emodin, the easily available active component, as the external standard to determine directly the content of emodin in rhubarb, and the same component as the internal standard to simultaneously determine the other six anthraquinones in rhubarb. Compared with the results obtained using the external standard method, this alternative method was found to have no statistically significant differences in our laboratory as verified by the F-test (p=95%, n=6). However, due to weak robustness caused by the fluctuation of relative response factors in different laboratories, such a method can only serve as a practical measure to solve the lack of so many chemical standard substances when relative response factors have been determined. This alternative method can then be used without reference substances. Once the corresponding chemical standards are available and are acceptable as well as cost-effective, then the external standard method for the simultaneous determination of multiple components (SDMC) in TCMs will prevail.
Ultrasonic Solvent Extraction Followed by Dispersive Solid Phase Extraction (d-SPE) Cleanup for the Simultaneous Determination of Five Anthraquinones in Polygonum multiflorum by UHPLC-PDA

Abstract:A rapid and effective ultra-high performance liquid chromatography (UHPLC) method was developed for the determination of five anthraquinones (emodin, physcion, aloe-emodin, rhein, and chrysophanol) in Polygonum multiflorum. The target compounds were ultrasonically extracted with 70% methanol, followed by dispersive solid-phase extraction (d-SPE) with HC-C18 and desorption with acetonitrile. The five anthraquinones were separated on an ACQUITY UPLC® HSS T3 column (2.1 × 100 mm, 1.8 μm) and detected by a photodiode array detector (PDA) at 254 nm. Under the optimized conditions, linear relationships were achieved in the range of 0.3~100 mg/L for emodin, 0.3~40 mg/L for physcion, 0.1~20 mg/L for aloe-emodin, and 0.05~20 mg/L for rhein and chrysophanol. The limits of detection of the five analytes ranged from 0.01 to 0.08 mg/L, and the recoveries were within the range of 82.8~118.4% with an RSD (n = 6) of 1.0~10.3%. The intra-day and inter-day precision (n = 5) of the five targets were in the range of 1.0~1.8% and 3.0~3.1%, respectively. Furthermore, this method was applied to analyses of Polygonum multiflorum samples collected from different regions in China with satisfactory results. All the results indicated that this method is suitable for the detection of five anthraquinones in Polygonum multiflorum.

Abstract:Rhamnus spp. is known to contain biologically active anthraquinone secondary metabolites but the presence of oxyprenylated ones is not reported. To this aim, a new simple, and accurate analytical method was developed to reveal chemical fingerprint of these analytes in plant extracts. Plant samples were analysed after extraction with n-hexane (first step) and methanol (second step) using a C(18) column with a mobile phase composed of 35% of water:65% of methanol (both with 1% formic acid, v/v) at 0.7 mL min⁻¹ flow rate in gradient elution mode. For quantitative analyses, selective detection was performed at 435 nm. The limit of quantification (LOQ) was 0.5 μM, with the only exception of 3-geranyloxyemodin for which the LOQ value was 5.0 μM, and external matrix-matched standard curves showed linearity up to 125 μM. The within- and between-batch precision (RSD%) values ranged from 0.2% to 12.9% while within- and between-batch trueness (bias%) values ranged from 12.2% to 12.7%. The method was applied to evaluate for the first time the presence and the quantities of oxyprenylated anthraquinones in Rhamnus spp. barks as well as the anthraquinone profile of Rhamnus pumila Turra. The proposed method could be directly applied to the selective quantification of these analytes in natural sources.
Determination of Anthraquinones in Different Rhubarb and Coptis Xiexin Decoctions by HPLC

Abstract:This is a study to determine 5 main compounds of rhubarb in Rhubarb and Coptidis Xiexin decoctions,using an assay method,in an attempt to compare the concentration of anthraquinones in different compatibility infusions.We developed a HPLC method,using a phenyl column(250mm×4.6mm,5μm)under an ambient temperature,and using gradient elution with mobile phase of methanol-0.01% phosphoric acid and detected at 254 nm,to determine the con- centration of 5 compounds in Rhubarb.The calibration curve of aloe-emodin,rhein,emodin,and chrysophanol indi- cating a good linearity was obtained within the range of 3.15～64.00μg/ml,and 1.70～33.92μg/ml of physcion.The RSD of 5 compounds were less than 2%.The average extraction recovery of the 5 main compounds was 102.1%,with an RSD less than 5%.The compatibility of rhubarb and coptis(scutellaris)has a significant effect on the concentration of anthraquinones.It is concluded that this method has been proved to be accurate and reliable for determining the concentration of anthraquinones in prescriptions containing Radix et rhizoma rhei.The assay results show that an in- creased or reduced scutellaris concentration in Rhubarb and Coptis Xiexin Decoction has a significant effect on the concentration of anthraquinones.

Abstract:The synthesis of three potent new antitumor agents is described: the A83586C-citropeptin hybrid (1), the A83586C-GE3 hybrid (2), and l-Pro-A83586C (3). Significantly, compounds 1 and 2 function as highly potent inhibitors of beta-catenin/TCF4 signaling within cancer cells, while simultaneously downregulating osteopontin (Opn) expression. A83586C antitumor cyclodepsipeptides also inhibit E2F-mediated transcription by downregulating E2F1 expression and inducing dephosphorylation of the oncogenic hyperphosphorylated retinoblastoma protein (pRb).

Abstract:A new HCl hydrolysis/HPLC method, by adopting L9(34) orthogonal test to optimize hydrolysis condition, has been developed for simultaneous determination of three flavonoid aglycones in Elsholtzia blanda benth. The HCl concentration, methanol concentration, hydrolysis temperature, hydrolysis time are taking as four inspecting foctors, and the contents of luteolin, apigenin, and 5-hydroxy-6,7-dimethoxyflavone in hydrolytic solution are used as the evaluation indexes. Agilent Zorbax SB-C18 is used as analytical column. The mobile phase is a mixture of methanol-0.2% phosphoric acid (70:30, v/v), and UV detector is set at 350 nm. The flow rate is 1.0 mL/min, the temperature of column is maintained at 30 degrees C. The optimal hydrolysis conditions are 3.0M HCl, 70% methanol, 85 degrees C hydrolytic temperature and 3 h hydrolytic time. Standard curves are linear over the concentration range 8.54-85.4 microg/mL, 1.2-12 microg/mL, 9.2-92 microg/mL, and their average recoveries are 96.8%, 98.0%, and 100.5% for luteolin, apigenin, 5-hydroxy-6, 7-dimethoxyflavone, respectively. Thus, the optimum hydrolysis condition is relatively gentle, and the HPLC method is proved to be simple, accurate, and sensitive, so it will be able be applied to quality control of medicinal plant of Elsholtzia blanda.
