Abstract:Paocai is a widely consumed Chinese traditional fermented vegetable product. To understand the effect of temperature on paocai fermentation flora, the bacterial community structure of paocai fermented at 10 degrees C, 15 degrees C, 25 degrees C and 35 degrees C was analyzed by culture-dependent and culture-independent methods. The results showed that increasing the fermentation temperature in a certain range is beneficial for rapid paocai acid production and shortening of the maturity period. Illumina Miseq sequencing was performed on 56 samples at different fermentation process temperatures using a culture-independent method. A total of 1,964,231 high-quality reads of 16S rRNA V3-V4 regions were obtained, and they were divided into 405 operational taxonomic units (OTUs) and identified as 213 bacterial genera. The bacterial diversity decreased with the progression of fermentation, and some spoiled samples had an increased diversity. The culture-independent method found that at 10 degrees C, Lactococcus appeared at the start of fermentation, Leuconostoc and Weissella appeared in the middle of fermentation, and Lactobacillus and Leuconostoc dominated fermentation in the late stage. At 15 degrees C, Lactococcus started fermentation, Leuconostoc appeared in the middle stage, and Lactobacillus was dominant in the late stage. At 25 degrees C, Lactococcus started fermentation, Weissella and Lactobacillus appeared in the middle stage, and Lactobacillus dominated fermentation in the late stage. Finally, at 35 degrees C, Lactococcus, Weissella, and Lactobacillus started fermentation, Weissella and Lactobacillus appeared in the middle stage, and Lactobacillus dominated fermentation in the late stage. A total of 647 strains of bacteria were isolated by culture-dependent methods and were divided into 12 genera and 19 species by randomly amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) and 16S ribosomal RNA gene (rDNA) sequencing technology. More types of bacteria were isolated in the early stage of fermentation. At 10 degrees C, Lactococcus lactis began fermentation, and Lactobacillus brevis and Leuconostoc mesenteroides dominated acid production in the middle and late stages of paocai fermentation. At 15 degrees C, L. lactis initiates fermentation, while Lactobacillus plantarum dominates the acid fermentation of paocai. At 25 degrees C and 35 degrees C, there were a large number of Enterobacteriaceae bacteria in the start-up fermentation stage, and L. plantarum was dominant after 1-2 days of fermentation. Redundancy analysis (RDA) found that the lower the temperature, the more bacterial species that are produced, and the higher the temperature and the longer the time, the more obvious are the effects of L. plantarum on paocai. The results of dominant bacteria studied by culture-dependent and culture-independent methods are similar. The results indicate that most of the dominant microorganisms in the paocai fermentation system are culturable. This discovery can provide data and physical support for modernization and regulation of different types of paocai production.

Abstract:Mustard green paocai is a salted vegetable product that is fermented with low salt in the initial stage and preserved with high salt in the middle stage. To reveal the key flavor-producing potential of core bacterial and fungal communities during the 365-day fermentation of industrial-scaled paocai. In this study, microbiome and molecular sensory science were used to analyze potential relationships between microbial diversity and key flavor compounds. The results showed that bacteria were more diverse than fungi. High salinity improved the flavor of industrial paocai and simplified the microbial community. A total of 74 bacterial and yeast strains were isolated and identified throughout the fermentation process. Lactiplantibacillus, Halanaerobium, Halomonas, Pseudomonas, and Enterobacteriaceae were the dominant bacterial genera, and Debaryomyces hansenii was the core fungal genera in paocai. 4-Ethylphenol, anethole, butanoic acid, 4-ethylguaiacol, and 3-methyl-1-butanol were identified as key flavor compounds in industrial paocai with odor activity values greater than 1. Spearman correlation analysis indicated that bacteria have a greater impact on the flavor of paocai than fungi. Halanaerobium and Halomonas were correlated with flavor and spoilage in paocai. This study provides guidance for the production of desirable flavors by multi-bacteria synergistic fermentation of industrial paocai.
Effect of non-core microbes on the key odorants of paocai

Abstract:Paocai is a widely consumed plant-based fermented food. We studied the effects of microbial community succession and non-core microbes on the key odorants of naturally fermented paocai. Microbiome and molecular sensory science were used to analyze potential relationships between microbial diversity and key flavor compounds. High-throughput analysis pointed out that paocai had a high level of biodiversity, with core microbes mainly represented by Weissella, Lactococcus, and Lactiplantibacillus and non-core microbes mainly represented by Acinetobacter, Raoultella, Enterobacteriales, Debaryomyces, Kazachstania, and Candida species. Ethyl acetate (OAV = 156.84), 1-hexanal (OAV = 76.18) and 1-nonanal (OAV = 57.90) were the key odorants, producing flavors described as etherial, fruity, green, and aldehydic. A total of 57 bacterial strains and 36 fungal strains were isolated and identified from paocai. A Spearman correlation analysis and in vitro simulation system comprehensively analyzed to the correlation of the microbial community and key flavors metabolism characteristics. Bacteria in non-core microbes are not conducive to the quality and safety of paocai, while yeast have the characteristics of “low quantity and great influence” on the key odorants of paocai. These findings will help elucidating the role of non-core microbial community plays in spoilage as well as flavor formation of paocai.

Abstract:The microbial community of lactic acid bacteria isolated from Chinese Paocai prepared with different salt concentrations (2％,moderate 5％ and 8％),was assessed by cultivable methods and various molecular biology methods.The results showed that lactic acid bacteria in Paocai brine exhibited faster growth rate at lower salt concentration during the fermentation process.A total of 563 lactic acid bacteria were isolated from Paocai brines fermented with different salt concentrations and identified by 16S rDNA sequencing analysis,multiplex-PCR method,and PCR-FRLP analyses.The isolates were identified to belong to 11 species of 5 genera respectively.Lactococcus lactis,Lactobacillus pentosus and Leuconostoc were found to be the dominant flora in Paocai samples during the early stage of fermentation with lower salt concentration (2％) while Lactobacillus pentosus dominated the late fermentation stage.Furthermore,Lactobacillus pentosus and Weissella were the most abundant isolates from Paocai brine during the early stage of fermentation with 5％ and 8％ salt concentrations,but the major microbes involved in the late fermentation stage were still Lactobacilluspentosus.
Characterization and correlations of dominant microorganisms and volatile compounds in fermentation process of Yangjiang douchi

Abstract:Background: Yangjiang douchi (YD) is a traditional fermented soybean product, which is popular in Chinese cuisine for its unique flavor. However, due to its high salt content and unstable flavor, its competitiveness in the international market is gradually weakening. Microorganisms have a key role in the production process of YD because it is a fermented food but the effect of microorganisms on the volatile compounds of YD is also not currently clear. Results: In this paper, aroma compounds and microbial diversity in different fermentation stages of YD were analyzed using gas chromatography-mass spectrometry/olfactometry (GC-MS/O) and IlluminaMiseq system sequencing. A total of 78 aroma-active compounds were detected throughout the fermentation process and they influenced the formation of flavor in YD. Fungi flora were relatively single in YD, and bacteria were rich and varied. A total of 418 species of bacteria were present during fermentation, with unclassified_Staphylococcus, Staphylococcus_kloosii, and Bacillus_velezensis_Bacillus predominating. There were 25 species of fungi at the species level, and Aspergillus minisclerotigenes (OTU 4) played a dominant role in the whole fermentation process. Conclusion: Staphylococcus and Bacillus in the bacterial genus were strongly correlated with most flavor compounds detected, and A. minisclerotigenes in the fungi were more relevant to flavor compounds. This research provides a theoretical basis for the enhancement of the flavor of traditional fermented douchi in China. © 2024 Society of Chemical Industry.
Effect of salt concentration on the quality and microbial community during pickled peppers fermentation

Abstract:This work aimed to investigate the effect of salt concentration on the quality and microbial community of pickled peppers during fermentation, and the cross-correlation between microorganisms and quality was also revealed. The results showed that 9 volatile flavor compounds were unique to the low salt concentration group (D group), which also contained higher content of FAA, lactic acid and acetic acid than high salt concentration group (G group). Meanwhile, the samples of D2 group have a better texture properties. Firmicutes, Proteobacteria, Ascomycota, Lactobacillus, Pectobacterium, and Pseudomonas were detected as the main microbial community during the fermentation with different salt concentrations. Furthermore, the correlations analysis results indicated that the salt concentration has a significant effect on the microbial community of pickled peppers (p < 0.001), and Pediococcus, Lactobacillus, Cedecca, Issatchenkia, Pichia, Kazachstania, and Hanseniaspora were significantly correlated with flavors, which played crucial roles in the unique flavor formation of pickled peppers.

Abstract:Ionizing radiation induces clustered DNA damage where two or more lesions are located proximal to each other on the same or opposite DNA strands. It has been suggested that individual lesions within a cluster are removed sequentially and that the presence of a vicinal lesion(s) may affect the rate and fidelity of DNA repair. In this study, we addressed the question of how 8-oxoguanine located opposite to normal or reduced abasic sites would affect the repair of these sites by the base excision repair system. We have found that an 8-oxoguanine located opposite to an abasic site does not affect either the efficiency or fidelity of repair synthesis by DNA polymerase β. In contrast, an 8-oxoguanine located one nucleotide 3′-downstream of the abasic site significantly reduces both strand displacement synthesis supported by DNA polymerase β or δ and cleavage by flap endonuclease of the generated flap, thus inhibiting the long-patch base excision repair pathway.
The dynamic changes of chemical components and microbiota during the natural fermentation process in Da-Jiang, a Chinese popular traditional fermented condiment

Abstract:There is limited information on the chemical components and microbiota changes involved in da-jiang, a Chinese popular traditional fermented soybean condiment. Therefore, dynamic changes both in physicochemical quality and microbial community diversity of 20 samples of naturally fermented da-jiang collected over 95 days were measured, and we classified the samples into 5 groups (each containing 4 time points) according to the results. Based on OTUs analysis, Group 2 and Group 3 might represent the shift-stage between bacteria and fungi. Also, our results demonstrated that the formation of flavour, nutrient and quality of the traditional da-jiang were actually related to all of microbial community, especially the increased bacteria, including Staphylococcus and Leuconostoc. Correspondingly, 'Membrane Transport', 'Carbohydrate Metabolism', 'Amino Acid Metabolism' and 'Replication and Repair' were the top 4 KEGG pathways based on bacterial genes. The findings are expected to be helpful for us not only to understand the formation of flavour, taste and quality of traditional fermented da-jiang, but also to improve the industrialised fermented da-jiang.
Organic Acids Drove the Microbiota Succession and Consequently Altered the Flavor Quality of Laotan Suancai Across Fermentation Rounds: Insights from the Microbiome and Metabolome

Abstract:Laotan Suancai, a popular traditional Chinese fermented vegetable, is manufactured in the industry via four fermentation rounds. However, the differences in flavor quality of Laotan Suancai from the four fermentation rounds and the causes of this variation remain unclear. Metabolome analysis indicated that the different content of five taste compounds and 31 aroma compounds caused the differences in flavor quality among the variated fermentation rounds of Laotan Suancai. Amplicon sequencing indicated that the microbial succession exhibited a certain pattern during four fermentation rounds and further analysis unveiled that organic acids drove the microbiota shift to more acid-resistant populations. Spearman correlation analysis highlighted that seven core microbes may be involved in the formation of differential flavor and the corresponding metabolic pathways were reconstructed by function prediction. Our findings offer a novel perspective on comprehending the deterioration of flavor quality across the fermentation rounds of Laotan Suancai.
Dynamic succession of microbial community in Nongxiangxing daqu and microbial roles involved in flavor formation

Abstract:Daqu, the fermentation starter of Chinese baijiu, is produced by barley and/or peas during open fermentation using natural inoculation. However, the succession of microbial communities and their roles during daqu manufacture remain unclear, which are closely related to the flavor and quality of baijiu. In this study, the dynamics of physicochemical properties, microbial communities, and volatile compounds were investigated at both fermentation (FM) and storage (ST) stages during the manufacture of Nongxiangxing daqu. High-throughput sequencing analysis showed that Weissella and Lactobacillus were the predominant bacterial genera, at the end of storage, the proportion was 31.45% and 13.14% respectively. Thermoascus, Rhizopus, Pichia and Rhizomucor were dominant fungi in daqu, and Thermoascus, Pichia and Rhizopus still occupied a large proportion at the end of daqu storage, accounting for 67.65%, 23.94% and 7.05% respectively. Redundancy analysis was employed to investigate the relationship between microorganisms and physicochemical indexes, and the results showed that acidity and Lactobacillus were positively correlated, saccharification power was positively correlated with Aspergillus, Rhizomucor, Rasamsonia and Thermoascu. The correlation networks between microorganisms and volatile substances were analyzed, a total of 60 compounds including 42 esters, 4 alcohols, 5 ketones and 5 aldehydes, 2 acids, 1 amine, 1 pyrazine were detected, the results suggested that esters are the main aroma components in daqu, and most of esters are positively correlated with fungi. In addition, key enzymes involved in carbohydrate hydrolysis, ethanol fermentation, and flavor formation were revealed. This study may further improve our understanding concerning the microbial system and microbial roles of Nongxiangxing daqu and may contribute to optimize the process during daqu manufacture.
Evaluation of microbial communities of Chinese Feng-flavor Daqu with effects of environmental factors using traceability analysis

Abstract:Analysis of the changes of microorganisms during Chinese Feng-flavor Daqu fermentation, and the specific contribution of different environmental factors to Daqu microorganisms. High throughput sequencing technology and SourceTracker software were used to analyze the microbial diversity of Feng-flavor Daqu before and after fermentation. 85 fungal and 105 bacterial were detected in the newly pressed Feng-flavor Daqu, while 33 fungal and 50 bacterial in the mature Daqu, and 202 fungal and 555 bacterial in the environmental samples. After fermentation, the microbial community structure of Daqu changed and decreased significantly. 94.7% of fungi come from raw materials and 1.8% from outdoor ground, 60.95% of bacteria come from indoor ground, 20.44% from raw materials, and 8.98% from tools. By comparing the changes of microorganisms in Daqu before and after fermentation, the microorganisms in mature Daqu may mainly come from not only the enhanced strains but also the environment.The source of main microorganisms in Feng-flavor Daqu and the influence of environmental factors on the quality of Daqu were clarified, which provided a basis for improving the quality of Feng-flavor Daqu.

The Fungal Communities and Flavor Profiles in Different Types of High-Temperature Daqu as Revealed by High-Throughput Sequencing and Electronic Senses

Abstract:Polymicrobial co-fermentation is among the distinct character of high-temperature Daqu. However, fungal communities in the three types of high-temperature Daqu, namely, white high-temperature Daqu, black high-temperature Daqu, and yellow high-temperature Daqu, are yet to be characterized. In this study, the fungal diversity, taste, and aroma profiles in the three types of high-temperature Daqu were investigated by Illumina MiSeq high-throughput sequencing, electronic tongue, and electronic nose, respectively. Ascomycota and Basidiomycota were detected as the absolute dominant fungal phylum in all types of high-temperature Daqu samples, whereas Thermomyces, Thermoascus, Aspergillus, Rasamsonia, Byssochlamys, and Trichomonascus were identified as the dominant fungal genera. The fungal communities of the three types of high-temperature Daqu differed significantly (p < 0.05), and Thermomyces, Thermoascus, and Monascus could serve as the biomarkers in white high-temperature Daqu, black high-temperature Daqu, and yellow high-temperature Daqu, respectively. The three types of high-temperature Daqu had an extremely significant difference (p < 0.01) in flavor: white high-temperature Daqu was characterized by sourness, bitterness, astringency, richness, methane, alcohols, ketones, nitrogen oxides, and sulfur organic compounds; black high-temperature Daqu was characterized by aftertaste-A, aftertaste-B, methane-aliph, hydrogen, and aromatic compounds; and yellow high-temperature Daqu was characterized by saltiness, umami, methane, alcohols, ketones, nitrogen oxides, and sulfur organic compounds. The fungal communities in the three types of high-temperature Daqu were significantly correlated with taste but not with aroma, and the aroma of high-temperature Daqu was mainly influenced by the dominant fungal genera including Trichomonascus, Aspergillus, Thermoascus, and Thermomyces. The result of the present study enriched and refined our knowledge of high-temperature Daqu, which had positive implications for the development of traditional brewing technique.
Biochemical changes and microbial community dynamics during spontaneous fermentation of Zhacai, a traditional pickled mustard tuber from China

Abstract:Zhacai is a traditional fermented vegetable that has been consumed in China for centuries. It is currently manufactured by spontaneous fermentation and therefore mostly relies on the activities of autochthonous microorganisms. Here, we characterized microbial community dynamics and associated biochemical changes in 12% salted Zhacai during a 90-day spontaneous fermentation process using high-throughput sequencing and chromatography-based approaches to identify associations between microorganisms and fermentation characteristics. Amplicon sequencing targeting bacterial 16S rRNA genes revealed that bacterial communities were dominated by halophilic bacteria (HAB, i.e., Halomonas and Idiomarina) and lactic acid bacteria (LAB, i.e., Lactobacillus-related genera and Weissella) after 30 days of fermentation. In addition, the relative abundances of the fungal genera Debaryomyces, Sterigmatomyces, and Sporidiobolus increased as fermentation progressed. Concomitantly, pH decreased while titratable acidity increased during fermentation, along with associated variation in biochemical profiles. Overall, the levels of organic acids (i.e., lactic and acetic acid), free amino acids (i.e., alanine, lysine, and glutamic acid), and volatiles (i.e., alcohols, esters, aldehydes, and ketones) increased in mature Zhacai. In addition, the abundances of Lactobacillus-related species, Halomonas spp., Idiomarina loihiensis, as well as that of the yeast Debaryomyces hansenii, were strongly correlated with increased concentrations of organic acids, amino acids, biogenic amines, and volatiles. This study provides new detailed insights into the succession of microbial communities and their potential roles in Zhacai fermentation.

Abstract:Appearance features serve as pivotal metrics for regulating the Daqu fermentation process and assessing its quality. Nevertheless, uncertainties persist regarding the variances in microbial community structure and potential functions during Daqu fermentation with distinct appearance features. Employing amplicon sequencing technology, this study scrutinized low-temperature Daqu sample groups (Y and C) exhibiting discernible differences in appearance features. The results elucidated that microbial α-diversity within the Y group aligned with fluctuations in temperature and moisture during LT-Daqu fermentation, a contrast observed in the C group. Concomitant β-diversity and dominant microbial succession analyses revealed disparate microbial community structures between these groups. Co-occurrence network analysis indicated heightened microbial interconnectivity within the Y group. Compared with the C group, the Y group showcased a greater abundance of relevant genes in the Glycolysis/Gluconeogenesis pathway. T test analysis delineated markedly distinct microorganisms at various fermentation stages, with Streptomyces, Staphylococcus, and Saccharomycopsis enriched in the Y group, and Lactobacillus, Wickerhamomyces, and Aspergillus in the C group. This comprehensive investigations revealed the difference of microbial community structure and function of LT-Daqu with different appearance features, providing insights for refining fermentation procedures and quality evaluation. Moreover, it contributes to an enhanced understanding of fermentation mechanisms in analogous domains.

This study comprehensively examined the microbial structure, microbial functions, and flavor characteristics of the three types of low-temperature Daqu using high-throughput sequencing and electronic senses, and it further clarified the relationship between the microbiota and flavor in low-temperature Daqu. The results showed that Hongxin had the highest bacterial richness and diversity, while Houhuo had the lowest. Both fungal richness and diversity were significantly higher in Qingcha than in Hongxin and Houhuo. The differences in peak temperature during Daqu-making led to significant differences in the structure of microbial communities, microbial functions, and flavor quality in the three types of low-temperature Daqu, and could be leveraged for screening and enriching functional microorganisms for Baijiu-making. Co-exclusion patterns between lactic acid bacteria and Bacillus in low-temperature Daqu resulted in a negative correlation between amino acid transport metabolism and carbohydrate transport metabolism. The different types of low-temperature Daqu had distinct flavor profiles, and the differences in the taste profiles were more significant. Dominated by Thermoactinomyces and Lactobacillus, and together with Saccharopolyspora, Bacillus, Streptomyces, Saccharomycopsis, and Thermoascus, they formed the core microbiota that influencing the flavor of low-temperature Daqu. The bacteria mainly influenced the taste of low-temperature Daqu, whereas the fungi mainly influenced the aroma. Each type of low-temperature Daqu contributed to the flavor of light-flavor Baijiu: Hongxin could elevate the levels of aromatic compounds, Houhuo could regulate the bitterness and sourness, and Qingcha could inhibit the generation of sulfur organic compounds. The results of the present study enrich and refine our knowledge of low-temperature Daqu, promoting the further evolution of traditional brewing methods.

Abstract:Daqu is an important saccharifying and fermenting agent. It provides various microorganisms and enzymes for the fermentation of Baijiu and plays a vital role in the formation of Baijiu flavor. However, it is difficult to obtain information on microbial growth and metabolism in time for Daqu production. Therefore, the “Qu Xiang” obtained by smelling is an important index in the traditional production process to evaluate the microbial fermentation in the process of Daqu-making, “Qu Xiang” mainly represents the volatile flavor compounds in Daqu. The microbial diversity and volatile metabolites on 0, 6, 16, and 29 days of the fermentation process were measured using high-throughput sequencing and gas chromatography–mass spectrometry. Significant differences were found in the composition of the microbial community. Pseudomonas , Weissella , Bacillus , and Pelomonas were the main bacterial genera. Alternaria , Rhizopus , and Pichia are the main fungal genera. A total of 32 differential volatile metabolites were detected in samples at four time points using differential metabolic analysis. The correspondence of prevailing microorganisms with differential metabolites distinguished by Spearman correlation and two-way orthogonal partial least square analysis show that Saccharopolyspora exhibited a significant connection for the 12 differential metabolites. A significant positive correlation was observed between Rhizomucor and 13 different metabolites. These findings further understanding of the metabolism of microorganisms in Daqu fermentation and also help to control the microorganisms in the Daqu-making process, to obtain more stable Baijiu products.

Abstract:Paocai is one of the most popular traditional fermented products in China. Different lactic acid bacteria (LAB) have an important influence on the microbial changes, the dynamic changes of metabolites, and the sensory qualities of paocai. In this study, five different paocai groups were designed which were fermented by Lactobacillus plantarum DMDL 9010 (LP), Lactobacillus brevis GIM 1.773 (LB), Leuconostoc mesenteroides GIM 1.774 (LM), mixed three kinds of LAB (LP + LB + LM), pasteurization (PST) and natural fermentation (NF) without LAB. The nitrite, nitrate, pH, titratable acidity, viable count (LAB and yeast) and metabolites (organic acids, sugars and volatile substances) of these paocai groups were measured. Compared with each single bacterial fermentation (LP, LB and LM), NF and PST, the mixed three kinds of LAB (LP + LB + LM) had a lower nitrite peak, which could be attributed to the significant accumulation of organic acids, and positively affected sensory quality of paocai. Flavor substances produced in paocai samples confirmed as alcohols, ketones and olefins. Sensory evaluation showed that multi-bacteria mixed fermentation of paocai had a better flavor than that of NF, PST and single bacterial fermentation. Therefore, the use of multiple LAB as starter cultures contributes to enhance paocai qualities and control the levels of nitrite.
The Microbial Communities and Flavour Compounds of Jiangxi Yancai, Sichuan Paocai and Dongbei Suancai: Three Major Types of Traditional Chinese Fermented Vegetables

Abstract:The microbial communities, their predicted functional roles and associated flavour compounds in three typical traditional Chinese fermented vegetable (TCFV) foods, viz. Jiangxi yancai (JX), Sichuan paocai (SC) and Dongbei suancai (DB) were revealed by gene sequencing, predictive functionality and correlation analysis. Bacterial community analysis showed that Lactobacilli made up a large proportion (up to 77.6%) of the species in each food type and revealed clear differences in bacterial community structure among the three types. Lactobacillus sakei, Lactobacillus acetotolerans, and Pediococcus pentosaceus were found to be markers of the DB, SC, JX types, respectively. Overall, 8 organic acids (OAs), 17 amino acids (AAs) and 97 volatile flavour compounds (VFCs) were detected. Several Lactobacilli were closely correlated with more than 20 flavour compounds. Based on their abundance and putative contributions to flavour, Lactobacillus acetotolerans and Lactobacillus sakei may be considered core functional microbiota of each of the three TCFV foods. Functional analysis showed that microbial metabolism of carbohydrates, amino acids and nucleotides is a major determinant of the characteristics of all three TCFV product types. The systematic method used in this study allows identification of the key microorganisms involved in vegetable fermentation and provides a deeper understanding of their contribution to TCFV products.
