Abstract:ABSTRACT The soil microbial community plays an important role in terrestrial carbon and nitrogen cycling. However, microbial responses to climate warming or cooling remain poorly understood, limiting our ability to predict the consequences of future climate changes. To address this issue, it is critical to identify microbes sensitive to climate change and key driving factors shifting microbial communities. In this study, alpine soil transplant experiments were conducted downward or upward along an elevation gradient between 3,200 and 3,800 m in the Qinghai-Tibet plateau to simulate climate warming or cooling. After a 2-year soil transplant experiment, soil bacterial communities were analyzed by pyrosequencing of 16S rRNA gene amplicons. The results showed that the transplanted soil bacterial communities became more similar to those in their destination sites and more different from those in their “home” sites. Warming led to increases in the relative abundances in Alphaproteobacteria , Gammaproteobacteria , and Actinobacteria and decreases in Acidobacteria , Betaproteobacteria , and Deltaproteobacteria , while cooling had opposite effects on bacterial communities (symmetric response). Soil temperature and plant biomass contributed significantly to shaping the bacterial community structure. Overall, climate warming or cooling shifted the soil bacterial community structure mainly through species sorting, and such a shift might correlate to important biogeochemical processes such as greenhouse gas emissions. This study provides new insights into our understanding of soil bacterial community responses to climate warming and cooling.
Responses of Soil Microbial Communities to Experimental Warming in Alpine Grasslands on the Qinghai-Tibet Plateau

Abstract:Global surface temperature is predicted to increase by at least 1.5°C by the end of this century. However, the response of soil microbial communities to global warming is still poorly understood, especially in high-elevation grasslands. We therefore conducted an experiment on three types of alpine grasslands on the Qinghai-Tibet Plateau to study the effect of experimental warming on abundance and composition of soil microbial communities at 0-10 and 10-20 cm depths. Plots were passively warmed for 3 years using open-top chambers and compared to adjacent control plots at ambient temperature. Soil microbial communities were assessed using phospholipid fatty acid (PLFA) analysis. We found that 3 years of experimental warming consistently and significantly increased microbial biomass at the 0-10 cm soil depth of alpine swamp meadow (ASM) and alpine steppe (AS) grasslands, and at both the 0-10 and 10-20 cm soil depths of alpine meadow (AM) grasslands, due primarily to the changes in soil temperature, moisture, and plant coverage. Soil microbial community composition was also significantly affected by warming at the 0-10 cm soil depth of ASM and AM and at the 10-20 cm soil depth of AM. Warming significantly decreased the ratio of fungi to bacteria and thus induced a community shift towards bacteria at the 0-10 cm soil depth of ASM and AM. While the ratio of arbuscular mycorrhizal fungi to saprotrophic fungi (AMF/SF) was significantly decreased by warming at the 0-10 cm soil depth of ASM, it was increased at the 0-10 cm soil depth of AM. These results indicate that warming had a strong influence on soil microbial communities in the studied high-elevation grasslands and that the effect was dependent on grassland type.

Abstract:Soil microbial communities are extremely complex, being composed of thousands of low-abundance species (<0.1% of total). How such complex communities respond to natural or human-induced fluctuations, including major perturbations such as global climate change, remains poorly understood, severely limiting our predictive ability for soil ecosystem functioning and resilience. In this study, we compared 12 whole-community shotgun metagenomic data sets from a grassland soil in the Midwestern United States, half representing soil that had undergone infrared warming by 2 degrees C for 10 years, which simulated the effects of climate change, and the other half representing the adjacent soil that received no warming and thus, served as controls. Our analyses revealed that the heated communities showed significant shifts in composition and predicted metabolism, and these shifts were community wide as opposed to being attributable to a few taxa. Key metabolic pathways related to carbon turnover, such as cellulose degradation (similar to 13%) and CO2 production (similar to 10%), and to nitrogen cycling, including denitrification (similar to 12%), were enriched under warming, which was consistent with independent physicochemical measurements. These community shifts were interlinked, in part, with higher primary productivity of the aboveground plant communities stimulated by warming, revealing that most of the additional, plant-derived soil carbon was likely respired by microbial activity. Warming also enriched for a higher abundance of sporulation genes and genomes with higher G+C content. Collectively, our results indicate that microbial communities of temperate grassland soils play important roles in mediating feedback responses to climate change and advance the understanding of the molecular mechanisms of community adaptation to environmental perturbations.

Abstract:Climate change is likely to alter the relative abundances of plant functional groups and the interactions between plants and soil microbes that maintain alpine meadow ecosystems. However, little is known about how warming-induced alterations to aboveground biomass (AGB) affect soil nutrients and microbial communities. We investigated plant community characteristics in 2002–2009 and analyzed soil properties and the soil microbial community in 2007–2009 to study the effects of warming in Qinghai Province, China. Sampling involved the use of warmed open top chambers, the monitoring of plant community characteristics, the quantification of total and available amounts of soil nutrients, and the evaluation of microbial community composition using phospholipid fatty acid (PLFA) analysis. Experimental warming initially significantly increased the number of plant functional groups and plant community AGB; however, plant community diversity and species richness decreased. Nevertheless, all these variables stabilized over time. Fungal and bacterial abundance, total nitrogen, available nitrogen and soil organic matter increased with warming, while microbial PLFAs decreased. These findings demonstrated that climate change drivers and their interactions may cause changes in soil nutrients and the abundance and content of soil microbial PLFAs. Elevated temperature has strong effects on aboveground grass biomass. Surface conditions and disturbance affect the soil microbial communities of deep soil layers.

Abstract:Identifying the drivers of the response of soil microbial respiration to warming is integral to accurately forecasting the carbon-climate feedbacks in terrestrial ecosystems. Microorganisms are the fundamental drivers of soil microbial respiration and its response to warming; however, the specific microbial communities and properties involved in the process remain largely undetermined. Here, we identified the associations between microbial community and temperature sensitivity (Q10) of soil microbial respiration in alpine forests along an altitudinal gradient (from 2974 to 3558 m) from the climate-sensitive Tibetan Plateau. Our results showed that changes in microbial community composition accounted for more variations of Q10 values than a wide range of other factors, including soil pH, moisture, substrate quantity and quality, microbial biomass, diversity and enzyme activities. Specifically, co-occurring microbial assemblies (i.e., ecological clusters or modules) targeting labile carbon consumption were negatively correlated with Q10 of soil microbial respiration, whereas microbial assemblies associated with recalcitrant carbon decomposition were positively correlated with Q10 of soil microbial respiration. Furthermore, there were progressive shifts of microbial assemblies from labile to recalcitrant carbon consumption along the altitudinal gradient, supporting relatively high Q10 values in high-altitude regions. Our results provide new insights into the link between changes in major microbial assemblies with different trophic strategies and Q10 of soil microbial respiration along an altitudinal gradient, highlighting that warming could have stronger effects on microbially-mediated soil organic matter decomposition in high-altitude regions than previously thought.

Abstract:Warming has been shown to cause soil carbon (C) loss in northern grasslands owing to accelerated microbial decomposition that offsets increased grass productivity. Yet, a multi-decadal survey indicated that the surface soil C stock in Tibetan alpine grasslands remained relatively stable. To investigate this inconsistency, we analyzed the feedback responses of soil microbial communities to simulated warming by soil transplant in Tibetan grasslands. Whereas microbial functional diversity decreased in response to warming, microbial community structure did not correlate with changes in temperature. The relative abundance of catabolic genes associated with nitrogen (N) and C cycling decreased with warming, most notably in genes encoding enzymes associated with more recalcitrant C substrates. By contrast, genes associated with C fixation increased in relative abundance. The relative abundance of genes associated with urease, glutamate dehydrogenase and ammonia monoxygenase (ureC, gdh and amoA) were significantly correlated with N2O efflux. These results suggest that unlike arid/semiarid grasslands, Tibetan grasslands maintain negative feedback mechanisms that preserve terrestrial C and N pools. To examine whether these trends were applicable to the whole plateau, we included these measurements in a model and verified that topsoil C stocks remained relatively stable. Thus, by establishing linkages between microbial metabolic potential and soil biogeochemical processes, we conclude that long-term C loss in Tibetan grasslands is ameliorated by a reduction in microbial decomposition of recalcitrant C substrates.
Warming-induced shifts in alpine soil microbiome: An ecosystem-scale study with environmental context-dependent insights

Abstract:Climate warming is a pressing global issue with substantial impacts on soil health and function. However, the influence of environmental context on the responses of soil microorganisms to warming remains largely elusive, particularly in alpine ecosystems. This study examined the responses of the soil microbiome to in situ experimental warming across three elevations (3850 m, 4100 m, and 4250 m) in the meadow of Gongga Mountain, eastern Tibetan Plateau. Our findings demonstrate that soil microbial diversity is highly resilient to warming, with significant impacts observed only at specific elevations. Furthermore, the influence of warming on the composition of the soil microbial community is also elevation-dependent, underscoring the importance of local environmental context in shaping microbial evolution in alpine soils under climate warming. Notably, we identified soil moisture at 3850 m and carbon-to-nitrogen ratio at 4250 m as indirect predictors regulating the responses of microbial diversity to warming at specific elevations. These findings underscore the paramount importance of considering pre-existing environmental conditions in predicting the response of alpine soil microbiomes to climate warming. Our study provides novel insights into the intricate interactions between climate warming, soil microbiome, and environmental context in alpine ecosystems, illuminating the complex mechanisms governing soil microbial ecology in these fragile and sensitive environments.

Abstract:High altitude alpine grasslands in the Qinghai-Tibetan Plateau (QTP) contain high soil organic C (SOC) stocks that are extremely vulnerable to climate warming. Microbial residues are increasingly recognized as a major source of SOC, however, how climate warming affects this component of SOC in this region remains largely unknown. In this study, we examined the response of microbial residues to a 3-year experimental warming and the degree to which they contributed to SOC storage in two Tibetan ecosystems—alpine steppe (AS) and swamp meadow (SM). The number of microbial residues was indicated by amino sugar analysis. Our results revealed that warming yielded divergent microbial residue accumulation that significantly altered their contribution to SOC storage in the two alpine grasslands. Warming increased microbial residue abundance by approximately 17.6% across 0 to 20 cm depth in SM soils, while causing a significant decline (about 6.2%) in AS soils. The higher microbial residue accumulation in SM could lessen potential positive feedbacks from climate warming, while the decrease in microbial residues in AS may indicate greater loss of microbial-derived C inputs in warmed soils. Moreover, we found that warming selectively increased fungal residues as compared with bacterial despite inconsistent responses to warming in the two grasslands. These changes were accompanied by significant shifts in fungal to bacterial residue C ratios and their contribution to SOC pool, indicating an alteration of SOC composition and stability in alpine grassland ecosystems. These findings demonstrate that a microbial-derived C feedback to climate change is ecosystem-specific that alters the direction and magnitude of the microbial community.

Warming Counteracts Grazing Effects on the Functional Structure of the Soil Microbial Community in a Tibetan Grassland

Abstract:Grazing intensity and global warming are expected to increase in the forthcoming decades, with uncertain consequences for their interaction on grassland ecosystems and their functions. We investigated the effects of warming, grazing and their interaction in a factorial warming (+1.2-1.7 degrees C) and grazing (moderate intensity with ca. 50% vegetation consumption) experiment in a Tibetan alpine meadow on microbial communities by studying functional genes involved in soil carbon and nitrogen cycles, using GeoChip technology. Our results showed that microbial functional gene structure and abundances were largely affected by the interactive effect of grazing and warming, rather than the main effect of warming or grazing. Compared to the control, grazing alone significantly increased the functional gene alpha diversity, changed the overall functional community structure, and increased the abundances of C fixation, C degradation, N mineralization and denitrification genes, likely due to the stimulating impact of urine and dung deposition. Warming alone did not change these microbial properties, possibly related to the unchanged soil nutrient status. Despite an increase in soil NO3- concentrations and the deposition of urine and dung, the combined treatment did not change functional gene alpha diversity, community structure, or C/N cycling gene abundances, possibly resulting from the limiting effect of water depletion in the combined treatment. Our study revealed antagonistic interactions between warming and grazing on microbial functional gene structure and abundances, which remained stable under the moderate intensity of grazing in future warming scenario in the Tibetan alpine meadow, raising potentially important implications for predicting future soil carbon and nitrogen processes in these systems.

Abstract:Global warming is likely to affect the underground microbial communities in various ecosystems, but the response of soil microbial communities along a vertical depth profile to global warming has been elusive. Herein, we leveraged a warming field experiment in Qinghai-Tibet Plateau grassland and investigated the community structure of prokaryotes and fungi from the upper (0–15 cm) and lower (15–30 cm) strata under ambient and elevated temperature treatments. Three-years continual warming only significantly shifted the prokaryotic community within the upper strata and there was no significant effect observed for the fungal community. Additionally, under ambient temperature, there were significant differences between the two strata in both the prokaryotic and fungal communities, but under warming, this effect was alleviated. Next, the prokaryotic and fungal community assembly processes were measured by a phylogenetic-bin-based null approach (iCAMP). Though deterministic and stochastic processes dominated the assembly of prokaryotic and fungal communities, respectively, the deterministic processes were strengthened under warming for both communities. Specifically, the increased portion of homogeneous selection, contributing to a homogenous state, led to a smaller difference between prokaryotic communities of the two soil strata under warming. The smaller difference in the stochastic process component, i.e. , dispersal limitation, contributed to the similar fungal community structures between the two strata under warming. This study deepens our understanding of warming effects on grassland microbial communities and gives greater insights on the underlying mechanisms for microbial assembly between upper and lower soil strata under warming scenarios.

Abstract:It is well established that climate warming has become a growing issue globally, posing a threat to native ecosystems. Alpine ecosystems, such as meadows of the Qinghai-Tibet Plateau, are expected to be particularly sensitive to warming given current temperature constraints. While many studies have explored the effects of warming on aboveground ecosystems and edaphic properties, few studies have assessed the effects on soil biota. We assessed edaphic, plant, microbial and nematode responses to warming in a long-term (8 year) multilevel warming experiment and applied piecewise structural equation modelling to reveal how warming affected nematode communities directly and indirectly via biotic and abiotic factors. We found that (1) warming had a significant effect on nematode community composition, which was mainly due to direct warming effects on herbivores and omnivore-predatory nematode composition; (2) warming affected nematode richness mainly through effects on bacterial richness, with a strong negative relationship between bacterial richness and bacterivore richness as well as bacterivore richness and omnivore-predatory richness; and, (3) the predominantly direct effect of warming on nematode biomass was mainly due to significant responses of omnivore-predatory biomass. Our study provides insight into the effects of long-term warming on nematode communities and highlights the contrasting responses of composition, diversity and biomass to warming. It contributes to forecasting warming effects on the structure of soil food webs and ecosystem functioning on the Qinghai-Tibet plateau in the future.

Abstract:I calculate the screening length and its relation to elastic constants from mean-field models of superionic conductors. The Debye formula for a continuous fluid's screening length is replaced by one that accounts for the discrete nature of the lattice. Interactions alter the screening length and in some cases give oscillations in the charge structure. The screening lengths derived here are exact not only at low temperature where there are few defects, but also at high temperature where the effective interaction strength is small. The mean-field treatment correctly gives the decrease in an elastic constant observed at the superionic transition for a variety of crystals. This decrease is best explained by a change in the defect creation entropy with density. The effect of doping on the elastic constant also agrees well with experiment. The extension to the mean-field models given here provides a consistent picture of the response of a superionic crystal to electrical, mechanical and thermal stresses, as well as to doping.
Unaltered soil microbial community composition, but decreased metabolic activity in a semiarid grassland after two years of passive experimental warming

Abstract:Soil microbial communities regulate soil carbon feedbacks to climate warming through microbial respiration (i.e., metabolic rate). A thorough understanding of the responses of composition, biomass, and metabolic rate of soil microbial community to warming is crucial to predict soil carbon stocks in a future warmer climate. Therefore, we conducted a field manipulative experiment in a semiarid grassland on the Loess Plateau of China to evaluate the responses of the soil microbial community to increased temperature from April 2015 to December 2017. Soil temperature was 2.0°C higher relative to the ambient when open‐top chambers (OTCs) were used. Warming did not affect microbial biomass or the composition of microbial functional groups. However, warming significantly decreased microbial respiration, directly resulting from soil pH decrease driven by the comediation of aboveground biomass increase, inorganic nitrogen increase, and moisture decrease. These findings highlight that the soil microbial community structure of semiarid grasslands resisted the short‐term warming by 2°C, although its metabolic rate declined.

Plant-Soil Mediated Effects of Long-Term Warming on Soil Nematodes of Alpine Meadows on the Qinghai–Tibetan Plateau

Abstract:Global warming is one of the most pressing environmental issues today. Our study aimed to investigate how warming affected plant and soil nematode communities in alpine meadows on the Qinghai–Tibetan Plateau over the past seven years. An artificial warming experiment with different gradients was conducted from 2011 to 2018, including temperature increases of 0 °C (CK), 0.53 °C (A), 1.15 °C (B), 2.07 °C (C), and 2.17 °C (D), respectively. Cyperaceae plants were shown to be eliminated by increasing temperature, and plant community composition tended to cluster differently under different warming gradients. The number of nematodes decreased with the increase in soil depth, and the majority of them were observed in the topsoil layer. The individual densities of soil nematodes were 197 ind.·100 g−1 dry soil at 10–20 cm and 188 ind.·100 g−1 dry soil at 20–30 cm in the A treatment, which was significantly higher than the CK (53 and 67 ind.·100 g−1 dry soil) (p < 0.05). The lowest relative abundance of bacterivore nematodes (Ba) was 31.31% in treatment A and reached the highest of 47.14% under the warming gradient of D (p < 0.05). The abundance of plant parasitic nematodes (Pp) was significantly reduced to 26.03% by excessive warming (2.17 °C increase) in comparison to CK (41.65%). The soil nematode community had the highest diversity with a 0.53 °C increase in soil temperature; 1.15 °C warming gradients were lower, and nematode communities tended to be simplified (p < 0.05). All nematode channel ratio (NCR) values were above 0.5, indicating that warming did not change the decomposition pathway of soil organic matter dominated by the bacterial channels. The Wasilewska Index (WI) in the D treatment increased significantly compared to other treatments (p < 0.05), indicating that the mineralized pathway of the food web was primarily involved with Ba and fungivores nematodes (Fu), which is conducive to the growth of micro-biophagous nematodes. The plant parasite index (PPI) decreased significantly in the D treatment compared with other treatments (p < 0.05), indicating that a high warming gradient caused a reduction in the maturity of Pp nematodes. The maturity index (MI) increased in the D treatment compared with A, B, and C treatments, indicating that overheating affected the nematode community in the later stage of succession and caused the soil to be less disturbed. A partial least squares path model (PLSPM) showed that warming indirectly affects Fu and Pp diversity by directly impacting the plant community as well as indirectly affecting Ba by directly affecting soil properties. In conclusion, plant diversity and community composition profoundly affect the soil nematode communities, thus reflecting the dynamic processes and evolution of soil ecosystems.

Abstract:The uncertainty response of soil fungi community to climate warming in alpine agroecosystems will limit our ability to fully exploit and utilize soil fungi resources, especially in alpine regions. In this study, a warming experiment was conducted in one perennial leguminous agroecosystem [i.e., alfalfa (Medicago sativa)], perennial gramineous agroecosystem (i.e., Elymus nutans) and annual gramineous agroecosystem [i.e., highland barley (Hordeum vulgare L)] in Tibet since 2016 to investigate the response of soil fungi community to climate warming. Soils at two layers (i.e., 0–10 cm and 10–20 cm) were collected in August 2017 to estimate soil fungi community based on the ITS method. The α-diversity, community composition and functional group abundance of soil fungi in the leguminous agroecosystem were more sensitive to climate warming. The α-diversity of soil fungi in the perennial gramineous agroecosystem were more sensitive to climate warming, but topology parameters of soil fungi species cooccurrence network in the annual gramineous agroecosystem were more sensitive to climate warming. Compared with 0–10 cm, soil fungal α-diversity, community composition and functional group abundance at 10–20 cm were more sensitive to climate warming. The topological parameters of soil fungi species cooccurrence network at 0–10 cm in the gramineous agroecosystem were more sensitive to climate warming, but those at 10–20 cm in the leguminous agroecosystem were more sensitive to climate warming. Warming increased the differences of soil fungi α-diversity and functional composition. For the Medicago sativa agroecosystem, warming increased the abundance of soil pathogenic fungi but decreased the abundance of soil symbiotic and saprophytic fungi at 10–20 cm. Therefore, responses of the soil fungi community to climate warming varied with agroecosystem types and soil depth. Climate warming can alter the differences of the soil fungi community among agroecosystems. Changes in soil fungi community caused by climate warming may be detrimental to the growth of alpine crops, at least for perennial Medicago sativa in Tibet.

Abstract:Exploring the feedback of the soil microbial community and associated processes to global warming represents a major global challenge. To date, the focus has been placed on the direct effects of warming on soil microbial communities, overlooking how concurrent changes in plant communities may mediate these effects. Additionally, few studies have examined long-term effects of warming in more than one environmental context. In the present study, we conducted a long-term simulated warming experiment to investigate how changes to the plant community within two different environmental contexts affect the responses of soil microorganisms and their respiration to warming. We analyzed the abundance, diversity, and community composition of plants and soil microbes, in addition to soil microbial interaction networks and soil microbial respiration, in two typical ecosystems of the Qinghai–Tibet Plateau. Following long-term warming, the soil microbial composition, structure, and interactions changed, and the shifts depended on the aboveground plant type. Specifically, the co-occurring networks containing different microbial communities tended to be more complex in a shrubland than in a grassland after warming, leading to higher carbon use efficiency. Additionally, long-term warming changed the structure of soil microbial communities, increasing the relative abundances of oligotrophic taxa in the shrubland but not in the grassland. The shifts in community structure and interaction patterns could be explained by vegetation community attributes, highlighting the strong effect of plants on soil microbial responses. These plant-mediated effects on community structure and interactions subsequently could explain changes in soil microbial respiration rates. Microbial respiration showed a positive response to elevated temperature in the grassland but no response to temperature in the shrubland. These results indicate that interactions between soil microbial communities and plant communities determine how soil microbes respond to global warming. Therefore, future research on soil microbial community composition and associated carbon feedbacks to the climate change should include plant-mediated effects, which can provide a scientific basis for effectively mitigating global warming.

Abstract:The alpine meadow ecosystem on the Qinghai-Tibetan Plateau (QTP) is very sensitive to warming and plays a key role in regulating global carbon (C) cycling. However, how warming affects the soil organic carbon (SOC) pool and related C inputs and outputs in alpine meadow ecosystems on the QTP remains unclear. Here, we combined two field experiments and a meta-analysis on field experiments to synthesize the responses of the SOC pool and related C cycling processes to warming in alpine meadow ecosystems on the QTP. We found that the SOC content of surface soil (0-10 cm) showed a minor response to warming, but plant respiration was accelerated by warming. In addition, the warming effect on SOC was not correlated with experimental and environmental variables, such as the method, magnitude and duration of warming, initial SOC content, mean annual temperature, and mean annual precipitation. We conclude that the surface SOC content is resistant to climate warming in alpine meadow ecosystems on the QTP.

Abstract:The grassland and shrubland are two major landscapes of the Tibetan alpine meadow, a region very sensitive to the impact of global warming and anthropogenic perturbation. Herein, we report a study showing that a majority of differences in soil microbial community functional structures, measured by a functional gene array named GeoChip 4.0, in two adjacent shrubland and grassland areas, were explainable by environmental properties, suggesting that the harsh environments in the alpine grassland rendered niche adaptation important. Furthermore, genes involved in labile carbon degradation were more abundant in the shrubland than those of the grassland but genes involved in recalcitrant carbon degradation were less abundant, which was conducive to long-term carbon storage and sequestration in the shrubland despite low soil organic carbon content. In addition, genes of anerobic nitrogen cycling processes such as denitrification and dissimilatory nitrogen reduction were more abundant, shifting soil nitrogen cycling toward ammonium biosynthesis and consequently leading to higher soil ammonium contents. We also noted higher abundances of stress genes responsive to nitrogen limitation and oxygen limitation, which might be attributed to low total nitrogen and higher water contents in the shrubland. Together, these results provide mechanistic knowledge about microbial linkages to soil carbon and nitrogen storage and potential consequences of vegetation shifts in the Tibetan alpine meadow.
Long-term Warming Rather Than Grazing Significantly Changed Total and Active Soil Procaryotic Community Structures

Abstract:There is a paucity of knowledge in understanding the effects of warming and grazing on soil microbes and their active counterparts, especially on the Tibetan Plateau which is extremely sensitive to global warming and human activities. A six-year field experiment was conducted to investigate the effects of asymmetric warming and moderate grazing on total and active soil microbes in a Tibetan Kobresia alpine meadow. Soil bacterial abundance and 16S rDNA transcriptional activity were determined using real-time PCR. Total and active soil procaryotic community structures were analyzed through MiSeq sequencing based on 16S rDNA and rRNA, respectively. The results showed that the soil procaryotic community was more sensitive to the warming than the grazing. The warming significantly decreased soil microbial respiration rates, 16S rDNA transcription activity, and dispersion of total procaryotic community structures, but significantly increased the α diversity of active procaryotes. Warming also significantly increased the relative abundance of oligotrophic microbes, whereas decreasing the copiotrophic lineage proportions. The functional profiles predicted from the total procaryotic community structures remained unaffected by warming. However, the rRNA-based predictions suggested that DNA replication, gene expression, signal transduction, and protein degradation were significantly suppressed under the warming. The grazing only significantly decreased the 16S rDNA transcription and total procaryotic richness. Overall, these findings suggest that warming can shift soil procaryotic community to a more oligotrophic and less active status, highlighting the importance of investigating active microbes to improve our understanding of ecosystem feedbacks to climate change and human activities.
Effects of Warming on Carbon and Nitrogen Cycling in Alpine Grassland Ecosystems on the Tibetan Plateau: A Meta-Analysis

Abstract:Anthropogenic activities have caused rapid warming globally, particularly in the alpine regions. The Tibetan Plateau is sensitive to global change and plays an important role in global carbon cycle due to its large area and high carbon density. Although a large number of field manipulative warming experiments have been conducted on the Tibetan Plateau, the general patterns of how warming affects ecosystem carbon (C) and nitrogen (N) cycles remain unclear. Here we conducted a meta-analysis from 65 publications to synthesize the responses of C and N cycles to warming in the alpine grassland ecosystems across the Tibetan Plateau. The results showed that warming significantly increased aboveground (mean 11.0%, 95% CI 1.6-21.4%) and belowground plant biomass (24.3%, 13.7-36.0%), soil dissolved organic C (13.6%, 5.4-22.5%), soil respiration (14.3%, 2.9-27.1%), ecosystem respiration (15.3%, 4.7-27.1%), and soil ammonium N (29.0%, 4.6-59.2%), but did not change soil total organic C (2.4%, -4.4 to 9.7%), soil total N (1.9%, -2.7 to 6.7%), microbial biomass C (5.5%, -4.1 to 16.2%) and microbial biomass N (4.7%, - 7.4 to 18.4%). There were negative relationships between the response ratio of aboveground (P = 0.08, R-2 = 0.05) and particularly below-ground (P < 0.01, R-2 = 0.23) biomass and the duration of warming. Moreover, the effect of warming on ecosystem respiration was positively correlated with the change in soil water content after warming (P = 0.03, R-2 = 0.12). Overall, this study showed that ecosystem C and N cycles were generally stimulated by warming, but soil C and N stocks showed minimal responses to warming in the alpine grassland ecosystems on the Tibetan Plateau. This finding suggests that the faster ecosystem C and N cycles under climate warming may not necessarily lead to the loss of soil C and N stocks in the alpine grassland on the Tibetan Plateau. Future studies should conduct long-term coordinated distributed experiments using consistent methods and pay more attention to subsurface soils and the less-studied central and western regions of the Tibetan Plateau.
