Abstract:Antimicrobial resistance (AMR) has emerged as a significant and pressing public health concern, posing serious challenges to effectively preventing and treating persistent diseases. Despite various efforts made in recent years to address this problem, the global trends of AMR continue to escalate without any indication of decline. As AMR is well-known for antibiotics, developing new materials such as metal containing compounds with different mechanisms of action is crucial to effectively address this challenge. Copper, silver, and chitosan in various forms have demonstrated significant biological activities and hold promise for applications in medicine and biotechnology. Exploring the biological properties of these nanoparticles is essential for innovative therapeutic approaches in treating bacterial and fungal infections, cancer, and other diseases. To this end, the present study aimed to synthesize silver@copper oxide (Ag@CuO) nanoparticles and its chitosan nanocomposite (Chi-Ag@CuO) to investigate their antimicrobial efficacy. Various established spectroscopic and microscopic methods were employed for characterization purposes, encompassing scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR). Subsequently, the antimicrobial activity of the nanoparticles was assessed through MIC (minimum inhibitory concentration), MBC (minimum bactericidal concentration), and well-disk diffusion assays against Pseudomonas aeruginosa, Acinetobacter baumannii Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans. The size of the CuO-NPs, Ag@CuO, and Chi-Ag@CuO NPs was found to be 70-120 nm with a spherical shape and an almost uniform distribution. The nanocomposites were found to possess a minimum inhibitory concentration (MIC) of 5 μg/mL and a minimum bactericidal concentration (MBC) of 250 μg/mL. Moreover, these nanocomposites generated varying clear inhibition zones, with diameters ranging from a minimum of 9 ± 0.5 mm to a maximum of 25 ± 0.5 mm. Consequently, it is evident that the amalgamation of copper-silver-chitosan nanoparticles has exhibited noteworthy antimicrobial properties in the controlled laboratory environment, surpassing the performance of other types of nanoparticles.

Chemical Synthesis of Innovative Silver Nanohybrids with Synergistically Improved Antimicrobial Properties.

Abstract:Background:The wide use of antibiotics has created challenges related to antibiotic-resistant bacteria, which have been increasingly found in recent decades. Antibiotic resistance has led to limited choices of antibiotics. Multiple old antimicrobial agents have high antimicrobial properties toward bacteria, but they unfortunately also possess high toxicity toward humans. For instance, silver (Ag) compounds were frequently used to treat tetanus and rheumatism in the 19th century and to treat colds and gonorrhea in the early 20th century. However, the high toxicity of Ag has limited its clinical use.Purpose:We aimed to reformulate Ag to reduce its toxicity toward human cells like osteoblasts and to optimize its antimicrobial properties.Results:Ag, an old antimicrobial agent, was reformulated by hybriding nanomaterials of different dimensions, and silver nanoparticles (AgNPs) of controllable sizes (95-200 nm) and varying shapes (cube, snowflake, and sphere) were synthesized on carbon nanotubes (CNTs). The obtained AgNP-CNT nanohybrids presented significantly higher killing efficacy against Staphylococcus aureus (S. aureus) compared to AgNPs at the same molar concentration and showed synergism in killing S. aureus at 0.2 and 0.4 mM. AgNPs presented significant osteoblast toxicity; in contrast, AgNP-CNT nanohybrids demonstrated significantly enhanced osteoblast viability at 0.04-0.8 mM. The killing of S. aureus by AgNP-CNT nanohybrids was fast, occurring within 15 min.Conclusion:Ag was successfully reformulated and Ag nanohybrids with various AgNP shapes on CNTs were synthesized. The nanohybrids presented significantly enhanced antimicrobial properties and significantly higher osteoblast cell viability compared to AgNPs, showing promise as an innovative antimicrobial nanomaterial for a broad range of biomedical applications.
Hybrid materials based on chitosan functionalized with green synthesized copper nanoparticles: Physico-chemical and antimicrobial analysis

Abstract:Recently, the development of materials with antimicrobial properties has become a challenge under scrutiny. The incorporation of copper nanoparticles (NpCu) into a chitosan matrix appears to represent a viable strategy to contain the particles and prevent their oxidation. Regarding the physical properties, the nanocomposite films (CHCu) showed a decrease in the elongation at break (5 %) and an increase in the tensile strength of 10 % concerning chitosan films (control). They also showed solubility values lower than 5 % while the swelling diminished by 50 %, on average. The dynamical mechanical analysis (DMA) of nanocomposites revealed two thermal events located at 113° and 178 °C, which matched the glass transitions of the CH-enriched phase and nanoparticles-enriched phase, respectively. In addition, the thermogravimetric analysis (TGA) detected a greater stability of the nanocomposites. Chitosan films and the NpCu-loaded nanocomposites demonstrated excellent antibacterial capacity against Gram-negative and Gram-positive bacteria, proved through diffusion disc, zeta potential, and ATR-FTIR techniques. Additionally, the penetration of individual NpCu particles into bacterial cells and the leakage of cell content were verified by TEM. The mechanism of the antibacterial activity of the nanocomposites involved the interaction of chitosan with the bacterial outer membrane or cell wall and the diffusion of the NpCu through the cells. These materials could be applied in diverse fields of biology, medicine, or food packaging.

Abstract:The production of silver nanoparticles (Ag NPs) is important for antibacterial and antifungal applications. Many studies utilized natural resources (e.g., biopolymers and rice husk silica) for Ag NPs' synthesis and stabilization, but few of them reported the effect of surface charge on the products' antifungal performance. This study uses biopolymers (i.e., chitosan (CTS) and carboxymethyl cellulose (CMC)) to synthesize Ag NPs and investigate their impact on Ag NPs morphologies, surface charge, and antifungal properties. Two functional nanocomposites (NCs) based on Ag NPs were developed via a chemical method using either (i) CTS or CMC as a stabilizing agent and (ii) rice husk silica as a supported material. Both stabilizers produce well-dispersed Ag NPs on the silica surface, with CTS-stabilized NCs showing a positive surface charge (51.1 mV) and CMC-stabilized NCs exhibiting a negative surface charge (− 64.1 mV). Antifungal experiments revealed that both NCs effectively inhibited the mycelial growth of P. palmivora and P. oryzae . The positively charged NC exhibited the strongest activity against P. palmivora , whereas the negatively charged NC was the most effective against P. oryzae . These results emphasize the benefit of using stabilizing agents to boost the antifungal performance of Ag NPs.

Dialdehyde cellulose nanofibrils/polyquaternium stabilized ultra-fine silver nanoparticles for synergistic antibacterial therapy

Abstract:Dialdehyde cellulose nanofibrils (DACNF) and Polyquaternium-10 (PQ: chloro-2-hydroxy-3-(trimethylamino) propyl polyethylene glycol cellulose) have become increasingly favored as antibacterial substances due to their advantageous characteristics. DACNF exhibits exceptional mechanical properties and biocompatibility, whereas PQ demonstrates a positive charge that enhances its antibacterial activity. Combined in a DACNF/PQ mixture, they provide an excellent template material for preparing and stabilizing ultra-fine (~ 10.3 nm) silver nanoparticles (AgNPs) at room temperature. Here, the dialdehyde group of DACNF functions as a reducing agent, while the quaternary ammonium of PQ and carboxylate groups of DACNF synergistically helped in-situ generation of AgNPs uniformly. The synthesized nanocomposites, namely PQ@AgNPs, AgNPs@DACNF, and AgNPs@DACNF/PQ, were subjected to comprehensive characterization using various advanced analytical techniques. The films containing AgNPs@DACNF and AgNPs@DACNF/PQ, fabricated via vacuum filtration, exhibited excellent mechanical properties of 9.78 ± 0.21 MPa, and demonstrated superior antibacterial activity against both Escherichia coli and Staphylococcus aureus. Additionally, the silver ion leaching from the prepared composite films was well controlled. The fabricated nanocomposites also effectively inhibited bacterial biofilm formation. It was also found to be highly biocompatible and non-toxic to human skin fibroblast cells. Furthermore, the nanocomposites exhibited enhanced migration of human dermal fibroblasts, suggesting their potential in facilitating wound healing processes.
Selective uptake of glutamine in the gastrointestinal tract: Confirmation in a human study

Abstract:Recent animal data suggest that the gut plays a far more important metabolic role than previously thought. During critical illness, disruption in bowel barrier function may result in a chronic hypermetabolic state and contribute to multiorgan failure. Animal studies have demonstrated that enterocytes of the gastrointestinal tract use glutamine as a respiratory fuel and during critical illness the consumption of glutamine by the gut significantly increases. The selective uptake of glutamine by the gut, to date, has not been confirmed in humans. Seven patients who sustained multisystem trauma necessitating laparotomy underwent portal venous catheterization. This was done by carefully reopening the obliterated umbilical vein and facilitating access to the left branch of the portal vein using a standard central venous catheter. Portal venous and systemic blood samples were recorded for 5 days after operation. Amino acid levels in both circulations were recorded at 48 h and 5 days. Using Student's t test for related samples, the differences between individual amino acids in portal and systemic circulations were compared. At 48 h, mean(s.d.) portal venous glutamine was 85(5) per cent of the systemic levels (253(80) compared with 296(90) μmol/ml, P<0·002). At 5 days, portal glutamine was 87(3) per cent of the systemic levels (255(69) compared with 292(83) μmol/ml, P<0·003). Levels of citrulline, a breakdown product of glutamine metabolism, were elevated in the portal venous circulation at 48 h (20(4) compared with 16(3) μmol/ml, P<0·005) and at 5 days (21(5) compared with 14(3) μmol/ml, P<0·002). No significant differences between any of the other amino acids analysed were identified. This study confirms, for the first time in humans, that selective uptake of glutamine occurs in the gut. In stressed states, glutamine deficiency is associated with gut mucosal atrophy. This has significant implications as glutamine is not provided in most commercially available parenteral and enteral nutrition formulations.
Synthesis of Biodegradable and Antimicrobial Nanocomposite Films Reinforced for Coffee and Agri-Food Product Preservation

Abstract:The antimicrobial activity of silver nanoparticles is widely known. However, their application to biodegradable polymeric materials is still limited. In this work, we report a strategy involving the green synthesis of nanocomposite films based on a natural biodegradable matrix. Nanometer-sized silver nanoparticles (C-AgNPs) were synthesized with the aid of ultrasound waves between the silver nitrate solution and the nanocurcumin solution. The green synthesized C-AgNPs were found to have particle sizes in the range of 5-25 nm and demonstrated good antimicrobial activity against Clostridium perfringens, Staphylococcus aureus, Bacillus subtilis, Macrophoma theicola, and Aspergillus flavus. Owing to their physical-chemical and mechanical properties and the excellent antimicrobial activities, the obtained AgNPs were used together with chitosan, cassava starch, and poly(vinyl alcohol) (PVA) to make nanocomposite films, which are suitable for the packaging requirements of various key agricultural and food products such as coffee beans, bamboo straws, and fruits. The nanocomposite films lost up to 85% of their weight after being buried in the soil for 120 days. This indicates that the films made with natural biodegradable materials are environmentally friendly.

Abstract:The aim of this investigation was to prepare novel hybrid materials with enhanced antimicrobial properties to be used in food preservation and packaging applications. Therefore, nanocomposite materials were synthesized based on two stimuli-responsive oligo(ethylene glycol methacrylate)s, namely PEGMA and PEGMEMA, the first bearing hydroxyl side groups with three different metal nanoparticles, i.e., Ag, TiO2 and ZnO. The in situ radical polymerization technique was employed to ensure good dispersion of the nanoparticles in the polymer matrix. FTIR spectra identified the successful preparation of the corresponding polymers and XRD scans revealed the presence of the nanoparticles in the polymer matrix. In the polymer bearing hydroxyl groups, the presence of Ag-NPs led to slightly lower thermal stability as measured by TGA, whereas both ZnO and TiO2 led to nanomaterials with better thermal stability. The antimicrobial activity of all materials was determined against the Gram-negative bacteria E. coli and the Gram-positive S. aureus, B. subtilis and B. cereus. PEGMEMA nanocomposites had much better antimicrobial activity compared to PEGMA. Ag NPs exhibited the best inhibition of microbial growth in both polymers with all four bacteria. Nanocomposites with TiO2 showed a very good inhibition percentage when used in PEGMEMA-based materials, while in PEGMA material, high antimicrobial activity was observed only against E. coli and B. subtilis, with moderate activity against B. cereus and almost absent activity against S. aureus. The presence of ZnO showed antimicrobial activity only in the case of PEGMEMA-based materials. Differences observed in the antibacterial activity of the polymers with the different nanoparticles could be attributed to the different structure of the polymers and possibly the more efficient release of the NPs.

Abstract:Research on preparing material via a greener approach paved the way to produce highly biocompatible nanoparticles for antibacterial and antifungal applications. These nanoparticles showed very good activity on some of the microbial elements; hence, it is used in food industries and food quality control divisions. The preparation of CuO, MgO, and CuO/MgO nanocomposite is discussed and its activity on some of the Gram-positive and Gram-negative programs is investigated. In this work, CuO, MgO, and CuO/MgO nanocomposite are prepared by using Madagascar periwinkle extract. The obtained microwave-irradiated powder samples are analyzed to identify their physio-chemical properties. The XRD pattern confirmed the phase purity and the sample of the crystalline nature. The UV-vis absorption spectra depict the band gap of CuO, MgO, and CuO/MgO nanocomposite. The presence of functional groups was confirmed using FTIR analysis. The agglomerated sphere-like morphology of the samples was confirmed by HRSEM analysis. According to the antibacterial investigation, pristine nanoparticles and their composite counterparts effectively combat both Gram-negative and Gram-positive bacteria. Therefore, the green synthesized CuO, MgO, and CuO/MgO nanocomposite are analyzed for antibacterial applications.

Abstract:Antimicrobial coatings are becoming increasingly popular in functional material modification and are essential in addressing microbial infection challenges. In this study, the phytochemical and antimicrobial potential of aqueous, 80% methanol and 80% ethanol pod extracts of Acacia concinna (Willd.) DC (AC) and its application in the green in situ (one pot) synthesis of silver nanoparticles on Cellulose nano fibrils (CNF) and Waterborne polyurethane (WPU) were prepared. The phytochemical evaluation of Acacia concinna crude extracts showed the presence of alkaloids, flavonoids, phenols, tannins, terpenoids, saponins, steroids. The surface plasmon Resonance peak of CNF/AC-AgNPs was 450 nm and the FTIR result confirmed functional groups such as carbonyl, phenols and carboxyl were present which was important for the bio-reduction of silver nanoparticles. The crude AC aqueous pods extract against Gram-positive and Gram-negative bacteria compared with AC ethanol and AC methanol extracts. The WPU/CNF/AC-AgNPs composite dispersion was also good in terms of its antibacterial activities. The WPU/CNF/AC-AgNPs nanocomposites could be applied as bifunctional nanofillers as an antimicrobial agent in food packaging systems and other biological applications.

Abstract:Silver carbonate and elemental silver nanoparticles embedded in the matrix of biological materials were successfully prepared via a bio-mechanochemical synthesis, i.e., one-step solvent-free approach utilizing high-energy ball milling. Namely, the powders of silver nitrate as a precursor of silver and a mixture of natural materials (eggshell waste and Thymus serpyllum L. plant) powders as reducing and stabilizing agents, were milled together. The optimization of experimental conditions was performed using a Taguchi design. The presence of nanoparticles was confirmed by a bunch of experimental techniques including XRD and TEM, their size being up to 15 nm. By means of AAS, the level of Ag stabilization was determined, being better when eggshell content was higher. The products also exhibited the antibacterial activity, and although the agar well diffusion method did show better results for the nanoparticles prepared using the eggshell or the plant alone, the respiratory tests have shown a good response also for the product prepared using a combination of the two. The strongest respiratory inhibition against Escherichia coli was observed for the nanoparticles prepared using only eggshell, but the ones synthesized using both reducing agents caused a prolonged inhibition against Staphylococcus aureus. The proposed synthetic approach offers the use of environmentally harmless technique and natural materials to yield antibacterially active nanoparticles.

Pre-exposure of rat anterior pituitary cells to somatostatin enhances subsequent growth hormone secretion.

Abstract:The precise roles of GH-releasing factor (GRF) and somatostatin (SRIF) in the orchestration of pulsatile GH secretion have not yet been fully determined. We examined the interactions of rat GRF and SRIF in the concentration ranges present in rat hypophysial-portal blood, on the secretion of GH from dispersed male rat anterior pituitary cells in monolayer culture. The effects of exposing cells to GRF and/or SRIF (0.01-1.nmol/l) for 1 h were compared with the effects of preincubation of cells with SRIF before experimental incubations. As anticipated, the stimulatory effects of 0.1-1 nmol GRF/1 were abolished by concurrent incubation with SRIF at an equimolar concentration, although SRIF, at these concentrations, did not significantly inhibit basal GH secretion. Conversely, pre-exposure to 0.1 nmol SRIF/1 for 30 or 60 min, resulted in an increase in GH secretion during a subsequent 60-min incubation period, both in the absence or in the presence of GRF (0.01-1 nmol/l). Pretreatment with GRF caused increased responsivity to GRF rather than significant sensitization of the GH response to GRF. These observations demonstrate actions of SRIF, at low and probably physiological concentrations, which are more complex than those of a pure inhibitor of GH secretion. Pre-exposure of the pituitary to SRIF enhances subsequent GH secretion, suggesting that SRIF may play an additional physiological role in amplifying the GRF signal.

Abstract:Abstract Nanomaterials have significantly contributed in the field of nanomedicine as this subject matter has combined the usefulness of natural macromolecules with organic and inorganic nanomaterials. In this respect, various types of nanocomposites are increasingly being explored in order to discover an effective approach in controlling high morbidity and mortality rate that had triggered by the evolution and emergence of multidrug resistant microorganisms. Current research is focused towards the production of biogenic silver nanoparticles for the fabrication of antimicrobial metallic-polymer-based non-cytotoxic nanocomposite system. An ecofriendly approach was adapted for the production of silver nanoparticles using fungal biomass ( Aspergillus fumigatus KIBGE-IB33). The biologically synthesized nanoparticles were further layered with a biodegradable macromolecule (chitosan) to improve and augment the properties of the developed nanocomposite system. Both nanostructures were characterized using different spectrographic analyses including UV–visible and scanning electron microscopy, energy dispersive X-ray analysis, dynamic light scattering, and Fourier transform infrared spectroscopic technique. The biologically mediated approach adapted in this study resulted in the formation of highly dispersed silver nanoparticles that exhibited an average nano size and zeta potential value of 05 nm (77.0%) and − 22.1 mV, respectively with a polydispersity index of 0.4. Correspondingly, fabricated silver–chitosan nanocomposites revealed a size of 941 nm with a zeta potential and polydispersity index of + 63.2 mV and 0.57, respectively. The successful capping of chitosan on silver nanoparticles prevented the agglomeration of nanomaterial and also facilitated the stabilization of the nano system. Both nanoscopic entities exhibited antimicrobial potential against some pathogenic bacterial species but did not displayed any antifungal activity. The lowest minimal inhibitory concentration of nanocomposite system (1.56 μg ml −1 ) was noticed against Enterococcus faecalis ATCC 29212. Fractional inhibitory concentration index of the developed nanocomposite system confirmed its improved synergistic behavior against various bacterial species with no cytotoxic effect on NIH/3T3 cell lines. Both nanostructures, developed in the present study, could be utilized in the form of nanomedicines or nanocarrier system after some quantifiable trials as both of them are nonhazardous and have substantial antibacterial properties.

Abstract:Antimicrobial resistance has become a major problem over the years and threatens to remain in the future, at least until a solution is found. Silver nanoparticles (Ag-NPs) and antimicrobial polymers (APs) are known for their antimicrobial properties and can be considered an alternative approach to fighting resistant microorganisms. Hence, the main goal of this research is to shed some light on the antimicrobial properties of Ag-NPs and APs (chitosan (CH), poly-L-lysine (PLL), ε-poly-L-lysine (ε-PLL), and dopamine (DA)) when used alone and complexed to explore the potential enhancement of the antimicrobial effect of the combination Ag-NPs + Aps. The resultant nanocomplexes were chemically and morphologically characterized by UV-visible spectra, zeta potential, transmission electron microscopy, and Fourier-transform infrared spectroscopy. Moreover, the Ag-NPs, APs, and Ag-NPs + APs nanocomplexes were tested against Gram-positive Staphylococcus aureus (S. aureus) and the Gram-negative Escherichia coli (E. coli) bacteria, as well as the fungi Candida albicans (C. albicans). Overall, the antimicrobial results showed potentiation of the activity of the nanocomplexes with a focus on C. albicans. For the biofilm eradication ability, Ag-NPs and Ag-NPs + DA were able to significantly remove S. aureus preformed biofilm, and Ag-NPs + CH were able to significantly destroy C. albicans biofilm, with both performing better than Ag-NPs alone. Overall, we have proven the successful conjugation of Ag-NPs and APs, with some of these formulations showing potential to be further investigated for the treatment of microbial infections.

Abstract:Green synthesis of metal nanoparticles is an attractive substitute for traditional methods using capping and reducing chemicals. In this study, silver nanoparticles (AgNPs) were synthesised using carbon dots (CDs) derived from bioresources as reducing, protecting, and stabilising agents in a single step using environmentally friendly and cost-effective synthetic methods. The optical and structural properties of prepared CD/AgNPs were explored using UV-vis (Ultraviolet-Visible Spectroscopy), Fluorescence spectroscopy, XRD (X-Ray Diffraction), DLS (Dynamic Light Scattering), SEM-EDX (Scanning Electron Microscopy with Energy-Dispersive X-ray Spectroscopy) and TEM (Transmission Electron Microscopy). The synthesised CD/AgNPs are stable as zeta potential value is -14.7mV. From TEM the particle size exhibited as ~12 nm. The prepared CD/AgNPs exhibited significant optical absorbance, good water dispersibility, stability and nano size. Also, CD/AgNPs revealed good biocidal effects against Gram-negative bacteria Escherichia coli (E. coli), Pseudomonas Aeruginosa (P. aeruginosa), Gram-positive Staphylococcus Aureus (S. aureus), Bacillus Cereus (B. Cereus), and good anti-fungal activity against Aspergillus Niger (A. niger). The CD/AgNPs were further analyzed by live/dead assay. E. coli and A. niger with zone of inhibition around 3.1 and 40 mm, respectively when compared to ciprofloxacin (2.2 mm) and fluconazole (25 mm). The above investigation proved that the developed CD/AgNPs will be a new platform as an alternative to the traditional antibiotics for the generation of new kind of antibacterial materials and also provide the pathway for various metal/CD nanomaterials for diverse biomedical applications.

Abstract:Copper is a cheaper alternative to various noble metals with a range of potential applications in the field of nanoscience and nanotechnology. However, copper nanoparticles have major limitations, which include rapid oxidation on exposure to air. Therefore, alternative pathways have been developed to synthesize metal nanoparticles in the presence of polymers and surfactants as stabilizers, and to form coatings on the surface of nanoparticles. These surfactants and polymeric ligands are made from petrochemicals which are non- renewable. As fossil resources are limited, finding renewable and biodegradable alternative is promising.The study aimed at preparing, characterizing and evaluating the antibacterial properties of copper nanoparticles. Copper nanoparticles were prepared using gelatin biopolymer, CuSO4.5H2O ions and hydrazine as stabilizer, precursor salt and reducing agent respectively. However, vitamin C and NaOH solution were also employed as an antioxidant and pH adjuster. The synthesized copper nanoparticles were characterized using UV-visible spectroscopy (UV-vis), thermogravimetric analysis (TGA), zeta potential measurements powder, X-ray diffraction (XRD), field emission scanning electron microscope and transmission electron microscope (TEM). The UV-visible absorption spectrum confirms the formation of the CuNPs, which showed maximum absorbance at 583 nm. Results obtained from TEM indicated a decrease in size of particle from a low concentration to high concentration of the supporting materials. The optimum concentration of gelatin was found to be 0.75 wt%. The supporting materials used for this synthesis are biocompatible and the obtained products are stable in air. The synthesized CuNPs display promising antibacterial activities against B. subtilis (B29), S. aureus (S276), S. choleraesuis (ATCC 10708) and E. coli (E266) as gram positive and negative bacteria respectively.
Lignin-facilitated growth of Ag/CuNPs on surface-activated polyacryloamidoxime nanofibers for superior antibacterial activity with improved biocompatibility

Abstract:Introduction: Nanofibers are one of the role-playing innovations of nanotechnology. Their high surface-to-volume ratio allows them to be actively functionalized with a wide range of materials for a variety of applications. The functionalization of nanofibers with different metal nanoparticles (NPs) has been studied widely to fabricate antibacterial substrates to battle antibiotic-resistant bacteria. However, metal NPs show cytotoxicity to living cells, thereby restricting their application in biomedicine. Objectives: To minimize the cytotoxicity of NPs, biomacromolecule lignin was employed as both a reducing and capping agent to green synthesize silver (Ag) and copper (Cu) NPs on the surface of highly activated polyacryloamidoxime nanofibers. The activation of polyacrylonitrile (PAN) nanofibers via amidoximation was employed for enhanced loading of NPs to achieve superior antibacterial activity. Methodology: At first, electrospun PAN nanofibers (PANNM) were activated to produce polyacryloamidoxime nanofibers (AO-PANNM) by immersing PANNM in a solution of Hydroxylamine hydrochloride (HH) and Na2CO3 under controlled conditions. Later, Ag and Cu ions were loaded by immersing AO-PANNM in different molar concentrations of AgNO3 and CuSO4 solutions in a stepwise manner. The reduction of Ag and Cu ions into NPs to fabricate bimetal-coated PANNM (BM-PANNM) was carried out via alkali lignin at 37 °C for 3 h in a shaking incubator with ultrasonication every 1 h. Results: AO-APNNM and BM-PANNM hold their nano-morphology except for some changes in fiber orientation. XRD analysis demonstrated the formation of Ag and CuNPs as evident from their respective spectral band. Maximum 8.46 ± 0.14 wt% and 0.98 ± 0.04 wt% Ag and Cu species were loaded on AO-PANNM, respectively as revealed by ICP spectrometric analysis. The hydrophobic PANNM turned into super hydrophilic, having WCA of 14 ± 3.32° after amidoximation which further reduced to 0° for BM-PANNM. However, the swelling ratio of PANNM reduced from 13.19 ± 0.18 g/g to 3.72 ± 0.20 g/g for AO-PANNM. Even at the third cycle test against S. aureus strains, 0.1Ag/Cu-PANNM, 0.3Ag/Cu-PANNM, and 0.5Ag/Cu-PANNM displayed bacterial reduction of 71.3 ± 1.64 %, 75.2 ± 1.91 %, and 77.24 ± 1.25 %, respectively. On 3rd cycle test against E. coli, above 82 % bacterial reduction was noticed for all BM-PANNM. Amidoximation increased COS-7 cell viability up to 82 %. The cell viability of 0.1Ag/Cu-PANNM, 0.3Ag/Cu-PANNM, and 0.5Ag/Cu-PANNM was found to be ∼68 %, ∼62, and 54 %, respectively. In LDH assay, almost no release of LDH was detected, suggesting the compatibility of the cell membrane in contact with BM-PANNM. The improved biocompatibility of BM-PANNM even at higher loading (%) of NPs must be ascribed to the controlled release of metal species in the early stage, antioxidant, and biocompatible lignin capping of NPs. Conclusions: BM-PANNM displayed superior antibacterial activity against E. coli and S. aureus bacterial strains and acceptable biocompatibility of COS-7 cells even at higher loading (%) of Ag/CuNPs. Our findings suggest that BM-PANNM can be used as a potential antibacterial wound dressing and other antibacterial applications where sustained antibacterial activity is needed.
Dialdehyde Cellulose Solution as Reducing Agent: Preparation of Uniform Silver Nanoparticles and In Situ Synthesis of Antibacterial Composite Films with High Barrier Properties

Abstract:The demand for antimicrobial materials is gradually increasing due to the threat of infections and diseases caused by microorganisms. Silver nanoparticles (AgNPs) are widely used because of their broad-spectrum antimicrobial properties, but their synthesis methods are often environmentally harmful and AgNPs difficult to isolate, which limits their application in several fields. In this study, an aqueous solution of dialdehyde cellulose (DAC) was prepared and used as a reducing agent to synthesize AgNPs in an efficient and environmentally friendly process. The synthesized AgNPs can be easily separated from the reducing agent to expand their applications. In addition, the AgNPs were immobilized in situ on dialdehyde cellulose to form antibacterial composite films. The results showed that the prepared silver nanoparticles were mainly spherical and uniformly dispersed, with an average size of about 25 nm under optimal conditions. Moreover, the dialdehyde cellulose–nanosilver (DAC@Ag) composite films had excellent mechanical properties, positive transparency, ultraviolet-blocking properties, and effective antibacterial activity against E. coli and S. aureus. Notably, the composite films exhibited excellent oxygen and water vapor barrier properties, with WVT and ORT of 136.41 g/m2·24 h (30 °C, 75% RH) and <0.02 cm3/m2·24 h·0.1 MPa (30 °C, 75% RH), respectively, better than commercial PE films. Hence, this study not only provides an environmentally friendly method for the preparation of silver nanoparticles, but also offers a simple and novel strategy for the in situ synthesis of silver-loaded antibacterial composite films.
