Abstract:Lonicera japonica Thunb. is a widely distributed plant with ornamental, economic, edible, and medicinal values. L. japonica is a phytoantibiotic with broad-spectrum antibacterial activity and a potent therapeutic effect on various infectious diseases. The anti-diabetic, anti-Alzheimer's disease, anti-depression, antioxidative, immunoregulatory, anti-tumor, anti-inflammatory, anti-allergic, anti-gout, and anti-alcohol-addiction effects of L. japonica can also be explained by bioactive polysaccharides isolated from this plant. Several researchers have determined the molecular weight, chemical structure, and monosaccharide composition and ratio of L. japonica polysaccharides by water extraction and alcohol precipitation, enzyme-assisted extraction (EAE) and chromatography. This article searched in the Chinese Pharmacopoeia, Flora of China, Web of Science, PubMed, and CNKI databases within the last 12 years, using "Lonicera. japonica polysaccharides", "Lonicera. japonica Thunb. polysaccharides", and "Honeysuckle polysaccharides" as the key word, systematically reviewed the extraction and purification methods, structural characteristics, structure-activity relationship, and health benefits of L. japonica polysaccharides to provide insights for future studies. Further, we elaborated on the potential applications of L. japonica polysaccharides in the food, medicine, and daily chemical industry, such as using L. japonica as raw material to make lozenges, soy sauce and toothpaste, etc. This review will be a useful reference for the further optimization of functional products developed from L. japonica polysaccharides.

Abstract:Lonicera japonica Thunb. flower is a well-known medicinal plant that has been widely used for the treatment of human disease. To explore the molecular mechanisms underlying the biological activities of L. japonica immature flower buds, a gel-free/label-free proteomic technique was used in combination with combinatorial peptide ligand libraries (CPLL) and polyethylene glycol (PEG) fractionation for the enrichment of low-abundance proteins and removal of high-abundance proteins, respectively. A total of 177, 614, and 529 proteins were identified in crude protein extraction, CPLL fractions, and PEG fractions, respectively. Among the identified proteins, 283 and 239 proteins were specifically identified by the CPLL and PEG methods, respectively. In particular, proteins related to the oxidative pentose phosphate pathway, signaling, hormone metabolism, and transport were highly enriched by CPLL and PEG fractionation compared to crude protein extraction. A total of 28 secondary metabolism-related proteins and 25 metabolites were identified in L. japonica immature flower buds. To determine the specificity of the identified proteins and metabolites for L. japonica immature flower buds, Cerasus flower buds were used, which resulted in the abundance of hydroxymethylbutenyl 4-diphosphate synthase in L. japonica immature flower buds being 10-fold higher than that in Cerasus flower buds. These results suggest that proteins related to secondary metabolism might be responsible for the biological activities of L. japonica immature flower buds.

Abstract:Lonicera Linn.is an important genus of the familyCaprifoliaceaecomprising of approximately 200 species, and some species of which have been usually used in traditional Chinese medicine for thousands of years. Some species of this genus can also be used in functional foods, cosmetics and other applications. The saponins, as one of most important bioactive components of theLonicera Linn.genus, have attracted the attention of the scientific community. Thus, a comprehensive and systematic review on saponins from the genus is indispensable. In this review, 87 saponins and sapogenin from the genus ofLonicera Linn., together with their pharmacological activities including hepatoprotective, anti-inflammatory, anti-bacterial, anti-allergic, anti-tumor, and immunomodulatory effects, and hemolytic toxicity were summarized.

Abstract:Lonicera japonica Thunb. is an important medicinal plant. The secondary metabolites in L. japonica are diverse and vary in levels during development, leading to the ambiguous evaluation for its medical value. In order to reveal the regulatory mechanism of secondary metabolites during the flowering stages, transcriptomic, proteomic, and metabolomic analyses were performed. The integration analysis of omic-data illustrated that the metabolic changes over the flower developmental stages were mainly involved in sugar metabolism, lipopolysaccharide biosynthesis, carbon conversion, and secondary metabolism. Further proteomic analysis revealed that uniquely identified proteins were mainly involved in glycolysis/phenylpropanoids and tricarboxylic acid cycle/terpenoid backbone pathways in early and late stages, respectively. Transketolase was commonly identified in the 5 developmental stages and 2-fold increase in gold flowering stage compared with juvenile bud stage. Simple phenylpropanoids/flavonoids and 1-deoxy-D-xylulose-5-phosphate were accumulated in early stages and upregulated in late stages, respectively. These results indicate that phenylpropanoids were accumulated attributing to the activated glycolysis process in the early stages, while the terpenoids biosynthetic pathways might be promoted by the transketolase-contained regulatory circuit in the late stages of L. japonica flower development. BIOLOGICAL SIGNIFICANCE: Lonicera japonica Thunb. is a native species in the East Asian and used in traditional Chinese medicine. Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Lonicera japonica Thunb. (Caprifoliaceae), a widely used traditional Chinese medicine, was known as Jin Yin Hua (Chinese: ), Ren Dong and Japanese honeysuckle. It was taken to treat the exopathogenic wind-heat, epidemic febrile diseases, sores, carbuncles and some infectious diseases. At the same time, Lonicera japonica could be used as healthy food, cosmetics, ornamental groundcover, and so on.AIM OF THE REVIEW: The present paper reviewed the ethnopharmacology, the biological activities, toxicology and phytochemistry of Lonicera japonica.MATERIALS AND METHODS: Information on Lonicera japonica was gathered via the Internet (using Google Scholar, Baidu Scholar, Elsevier, ACS, Medline Plus, CNKI and Web of Science) and libraries. Additionally, information also was obtained from some local books and brilliant scholars on ethnopharmacology.RESULTS: More than 140 chemical compounds have been isolated, and the main compositions are essential oils, organic acids and flavones, etc. Lonicera japonica and its active principles possess wide pharmacological actions, such as anti-inflammatory, antibacterial, antiviral, antioxidative and hepatoprotective activities.CONCLUSIONS: As an important traditional Chinese medicine, further studies on Lonicera japonica can lead to the development of new drugs and therapeutics for various diseases, and how to utilize it better should be paid more attentions.

Abstract:O. japonicus and L. spicata var. prolifera are distinguished as sources of highly promising yin-tonifying medicinals, namely Ophiopogonis Radix and Liriopes Radix. Liriopes Radix is generally medicinally used as a substitute for Ophiopogonis Radix in various prescriptions due to their extremely similar nature. Ophiopogonis Radix and Liriopes Radix are both very rich in bioactive polysaccharides, especially β‑fructans. Over the past twelve years, except for work on physical entrapment and chemical modification of obtained β‑fructans, the vast majority of studies are carried out to investigate the bioactivities of O. japonicus polysaccharides (OJP) and L. spicata var. prolifera polysaccharides (LSP), mainly including anti-diabetes, immunomodulation, anti-inflammation, antioxidation, anti-obesity, cardiovascular protection, etc. In addition, OJP and LSP are considered to have the potential to regulate intestinal flora. The main purpose of this review is to provide systematically reorganized information on structural characteristics and bioactivities of OJP and LSP to support their further therapeutic potentials and sanitarian functions.

Abstract:A simple ellipsometer and an inexpensive 13-MHz sputteretch assembly have been used with a conventional evaporator to make in, situ measurements of the growth of thin oxide layers (≤ 10 nm) on pure lead and pure indium films, and also on a series of alloy films containing up to 46 at % In. Our lead results agree with results of earlier workers. For pure indium, we find that equilibrium oxide thicknesses are typically smaller than for lead, and that equilibrium time constants are longer; moreover, increased RF power leads to increased, rather than decreased, oxide thicknesses. Alloy films show an intermediate behavior in general, but in the range 26-36 at % In the oxide thickness rises to a maximum before falling to its equilibrium value. We agree with results obtained by other methods of oxidation and analysis which show the oxide to have a disproportionately large In2O3content, and, in particular, that there is 100 percent In2O3for all alloys containing more than 35 at % In. We show that the difference between PbO and In2O3as regards the dependence of oxide thickness on RF power can be accounted for by slightly modifying the oxide growth model of Greiner, to make the diffusion length of oxygen ions very slightly dependent on RF voltage. To account for the intermediate behavior, we introduce a simple coupled Greiner model involving separate columns of two oxides, in which oxygen ions can interdiffuse between columns as they find their way to the alloy surface. As the parameters of the model are changed, the growth behavior changes from a simple monotonic growth of thickness with time, through a behavior in which there is an intermediate maximum, and then back to a monotonic growth behavior again.

Abstract:In this study, a novel polysaccharide, LPP, was obtained from Lonicera caerulea L. pomace by ultrasonic-assisted heating and was purified by Sephadex G-100. The structural characteristics of LPP showed that the molecular weight (Mw) was 8.53 × 104 Da; that it was mainly composed of galacturonic acid, followed by galactose; that it possessed the characteristic functional groups of polysaccharides; and that it had an absence of O-glycosidic bonds and crystalline and triple helix structures. Furthermore, LPP exhibited a favorable thermodynamic stability and antioxidant, hypoglycemic, and hypolipidemic activities in a dose-dependent manner in vitro, demonstrating that LPP can be used as an agent to regulate glycolipid metabolism. Additionally, the relationship between its bio-activities is discussed in this paper. The results revealed that the RP, •OH, and NO2− radicals had synergistic promoting effects, and polysaccharides with a strong antioxidant ability may have excellent hypoglycemic and hypolipidemic effects. Collectively, these results suggest that LPP has a strong bio-activity, and that Lonicera caerulea L. pomace can be used as a potential polysaccharide source.

Abstract:Lonicerae Japonicae Flos (LJF, called Jinyinhua in China), comes from the dried flower buds or flowers to be opened of Lonicera japonica Thunb. in the Lonicera family. It has a long history of medicinal use and has a wide range of application prospects. As modern research advances, an increasing number of scientific experiments have demonstrated the anticancer potential of LJF. However, there is a notable absence of systematic reports detailing the anti-tumor effects of LJF. This review integrates the principles of Traditional Chinese Medicine (TCM) with contemporary pharmacological techniques, drawing upon literature from authoritative databases such as PubMed, CNKI, and WanFang to conduct a comprehensive study of LJF. Notably, a total of 507 compounds have been isolated and characterized from the plant to date, which include volatile oils, organic acids, flavonoids, iridoids, triterpenes and triterpenoid saponins. Pharmacological studies have demonstrated that LJF extract, along with components such as chlorogenic acid, luteolin, rutin, luteoloside, hyperoside and isochlorogenic acid, exhibits potential anticancer activities. Consequently, we have conducted a comprehensive review and summary of the mechanisms of action and clinical applications of these components. Furthermore, we have detailed the pharmacokinetics, quality control, and toxicity of LJF, while also discussing its prospective applications in the fields of biomedicine and preventive healthcare. It is hoped that these studies will provide valuable reference for the clinical research, development, and application of LJF.

Is legacy a myth or a reality? We should know, and we do not.

Abstract:T he concept of cardiovascular risk factor is spontaneously, intuitively, associated with the notion of legacy, because individuals presenting high levels of risk factors will not suffer immediately from this situation. For example, in the pooled analyses of several trials of antihypertensive drugs against placebo or no treatment in mild-to-moderate hypertension [1], stroke incidence was less than 4% at 5 years in the control groups (Fig. 1). Three periods do exist in the disease process: complete latency, symptomatic phase, and cardiovascular complications phase. The symptomatic phase does not exist with cholesterol levels, whereas type 2 diabetes and hypertension are possibly associated with symptoms such as polyuria and polydipsia for the former and headaches for the latter. The duration of the first two phases is highly variable from an individual to another, but the evolution of arterial disease can spread over several years or tens of years, before the occurrence of symptoms of complications. A logical hypothesis comes naturally to mind that the duration of the exposure to high levels of risk factors has per se a prognostic value, the severity of the disease and of the complications being attributed to the legacy from this exposure. We can also view legacy in negative terms regarding treatment exposure when treatment successfully decreases the risk associated with the risk factor: the legacy of treatment consists in a decreased probability of complications extending beyond the period of the exposure to treatment. If the concept of legacy, regarding risk factor or its treatment, correctly captures the reality, physicians should consider it, screening the risk factors and treating them as early as possible. Prolonging treatment exposure as much as possible is usually associated with this reasoning, because the concept of lifelong treatment of risk factors prevails. This conclusion contradicts the current guidelines toward cardiovascular prevention, which base therapeutic decision on risk level through scores that integrates several risk factors rather than on the level of the risk factor targeted by the drug treatment. As age is by far the strongest risk factor, guidelines promote a late treatment initiation, leaving people exposed to a high level of risk factors. The risk is low during this abstention period, but under the legacy theory, the unopposed exposure to high risk factor levels would represent an accumulation of risk for subsequent periods. Of note, legacy effect is a very good concept for increasing the drug market. In this issue of the Journal of Hypertension, Nelson et al. [2] explore to what extent the past exposure to antihypertensive treatment of individuals without cardiovascular history and recruited in the second Australian National Blood Pressure Study was associated with a difference of prognosis. Although they expected that legacy (of unopposed hypertension or of treatment exposure) would lead to a better prognosis in those previously treated, they did not observe any difference in cardiovascular mortality at 10 years. On the contrary, previous treatment was associated during the trial with higher cardiovascular disease, all-cause mortality rate, and incidence of new-onset diabetes. This clearly contradicts the expectations from a legacy effect and supports the strategy of delaying the start of drug treatment in young hypertensive individuals whose risk within 5 or 10 years is low. Considering as a risk factor per se, the duration of the exposure to a risk factor raises the issue of the precise date at which the risk factor was elevated through life. Prescription habits bias the answer to this question: once the high level of risk factor is diagnosed and a drug treatment is started, the latter is exceptionally interrupted by prescribers, who usually consider that treatment is responsible for the good control of the risk factor. Given the large withinindividual variability of risk factors levels, the individuals with false-negative diagnoses of hypertension are submitted to lifelong treatment, without leaving a chance to patients to check whether the treatment is appropriate. This statement concerns blood pressure, glycemia, or cholesterol. A simple simulation illustrates that after 10 medical visits with blood pressure measurements, the individuals presenting the definition criteria of hypertension will include 30% of false positive, or an individual with systolic blood pressure of 130 mmHg will have 64% probability to be misdiagnosed as hypertensive [3]. Abstract:A novel polysaccharide (LJCP-2b) was isolated from Lonicerae Japonicae Caulis, and its structural features were identified by molecular weight distribution, infrared spectrum, monosaccharide composition, methylation analysis and NMR. LJCP-2b was a homogeneous heteropolysaccharide with a molecular weight of 7.0 kDa and composed of glucose, galacturonic acid, galactose, arabinose, rhamnose, xylose, mannose and glucuronic acid. Structural analysis revealed that LJCP-2b was mainly consisted of 1,3,6-β-D-Galp, 1,4-α-D-Glcp, 1,4,6-α-D-Glcp, 1,4-β-D-Galp, 1,2,4-α-L-Rhap and 1,4-α-D-GalpA. In vitro experiments showed that LJCP-2b could affect the function of TNF-α-induced rheumatoid arthritis fibroblast-like synoviocytes (RA-FLS), including weakening cell viability, increasing apoptosis rate, decreased migration number and adhesion capacity, and reduced the levels of IL-6 and IL-1β. These results indicated that LJCP-2b exhibited the activity of inhibiting the hyperproliferation and inflammatory response of RA-FLS, which may be developed for prevention or treatment of RA.

Abstract:Lonicerae Japonicae Flos has been used as a tea and medicine for more than 1,500 years. It has the functions of clearing heat, detoxification, and is often used to treat carbuncle, furuncle, throat arthralgia, erysipelas, heat-toxic blood dysentery, febrile fever. This paper summarizes the botany, ethnopharmacology, chemical composition and pharmacological action of Lonicerae Japonicae Flos from 1986 to 2022, and looks forward to the future research direction of Lonicerae Japonicae Flos. At present, the components isolated from Lonicerae Japonicae Flos include essential oils, organic acids, flavonoids, iridoids, saponins and other compounds. It has the effects of anti-inflammation, anti-virus, anti-bacteria, anti-oxidation, anti-tumor, protect liver and galltesticles, hypotensive, hypolipidemic, anti-thrombosis, anti-allergy, immune regulation and so on. It is often used in clinical treatment of diarrhea, hematochezia, febrile disease, exogenous wind-heat, and cold, swelling and toxin of carbuncle, sore throat and so on. The comprehensive evaluation of the quality of Lonicerae Japonicae Flos and the understanding of multi-target network pharmacology also need to be studied. As a kind of health food with high value, LJF is worthy of further promotion and development.

Protective Role of a New Polysaccharide Extracted from Lonicera japonica Thunb in Mice with Ulcerative Colitis Induced by Dextran Sulphate Sodium

Abstract:Lonicera japonica Thunb is a traditional Chinese herbal medicine for treating intestinal inflammation. The extraction method of Lonicera japonica Thunb polysaccharide (LJP) has been developed previously by our research group. In this study, a Fourier transform infrared spectrometer (FT-IR) was used to perform a qualitative analysis of LJP and a precolumn derivatization high-performance liquid chromatography (HPLC) ((Palo Alto, CA, USA) method was used to explore the monosaccharide composition of LJP. Then, we studied the effect of LJP on the intestinal flora and immune functions of dextran sulfate sodium- (DSS-) induced colitis ulcerative mouse models. The results showed that LJP was consisted of 6 types of monosaccharides and had the characteristic absorption of typical polysaccharides. LJP can increase significantly the weight, organ index, serum cytokines (interleukin, tumor necrosis factor, and interferon-γ), secretory immunoglobulin A (SIgA) concentration, and natural killer (NK) cell and cytotoxic lymphocyte (CTL) activities in DSS-treated mice. The results of intestinal flora showed that a high dose (150 mg kg-1) of LJP had the best effects on improving the intestinal probiotics (Bifidobacterium and Lactobacilli) and antagonizing the pathogenic bacteria (Escherichia coli and Enterococcus). In addition, the measurement results of the spleen lymphocyte apoptosis confirmed from another perspective that LJP had protective effects of immune cells for DSS-treated mice.

Abstract:Laminaria japonica is widely consumed as a key food and medicine. Polysaccharides are one of the most plentiful constituents of this marine plant. In this study, several polysaccharide fractions with different charge numbers were obtained. Their physicochemical properties and anticoagulant activities were determined by chemical and instrumental methods. The chemical analysis showed that Laminaria japonica polysaccharides (LJPs) and the purified fractions LJP0, LJP04, LJP06, and LJP08 mainly consisted of mannose, glucuronic acid, galactose, and fucose in different mole ratios. LJP04 and LJP06 also contained minor amounts of xylose. The polysaccharide fractions eluted by higher concentration of NaCl solutions showed higher contents of uronic acid and sulfate group. Biological activity assays showed that LJPs LJP06 and LJP08 could obviously prolong the activated partial thromboplastin time (APTT), indicating that they had strong anticoagulant activity. Furthermore, we found that LJP06 exerted this activity by inhibiting intrinsic factor Xase with higher selectivity than other fractions, which may have negligible bleeding risk. The sulfate group may play an important role in the anticoagulant activity. In addition, the carboxyl group and surface morphology of these fractions may affect their anticoagulant activities. The results provide information for applications of L. japonica polysaccharides, especially LJP06 as anticoagulants in functional foods and therapeutic agents.

Abstract:At present, the structure-activity relationship of polysaccharides is a common and important focus in the fields of glycobiology and carbohydrate chemistry. To better understand the effect of specific polysaccharide structures on bioactive orientation, four homogeneous polysaccharides from Lycii fructus, one neutral along with three acidic polysaccharides, were purified, structurally characterized and comparatively evaluated on the antioxidative and anti-aging activities. The GC–MS-based monosaccharide composition analysis and methylation results showed that the LFPs had similar glycosyl types but varied proportions. Nuclear magnetic resonance (NMR) spectroscopy showed that LFPs consisted of arabinogalactan, rhamnogalacturonan and homogalacturonan structural domains. The results of the structure-activity relationship indicated that the antioxidative activity was positively correlated with the galacturonic acid (GalA) content, while the neutral multi-branched chains might be responsible for the anti-aging activity. This study is the first time to compare the principal structures and multiple biological activities of LFPs, which provided a reference for the industrial development and deep excavation of the health value of LFPs.

Abstract:Plants of the genus Atractylodes are perennial herbaceous plants in the family Asteraceae, whose rhizome is often used in the production of medicines and health products. There are 6 main species in this genus, namely A. macrocephala , A. lancea , A. chinensis , A. carlinoides , A. koreana and A. japonica . Among them, A. lancea and A. macrocephala are the most extensively investigated. Polysaccharides as the main active ingredients extracted and isolated from plants in this genus, show good pharmacological activities in vivo and in vitro , such as immunomodulatory, antioxidant, antidiabetic and intestinal protective activities. The pharmacological activities of polysaccharides are closely related to their extraction methods and physicochemical properties. This article discusses the extraction and separation methods, molecular weight, monosaccharide composition, chemical structure characteristics and pharmacological activities of polysaccharides from the genus Atractylodes . Furthermore, a comparative analysis of the relationship of monosaccharide composition, relative molecular weight and structural modifications with the pharmacological activities of polysaccharides of the genus Atractylodes was carried out, which provided a reference for the development and utility of polysaccharides.

Abstract:Ethnopharmacological relevance: Lonicerae Japonicae Caulis, the dried stem and branch of Lonicera japonica Thunb., is a Chinese Materia Medica known as Ren Dong Teng in Chinese with long use history in the traditional Chinese medicine (TCM) prescriptions. Lonicerae Japonicae Caulis possesses heat-clearing and detoxifying functions according to the TCM theory. In recent years, a large amount of experimental and clinical studies proved good anti-inflammatory effects of some heat-clearing and detoxifying herbs. The present study aims to reveal the anti-inflammatory property and functional substances of Lonicerae Japonicae Caulis. Materials and methods: For anti-inflammatory activity test, LPS-induced RAW 264.7 macrophages, DSS-induced SPF male C57BL/6J mice model, and LPS-induced SPF male ICR mice model were used in vitro and in vivo, respectively. The behavioral changes, organ damage, and the expression of inflammatory factors such as TNT-α and IL-6 mRNA expression were measured for activity evaluation. Lonicerae Japonicae Caulis samples were prepared by solvent extraction and subsequent column chromatography. The main components were identified and determined using UPLC-UV analysis as well as NMR interpretation after purification. To testify the contribution of main components for the anti-inflammatory activity, different samples were also prepared by compound-knockout strategy. Results: Ethanol extract of Lonicerae Japonicae Caulis could attenuate sickness symptoms in mice such as diarrhea, less activity, and depression. It could also alleviate multiple organ damage, and significantly inhibit the expression of pro-inflammatory factors such as TNF-α, IL-1β, IL-6 and IFN-γ in mice. Furthermore, the isochlorogenic acid-rich and biflavonoid-rich fractions and isochlorogenic acids A and C, and ochnaflavone could significantly down-regulate the mRNA expression of TNF-α and IL-6 in LPS-induced RAW 264.7 macrophages. Conclusions: Lonicerae Japonicae Caulis possesses anti-inflammatory property. Its isochlorogenic acid-rich and biflavonoid-rich fractions do the major contribution. And their main components, isochlorogenic acids A and C, and ochnaflavone, take main responsibility for the anti-inflammatory property.

Abstract:The polysaccharides in honeysuckle leaves (PHL) were separated and characterized for the first time. The nano-emulsion stabilized by PHL and whey protein isolate (WPI) were also fabricated based on the ultrasonic method. The results indicated that PHL was mainly composed of glucose (47.40 mol%), galactose (19.21 mol%) and arabinose (20.21 mol%) with the weight-average molecular weight of 137.97 +/- 4.31 kDa. The emulsifier concentration, WPI-to-PHL ratio, ultrasound power and ultrasound time had significant influence on the droplet size of PHL-WPI nano-emulsion. The optimal preparation conditions were determined as following: emulsifier concentration, 1.7%; WPI/PHL ratio, 3:1; ultrasonic power, 700 W; ultrasonic time, 7 min. Under the above conditions, the median diameter of the obtained nano-emulsion was 317.70 +/- 5.26 nm, close to the predicted value of 320.20 nm. The protective effect of PHL-WPI emulsion on beta-carotene against UV irradiation was superior to that of WPI emulsion. Our results can provide reference for the development of honeysuckle leaves.
