Abstract:Silk has a long history of use in medicine as sutures. To address the requirements of a mechanically robust and biocompatible material, basic research to clarify the role of repeated sequences in silk fibroin in its structures and properties seems important as well as the development of a processing technique suitable for the preparation of fibers with excellent mechanical properties. In this study, three silk-like protein analogs were constructed from two regions selected from among the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(2), the crystalline region of Samia cynthia ricini silk fibroin, (Ala)(12,) the crystalline region of spider dragline silk fibroin, (Ala)(6), and the Gly-rich region of spider silk fibroin, (GGA)(4). The silk-like protein analog constructed from the crystalline regions of the spider dragline silk and B. mori silk fibroins, (A(6)SCS)(8), that constructed from the crystalline regions of the S. c.ricini and B. mori silk fibroins, (A(12)SGS)(4), that constructed from and the crystalline region of S. c.ricini silk fibroin and the glycine-rich region of spider dragline silk fibroin, (A(12)SGS)(4),were expressed their molecular weights being about 36.0 kDa, 17.0 kDa and 17.5 kDa, respectively in E. coli by means of genetic engineering technologies. (A(12)-SCS)(4) and (A(12)SGS)(4) undergo a structural transition from alpha-helix to beta-sheet on a change in the solvent treatment from trifluoroacetic acid (TFA) to formic acid (FA). However, (A(6)SCS)(4) takes on the P-sheet structure predominantly on TFA treatment and FA treatment. Structural analysis was performed on model peptides selected from spider dragline and S. c.ricini silks by means of C-13 CP/MAS NAIR.

Abstract:The C-13 CP/MAS NMR, X-ray diffraction, and Raman spectroscopies were used for monitoring the structural transition of Samia cynthia ricini (S. c. ricini) silk fibroin induced by stretching. Here the silk fibroin was obtained from the aqueous solution stored in the silk gland. All of these spectroscopic data indicate that the structural transition from a-helix to P-sheet occurs with increasing the stretching ratio, especially between the stretching ratios, x 4 and x 6. The C-13 chemical shifts of Ala C peak in the C-13 CP/MAS NMR spectrum change significantly depending on alpha-helix, random coil, and two kinds of beta-sheet structure, which make it possible to clarify the local structure and structural transition. Actually, the fraction of individual structure in the silk fibroin samples was determined from decomposition of the Ala Cbeta peak by assuming a Gaussian line shape. To examine the conformational change of Ala residues of the polyalanine region in S. c. ricini silk fibroin observed with these spectroscopic methods, MD simulations for four peptide molecules, AGGAGG(A)(12)GGAGAG, with alpha-helix conformation were performed in the presence of water molecules under different tensile strengths, and then additional MM calculations were performed after removal of water molecules. The change in the conformational character of Ala residues induced by stretching is explicable by these calculations.
Structure of the Spinning Apparatus of a Wild Silkworm Samia Cynthia Ricini and Molecular Dynamics Calculation on the Structural Change of the Silk Fibroin

Abstract:Silkworms have been developed over thousands years to optimize folding and crystallization of fibroin under highly controlled conditions which have resulted in their efficient fiber formation. In this paper, we reconstructed the three-dimensional architecture of the spinneret of a wild silkworm Samia cynthia ricini from approximately 1000 optical micrographs of the semi-thin cross sections. The chitin plates and muscles were observed in the silk press part together with large change in the diameter of the spinneret lumen at the press part by large shear stress. This is similar to the case of the spinneret of Bombyx mori silkworm, indicating that the structural change in the silk fibroin of S.c. ricini silkworm occurs exclusively at the silk press part due to large shear stresses. Molecular dynamics (MD) calculations were then performed to study the structural change that occurs in the crystalline region of S.c. ricini silk fibroin under shear stress. Namely, using the peptide AGGAGG(A)12GGAGAG as a model of the crystalline part of the silk fibroin under different shear stresses in the presence of water molecules and followed by molecular mechanics (MM) calculation after removal of water molecules. The simulation indicates that the Ala residues in the model peptides adopt a predominantly β-sheet structure under shear stresses of above 1.0GPa.
Development of Silk-Like Materials Based on Bombyx Mori and Nephila Clavipes Dragline Silk Fibroins

Abstract:In order to develop new silk-like materials in the form of fiber and non-woven nano-fiber, this study synthesized a new silk-like protein by selecting the sequence from the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(6), to imitate the processing condition of the silk fibroin with the combination of the sequence YGGLGSQGAGRG, the hydrophilic motif of spider dragline silk which was considered as the origin of supercontraction of spider dragline silk (Yang Z, et al. J Am Chem Soc 2000;122: 9019-25). The CD pattern of the silk-like protein in hexafluoroacetone (HFA) indicates that it takes helical structure. The solid-state structures of the silk-like protein before and after methanol treatment were determined to be random coil and Silk II structure (mainly beta-sheet structure), respectively, using C-13 CP/MAS NMR. The C-13 CP/MAS NMR spectra of the protein after methanol treatment in hydrated state showed that the random coil peaks from Ala C beta and Set C beta carbons become sharper compared with the corresponding peaks in the dry state. The 2D-WISE spectra in the hydrated state also showed increase of the narrow components for these carbons. Thus, water molecules are relatively easy to access at the random coil regions of the protein. The fiber formation of the silk-like protein was possible with wet-spinning or electrospinning methods using HFA as dope solvent and methanol as coagulation solvent. (C) 2008 Elsevier Ltd. Abstract:As a natural protein polymer, Bombyx mori silk fibroin (SF) has superior characteristics such as excellent mechanical properties, biocompatibility and biodegradation which make SF to be a potential candidate in the field of biomaterials. It has been reported that the mineralized SF can be used for bone repair and regeneration in the bone tissue engineering. In order to map out the growth process of hydroxyapatite (HAp) induced by SF in the mineralized buffer solution, the co-solution of SF and supersaturated simulated body fluid (1.5 SBF) was incubated at 37.2 degrees C for 7 days and observed by using TEM, DSC, DTA and TGA in this study. The TEM results indicated that nanoscale crystal nuclei were found along the surface of SF in the initial stage of mineralization and finally these crystals aggregated into three-dimensional network. DSC and DTA results showed that the thermal stability of HAp/SF was greatly improved by increasing incubation duration and that the phase transition of HAp occurred from metastable state to steady state. TGA results suggested the organic content of HAp/SF complex was about 21% which was similar to the natural bone.

Abstract:The mechanical properties of biomaterials are dictated by the interactions and conformations of their building blocks, typically proteins. Although the macroscopic behavior of biomaterials is widely studied, our understanding of the underlying molecular properties is generally limited. Among the noninvasive and label-free methods to investigate molecular structures, infrared spectroscopy is one of the most commonly used tools because the absorption bands of amide groups strongly depend on protein secondary structure. However, spectral congestion usually complicates the analysis of the amide spectrum. Here, we apply polarized two-dimensional (2D) infrared spectroscopy (IR) to directly identify the protein secondary structures in native silk films cast from Bombyx mori silk feedstock. Without any additional peak fitting, we find that the initial effect of hydration is an increase of the random coil content at the expense of the helical content, while the β-sheet content is unchanged and only increases at a later stage. This paper demonstrates that 2D-IR can be a valuable tool for characterizing biomaterials.

Abstract:Two silklike proteins, [TGRGDSPAGG(GAGA GS)(3)AS](5) (FS5) and [TGRGDSPA-(GVPGV)(2)GG(GAGA GS)(3)AS](8) (FES8) were designed to demonstrate the superior performance as biomaterials of silklike proteins. The former protein consists of the crystalline domain sequence, (GAGAGS)n from Bombyx mori silk fibroin and cell-adhesive sequence TGRGDSPA coming from fibronectin-containing RGD triplet. The additional sequence (GVPGV)(n) from elastin was included in the latter protein. The considerably higher cell-adhesion activities of these proteins for NHDF and VERO cells were observed by comparing with those of silklike materials without RGD sequences and also the crystalline fraction of B. mori silk fibroin. This tendency was independent of the treatments, 4.5M LiClO4 or formic acid (FA), on silklike proteins. Their activities are also higher than those of commercial Fibronectin F (TM) for NHDF cell. Their structural characterization was studied using C-13 solid-state NMR. Although the overlapped peaks in usual C-13 CP/MAS NMR spectra make the detailed structural analysis difficult, the methyl resonance regions observed using dipolar dephasing NMR were very useful for the analysis. The presence of both random coil and P-sheet structures was observed in these proteins clearly. The content of P-sheet structure in both proteins increases after FA treatment when compared with the lyophilized samples. The production of electrospun nanofibers from their hexafluoroacetone solution was also tried. The silklike protein FES8 could prepare nonwoven silk fibers although FS5 could not. Nucleation of Hydroxyapatite Crystals by Self-Assembled Bombyx Mori Silk Fibroin

Abstract:Bombyx mori silk fibroin (SF) is known to be capable of facilitating nucleation of the hydroxyapatite crystals (HAps). To find out how SF mediates the nucleation of HAps, self-assembly of SF in 1.5 simulated body fluid (SBF) was observed in this study through design of a co-solution of SF and 1.5-times SBF (SF/1.5 SBF). After the co-solution of SF/1.5 SBF was incubated at 37.2 degrees C up to 7 days, SEM, X-ray, and Fourier transform infrared (FTIR) observations indicated that nucleation of HAps was increased. In addition, the structure of SF was transited from random coil into -sheet indicated by FTIR spectra. The -sheet assembly of SF in 1.5 SBF was also supported by CD spectra. Atomic force microscopy provided detailed progress of the self-assembly that SF incubated in 1.5 SBF was self-assembled in the form from dot, through rod to final net. Direct Observation of Native Silk Fibroin Conformation in Silk Gland of Bombyx mori Silkworm

Abstract:To understand the natural silk spinning mechanism, synchrotron Fourier transform infrared (S-FTIR) microspectroscopy was employed in this study to monitor the conformation changes of silk protein in the silk gland of Bombyx mori silkworm. The ultrahigh brightness of S-FTIR microspectroscopy allowed the imaging of the silk gland with micrometer-scale spatial resolution. Herein, tissue sections of a silk gland, including cross-section slices and longitudinal-section slices, were characterized. The results obtained clearly confirm that the conformation of the silk fibroin changes gradually along the silk gland from the tail to the spinneret. In the middle silk gland, silk fibroin mainly contains random coil/helix conformation. When it comes to the spinneret through the anterior silk gland, the content of β-sheet increases, but the content of random coil/helix instead reduces gradually. Further, the β-sheet distribution in the cross-section of the anterior silk gland was imaged using S-FTIR mapping technique. The results show that the structural distribution of the silk fibroin in cross-section is uniform without significant shell–core structure, which implies that the primary driving force to induce the conformation transition of silk fibroin from random coil/helix to β-sheet during the spinning process is elongational flow of silk fibroin in the silk gland and not the shear force between the silk fibroin and the lumen wall of silk gland. These direct pieces of evidence of silk fibroin structure in the silk gland would definitely promote a deeper understanding of the natural spinning process.

Optical spectroscopy to investigate the structure of regenerated Bombyx mori silk fibroin in solution.

Abstract:Fluorescence and circular dichroism spectroscopy were used to monitor the conformational transition of regenerated Bombyx mori silk fibroin (RSF) in aqueous solutions under different conditions. According to the analysis of fluorescence spectra using anilinonaphthalene-8-sulfonic acid magnesium salt (ANS) as ail external probe, the destruction of the hydrophobic core prior to the secondary structure change suggests that this collapse may initiate the conformational transition from random coil to beta-sheet for RSF. The temperature dependence of the structural changes of RSF, detected by both fluorescence spectroscopy and circular dichroism, shows a reversible process upon heating and recooling, with the midpoint around 45degreesC. The results also indicate that most of the tryptophan (Trp) residues contained in silk fibroin lire concentrated on the surface of the unfolded protein. However, they will change their location in the highly ordered structure (e.g., becoming more homogeneous) with the conformational transition of silk fibroin. Moreover, our studies also suggest that the presence of water plays a crucial role during the structure changes of fibroin.
Structural analysis of Bombyx mori silk fibroin peptides with formic acid treatment using high-resolution solid-state 13C NMR spectroscopy

Abstract:Bombyx mori silk fibroin fiber is a fibrous protein produced by the silkworm at room temperature and from an aqueous solution whose primary structure is highly repetitive. In this study we analyzed the structural characteristics of native peptides, derived from B. mori silk fibroin, with formic acid treatment using high-resolution solid-state 13C NMR. We establish that the Ser residue bearing a short polar side chain has the ability to stabilize the conformation formed in the model peptides due to its ability to form intermolecular hydrogen bonds involving its hydroxyl group as a donor and the carbonyl groups of other residues as acceptors. On the other hand, insertion of Tyr residue in the basic (AG)n and (AGSGAG)n sequence motifs usually exhibited disruptive effects on the preferred conformations. Moreover, the environmental effect was investigated by mixing the native Cp fraction with the model peptides, showing that there is no significant structural difference on the Ser-containing peptides, while structural transformation was observed on the peptides containing the GAAS unit. This may be attributed to the fact that the Cp fraction promotes the formation of an antiparallel beta-sheet in the Ala-Ala unit. Such periodically disrupted ordered structures in the semicrystalline region of B. mori silk fibroin may be critical not only for facilitating the conformational transformation from silk I to silk II structural form but also for having some correlation with the unique properties of the silk materials.
On the Strength of Β-Sheet Crystallites of Bombyx Mori Silk Fibroin

Abstract:Silk fibroin, a natural multi-domain protein, has attracted great attention due to its superior mechanical properties such as ultra-high strength and stretchability, biocompatibility, as well as its versatile biodegradability and processability. It is mainly composed of β-sheet crystallites and amorphous domains. Although its strength is well known to be controlled by the dissociation of protein chains from β-sheet crystallites, the way that water as the solvent affects its strength and the reason that its theoretically predicted strength is several times higher than experimental measurement remain unclear. We perform all-atom molecular dynamics simulations on a β-sheet crystallite of Bombyx mori silk. We find that water solvent reduces the number and strength of hydrogen bonds between β-chains, and thus greatly weakens the strength of silk fibroin. By dissociating protein chains at different locations from the crystallite, we also find that the pulling strength for the interior chains is several times higher than that for the surface/corner chains, with the former being consistent with the theoretically predicted value, while the latter on par with the experimental value. It is shown that the weakest rupture strength controls the failure strength of silk fibre. Hence, this work sheds light on the role of water in the strength of silk fibroin and also provides clues on the origin of the strength difference between theory and experiment.

Abstract:Synchrotron FTIR (S-FTIR) microspectroscopy was used to monitor the silk protein conformation in a range of single natural silk fibers (domestic and wild silkworm and spider dragline silk). With the selection of suitable aperture size, we obtained high-resolution S-FTIR spectra capable of semiquantitative analysis of protein secondary structures. For the first time, we have determined from S-FTIR the β-sheet content in a range of natural single silk fibers, 28 ± 4, 23 ± 2, and 17 ± 4% in Bombyx mori, Antheraea pernyi, and Nephila edulis silks, respectively. The trend of β-sheet content in different silk fibers from the current study accords quite well with published data determined by XRD, Raman, and (13)C NMR. Our results indicate that the S-FTIR microspectroscopy method has considerable potential for the study of single natural silk fibers.
In Situ ATR-FTIR Studies on the β-Sheet Formation of Native and Regenerated Bombyx mori Silk Material in Solution and Its Potential for Drug Releasing Coatings

Abstract:Dynamic attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy at both solutions and coatings of a semicrystalline silk material derived from Bombyx mori was applied to monitor the β-sheet conformation, which is known to correlate with silk protein crystallinity. The secondary structure-sensitive Amide I band was analyzed. Two silk protein samples were studied: native-based silk buffer fibroin (NSF) was extracted from silk glands and regenerated silk fibroin (RSF) was extracted from degummed cocoons. Solutions of both NSF and RSF at 2 mg/mL featured low initial β-sheet contents of 5-12%, which further increased to 47-53% after 24 h. RSF and NSF solutions at 23 mg/mL also featured low initial β-sheet contents of 9-10%, which yet only slightly increased to 16-17% after 24 h. Coatings deposited from RSF solutions showed high surface integrity (Q > 99%) after rinsing in mineralized water, enabling interfacial drug delivery applications. RSF coatings were post-treated with either formic acid (FA) or pure methanol (MeOH) vapor to showcase inducibility of crystalline domains in RSF coatings. Such coatings were loaded with the model antibiotic drugs tetracycline (TCL) and streptomycin (STRP), and the sustained release of TCL was followed in contact with (4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid) (HEPES) buffer. RSF/TCL coatings post-treated with formic acid (FA) vapor followed by methanol (MeOH) vapor showed a significantly lower (52%) initial burst of rather hydrophobic TCL compared to untreated RSF/TCL coatings (72%), while no such significant release difference was observed for hydrophilic STRP. This was rationalized by a specific interaction between nonpolar TCL and hydrophobic crystalline RSF domains.

Abstract:The chemical and conformational structures of Bombyx mori silk have been studied using the complementary techniques of FT-Raman spectroscopy and FT-IR spectroscopy.The FT-Raman spectrum of silk shows strong bands for the photo-sensitive aromatic amino acids tyrosine, tryptophan and phenylalanine. Intensive UV/ozone irradiation decreases Raman intensities of Amide III band and the tyrosine signals due to peptide chain scission of silk polymer and the photo-chemical changes of tyrosine residues on silk molecules. However, the Raman spectral changes for tryptophan are less clearly defined due to peak overlapping with other amino acid signals and the low concentration of tryptophan.The FT-IR (ATR) technique coupled with second derivative spectroscopic analysis has demonstrated the decrease in crystillinity degree and tyrosine content of UV/ozone irradiated silk, as indicated by the changes in FT-IR bands of Amide III doublet and tyrosine signals.
Broadband Multidimensional Spectroscopy Identifies the Amide II Vibrations in Silkworm Films

Abstract:We used two-dimensional infrared spectroscopy to disentangle the broad infrared band in the amide II vibrational regions of Bombyx mori native silk films, identifying the single amide II modes and correlating them to specific secondary structure. Amide I and amide II modes have a strong vibrational coupling, which manifests as cross-peaks in 2D infrared spectra with frequencies determined by both the amide I and amide II frequencies of the same secondary structure. By cross referencing with well-known amide I assignments, we determined that the amide II (N-H) absorbs at around 1552 and at 1530 cm–1 for helical and β-sheet structures, respectively. We also observed a peak at 1517 cm−1 that could not be easily assigned to an amide II mode, and instead we tentatively assigned it to a Tyrosine sidechain. These results stand in contrast with previous findings from linear infrared spectroscopy, highlighting the ability of multidimensional spectroscopy for untangling convoluted spectra, and suggesting the need for caution when assigning silk amide II spectra.

Fourier Transform Raman And Fourier Transform Infrared Spectroscopy Studies Of Silk Fibroin

Abstract:The chemical and conformational structures of Bombyx mori silk were Studied with the complementary techniques of Fourier transforrn Raman spectroscopy and Fourier transform infrared spectroscopy. The Fourier transform Raman spectrum of silk showed strong bands for the photosensitive aromatic amino acids tyrosine, tryptophan, and phenylalamne. Intensive UV/ozone irradiation reduced the Raman intensities of the amide TII band and the tyrosine signals due to peptide chain scission of the silk polymer and the photochemical changes in the tyrosine residues on the silk molecules. However, the Raman spectroscopy changes for tryptophan were less clearly defined because of peak overlapping with other amino acid signals and the low concentration of tryptophan. The Fourier transform infrared (attenuated total reflectance) technique, coupled with second-derivative spectroscopy analysis, demonstrated a decrease in the crystallinity degree and tyrosine content of UV/ozone-irradiated silk, as indicated by changes in the Fourier transform infrared bands of amide III doublet and tvrosine sianals. Abstract:The conformation transition of silk fibroin in the silk glands of Bombyx mori was studied by scanning tunneling microscopy, The dilute aqueous solution of fibroin synthesized in the posterior glands is in a form of random coils, which are transformed into coiled-coil and later into individual alpha-helices in the middle glands. During spinning, they turned into beta-strands and finally to beta-sheets. The formation of liquid crystalline state aids the process of spinning. The STM images were taken dozens of time for each sample at different spots, which can represent the general nature of fibroin in the glands of silkworm, So far as we know, this is the first time to have the individual fibroin, or more specifically, the alpha-helix and beta-sheet structure of protein, to be imaged.

Abstract:A new atomic force microscopy (AFM)-based technique, single-molecule force spectroscopy (SMFS), was used to study the nanomechanics of Bombyx mori silk fibroin. Three types of force-extension curves were found in the system. A modified freely jointed chain (MFJC) model can fit two of the three types well, but the fit parameters are different. The third type of force curve, in which there exists a plateau, cannot be modeled by MFJC. These results may show that there are three kinds of conformations in the silk fibroin system and SMFS can "distinguish" different conformations of the polymer chains.
