Abstract:Using a simple fluorogenic reaction and the inner filter effect, a label-free ratiometric method for biothiol detection is developed. The reaction between dopamine and resorcinol can generate azamonardine with strong fluorescence. Since the absorption spectrum of azamonardine overlaps with the excitation spectrum of the mercaptopropionic acid-stabilized CdTe quantum dots (MPA-CdTe QDs), the emission of QDs can be quenched via the inner filter effect in the presence of azamonardine. Biothiols can inhibit the above fluorogenic reaction, resulting in the weakening of the fluorescence of azamonardine and the recovery of the emission of MPA-CdTe QDs. The fluorescence intensity ratio of azamonardine to MPA-CdTe QDs can be used to detect biothiols. This approach exhibited a linear trend to the biothiol concentrations in the range of 2 μM to 12 μM, with a limit of detection of 0.6 μM (S/N = 3). This ratiometric analysis strategy was further applied to detect biothiols in human serum samples, showing their great application potentials in real biological samples. The constructed method is simple, easy to prepare, and low-cost.

Abstract:Fluorescent materials with lanthanide cations encapsulated in MOFs are currently used in numerous applications, especially in biosensors. Therefore, herein, two novel composites were designed and developed based on a Tb(iii)&Yb(iii)-functionalized Cu(ii)-coordination polymer, possessing higher thermal and water stability and fascinating fluorescence properties. The first bimetallic composite Tb@Cu-Hcbpp demonstrated broad ligand-centered emission and weak typical Tb3+ ion emission; moreover, it was used as an excellent ratiometric fluorescent sensor for the metabolic product NMF of DMF in the human body (LOD = 0.02 mu M). In addition, the Yb3+ ions were doped into Tb@Cu-Hcbpp to improve the fluorescence performance of the green Tb3+ ion emission. Among the series of Tb1-xYbx@Cu-Hcbpp samples (x = 0.05, 0.10, 0.15, 0.20, 0.25, 0.30, 0.35 and 0.40), Tb0.85Yb0.15@Cu-Hcbpp showed maximum enhanced fluorescence intensity (almost 9.6 times that of the pure terbium system), but exhibited high fluorescence quenching efficiency for methylglyoxal (MGO), which could be used for the sensitive detection of MGO (LOD = 0.25 mu M). Furthermore, the developed biosensors were successfully applied for the detection of NMF and MGO in urine and serum samples, and satisfactory results were obtained, showing good potential of these biosensors in practical applications such as in disease diagnosis and biochemical research.

Abstract:The rapid advance and growth of the point-of-care diagnosis industry has provided an impetus for the development of novel signal labels for highly sensitive bio-molecule detections. Colloidal quantum dots (QDs) exhibit superior brightness, facile surface functionalization and exceptional photostability, making them the preferred option for these biological applications. However, the significantly reduced fluorescence intensity and limited photochemical stability in complex biological environments have greatly hampered their further use. Herein, with an innovative alloying engineering strategy, the high-quality water-soluble CdSe/CdxZn1−xS QDs with near-unity PL quantum yield and monoexponential PL decay dynamics are obtained. Notably, for the first time, a record-breaking stability at the single QD level in water with nonblinking behavior persisting for an hour is achieved, which approaching those of state-of-the-art hydrophobic QDs. Furthermore, the nanocomposites formulated with these novel alloyed QDs demonstrate a remarkable PL QY of 96 % and provide an ultrasensitive detection for prostate specific antigen on lateral flow immunoassays. These findings presented here shed new light on the design of high-brightness water-soluble QDs in single-molecule level and QDs-based nanocomposites, significantly pushing ahead toward high-sensitivity biomedical detections and diagnosis.
Luminescent Lanthanide-Based Organic/Inorganic Hybrid Materials for Discrimination of Glutathione in Solution and Within Hydrogels.

Abstract:Glutathione (GSH) as a biothiol is an essential peptide related to various diseases. Although multiple strategies for biothiols detection have been developed, there is increasing demand for sensors that can differentiate GSH from cysteine (Cys) and homocysteine (Hcy), owing to the similar structures and thiol groups in these amino acids. Herein, we report a novel Eu3+/LAPONITE (Lap)-based organic/inorganic hybrid material for selective detection of GSH via an "off-on" process. The fluorescence of Eu(DPA)3@Lap-Tris can be quenched by Cu2+ through photoinduced electron transfer (PET). The addition of GSH into the Eu(DPA)3@Lap-Tris/Cu2+ system induces the removal of Cu2+ from Eu(DPA)3@Lap-Tris and blocks PET, resulting in the recovery of fluorescence. This proposed assay demonstrates higher selectivity toward GSH than Cys and Hcy, and showed a detection limit of 162 nM within a linear range of 0.5-30 μM. Unlike other GSH selective sensors, this platform could be formed into a hydrogel while its sensitivity was maintained. The sensitive response to GSH in serum samples makes this platform an efficient tool for biological applications because of its ease of preparation, high selectivity, good biocompatibility, and low toxicity.
A Fluorescent Biosensor for Cardiac Biomarker Myoglobin Detection Based on Carbon Dots and Deoxyribonuclease I-aided Target Recycling Signal Amplification

Abstract:A sensitive biosensor using carbon dots and deoxyribonuclease I-aided target recycling signal amplification has been developed to detect myoglobin (MB), which is an important cardiac biomarker and plays a major role in the diagnosis of acute myocardial infarction (AMI). Here, in the absence of MB, the MB aptamer (Ap) is absorbed on the surface of carbon dots (CDs) through π-π stacking interactions, resulting in quenching of the fluorescent label by forming CD-aptamer complexes. Upon adding MB, the Ap sequences could be specifically recognized by MB, leading to the recovery of quenched fluorescence. Thus, quantitative evaluation of MB concentration has been achieved in a broad range from 50 pg mL-1 to 100 ng mL-1, and the detection limit is as low as 20 pg mL-1. This strategy is capable of specific and sensitive detection of MB in human serum, urine, and saliva and can be used for the diagnosis of AMI in the future.

Abstract:Transition metal dichalcogenide (TMD) dots exhibit excellent photoluminescence performance due to the quantum confinement effect and edge effect, and are extensively applied in electronic and optical devices, sensors, catalysis, and bioimaging. In this work, WS2 quantum dots (WS2 QDs) were prepared under a simple one-step hydrothermal method by optimizing the reaction conditions, and a quantum yield of 11.23% was achieved. The as-prepared WS2 QDs possess good photo-bleaching resistance, salt tolerance, and pH stability. The fluorescence investigations showed that the WS2 QDs acted as a highly efficient fluorescent sensor to detect hemoglobin (Hb) and cardiac biomarker myoglobin (Myo). The linear range was 1–600 μg/mL for Hb and 0.01–120 μg/mL for Myo, with detection limits as low as 260 and 7.6 ng/mL, respectively. Importantly, the WS2 QDs were used to determine the Hb/Myo content in human blood/serum samples, with satisfactory results, indicating that this technique holds promise for application in clinical diagnosis associated with Hb/Myo levels. To the best of our knowledge, this is the first example of TMD QDs without any modification as a fluorescent sensor for detecting Hb and Myo simultaneously.Graphical abstract

Abstract:Developing a highly sensitive fluorescence probe for biomolecule detection has a significant meaning in clinical diagnosis and research. Tuning the optical property of fluorescent nanomaterials by heteroatom doping is generally feasible to improve the application performance. In this work, a kind of cesium-doped graphene quantum dots (Cs-GQDs) are synthesized, characterized, and explored as a fluorophore in ultrasensitive determination of H2O2 and glucose. The prepared Cs-GQDs with a quantum yield of 39.8% eradiate intense blue fluorescence under UV irradiation. The emission of Cs-GQDs is almost excitation-independent. Doping Cs into the GQDs leads to a clear blue shift of the fluorescence emission peak, probably related to the increased band gap. In addition, Cs-GQDs show a pH-sensitive behavior under acidic conditions. In the presence of horseradish peroxidase, o-phenylenediamine could react with H2O2 to form a yellow fluorescent compound of 2,3-diaminophenazine (DAP). A ratiometric fluorescence probe toward H2O2 and glucose is established using the fluorescence resonance energy transfer mechanism between DAP and Cs-GQDs. This sensing strategy shows high sensitivity for H2O2 and glucose detection with detection limits of 25 and 23 nM, respectively, and excellent selectivity toward glucose out of various interferences. This method is extended to glucose detection in human serum samples. Abstract:A novel magnetic/fluorometric bimodal sensor was built from carbon dots (CDs) and MnO2. The resulting sensor was sensitive to glutathione (GSH), leading to apparent enhancement of magnetic resonance (MR) and fluorescence signals along with visual changes. The bimodal detection strategy is based on the decomposition of the CDs-MnO2 through a redox reaction between GSH and MnO2. This process causes the transformation from non-MR-active MnO2 to MR-active Mn(2+), and is accompanied by fluorescence restoration of CDs. Compared with a range of other CDs, the polyethylenimine (PEI) passivated CDs (denoted as pCDs) were suitable for detection due to their positive surface potential. Cross-validation between MR and fluorescence provided detailed information regarding the MnO2 reduction process, and revealed the three distinct stages of the redox process. Thus, the design of a CD-based sensor for the magnetic/fluorometric bimodal detection of GSH was emphasized for the first time. This platform showed a detection limit of 0.6 μM with a linear range of 1-200 μM in the fluorescence mode, while the MR mode exhibited a linear range of 5-200 μM and a GSH detection limit of 2.8 μM with a visible change being observed rapidly at 1 μM in the MR images. Furthermore, the introduction of the MR mode allowed the biothiols to be easily identified. The integration of CD fluorescence with an MR response was demonstrated to be promising for providing detailed information and discriminating power, and therefore extend the application of CDs in sensing and imaging.
Rapid and facile electrospray preparation of CsPbBr3@PMMA fluorescent microspheres for fluorescent detection of ALP in biological samples

Abstract:All-inorganic cesium lead halide perovskite quantum dots (QDs) have attracted more attention due to their excellent optoelectronic and photovoltaic properties. However, there are few applications in biochemical detection based on their unique photoluminescence property mainly because of the instability to external stimulus. In this work, we focus on a rapid preparation method of CsPbBr3@poly (methyl methacrylate) (PMMA) fluorescent composite microspheres based on electrospray to protect CsPbBr3 QDs from the external stimuli and stabilize them in aqueous environment. The CsPbBr3@PMMA composite microspheres effectively retained over 70% fluorescence intensity after being immersed in deionized water for 20 days. Based on the effective quenching of CsPbBr3@PMMA composite microspheres by MnO2 nanosheets, we developed a simple, label-free, low-cost, and facile fluorescent detection system for assay of alkaline phosphatase (ALP) in biological samples. The method shows a good linear relationship from 10.0 mU/mL to 100 mU/mL (R2 = 0.998) with the detection limit of 4.85 mU/mL and the recovery range of 95.1–110%. In , the PMMA coating can simply and effectively stabilize the CsPbBr3 QDs and preserve the luminescence of the CsPbBr3 QDs. The excellent applicability of the composite microspheres-based fluorescent detection system in human serum samples further demonstrates the potential application in the aqueous phase-based biochemical detection for clinical diagnosis and biomedical research.

Abstract:In this work, 3-aminophenylboronic acid (APBA) functionalized dual-emission quantum dots (QDs) were prepared and utilized as the ratiometric fluorescence probe for the onsite naked eye determination of glucose. The unique ratiometric fluorescence probe, which combined ratiometric fluorescence technique with boric acid functional materials into one system, has been prepared by hybridizing red-emitting CdTe QDs (r-QDs) and green-emitting CdTe QDs (g-QDs). The fluorescence of the embedded r-QDs stays constant, whereas the g-QDs functionalized with APBA can selectively bind glucose by the chemistry of boronic acid and cis diol compounds, leading to the fluorescence of g-QDs quenched due to the surface quenching states induced mechanism and resulting in continuous fluorescence color changes (from green to red) of the ratiometric fluorescence probe system. Under optimum conditions, the proposed ratiometric probe was used for selective detection of glucose in a concentration range of 0.1-2.0 mmol/L with a detection limit of 4.5 timol/L. Moreover, the developed ratiometric fluorescence probe was successfully applied to the detection of glucose in human serum samples. The present study provides an efficient and facile ratiometric fluorescence probe for rapid, convenient, sensitive and selective visual identification of glucose in blood glucose sensing without the need of complex equipment. Abstract:The poor stability and aqueous-quenching of fluorescence of perovskite nanocrystals (NCs) hinder their application in bio-detection and bio-imaging. Herein, through the synergistic effects of polymer surface ligand and silica encapsulation, highly stable and enhanced aqueous fluorescent CsPbBr3-mPEG@SiO2 NCs were synthesized and used as a novel "on-off-on" fluorescent probe for highly sensitive and selective detection of mercury ions (Hg2+) and glutathione (GSH) in aqueous solutions. The effects of the methoxypolyethylene glycol amine (mPEG-NH2) ligand and silica encapsulation on the stability and aqueous fluorescence of the CsPbBr3 NCs were studied. It indicated that the aqueous fluorescence of perovskite NCs was increased by 2.59 times. The water stability was also greatly improved, with the NCs maintaining 73% of their original fluorescence after storage for 30 days in water. X-ray diffraction (XRD), transmission electron microscopy (TEM) and Fourier transform infrared spectroscopy (FT-IR) analyses further demonstrated that the NCs were successfully passivated by mPEG-NH2 and silica. The fluorescence of the CsPbBr3-mPEG@SiO2 nanocrystals was effectively quenched by Hg2+ which is attributed to the electron transfer process between NCs and Hg2+. Then, through the interaction between Hg2+ and GSH, the restoration of fluorescence for CsPbBr3-mPEG@SiO2 was realized. The "on-off-on" fluorescent probe can be used for the detection of Hg2+ and GSH with a low detection limit of 0.08 nM and 0.19 μM, respectively. It also shows a fast response time and high accuracy for practical sample detection. The simple and sensitive fluorescent probe of CsPbBr3-mPEG@SiO2 shows great potential in environmental and biological sensing.

Abstract:Despite the excellent optoelectronic properties exhibited by CsPbBr3 QDs (PQDs) for sensing applications, their poor resistance to water does not allow their utilization as probes to detect analytes in aqueous media. The present work provides water soluble PQDs (dispersed in water) prepared by an appropriate phase engineering of the ligand. The dicarboxylate functional ligands at a particular pH allow the protonated state to form solvated carboxyl dimers, which interconnects PQDs, thus avoiding Ostwald ripening and enhancing the photoluminescence quantum yield (PLQY). As a proof of concept, this probe was applied to detect bioamines in water, namely histamine, hexamethylenediamine, phenethylamine, dopamine and thiamine. The probe is highly selective to histamine at concentrations below 500 nM and this selectivity of histamine over dopamine is very interesting and rarely reported. More importantly, this work offers a standard protocol for transferring PQDs from the organic to aqueous phase, for the detection of such biomolecules in water.

Aqueous, Non-Polymer-Based Perovskite Quantum Dots for Bioimaging: Conserving Fluorescence and Long-Term Stability via Simple and Robust Synthesis

Abstract:Perovskite quantum dots (PQDs) offer high photoluminescence quantum yields; however, due to their limited stability in aqueous media, to date their utilization in biomedical applications has been limited. The present work demonstrates highly fluorescent and stable aqueous PQDs that were synthesized using a facile engineered phase transfer method. Ligands were engineered to have a dual functionality, i.e., they could simultaneously mediate the strong binding of PQDs and the interactions with water molecules. The resultant water-soluble PQDs demonstrated robust structural and optical properties. The extracted aqueous PQDs remained stable in pellet form for 8 months, which was the entire test duration. Notably, 100% of their fluorescence was also retained. As a proof-of-concept experiment, the water-soluble PQDs were successfully tagged to polyclonal antibodies and used to image Escherichia coli cells in aqueous media. No structural or optical disturbance in PQDs was detected throughout the process. This work marks the beginning of the use of nonpolymeric aqueous PQDs and shows their strong potential to be used in biological applications.
Quantum Dots Based Mesoporous Structured Imprinting Microspheres for the Sensitive Fluorescent Detection of Phycocyanin

Abstract:Phycocyanin with important physiological/environmental significance has attracted increasing attention; versatile molecularly imprinted polymers (MIPs) have been applied to diverse species, but protein imprinting is still quite difficult. Herein, using phycocyanin as template via a sol-gel process, we developed a novel fluorescent probe for specific recognition and sensitive detection of phycocyanin by quantum dots (QDs) based mesoporous structured imprinting microspheres (SiO2@QDs@ms-MIPs), obeying electron-transfer-induced fluorescence quenching mechanism. When phycocyanin was present, a Meisenheimer complex would be produced between phycocyanin and primary amino groups of QDs surface, and then the photoluminescent energy of QDs would be transferred to the complex, leading to the fluorescence quenching of QDs. As a result, the fluorescent intensity of the SiO2@QDs@ms-MIPs was significantly decreased within 8 min, and accordingly a favorable linearity within 0.02-0.8 μM and a high detectability of 5.9 nM were presented. Excellent recognition specificity for phycocyanin over its analogues was displayed, with a high imprinting factor of 4.72. Furthermore, the validated probe strategy was successfully applied to seawater and lake water sample analysis, and high recoveries in the range of 94.0-105.0% were attained at three spiking levels of phycocyanin, with precisions below 5.3%. The study provided promising perspectives to develop fluorescent probes for convenient, rapid recognition and sensitive detection of trace proteins from complex matrices, and further pushed forward protein imprinting research.

Here, a pore-functionalized metal–organic framework (MOF) becomes a versatile platform for fluoro-sensing various preservatives and amino acids with higher emission and multicolorful scaffolds and allows for visible detection. The carbazole groups in a Zr-based MOF UiO-67-CDC (CDC = 9H-carbazole-2,7-dicarboxylic acid) are partially substituted into dimethylation through postsynthetic N-methylation to form a positively charged framework UiO-67-CDC-(CH3)2. Because of electrostatic induction, the postmodification skeleton can efficaciously encapsulate the anionic dye phloxine B (PB) in the cavity of the electron-deficient MOF, and a newfangled fluorescence “turn-on” behavior is ascribed due to the inhibition of photoinduced electron transfer (PET) (from the benzene rings to the quaternary ammonium group on the ligand) and the stunning synergistic effect of the confinement-induced emission (CIE) enhancement. PB@UiO-67-CDC-(CH3)2 shows a unique fluorescence response for preservatives and amino acids with variable colors and visual, quantitative discrimination effects, which could be applied for sensing, switching, and anticounterfeiting identification. Notably, the fluorescence emission peak of dye (PB) and MOF presents a ratiometric change, further improving the detection’s resolution and veracity. The sodium acetate and leucine detection limits are as low as 0.0157 μM and 0.1084 μM, respectively. In addition, a smartphone was used to visualize sodium acetate by identifying the RGB value of the fluorescent changes, and the LOD can be as low as 7.23 μM. These features make the dye-embedded MOF composite a simple, efficient, and instrument-free stratagem for the on-site and convenient detection of preservatives in various samples.

Abstract:Abstract Real‐time in vitro detection of glioma cells facilitates precise tumor removal. However, the fluorescent labeling of tumor cells in clinical practice is limited by many factors, including the time consumed, low recognition efficiency, and fluorescence quenching. Here, a general strategy for building perovskite quantum dot (PQD)‐based biological probes utilizing the attraction between positive and negative electric charges is reported. Poly (lactic‐co‐glycolic acid) (PLGA) is chosen for encapsulating PQDs to completely prevent their aggregation, decomposition, or release in water or oxygen. The carboxyl group of PLGA has anchoring coordination with the PQDs, which reduces the surface defects. Moreover, it causes PQD‐based nanocrystals (P‐PNCs) to be surrounded by a positively charged layer in water. Given the specific recognition of chlorotoxin for the channels, rapid imaging of glioma cells is successfully performed in 15 min using P‐PNCs modified with chlorotoxin via charge attraction. The photoluminescence quantum yield of P‐PNC probes reached 87% and remained at 93% after 30 days of dispersion in water, while maintaining a much longer fluorescence lifetime of 15 µs. Therefore, this promising biological probe will be a general nanoplatform for identifying distinct cellular compartments using different biomarker imaging methods.

Glucose and pH responsive fluorescence detection system based on simple synthesis of silicon-coated perovskite quantum dots

Abstract:Perovskite quantum dots (PQDs) are extremely unstable in ambient air due to their inherent structural instability, which limits the wide application of PQDs. In this work, silicon-coated CsPbBr3 PQDs (CsPbBr3@SiO2) was synthesized via a simple method. The SiO2 coating effectively isolated PQDs from water and oxygen in the environment, which were the main elements that destroyed the structure stability of PQDs. The synthesized CsPbBr3@SiO2 can be stored in water for more than 2 months and posessed wonderful dispersibility in aqueous solution. The fluorescence intensity remained unchanged within 7 days and only decreased by 11.9 % within 2 months. We found that CsPbBr3@SiO2 was extremely sensitive to environmental pH, and the fluorescence intensity decreased with the reduction of pH. In addition, an excellent linear relationship with pH value in the range of 1.0 ∼ 5.0 was achieved. As we all known that glucose can be catalyzed by glucose oxidase to produce gluconic acid and hydrogen peroxide, in which a good deal of protons were produced and the pH was gradually lowered. Since CsPbBr3@SiO2 was stable to water and oxygen, and sensitive to ambient pH, we applied CsPbBr3@SiO2 to the detection of glucose. CsPbBr3@SiO2 showed fantastic selectivity and sensitivity to glucose, and the detection limit can even reach 18.5 μM. Furthermore, CsPbBr3@SiO2 was successfully applied to the detection of glucose in the human serum with satisfactory performance.

Abstract:An "off-on"-type electrochemiluminescence (ECL) system for myoglobin (Myo) assay was proposed by coupling the resonance energy transfer (RET) strategy with functional liposome as probes. Specifically, black phosphorus nanosheets (BP NSs) as the energy donors were introduced into the ECL system and MnO 2 NSs as the energy acceptors were reduced on the surface of BP NSs in situ to form the BP/MnO 2 nanocomposites. Attributing to the perfect spectral match between BP NSs and MnO 2 NSs, efficient ECL-RET occur in the BP/MnO 2 nanocomposites. Glutathione (GSH) loaded liposomes as probes for signal regulation and signal amplification were immobilized in a 96-well plate through a sandwich immunoreaction. The abundant GSH lysed from liposomes could effectively reduce MnO 2 to Mn 2+ and destroyed the RET, thus regulating the recovery degree of ECL signal from BP NSs. The absence and presence of GSH generated the "signal off" and "signal on" states of the BP/MnO 2 nanocomposites-based ECL system. The amount of Myo can be estimated by the recovery degree of the signal. Benefiting from that, a good linear relationship ranging from 1.0 × 10 -13 to 1.0 × 10 -7 g mL -1 was found and the detection limit is down to 2.5 × 10 -14 g mL -1 for Myo. Furthermore, the ECL system revealed favorable performance in the determination of Myo in human serum samples.

Abstract:In this paper, a new fluorescence bioassay for Golgi protein-73 (GP73), a promising marker for monitoring liver tumor, was developed by using anti-GP73 antibody (GP73 Ab) capped quantum dots (QDs) coupled with protein A/G agarose beads in an attempt to improve the analysis time, cost and operation. First, carboxylic-functionalized Mn modified CdTe/CdS QDs were synthesized and covalently conjugated with GP73 Ab, then protein A/G agarose beads were specifically combined with the QDs-conjugated Ab to form the QDs-Ab-beads conjugate, which could capture and separate GP73 from the sample through simple centrifugation. It was found that the fluorescence intensity of the above QDs-Ab-beads biosensor could be specifically quenched by GP73 added. A simple, rapid and specific quantitative method for GP73 protein was proposed using the as-prepared QDs-Ab-beads as a biosensor. Under the optimized conditions, the calibration curve of the proposed assay showed good linearity with a correlation coefficient of 0.9935 in the concentration range of 20-150 ng/mL of GP73 protein. The limit of detection (defined as 3σ/K) was 10 ng/mL. The method built exhibited a great potential in the clinic test of GP73.
